Background: In many real-world situations there are many components in a mixture that produce an enormous amount of information., Objective: The main task is to build up balanced models that convert these data into meaningful information to deal with. Hence, different chemometric models were applied for the analysis of data obtained from a mixture containing sofosbuvir, ledipasvir, velpatasvir, daclatasvir, and valacyclovir that were recently used internationally for their antiviral activity., Methods: Partial Least Squares, Spectral Residual Augmented Classical Least Squares, and Concentration Residual Augmented Classical Least Squares designs were applied with and without variable selection procedure [Genetic Algorithm (GA)]. The methods were used for the quantitative analysis of the drugs in laboratory prepared mixtures and real market sample through handling the UV spectral data., Results: Robust models were obtained by applying GA. Herein, two new swarm intelligence based algorithms namely; grey wolf optimization (GWO) and antlion optimization (ALO) algorithms were presented, for the first time, as variable selection tools in spectroscopic data analysis. In order to assess the performance of these algorithms, they were applied along with the recently introduced firefly algorithm (FFA) and the well-established genetic algorithm (GA) and particle swarm optimization (PSO) algorithm on four different spectroscopic datasets of varying sizes and nature (UV and IR). Partial least squares (PLS) regression models were built using the selected variables by these algorithms along with the full spectral data as the reference models. The obtained results prove that the ALO and GWO optimization algorithms select variables in most cases less than GA and PSO while keeping the PLS performance almost the same. Six stability-indicating UV-spectrophotometric methods manipulating ratio spectra were utilized for the analysis of cefradine in presence of its alkaline degradate. These methods are different forms of transformations; ratio difference, mean centering, derivative ratio using numerical differentiation, derivative ratio using Savitsky-Golay filter, continuous wavelet transform and derivative continuous wavelet transform. Water was used as a solvent and the linearity ranges were 6-26 μg/mL. Determination of accuracy and precision for the suggested procedures were executed. Assessment of specificity was run through analyzing laboratory prepared mixtures containing cefradine and its alkaline degradate. The suggested methods were useful for cefradine estimation in tablets. In this paper, three rapid, simple, accurate and precise spectrophotometric methods were developed for the determination of meclizine hydrochloride in the presence of pyridoxine hydrochloride without previous separation. The methods under study are dual wavelength (DWL), ratio difference (RD) and continuous wavelet transform (CWT). On the other hand, pyridoxine hydrochloride (PYH) was determined directly at 291nm. The methods obey Beer's law in the range of (5-50μg/mL) for both compounds. All the methods were validated according to the ICH guidelines where the accuracy was found to be 98.29, 99.59, 100.42 and 100.62% for DWL, RD, CWT and PYH; respectively. Moreover the precision of the methods were calculated in terms of %RSD and it was found to be 0.545, 0.372, 1.287 and 0.759 for DWL, RD,CWT and PYH; respectively. The selectivity of the proposed methods was tested using laboratory prepared mixtures and assessed by applying the standard addition technique. A reversed-phase HPLC method (RP-HPLC) with UV detection was developed and validated for the quantitative determination of cefprozil, a second-generation cephalosporin. Due to β-lactam ring instability under alkaline conditions, this RP-HPLC method was applied for the determination of cefprozil in the presence of its possible degradation product. The interactions that govern the separation process with stationary phase were investigated at both molecular and quantum mechanical levels. Moreover, electrostatic potential maps were generated to determine the sites of interaction with mobile phase. The suggested method was validated in compliance with International Conference on Harmonization guidelines and successfully applied for the determination of cefprozil in its commercial pharmaceutical formulation.

Three chemometric methods namely, concentration residual augmented classical least squares (CRACLS), spectral residual augmented classical least squares (SRACLS) and partial least squares (PLS) were applied for the simultaneous quantitative determination of Cinnarizine and Dimenhydrinate in their binary mixtures. All techniques were applied with and without variable selection using genetic algorithm (GA) resulting in six models (CRACLS, GA-CRACLS, SRACLS, GA-SRACLS, PLS, GA-PLS). These models were applied for the simultaneous determination of the drugs in their laboratory prepared mixtures and pharmaceutical dosage form via handling their UV spectral data. It was found that GA based models are simpler and more robust than those built with the full spectral data. For the first time, a new variable selection method based on swarm intelligence namely firefly algorithm is coupled with three different multivariate calibration models namely, concentration residual augmented classical least squares, artificial neural network and support vector regression in UV spectral data. A comparative study between the firefly algorithm and the well-known genetic algorithm was developed. The discussion revealed the superiority of using this new powerful algorithm over the well-known genetic algorithm. Moreover, different statistical tests were performed and no significant differences were found between all the models regarding their predictabilities. This work represents a comparative study of different aspects of manipulating ratio spectra, which are: double divisor ratio spectra derivative (DR-DD), area under curve of derivative ratio (DR-AUC) and its novel approach, namely area under the curve correction method (AUCCM) applied for overlapped spectra; successive derivative of ratio spectra (SDR) and continuous wavelet transform (CWT) methods. The proposed methods represent different aspects of manipulating ratio spectra of the ternary mixture of Ofloxacin (OFX), Prednisolone acetate (PA) and Tetryzoline HCl (TZH) combined in eye drops in the presence of benzalkonium chloride as a preservative. The proposed methods were checked using laboratory-prepared mixtures and were successfully applied for the analysis of pharmaceutical formulation containing the cited drugs. The proposed methods were validated according to the ICH guidelines. A comparative study was conducted between those methods regarding simplicity, limitation and sensitivity.
