Five adult Saanen goats received a single oral dose of Heterophyllaea pustulata containing 42.25 μg/kg rubiadin (anthraquinone) and 3 adult goats were untreated controls. All goats were exposed to sunlight and sequential ear skin biopsies were collected before treatment and at 32 hours, 3 days, 8 days, and 15 days after treatment. Changes at 32 hours after dosing included epidermal spongiosis, single cell death and acantholysis, an increased BAX/BCL-2 protein ratio, and dermal edema. Lesions at day 3 included epidermal and adnexal necrosis, crust formation, and acanthosis. Acanthosis, hyperkeratosis, and dermal fibrosis and neovascularization were present at day 15. The pro-apoptotic (BAX)/anti-apoptotic (BCL-2) protein ratio increased at 32 hours, whereas epidermal and dermal PCNA immunolabeling increased between days 8 and 15 after treatment. The cutaneous lesions were consistent with sunlight-induced damage, and the occurrence in treated but not control goats indicates photosensitization.

The Solanum glaucophyllum Desf. has been used to treat and prevent diseases in human and veterinary medicine. On the other hand, plant poisoning causes several bone diseases, among them osteoporosis, which is characterized by osteoblastic hypoplasia. Because the osteoblast is a cell derived from the differentiation of mesenchymal stem cells (MSCs) from bone marrow, the hypothesis is that the plant reduces the osteogenic differentiation of MSCs. The objective of this study was to evaluate the effects of S. glaucophyllum Desf. extract on MSCs cultured in osteogenic differentiation medium. We determined by liquid chromatography that 1 ml of plant extract contained 3.8 μl of 1,25(OH)2 D3 (calcitriol). Four groups of MSCs cultivated in osteogenic medium were evaluated as follows: (a) treated with 100 μl of extract/L containing 0.4 μg/L of calcitriol; (b) treated with 1 ml of extract/L containing 4 μg/L of calcitriol; (c) treated with 5 ml of extract/L containing 20 μg/L of calcitriol; and (d) a control group without extract. We performed alkaline phosphatase activity assay, analysis of MTT conversion to formazan, and evaluated the percentage of cells, and number and diameter of mineralization nodules. The expression of gene transcripts for osteopontin, bone sialoprotein and BMP-2 was analysed by RT-qPCR. After 21 days, there was a significant reduction in MTT conversion to formazan in treated groups, of the cellularity in the group with 5 ml of extract/L, and in the number and size of mineralization nodules in the groups treated with 1 and 5 ml of extract/L. The 5 ml extract/L concentration also reduced transcript expression of osteopontin. It is concluded that S. glaucophyllum Desf. at concentrations of 1 and 5 ml extract/L reduced mineralized matrix synthesis in MSCs cultivated in osteogenic differentiation medium, which suggests that this is one of the mechanisms by which osteoporosis occurs in intoxicated animals.

It is well known that several of the swainsonine-containing plant species found widespread around the world have a negative economic impact in each country. In Argentina, most of the information on the poisonous plant species that produce α-mannosidosis is published in Spanish and thus not available to most English speaking researchers interested in toxic plants. Therefore, the aim of this review is to summarize the information about swainsonine-containing plants in Argentina, which are extensively distributed throughout different ecoregions of the country. To date, five species from three genera have been shown to induce α-mannosidosis in livestock in Argentina: Ipomoea carnea subsp. fi stulosa, Ipomoea hieronymi subsp. calchaquina (Convolvulaceae), Astragalus garbancillo, Astragalus pehuenches (Fabaceae), and Sida rodrigoi (Malvaceae). These species contain the indolizidine alkaloid swainsonine, which inhibits the lysosomal enzyme α-mannosidase and consequently affects glycoprotein metabolism, resulting in partially metabolized sugars. The prolonged consumption of these poisonous plants produces progressive weight loss and clinical signs related to a nervous disorder, characterized by tremors of head and neck, abnormalities of gait, difficulty in standing, ataxia and wide-based stance. Histological lesions are mainly characterized by vacuolation of different cells, especially neurons of the central nervous system. The main animal model used to study α-mannosidosis is the guinea pig because, when experimentally poisoned, it exhibits many of the characteristics of naturally intoxicated livestock. The lectin histochemical pattern (LHP) was characterized and compared in normal and cystic ovaries of sows. Six biotinylated lectins (PNA, SBA, WGA, RCA-1, DBA and UEA-1) were used on tissue sections. In the normal ovaries, the reaction to UEA-1 and SBA was mild to moderate in mesothelial and endothelial cells. RCA-1 staining was mild to moderate in theca interna of growing follicles, corpora luteum and mesothelium. In addition, this lectin presented strong reaction in endothelial cells, granulosa cells of atretic follicles, zona pellucida of growing follicles and plasma. DBA showed strong intensity in mesothelial and endothelial cells. There was mild to moderate reactivity to WGA in granulosa cells, corpus luteum and theca interna of follicles in development, and moderate in zona pellucida, in granulosa cells of atretic follicles and mesothelium. PNA staining was mild to moderate in oocytes and in the adventitia and media of medullary arteries. Changes in the LHP of the cystic ovaries were noted; however, there were no differences in these findings between the follicular and luteinized cysts. UEA-1 reactivity in the cystic ovaries was moderately reduced in the mesothelial and endothelial cells, whereas there was mild reduction in the DBA staining in the granulosa cells. Reaction to RCA-1 and WGA in the cysts also was decreased in theca interna, zona pellucida and granulosa cells of atretic follicles. Furthermore, endothelium and theca interna in the cystic ovaries presented mild reduction of marcation to SBA, whereas there was decreased reactivity to PNA in the oocytes and adventitia and media layers of the medullary arteries. The results of the current study show that cysts modify the LHP in swine ovaries. These changes of glycoconjugates in many ovarian structures could modify diverse process and may be one of the reasons for decreased fertility in sows.

The aim of this study was to investigate the clinical, biochemical and toxicological findings of the experimentally poisoning induced by Heterophyllaea pustulata in goats. Ten healthy adult female Saanen breed goats were used in the experiment. The goats were randomly assigned to two groups of five individuals: control and experimental group (CG and EG). Both groups were kept in the same enclosure devoid of shade for 8 h daily. The EG received only H. pustulata samples (leaves and thin steam) and water ad libitum. The CG received lucerne hay. Blood samples were taken at different times after oral administration of vegetal samples, and level of hepatic enzymes, total bilirubin, conjugated and non-conjugated bilirubin was measured, together with the detection of anthraquinones (AQs) and phylloerythrin by High Performance Liquid Chromatography with Diode-Array Detector and Mass Spectrometry with Electron Spray Ionization and Quadrupole Time Of Fly analysis. At the same time, skin biopsy samples were collected for AQs determinations. For histopathological examination, hepatic biopsy samples were collected on day 8. Clinically, all goats of the EG revealed photophobia, dermatitis and photosensitization. None of these goats developed jaundice or died during the experiment (15 days). In addition, affected goats exhibited a significant elevation in the serum levels of glutamic oxaloacetic transaminase, direct bilirubin, and total bilirubin. Microscopic examination of the liver samples revealed slight degenerative lesions. Although phylloerythrin was not detected in sera, a high level of two predominant AQs in H. pustulata (rubiadin/soranjidiol) were noted between 24 and 72 h after plant consumption, which coincided with the period in which the clinical signs were more obvious. Since those AQs were not identified in skin samples, the clinical findings were supported by the presence of AQs in sera. Finally, toxicological studies of the AQs are important, since many current works suggest their potential use in the photodynamic therapy.

The aim of this paper was to present the first report of Prosopis nigra poisoning of cattle in Argentina. Outbreaks occurred in five farms located in Salta and Santiago del Estero provinces. All animals were examined, euthanized and necropsied. Clinical signs included tongue protrusion, twitches and tremors of muscles of mastication, weight loss and lethargy. Severe atrophy of the masseter, buccinator and lingual muscles was observed, along with neuronal vacuolation in the nuclei of the trigeminal, facial, and hypoglossus nerves. These findings and the clinical signs are consistent with results obtained in animals, spontaneously and experimentally intoxicated with Prosopis juliflora in previous studies. Several species of this genus are native to Argentina. Farmers should be warned about the suspected toxicity by Prosopis nigra, since this species has wide geographical distribution in the country.

Summary: Strain "LP-3" of equine herpesvirus 1 (EHV-1), isolated from plasma of horses with clinical signs of rhinopneumonitis, was characterised by cultural, physicochemical, electron microscopic, serological and immunohistochemical studies. This was the first isolation from Argentine horses with respiratory disease. Another strain, SP-1, was isolated in the same laboratory from an aborted fetus in 1980.

Enzootic calcinosis (EC), is a chronic disease, caused by toxic plants, which has mainly affected ruminants for more than 100 years. It is characterized by a soft tissue calcification, hypercalcemia, hyperphosphatemia, hypoparathiroidism, hypercalcitoninism, osteonecrosis, and osteopetrosis. The toxic compound reported in some of these plants are 1,25(OH)2D3 glycosides. Characterization of calcinogenic principles were performed with Solanum glaucophyllum, Trisetum flavescens, Cestrum diurnum, and Stenotaphrum secundatum. Other plant species involved in EC are: Nierembergia rivularis, Nierembergia veitchii, Solanum torvum, and Solanum stuckertii. Clinical and pathological findings are well characterized. We present different aspects of the pathogenic mechanism of EC, including genetic modulation, expression of bone proteins, cellular de-differentiation of arterial smooth muscle cells, phenotypic changes, production of a calcifiable matrix, and modulation of the calcium and phosphorus balance. The use of comparative pathology can bring significant advances in the understanding of the pathogenic mechanism. In this review we present a broad approach to the different aspects of intoxication: history of EC on different continents; plant species, distribution and morphology; toxic principle; pathogenesis; epidemiology; clinical signs; diagnosis; and control and prophylaxis.

Lysosomal storage diseases are inherited and acquired disorders characterized by dysfunctional lysosomes. Intracytoplasmic accumulation of undegraded substrates leads to impaired cellular function and death. Several plant species are toxic to livestock because of the presence of indolizidine alkaloids, including swainsonine, which cause a storage disease. Swainsonine-induced nervous disease (i.e., locoism) of sheep and cattle is well recognized in several parts of the world, particularly in the western United States and in parts of Australia. Spontaneous intoxication by Astragalus garbancillo var. garbancillo was suspected in a group of 70 llamas (Lama glama) in Jujuy Province, northwestern Argentina. The animals grazed an area dominated by stands of A. garbancillo var. garbancillo. Clinical signs were staggering, ataxia, hypermetria, and progressive weight loss. The clinical course in individual animals was ~50 d. The main microscopic changes were Purkinje cell degeneration, necrosis, and loss, associated with intracytoplasmic vacuolation, meganeurite formation, and Wallerian degeneration. Specific positive labeling for ubiquitin was observed in axonal spheroids. Composite leaf and stem samples of A. garbancillo var. garbancillo analyzed by high-performance liquid chromatography contained 0.03% swainsonine. Based on the microscopic lesions, clinical history, and plant analysis, a diagnosis was made of storage disease caused by consumption of swainsonine-containing A. garbancillo var. garbancillo. In Argentina there is little information about diseases that affect exotic ungulates and the health risks that they pose to native wildlife, livestock, and humans. The aim of this study was to evaluate the health status of red deer ( Cervus elaphus) in the Nahuel Huapi National Park and surrounding areas in Patagonia, Argentina. During three consecutive hunting seasons, necropsies were performed on 101 red deer, and tissues were examined histologically. The most common lesions were those associated with hepatic and pulmonary parasites. Fasciola hepatica was observed in 15 red deer and was associated with cholangiohepatitis (8%) and/or cholangitis (10%). Dictyocaulus sp. (likely Dictyocaulus eckerti) was associated with interstitial pneumonia (5%), bronchitis (5%), pulmonary emphysema (13%), and bronchus-associated lymphoid tissue hyperplasia (13%). Other findings included Sarcocystis spp. cysts in the myocardium (89%) associated with interstitial, focal, lymphoplasmacytic myocarditis (8%); periportal lymphoplasmacytic hepatitis (8%); hepatic centrilobular necrosis (6%); lymphoplasmacytic interstitial nephritis (25%); and follicular hyperplasia in mediastinal, prescapular, and prefemoral lymph nodes (86%). Our report of lesions caused by endemic parasites of livestock in free-ranging exotic red deer in Patagonia sets the foundation for a health-monitoring and -surveillance system of wildlife in this region, which is essential for the sustainable management of threatened Argentinean native fauna.

Los sarcomas asociados a sitios de inoculación son neoplasias de origen mesenquimático que aparecen en las regiones corporales utilizadas rutinariamente para la aplicación de vacunas u otros inóculos. Con el objetivo de caracterizar estas neoplasias se seleccionaron 117 tumores diagnosticados como sarcomas posinoculación. Se realizaron estudios histopatológicos e inmunohistoquímicos que permitieron reclasificar los diferentes sarcomas hallados y establecer el porcentaje de células proliferantes. Además, se consideraron los datos demográficos y se realizaron encuestas a los médicos veterinarios para conocer la evolución de los pacientes. El diagnóstico más frecuente fue el de fibrosarcoma (87,18%). También, se encontraron, en menor proporción, otros tumores como: sarcomas pleomórficos (HFM), osteosarcomas extraesqueléticos y mixosarcomas. El grado histológico más frecuente fue el II y la mayoría presentó abundante inflamación. El promedio de células proliferantes alcanzó el 50,12%. La edad de los pacientes al momento del diagnóstico fue mayor que la informada por otros autores. Si bien los sarcomas asociados a sitios de inoculación en la República Argentina comparten características histopatológicas y clínicas con aquellos informados en otros países, existen diferencias vinculadas a la edad al momento del diagnóstico, a la presentación de las recidivas y al tiempo de sobrevida, entre otras características., Feline injectionsite sarcomas are neoplasms of mesenchymal origin frequently found in sites of the body routinely used for inoculation of vaccines and other drugs. With the aim of characterizing these neoplasms, 117 postinoculation sarcomas were selected and analyzed. Histopathological and immunohistochemical techniques were performed to reclassify different sarcomas and determine cellular proliferation percentages. In addition, demographic data were considered, and surveys were conducted to obtain information about the clinical followup of patients. Of all tumors analyzed, fibrosarcoma was the most frequently diagnosed (87.18%), followed by pleomorphic sarcomas (HFM), extra skeletal osteosarcomas and mixosarcomas. The most frequent histological grade was type II which was associated with great inflammation. Cell proliferation percentage of these tumors was 50.12%. The average age of diagnosis for these sarcomas was higher than that reported by other authors. Solanum glaucophyllum, a toxic plant known for its calcinogenic effects, causes enzootic calcinosis in ruminant and monogastric animals. We describe an outbreak of enzootic calcinosis that occurred in a herd of 110 horses grazing pastureland heavily contaminated with S. glaucophyllum in Buenos Aires province, Argentina. Ten horses developed clinical signs, and 6 horses died. Clinical signs included abnormal gait (stiff-legged action, short strides), stiffness, thoracolumbar kyphosis, reluctance to move, wide stance, chronic weight loss, weakness, recumbency, and difficulty standing. Autopsy of 2 horses revealed severe mineralization of the aorta, pulmonary arteries, heart, and lungs, consistent with enzootic calcinosis. Although horses usually have very selective grazing behavior, under food restriction conditions, they can ingest the toxic plants and can develop the disease. Bovine enzootic haematuria (BEH) is caused by prolonged ingestion of toxic principles of bracken fern, essentially by Pteridium spp. In northwestern Argentina, this disease has a great economic impact ant it is attributed a chronic consumption to Pteridium arachnoideum. This paper describes two endemic areas for enzootic hematuria due to the consumption of Pteris deflexa and Pteris plumula. Two areas where P. deflexa and P. plumula are endemic, but free of Pteridium species, were devised and seven farms were visited. The disease was confirmed based on the presence of clinically affected animals. In four necropsies bleeding neoplastic lesions were observed in the mucosa of the urinary bladder. At phytochemical analysis, both ptaquiloside and pterosin B were found in P. deflexa and P. plumula. Thus, the consumption of P. deflexa and P. plumula can also cause BEH.

Hypertrophic osteopathy is commonly associated with primary lung neoplasms. It may also be secondary to intrathoracic lesions such as pulmonary metastasis, granulomatous pleurisy and chronic pneumonia, among others. It is characterized by progressive proliferation of sub periosteal bone tissue in limb bones. Clinically, patient usually exhibits lameness. It is a rare disease most frequently observed in dogs among domestic animals. A case of a dog with pulmonary hypertrophic osteopathy associated with pulmonary metastases coming from an appendicular periosteal osteosarcoma is reported., Facultad de Ciencias Veterinarias

There are numerous poisonous plants that can induce intralysosomal accumulation of glycoproteins and neurologic syndromes. Here we describe for the first time, a disease caused by ingesting Sida rodrigoi Monteiro in goats in North-western Argentina. The animals showed weight loss, indifference to the environment, unsteady gait and ataxia. Histopathologic studies showed vacuolization in cells of various organs, mainly in the CNS. The material deposited in the cells was positive for LCA (Lens culinaris agglutinin), WGA (Triticum vulgaris agglutinin), sWGA (succinyl-Triticum vulgaris agglutinin) and Con-A (Concanavalia ensiformis agglutinin) lectins. Finally, toxic levels of swansonine were identified in the plant. The present investigation allowed to recognize S. rodrigoi Monteiro poisoning as a plant induced α-mannosidosis. This study describes the changes observed in the placentas of mice experimentally infected with an abortigenic strain of EHV-1 at mid-pregnancy and euthanized at days 3 and 4 post-infection. We analyzed microscopic vascular alterations, cell proliferation and death by immunohistochemistry, and the expression of IFN-γ, TNF-α and the IL-10 by qPCR and flow cytometry. Infected mice showed slight respiratory signs and ruffled fur during the first two days post-infection. Virus isolation and DNA detection were positive only in the lungs of the infected mice. Vascular congestion, increase in the labyrinth area, and a significant reduction in fetal capillary endothelium surface of infected placentas were found. Cell proliferation was significantly reduced in the infected placentas, whereas the apoptosis was significantly increased. IL10, TNF and IFN-γ showed different expression in the infected placentas and uteri. The effects of EHV-1 during pregnancy depend on different pathogenic mechanisms in which vascular alterations, and cell death and proliferation and local cytokine changes are compromised. Commerson’s dolphins Cephalorhynchus c. commersonii are the small cetacean most frequently found stranded along the coast of Tierra del Fuego, Argentina, but little is known about their pathologies. We examined the postcranial axial skeleton of 425 Commerson’s dolphins collected in the period 1974−2011 for the presence of pathologies. Miscellaneous lesions were detected in 107 (25.2%) of the specimens. Among them, idiopathic hyperostosis was the most frequent pathology (73.8%), followed by spondyloarthropathy (40.2%). Traumata (25.2%), spondylitis (10.3%) and osteoarthritis (11.2%) occurred less frequently. The type of lesion was statistically associated with the region of the vertebral column. Idiopathic hyperostosis occurred significantly more frequently in the lumbar vertebrae than in other parts of the vertebral column, while spondyloarthropathy was more frequent in the thoracic and caudal vertebrae. Both conditions were predominantly seen in mature dolphins. Osteoarthritis affected the transverse processes of the thoracic vertebrae of 6 males, 3 females and 3 individuals of undetermined sex, all mature. Healed and unhealed fractures were observed in the ribs of 27 specimens and in the transverse processes of 3 others. Spondylitis of various degrees of severity affected the cervical, thoracic, lumbar and caudal regions of 11 individuals. Together these data indicate that bone lesions and traumata commonly occur in Commerson’s dolphins, likely causing pain and morbidity in severe cases. Fil: San Martín, Analía Anahí. The hypothesis of this experiment is that mesenchymal stem cells (MSCs) are involved in the genesis of the bone metaplasia caused by Solanum glaucophyllum intoxication. We determined using liquid chromatography that 1 mL of plant extract contained 3.8 μl of 1,25(OH)2D3. The ability of 100 μL, 1 mL and 5 mL of extract/L, containing 1 nM (0.4 μg/L), 10 nM (4 μg/L) and 50 nM (20 μg/L) of 1,25(OH)2D3, respectively, in inducing the osteogenic differentiation in bone marrow MSCs from rats was tested. At the concentrations of 1 and 5 mL of extract/L of culture medium without osteogenesis-inducing factors, the plant extract induced the osteogenic differentiation of the MSCs, as was evidenced by the greater synthesis of mineralized matrix. At the higher concentration (5 mL of extract/L), an increase in the relative expression of BMP-2 gene was observed. It was concluded that rat bone marrow MSC culture is a good model for studying the effects of the S. glaucophyllum extract on the osteogenic differentiation of undifferentiated cells. Also, S. glaucophyllum extracts containing 10 nM (4 μg/L) and 50 nM (20 μg/L) of 1,25(OH)2D3 induce the osteogenic differentiation of MSCs, suggesting that this is one of the mechanisms by which S. glaucophyllum causes bone metaplasia.

Anthelmintic resistance (AR) of gastrointestinal nematodes to macrocyclic lactones is an increasingly common worldwide phenomenon limiting cattle production. This has motivated the search for alternatives, such as new active compounds, added drug synergisms, different doses, and alternate administration routes. The aim of this study was the assessment of moxidectin (MXD) performance in feedlot calves with a history of AR to ivermectin (IVM). Crossbred female calves aged 6-7 months and weighing 163 kg (SD = 34 kg) were divided into 3 groups of 35 animals each. They were assigned to the following antiparasitic treatment groups: IVM group (0.2 mg/kg IVM); MXD group (0.2 mg/kg MXD), and ricobendazole + levamisole (RBZ + LEV) group (7.5 mg/kg RBZ + 8 mg/kg LEV). On days 0, 26, and 47, fecal samples were taken and the weight of each animal was registered. Anthelmintic efficacy (by fecal egg count reduction), total weight gain (TWG) and average daily weight gain (AWG) were compared between the groups. A mixed SAS procedure was used for statistical analysis. Fecal egg count reduction 26 days post-treatment (PT) was calculated at 28% for the IVM group, 85% for the MXD group, and 99% for the RBZ + LEV group. AWGs (Standard Error) of 1.095 g (56), 1.264 g (49), and 1.340 g (52) were registered for the IVM, MXD, and RBZ + LEV groups, respectively (p < 0.05). Coprocultures revealed that MXD more effectively reduced Haemonchus spp. and Cooperia spp. egg counts than IVM. This resulted in higher AWGs and TWGs for this group; similar results were seen for the RBZ + LEV group as well. In this study, animals treated with MXD gained about 160 more g/day than animals treated with IVM. This represents a gain of 16 USD per animal over the 47 day trial. Fil: Fazzio, Luis Emilio. Feline injection site sarcomas (FISSs) are mesenchymal neoplasms that develop at the sites of delivery of vaccines or other injectable products. Vaccine adjuvants can trigger an intense and persistent inflammatory response that may lead to neoplastic transformation. The proinflammatory role of cyclo-oxygenase (COX)-2 is well known and its overexpression has prognostic value in multiple neoplastic processes. One hundred and seventeen FISSs were evaluated for the degree of inflammation and anaplasia. Immunohistochemistry was used to determine the expression of COX-2 in these sarcomas. There was a significant association between the degree of inflammation and the expression of COX-2 by neoplastic cells. COX-2 expression was lower in tumours with higher degrees of anaplasia. These findings may be useful in predicting the sensitivity of FISSs to treatment with COX-2 inhibitors. The potential therapeutic use of such agents could then be restricted to tumours with lower degrees of anaplasia.

Equid alphaherpesvirus 1 (EHV-1) infection causes abortion, respiratory disease, perinatal deaths and neurological disorders in horses. The natural infection and available vaccines provide only partial and short-lived protection against reinfections. In the present study, we analyzed the ability of purified baculovirus-expressed glycoprotein D (gD) administered by different routes to induce protective immunity in BALB/c mice after challenge with the EHV-1 AR8 strain. Clinical signs varied among the different groups of mice immunized by parenteral routes, and, although gD induced a specific serum IgG response, it did not prevent the virus from reaching the lungs. Intranasally immunized mice showed no clinical signs, and virus isolation from lungs, histological lesions and antigen detection by immunohistochemistry were negative. In addition, by this route, gD did not stimulate the production of serum IgG and IgA. However, a specific IgA response in the respiratory tract was confirmed in intranasally immunized mice. Thus, we conclude that the mucosal immune response could reduce the initial viral attachment and prevent the virus from reaching the lungs. Our findings provide additional data to further study new immunization strategies in the natural host., La infección con alfaherpesvirus equino 1 (EHV-1) causa abortos, enfermedad res-piratoria, muertes perinatales y desórdenes neurológicos en equinos. La infección natural ylas vacunas disponibles solo proporcionan protección parcial y de corta duración contra lasreinfecciones. En el presente estudio se analizó la inducción de inmunidad protectiva de laglicoproteína D (gD) expresada en baculovirus y purificada al ser administrada por diferentesrutas en ratones BALB/c desafiados con la cepa AR8 de EHV-1. Los signos clínicos fueron varia-bles entre los grupos de ratones inmunizados por rutas parenterales y, aunque la gD indujorespuesta específica de IgG en suero, no logró prevenir la llegada del virus al pulmón. En losratones inmunizados intranasalmente no se observaron signos clínicos ni lesiones histopatológi-cas, y el aislamiento viral y la detección de antígenos por inmunohistoquímica en pulmón fueronnegativos. Además, por esta ruta la gD no estimuló la producción de IgG y de IgA en suero. Sinembargo se confirmó la respuesta de IgA específica en el tracto respiratorio de ratones inmu-nizados intranasalmente. Esta respuesta inmune mucosal podría haber reducido la unión inicialdel virus a la célula huésped y, de este modo, prevenir la llegada del virus al pulmón. Many viruses alter different stages of apoptosis of infected cells as a strategy for successful infection. Few studies have addressed mechanisms of equine herpesvirus 1 (EHV-1) strain-induced cell death. We investigated the effect of an abortigenic strain (AR8 strain) on heterologous Madin–Darby bovine kidney cells and homologous equine dermis (ED) cells cell lines. We compared morphologic and biochemical features of early and late apoptosis at different postinfection times. We investigated translocation of phosphatidylserine to the cell surface, nuclear fragmentation and changes in the cytoskeleton using flow cytometry and annexin V/propidium iodide staining, DNA laddering, terminal deoxynucleotidyl transferase UTP nick-end labeling assay and immunofluorescence staining of cytokeratin 18 cleavage. AR8 EVH-1 strain interfered with apoptosis in both cell lines, particularly during the middle stage of the replication cycle; this was more evident in ED cells. Intoxication with Solanum bonariense in cattle causes cerebellar cortical degeneration with perikaryal vacuolation, axonal swelling, and death primarily of Purkinje cells, with accumulation of electron-dense residual storage bodies in membrane-bound vesicles. The pathogenesis of this disease is not fully understood. Previously, we proposed that inhibition of protein synthesis in Purkinje cells among other altered metabolic pathways could lead to cytoskeletal alterations, subsequently altering cell-specific axonal transport. In the present study, immunohistochemical and histochemical methods were used to identify neuronal cytoskeletal alterations and axonal loss, demyelination, and astrogliosis in the cerebellum of intoxicated bovines. Samples of cerebellum from 3 natural and 4 experimental cases and 2 control bovines were studied. Immunoreactivity against neurofilament (NF)-200KDa confirmed marked loss of Purkinje neurons, and phospho-NF protein, β-tubulin, and affinity reaction against phalloidin revealed an altered perikaryal distribution of neuronal cytoskeletal proteins in the remaining Purkinje cells in intoxicated cattle. Reactive astrogliosis in every layer of the cerebellar cortex was also observed with anti–glial fibrillary acidic protein immunohistochemistry. In affected cattle, demyelination and axonal loss in the cerebellar white matter, as well as basket cell loss were demonstrated with Klüver–Barrera and Bielschowsky stains, respectively. Based on these results, we propose that neuronal cytoskeletal alterations with subsequent interference of the axonal transport in Purkinje cells may play a relevant role in the pathogenesis of this neurodegenerative disorder, and also that demyelination and axonal loss in the cerebellar white matter, as well as astrogliosis in the gray matter, likely occur secondarily to Purkinje cell degeneration and death., Facultad de Ciencias Veterinarias

The expression of growth factors was evaluated immunohistochemically in normal and cystic ovaries of sows. The immunohistochemically stained area (IHCSA) was quantified by image analysis to analyse the expression of these proteins in the follicular wall of secondary, tertiary and cystic follicles. IGF-I immunoreactivity was strong in the granulosa cell layer (GC), moderate in the theca interna (TI) and mild in the theca externa (TE) of the normal follicles. There was severe reduction of the labelling to IGF-I in the GC of the follicular and luteinized cysts. In the normal follicles, the reactivity for IGF-II was very similar to pattern noted in IGF-I. There was reduction of the IHCSAs in the GC of the follicular and luteinized cysts, but the decrease was not significant. The staining of the IGF-II in the TI and TE of the cysts was increased, in comparison with normal follicles. The IHCSAs for VEGF were higher in the GC and TE of the normal follicles in contrast to TI, but this difference was noted only in the tertiary follicle. The VEGF reactivity increased in the GC of the cysts, in relation to normal follicles. The results of the current study show that the formation of ovarian cysts in sows is associated with alterations in the immunohistochemical expression of some growth factors.

The expression of cytoskeletal proteins was evaluated immunohistochemically in 36 normal ovaries sampled from 18 sows and 44 cystic ovaries sampled from of 22 sows, was evaluated. All sows had history of reproductive problems, such as infertility or subfertility. The immunohistochemically stained area (IHCSA) was quantified through image analysis to evaluate the expression of these proteins in the follicular wall of secondary, tertiary, and cystic follicles. Cytokeratins (CK) immunoreactivity was strong in the granulosa cell layer (GC) and mild in the theca interna (TI) and externa (TE) of the normal follicles. There was severe reduction of the reaction to CK in the GC in the cystic follicles, mainly in the luteinized cysts. The immunoreactivity for vimentin was higher in the GC from normal and cystic follicles in contrast with the other follicular structures. In the luteinized cysts, the IHCSA for vimentin was significantly higher in TI and in both observed cysts, the labeling was more accentuated in TE. Immunohistochemical detection of desmin and α-SMA was restricted to the TE, without differences between the normal and cystic follicles. The results of the current study show that the development of ovarian cysts in sows is associated to changes in the expression of the cytoskeletal proteins CK and vimentin. A expressão de proteínas do citoesqueleto foi avaliada por imuno-histoquímica em ovários normais e císticos de porcas matrizes. Amostras de 36 ovários normais (18 porcas) e de 44 císticos (22 porcas) foram avaliadas. Todas as matrizes apresentaram histórico de problemas reprodutivos, como infertilidade ou subfertilidade. As áreas coradas por imuno-histoquímica (IHCSA) foram quantificadas por avaliação de imagens avaliando a expressão dessas proteínas na parede folicular de folículos secundários, terciários e císticos. A imuno-reatividade para citoqueratina (CK) foi forte na camada de células da granulosa (GC) e discreta nas tecas interna (TI) e externa (TE) dos folículos normais. Houve redução acentuada da reação de CK na CG dos folículos císticos, principalmente nos cistos luteinizados. A reação para vimentina foi mais intensa na CG dos folículos normais e císticos em comparação com outras estruturas foliculares. Nos cistos luteinizados, a IHCSA para vimentina foi significativamente maior na TI e, em ambos os cistos observados, a marcação foi mais acentuada na TE. A marcação de desmina e actina alfa de músculo liso foi restrita a TE, sem diferenças entre os folículos normais e císticos. Os resultados do presente estudo mostram que o desenvolvimento de cistos ovarianos em porcas matrizes está associado a alterações na expressão das proteínas do citoesqueleto CK e vimentina. This study describes an occurrence of pink morning glory (Ipomoea carnea) intoxication in goats in northern Argentina. The clinical signs displayed by the affected animals were ataxia, lethargy, emaciation, hypertonia of the neck muscles, spastic paresis in the hind legs, abnormal postural reactions and death. The clinico-pathologic examination revealed that the affected animals were anemic and their serum level of aspartate aminotransferase was significantly increased. Cytoplasmic vacuolation in the Purkinje cells and pancreatic acinar cells was observed by histological examination. The neuronal lectin binding pattern showed a strong positive reaction to WGA (Triticum vulgaris), sWGA (succinylated T. vulgaris) and LCA (Lens culinaris). Although I. carnea is common in tropical regions, this is the first report of spontaneous poisoning in goats in Argentina.

The aim of this study was to evaluate, in a commercial feedlot, the effect of different anthelmintic drugs on the productivity of naturally infected calves from a cow-calf operation, where resistance to ivermectin (IVM) has been previously detected. The study began with the random selection of 80 calves whose weight was 132 ± 12. kg. Four groups were made: IVM, ricobendazole (RBZ), ricobendazole plus levamisol (RBZ + LEV) and a control group (CG) without treatment. On days 0, 21, 42, 70, 98 and 126, manual collection of fecal matter and individual weight were registered. Mixed SAS procedure was used for statistical analysis. The percentages of fecal egg count reduction test (FECRT) calculated 21days post treatment (PT) were 18%, 96% and 100% for the IVM, RBZ and RBZ + LEV groups, respectively. Body weight (± SEM) at the end of the trial was 266. kg (±0.9), 269. kg (±1.1), 276. kg (±1.3), 280. kg (±1.9) for CG, IVM, RBZ and RBZ + LEV groups, respectively. The effect on live weight was highly significant (p < 0001). After 126 days of fattening, the deleterious effect of the combination of Cooperia and Haemonchus in the IVM group on body weight was evident. "Mascadera" is a chronic emaciating neuropathy affecting goats; it produces significant economic losses in many regions and its cause is unknown. Here, the histological lesions found in 15 animals naturally affected by the disease are described. Complete necropsy was performed and tissue samples were collected for histopathological study. Severe atrophy of the masseter and buccinator muscles and tongue was observed, as well as vacuolar degeneration of neurons in the nuclei of the trigeminal, facial, and glossopharyngeal nerves. No relevant lesions were observed in other tissues. These findings and the clinical signs are consistent with those observed by other authors in animals spontaneously and experimentally intoxicated with Prosopis juliflora. The disease may be due to consumption of a similar species present in our country that is still unknown. Heterophyllaea pustulata Hook. f. (Rubiaceae) is a phototoxic plant. It grows in the Andean area of northwest of Argentina, and it causes significant economic losses in the livestock. This plant induces dermal lesions by photosensitization probably due to its content of photosensitizing anthraquinones. This paper describes an outbreak of poisoning in Corriedale sheepfold, which had an incidence of 49%. Ear skin biopsies and blood samples were collected of six affected animals. Liver enzymes remained within the reference limits. Histopathologically, a deep necrotizing dermatitis was identified in all samples. H. pustulata was identified in the areas of grazing. Anthraquinone concentration in leaves was 0.84% p/p, expressed as rubiadin. All findings allow us to conclude that the diagnosis is a primary photosensitization. The aim of this study was to characterize cytoskeletal intermediate filament proteins and glycoconjugates of syncytiotrophoblast, cytotrophoblast and decidual cells of feline endotheliochorial placenta. Samples from 12 normal pregnant female cats, after 45 ± 5 days of gestation, were obtained removing the uterine horns by hysterectomy. Sections were processed for routine observation and for immunohistochemistry using anticytokeratin, antivimentin and antidesmin antibodies. In addition, lectin histochemistry was performed using a panel of several biotinylated lectins to characterize glycosides expression profile. Cytotrophoblast and syncytiotrophoblast showed immunoreactivity only with acidic and basic cytokeratins. Decidual cells were only positive to vimentin, consistent with their origin from endometrial fibroblasts. Trophoblast expressed a broad population of glycans, highly exposing terminal N-acetyl glucosamine residues and non-sialylated galactose and N-acetyl galactosamine oligomers. Oligosaccharides bound by Phaseolus vulgaris erythroagglutinin were the only highly branched N-linked residues evidenced in cats, and they were restricted to the syncytium. Unlike results reported on humans, mice and rats on lectin affinity of decidual cells, sialid acids and complex N-linked oligosaccharides were not demonstrated in cats. Glycosylation of proteins determines many of their final properties, thus becoming essential for the embryo-maternal dialogue during implantation and placentation. Changes in glycosylation pattern have been related to pathological pregnancies in other species. Hence, the knowledge about glycosylation profile of the normal cat placenta may lead to a better understanding of both normal and pathological reproductive events. Fil: Fernández, Patricia Elena. Objective: This study was conducted to evaluate, by means of lectinhistochemistry (LHC), the expression of carbohydrates in granulomas induced by the bacillus Calmette-Guerin (BCG) in muscle tissue of Piaractus mesopotamicus after 33 days. Material and methods: Histological sections with 3 µm thick were incubated with the following lectins: WGA (Wheat germ agglutinin), DBA (Dolichos biflorus agglutinin) and HPA (Helix pomatia agglutinin), and the results were evaluated by light microscopy. Results: Acid fast bacilli were stained by Ziehl Neelsen (ZN) and strong labeled by WGA in the cytoplasm of macrophages. Labeling with DBA was intense in fibroblasts and weak in macrophages. On the other hand, HPA binding was stronger in macrophages, especially in those that were in close contact with epithelioid cells, without evidence of binding to fibroblasts. The epithelioid cells were not labeled by the used lectins, but they were identified by Hematoxilin-Eosin (HE). The lectins labeled specific type saccharides in glycoproteins, as N-acetylglucosamine present in bacilli and macrophages, as well as N-acetyl-galactosamine in macrophages. The control group showed no inflammation or lectin binding. Conclusions. This technique may be useful in identifying receptors for WGA, DBA and the HPA lectins in epithelioid granuloma induced by BCG in P. mesopotamicus., Objetivo: El presente estudio fue realizado para evaluar por medio de lectinhistoquímica (LHC), la expresión de carbohidratos en granulomas inducidos por el bacilo de Calmette-Guérin (BCG) en músculo de Piaractus mesopotamicus después de 33 días. El marcaje con DBA fue intenso en fibroblastos y débil en macrófagos. Con la lectina HPA el marcaje fue intenso en macrófagos, principalmente en los que estaban en estrecho contacto con las células epitelióides, externamente se observó marcaje débil en fibroblastos. Las células epitelióides no fueron marcadas por las lectinas, pero fueron identificadas con la tinción de Hematoxilina-Eosina (HE). Las lectinas tuvieron un tipo de marcaje específico en algunos monosacáridos, como N-acetilglucosamina presente en los bacilos y en macrófagos, y N-acetilgalactosamina en macrófagos. En el grupo control no fue observada inflamación así como tampoco marcaje con las lectinas. Conclusiones. Bovine genital tritrichomonosis is caused by the protozoon Tritrichomonas foetus and leads to embryonic death and abortion. The complexity of handling bovine experimental systems has led to the development of alternative models. The infection has been reproduced in pregnant BALB/c mice. In the pathogenesis of the disease, adhesion of the protozoon to host cell surface glycoproteins is important. Labelling with soya bean agglutinin (SBA) and peanut agglutinin (PNA) lectins increases in the luminal and glandular uterine epithelium of non-pregnant infected mice. The aim of the present study was to determine whether these changes also occur in pregnant infected BALB/c mice. Female BALB/c mice were inoculated intravaginally with T. foetus and, 15±3days post infection, were paired with males overnight. Infected and control mice were sacrificed 6, 8 and 10days later. Samples of uterus were labelled with a panel of biotinylated lectins. Infected mice showed increased binding of PNA and SBA. There was also increased binding of concanavalin (Con-A) by luminal epithelium and Ricinus communis agglutinin (RCA-1) by glandular epithelium at day 6 post coitum. These changes may be due to the production of enzymes by T. foetus, which could act to enhance adhesion and colonization and thus favour infection. Fil: Woudwyk, Mariana Andrea. Here, we used a murine model to describe and compare the pathogenic potential of the Argentinean equid herpesvirus 1 (EHV-1) AR8 strain with the Japanese HH1 reference strain. In AR8-inoculated animals, clinical signs began earlier, but the viremic phase was shorter. Virus isolation and DNA detection in the lungs, liver and spleen were positive for both strains at different times postinfection (pi). Infection foci produced by both strains were immunohistochemically detected in lungs from day 1 to day 4 pi. The uropygial glands of birds are sebaceous organs that contribute to the water-repellent properties of the feather coat. We studied the histological and histochemical characteristics of the uropygial gland of chimango caracara using hematoxylin and eosin (H & E), Gomorís trichrome, orcein, Gomorís reticulin, periodic acid-Schiff (PAS), Alcian blue (AB) and a variety of lectins. The gland is composed of two lobes and a papilla with 20 downy feathers. It is surrounded by a capsule of dense connective tissue that contains elastic, reticular and smooth muscle fibers. The papilla is delicate and has two excretory ducts. The gland mass relative to body mass was 0.143%. Both adenomer cells and their secretions were stained with Sudan IV, PAS and AB, and were positive for numerous lectins that indicated the presence of lipids and carbohydrates. Immunohistochemical techniques to detect PCNA confirmed cell proliferation in the basal stratum of the adenomer cells. The aim of this study was to describe the effects of a single dose of the gonadotrophin releasing hormone (GnRH) antagonist acyline on testicular characteristics of the domestic cat. Twelve mature cats were orchidectomised unilaterally (right testis) on Day -7 (n=7) or Day 15 (n=5). On Day 0, 330 μg/kg acyline was administered s.c. to all the animals. Ipomoea carnea subsp. fistulosa, aguapei or mandiyura, is responsible for lysosomal storage in goats. The shrub contains several alkaloids, mainly swansonine which inhibits lysosomal α-mannosidase and Golgi mannosidase II. Poisoning occurs by inhibition of these hydrolases. There is neuronal vacuolation, endocrine dysfunction, cardiovascular and gastrointestinal injury, and immune disorders. Clinical signs and pathology of the experimental poisoning of goats by Ipomoea carnea in Argentina are here described. Five goats received fresh leaves and stems of Ipomoea. At the beginning, the goats did not consume the plant, but later, it was preferred over any other forage. High dose induced rapid intoxication, whereas with low doses, the course of the toxicosis was more protracted. The goats were euthanized when they were recumbent. Cerebrum, cerebellum, medulla oblongata, pons and colliculi, were routinely processed for histology. In nine days, the following clinical signs developed: abnormal fascies, dilated nostrils and abnormal postures of the head, cephalic tremors and nystagmus, difficulty in standing. Subsequently, the goats had a tendency to fall, always to the left, with spastic convulsions. There was lack in coordination of voluntary movements due to Purkinje and deep nuclei neurons damage. The cochlear reflex originated hyperreflexia, abnormal posture, head movements and tremors. The withdrawal reflex produced flexor muscles hypersensitivity at the four legs, later depression and stupor. Abnormal responses to sounds were related to collicular lesions. Thalamic damage altered the withdrawal reflex, showing incomplete reaction. The observed cervical hair bristling was attributed to a thalamic regulated nociceptive response. Depression may be associated with agonists of lysergic acid contained in Ipomoea. These clinical signs were correlated with lesions in different parts of the CNS., Facultad de Ciencias Veterinarias

Contents Bovine campylobacteriosis (BC) is a venereal disease caused by Campylobacter fetus characterized by temporary infertility with mild endometritis, early embryonic death and occasional abortions. The objectives of this study were to describe and identify C. fetus in spontaneous bovine abortion on the basis of histopathological, immunohistochemical, lectinhistochemical and polymerase chain reaction (PCR) techniques. The most frequent foetal lesion was neutrophilic bronchopneumonia and interstitial pneumonia. Other commonly observed lesions included non-suppurative interstitial enteritis, hepatitis, pericarditis, myositis, myocarditis, and meningitis. In this study, C. fetus fetus was phenotypically classified in all bovine foetuses from lungs and abomasal fluids. Immunohistochemistry (IHC) staining revealed positive stained Campylobacter organisms with typical morphology. Lectin binding patterns not showed great differences between the infected and the non-infected groups. The most important changes were a minor peanut agglutinin (PNA) and DBA binding in the alveolar cells of the lungs and DBA globet cells in some of the C. fetus–positive foetuses. Individual variations in each lectin binding pattern complicate the evaluation of the lectins results. All foetuses positive to IHC were positive by PCR. Better efficiency of PCR was obtained from abomasal fluids than from lung tissues. The association of culture and phenotypic techniques with histopathology, IHC and PCR allowed a better characterization and description of BC.

A study was conducted to evaluate the predictive diagnostic value of different copper (Cu) parameters as indicators of average daily gain (ADG) in growing calves. The effects in calves of cow Cu supplementation in the last one-third gestation period were also evaluated. Five supplementation trials, with a total of 300 calves, were carried out. Two groups of 30 calves were randomly assigned to each trial, one group was parenterally supplemented (SG) and the other was not supplemented (NSG). Trials began when calves were three-month-old and ended at weaning time. At each sampling calves were weighed and blood was taken to determine Cu concentrations in plasma, Whole Blood (WB), Red Cells (RC) and Packed Cell Volume (PCV). Liver samples from six animals of each group were taken both at the beginning and at the end of the trial. In two trials the mothers of the SG received Cu supplementation at the last one- third gestation period. Four of the five trials exhibited low ADG in the NSGs. In these groups, plasma Cu concentration decreased rapidly before low ADG was detected, which occurred with values remaining below 25μg/dl. The decrease of RC Cu concentration was considerably slow. WB showed an intermediate position. PCV in the SGs was higher than in the NSGs in all trials. Cow supplementation was insufficient to generate a liver storage able to last after calves reached the 3 months of age. Alterations in the ovarian micro-environment of females with follicular cysts could alter the normal processes of proliferation and programmed cell death in ovarian cells. Thus, the objective in the present study was to evaluate apoptosis and proliferation in induced ovarian cystic follicles in cows to investigate the follicular persistence. Solanum bonariense intoxication is characterized by cerebellar neuronal vacuolation, degeneration, and necrosis. Cerebellar Purkinje cells seem especially susceptible, but more research is needed to determine the pathogenesis of neuronal necrosis and the mechanism of Purkinje cell susceptibility. Calbindin D28k (CbD28k) is highly expressed in Purkinje cells and has been used as a marker for normal and degenerative Purkinje cells. The goal of this study was to describe S bonariense-induced disease by ascertaining Purkinje cell-specific degenerative changes using CbD28k expression and to correlate this with apoptosis in Purkinje cells, as determined using TUNEL (transferase-mediated dUTP-biotin nick end-labeling) and ultrastructural changes. In all cases, an increase in both dose and duration of S bonariense intoxication resulted in a decrease in the number of Purkinje cells. CbD28k immunohistochemistry was an excellent marker for Purkinje cells because immunoreactivity did not change in normal or degenerative tissues. This finding suggests that excessive calcium excitatory stimulation does not induce rapid neuronal degeneration and death. As found in previous studies, TUNEL tests and electron microscopy suggest that Purkinje cell degeneration and death are not occurring via an apoptotic process. Lagostomus maximus is a South American Hystricognathi rodent whose reproductive behaviour shows characteristics unusual for mammals, such as polyovulation (200–800 oocytes) and a high rate of embryo mortality. Thirty-six mature females captured in the province of Buenos Aires showed different physiological reproductive stages. Most of them presented a postpartum oestrus in August–September. This characteristic is different from that reported in other geographical areas. The stages considered were: anoestrus, follicular phase, early pregnancy and late pregnancy. The ovaries were light-pink and smooth and presented a tortuous cord-like aspect. Many primordial follicles were found in all the females studied. Follicles in different states of maturation and primary and accessory corpora lutea were observed in the cortex. These structures were smaller than those present in other related species. Follicles did not project into the surface of the organ. Calcified follicles of diverse size were found in all the ovaries. Atretic follicles were found in all the stages analysed. Interstitial tissue surrounding the follicles and the corpora lutea was also observed. The number and proportion of different cortical structures varied in the physiological stages analysed. The ovaries of the viscacha have differential characteristics in comparison to other Hystricognathi, some of them related to polyovulation.
