To develop and utilize the oil of Pinus seeds and explore natural resources rich in pinolenic acid (PNLA), twenty-one Pinus taxa were evaluated in a search of Δ5-unsaturated polymethylene-interrupted fatty acids (Δ5-UPIFA)-rich oils. While the fatty acid (FA) composition was determined by GC-FID and GC-MS, NMR of crude oils proved to be a fast method for establishing the ratio between Δ5-UPIFA and total FA. For all analyzed taxa, both the geographical origin and the concentration of total FA in the seeds are provided. PNLA and sciadonic acids occurred in all samples, while taxoleic and bishomopinolenic acids were present in most taxa. PNLA reached a maximum of 28.3% of total FA in P. mugo, and P. koraiensis showed the highest total FA amount (66.8 g/100 g seeds). The previously unanalyzed taxon P. ponderosa var. scopulorum can be considered a new PNLA source (17.1%). Principal Component Analysis showed that the similarities in FA profiles allow the grouping of the various taxa within Pinus subsections and confirmed the differential metabolic activities of Δ5 and Δ9 desaturase enzymes. This study showed that several underutilized Pinus taxa could be developed into renewable woody oil species, and their seeds could be used as raw materials for Δ5-UPIFA-rich oils extraction.

γ-linolenic acid (GLA, 18:3n-6) is a bioactive fatty acid (FA) that exerts several healthy actions; however, its occurrence is restricted to a few oils. The goal of this study was to detect GLA-rich Ribes species and cultivars (cv), and to achieve this the seeds of 7 Ribes taxa and 50 Ribes cv were surveyed for FA profiles. The highest GLA percentages were found in R. nigrum cv 'Plotnokistnaya', 'Volshebnica', 'Atlant' and 'Nara' (22.6, 22.1, 20.9, and 20.0% of total FA, respectively) and also in R. komarovii (19.6%) and R. nigrum var. sibiricum (18.3%). Stearidonic acid (SDA, 18:4n-3) had the highest values in both R. rubrum 'Konstantinovskaya' and R. niveum 'Smolyaninovskaya' (4.8%). GLA content ranged from 0.4 in some R. rubrum cv and R. niveum 'Smolyaninovskaya' to 3.5 g/100 g seeds in R. nigrum 'Plotnokistnaya'. Principal component analysis (PCA) was performed using PUFA profiles, which allowed grouping Ribes sections as well as black currant cv derived from different pedigree within the section Coreosma. All taxa and cv checked here are valuable by-product sources, given the high GLA percentages contained in their seed oils. Such cv could be used for healthy oils production, as well as for breeding to obtain new cv with improved GLA concentrations.

Ahiflower oil© is extracted from the seeds of Buglossoides arvensis, which contains high amounts of stearidonic acid (SDA, 18:4n-3), while its phenolic composition still is unreported. Moreover, several Buglossoides taxa remain unstudied and could become natural sources of SDA. In this work, seeds of several Buglossoides taxa and Ahiflower oil© were screened for fatty acids, phenolic compounds, and in vitro antiproliferative activities against colorectal cancer cells. Four flavonoids and 16 phenolic acids were identified and quantified. Among Buglossoides taxa, the highest amounts of phenolic compounds were found in samples collected in Spain, under a warm Mediterranean climate. In this work, seeds from selected Mertensia species were analyzed for γ-linolenic acid-rich oils and minor functional compounds. Fatty acids (FA) were analyzed by GC-FID; tocopherols, sterols, squalene, and phenolics compounds by HPLC-DAD, and the structures of the latter were confirmed by LC-MS. M. maritima spp. asiatica and M. sibirica showed the highest amounts of γ-linolenic acid (22.8 and 18.7%, respectively) and total FA (15.9 and 10.9 g/100 g seeds, respectively). M. sibirica had the greatest levels of sterols, phenolics and tocopherols (244.8, 243.9 and 66.3 mg/100 g, respectively), in which stigmasterol, rosmarinic acid and α-tocopherol (237.7, 180.1 and 53.6 mg/100 g, respectively) were the most abundant components. M. maritima spp. asiatica and M. arizonica showed the highest amounts of squalene (2.5 and 1.1 mg /100 g seeds). Mertensia species constitute a new source of GLA-rich oils, suitable to be marketed by the pharmaceutical and food industries.

A feed-forward back-propagation neural network (FBN) was used as a tool for predicting the growth dynamics of the microalga Karlodinium veneficum in a culture medium with any specified concentrations of the key nutrients. A 3-layered FBN configuration of 27-25-1 nodes was used. This FBN satisfactorily represented the nonlinear interactions among all the nutrients of a culture medium containing up to 25 different components. The FBN model was trained using the growth dynamics data from more than 420 batch culture experiments involving different media compositions. The relative impact of individual nutrients, the initial cell concentration and the culture duration on growth profiles were determined through a systematic analysis of the partitioning of the FBN connection weights. Microelements and vitamins together had a higher relative impact on growth compared to the impact of the macronutrients. The trained FBN successfully predicted the cell concentration dynamics in cultures with previously untested initial conditions. The FBN proved to be an excellent tool for predicting the growth curves in the range of culture conditions that were relevant to this study.

Argan oil (AO) has traditionally been used for its dermatologic and hepatoprotective effects and for human nutrition. Traditionally, AO extraction is carried out by Moroccan women with the help of stone mill pressing. However, it is long and slow process that frequently is achieved at deficit hygiene conditions. In this research, unroasted seeds (US) and roasted seeds (RS) of argan tree have been extracted using different extraction systems: n-hexane, n-hexane:acetone (1:1, v/v), ethyl acetate, acetone, ethanol:water (96:4, v/v), and water. The main results are; Hexane: acetone extraction yielded the highest FA amounts, at 39.7 g FA/100 g of total US seed, followed by acetone (36.5 g FA/100 g US seed). The main FA in all extracts were oleic acid, from 52.1 (ethanol:water) to 57.7% (hexane), and linoleic acid from 22.9 (hexane, US) to 29.3% (ethanol:water, RS), both of total FA. Oxidation levels, computed by conjugated diene index (CD), was extremely low for all extraction processes. CD were higher in RS in comparison with US, due to compounds produced in Maillard reactions. Colour parameters indicated a color variation from yellow (US) to brown (RS). Therefore, the conclusions can be summarized as follows. All extracting systems yielded good quality argan oil, thus, the tested procedures constitute a suitable alternative to the traditional extraction processes. The obtained oils could be used in the alimentary and cosmetic industries with different applications. Future research should be directed to check the unsaponifiable fractions of the oils here obtained. Overall, this research opens new perspectives for industries focused on argan oil extraction.

Large scale culture of marine dinoflagellate microalgae has proved difficult owing to their extreme sensitivity to hydrodynamic forces in photobioreactors. This work discusses the scale up of Protoceratium reticulatum dinoflagellate culture from a successful 2 L stirred-tank photobioreactor operation to a 15 L stirred photobioreactor. Both bioreactors were equipped with internal spinfilters for cell retention. A semicontinuous perfusion culture in the 15 L photobioreactor proved to be more productive than fedbatch perfusion culture. Under the best operational conditions, the average cell productivity in the 15 L photobioreactor was 5228 cell mL−1 day−1, or nearly 3.7-fold greater than the best attainable value in static flask cultures, but similar to the results obtained in the 2 L stirred tank. At 9.16 gL −1 day −1 the average volumetric productivity of yessotoxin in the scaled up operation in the semicontinuous perfusion mode was comparable to the results obtained in the small bioreactor. P. reticulatum has a cell damage threshold shear rate of as low as 0.1 s −1 . Notwithstanding this extreme shear sensitivity, the results suggest a good potential for mass scale culture of this dinoflagellate in suitably designed photobioreactors.

The red-tide dinoflagellate Protoceratium reticulatum is shown to be protected against turbulence-associated damage by the use of the additives Pluronic F68 (PF68) and carboxymethyl cellulose (CMC) in the culture medium. Relative to agitated controls, these additives had a dose-dependent protective effect at concentrations of up to 0.4 and 0.5 g L(-1) for CMC and F68, respectively. In static cultures, these additives inhibited growth directly or indirectly at a concentration of0.5 g L(-1). Compared to CMC, PF68 was a better protectant overall. Cell-specific production of yessotoxins was enhanced under elevated shear stress regimens so long as the turbulence intensity was insufficient to damage the cells outright. Shear-induced production of reactive oxygen species and direct effects of turbulence on the cell cycle contributed to the observed shear effects.

Dinoflagellates are potentially important sources of high-value toxins in biomedical, toxicological and chemical research programs. However, the difficulty to culture them in bioreactors limits the development of new products. L1 and other usual dinoflagellates media have been shown to not support an elevated cell growth. The present work evaluated the nutrient uptake rates and nutrient cell yields of Protoceratium reticulatum in different culture modes that allowed to maintain a high concentration of cells under a quasi-steady-state concentration of nutrients. Monitoring of the cellular ROS and lipoperoxides contents in low-shear bioreactor cultures is proposed as culture health-indicating parameters. The amount of yessotoxins (YTXs) produced by the strains studied to date was very different among them and are importantly conditioned by the culture conditions; therefore the strain selection is a key issue. In this work, we evaluated the growth and toxin production of two strains of P. reticulatum which are known to produce very different YTX quantities. Dinoflagellates have proven extremely difficult to culture because they are inhibited by low-level shear forces. Specific growth rate of the toxic dinoflagellate Protoceratium reticulatum was greatly decreased compared with static control culture by intermittent exposure to a turbulent hydrodynamic environment with a bulk average shear rate that was as low as 0.3 s−1. Hydrodynamic forces appeared to induce the production of reactive oxygen species (ROS) within the cells and this caused peroxidation of cellular lipids and ultimately cell damage. Exposure to damaging levels of shear rate correlated with the elevated level of lipoperoxides in the cells, but ROS levels measured directly by flow cytometry did not correlate with shear induced cell damage. This was apparently because the measured level of ROS could not distinguish between the ROS that are normally generated by photosynthesis and the additional ROS produced as a consequence of hydrodynamic shear forces. Continuously subjecting the cells to a bulk average shear rate value of about 0.3 s−1 for 24-h caused an elevation in the levels of chlorophyll a, peridinin and dinoxanthin, as the cells apparently attempted to counter the damaging effects of shear fields by producing pigments that are potential antioxidants. In static culture, limitation of carbon dioxide produced a small but measureable increase in ROS. The addition of ascorbic acid (0.1 mM) to the culture medium resulted in a significant protective effect on lipid peroxidation, allowing cells to grow under damaging levels of shear rates. This confirmed the use of antioxidant additives as an efficient strategy to counter the damaging effects of turbulence in photobioreactors where shear sensitive dinoflagellates are cultivated. © 2009 American Institute of Chemical Engineers Biotechnol. Marine dinoflagellates are potentially important innovative sources of high-value toxins in biomedical, toxicological and chemical research programs. However, little is known about the difficulties related to dinoflagellate cultures. In this article, we demonstrate that the shear sensitivity of cells may be one of the main causes. The red-tide Protoceratium reticulatum , a producer of yessotoxins, was used to examine the effect of hydromechanical shear stress associated with intermittent fluid agitation on cell growth. Shaken flasks, widely used in biotechnological process research, were used as model bioreactors, as hydrodynamic shear stress is relatively easy to quantify in them. Intermittent turbulence regime was characterized by three key operating variables: shear stress, cycle time or shaking frequency, and fraction of time shaken per agitation cycle. The light/dark cycle was also used as another variable. Cell damage depended on the combination of the above-mentioned variables. A damage threshold was observed at an average shear stress of approximately 0.16 mN m −2 (equivalent shear rate of 0.12 s −1 ). Cell damage from exposure to average detrimental shear stress was also shown to be greater in the dark than in the light period. Preliminary experiments demonstrated that dinoflagellates are also much more sensitive to bubbling than the majority of common fragile microalgae. Although slight toxicity of Pluronic F-68 was observed at a concentration of 0.05% (w/v), this protective medium additive considerably reduced cell breakage. On the other hand, no cell adaptation to stronger shear stress was observed. Finally, the implications of the proposed approach for the hypothetical mass culture of dinoflagellates in bioreactors were also thoroughly assessed.

Dinoflagellates are microalgae that are associated with the production of many marine toxins. These toxins poison fish, other wildlife and humans. Dinoflagellate-associated human poisonings include paralytic shellfish poisoning, diarrhetic shellfish poisoning, neurotoxic shellfish poisoning, and ciguatera fish poisoning. Dinoflagellate toxins and bioactives are of increasing interest because of their commercial impact, influence on safety of seafood, and potential medical and other applications. This review discusses biotechnological methods of identifying toxic dinoflagellates and detecting their toxins. Potential applications of the toxins are discussed. A lack of sufficient quantities of toxins for investigational purposes remains a significant limitation. Producing quantities of dinoflagellate bioactives requires an ability to mass culture them. Considerations relating to bioreactor culture of generally fragile and slow-growing dinoflagellates are discussed. Production and processing of dinoflagellates to extract bioactives, require attention to biosafety considerations as outlined in this review.

Marine sponges are sources of high-value bioactives. Engineering aspects of in vitro culture of sponges from cuttings (explants) are poorly understood. This work develops a diffusion-controlled growth model for sponge explants. The model assumes that the explant growth is controlled by diffusive transport of at least some nutrients from the surrounding medium into the explant that generally has a poorly developed aquiferous system for internal irrigation during early stages of growth. Growth is assumed to obey Monod-type kinetics. The model is shown to satisfactorily explain the measured growth behavior of the marine sponge Crambe crambe in two different growth media. In addition, the model is generally consistent with published data for growth of explants of the sponges Disidea avara and Hemimycale columella. The model predicted that nutrient concentration profiles for nutrients, such as dissolved oxygen within the explant, are consistent with data published by independent researchers. In view of the proposed model's ability to explain available data for growth of several species of sponge explants, diffusive transport does play a controlling role in explant growth at least until a fully developed aquiferous system has become established. According to the model and experimental observations, the instantaneous growth rate depends on the size of the explant and all those factors that influence the diffusion of critical nutrients within the explant. Growth follows a hyperbolic profile that is consistent with the Monod kinetics.

Fractions of viable cells, apoptotic and irreversibly damaged cells, dead whole cells and cell fragments were measured by flow cytometry during the production of freely suspended primary cells from explants of the marine sponge Axinella damicornis. The explants were disintegrated using the well-known Muller protocol [W.E.G. Muller, M. Wiens, R. Batel, R. Steffen, R. Borojevic, M.R. Custodio, Establishment of a primary cell culture from a sponge: primmorphs from Suberites domuncula, Mar. Ecol. Progr. Ser. 178 (1999) 205–219]. Supplementation of the standard Ca2+- and Mg2+-free artificial seawater of the Muller protocol, with the shear protectant Pluronic F68 (0.1%, w/v) greatly reduced the cell damage and enhanced the recovery of viable cells at each of the four stages of the protocol. Agitation of cells on an orbital shaker at 75 rpm essentially killed all the viable cells within 2.5 h, but no loss of viability occurred at a higher agitation speed of 100 rpm for up to 6 h when the cells were supplemented with Pluronic F68. This time-dependent loss in viability could be significantly reduced by processing at 3 °C instead of the normal 17 °C. A four-step mechanistic model was shown to describe the kinetics of cell death and fragmentation within ±10% of the measured values. The damage to cells was modeled as a web of first-order processes that did not depend on cell–cell interactions. The forces in the agitated fluid killed the viable cells by impact, which was not accompanied by cell rupture (i.e. the cell was left dead, but intact).

Pilot-scale (0.19 m column diameter, 2 m tall, and 60 L working volume) outdoor vertical bubble column (BC) and airlift photobioreactors (a split-cylinder (SC) and a draft-tube airlift device (DT)) were compared for fed-batch mixotrophic culture of the microalga Phaeodactylum tricornutum UTEX 640. The cultures were started photoautotrophically until the onset of a quasi-steady-state biomass concentration of 3.4 g L(-)(1). After this, the cultures were supplemented with organic nutrient (glycerol 0.1 M) and a reduced nitrogen source, resulting in an immediate growth rate boost, which was repeated with successive additions of nutrients in all three photobioreactors. During this period the biomass productivity was enhanced compared to photoautotrophic cultures in the three reactors, although differences were found among them. These could be attributed to the different hydrodynamic behavior influencing the transport phenomena inside the cultures. A 25.4 g L(-)(1) maximum biomass concentration was attained in the SC. Further additions of nutrients did not promote any more growth. The consumption of glycerol was quantitative in the first additions but slowed at high biomass concentration, suggesting that a minimum amount of light is needed to sustain growth. No significant effect of the supplied organic nutrient on carotenoids and chlorophylls content was observed, although it had a profound effect on the fatty acid composition. Eicosapentaenoic acid (EPA) content was increased up to 3% (DW) in mixotrophic growth, giving a productivity of 56 mg L(-)(1) d(-)(1), a significant increase compared to the photoautotrophic control, which yielded a maximum EPA content of 1.9% (DW) and a productivity of 18 mg L(-)(1) d(-)(1). The maximum biomass and EPA volumetric yields obtained in this work are comparable with those reported for commercial photoautotrophic monoculture of large quantities of P. tricornutum in closed continuous-run tubular loop bioreactors with tubes that are typically less than 0.08 m in diameter. When the comparison is established in terms of productivities based on the land area occupied, the vertical airlift and bubble-column bioreactors are extraordinarily more productive.

Marine sponges are potential sources of many unique metabolites, including cytotoxic and anticancer compounds. Natural sponge populations are insufficient or inaccessible for producing commercial quantities of metabolites of interest. This review focuses on methods of producing sponge biomass to overcome supply limitations. Production techniques discussed include aquaculture in the sea, the controlled environments of aquariums, and culture of sponge cells and primmorphs. Cultivation in the sea and aquariums are currently the only practicable and relatively inexpensive methods of producing significant quantities of sponge biomass. In the future, metabolite production from cultured sponge cells and primmorphs may become feasible. Obtaining a consistent biomass yield in aquariums requires attention to many factors that are discussed in this work.

Explants of the sponge Crambe crambe were cultured in natural seawater, with or without marine microalga (Phaeodactylum tricornutum) in discontinuous flow through systems and in continuous flow-through systems (DFTHS and CFTHS, respectively). Growth was measured as the increase in underwater weight. In the experiment carried out in the CFTHS, the explants average underwater weight increased by up to 1380% of the initial weight in 22–45 days. Growth in DFTHS was much slower producing a gain of up to an average value of 322% of the initial weight in 100–210 days. Growth kinetics varied considerably for different explants. Explants grew fastest in the first 10-days of subculture. The sponges grew better in CFTHS compared with the DFTHS. The high growth rates observed in CFTHS suggest that this technique is a promising method for culturing C. crambe in closed systems.

The polyunsaturated fatty acids (PUFA) eicosapentaenoic acid (EPA, 20∶5n−3) and docosahexaenoic acid (DHA, 22∶6n−3), which have several pharmaceutical properties, have been purified from cod liver oil. The process consisted of four main steps: (i) saponification of the oil, (ii) use of urea inclusion adducts method to obtain PUFA, (iii) PUFA methylation, and (iv) argentation silica gel column chromatography of the methylated PUFA. Argentation silica gel chromatography yielded highly pure DHA in the process (100% purity, 64% yiild). For EPA, the recovery in the combined process was 29.6%, and the final purity was 90.6%, owing to the simultaneous elution of other polyunsaturated fatty esters. The recovery in the urea inclusion method was strongly enhanced by application of orbital agitation during the crystallization process, in which EPA yield increased from 60–70% without agitation to 90–97% at 800 rpm; stearidonic acid (18∶4n−3) yield ranged from 60–75% without agitation to 87–95% at 800 rpm, and DHA yield varied from 53–73% without agitation to 85–99% at 800 rpm

The free fatty acids eicosapentaenoic acid (C20:5ω3, EPA) and docosahexaenoic acid (C22:6ω3), obtained from the microalgae Phaeodactylum tricornutum, and the EPA methyl ester were compared with regard to their extent of peroxidation using different storage conditions. Several series were stored according to selected variables: hexane addition versus no addition, 4 °C versus 25 °C, and antioxidant addition (octyl gallate) versus no antioxidant addition, always in the dark. Previously, the EPA and methyl EPA structures were confirmed by NMR spectra to discard the formation of conjugated dienes after the downstream process. The results showed that the stability was higher for methyl EPA than for the free fatty acid, and that peroxidation can be retarded by low temperature storage and mainly by hexane addition. The peroxidation process was evaluated by the peroxide value (spectrophotometric method by iodine absorption), although the conjugated diene absorbance and the loss in percentage of the fatty acid have been tested as good indicators of the peroxidation process. A simple kinetic model that explains the peroxidation process during the initiation and propagation steps is given.

A low expense process is developed for recovering esterified eicosapentaenoic acid (EPA) from microalgae and fish oil. Over 70% of the EPA content in the esterified crude extract of microalgae were recovered at purities exceeding 90%. The recovery scheme utilizes either wet or freeze-dried algal biomass. The process consists of only three main steps: 1) simultaneous extraction and transesterification of the algal biomass; 2) argentated silica gel column chromatography of the crude extract; and 3) removal of pigments by a second column chromatographic step. Argentated silica gel chromatography recovered about 70% of the EPA ester present in the crude fatty ester mixture of fish oil, but at a reduced purity ( approximately 83% pure) compared to the microalgal derived EPA. The optimal loading of the fatty ester mixture on the chromatographic support was about 3% (w/w) but loadings up to 4% did not affect the resolution significantly. The process was scaled up by a factor of nearly 320 by increasing the diameter of the chromatography columns. The elution velocity remained constant. Compared to the green alga Monodus subterraneus, the diatom Phaeodactylum tricornutum had important advantages as a potential commercial producer of EPA. For a microalgal EPA process to be competitive with fish oil derived EPA, P. tricornutum biomass (2.5% w/w EPA) needs to be obtained at less than $4/kg. If the EPA content in the alga are increased to 3.5%, the biomass may command a somewhat higher price. The quality of microalgal EPA compares favorably with that of the fish oil product. Compared to free fatty acid, EPA ester is more stable in storage. Shelf-life is extended by storing in hexane. The silver contamination in the final purified EPA was negligibly small (210 ppb).

Acyl-lipid composition of Isochrysis galbana, Porphyridium cruentum and Phaeodactylum tricornutum from stationary-phase cultures has been analyzed by TLC and GC. Additionally, P. tricornutum from an outdoor tubular photobioreactor was also studied. Neutral (NLs) and glyco (GLs) lipids were found in similar amounts of ca 40–45% each, with phospholipids (PLs) representing ca 10–20%. The major lipid classes were triacylglycerol (TAG), monogalactosylaylglycerols (MGD), and galactosylacylglycerols (DGD), usually in that order. The P. tricornutum biomass taken from the bioreactor showed a distinctive acyl-lipid composition. GL content was nearly twice (56%) that of the indoor culture (31%) and NL content (31%) was nearly half that of the indoor culture (54%). These changes mainly involved TAG and MGD. The fatty acid composition of lipids in the outdoor culture generally remained unaffected, except for MGD, DGD, phosphatidylcholine (PC) and phosphatidylethanolamine (PE), in which eicosapentaenoic acid (EPA) content was greatly increased. TAG, the main class of lipids, always contained a high proportion of EPA, 16:0 and 16:1. In the typical (SQD) composition, 14:0, 16:0 and 16:1 accounted for around 70% of fatty acids, with small amounts of polunsaturated fatty acids (PUFAs). The lipid classes which typically had the highest PUFA content in the microalgae were MGD and DGD. In P. tricornutum , these lipids characteristically contained a large proportion of 16:1, 16:2, 16:3 and 16:4. PC, which was composed of the main C 16 fatty acids and the major PUFAs, was usually the most abundant phospholipid. Phosphatidylgycerol showed an accumulation of 16:1 (probably a mixture of 16:1n7 and 16:1n3 trans ) as a distinctive feature. Phosphatidylinositol was characterized by 16:0 and 16:1 (and 14:0 in I. galbana ), which together accounted for over 50% of fatty acids, and significant presence of the main PUFAs. There was no consistent fatty acid pattern for PE in the three microalgae studied. PE was exceptional in I. galbana , containing 61% docosahexaenoic acid.

The 55-6 murine B cell hybridoma line not constitutively expressing CD40 was treated with increasing amounts of intact anti-mouse surface immunoglobulin G antibody (anti-mIgG) either not preincubated or preincubated for 48 h with lipopolysaccharide (LPS). In vitro, cross-linking of surface immunoglobulin G (sIgG) with the whole molecule of anti-IgG antibodies induced the expression of CD69, CD40, and CD19 surface antigens on 55-6 cells. The effect of sIgG ligation was dose-dependent, and preincubation with LPS enhanced their responsiveness to anti-mIgG stimulation. The expression of these surface molecules reached the maximum value during the first part of the cell cycle, corresponding to the position of the G1 peak of the DNA distribution. Stimulation of cells with anti-mIgG did not induce changes either in the number of viable cells or in the fraction of cells undergoing proliferation (mitosis). However, preincubation of 55-6 cells with LPS for 48 h before stimulation with anti-mIgG increased both the maximum specific growth rate (micromax) and the percentage of cells in the G2/M phase, in comparison with non-preincubated cells. Moreover, on cells preincubated with LPS prior to anti-mIgG treatment, specific IgG2a production rate was enhanced significantly compared to that obtained in control cultures. The correlation between the antibody production rate and the amount of IgG that is detectable on the cell surface was analyzed by flow cytometry. A good correlation between secreted and surface IgG was observed, and the results of cell cycle analyses demonstrated that the 55-6 hybridoma cell line has a substantially higher sIgG content in G1 phase.

Fed-batch and perfusion cultures were carried out in a traditional glass 2-L bioreactor with the toxic dinoflagellate Protoceratium reticulatum. The maximum cell concentration obtained was 2.3 x 10 5 cell·mL -1 , which is almost 1 order of magnitude higher than the maximum previously referenced for this species. L1 medium was shown to be clearly deficient in nitrate and phosphate for this strain, and addition of highly concentrated aliquots of these nutrients allowed higher cell concentrations to be obtained. This species consumed high amounts of nitrate and phosphate, 2.1 x 10 -3 and 2.3 x 10 -4 μmol·h -1 ·cell -1 , respectively. However, this consumption produced a very low number of cells compared to other classes of microalgae, indicating that this species is, like other dinoflagellates, a poor competitor in terms of utilization of inorganic nutrients. Higher production of toxins and pigments was strongly associated with cell number in the culture, with maximum values of 700 ng·mL -1 and 1321 μg·mL -1 , respectively. Most yessotoxins remained within the cells and not in the cell-free culture medium, and their production was not related to either the age of the culture or the cell growth phase.

Marine sponges are potential sources of many unique metabolites, including cytotoxic and anticancer compounds. Natural sponge populations are insufficient or inaccessible for producing commercial quantities of metabolites of interest. It is commonly accepted that tissue (fragments, explants, and primmorphs) and in vitro cell cultivation show great potential. However, there is little knowledge of the nutritional requirements of marine sponges to carry out efficient and sustained in vitro culture and progress has been slow. In marine invertebrate fila many unsuccessful attempts have been made with in vitro cultures using typical commercial animal cell media based on sources of dissolved organic carbon (DOC) (e.g., DMEM, RPMI, M199, L-15, etc.). One of the reasons for this failure is the use of hardly identifiable growth promoters, based on terrestrial animal sera. An alternative is the use of extracts from marine animals, since they may contain nutrients necessary for growth. In this work we have cultivated in vitro explants of the encrusting marine sponge Crambe crambe. It is one of the most abundant sponges on the Mediterranean coastline and also possesses an array of potentially active metabolites (crambines and crambescidins). Initially a new approach was developed in order to show consumption of DOC by explants. Thus, different initial DOC concentrations (300, 400, 700 and 1200 mg DOC L(-1)) were assayed. Consumption was evident in all four assays and was more marked in the first 6 h. The DOC assimilation data were adjusted to an empirical model widely used for uptake kinetics of organic dissolved compounds in marine invertebrates. Second, a protocol was established to cultivate explants in vitro. Different medium formulations based on RPMI 1640 commercial medium enriched with amino acids and inorganic salts to emulate seawater salinity were assayed. The enrichment of this medium with an Octopus aqueous extract in the proportions of 10% and 20% (v/v) resulted in an evident sustained long-term growth of C. crambe explants. This growth enhancement produced high metabolic activity in the explants, as is confirmed by the high ammonium and lactate content in the medium a few days after its renewal and by the consumption of glucose. The lactate accumulation increased with the size and age of explants. Prior to these experiments, we successfully developed a robust new alternative method, based on digital image treatment, for accurate determination of the explant apparent volume as growth measure.

Commercial production of intracellular microalgal metabolites requires the following: (1) large-scale monoseptic production of the appropriate microalgal biomass; (2) recovery of the biomass from a relatively dilute broth; (3) extraction of the metabolite from the biomass; and (4) purification of the crude extract. This review examines the options available for recovery of the biomass and the intracellular metabolites from the biomass. Economics of monoseptic production of microalgae in photobioreactors and the downstream recovery of metabolites are discussed using eicosapentaenoic acid (EPA) recovery as a representative case study.

Explants of the tropical sponge Pseudosuberites andrewsi were fed with the marine diatom Phaeodactylum tricornotum. The food was supplied either as intact algae or as a filtered crude extract. Growth (measured as an increase in underwater weight) was found in both experiments. The explants fed with intact algae increased to an average underwater weight of 255% of the initial weight in 45-60 days. The explants fed with crude extract increased to an average of 200% of the initial weight in 30 days. These results show that it is possible to grow a sponge using a single microorganism species as a food source. In addition, it was demonstrated that sponges are also capable of growing on non-particulate food. The influence of culture age and nitrogen concentration on the distribution of fatty acids among the different acyl lipid classes has been studied in continuous cultures of the microalga Phaeodactylum tricornutum. The culture age was tested in the range of 1.15-7 days, controlled by adjusting the dilution rate of fresh medium supplied. The effect of nitrogen concentration was tested from saturating conditions to starvation by modifying nitrate concentration in the fresh medium. Culture age had almost no influence on the fatty acid content; 16:0, 16:3 and 20:5 increased moderately wherein the level of 16:1 decreased when the culture age decreased. Culture age had no effect on the total fatty acid content that remained around 11% of dry weight. Conversely, culture age had a greater impact on lipid classes, producing changes in amounts of triacylglycerols (TAG) which ranged between 43% and 69%, and galactolipids (GLs) that oscillated between 20% and 40%. In general, the content of polar lipids of the biomass decreased with culture age. The other factor assayed, nitrogen content, affected the fatty acid profile. Saturated and monounsaturated fatty acids accumulated when the nitrogen concentration was decreased. The experiments regarding the effect of nitrogen concentration on lipid species were carried out with cells of an average age of 3.5 days. A decrease of the nitrogen concentration caused the GL fraction to decrease from 21 to 12%. Conversely, both neutral lipids (NLs) and phospolipids (PLs) increased from about 73 to 79% and from 6 to 8%, respectively. In these experiments, TAG was the lipid class with the highest increase, from 69 to 75%.
