Abstract: We present cosmological results from the measurement of clustering of galaxy, quasar and Lyman-$\alpha$ forest tracers from the first year of observations with the Dark Energy Spectroscopic Instrument (DESI Data Release 1). We adopt the full-shape (FS) modeling of the power spectrum, including the effects of redshift-space distortions, in an analysis which has been validated in a series of supporting papers. In the flat $\Lambda$CDM cosmological model, DESI (FS+BAO), combined with a baryon density prior from Big Bang Nucleosynthesis and a weak prior on the scalar spectral index, determines matter density to $\Omega_\mathrm{m}=0.2962\pm 0.0095$, and the amplitude of mass fluctuations to $\sigma_8=0.842\pm 0.034$. The addition of the cosmic microwave background (CMB) data tightens these constraints to $\Omega_\mathrm{m}=0.3056\pm 0.0049$ and $\sigma_8=0.8121\pm 0.0053$, while further addition of the the joint clustering and lensing analysis from the Dark Energy Survey Year-3 (DESY3) data leads to a 0.4% determination of the Hubble constant, $H_0 = (68.40\pm 0.27)\,{\rm km\,s^{-1}\,Mpc^{-1}}$. In models with a time-varying dark energy equation of state, combinations of DESI (FS+BAO) with CMB and type Ia supernovae continue to show the preference, previously found in the DESI DR1 BAO analysis, for $w_0>-1$ and $w_a<0$ with similar levels of significance. DESI data, in combination with the CMB, impose the upper limits on the sum of the neutrino masses of $\sum m_\nu < 0.071\,{\rm eV}$ at 95% confidence. DESI data alone measure the modified-gravity parameter that controls the clustering of massive particles, $\mu_0=0.11^{+0.45}_{-0.54}$, while the combination of DESI with the CMB and the clustering and lensing analysis from DESY3 constrains both modified-gravity parameters, giving $\mu_0 = 0.04\pm 0.22$ and $\Sigma_0 = 0.044\pm 0.047$, in agreement with general relativity. Abstract: We present the samples of galaxies and quasars used for DESI 2024 cosmological analyses, drawn from the DESI Data Release 1 (DR1). We describe the construction of large-scale structure (LSS) catalogs from these samples, which include matched sets of synthetic reference `randoms' and weights that account for variations in the observed density of the samples due to experimental design and varying instrument performance. We detail how we correct for variations in observational completeness, the input `target' densities due to imaging systematics, and the ability to confidently measure redshifts from DESI spectra. We then summarize how remaining uncertainties in the corrections can be translated to systematic uncertainties for particular analyses. We describe the weights added to maximize the signal-to-noise of DESI DR1 2-point clustering measurements. We detail measurement pipelines applied to the LSS catalogs that obtain 2-point clustering measurements in configuration and Fourier space. The resulting 2-point measurements depend on window functions and normalization constraints particular to each sample, and we present the corrections required to match models to the data. We compare the configuration- and Fourier-space 2-point clustering of the data samples to that recovered from simulations of DESI DR1 and find they are, generally, in statistical agreement to within 2\% in the inferred real-space over-density field. 4. A Mechanical Model for the Failure of Reconstructive Breast Implant Surgery Due to Capsular Contracture

Abstract: Capsular contracture is a pathological response to implant-based reconstructive breast surgery, where the ``capsule'' (tissue surrounding an implant) painfully thickens, contracts and deforms. It is known to affect breast-cancer survivors at higher rates than healthy women opting for cosmetic cosmetic breast augmentation with implants. We model the early stages of capsular contracture based on stress-dependent recruitment of contractile and mechanosensitive cells to the implant site. We derive a one-dimensional continuum spatial model for the spatio-temporal evolution of cells and collagen densities away from the implant surface. Various mechanistic assumptions are investigated for linear versus saturating mechanical cell responses and cell traction forces. Our results point to specific risk factors for capsular contracture, and indicate how physiological parameters, as well as initial states (such as inflammation after surgery) contribute to patient susceptibility.

5. Travelling waves and wave pinning (polarity): Switching between random and directional cell motility

Abstract: We derive a simple model of actin waves consisting of three partial differential equations (PDEs) for active and inactive GTPase promoting growth of filamentous actin (F-actin, $F$). The F-actin feeds back to inactivate the GTPase at rate $sF$, where $s\ge 0$ is a ``negative feedback'' parameter. In contrast to previous models for actin waves, the simplicity of this model and its geometry (1D periodic cell perimeter) permits a full PDE bifurcation analysis. Based on a combination of continuation methods, linear stability analysis, and PDE simulations, we explore the existence, stability, interactions, and transitions between homogeneous steady states (resting cells), wave-pinning (polar cells), and travelling waves (cells with ruffling protrusions). We show that the value of $s$ and the size of the cell (length of perimeter) can affect the existence, coexistence, and stability of the patterns, as well as dominance of the one or another cell state. Abstract: Hundreds of exoplanets between 1-1.8 times the size of the Earth have been discovered on close in orbits. However, these planets show such a diversity in densities that some appear to be made entirely of iron, while others appear to host gaseous envelopes. To test this diversity in composition, we update the masses of 5 rocky exoplanets (HD 93963 A b, Kepler-10 b, Kepler-100 b, Kepler-407 b, and TOI-1444 b) and present the confirmation of a new planet (TOI-1011) using 187 high precision RVs from Gemini/MAROON-X and Keck/KPF. Our updated planet masses suggest compositions closer to that of the Earth than previous literature values for all planets in our sample. In particular, we report that two previously identified ``super-Mercuries'' (Kepler-100 b and HD 93963 A b) have lower masses that suggest less iron-rich compositions. We then compare the ratio of iron to rock-building species to the abundance ratios of those elements in their host stars. These updated planet compositions do not suggest a steep relationship between planet and host star compositions, contradictory to previous results, and suggest that planets and host stars have similar abundance ratios., Comment: Submitted to AJ 09/30/2024

7. Restoring Causality in Higher Curvature Gravity

Abstract: Incorporating higher curvature terms into gravity theories modifies the classical field equations, potentially leading to theoretical issues like Shapiro time advancements that violate the Camanho, Edelstein, Maldacena, and Zhiboedov (CEMZ) causality criterion. We explore this criterion within the context of Generalised Quadratic Gravity (GQG), a higher curvature theory with a distinct graviton three-point coupling from General Relativity (GR). By constructing an exact shock wave solution of GQG and calculating the Shapiro time shift for a massless probe graviton, we demonstrate that it can remain strictly positive within a classically allowed parameter space of couplings, ensuring the theory's adherence to the CEMZ criterion. This finding indicates that GQG can offer a causal extension beyond GR, paving the way for further exploration into the consistency of classical higher curvature gravity theories., Comment: 5 pages, 2 figures

Abstract: Asteroseismology of dwarf stars cooler than the Sun is very challenging due to the low amplitudes and rapid timescales of oscillations. Here, we present the asteroseismic detection of solar-like oscillations at 4-minute timescales ($\nu_{\mathrm{max}}\sim4300\mu$Hz) in the nearby K-dwarf $\sigma$ Draconis using extreme precision Doppler velocity observations from the Keck Planet Finder and 20-second cadence photometry from NASA's Transiting Exoplanet Survey Satellite. The star is the coolest dwarf star to date with both velocity and luminosity observations of solar-like oscillations, having amplitudes of $5.9\pm0.8\,$cm$\,\text{s}^{-1}$ and $0.8\pm0.2$ ppm, respectively. These measured values are in excellent agreement with established luminosity-velocity amplitude relations for oscillations and provide further evidence that mode amplitudes for stars with $T_{\mathrm{eff}}<\,5500\,$K diminish in scale following a $(L/M)^{1.5}$ relation. By modeling the star's oscillation frequencies from photometric data, we measure an asteroseismic age of $4.5\pm0.9\,\rm{(ran)} \pm 1.2\,\rm{(sys)}$ Gyr. The observations demonstrate the capability of next-generation spectrographs and precise space-based photometry to extend observational asteroseismology to nearby cool dwarfs, which are benchmarks for stellar astrophysics and prime targets for directly imaging planets using future space-based telescopes., Comment: Accepted for publication in The Astrophysical Journal

Abstract: TOI-6255~b (GJ 4256) is an Earth-sized planet (1.079$\pm0.065$ $R_\oplus$) with an orbital period of only 5.7 hours. With the newly commissioned Keck Planet Finder (KPF) and CARMENES spectrographs, we determined the planet's mass to be 1.44$\pm$0.14 $M_{\oplus}$. The planet is just outside the Roche limit, with $P_{\rm orb}/P_{\rm Roche}$ = 1.13 $\pm0.10$. The strong tidal force likely deforms the planet into a triaxial ellipsoid with a long axis that is $\sim$10\% longer than the short axis. Assuming a reduced stellar tidal quality factor $Q_\star^\prime \approx10^7$, we predict that tidal orbital decay will cause TOI-6255 to reach the Roche limit in roughly 400 Myr. Such tidal disruptions may produce the possible signatures of planet engulfment that have been on stars with anomalously high refractory elemental abundances compared to its conatal binary companion. TOI-6255 b is also a favorable target for searching for star-planet magnetic interactions, which might cause interior melting and hasten orbital decay. Abstract: Formation of organs and specialized tissues in embryonic development requires migration of cells to specific targets. In some instances, such cells migrate as a robust cluster. We here explore a recent local approximation of nonlocal continuum models by Falc\'o, Baker, and Carrillo (2023). We apply their theoretical results by specifying biologically-based cell-cell interactions, showing how such cell communication results in an effective attraction-repulsion Morse potential. We then explore the clustering instability, the existence and size of the cluster, and its stability. Abstract: We present cosmological results from the measurement of baryon acoustic oscillations (BAO) in galaxy, quasar and Lyman-$\alpha$ forest tracers from the first year of observations from the Dark Energy Spectroscopic Instrument (DESI), to be released in the DESI Data Release 1. DESI BAO provide robust measurements of the transverse comoving distance and Hubble rate, or their combination, relative to the sound horizon, in seven redshift bins from over 6 million extragalactic objects in the redshift range $0.1-1$ and $w_a<0$. This preference is 2.6$\sigma$ for the DESI+CMB combination, and persists or grows when SN~Ia are added in, giving results discrepant with the $\Lambda$CDM model at the $2.5\sigma$, $3.5\sigma$ or $3.9\sigma$ levels for the addition of Pantheon+, Union3, or DES-SN5YR datasets respectively. For the flat $\Lambda$CDM model with the sum of neutrino mass $\sum m_\nu$ free, combining the DESI and CMB data yields an upper limit $\sum m_\nu < 0.072$ $(0.113)$ eV at 95% confidence for a $\sum m_\nu>0$ $(\sum m_\nu>0.059)$ eV prior. These neutrino-mass constraints are substantially relaxed in models beyond $\Lambda$CDM. Abstract: We present the measurement of Baryon Acoustic Oscillations (BAO) from the Lyman-$\alpha$ (Ly$\alpha$) forest of high-redshift quasars with the first-year dataset of the Dark Energy Spectroscopic Instrument (DESI). Our analysis uses over $420\,000$ Ly$\alpha$ forest spectra and their correlation with the spatial distribution of more than $700\,000$ quasars. An essential facet of this work is the development of a new analysis methodology on a blinded dataset. We conducted rigorous tests using synthetic data to ensure the reliability of our methodology and findings before unblinding. Additionally, we conducted multiple data splits to assess the consistency of the results and scrutinized various analysis approaches to confirm their robustness. For a given value of the sound horizon ($r_d$), we measure the expansion at $z_{\rm eff}=2.33$ with 2\% precision, $H(z_{\rm eff}) = (239.2 \pm 4.8) (147.09~{\rm Mpc} /r_d)$ km/s/Mpc. Similarly, we present a 2.4\% measurement of the transverse comoving distance to the same redshift, $D_M(z_{\rm eff}) = (5.84 \pm 0.14) (r_d/147.09~{\rm Mpc})$ Gpc. Abstract: Neuropsychiatric genome-wide association studies (GWASs), including those for autism spectrum disorder and schizophrenia, show strong enrichment for regulatory elements in the developing brain. However, prioritizing risk genes and mechanisms is challenging without a unified regulatory atlas. Across 672 diverse developing human brains, we identified 15,752 genes harboring gene, isoform, and/or splicing quantitative trait loci, mapping 3739 to cellular contexts. Gene expression heritability drops during development, likely reflecting both increasing cellular heterogeneity and the intrinsic properties of neuronal maturation. Isoform-level regulation, particularly in the second trimester, mediated the largest proportion of GWAS heritability. Through colocalization, we prioritized mechanisms for about 60% of GWAS loci across five disorders, exceeding adult brain findings. Finally, we contextualized results within gene and isoform coexpression networks, revealing the comprehensive landscape of transcriptome regulation in development and disease.

Abstract: Nucleotide changes in gene regulatory elements are important determinants of neuronal development and diseases. Using massively parallel reporter assays in primary human cells from mid-gestation cortex and cerebral organoids, we interrogated the cis-regulatory activity of 102,767 open chromatin regions, including thousands of sequences with cell type-specific accessibility and variants associated with brain gene regulation. In primary cells, we identified 46,802 active enhancer sequences and 164 variants that alter enhancer activity. Activity was comparable in organoids and primary cells, suggesting that organoids provide an adequate model for the developing cortex. Using deep learning we decoded the sequence basis and upstream regulators of enhancer activity. This work establishes a comprehensive catalog of functional gene regulatory elements and variants in human neuronal development.

Abstract: Two-dimensional shape-morphing networks are common in biological systems and have garnered attention due to their nontrivial physical properties that emanate from their cellular nature. Here, we present the fabrication and characterization of inhomogeneous shape-morphing networks composed of thermoresponsive microfibers. By strategically positioning fibers with varying responses, we construct networks that exhibit directional actuation. The individual segments within the network display either a linear extension or buckling upon swelling, depending on their radius and length, and the transition between these morphing behaviors resembles Landau's second-order phase transition. The microscale variations in morphing behaviors are translated into observable macroscopic effects, wherein regions undergoing linear expansion retain their shape upon swelling, whereas buckled regions demonstrate negative compressibility and shrink. Manipulating the macroscale morphing by adjusting the properties of the fibrous microsegments offers a means to modulate and program morphing with mesoscale precision and unlocks novel opportunities for developing programmable microscale soft robotics and actuators.

Abstract: New development approaches, including launch vehicles and advances in sensors, computing, and software, have lowered the cost of entry into space, and have enabled a revolution in low-cost, high-risk Small Satellite (SmallSat) missions. To bring about a similar transformation in larger space telescopes, it is necessary to reconsider the full paradigm of space observatories. Here we will review the history of space telescope development and cost drivers, and describe an example conceptual design for a low cost 6.5 m optical telescope to enable new science when operated in space at room temperature. It uses a monolithic primary mirror of borosilicate glass, drawing on lessons and tools from decades of experience with ground-based observatories and instruments, as well as flagship space missions. It takes advantage, as do large launch vehicles, of increased computing power and space-worthy commercial electronics in low-cost active predictive control systems to maintain stability. Abstract: An extensive library of symptom inventories has been developed over time to measure clinical symptoms, but this variety has led to several long standing issues. Most notably, results drawn from different settings and studies are not comparable, which limits reproducibility. Here, we present an artificial intelligence (AI) approach using semantic textual similarity (STS) to link symptoms and scores across previously incongruous symptom inventories. We tested the ability of four pre-trained STS models to screen thousands of symptom description pairs for related content - a challenging task typically requiring expert panels. Models were tasked to predict symptom severity across four different inventories for 6,607 participants drawn from 16 international data sources. The STS approach achieved 74.8% accuracy across five tasks, outperforming other models tested. This work suggests that incorporating contextual, semantic information can assist expert decision-making processes, yielding gains for both general and disease-specific clinical assessment.

Abstract: Induction of mycobacterial efflux pumps is a cause of Mycobacterium tuberculosis (Mtb) drug tolerance, a barrier to shortening antitubercular treatment. Verapamil inhibits Mtb efflux pumps that mediate tolerance to rifampin, a cornerstone of tuberculosis (TB) treatment. Verapamils mycobacterial efflux pump inhibition also limits Mtb growth in macrophages in the absence of antibiotic treatment. These findings suggest that verapamil could be used as an adjunctive therapy for TB treatment shortening. However, verapamil is rapidly and substantially metabolized when co-administered with rifampin. We determined in a dose-escalation clinical trial of persons with pulmonary TB that rifampin-induced clearance of verapamil can be countered without toxicity by the administration of larger than usual doses of verapamil. An oral dosage of 360 mg sustained-release (SR) verapamil given every 12 hours concomitantly with rifampin achieved median verapamil exposures of 903.1 ng.h/mL (area under the curve (AUC)0-12 h ) in the 18 participants receiving this highest studied verapamil dose; these AUC findings are similar to those in persons receiving daily doses of 240 mg verapamil SR but not rifampin. Moreover, norverapamil:verapamil, R:S verapamil, and R:S norverapamil AUC ratios were all significantly greater than those of historical controls receiving SR verapamil in the absence of rifampin. Thus, rifampin administration favors the less-cardioactive verapamil metabolites and enantiomers that retain similar Mtb efflux inhibitory activity to verapamil, increasing overall benefit. Finally, rifampin exposures were 50% greater after verapamil administration, which may also be advantageous. Our findings suggest that a higher dosage of verapamil can be safely used as adjunctive treatment in rifampin-containing treatment regimens.

25. Incidence of Natural Conception in Fertility Couples Delayed Treatment in the COVID-19 Shutdown in Australia

Abstract: Background: In vitro fertilization (IVF) is the most widely applied fertility treatment. While the indication for IVF is not disputed in couples where natural conception chances are close to zero (blocked fallopian tubes or severe male infertility), it is less clear when IVF should be started in other couples. Natural conception occurs frequently and has been reported in patients pre- and post-IVF treatment. The outbreak of COVID-19 in the early months of 2020, resulted in Australian fertility clinics stopping treatment. This enabled the opportunity for fertility patients to try and conceive naturally. Methods: We studied couples who were scheduled to start IVF in March 2020 but who faced delayed treatment due to COVID-19 shutdowns. Outcomes were recorded as couples returned for treatment or were contacted via phone and asked why they did not return. We collected baseline characteristics including maternal age, day of next Day 1, and type of infertility, as well as the pregnancy outcome or reason for not returning. Results: We studied 533 couples who were delayed IVF treatment for a total of 1,204 observation months. From the initial cohort of 533 couples delayed treatment, 16 couples achieved pregnancy (3%) with 12 resulting in live birth (2.3%). When analyzed by indication, patients with ovulatory disorders (4 of 55 [7.2%]) or reduced ovarian reserve (7 of 141 [5%]) proved to have higher rates of natural conception compared to other diagnostic categories. There were 39 couples who did not return to treatment and did not achieve natural conception. Conclusion: There is a significant emotional toll that patients experience when undergoing fertility treatment. Natural conception is evident within cohorts scheduled for IVF treatment, and this study indicates that the chance per cycle of live birth in couples waiting to undergo medically assisted reproduction during the COVID pandemic was 2.3%.

Abstract: Weather conditions that can enhance wildfire potential are a problem faced by many countries around the world. Wildfires can have major economic impacts as well as prolonged effects on populations and ecosystems. Distributing information on fire hazards to the public and first responders in real time is crucial for fire risk management and risk reduction. Although most fires today are caused by people, weather conditions determine if and how fast the fire spreads. In particular, research has shown that atmospheric vapor pressure deficit (VPD) is a key parameter predicting the dryness of vegetation and the available fuel for fires. VPD is determined from the environmental air temperature and relative humidity, both of which are readily obtained from smartphones carried by the public. In this study we use smartphone data from the company Opensignal, collected over almost 4 years and from more than 40 000 users per day, to estimate VPD values. We have found that smartphone data can provide useful information about fire risk and danger. Here we present two case studies from wildfires in Israel and Portugal in which VPD is calculated using calibrated temperature and relative humidity measurements from smartphones. Given the rapid growth in the number of smartphones around the globe, we propose applying smartphone data for meteorological research and fire weather applications. Possible users of these results could be wildfire researchers; public policy specialists in wildfire, climate, and disaster management; engineers working with big data; low-income countries; and citizen science advocates.

We propose three different regularizers and provide analytical solutions in special cases, as well as corresponding efficient optimization algorithms. IBS with constipation (IBS-C) accounts for approximately one-third of IBS cases and is associated with substantial burden of illness and decreased quality of life. This narrative review provides an overview of the current and upcoming treatment options and disease management for IBS-C from a US perspective and discusses the importance of the relationship between patient and health care provider in diagnosis and treatment. A positive diagnostic strategy for IBS-C is recommended, based on clinical history, physical examination, and minimal laboratory tests. An effective communication strategy between patients and health care professionals is essential to ensure early diagnosis and reduce both health care costs and overall disease burden. Treatment typically begins with lifestyle interventions and nonpharmacologic options, such as dietary interventions, fiber supplements, and osmotic laxatives. In patients with inadequate response to these therapies, 4 currently available therapies (lubiprostone, linaclotide, plecanatide, and tenapanor) approved by the US Food and Drug Administration may relieve IBS-C symptoms. These agents are generally well tolerated and efficacious in improving IBS-C symptoms, including constipation and abdominal pain. Our proposed framework provides a complementary method that does not rely on conventional contact tracing or laboratory involvement, including the collection, processing, and analysis of biological samples. We use a text classifier to identify reports of people tweeting about themselves and/or members of their household having COVID-19 infections. A probabilistic analysis is then performed to estimate the hSAR based on the number of self or household, and self and household tweets of COVID-19 infection. The analysis includes adjustments for a reluctance of Twitter users to tweet about household members, and the possibility that the secondary infection was not acquired within the household. Experimental results for the UK, both monthly and weekly, are reported for the period from January 2020 to February 2022. Our results agree with previously reported hSAR estimates, varying with the primary variants of concern, e.g. delta and omicron. The serial interval (SI) is based on the time between the two tweets that indicate a primary and secondary infection. Experimental results, though larger than the consensus, are qualitatively similar. The estimation of hSAR and SI using social media data constitutes a new tool that may help in characterizing, forecasting and managing outbreaks and pandemics in a faster, affordable, and more efficient manner.

Abstract: Abstract The growing number of portable consumer-grade electroencephalography (EEG) wearables offers potential to track brain activity and neurological disease in real-world environments. However, accompanying open software tools to standardize custom recordings and help guide independent operation by users is lacking. To address this gap, we developed HEROIC, an open-source software that allows participants to remotely collect advanced EEG data without the aid of an expert technician. The aim of HEROIC is to provide an open software platform that can be coupled with consumer grade wearables to record EEG data during customized neurocognitive tasks outside of traditional research environments. This article contains a description of HEROIC’s implementation, how it can be used by researchers and a proof-of-concept demonstration highlighting the potential for HEROIC to be used as a scalable and low-cost EEG data collection tool. Specifically, we used HEROIC to guide healthy participants through standardized neurocognitive tasks and captured complex brain data including event-related potentials (ERPs) and powerband changes in participants’ homes. Our results demonstrate HEROIC’s capability to generate data precisely synchronized to presented stimuli, using a low-cost, remote protocol without reliance on an expert operator to administer sessions. Together, our software and its capabilities provide the first democratized and scalable platform for large-scale remote and longitudinal analysis of brain health and disease.

Abstract: Abstract Naturally generated lipid nanoparticles termed extracellular vesicles (EVs) hold significant promise as engineerable therapeutic delivery vehicles. However, active loading of protein cargo into EVs in a manner that is useful for delivery remains a challenge. Here, we demonstrate that by rationally designing proteins to traffic to the plasma membrane and associate with lipid rafts, we can enhance loading of protein cargo into EVs for a set of structurally diverse transmembrane and peripheral membrane proteins. We then demonstrate the capacity of select lipid tags to mediate increased EV loading and functional delivery of an engineered transcription factor to modulate gene expression in target cells. We envision that this technology could be leveraged to develop new EV-based therapeutics that deliver a wide array of macromolecular cargo.

Abstract: Objective. To study epidemiology of resistance to macrolides and fluoroquinolones in Mycoplasmagenitalium isolated from pregnant women in Russia and Belarus. Materials and Methods. A total of 107 M. genitalium DNA samples isolated from pregnant women in Russia (n = 58) and Belarus (n = 49) was included in the study. In order to detect markers of macrolide or fluoroquinolone resistance, polymerase chain reaction in real time with the effect of quenching fluorescence of the probe by a primer was used. Confirmation of mutation type was performed using Sanger sequencing. Results. The prevalence of M. genitalium resistance mutations to macrolides in Russia (Tula, Smolensk) and Belarus (Vitebsk, Brest, Minsk) was 3.4% (2/58) and 6.1% (3/49), respectively. Markers of M. genitalium resistance to fluoroquinolones were detected in 5.2% (3/58) of samples from Russia and 8.2% (4/49) of samples from Belarus. The most common mutations associated with macrolide resistance were A2059G (n = 4), fluoroquinolone resistance – D84N (n = 3) and S80I (n = 3) (numbering according to E. coli). Conclusions. The prevalence of M. genitalium resistance markers isolated in pregnant women highlights a need for regular macrolide and fluoroquinolone resistance surveillance.

The layer of epithelial cells that lines the intestine is permeable to a limited degree, and the amount of paracellular permeability is tightly controlled to enable the absorption of ions, nutrients, and water from the lumen. Increased intestinal permeability to macromolecules can be triggered by a variety of insults, including infections, toxins from food poisoning, or allergens, which in turn cause an inflammatory response and are associated with abdominal pain in patients with IBS. This review article discusses increased intestinal permeability in IBS, focusing on IBS with constipation (IBS-C) through the lens of a patient case with a reported prior diagnosis of “leaky gut syndrome” upon initial contact with a gastrointestinal specialist. We review advantages and disadvantages of several methods of measuring intestinal permeability in patients and discuss when measuring intestinal permeability is appropriate in the therapeutic journey of patients with IBS-C. Furthermore, we discuss a possible mechanism of restoring the intestinal barrier to its healthy state through altering intracellular pH by inhibiting sodium-hydrogen exchanger isoform 3 (NHE3). Tenapanor is a minimally absorbed, small-molecule inhibitor of NHE3 that has been approved by the US Food and Drug Administration for the treatment of IBS-C in adults. Preclinical studies showed that tenapanor may restore the intestinal barrier in IBS-C by affecting the conformation of tight junction proteins via NHE3 inhibition to block the paracellular transport of macromolecules from the intestinal lumen. 50. AmiGo: Computational Design of Amigurumi Crochet Patterns

Abstract: We propose an approach for generating crochet instructions (patterns) from an input 3D model. We focus on Amigurumi, which are knitted stuffed toys. Given a closed triangle mesh, and a single point specified by the user, we generate crochet instructions, which when knitted and stuffed result in a toy similar to the input geometry. Our approach relies on constructing the geometry and connectivity of a Crochet Graph, which is then translated into a crochet pattern. We segment the shape automatically into chrochetable components, which are connected using the join-as-you-go method, requiring no additional sewing. We demonstrate that our method is applicable to a large variety of shapes and geometries, and yields easily crochetable patterns., Comment: 11 pages, 10 figures, SCF 2022
