6-N-Cyclohexyl-2′-O-methyladenosine was formulated into controlled release formulations exhibiting comparable in vitro release profiles using different formulation principles, i.e. osmotic pump tablets, membrane-coated pellets and hydrophilic matrix tablet. Dissolution behaviour of these formulations was evaluated in vitro under various testing conditions to assess the effect of pH and hydrodynamic conditions. It was found that osmotic tablets were not sensitive to dissolution media pH and hydrodynamics change, while drug release from monolithic hydrophilic matrix tablets were pH-dependent. When tested in vivo in dogs, it was found that metabolism of 6-N-Cyclohexyl-2′-O-methyladenosine was extensive and appeared to be saturable based on a pharmacokinetic study. Cumulative percent input in vivo (%dose) was obtained by numerical deconvolution, and compared to in vitro release profiles. A linear correlation between fraction absorbed (FRA) in vivo and fraction dissolved (FRD) in vitro was established for osmotic tablets—a true zero-order release formula, whereas only a nonlinear correlation was obtained for membrane-coated pellets. The difference in the in vivo behaviour of these formulations, despite their similar in vitro release characteristics, demonstrated the effect of different controlled release principles on their in vivo bioavailability. The curvature of fraction absorbed in vivo vs. fraction dissolved in vitro for membrane-coated pellets indicated that there was a time-scale difference between in vivo and in vitro testing. In conclusion, drug release from the osmotic system was independent of in vitro and in vivo conditions, where best sustained release effect was achieved, whereas the in vitro dissolution test employed for membrane-coated pellets and hydrophilic matrix tablets needed to be optimized to be biorelevent.

This report investigated dissolution stability of benazepril hydrochloride tablets.Reduction in dissolution rate was observed for benazepril hydrochloride tablets when they were subjected to stressed storage condition (40 degrees C/75% RH) for prolonged periods of time (1-3 months). Moisture contents of initial and stressed tablets were measured by Karl Fischer method. Comparative thermal and physical characterizations of initial and stressed tablets were also performed. A mathematical model that was used to predict possible reduction in dissolution rate was proposed and validated using experimental data.It was found that there was a direct correlation between moisture content of benazepril hydrochloride tablets and their percentage of dissolution at 10 min. At moisture content below 3.5%, there were no significant changes in dissolution values. Beyond that point, however, a close to linear decrease in dissolution was observed as a function of increase in moisture content. Results from thermal and X-ray analysis have ruled out possible changes in drug substance. Other physical characterization, such as scanning electron microscope and mercury porosimetry measurements, revealed changes in core structure of stressed tablets vs. initial tablets. Based on results from these measurements, "preactivation" of disintegrant was identified as the mechanism for reduction in dissolution rate above critical moisture content. A simple physical model for moisture uptake of benazepril hydrochloride tablets was also proposed for predicting when, based on water vapor transmission and critical moisture content, dissolution rate will decline.Physical changes of tablets mediated by moisture were the main cause for reduction in dissolution. Inclusion of desiccant, although beneficial, cannot prevent reduction in dissolution completely. The simple physical model proposed in this report was found to be useful in predicting the dissolution stability of the dosage form.

The solution of a poorly water-soluble drug in a liquid lipid-surfactant mixture, which served as a microemulsion preconcentrate, was converted into a solid form by incorporating it in a solid polyethylene glycol (PEG) matrix. The solid microemulsion preconcentrates thus formed consisted of Capmul PG8 (propylene glycol monocaprylate) as oil, Cremophor EL (polyoxyl 35 castor oil) as surfactant, and hydrophilic polymer PEG 3350 as solid matrix. The drug (aqueous solubility: 0.17 microg/mL at pH 1-8 and 25 degrees C) was dissolved in a melt of the mixture at 65-70 degrees C and then the hot solution was filled into hard gelatin capsules; the liquid gradually solidified upon cooling below 55 degrees C. The solid system was characterized by differential scanning calorimetry (DSC), scanning electron microscopy (SEM), confocal Raman microscopy (CRM), and the dispersion testing in water. It was confirmed that a solid microemulsion preconcentrate is a two-phase system, where clusters of crystalline PEG 3350 formed the solid structure (m.p. 55-60 degrees C) and the liquid microemulsion preconcentrate dispersed in between PEG 3350 crystals as a separate phase. The drug remained dissolved in the liquid phase. Two methods for the measurement of surface pH of pharmaceutical solids, namely, the dye-sorption method and the slurry pH method, were compared. High purity drug substances, instead of excipients, were used as model solids, because acidic or basic impurities present in excipients could influence slurry pH. Solid test samples were prepared by sorption of methanol-water solutions of several indicator dyes, and their diffuse reflectance UV-visible spectra were measured. The solid surface pH values were estimated by comparing base-to-acid peak ratios of the diffuse reflectance UV-visible spectra of solid samples to the calibration plots of dye solutions in aqueous standard buffers of known pH. In the slurry pH method, pH values of concentrated slurries of the compounds in water were considered to represent solid surface pH. The agreement between the two methods was mixed and depended on the compound or the indicator used. It was concluded that in many cases calibration plots of indicator dye spectra in aqueous buffers were not applicable to the solid state, and, as a result, the reliability of the method was low. The slurry method provided a simple and reliable measurement of surface pH indicating that concentrated slurry may closely represent solid surface pH.

The effect of chloride ion (Cl-) on dissolution rates of hydrochloride, mesylate (methanesulfonate) and phosphate salt forms of a model drug, haloperidol, was investigated. The dissolution rates of the salts in 0.01 M HCl from rotating disks followed the order of mesylate>>phosphate>hydrochloride. With additional chloride ion, a decrease in dissolution rate of the hydrochloride salt was observed due to the common ion effect. Dissolution rates of mesylate and phosphate salts also decreased due to their conversion to the HCl salt form on the surfaces of dissolving disks, however, the dissolution rates of mesylate and phosphate salts under identical chloride ion concentrations were still higher than that of the HCl salt.
