18. Influence of hypoxia on the domiciliation of Mesenchymal Stem Cells after infusion into rats: possibilities of targeting pulmonary artery remodeling via cells therapies?

Abstract

Abstract Background Bone marrow (BM) cells are promising tools for vascular therapies. Here, we focused on the possibility of targeting the hypoxia-induced pulmonary artery hypertension remodeling with systemic delivery of BM-derived mesenchymal stem cells (MSCs) into non-irradiated rats. Methods Six-week-old Wistar rats were exposed to 3-week chronic hypoxia leading to pulmonary artery wall remodeling. Domiciliation of adhesive BM-derived CD45- CD73+ CD90+ MSCs was first studied after a single intravenous infusion of Indium-111-labeled MSCs followed by whole body scintigraphies and autoradiographies of different harvested organs. In a second set of experiments, enhanced-GFP labeling allowed to observe distribution at later times using sequential infusions during the 3-week hypoxia exposure. Results A 30% pulmonary retention was observed by scintigraphies and no differences were observed in the global repartition between hypoxic and control groups. Intrapulmonary radioactivity repartition was homogenous in both groups, as shown by autoradiographies. BM-derived GFP-labeled MSCs were observed with a global repartition in liver, in spleen, in lung parenchyma and rarely in the adventitial layer of remodeled vessels. Furthermore this global repartition was not modified by hypoxia. Interestingly, these cells displayed in vivo bone marrow homing, proving a preservation of their viability and function. Bone marrow homing of GFP-labeled MSCs was increased in the hypoxic group. Conclusion Adhesive BM-derived CD45- CD73+ CD90+ MSCs are not integrated in the pulmonary arteries remodeled media after repeated intravenous infusions in contrast to previously described in systemic vascular remodeling or with endothelial progenitor cells infusions.

19. Synergistic effect of human Bone Morphogenic Protein-2 and Mesenchymal Stromal Cells on chronic wounds through hypoxia-inducible factor-1 α induction

Abstract

Chronic skin ulcers and burns require advanced treatments. Mesenchymal Stromal Cells (MSCs) are effective in treating these pathologies. Bone Morphogenic Protein-2 (BMP-2) is known to enhance angiogenesis. We investigated whether recombinant human hBMP-2 potentiates the effect of MSCs on wound healing. Severe ulceration was induced in rats by irradiation and treated by co-infusion of MSCs with hBMP-2 into the ulcerated area which accelerated wound healing. Potentiation of the effect of MSCs by hBMP-2 on endothelial repair improved skin healing. HBMP-2 and MSCs synergistically, in a supra additive or enhanced manner, renewed tissue structures, resulting in normalization of the epidermis, hair follicles, sebaceous glands, collagen fibre density, and blood vessels. Co-localization of MSCs with CD31 + cells suggests recruitment of endothelial cells at the site of injection. HBMP-2 and MSCs enhanced angiogenesis and induced micro-vessel formation in the dermis where hair follicles were regenerated. HBMP-2 acts by causing hypoxia-inducible factor-1 α (HIF-1α) expression which impacts endothelial tube formation and skin repair. This effect is abolished by siRNA. These results propose that new strategies adding cytokines to MSCs should be evaluated for treating radiation-induced dermatitis, burns, and chronic ulcers in humans.

24. USPIO size control through microwave nonaqueous sol-gel method for neoangiogenesis T2 MRI contrast agent.

Abstract

Aim: Tunable size ultrasmall superparamagnetic iron oxide (USPIO) nanoparticles from 3 to 9 nm coated with polyethylene glycol phosphonate moieties have been designed for neovascularization MRI. Materials & methods: USPIO were synthesized using a nonaqueous sol-gel method. An ischemia-reperfusion rat model has been chosen for neo-angiogenesis and scanned on MRI after injection of different sized USPIO. Histological studies have been performed for USPIO localization within the tissue. Results: The magnetic properties and consequently their MRI relaxivities are drastically dependent on the crystalline core size. In vivo MRI spots were observed specifically in the ischemic area. The best MRI contrast within neovascularization is generated by 6 nm nanoparticles proving that compromise between T2 relaxivity and physico-chemical properties is essential for the design of effective MRI contrast agent. Abstract

Chronic hypoxia (CH)-induced pulmonary hypertension (PHT) is well known to alter K+ channels in pulmonary myocytes. PHT induces right ventricle hypertrophy that increases oxygen demand; however, coronary blood flow and K+ channel adaptations of coronary myocytes during PHT remain unknown. We determined whether CH and PHT altered K+ currents and coronary reactivity and what impact they might have on right myocardial perfusion. Right ventricle perfusion, as attested by microspheres, was redistributed toward hypertrophied right ventricle [RV/LV (%) = 0.59 ± 0.07% in CH rats vs. 0.29 ± 0.03 in control rats, P < 0.05]. Whole-cell patch clamping showed a reduction of global outward current in hypoxic right coronary artery myocytes (H-RCA), whereas hypoxic left coronary artery myocytes exhibited an increase. K+ channel blockers revealed that a 4-aminopyridine (4AP)-sensitive current (Kv current) was decreased in H-RCA (14.3 ± 1.1 vs. 23.4 ± 2.5 pA/pF at 60 mV in control RCA, P < 0.05) and increased in hypoxic left coronary artery myocytes (H-LCA; 26.4 ± 3.8 vs. 11.8 ± 1.6 pA/pF at 60 mV in control LCA, P < 0.05). Constriction to 4AP was decreased in H-RCA when compared to normoxic control and increased in H-LCA when compared to LCA. Finally, we observed that the expression of Kv1.2 and Kv1.5 were lower in H-RCA than that in H-LCA. This study reveals that CH differentially regulates Kv channels in coronary myocytes. Hypoxia decreases Kv currents and therefore reduces vasoreactivity that contributes to an adaptative response leading to right hypertrophied ventricle perfusion enhancement at rest. Abstract

Coronary blood flow control is not uniform along the vascular tree and particularly between the right coronary artery and the left anterior descending artery. Resting membrane potential that contributes largely to the vascular tone is mainly regulated by K+ channels in coronary myocytes. In the present study, we hypothesized that right coronary artery (RCA) and left coronary artery (LCA) exhibited a cell-specific function of K+ channels. The net outward current was markedly greater in RCA compared with LCA cells, and this difference was due to a larger 4-aminopyridine (4-AP)-sensitive voltage-gated potssium (Kv) current in RCA cells, whereas the iberiotoxin (IbTx)-sensitive, large conductance Ca2+-dependent potassium (BKCa) current was smaller in RCA cells. To go further in the molecular identity of this Kv current, we used 50 nM correolide, which specifically blocked Kv1 family α-subunits. Outward currents generated by ramp depolarization protocols were highly sensitive to correolide in both RCA and LCA cells, suggesting that Kv1 contributed for a large part to the net outward current. 4-AP-induced contractions in isolated RCA, and LCA were greater than IbTx-induced contraction. Furthermore, the 4-AP-induced contraction in RCA was significantly greater than that in LCA, which is in agreement with the electrophysiological data. Finally, the Kv1.2 α-subunit but not the Kv1.5 was detected in both RCA and LCA using primary specific antibody in Western blotting and immunofluorescence assay, and expression of Kv1.2 α-subunit was markedly higher in RCA compared with LCA. In summary, we reported for the first time a heterogeneous function and expression of Kv1 α-subunits in rat coronary myocytes isolated from RCA or LCA. [ABSTRACT FROM AUTHOR]

Abstract

International audience; The association between peripheral myopathies and cardiac complications is well established. However, until recently, the clinical and genetic variability of these pathologies limited our ability to recognise individual risk of complications, particularly in the more rare pathologies. Advances have been made in the understanding of the progression, in the physiopathology of molecular deficits and cardiac complications of the different types of muscular dystrophy. This has partially helped to identify the risk of cardiac complications. The commonest condition, Steinert's disease, is associated with a high incidence of atrioventricular block and atrial arrhythmias. Prophylactic implantation of a dual chamber pacemaker with diagnostic functions may be envisaged when the HV interval is greater than 70 ms, on endocavitary electrophysiological investigations. In other patients, follow-up by standard ECG and/or amplified averaged ECG and Holter monitoring is essential. The natural history of Duchenne and Becker muscular dystrophies and the Emery Dreifuss dystrophy have been better described in the last few years. Recommendations have been proposed for the cardiological follow-up of these patients. Empiric recommendations of the same type have been proposed for patients with shoulder and girdle myopathies and propositions for their management have also been made, the pertinence of which is still being evaluated. Our understanding of the incidence, the type, the physiopathology and molecular biology of the various peripheral myopathies and their cardiac complications has advanced considerably in recent years. This has led to the elaboration of new recommendations for diagnostic and therapeutic strategies in these patients.
