This thesis expands the scope for using the permethylpentalene ligand and its precursors in the synthesis of organometallic complexes. Chapter one begins with a brief review of linked metallocenes, with which multimetallic compounds bridged by pentalene ligands have often been compared, followed by a comprehensive review of the routes used to make pentalenes and substituted pentalenes. Organometallic compounds of pentalenes are introduced, with a focus on bimetallic systems. Chapter two explores the diversification of substituents added to the permethylpentalene (Pn*) precursor WeissH4, to include ethyl and isopropyl groups. Low-symmetry mono-, di-, tri- and tetraalkylated products are formed, eight such organic molecules have been identified by NMR spectroscopy, and two characterised crystallographically. It has been demonstrated that subsequent hydrolysis and decarboxylation of two of these products produces low-symmetry alkylpentalene precursors. The chapter concludes with discussions on the selectivity exhibited in these reactions, and the assignment of stereochemistry. Chapter three describes the synthesis of the first homoleptic double metallocene complex of iron. Fe2Pn*2 has been characterised by X ray diffraction, and cyclic voltammetry studies demonstrate four accessible oxidation states (-1, 0, +1, +2). Magnetic measurements in the solid and solution state reveal an unusual triplet configuration, and DFT calculations indicate the origin of a high magnetic moment likely resides in unquenched orbital angular momentum contributions from SOMOs which have metal d character. Fe2Pn*2 is EPR silent at 5, 40, and 300 K both in solution and the solid state, suggesting a large zero-field splitting parameter. The reaction of the di-iron complex with carbon monoxide, ethylene and H2 is reported; the bimetallic CO adduct, Fe2(μ η5,η3 Pn*)(μ η5,η1 Pn*)(CO)2, has been crystallographically characterised, and contains a highly distorted allylic bonding motif, which to the author’s knowledge is believed to be unique among iron complexes. Chapter four discusses the interaction of the bidentate Pn* ligand in anti bimetallic fused metallocenes. A new ligand exchange route has been developed to access the complexes (MCp)2Pn* (M = Co, Ni), and the isostructural complexes (MCp*)2Pn* have been made for M = Fe, Co, Ni by salt metathesis reactions. All five complexes have been characterised by single crystal X-ray crystallography, and have diamagnetic ground states in solution in common with their Pn bridged analogues. Variable temperature NMR studies reveal a spin-equilibrium between S = 0 and S = 1 in the dinickel complexes. DFT calculations reproduce the spin states found, and suggest the distortion towards η3 coordination observed on crossing from Fe, to Co, to Ni, results from population of orbitals with M―bridgehead antibonding character. The electronic structures show it is important to draw comparisons between isoelectronic linked metallocenes. Electrochemical studies on the diiron, dicobalt, and (NiCp)2Pn* complexes reveal at least three redox events for each. Chapter five documents the successful synthesis and characterisation of monometallic complexes of iron and manganese with Pn*H ligands. The isostructural complexes Fe(Pn*H)2 and Mn(Pn*H)2 can have been characterised crystallographically, and are potential precursors for accessing heterometallic, and multimetallic complexes. Mn(Pn*H)2 is a rare example of a manganese sandwich compound and magnetic studies on a single isomer in the solution and solid states suggest it adopts intermediate spin states of S = 2 in solution, and S = 3/2 in the solid state. Chapter six gives experimental details for all syntheses and studies described in the preceding chapters. Chapter seven provides characterising data for all new compounds. Fitting data for VT NMR and SQUID studies are provided in the appendix at the end of this thesis. Crystallographic data in the form of .cif files, DFT output files, and raw SQUID data, can be found in the electronic appendix.

The work in this Thesis addresses the challenges associated with using redox enzymes for chemical synthesis. The use of enzymes as catalysts in the synthesis of fine chemicals is becoming more wide spread, in part due their ability to catalyse reactions with incredible selectivity under relatively mild conditions. In particular, enzymes are useful for selective reduction of ketones to enantiomerically pure alcohols or amines, and partial oxidations of alkanes to alcohols. However, a key limitation to exploiting redox enzymes in these reaction pathways is the requirement for a specialised electron source, usually the expensive nicotinamide cofactors NADH or NADPH. Existing cofactor regeneration methods use a second enzyme with a sacrificial substrate which is oxidised to generate a stoichiometric waste product; this complicates isolation of the desired product and prevents the environmental benefits of biocatalysis from being fully realised. In order to provide clean and efficient biocatalytic routes, improved recycling methods for these cofactors are crucial. This Thesis develops two novel methods for in situ cofactor recycling. The first is an electro-enzymatic system; an NAD+-reductase enzyme is shown to use electrons directly from an electrode for supply of NADH to a co-immobilised cofactor-dependent enzyme. The second uses a hydrogenase, NAD+ reductase and cofactor-dependent enzyme immobilised on conducting particles for H2-driven NADH regeneration. This relies on the thermodynamically favourable reduction of NAD+ by H2 when the hydrogenase and NAD+-reductase are in electronic contact, provided by the conducting particle. The electro-enzymatic approach to NAD+ reduction is then adapted for electrochemical devices; an enzyme catalysed fuel cell and a self-powered biosensor were considered.

The covalent chemistry of individual reactants bound within a protein nanopore can be monitored by observing the ionic current flow through the pore, which acts as a nanoreactor responding to bond-making and bond-breaking events. However, chemistry investigated in this way has been largely confined to the reactions of thiolates, presented by the side chains of cysteine residues. The introduction of unnatural amino acids would provide a large variety of reactive side chains with which additional single-molecule chemistry could be investigated. An efficient method to incorporate unnatural amino acid is semisynthesis, which allows site-specific modification with a chemically-defined functional group. However, relatively little work has been done on engineered membrane proteins. This deficiency stems from attributes inherent to proteins that interact with lipid bilayer, notably the poor solubility in aqueous buffer. In the present work, four different derivatives α-hemolysin (αHL) monomer were obtained either by two- or three-way native chemical ligation. The semisynthetic αHL monomers were successfully refolded to heptameric pores and used as nanoreactors to study single-molecule chemistry. The semisynthetic pores show similar biophysical properties to native αHL pores obtained from an in vitro transcription and translation technique. Interestingly, when αHL pores with one semisynthetic subunit containing a terminal alkyne group were used to study Cu(I)-catalyzed azide-alkyne cycloaddition, a long-lived intermediate in the reaction was directly observed.

In this work, the use of modified diamond as an electrode material with superlative physical and electrochemical properties was investigated in a number of electrochemical applications. The surface chemistry of three differing forms of diamond, namely boron-doped microcrystalline diamond, boron-doped diamond powder and detonation nanodiamond powder was modified utilising such strategies as hydrogen plasma treatment, reactive ion plasma etching along with various chemical treatments. The surface and functional properties of the modified diamond electrodes were studied using a wide spectrum of techniques. The electrochemical activity of these materials was concomitantly investigated in order to expand the knowledge of diamond electrochemistry and to establish an understanding of how the surface chemistry of these materials impacts their electrochemical performance. In the first study, the nanostructuring strategies of boron-doped diamond surface with platinum nanoparticles were developed. In particular, two types of diamond nanostructures were produced: one consisting of platinum particles located on the top of diamond nanorods, the other with platinum particles located in the bottom of diamond nanopits. For the first time, the experimental evidence proving the mechanism of the diamond nanostructuring process was reported. The electrochemical activity of these nanostructured diamond electrodes with regard to the electrochemical oxidation of glucose and methanol was investigated. In the second study, the relationship between the surface chemistry of three differing forms of diamond, including microcrystalline boron-doped diamond, boron-doped diamond powder as well as detonation nanodiamond powder, and the electrode fouling in the result of the adsorption processes in methyl viologen and anthraquinonedisulfonate solutions was investigated. The influence of two dissimilar surface terminations: hydrophobic H-terminated and hydrophilic O-terminated on the electrode performance was studied in detail. This work provides a useful insight on the likely reasons for the undesirable adsorption occurrence which may be experienced in many electroanalytical applications that utilise solid and powdered forms of diamond. The third project extends the discussion on the study of the diamond electrodes, modified with detonation nanodiamond and boron-doped diamond powders and investigates the electrochemical behaviour of these materials. In this work, charge transport within the diamond powder films, partition coefficients of different redox mediators along with heterogeneous electron transfer constants were identified. The chemical modification of these electrodes with platinum nanoparticles along with the mechanism of nucleation and growth of the latter were studied. The enhanced electrode performance with regard to methanol electrooxidation reaction was demonstrated. The fourth study investigates the preparation of nickel modified boron-doped diamond electrodes and ascertains the relationship between the surface chemistry of the modified diamond and the associated electrocatalytic performance of nickel nanoparticles in hydrogen peroxide and glucose electrooxidation. The fifth study reports on the development of a novel surface functionalization strategy, based on porphyrin and amide coupling chemistry, which allows the creation of hybrid biomimetic diamond interface that was used as the artificial β-alanine receptor.

Fuel cell is a device that can directly convert chemical energy into electrical energy. For low-temperature fuel cells, catalysts are required. Fuel cells using Pt-based or other non-biological materials as catalysts are known as conventional fuel cells. Inspired from Nature, enzymes can be used as catalysts in fuel cells known as enzyme-based fuel cells. The conventional and enzymatic fuel cells share the same underlying electrochemical principles, while enzyme-based fuel cells have their intrinsic advantages and disadvantages due to enzyme properties. The objective of this thesis is to investigate the current/voltage/ power/stability capabilities of enzyme-based membrane-less H2 fuel cells in order to design the enzymatic fuel cells with improved performance. This thesis presents a facile, effective method for the construction of 3D porous carbon electrodes. The 3D porous carbon electrodes are constructed by compacting suitable carbon nanomaterials into discs. The 3D porous carbon electrodes, with large roughness, high specific surface area, and optimized pore size distribution, are able to increase the loading density of enzymes, that is, reaction sites per unit geometric electrode area. The high loading density of enzymes can result in the high current/power density of the enzyme-based membrane-less H2 fuel cells. Moreover, the large enzyme loading can bring about the improvement in fuel cell stability because current becomes limited by mass transport of dissolved gases rather than enzyme immobilization so that neither inactivation nor desorption of enzymes would influence the current output. Based on one type of 3D porous carbon electrodes, the maximum power density of enzyme-based membrane-less H2 fuel cells has increased to the mW•cm2 level by at least one order of magnitude and the half-life has also increased from several hours to one week. This thesis presents a method for the increase in power density otherwise limited by low cathodic currents due to meagre O2 in non-explosive H2-rich H2-air mixtures. The power density of enzyme-based membrane-less H2 fuel cells can be increased by re-proportioning cathode/anode geometric area ratio to balance the cathodic and anodic currents under such an unusual H2-air mixture. This thesis also demonstrates that the 3D porous carbon electrode can improve the apparent O2 tolerance of anodic catalysts – hydrogenases, which are very important for the fuel cell performance. The degrees of apparent O2 tolerance for both O2-tolerant and O2-sensitive [NiFe]-hydrogenases are greatly increased based on the 3D porous carbon electrodes, so that even an O2-sensitive [NiFe]-hydrogenase can be used as an anodic catalyst in the enzyme-based membrane-less H2 fuel cell under a non-explosive H2-rich H2-air mixture. This thesis presents a design of a test bed in which series and parallel connections of sandwich-like electrode stacks can be varied. The fuel cell test bed has demonstrated low-loss interconnects and efficient stack configuration. Operated under a non-explosive H2-air mixture containing only 4.6% O2 at 20 °C, the maximum volume power density of the fuel cell test bed exceeds 2 mW•cm3, capable of powering electronic gadgets, which is a good demonstration of electricity that originates from the buried active sites of enzymes and is transmitted by long-range electron hopping in accordance with Marcus theory.

This thesis explores the capacity of sterically and electronically unsaturated boranes to bind substrates of biological and environmental interest, and transduce such binding events into a photo-physical and/or electrochemical response, hence reporting the presence of these substrates. Chapter three details the synthesis of a range of novel ferrocenyl boranes featuring either a proximal hydrogen-bond donor or a second Lewis acidic centre. These novel boranes were shown to be competent at binding both cyanide and fluoride anions, with the role played by a proximal hydrogen-bond donor or a second Lewis acidic centre in anion binding investigated by both NMR and crystallographic studies. Chapter four reports the synthesis of novel pyridinyl and related boronic esters, as well as unexpected mixed alkenyl/aryl boranes. The capacity of both types of system to bind fluoride or cyanide anions in solution was investigated by UV-Vis and NMR studies. The photo-physical responses to these anions were also probed, leading to the establishment of both switch-on and switch-off fluorescent responses. Chapter five extends the knowledge derived from selective anion receptor design and combines this with recent developments in the field of frustrated Lewis pairs (FLPs) to activate, bind and report the presence of nitrous oxide (N2O) molecule. Thus, the syntheses of novel, highly Lewis acidic ferrocenyl boranes that incorporate a high degree of steric loading around the boron centre are reported. The electrochemical and photo-physical response of an FLP system to the presence of N2O was investigated leading to the development of a novel N2O reporting system.

Nanoelectrochemistry explores the differences in chemical behaviour at the nanoscale as compared to the macro-scale. This thesis is concerned with nanoelectrochemistry and aims to develop and apply novel experiments for the unambiguous identification of changed electrode kinetics at the nanoscale. This is challenging since electrochemical responses are controlled by diverse factors like enhanced mass transport and adsorption as well as electron transfer kinetics. A joint computational and experimental strategy is employed. Chapter 1, 2 and 3 cover essential introductory material and basic experimental details relevant to all experiment. Fuller descriptions and details are given in the following chapters as and when needed. Chapter 4 reports the development of an electrochemical characterization method, to achieve a fast and simple quantification of the average particle size and the number of nanoparticles deposited on a glassy carbon electrode. The method consists of surface area characterization by underpotential deposition of lead particles and the determination of the amount of gold from anodic stripping in HCl. This method is also proven to be effective by comparing the results with SEM measurements. Next, in chapter 5, a generic strategy combining computation and experimental approach is developed in order to study the electron transfer kinetics of gold nanoparticles. The modelling part considers the kinetics of the electrochemical process on the bulk materials for different regions in the electrode, that is, the substrate (glassy carbon) and the nanoparticles (gold). Comparison of experimental and theoretical results enables the detection of changes in the electrode kinetics at the nanoscale. This approach is applied into the electro-oxidations of nitrite and L-ascorbic acid for gold nanoparticles from 20 - 90 nm. In the former, analysing the system shows that no change in electron transfer kinetics is involved in the process, even though a decrease of the over-potential and an increase in the peak current are observed. But these changes reflect mass transport effects, not electrocatalysis. A case where an authentic enhanced electron transfer kinetic change occurs is shown for the ascorbic acid system. Finally, in chapter 6 , the above strategy is exploited further to apply a quantitative study of electron transfer kinetics for various sizes of gold nanoparticles in the oxygen reduction reaction system in sulphuric acid at 298 K. The latter is at the heart of energy transformation techniques (fuel cells, battery and so on). Compared with the electron transfer kinetics on macro gold electrodes, there is no change at gold nanoparticles from size 5 nm to 40 nm. However, in the presence of Pb(II), a strong enhancement of electron transfer kinetics is observed on 5 nm citrate capped gold nanoparticles for ORR. On the other hand, a significant decrease of electron transfer kinetics has been found for gold nanoparticles of size 2 nm for ORR. The latter observation of strong negative electrocatalysis is also observed for the hydrogen evolution reaction (HER). This represents the first report of such effects with the HER system. Overall the thesis has established a rigurous, theoretical basis for evaluating electrocatalysis in nanoparticulate system.

Electrochemical analysis using mercury electrodes has been a prominent methodology since the beginning of voltammetry, however, due to its toxic nature their use is being slowly phased out in favour of environmentally friendlier alternatives. In this thesis, carbon electrodes feature heavily as a means to provide a cheaper and non-toxic approach to voltammmetric sensing. Pulse techniques have been used for the sensitive detection of commonly studied analytes, including antimony and iron. A simple fabrication of a vibrating electrode is presented by modifying a commercially available vibrating toothbrush with platinum foil as a means to enhance mass transport for electroanalytical work. Finally, pH determination at carbon electrodes is investigated. Carbon electrodes have been proposed as a simple sensor for pH determination by exploiting the pH sensitive nature of surface quinone groups intrinsic within carbon edge-plane like sites. Using this approach, both EPPG and GC electrodes are suggested as new materials for pH determination and can be used in aqueous solutions over the pH range 1.0 to 13.0, as a cheap and simple alternative to the classic glass electrode.

[NiFe] hydrogenases are metalloenzymes that act as highly efficient molecular electrocatalysts for the interconversion of protons and molecular hydrogen. Unlike any other known molecular electrocatalyst, the members of a subgroup of respiratory membrane-bound [NiFe] hydrogenases are able to maintain H2 catalysis in the sustained presence of O2. This O2-tolerance depends on the ability to respond to oxidative inactivation by O2 by exclusively forming rapidly reactivated active site states, thus implying a catalytic cycle in which O2 acts as a competing substrate to H2. Using isotope ratio mass spectrometry it is proven that the O2-tolerant Escherichia coli Hydrogenase 1 responds to O2 attack by acting as a four-electron oxidoreductase, catalysing the reaction 2 H2 + O2 → 2 H2O, equivalent to hydrogen combustion. Special features of the enzyme’s electron relay system enable delivery of the required electrons. A small fraction of the H2O produced arises from side reactions proceeding via reactive oxygen species, an unavoidable consequence of the presence of low-potential relay centres that release electrons from H2 oxidation. While the ability to fully reduce O2 to harmless H2O at the active site to generate the rapidly reactivated state Ni-B, determines if a hydrogenase is O2-tolerant, the ratio of oxidative inactivation to reductive reactivation rates determines how tolerant the enzyme is. It is shown by protein film electrochemistry that the (αβ)2 dimeric assembly of Hyd-1 plays an important role in O2-tolerance by aiding reactivation of one catalytic unit through electron transfer from the other. The teamwork between two redundant partners implicates a new role for dimerisation and represents a new example of cooperativity in biology. Finally, the non-natural amino acid p-azido-L-phenylalanine was synthesised and incorporated into Hyd-1, testing the possibility of introducing labels at specific sites.

Hydrogenases catalyse the interconversion of H2 and H+. Protein Film Electrochemistry (PFE), a technique in which a redox enzyme is adsorbed directly onto an electrode, enables a detailed description of the catalytic function of these metalloenzymes to be obtained. Unlike small-molecule electrocatalysts, the hydrogenase active site is surrounded by a protein structure ensuring that it is relatively unperturbed by the electrode surface. In this thesis, PFE is used alongside mathematical modelling to explain differences between [NiFe]- and [FeFe]-hydrogenases, highlighting some important considerations for efficient, reversible electrocatalysis. This thesis probes the unusual reaction between [NiFe]-hydrogenases and cyanide. Through a detailed study utilising PFE, Electron Paramagnetic Resonance (EPR) and Attenuated Total Reflection Infrared spectroelectrochemistry (ATR-IR), it is demonstrated that cyanide promotes the formation of the inactive Ni-B state. Preferred formation of the Ni-B state over more slowly reactivating Unready states is considered an important characteristic of the O2-tolerant class of [NiFe]-hydrogenases. The nature of the Ni-L state, commonly thought to be an artefact formed when a [NiFe]-hydrogenase is exposed to visible light, is probed via EPR and ATR-IR. In this thesis, the Ni-L state is shown to occur in samples of Hydrogenase-1 from Escherichia coli that have not been exposed to visible light, calling into question the true nature of this state. Finally, this thesis details the first study in which PFE is used to investigate the spontaneous incorporation of a synthetic active site mimic complex into apo-hydrogenase. Incorporation into apo-hydrogenase from Chlamydomonas reinhardtii and Clostridium pasteurianum is discussed, in both cases resulting in fully functional [FeFe]-hydrogenase, electrochemically indistinguishable from the native enzyme.

This thesis presents the development and redesign of a commercial electrochemical quartz crystal microbalance with dissipation (E–QCM–D). This was used to study factors affecting the efficiency of the four electron reduction catalysed by the fuel cell enzyme bilirubin oxidase from Myrothecium verrucaria immobilised on thiol modified gold surfaces. Within this thesis, the E–QCM–D was used to show that application of a constant potential to bilirubin oxidase adsorbed to thiol-modified gold surfaces causes activity loss that can be attributed to a change in structural arrangement. Varying the load by potential cycling distorts the enzyme by inducing rapid mass loss and denaturation. Attaching the enzyme covalently reduces the mass loss caused by potential cycling but does not mitigate activity loss. Covalent attachment also changes the orientation of the surface bound enzyme as verified by the position of the catalytic wave (related to the overpotential for catalysis) and reactive labelling followed by mass spectrometry analysis. The E–QCM–D was used to show how electrostatic interactions affect enzyme conformation where high pH causes a reduction in both mass loading at the electrode and a reduction in activity. At pH lower than the enzyme isoelectric point, there is a build up of multilayers in a clustered adsorption. When enzyme adsorbs to hydrophobic surfaces there is a rapid denaturation which completely inactivates the enzyme. Changing the surface chemistry from carboxyl groups to hydroxyl and acetamido groups shows that catalysis is shifted to more negative potentials as a result of an enzyme misorientation. Further to this, increasing the chain length of the thiol modifier indicates that an increased distance between surface and enzyme reduces activity, enzyme loading and results in a conformational rearrangement that permits electron transfer over longer distances.

Graphene, as a recently discovered carbon allotrope, possesses with it many outstanding properties ranging from high electrical conductivity to great mechanical strength. Single layer graphene can be prepared by mechanical cleavage of graphite or by a more sophisticated method, CVD. However, the scale-up process for these preparation techniques is still unconvincing. Solution-processed graphene from exfoliation of graphite oxide on the other hand provides an alternative prospect resulting in the formation of graphene nanoplatelets (GNPs), which can be readily manipulated to tailor-suit various application demands. The main aim of the thesis is to explore the possibility and availability of this versatile method to produce graphene nanoplatelet and its composites with good all-round performance in energy and bioanalytical applications. A range of physical and chemical characterisation techniques were utilised including SEM, TEM, AFM, XPS, XRD, DLS, FTIR, Raman and UV-Vis spectroscopy in order to investigate the structural and chemical information of the graphene-based materials prepared. Functionalisation of graphene oxide with polyelectrolyte polymer could facilitate deposition of platinum nanoparticles in the formation of Pt-GNPs composites. The resultant composite was employed for bioanalytical application in the detection of an important neurotransmitter, glutamate, based on glutamate oxidase enzyme. The performance of Pt-GNPs based glutamate sensor exhibited enhanced sensitivity and prolonged stability compared to the sensors based on Pt decorated diamond or glassy carbon electrodes. The significant interfering effect from concomitant electrochemically active biological compounds associated with Pt-GNPs electrode however could be alleviated via opting for Prussian blue deposited GNPs electrode instead. The oppositely charged Pt-GNPs due to different functionalising polymers were also subject to self-assembly, which was enabled by the electrostatic interaction of the opposite charges of Pt-GNPs. The self-assembled film showed enhanced mechanical stability than the conventional drop-casted film and provided reasonably good activity towards oxidation of hydrogen peroxide. Three-component composite of graphene, nanodiamond and polyaniline was prepared via in-situ polymerisation for usage as an electrode material in electrochemical capacitors ("supercapacitors"). The addition of graphene was shown to significantly enhance specific capacitance while nanodiamond could improve the stability of the electrode by strengthening the polymer core. Another approach to produce a supercapacitor was via electrodeposition of nickel and cobalt hydroxides on graphene oxide film corporated with bicarbonate salt. The film was then subject to thermal reduction of GO and expansion of graphene layers within the film was observed. This leavening process enhanced the surface area of graphene film and thus the higher specific capacitance was obtained. The decoration of nickel and cobalt hydroxides onto the film also boosted the specific capacitance further however the poor cycling stability of the heated film still remained an issue. Graphene nanoplatelets were also used as a support for electrodeposition of Pt nanoparticles for methanol oxidation in acidic media. The preferential phase of the Pt deposited and large surface area of graphene in comparison to other carbon supports studied led to good catalytic activity being observed.

A major problem in the quantitative analysis of AC voltammetric data has been the variance in results between laboratories, often resulting from a reliance on "heuristic" methods of parameter estimation that are strongly dependent on the choices of the operator. In this thesis, an automatic method for parameter estimation will be tested in the context of experiments involving electron-transfer processes in solution-phase. It will be shown that this automatic method produces parameter estimates consistent with those from other methods and the literature in the case of the ferri-/ferrocyanide couple, and is able to explain inconsistency in published values of the rate parameter for the ferrocene/ferrocenium couple. When a coupled homogeneous reaction is considered in a theoretical study, parameter recovery is achieved with a higher degree of accuracy when simulated data resulting from a high frequency AC voltammetry waveform are used. When surface-confined reactions are considered, heterogeneity in the rate constant and formal potential make parameter estimation more challenging. In the final study, a method for incorporating these "dispersion" effects into voltammetric simulations is presented, and for the first time, a quantitive theoretical study of the impact of dispersion on measured current is undertaken.

This thesis is concerned with the simulation of various electrochemical experiments and its application to electroanalysis. Chapter 1 outlines the fundamental principles of electrochemistry which are of importance for the reading of this thesis. Chapter 2 then outlines the methods used in the numerical simulation of electrochemical experiments. Chapters 3 and 4 are concerned with the electrochemistry of nanoparticles, and how this is affected by the presence of near wall hindered diffusion. In Chapter 3, a computational model to simulate anodic particle coulometry of nanoparticles in the presence of hindered diffusion is developed, and the effect of this hindered diffusion investigated. The model is then applied to simulate experimental data. Chapter 4 looks at the effect of hindered diffusion on the adsorption of nanoparticles on electrode surfaces, and investigates the effects of this adsorption on electrochemical experiments with nanoparticles generally. Chapters 5, 6 and 7 are concerned with band electrodes in isolation, in a pair and in an array respectively. In Chapter 5, a model to simulate double potential step chronoamperometry at an individual band electrode is developed, and used to successfully simulate experimental data. Chapter 6 looks at dual band electrodes used in generator-collector mode, and how this can be used to simultaneously measure the concentration of two species in solution. Chapter 7 looks at interdigitated arrays of band and ring electrodes in generator-collector mode, and develops a model to simulate cyclic voltammetry in both cases, as well as investigating under what conditions interdigitated ring arrays may be modelled as interdigitated band arrays. Chapter 8 develops a model to simulate chronoamperometry and cyclic voltammetry at porous electrodes, and investigates the consequences for electroanalysis of having a porous layer. Finally, Chapter 9 investigates the Marcus-Hush theory of electron transfer kinetics, and looks at the effect of the kinetically limited currents resulting from this theory to the equivalence relation between microdisc electrodes and sphere-on-a-surface electrodes.

This thesis presents experimental work with the primary aim of developing new approaches for the detection and characterisation of nanoparticles via electrochemical methods. The first chapter introduces the fundamental aspects of electrochemistry while the second chapter discusses the need for nanoparticle detection methods and the nonelectrochemical and electrochemical techniques that are currently used in the measurement of nanoparticles. A novel way to quantify silver nanoparticles in aqueous solution is proposed via nanoparticle-electrode impact experiments. In this technique a suitably potentiostatted electrode is immersed in a nanoparticle solution so as to bring about the oxidation or reduction of a single nanoparticle upon its collision with the electrode surface. This “direct” nanoparticle impact technique is then employed to detect laboratory synthesised silver nanoparticles in seawater. It is further shown that this method is capable of sizing silver nanoparticles contained in a commercially available cleaning product. Commercial silver nanoparticles are subsequently monitored via a sticking and stripping technique where homemade gold electrodes fabricated from CDs are immersed in a seawater sample spiked with nanoparticles prior to stripping voltammetry. The reduction of hydrogen peroxide on the surface of silver nanoparticles impacting upon an electrode is also examined. This “indirect” nanoparticle detection method is shown to provide an accurate route to nanoparticle sizing. A Fickian model is subsequently proposed to describe nanoparticle transport to the substrate electrode in both direct and indirect nanoparticle detection techniques. The importance of determining the proportion of nanoparticles which adhere to the electrode surface upon impact is highlighted and the sticking coefficient of a gold nanoparticle at a carbon surface determined. This technique to monitor nanoparticle sticking is optimised by chemical modification of the substrate electrode in order to achieve a “sticky” surface improving the rate of silver nanoparticle sticking. Finally, the nanoparticle collision method is shown to be applicable to C60 nanoparticles where their detection and sizing is achieved in non-aqueous conditions. The methods developed in this thesis make a significant contribution to the promising application of electrochemical techniques in the detection and characterisation of single nanoparticles.

In this thesis, mathematical modelling is used to theoretically investigate the electrochemical behaviour of surfaces which can be broadly classified as being ‘electrochemically heterogeneous’. Simulated voltammetry is used in the exploration of a number of specific systems as listed below. The cyclic voltammetry of electrodes composed of two different electroactive materials that differ in terms of their electrochemical rate constants towards any given redox couple. The effect of the distribution of the two materials was investigated and the occurrence of split peak cyclic voltammetry where two peaks are observed in the forward sweep, was studied. The technique was specifically applied to the modelling of highly-ordered pyrolytic graphite (HOPG). The steady-state voltammetry of a conducting spherical particle resting on an insulating supporting surface. An algebraic expression that completely describes the voltammetric waveform in the limit of irreversible kinetics was developed. The cyclic voltammetry of the EC′ (catalytic) mechanism at a regularly distributed array of hemispherical particles on an insulating supporting surface. Particular attention was paid to the ‘split-wave’ phenomenon, where two peaks are observed in the forward scan of a cyclic voltammogram and the conditions under which these peaks are resolvable were elucidated. The linear sweep voltammetry of micro- and nano-particle modified electrodes and other electrodes of partially covered and non-planar geometry. It was demonstrated that the apparent electrochemical rate constant of the reaction and thus the peak position of the voltammetry is dependent only on the relative electroactive surface area of the particles on the surface and not upon their shape or distribution. This has wide reaching implications as it can be used to explain some instances of a purported nano-catalytic effect without appeal to altered properties at the nanoscale. The linear sweep voltammetry of the interior of a partially electroactive cylindrical pore. Four limiting cases were observed and fully characterised. The linear sweep voltammetry of porous surfaces. It was established that if the pores are less than a certain threshold depth, then a porous surface will also display an apparent catalytic effect that is dependent on the relative electroactive surface area (including the area in the interior of the pores).

Carbon monoxide dehydrogenase (CODH) is known to catalyze CO oxidation and CO₂ reduction reversibly with the minimal overpotential. A great advantage of protein film electrochemistry (PFE) is its ability to probe catalysis over a wide range of potentials, especially in the low potential region required for CO₂ reduction. CODH I and CODH II from Carboxydothermus hydrogenoformans(Ch) and the composite enzyme acetyl-CoA synthase/carbon monoxide dehydrogenase (ACS/CODH) from Moorella thermoacetica(Mt) are intensively studied throughout this thesis. The different catalytic redox-states in CODH, Cox (inactive state), Cred1 (for CO oxidation) and Cred2 (for CO₂ reduction) as characterized by spectroscopy, are studied by PFE in the presence of substrate-mimic inhibitors. Cyanide, isoelectronic with CO, mainly inhibits CO oxidation, whereas cyanate, isoelectronic with CO₂, mainly targets CO₂ reduction. Sulfide inhibits CODH rapidly when the potential is more positive than −50 mV, which suggests that sulfide reacts to form a state at the oxidation level of Cox in CODH and is not an activator for CODH catalysis as suggested before. Thiocyanate only partially inhibits CO oxidation. No inhibition of CODH by azide is detected, which is in contrast with previous studies with ACS/CODHMt. The main differences between CODH ICh and CODH IICh are the stronger CO product inhibition and inhibition of CODH IICh by cyanide. These discoveries might shed light on the possible role of CODH II_{Ch,/sub> in biological systems. In comparison with bidirectional (reversible) electrocatalysis by CODH ICh and CODH IICh, only unidirectional electrocatalysis for CO oxidation by ACS/CODHMt is observed with an overpotential of 0.1 V and the electrocatalytic current is much smaller. In order to identify whether ACS influences the performance of CODH, several chemical reagents, such as sodium dodecyl sulfate (which separates CODH and ACS partially), 1, 10-phenanthroline, (which inhibits the active site in ACS) and acetyl-CoA (the product of the reaction carried out by ACS/CODHMt) are added. However, we have yet to observe any electrocatalytic current from CO₂ reduction. Inhibition of ACS/CODHMt by cyanide, cyanate and azide is consistent with previous studies by spectroscopy. Oxygen attack toward the active site in CODH is proved by cyanide protection. The inactive state, Cox can prevent oxygen attack and reductive reactivation restores CODH activity. In contrast, oxygen damages the active site irreversibly when CODH is in the Cred1 state. The new substrate, nitrous oxide (N₂O), isoelectronic with CO₂, is reduced by CODH and acts as the suicide substrate. Finally, hydrogen formation in the direction of CO oxidation and formate formation in the direction of CO₂ reduction by CODH are detected. The small solvent kinetic isotope effect is observed in CO oxidation. These findings suggest metal-hydride should play a role in CODH catalysis, which might provide a new direction to design better catalysts for CO₂ reduction.}

A spray deposition manufacturing route has been developed for the fabrication of carbon nano-structured and micro-structured energy storage devices in a thin film format, with controlled film thickness, homogeneous film surface morphology and high electrochemical performance for both supercapacitors and lithium ion battery anodes. Three types of low cost commercially available carbon materials (graphite, activated carbon and carbon black) have been investigated, and electrodes characterised in terms of surface morphology, surface chemistry, microstructure and electrochemical properties. By using ball milling, CO2 activation and adding suitable carbon conductive additives, nano-graphite-based film electrodes (one meter long and ~ 3 µm thickness) have been fabricated, with excellent ion transport and low electrical resistance (< 1.8 Ω). Specific capacitance of 110 F/g at a scan rate of 100 mV/s in 1 M H2SO4 was achieved. The high rate performance of activated carbon-based electrodes ( ~2 µm thickness) has been enhanced by reducing the contact resistance of electrode/current collector interface and building a well-interconnected and hierachical meso/macro-porous structure. A specific capacitance of over 120 F/g at a scan rate of 600 mV/s or 20 A/g current density in 1 M H2SO4 was achieved. The performance of carbon black-based electrodes (~4 µm thickness) in different electrolytes has been studied in both two- and three-electrode cells. High specific capacitances of 260 F/g at 1 A/g was achieved in 6 M KOH, together with energy and power densities of 21 kW/kg and 18 Wh/kg in 1 M Na2SO4. Finally, graphite-based electrodes for rechargeable lithium-ion batteries have also been fabricated with controlled film thickness from ~ 900 nm to ~ 40 µm and 98% capacity retention of 371 mA/g after 20 cycles. Spray deposition has been demonstrated to have the potential for scalability in the manufacture of carbon-based thin film electrodes with competitive properties.

This thesis is concerned with the study of electrode kinetics, which we shall examine via comparison between theory with experiment. As such the first two chapters outline the basic principles of electrochemical experiments and their simulation. First, we examine the properties of voltammetry at porous electrodes by means of both simulations and experiments. We then introduce the symmetric Marcus-Hush (SMH) model of electrode kinetics as an alternative to the empirical Butler-Volmer model. First, we examine different methods for modeling the voltammetry of kinetically inhomogeneous electroactive monolayers. Next, we perform a critical evaluation of the SMH model for solution-phase systems through extensive comparison to experiments under diffusion-only and convective mass transport conditions using both cyclic and square wave voltammetry. The model is compared with the Butler-Volmer model throughout and is ultimately found to be poorly suited to the parameterisation of electrode kinetics, despite its foundations in the microscopic Marcus theory. We then introduce the asymmetric Marcus-Hush model, which removes the assumption that the Gibbs energy curves for reactant and product have the same curvature. This modification results in an additional parameter which quantifies the asymmetry of the system. A similar evaluation of this model is then undertaken for both surface-bound and solution phase systems and the asymmetric model is found to be a great deal more successful than its symmetric predecessor. Finally we outline a novel technique for extracting kinetic information directly from experimental cyclic voltammetry. The method is simple to implement and is general to all electrode geometries with one-dimensional symmetry.

The work presented in this thesis examines the electrochemical behaviour of a number of species, both surface-bound and in the solution phase, in various room-temperature ionic liquids (RTILs), in order to identify systems which show altered reactivities in room-temperature ionic liquids, compared to that in conventional, molecular, aprotic solvents. The fundamentals of electrochemistry are outlined and an introduction to room-temperature ionic liquids is given, after which the results of six original investigations are presented, as follows; The electrochemical response of surface-bound anthraquinonyl films was investigated as a function of RTIL cation size. The reduction of oxygen to superoxide in the [C2mim][NTf2] RTIL was studied at different carbon electrodes. The mechanisms of electrodeposition and stripping of Zn(II) onto bulk zinc deposits, on glassy carbon electrodes, in the [C4mPyrr][NTf2] RTIL were investigated. A novel and successful method for the fabrication of zinc microdisk electrodes was developed and the electrochemical behaviour of these electrodes was investigated. The mechanisms for the electrochemical reductions of some bromo- and nitrobenzenes at platinum microelectrodes were determined in the [C4mPyrr][NTf2] ionic liquid, in order to identify systems which displayed changed reactivities in RTILs compared to those in molecular aprotic solvents. The altered reactivity of 1-bromo-4-nitrobenzene in the [C4mPyrr][NTf2] ionic liquid was utilised via electrochemical reduction at zinc microelectrodes in order to form arylzinc compounds. The work presented in this thesis shows that the oft-taken view that the reactivity of a given species in an ionic liquid will mimic that in conventional aprotic solvents is not always the case. Reactivities can be markedly different between RTILs and aprotic solvents and can also be highly dependent on the specific ionic liquid employed.

Electron paramagnetic resonance (EPR) spectroscopy is a powerful tool for obtaining structural information about chemical centres with unpaired electrons. In complex biological systems, EPR spectroscopy can be used to probe these paramagnetic centres and the long-range interactions between them. This thesis investigates two important types of enzymes, and in particular the role of the iron-sulfur electron-transfer centres they contain, with a variety of EPR techniques. Complex I (NADH:Ubiquinone Oxidoreductase) plays a key role in the electron transfer chain essential to the formation of ATP, and its malfunction has been related to numerous human diseases. It is a giant enzyme that contains the longest relay of iron-sulfur clusters known. EPR experiments conducted on complex I from bovine mitochondria yield crucial insight into the mechanism of efficient long-range electron transfer and bring us a step closer to understanding the functioning of this important complex. Hydrogenases are produced by micro-organisms and catalyse the reversible oxidation of H2. Most hydrogenases, including Hyd-2 from Escherichia coli, are very air-sensitive, but some, including E. coli Hyd-1 and Salmonella Hyd-5, are able to function in the presence of atmospheric levels of O2. Understanding the origins of this 'O2-tolerance' is of paramount importance if hydrogenases are to be exploited in future energy technologies. In this thesis, native E. coli Hyd-1 and Hyd-2, Salmonella Hyd-5, as well as O2-tolerant and O2-sensitive variants of E. coli Hyd-1 are characterised using EPR. The EPR investigations elucidate properties of the active site and the electron-transfer relay and, in conjunction with other techniques, reveal structural and mechanistic details of how a highly unusual iron-sulfur cluster in the electron-transfer chain enables some hydrogenases to sustain catalytic activity in the presence of O2.

The use of H2 as an energy carrier has in recent years been identified as a promising future solution to the current energy crisis. Hydrogenases are metalloenzymes found in many microorganisms and are used to catalyse the reversible inter-conversion of protons and H2. These enzymes and their synthetic analogues have been recognised as a way to facilitate the use of H2 as a fuel. A major challenge to the future use of these catalysts is their reactions with small molecule inhibitors, such as oxygen and carbon monoxide. Detailed understanding of the structure and catalytic mechanism of these highly efficient catalysts is vital for the design of bio-inspired functional analogues for use in technological applications. In this thesis electrochemical studies of three [FeFe]-hydrogenases are presented, performed using the technique of protein film electrochemistry. The strong potential dependence of the reaction of these hydrogenases with carbon monoxide and formaldehyde is characterised and rationalised. These studies provide compelling evidence for the formation of a previously ambiguous super-reduced state of [FeFe]-hydrogenase at low potential. This state is shown to be active and stable, and it is proposed that this state is involved in catalytic H2 production. This super-reduced state is shown to have a high affinity for the reversible binding of formaldehyde, but a very low affinity for both carbon monoxide and oxygen. Activation of carbon monoxide inhibited [FeFe]-hydrogenase can be very rapidly induced by the application of a sufficiently reducing potential. These enzymes, considered to be oxygen sensitive, are shown to be extremely tolerant to irreversible oxygen damage at very reducing potentials where the super-reduced state is formed.

Understanding the factors influencing the ET characteristics of redox proteins confined at an electrochemical interface is of fundamental importance from both pure (fundamental science) and applied (biosensory) perspectives. This thesis reports on progress made in the emerging field of coupled electrochemical characterization and optical imaging in moving the analysis of redox-active films to molecular scales. More specifically the combination of cyclic voltammetry and wide-field Total Internal Reflection (TIRF) microscopy, here named ‘Fluorescent Cyclic Voltammetry’ (FCV), was applied to monitoring the response of surface-confined redox active proteins at submonolayer concentrations. The combined submicrometre spatial resolution and photon capture efficiency of an inverted TIRF configuration enabled the redox reactions of localized populations of proteins to be directly imaged at scales down to a few hundreds of molecules. This represents a 6-9 orders of magnitude enhancement in sensitivity with respect to classical current signals observed in bioelectrochemical analysis. Importantly, measurements of redox potentials at this scale could be achieved from both natural and artificially designed bioelectrochemical fluorescent switches and shed fundamental light on the thermodynamic and kinetic dispersion within a population of surface confined metalloproteins. The first three chapters of this thesis provide an overview of the relevant literature and a theoretical background to both the rapidly expanding fields of electroactive monolayers bioelectrochemistry and TIRF imaging. The initial design and construction of a robust electrochemically and optically addressable fluorescent switch, crucial to the applicability of FCV is reported in chapter 5. The generation of optically transparent, and chemically modifiable electrode surfaces suitable for FCV are also described. Chapter 6 describes the response of the surface confined azurin-based switch. Analysis of the spatially-resolved redox reaction of zeptomole samples in various conditions enables the mapping of thermodynamic dispersion across the sampled areas. In chapter 7 the newly developed FCV detection method was extended to investigate more complex bioelectrochemical systems containing multiple electron transferring redox centres and responding optically at different wavelengths. This approach provides a platform for spectral resolution of different electrochemical processes on the same sample. Finally in chapter 8 an electrochemical procedure is proposed for investigating the kinetic response of redox proteins using a fundamentally new methodology based on interfacial capacitance. In using variations in the surface chemistry to tune the rate of electron transfer, the approach was shown to be a robust and facile means of characterising redox active films in considerably more detail than possible through standard electrochemical methodologies. Ultimately, it can be applied to probe dispersion within protein populations and represents a powerful means of analysing molecular films more generally.

Nicotinamide cofactors in their reduced and oxidised forms are important redox agents in biology. Of about 3000 dehydrogenases available to date, many require these cofactors for their activity. Dehydrogenases are of interest to chemists as they offer asymmetric catalysis to yield chiral products. The requirement of dehydrogenases for nicotinamide cofactors necessitates research into finding the best way of recycling the oxidised or reduced forms of these cofactors. Electrocatalytic NAD(P)H oxidation and NAD(P)⁺ reduction on standard electrodes is problematic due to unwanted side reactions and high overpotential requirements, but in Nature efficient enzyme catalysts are available to facilitate these reactions. The focus of this Thesis, the Soluble Hydrogenase of R. eutropha (SH) is a multimeric bidirectional hydrogenase that couples H2 oxidation to the reduction of NAD⁺ to NADH. Protein Film Electrochemistry (PFE) has been employed to study NAD⁺-reducing catalytic moieties of the SH for the first time. It is shown that SH subunits on an electrode are able to catalyse NADH oxidation and NAD⁺ reduction efficiently with minimal overpotential, which is significant because in vivo, NAD(H) cycling is coupled to 2H⁺/H₂ cycling and these reactions are closely spaced in potential. Substrate affinities and inhibition constants for the SH, determined using PFE are discussed in the context of the SH function and the related catalytic domains of respiratory Complex I. A range of molecules that are known to inhibit the related Complex I have been investigated for their ability to inhibit the SH moieties: the similarity between inhibition constants is consistent with structural and functional similarity between the SH and Complex I. The ability of the SH moieties to sustain NAD(H) catalysis in the presence of O₂ is also demonstrated and is consistent with the requirement for the SH to function under aerobic conditions and to reactivate the inactivated hydrogenase moiety by supplying low potential electrons from NADH. Engineered variants of the SH, designed to enhance the affinity towards NADP⁺, were investigated for the first time, using PFE. Electrochemical characterisation of the variants is presented and results are discussed alongside findings on the wild type SH. The variants are shown to exhibit NADP⁺ reduction, and to have higher affinity towards NADP⁺ than the wild type SH. The first efficient NADP⁺ reduction and NADPH oxidation is observed for one of the variants on a graphite electrode and the best variant showed a KM of 1.7 mM for NADP⁺. This Thesis also provides evidence for the ability of moieties of the SH to be used in cofactor regeneration systems. Two novel systems are demonstrated. The first involves H₂ driven NADH recycling based on the NAD⁺-reducing moiety of the SH immobilised on graphite particles together with a hydrogenase or platinum, with electrons from H₂ passed from the hydrogenase through the graphite to the NAD⁺-reducing moiety. The second involves an electrode modified with the NAD⁺-reducing moiety of the SH, and is demonstrated as an electrochemical NADH recycling system coupled with NADH-dependent pyruvate reduction to lactate by lactate dehydrogenase. The ability of variants of the SH to catalyse NADP⁺ reduction suggests that it may also be possible to use these systems for recycling NADPH for catalysis of important biotransformation reactions by NADPH-dependent dehydrogenases.

Carbon-based materials are poised to be an important class of 21st century materials, for bio-medical, bio-electronic, and bio-sensing applications. Diamond and polymers are two examples of carbon-based materials of high interest to the bio-materials community. Diamond, in its conductive form, can be used as an electrochemical bio-sensor, whilst its nanoparticle form is considered a non-inflammatory platform to deliver drugs or to grow neuronal cells. Polymers, especially when chemically modified, have been used extensively in biological environments, from anti-microbial use to drug delivery. The large-scale use of either material for biological use is limited by two factors: ease of chemical modification and the paucity of knowledge of their surface chemistry in aqueous media. This thesis addresses aspects of both these issues. The first study reported is an in situ study of the adsorption dynamics of an exemplar globular protein (bovine serum albumin, BSA) on nanodiamond using the relatively novel quartz crystal microbalance with dissipation (QCM-D) technique. For the first time, QCM-D enabled the detailed study of protein dynamics (i.e. kinetics, viscoelastic properties, overlayer structure, etc.) onto nanodiamond thin films having various surface chemistry and roughness. The dynamics of protein adsorption is found to be sensitive to surface chemistry at all stages of adsorption, but it is only sensitive to surface roughness during initial adsorption phases. Our understanding of the nanodiamond-biology interface is enhanced by this study, and it suggests that QCM-D is useful for the study of the surface chemistry of nanoparticle forms of inorganic materials. A second study concerns a novel surface functionalization scheme, based on carbene and azo-coupling chemistry, which has been recently introduced as a practical, facile method for modifying the surfaces of polymers. Using modern surface characterization techniques, it is demonstrated that a chemical linker can be attached to polystyrene surfaces using carbene-based chemistry, and that further chemical functionality can be added to this chemical linker via an azo-coupling reaction. In situ studies of protein dynamics at these interfaces were conducted using QCM-D, thus enabling a link between specific protein behaviour and the polymer surface chemical termination chemistry to be made. A third area of study of investigates the use of diamond electrodes as a bio-sensor for dopamine under physiological conditions. For these conditions, ascorbic acid interferes with the dopamine oxidation signal, in ways that render the two signals irresolvable. Various modifications are used in attempts to reduce this interference, including: small and large cathodic treatments, grafting of electro-active polymers, addition of carbon nanotubes, and hydrogen plasma treatment. Those modifications leading to the hydrogen-termination of diamond are shown to work the best. Notably, hydrogen plasma treatment effects the complete electrochemical separation of dopamine and ascorbic acid at a diamond electrode. This is the first time this has been accomplished without adding non-diamond materials to the diamond electrode surface.

One of today’s most pressing scientific challenges is the conception, development and deployment of renewable energy technologies that will meet the demands of a rapidly increasing population. The motivation is not only dwindling fossil fuel reserves, but also the necessary curtailment of emissions of the greenhouse gas carbon dioxide (a product of burning fossil fuels). The sun provides a vast amount of energy (120,000 TW globally), and one major challenge is the conversion of a fraction of this energy into chemical energy, thereby allowing it to be stored. Dihydrogen (H₂) that is produced from water is an attractive candidate to store solar energy (a ‘solar fuel’), as are high energy carbon-containing molecules (such as CO) that are formed directly from carbon dioxide. One key aspect is the development of catalysts that are able to offer high rates and efficiencies. In biology, some microbes acquire energy from the metabolism of H₂ and CO. The biological catalysts - enzymes - that are responsible are hydrogenases (for the oxidation of H₂ to protons); and carbon monoxide dehydrogenases (CODHs, for the oxidation of CO to CO₂). These redox enzymes, containing nickel and iron as the only metals, are extraordinary in terms of their catalytic characteristics: many are fully reversible catalysts and offer very high turnover frequencies (thousands per second are common), with only tiny energy input requirements. This Thesis uses a hydrogenase from the bacterium Escherichia coli, and two CODHs from the bacterium Carboxydothermus hydrogenoformans, as the catalysts in H2 production and CO₂ reduction systems. Chapter 3 describes the concept and development not of a solar fuel system, but of a device that catalyses the water-gas shift reaction (the reaction between CO and water to form H₂ and CO₂) - a process of major industrial importance for the production of high purity H₂. Chapters 4, 5 and 6 detail photochemical CO₂ reduction systems that are driven by visible light. These systems, operating under mild, aqueous conditions, involve CODHs attached either to TiO₂ nanoparticles that are sensitised to visible light by the co-attachment of a ruthenium-based dye complex, or to cadmium sulfide nanomaterials that, having a narrow band gap, are inherently photoexcitable by visible light. The motivation here is not the construction of technological devices; indeed, the enzymes that are used are fragile, highly sensitive to oxygen, and impossible to scale to industrial levels. Rather, the drivers are those of scientific curiosity (can the incorporation of these remarkable biological catalysts enable the creation of outstanding solar fuel devices?), and of producing systems that serve as benchmarks and inspiration for the development of fully synthetic systems that are robust and scalable.

Hydrogenases are a class of metalloenzymes which catalyse H₂ oxidation and its reverse reaction, H⁺ reduction. There is interest in investigating how H₂ as an energy carrier is cycled in biology. Hydrogenases have also been studied extensively because there are potential applications for them as catalysts for H₂ oxidation in fuel cells or H₂ production via light-driven water splitting. For these applications, the ability for the hydrogenase to work in the presence of O₂ is an important issue. The microorganism Ralstonia eutropha is a well-studied model aerobic H₂ oxidiser: it can adopt H₂ as the sole energy source to grow cells in the presence of O₂. It produces at least three distinct O₂-tolerant NiFe hydrogenases: the membrane-bound hydrogenase (MBH), the NAD⁺-reducing soluble hydrogenase (SH) and the regulatory hydrogenase (RH). This Thesis employs protein film electrochemistry (PFE) to study the SH and RH. It is found that the SH is able to work in both direction (H₂ oxidation and H⁺ reduction) with minimum overpotential, which is critical in coupling 2H⁺/H₂ cycling with the closely spaced NAD⁺/NADH potential. Reactions of the SH with O₂ have been investigated, revealing at least two distinct O₂-inactivated states, but consistent with the requirement for the SH to function in air, it can be reactivated in the presence of O₂ at low potentials which could be provided by the NAD⁺/NADH pool in vivo. The affinity of the RH for H₂ was determined by PFE and found to be slightly higher than that of the SH and MBH. This may provide a way for the microbe to regulate hydrogenase expression in response to the H₂ availability. Carbon monoxide and O₂-inactivated states of the RH have been identified for the first time, confirming that a constricted gas channel is not sufficient to explain its O₂ tolerance. Observation of potential dependent reactions in hydrogenases means that it is important to have spectroscopic methods for characterising states triggered by inhibitors and potential. An Infrared spectroelectrochemical approach suitable for studying metalloenzymes has been developed and preliminary spectra on RH recorded. This method should provide many opportunities for future studies of redox states of hydrogenases.

This thesis addresses simulation of electrochemical experiments, with an emphasis on processes of diffusional mass transport to electrode surface. Following system has been studied: • Applying theoretical modeling and experimentation is shown that even significant surface roughness produced by deliberate polishing or scratching is not sufficient to be distinguished in cyclic voltammetry experiments conducted under the usual conditions. In stripping voltammetry experiment the shape of the voltammograms strongly depends on the model of the electron transfer but is not always sensitive to the precise model of the electrode surface; the conditions under which this is the case are identified, and generic roughness effects on stripping voltammetry are quantified. Electrode roughness can have a significant effect on the stripping of the metals from the solid electrode especially in respect of the voltammetric waveshape. • We first consider two different models of electrodes covered with electroinactive layers: the electrode is covered with a uniform layer and the layer contains pinholes. Both models are simulated and then compared to identify conditions under which they can be distinguished. Next we propose generic model to predict the influence of electroactive layer on the cyclic voltammetric. The conditions under which deviation from the behavior of a planar electrode are predicted. • We first consider one electron, one proton and next two electron, two proton reduction of surface bound species. Two mechanisms of reaction are considered: stepwise and concerted. Voltammetry studied under the three regimes of protons mass transport: infinitely fast (fully buffered solution), infinitely slow (infinitely high surface coverage of electrode) and intermediate case of finite rate of diffusional mass transport to electrode surface. Types of voltammograms observed in each case are presented and discussed. • Theory of chronoamperometry on disc and ring-recessed microelectrodes and their arrays is reported. Three and four different regimes of transient current versus time can be observed at microelectrode arrays of disc and ring electrodes, accordingly. A generic, accurate and easy to use method of experimental chronoamperometric data analysis is proposed. It is shown that the method can be applied to the simultaneous measurement of D and nC in solution. • The fabrication, characterization, and use of arrays of ring-recessed disk generator-colector microelectrodes are reported. Experiments and simulations relating to time- of-flight experiments in which material electrogenerated at a disk is diffusionally transported to the ring are reported. We further study voltammetry of electrochemically active species which undergoes first and second order chemical reactions. Current transients are found to be sensitive to the diffusion coefficient of both the reduced and oxidised species as well as to the rate of the chemical reaction and its mechanism.

Electrochemistry is the science of electron transfer. The subject is of great importance and appeal because detailed information can be obtained using relatively simple experimental techniques. In general, the raw data is sufficiently complicated to preclude direct interpretation, yet is readily rationalised using numerical procedures. Computational analysis is therefore central to electrochemistry and is the main topic of this thesis. Chapters 1 and 2 provide an introductory account to electrochemistry and numerical analysis respectively. Chapter 1 explains the origin of the potential difference and describes its relevance to the thermodynamic and kinetic properties of a redox process. Voltammetry is introduced as an experimental means of studying electrode dynamics. Chapter 2 explains the numerical methods used in later chapters. Chapter 3 presents a review of the use of nanoparticles in electrochemistry. Chapter 4 presents the simulation of a random array of spherical nanoparticles. Conclusions obtained theoretically are experimentally confirmed using the Cr3+/Cr2+ redox couple on a random array of silver nanoparticles. Chapter 5 presents an investigation into the concentration of supporting electrolyte required to make a voltammetric experiment quantitatively diffusional. This study looks at a wide range of experimental conditions. Chapter 6 presents an investigation into the deliberate addition of insufficient supporting electrolyte to an electrochemical experiment. It is shown that this technique can be used to fully study a stepwise two electron transfer. Conclusions obtained theoretically are experimentally confirmed using the reduction of anthracene in acetonitrile. Chapter 7 presents a new method for simulating voltammetry at disc shaped electrodes in the presence of insufficient supporting electrolyte. It is shown that, under certain conditions, the results obtained from this complicated simulation can be quantitatively obtained by means of a much simpler ‘hemispherical approximation’. Conclusions obtained theoretically are experimentally confirmed using the hexammineruthenium ([Ru(NH3)6]3+/[Ru(NH3)6]2+) and hexachloroiridate ([IrCl6]2−/[IrCl6]3−) redox couples. Chapter 8 presents an investigation into the voltammetry of stepwise two electron processes using ionic liquids as solvents. It is shown that these solvents can be used to fully study a stepwise two electron transfer. Conclusions obtained theoretically are experimentally confirmed using the oxidation of N,N-dimethyl-p-phenylenediamine in the ionic liquid 1-butyl-3-methylimidazolium tetrafluoroborate ([C4 mim][BF4]). The work presented in this thesis has been published as 7 scientific papers.

This thesis concerns several advances in electroanalytical chemistry which are separated into four parts: the electrochemical investigation of diffusional behaviour, the mechanistic and kinetic study of electrochemistry with room temperature ionic liquids (RTILs), the study of weakly-supported electrochemistry and a comparison of the Butler-Volmer and Marcus-Hush kinetic theories of electron transfer. A study of the diffusional behaviour of electroactive species is essential for further studies, especially in the case when electrochemistry is complicated through ion-pairing interactions between the electroactive species and other electrolytes. In Part II of this thesis, the possibility of the ferricenium ion-paired with perchlorate and hexauorophosphate in acetonitrile was discussed firstly employing chronoamperometric technique. Afterwards, the hexaammineruthenium III/II couple supported by chloride, nitrate and sulfate respectively was studied by a similar method. In order to avoid unwanted ion-pairing effects, room temperature ionic liquids can be applied as solvent, which provide high conductivity by their own ionic nature so that experiments can be conducted without adding additional supporting ions. Because of RTILs have distinctive properties, for example, high viscosity, high conductivity and ionic nature, electrochemistry could be greatly changed compared to those in conventional solvents. Part III of this thesis gives a detailed description of this topic. First, a study of the reduction of 1,4-benzonquinone in 1-ethyl-3-methylimidazolium bis(triuoromethanesulfonyl)imide is presented to show the new mechanistic insight into comproportionation in a electrochemical process. Second, a discussion of the oxidation of hydroquinone in the same RTIL is introduced to suggest a possible ECE scheme which was never reported before. The interest of weakly supported electrochemistry is also well-established, which not only provides another alternative strategy to avert ion-pairings but also offers more physical insights into electrochemical processes. Quantitative methods analysing voltammetries without an excess amount of supporting electrolyte are developed by introducing a migration term into the mass transport equation. In Part IV, new mechanistic insights into the reduction of 2-nitrobromobenzene and the dimerisation of 2,6-diphenylpyrylium in acetonitrile were provide by using weakly-supported cyclic voltammetry. Also, pulse techniques was also adopted to investigate the reduction of cobaltocenium and cobalt(III) sepulchrate, giving an alternative way for electrochemical analysis. A major application of electroanalytical chemistry is investigating electrochemical kinetics. Two kinetic models mostly concerned by electrochemists are Butler-Volmer and Marcus-Hush formalisms. The classic phenomenological model, Butler-Volmer formalism successfully describes most common electron transfer kinetics but shows little reference with nature of the involved species, solution and electrode material, while a more physically insightful theory, the Marcus-Hush formalism, takes species natural properties, for instance, a change of distances or geometry in the solvation or coordination shells of the redox, into account although it requires more complex formulations. Comparative studies of these two theories are presented in Part V in order to improve our understanding of the electron transfer kinetics under different circumstances. First, comparison of cyclic voltammograms of the reduction of europium(III) and 2-methyl-2-nitropropane at mercury microhemispherical electrodes was carried out. Second, square wave and differential pulse voltammetric techniques were also employed to further discriminate the two kinetic models. These studies all find that the symetric Marcus-Hush theory assuming the reactants and products have identical force constant dose not satisfactorily agree with the experimental results. Hence, the introduction of asymmetric Marcus-Hush theory was presented considering different oxidative and reductive reorganization energies, which gives reasonable agreement with experiments and makes this theory more insightful.

This thesis primarily focuses on the electrochemical properties of the H2/H+ redox couple, at various metallic electrodes in room temperature ionic liquids. Initially, a comprehensive overview of room temperature ionic liquids, RTILs, compared to conventional organic solvents is presented which identifies their favourable properties and applications, followed by a second chapter describing the basic theory of electrochemistry. A third chapter presents the general experimental reagents, instruments and measurements used in this thesis. The results presented in this thesis are summarized in six further chapters and shown as follows. (1) Hydrogenolysis, hydrogen loaded palladium electrodes by electrolysis of H[NTf2] in a RTIL [C2mim][NTf2]. (2) Palladium nanoparticle-modified carbon nanotubes for electrochemical hydrogenolysis in RTILs. (3) Electrochemistry of hydrogen in the RTIL [C2mim][NTf2]: dissolved hydrogen lubricates diffusional transport. (4) The hydrogen evolution reaction in a room temperature ionic liquid: mechanism and electrocatalyst trends. (5) The formal potentials and electrode kinetics of the proton_hydrogen couple in various room temperature ionic liquids. (6) The electroreduction of benzoic acid: voltammetric observation of adsorbed hydrogen at a Platinum microelectrode in room temperature ionic liquids. The first two studies show electrochemically formed adsorbed H atoms at a metallic Pt or Pd surface can be used for clean, efficient, safe electrochemical hydrogenolysis of organic compounds in RTIL media. The next study shows the physicochemical changes of RTIL properties, arising from dissolved hydrogen gas. The last three studies looked at the electrochemical properties of H2/H+ redox couple at various metallic electrodes over a range of RTILs vs a stable Ag/Ag+ reference couple, using H[NTf2] and benzoic acid as proton sources. The kinetic and thermodynamic mechanisms of some reactions or processes are the same in RTILs as in conventional organic or aqueous solvents, but other remarkably different behaviours are presented. Most importantly significant constants are seen for platinum, gold and molybdenum electrodes in term of the mechanism of proton reduction to form hydrogen.

Herein, the influence of protonation and adsorption upon the redox and electrocatalysis of quinone species - specifically anthraquinone derivatives – is investigated. Through the comparison of the measured rate constants of one-electron reductions of a family of quinones in acetonitrile at both graphite and gold electrodes, it was confirmed that the redox potential indirectly influences the rate of electron transfer in a manner consistent with the potential-dependence of the density of states. In aqueous media, the voltammetric response of both anthraquione-2-sulfonate (AQMS) and anthraquinone-2,6-disulfonate (AQDS) was measured over the full aqueous pH range. A model is provided which is able to describe not just the variation in the formal potential but also the peak height as a function of pH. Importantly, this model predicts that the formal potential for the first (Ef1) and second (Ef2) electron transfers are comparable in magnitude (E^θ _f2−E^_θf1 equals -15mV for AQMS and -36mV for AQDS). This quantitative model is then further extended to consider the situation in which the system is not fully buffered, giving insight into the change of pH at the electrode surface during experimentation. Adsorption to graphitic electrodes can impart a strong influence on the measured voltammetric response. It is demonstrated that through the pre-exposure of a newly prepared graphitic electrode to organic solvents, these adsorption processes can be predominantly blocked. Moreover, it is shown that the electroactivity of the electrode is not significantly altered. This thesis also highlights two cases in which adsorption of the electroactive species may be used to positive effect. First, the surface adsorption of anthraquinone-2-monosulfonate is studied on a graphite electrode, where it is demonstrated that the heterogeneity of the electrode surface may be probed through studying the electrochemical response of the adsorbed species. From this work it is concluded that the rate of electron transfer at the graphitic basal plane is 2-3 orders of magnitude lower than that observed on the edge plane sites. Second, the co-adsorption of DNA and anthraquinone-2-monosulfonate is used as an indirect method to measure the solution phase concentration of DNA (LOD = 8.8μM). The reduced form of anthraquinone is also known to readily reduce oxygen. Through the use of a boron-doped diamond electrode it was possible to directly study the anthraquinone mediated reduction mechanism. Significantly, the voltammetric response indicates the reduction of the oxygen via the semi-quinone intermediate (kf = 4.8 × 10⁹ mol⁻¹ dm³ s⁻¹) is over two orders of magnitude faster than the reaction involving the di-reduced form (kf = 1 × 10⁷ mol⁻¹ dm³ s⁻¹). More importantly, this work provides voltammetric evidence for the existence of the semi-quinone species. This work is subsequently extended through the investigation of the poorly soluble anthraquinone derivative quinizarin. Not only is it possible to detect voltammetrically this biologically relevant species to concentrations as low as 5nM (100ppt), but the methodology also allows the electrochemistry of the quinizarin species to be probed, something which was not previously possible.

Chapter1 gives an overview of the basic principles of electrochemistry. A rigorous electrochemical study on the solution phase and solid phase cobalt phthalocyanine (CoPC) is presented in chapter2. The formof CoPC on carbon electrodes was characterized by scanning electron microscope (SEM). The use of CoPC modified edge plane pyrolytic graphite (CoPC-EPPG) for sensing nitrite (NO₂⁻) was also investigated. It was found that the claimed mediator CoPC has no influence on the process. A bare glassy carbon (GC) electrode was successfully applied for the quantitative determination of nitrite as a simple alternative to the modified electrodes reported in the literature (chapter3). Chapter4 compares the voltammetric responses of an edge plane pyrolytic graphite electrode covalently modifed with 2-anthraquinonyl groups (EPPG-AQ2) and solution phase anthraquinone monosulphonate (AQMS) in the presence of a limited concentration of protons. The solution phase and surface bound species show analogous responses resulting in split waves. Digisim™ simulation of the AQMS voltammetry have shown that the pH adjacent to the electrode may be altered by up to 5-6 pH units in low buffered solutions; this is caused by the consumption of protons during the electrochemical reaction. Chapters5 and 6 compare the electrochemical properties of 2-anthraquinonyl groups covalently attached to an edge plane pyrolytic graphite (EPPG) and to a gold electrode. In both cases simulations using newly developedMarcus-Hush-Chidsey theory for a 2e⁻ process assuming a uniform surface did not achieve a good agreement between theory and experiment. Subsequently two models of surface inhomogeneity were investigated: a distribution of formal potentials, EӨ, and a distribution of electron tunneling distances, r₀. For both EPPG-AQ2 and Au-AQ2 modified electrodes the simulation involving EӨ distribution turned out to be the most adequate. This is the first time that Marcus-Hush-Chidsey theory has been applied to a 2e⁻ system. Chapter7 briefly summarizes the obtained results.

The experimental work discussed in this thesis explores the effects of metal modification on the electroanalytical ability of boron doped diamond electrodes. Boron doped diamond (BDD) electrodes have found increased application to electroanalysis in the past two decades, yet relatively little of the literature is focused on metal, nano and microparticle modification of the substrate. In this thesis three metals have been used to modify the BDD electrode; bismuth, antimony and nickel. Bismuth and antimony nanoparticle modified BDD electrodes were directly compared to unmodified BDD and a bulk bismuth electrodes in the determination of trace levels of cadmium and lead using anodic stripping voltammetry. In both instances, the modified electrode allowed for the simultaneous determination of each analyte that was otherwise unattainable at the unmodified BDD electrode. The nickel modified BDD (Ni-BDD) electrode was used in the determination of organic analytes, namely glucose, methanol, ethanol and glycerol. The nickel nano and microparticle electrodes gave the characteristic Ni(OH)₂/NiOOH redox couple in alkali pH, the oxidised form of which (NiOOH) catalysed the oxidation of the organic analytes. The chapter on glucose sensing with the Ni-BDD electrode is preceded by an extensive literature review on the advances of non-enzymatic glucose sensing, and the application of catalytic metals and nanomaterials in this field. Throughout the course of this DPhil, there has been a collaborative project between Asylum Research and myself within the Compton group to develop a commercial electrochemical atomic force microscope (EC-AFM) cell. The aim was to produce an adaptable EC-AFM cell capable of dynamic electrochemical experiments whilst simultaneously or instantaneously acquiring an AFM image of the modified surface, in-situ. This project was successful, and the EC-AFM cell has contributed to a number of chapters in this thesis, and has now been commercialised.

This thesis presents algorithm development in computational chemistry, and applies new computer science concepts to voltammetric simulation. To begin, this thesis discusses why algorithm development is necessary, and inherent problems found in commercial simulation solvers. As a result of this discussion, this thesis describes the need for simulators to keep abreast of recent computational developments. Algorithm development in this thesis is taken through stages. Chapter 3 applies known theory relating to the stripping voltammetry at a macroelectrode to the diffusional model of a microdisk, using finite difference and alternating direction implicit simulation techniques. Chapter 4 introduces the concept of parallel computing, and how computational hardware has developed recently to take advantage of out-of-order calculations, by processing them in parallel to reduce simulation time. The novel area of graphics card simulation for highly parallel algorithms is also explained in detail. Chapter 5 discusses the adaptation of voltammetric finite difference algorithms to a purely parallel format for simulation by explicit solution. Through explicit solution, finite difference algorithms are applied to electrode geometries which necessitate a three-dimensional solution – elliptical electrodes; square, rectangular, and microband electrodes; and dual microdisk electrodes in collector-generator mode. Chapter 6 introduces 'Random Walk' simulations, whereby individual particles in the simulation are modelled and their trajectories over time are calculated. The random walk technique in this thesis is improved for pure three-dimensional diffusion, and adapted to graphics cards, allowing up to a factor 4000 increase in speed over previous computational methods. This method is adapted to various systems of low concentration confined voltammetry (chapter 6.4) and single molecule detection, ultra low concentration cyclic voltammetry (chapter 6.5), and underpotential deposition of thallium on mobile silver nanoparticles (chapter 6.6). Overall, this thesis presents, and applies, a series of algorithm development concepts in computational electrochemistry.

Electrolytic solutions contain mobile ions that can pass current, and are essential components of any solution-phase electrochemical system. The Nernst–Planck–Poisson equations describe the electrodynamics and transport dynamics of electrolytic solutions. This thesis applies modern numerical and mathematical techniques in order to solve these equations, and hence determine the behaviour of electrochemical systems involving charge transport. The following systems are studied: a liquid junction where a concentration gradient causes charge transport; an ideally polarisable electrode where an applied potential difference causes charge transport; and an electrochemical cell where electrolysis causes charge transport. The nanometre Debye length and nanosecond Debye time scales are shown to control charge separation in electrolytic solutions. At equilibrium, charge separation is confined to within a Debye length scale of a charged electrode surface. Non-equilibrium charge separation is compensated in solution on a Debye time scale following a perturbation, whereafter electroneutrality dictates charge transport. The mechanism for the recovery of electroneutrality involves both migration and diffusion, and is non-linear for larger electrical potentials. Charge separation is an extremely important consideration on length scales comparable to the Debye length. The predicted features of capacitive charging and electrolysis at nanoelectrodes are shown to differ qualitatively from the behaviour of larger electrodes. Nanoscale charge separation can influence the behaviour of a larger system if it limits the overall rate of mass transport or electron transfer. This thesis advocates the use of numerical methods to solve the Nernst–Planck–Poisson equations, in order to avoid the simplifying approximations required by traditional analytical methods. As this thesis demonstrates, this methodology can reveal the behaviour of increasingly elaborate electrochemical systems, while illustrating the self-consistency and generality of fundamental theories concerning charge transport.

The work presented in this thesis involves the use of room temperature ionic liquids (RTILs) as solvents in electrochemical experiments for the detection of hydrogen sulfide. The fundamentals of electrochemistry are presented, followed by an overview of RTILs in terms of their properties, applications and their behaviour as electrochemical solvents compared to conventional solvents. This is followed by an outline of electrochemical detection of various gases in aqueous, organic and ionic solvents. The results of 8 original studies are then presented as follows:

• The study of the electrochemical window of twelve different room temperature ionic liquids using cyclic voltammetry vs. an internal redox couple for two defined current densities, and observation of water uptake of different ionic liquids under different conditions using a Karl Fischer titrator.
• The reduction of hydrogen sulfide in various room temperature ionic liquids at a platinum electrode, measured using cyclic voltammetry. Also, solubilities and diffusion coefficients of hydrogen sulfide determined by potential step chronoamperometry.
• The oxidation of hydrogen sulfide in various room temperature ionic liquids at a platinum electrode measured using cyclic voltammetry and the simulation of the electrochemical signal using experimentally defined parameters.
• The disproportionation of N,N-dimethyl-p-phenylenediamine (DMPD) in room temperature ionic liquids using cyclic voltammetry, and computational simulation of the voltammetry of DMPD using experimentally defined parameters to elucidate kinetic and thermodynamic data. DMPD was examined as a mediating species for hydrogen sulfide detection.
• The oxidation of catechol and dopamine in ionic liquids using cyclic voltammetry and observing adsorption effects when varying solvent anion. Catechol was examined as a mediating species for hydrogen sulfide detection.
• The electrochemical oxidation of NADH in ionic liquids using cyclic voltammetry and observing the ”switching on or off” of the electrochemical signal when varying the solvent anion. NADH was examined as a mediating species for hydrogen sulfide detection.
• The mediated detection of hydrogen sulfide utilizing various mediating species in several ionic liquids using cyclic voltammetry, and the elucidation of the mediating mechanism of hydrogen sulfide in 3,5-tert-butyl-o-benzoquinone.
• The observation of the diffusion of ferrocene in an ionic liquid at ring-recessed disc microelectrode arrays in generator-collector mode using potential step chronoamperometry.
The results presented show that room temperature ionic liquids perform well as solvents in gas sensors, and could be viable alternatives to traditional organic solvents. Ionic liquids have also been observed to be tuneable in their reactions with analytes depending on the constituent cations and, in particular, anions. This tuneability is advantageous as specific combinations of cations and anions can be chosen to suit particular experiments.

This thesis is concerned with dynamic electrochemical experiments with different concentrations of supporting electrolyte. Normally supporting electrolyte is added to a solution in order to avoid undesirable effects as migration and potential drop in solution. However, in the present thesis we focus on the study and understanding of such effects as the concentration of supporting electrolyte decreases. First a theoretical treatment is proposed, based on numerical simulation using the Nernst-Planck- Poisson system of equations. The theoretical treatment is compared with previous works as electroneutrality, the differences between both models are explained. The model is also compared with theoretical results to validate the theoretical treatment. Experimental results of chronoamperometry and cyclic voltammograms are compared with theoretical results obtaining remarkable agreement. Is noteworthy that to the best of the author’s knowledge this is the first time that experimental dynamic voltammetry under weakly supported conditions has been successfully modeled by a theoretical treatment. The electrochemical reaction of a non-charged electroactive species is presented for the system ferrocene/ferrocenium in acetonitrile in which the oxidized and reduced species are soluble in solution, the reaction is studied at different concentrations of supporting electrolyte. Comparison is presented between theoretical simulations and experimental results, for which potential drop in solution is studied. Then systems involving charged electroactive species are treated, in these cases the decrease of supporting electrolyte influence the mass transport of the electroactive species due to migration, comparison between different experimental systems as hexaammineruthenium (III)/(II), cobaltoceniun/cobaltocene and hexacyanoferrate (III)/(II) are presented in comparison with theoretical simulations. More complex mechanistic paths are also investigated, such as deposition and stripping, in which it is established that the level of support required to achieve ‘diffusion only’ voltammetry is on dependence of the concentration of amalgamated electroactive species prior to the stripping step. Comparison between theoretical simulation and experimental results of the deposition and stripping of thallium at a mercury hemisphere are presented, and found to be in good agreement for either chronoamperometry and cyclic voltammetry Simulations are also presented showing the necessary required amount of supporting electrolyte required to achieve ‘diffusion only’ cyclic voltammetry. This is obtained by comparison between diffusion only software and the simulation described in the present thesis. The required amount of supporting electrolyte is shown to depend on the concentration of the electroactive species and supporting electrolyte in the media, the electrode radius, the diffusion coefficient of species and the scan rate. Finally, the cyclic voltammetry in weakly supporting media is used to obtain mechanistic information, by using the migration of electroactive species to differentiate the mass transport of electroactive species to the electrode. The two single electron reductions of anthraquinone in acetonitrile is presented, and the comproportionation mechanistic path is observed in weakly supported media, diffusion only voltammetry is normally unable to present whether this mechanism path takes place, due to the similarity in diffusion coefficients of the electroactive species. In contrast in weakly support conditions the diffusion controlled comproportionation mechanistic path is observed experimentally and constraints for the rate constant are discussed.

The experimental work discussed in this thesis examines the effect of voltammetry of nanoparticle arrays with a specific focus on the effect of nanoparticle size and surface coverage on the substrate electrode. These effects are investigated in relation to the reduction of hydrogen peroxide, 4-nitrophenol and the hydrogen evolution reaction. In each case, the experimental data obtained is subsequently fitted using numerical simulations to extract quantitative kinetic data. Distinct differences are noted between macro – and nano – scale. The reduction of H₂O₂ reveals the absence of autocatalysis at the nano – scale. Furthermore, peak potential shifts positively with surface coverage under Case 3 diffusion conditions. On the other hand, at essentially isolated particles (Case 1) Ep varies logarithmically with nanoparticle radius. By decreasing nanoparticle size, we promote convergent diffusion and enhance irreversibility of the process. The shift in Ep with surface coverage can be accounted for as the diffusion layers begin to overlap heavily, tending towards more macro – disk type behaviour. We also study the hydrogen evolution reaction at an array of AgNPs. By fitting of the experimental data with numerical simulations we demonstrate altered kinetics between the macro – and nano – scale. Voltammetry at AgNP – arrays also display dependence between surface coverage and current. We attribute this to increased electro – active surface area and decrease in irreversibility of the process, with increasing surface coverage. Numerical simulations are also used to fit experimental data obtained for the reduction of 4 – nitrophenol in acidic media. The AgNP – arrays exhibit significantly different electrode kinetics compared to a macro – disk. Examination of the data obtained for AgNP – arrays at two different acid concentrations implies the rate - determining step is likely the electron transfer process. We therefore infer a change in α the mechanism of the rate – determining step between macro – and nano – scale. An unusually low value of α at the NP – array may indicate that adsorption plays some role in the process. Furthermore, we discuss the size – dependent adsorption behaviour exhibited by silver nanoparticle arrays, such that small particles with diameters below ~ 50 nm do not display the typical underpotential deposition characteristics of corresponding bulk materials or larger nanoparticles. This phenomenon is reported for the deposition of heavy metals (thallium, lead and cadmium) at silver nanoparticle arrays. The stripping voltammetry of arrays of silver nanoparticles has also been investigated. The stripping peak potential is dependent on the degree of surface coverage. Modelling of this system has shown that a one – dimensional diffusion model is appropriate for high surface coverage; essentially it is experiencing planar diffusion. However, for an array of widely dispersed particles, the individual nanoparticle size and convergent diffusion begin to dominate behaviour. A detailed overview of the literature is first discussed, relating to the synthesis of a wide variety of metallic nanoparticles and their practical applications, such as the determination of pH using platinum nanoparticles as covered in Chapter 7. We also discuss the fundamental principles governing the voltammetric behaviour of nanoparticles.

Hydrogenases are a family of enzyme that catalyses the bidirectional interconversion of H+ and H2. There are two major classes of hydrogenases: the [NiFe(Se)]- and [FeFe]-hydrogenases. Both of these benefit from characteristics which would be advantageous to their use in technological devices for H2 evolution and the generation of energy. These features are explored in detail in this thesis, with a particular emphasis placed on defining the conditions that limit the activity of hydrogenases when reducing H+ to produce H2. Electrochemistry can be used as a direct measure of enzymatic activity; thus, Protein Film Electrochemistry, in which the protein is adsorbed directly onto the electrode, has been employed to probe catalysis by hydrogenases. Various characteristics of hydrogenases were probed. The catalytic bias for H2 production was interrogated and the inhibition of H2 evolution by H2 itself (a major drawback to the use of some hydrogenases in technological devices to produce H2) was quantified for a number of different hydrogenase. Aerobic inactivation of hydrogenases is also a substantial technological limitation; thus, inactivation of both H2 production and H2 oxidation by O2 was studied in detail. This was compared to inhibition of hydrogenases by CO so as to elucidate the mechanism of binding of diatomic molecules and determine the factors limiting inactivation. This allows for a preliminary proposal for the genetic redesigning of hydrogenases for biotechnological purposes to be made. Finally, preliminary investigation of the binding of formaldehyde, potentially at a site integral to proton transfer, opens the field for further research into proton transfer pathways, the structural implications thereof and their importance in catalysis.

This thesis concerns the electrochemical investigation of high-potential laccases. These multicopper oxidases are efficient electrocatalysts for the dioxygen reduction reaction. A method for stabilising laccase on a graphite electrode was established. The method involved modification of the graphite surface by diazonium coupling of a 2-anthracene molecule. A laccase ‘film’ adsorbed on this modified surface remained stable for over two months and, typically, the current density for dioxygen reduction was doubled compared to a laccase ‘film’ on an unmodified surface. Protein film voltammetry was used to investigate thermodynamic and kinetic aspects of the electrochemical behaviour of laccase. The effect of inhibitors on the magnitude of reduction current and the position of the wave (related to the overpotential for the reaction) was also studied. Fluoride, chloride and azide showed different modes of inhibition and inhibition constants ranged from micromolar for azide to millimolar for chloride. In cyclic voltammetry experiments it was only in the presence of high concentrations of the inhibitors fluoride, chloride and azide that a non-turnover signal, corresponding to a one electron transfer process, was revealed. The evidence suggested that the non-turnover signal arose from interfacial electron transfer between the electrode and the type 1 or ‘blue’ copper. Evaluation of the peak areas allowed determination of the catalytic rate constant, kcat, as 300 s–1, and the electroactive surface coverage as four pmol cm–2. The rate of interfacial electron transfer was rapid enough to not limit catalysis at high overpotentials. A spectroelectrochemical cell was designed to investigate the behaviour of the type 1 copper in the presence of inhibitors and at different pH values. The inhibitors fluoride, chloride and azide had little effect on the reduction potential of the type 1 copper, but at higher pH values the reduction potential of the type 1 copper was decreased.

This thesis is concerned with dynamic electrochemistry experiments in which faradaic processes are driven by the application of potential to an electrode immersed in an electrolyte solution. In particular, experimental methods are considered which could be used to study electrochemical systems in a more informative way if the processes occurring at the electrode were better understood. The work develops the theoretical models which describe these experiments, and details the approximations made in each model and the conditions under which they are appropriate. Numerical simulations are reported which demonstrate how the models can be used to infer quantitative details of chemical behaviour from experimentally recorded data. The first system studied in detail is linear sweep voltammetry at a microband electrode array. The diffusional behaviour of an electroactive species is shown to depend on the configuration of the microband array and on the potential scan rate used. Details are given on how experimental conditions can be optimised for the study of electrochemical systems. The next area of work develops the theory of nanoparticle-modified electrodes. Experiments are considered in which an electron transfer reaction is catalysed only at the site of the nanoparticles, whilst the supporting planar electrode remains electrochemically inert. Numerical simulations show how the current measured at these modified electrodes depends on the size and shape of the particles, on the distribution of the particles on the electrode surface, and on the timescale of the experiment. The final theme of work is on electrochemical experiments in poorly conducting solutions. A theoretical model is developed which takes into account the effects of an electric field on the mass transport of electroactive species and on the charge transfer kinetics at the electrode. The model is then used to rationalise the unusual current behaviour that is observed in the anodic stripping of thallium from an amalgam.

SPM, optical microscopy, and electrochemical techniques are used to study a range of electrochemical deposition processes on carbon electrodes, particularly those associated with diffusion-controlled multiple nucleation. Anodic stripping voltammetry for analytical measurements using solid electrodes is addressed in the light of limitations arising from electrode heterogeneity, electrode morphology, inhibited electrodeposition, and incomplete stripping of deposited metal. It is shown, using direct imaging of electrode surfaces, that each of the preceding factors may produce significant deviations from ideal electrode behaviour. The electrochemical nucleation of silver on BDD is examined. Data are obtained for the nucleation rate by interpretation of the deposition voltammetry, and by inspection of in-situ optical microscopic images. The particle distributions are analyzed and a stochastic model of nucleation developed. A model for the potentiostatic nucleation and three dimensional growth of deposits on an electrode surface under hydrodynamic conditions is examined. A wall-tube and stirred cell are used to generate conditions in which the diffusion layer thickness is in the range 10 – 40 μm. It is shown that the model provides excellent fits to the experimental data. A previously unrecognised correlation between the morphology of the PbO2 deposits and their electrocatalytic activity is established. The morphology of the films are observed as a function of time and potential using in-situ AFM. Nanotrench arrays are fabricated on HOPG surfaces. Cyclic voltammetry in simple redox couples is used to provide experimental evidence that the voltammetric response of a graphite electrode is solely due to the edge plane sites, with the basal plane sites having no measurable contribution. Nanotrenches are used as templates in a simple method for generating random assemblies of metal nanobands. This method is shown to be effective for generating gold, silver and copper nanowires. The electrochemical properties of the array are investigated via cyclic voltammetry.
