1. The Relation between ACE Gene Polymorphism and the Severity of COVID-19 Infection

Abstract

Introduction. The coronavirus disease 2019 (COVID-19) pandemic, which emerged in China at the end of 2019, rapidly spread worldwide. The angiotensin-converting enzyme (ACE) gene contains an insertion/deletion (I/D) polymorphism that leads to a higher serum ACE level which is associated with several diseases and also with a high mortality rate in SARS. Therefore, this study aimed at assessing the association between ACE gene polymorphism and the risk and severity of COVID-19 disease in patients. Methodology. Forty-five SARS-CoV-2 infected patients and another random control group of 45 healthy individuals were included. The detection of ACE I/D gene polymorphism in both groups was done by PCR. Results. 53% of infected patients with SARS-CoV-2 had an ACE deletion/deletion genotype (D/D), 27% had an ACE deletion/insertion genotype (D/I), and 20% had an ACE insertion/insertion genotype (I/I). On the one hand, the D/D variant was significantly detected in the COVID-19 patients compared to the control subjects, whereas the I/I variant was significantly detected in the control subjects compared to the COVID-19 patients (p = 0.004). The D/D variant subgroup showed the lowest lymphocytic count compared to the D/I or I/I subgroups. In addition, the C-reactive protein was significantly higher and the oxygen saturation was significantly lower in patients with the D/D allele compared to the other subgroups. Conclusions. ACE gene polymorphism, particularly the DD genotype, was observed to affect the severity of COVID-19 infection.

Background. Alopecia areata (AA) is an acquired hair loss disorder induced by a cell-mediated autoimmune attack against anagen hair follicles. CD27-CD70 is a receptor-ligand complex which enhances T helper and cytotoxic T cell activation, survival, and proliferation. The overstimulation of this complex can lead to a lack of tolerance and the development of autoimmunity. Objectives. This study aimed to assess the gene expression of CD27 and CD70 in patients with AA. Methods. CD70 and CD27 mRNA expressions were evaluated by a quantitative real-time polymerase chain reaction in scalp biopsies from 40 AA patients (both AA lesions and non-lesional areas) and 40 healthy controls (HCs). The Severity of Alopecia Tool (SALT) score was used to assess AA severity. Abstract

Abstract Background Vascular endothelial growth factor (VEGF) isoforms, particularly the diffusible VEGF-121, could play a major role in the response of recurrent glioblastoma (GB) to anti-angiogenetic treatment with bevacizumab. We hypothesized that circulating VEGF-121 may reduce the amount of bevacizumab available to target the heavier isoforms of VEGF, which are the most clinically relevant. Methods We assessed the plasma level of VEGF-121 in a brain xenograft model, in human healthy controls, and in patients suffering from recurrent GB before and after bevacizumab treatment. Data were matched with patients’ clinical outcome. Results In athymic rats with U87MG brain xenografts, the level of plasma VEGF-121 relates with tumor volume and it significantly decreases after iv infusion of bevacizumab. Patients with recurrent GB show higher plasma VEGF-121 than healthy controls (p = 0.0002) and treatment with bevacizumab remarkably reduced the expression of VEGF-121 in plasma of these patients (p = 0.0002). Higher plasma level of VEGF-121 was significantly associated to worse PFS and OS (p = 0.0295 and p = 0.0246, respectively). Conclusions Quantitative analysis of VEGF-121 isoform in the plasma of patients with recurrent GB could be a promising predictor of response to anti-angiogenetic treatment.

There were significant positive correlations between AA severity and gene expression of CD70 ( p < 0.001 ) and CD27 ( p = 0.030 ) in AA lesions. Significant associations were detected between signs of AA activity and lesional gene expression of CD70 and CD27. Additionally, CD70 and CD27 gene expression was significantly lower in non-lesional biopsies compared to HCs ( p < 0.001 ). Conclusion. Gene expression of CD70 and CD27 was increased in AA lesions and was associated with disease severity and activity. Thus, both molecules can be a predictor of AA severity and activity. Furthermore, the expression was reduced in non-lesional scalp areas. Thus, a lack of CD27 and CD70 expression may initially predispose to immunological dysregulation and the development of AA.

Abstract

International audience; This declaration, signed by an interdisciplinary task force of 234 experts from 83 different countries with different backgrounds, highlights the threat posed by antimicrobial resistance and the need for appropriate use of antibiotic agents and antifungal agents in hospitals worldwide especially focusing on surgical infections. As such, it is our intent to raise awareness among healthcare workers and improve antimicrobial prescribing. To facilitate its dissemination, the declaration was translated in different languages.

This declaration, signed by an interdisciplinary task force of 234 experts from 83 different countries with different backgrounds, highlights the threat posed by antimicrobial resistance and the need for appropriate use of antibiotic agents and antifungal agents in hospitals worldwide especially focusing on surgical infections. As such, it is our intent to raise awareness among healthcare workers and improve antimicrobial prescribing. To facilitate its dissemination, the declaration was translated in different languages.

Abstract

Background: Escherichia coli (E.coli) is responsible for over 80 percent of urinary tract infections (UTI). The emergence and development of multi-drug resistant (MDR) strains is due to inappropriate use of antibiotics and the horizontal gene transfer between bacteria. The MDR strains of E. coli are highly associated with the presence of integrons; also, extended-spectrum beta lactamase (ESBL) producing isolates are usually resistant to various antibiotics. This study aimed to determine the incidence of class 1 integrons and its association with drug resistance in ESBL producing E. coli isolated from patients who were suffering from UTI. Methods: This study was conducted on 232 hospitalized patients with UTI, from which 160 E. coli strains were isolated. Antibiotic susceptibility testing and screening for ESBL production were performed by Kirby-Bauer disk diffusion method on Mueller-Hinton agar. Confirmation for ESBL production was performed by combined disc diffusion test. All the MDR ESBL producing E. coli isolates were examined by conventional polymerase chain reaction (PCR) for the presence of intI1 gene and related gene cassettes. Results: One hundred sixty E. coli strains (69%) were isolated from 232 hospitalized patients. The highest percentage of resistance was to aztreonam (92%) followed by ceftazidime and cefotaxime (90%) then ciprofloxacin (79%). Seventy-two E. coli isolates (45%) were found to be ESBL producers and out of them, 61 isolates (84.7%) were MDR. Out of the 61 MDR ESBL-producing isolates, class I integron was identified in 56 isolates (91.8%). Conclusion: Our findings indicate a high rate of MDR. Most ESBL-producing isolates are MDR and the high prevalence of class 1 integrons and gene cassettes suggests possible risk for the dissemination of resistance genes and the spread of MDR bacteria. Abstract

Neural stem cells are multipotent self-renewing cells that could be used in cellular-based therapy for a wide variety of neurodegenerative diseases. Here, to assess the ability of Carbon Nanotubes (CNTs) to enhance the therapeutic potential of human Olfactory Bulb Neural Stem Cells (OBNSCs) for restoring cognitive deficts and neurodegenerative lesions, we co-engrafted CNTs and human OBNSCs in trimethyltin (TMT)-neurodegenration rat model. The present study revealed that engrafted human OBNSCs-CNTs restored cognitive deficits and neurodegenerative changes associated with TMT-induced rat neurodegeneration model. Moreover, the CNTs seemd to provide a support for engrafted OBNSCs with increasing their tendecny to differentiate into neurons rather than into glia cells.
