Background: Members of the ACR gene family are commonly involved in various physiological processes, including amino acidmetabolismand stress responses. In recent decades, significant progress has been made in the study of ACR genes in plants. However, little is known about their characteristics and function in maize. Methods: In this study, ACR genes were identified from the maize genome, and their molecular characteristics, gene structure, gene evolution, gene collinearity analysis, cis-acting elements were analyzed. qRT-PCR technology was used to verify the expression patterns of the ZmACR gene family in different tissues under salt stress. In addition, Ectopic expression technique of ZmACR5 in Arabidopsis thaliana was utilized to identify its role in response to salt stress. Results: A total of 28 ZmACR genes were identified, and their molecular characteristics were extensively described. Two gene pairs arising from segmented replication events were detected in maize, and 18 collinear gene pairs were detected between maize and 3 other species. Through phylogenetic analysis, three subgroups were revealed, demonstrating distinct divergence between monocotyledonous and dicotyledonous plants. Analysis of ZmACR cis-acting elements revealed the optional involvement of ZmACR genes in light response, hormone response and stress resistance. Expression analysis of 8 ZmACR genes under salt treatment clearly revealed their role in the response to salt stress. Ectopic overexpression of ZmACR5 in Arabidopsis notably reduced salt tolerance compared to that of the wild type under salt treatment, suggesting that ZmACR5 has a negative role in the response to salt stress. Conclusion: Taken together, these findings confirmed the involvement of ZmACR genes in regulating salt stress and contributed significantly to our understanding of the molecular function of ACR genes in maize, facilitating further research in this field. Key message: GhHB14_D10 and GhREV_D5 regulated secondary cell wall formation and played an important role in fiber development. Cotton serves as an important source of natural fiber, and the biosynthesis of the secondary cell wall plays a pivotal role in determining cotton fiber quality. Nevertheless, the intricacies of this mechanism in cotton fiber remain insufficiently elucidated. This study investigates the functional roles of GhHB14_D10 and GhREV_D5, two HD-ZIP III transcription factors, in secondary cell wall biosynthesis in cotton fibers. Both GhHB14_D10 and GhREV_D5 were found to be localized in the nucleus with transcriptional activation activity. Ectopic overexpression of GhHB14_D10 and GhREV_D5 in Arabidopsis resulted in changed xylem differentiation, secondary cell wall deposition, and expression of genes related to the secondary cell wall. Silencing of GhHB14_D10 and GhREV_D5 in cotton led to enhanced fiber length, reduced cell wall thickness, cellulose contents and expression of secondary cell wall-related genes. Moreover, GhHB14_D10’s direct interaction with GhREV_D5, and transcriptional regulation of cellulose biosynthesis genes GhCesA4-4 and GhCesA7-2 revealed their collaborative roles in secondary cell wall during cotton fiber development. Overall, these results shed light on the roles of GhHB14_D10 and GhREV_D5 in secondary cell wall biosynthesis, offering a strategy for the genetic improvement of cotton fiber quality. Cymbidium sinense is one of the most important traditional Chinese Orchids due to its unique and highly ornamental floral organs. Although the ABCDE model for flower development is well-established in model plant species, the precise roles of these genes in C. sinense are not yet fully understood. In this study, four SEPALLATA-like genes were isolated and identified from C. sinense. CsSEP1 and CsSEP3 were grouped into the AGL9 clade, while CsSEP2 and CsSEP4 were included in the AGL2/3/4 clade. The expression pattern of CsSEP genes showed that they were significantly accumulated in reproductive tissues and expressed during flower bud development but only mildly detected or even undetected in vegetative organs. Subcellular localization revealed that CsSEP1 and CsSEP4 were localized to the nucleus, while CsSEP2 and CsSEP3 were located at the nuclear membrane. Promoter sequence analysis predicted that CsSEP genes contained a number of hormone response elements (HREs) and MADS-box binding sites. The early flowering phenotype observed in transgenic Arabidopsis plants expressing four CsSEP genes, along with the expression profiles of endogenous genes, such as SOC1, LFY, AG, FT, SEP3 and TCPs, in both transgenic Arabidopsis and C. sinense protoplasts, suggested that the CsSEP genes played a regulatory role in the flowering transition by influencing downstream genes related to flowering. However, only transgenic plants overexpressing CsSEP3 and CsSEP4 caused abnormal phenotypes of floral organs, while CsSEP1 and CsSEP2 had no effect on floral organs. Proteinprotein interaction assays indicated that CsSEPs formed a protein complex with B-class CsAP3-2 and CsSOC1 proteins, affecting downstream genes to regulate floral organs and flowering time. Our findings highlighted both the functional conservation and divergence of SEPALLATA-like genes in C. sinense floral development. Abstract Background Long noncoding RNAs (lncRNAs) play important roles in many physiological and pathological processes, this indicates that lncRNAs can serve as potential targets for gene therapy. Stable expression is a fundamental technology in the study of lncRNAs. The lentivirus is one of the most widely used delivery systems for stable expression. However, it was initially designed for mRNAs, and the applicability of lentiviral vectors for lncRNAs is largely unknown. Results We found that the lentiviral vector produces lncRNAs with improper termination, appending an extra fragment of ~ 2 kb to the 3ʹ-end. Consequently, the secondary structures were changed, the RNA–protein interactions were blocked, and the functions were impaired in certain lncRNAs, which indicated that lentiviral vectors are not ideal delivery systems of lncRNAs. Here, we developed a novel lncRNA delivery method called the Expression of LncRNAs with Endogenous Characteristics using the Transposon System (ELECTS). By inserting a termination signal after the lncRNA sequence, ELECTS produces transcripts without 3ʹ-flanking sequences and retains the native features and function of lncRNAs, which cannot be achieved by lentiviral vectors. Moreover, ELECTS presents no potential risk of infection for the operators and it takes much less time. ELECTS provides a reliable, convenient, safe, and efficient delivery method for stable expression of lncRNAs. Conclusions Our study demonstrated that improper transcriptional termination from lentiviral vectors have fundamental effects on molecular action and cellular function of lncRNAs. The ectopic expression of the EsMYB90 transcription factor gene from halophytic Eutrema salsugineum has been reported to enhance the level of anthocyanin and other flavonoid metabolites in transgenic tobacco. In this study, the wheat JW1 overexpressing EsMYB90 showed longer roots and higher fresh weight than that in wild type (WT) under salt stress. In addition, the transgenic wheat plants displayed significantly higher peroxidase (POD) and glutathione S-transferase (GST) activity, as well as markedly lower malondialdehyde (MDA) content than that of the WT during salt stress conditions. The analysis of histochemical staining and H2O2 level indicated that the accumulation of reactive oxygen species (ROS) was significantly lower in the roots of transgenic wheat plants compared to the WT under salt stress. Transcriptome analysis revealed that the EsMYB90 gene affected the expression of considerable amounts of stress-related genes that were involved in phenylpropanoid biosynthesis and antioxidant activity in transgenic plants subjected to NaCl treatment. Importantly, the significantly upregulated expression genes in transgenic wheat under salt stress were mainly associated with the antioxidative enzymes POD and GST encoding genes compared with the WT. Furthermore, EsMYB90 is suggested to bind with the MYB-binding elements of pTaANS2 and pTaDFR1 by dual luciferase assay, to activate the transcription of TaANS2 and TaDFR1 genes that are encoding key enzymes of anthocyanin biosynthesis in transgenic wheat plants. All the results indicated that, under salt stress, the EsMYB90 gene plays a crucial role in preventing wheat seedlings from oxidative stress damage via enhancing the accumulation of non-enzymatic flavonoids and activities of antioxidative enzymes, which suggested that EsMYB90 is an ideal candidate gene for the genetic engineering of crops. The glutathione S-transferases (GSTs) are encoded by a large gene family and well conserved in all living organisms; however they have evolved and are classified clearly according to each kingdom. GSTs are a dimeric protein that has been reported to maintain redox homeostasis in cells, and to protect organisms against oxidative damage. Recently, we isolated a GST coding gene from stress-treated teff grass (Eragrostis tef) and identified it as a plant-specific phi class GST (EtGSTF1) possessing conserved phi class-specific GST N- and C-terminal domains, GSH binding site, substrate binding pocket, and dimer interface. We found that overexpression of plant-specific phi class EtGSTF1 confers diverse abiotic stress tolerances including salt, osmotic, and heat stresses in E. coli which does not possess phi class GSTs. In addition, EtGSTF1 expression helps the E. coli cells tolerate arsenic (As)-induced cell toxicity. Collectively, although plantae and prokaryotae have differentiated a few billions of years ago, the plant-specific phi class EtGSTF1 could protect prokaryotic organisms by detoxification of molecules under diverse abiotic stresses. Key message: A heat stress transcription factor LlHSFA1 in lily and its relationship with LlHSFA2 was investigated, and its function in enhancing thermotolerance was confirmed by analyzing transgenic Arabidopsis thaliana overexpressed LlHSFA1. Abstract: A large family of heat stress transcription factors that are involved in the heat stress response in plants can induce the expression of multiple genes related to thermotolerance including heat-shock proteins. In this study, a novel class A1 HSF named LlHSFA1 was isolated from leaves of lily ( Lilium longiflorum cv. 'White Heaven') using the rapid amplification of cDNA ends technique. Analysis of the deduced amino acid sequence and construction of a phylogenetic tree showed that LlHSFA1 contained five critical domains and motifs and belonged to the A1 family of HSFs. Following the heat treatment of lily leaves, transcription of LlHSFA1 was induced to a varying extent, related to the time of measurement. The induced expression peak of LlHSFA1 occurred prior to that of LlHSFA2, during the early phase of heat stress. Following transient expression of LlHSFA1 in Nicotiana benthamiana, LlHSFA1 was found to be localized in both the nucleus and the cytoplasm. Analysis using bimolecular fluorescence complementation and a yeast two-hybrid assay demonstrated that LlHSFA1 could interact with LlHSFA2. Use of a yeast one-hybrid assay confirmed that LlHSFA1 had transcriptional activation activity. In transgenic Arabidopsis lines overexpressing LlHSFA1 under unstressed conditions, the expression of some putative target genes was up-regulated, in comparison with expression in wild-type plants, and furthermore, the thermotolerance of the transgenic lines was enhanced. Overall, LlHSFA1 was demonstrated to play an important role in the heat stress response of lily and to be a novel candidate gene for application in lily breeding, using genetic modification approaches. Heat shock transcription factors (Hsfs) are the terminal components of the signal transduction chain mediating the activation of genes responsive to both heat stress and a large number of chemical stressors. This paper aims to clone Hsf from lily and characterize its function by analyses of mRNA expression, transactivation activity and thermotolerance of transgenic Arabidopsis. In this study, the gene encoding HsfA2 with 1,053 bp open reading frame (ORF) was cloned by rapid amplification of cDNA ends (RACE) technique from Lilium longiflorum ‘White heaven’. Multiple alignment and phylogenetic analyses showed that the deduced protein was a novel member of the Hsf class A2. Expression analyses by RT-PCR indicated that LlHsfA2 expression was induced by heat shock and H2O2 treatment, but not by NaCl. It was also found that the expression of LlHsfA2 correlated with thermotolerance in Lilium longiflorum ‘White heaven’ and Oriental hybrid ‘Acapulco’ under heat stress. Furthermore, yeast one-hybrid assay showed that LlHsfA2 had transactivation activity. In addition, overexpression of LlHsfA2 activated the downstream genes including Hsp101, Hsp70, Hsp25.3 and Apx2 and enhanced the thermotolerance of transgenic Arabidopsis plants. Taken together, our data suggest that LlHsfA2 is a novel and functional HsfA2, involved in heat signaling pathway in lily and useful for improvement of thermotolerance in transgenic plants. Background and Objectives: Prostate stem cell antigen (PSCA) is a cell surface, glycosylphosphatidylinositol (GPI)-anchored glycoprotein. Its overexpression has been detected in both local and metastatic prostate cancer (PCa), making it a potential therapeutic target. We previously reported that silencing PSCA by small interfering RNA targeting human PSCA (siRNA-PSCA) inhibited biological activity of PSCA-positive PCa cells leading to reduced proliferation, motility and invasion in vitro. In this study, we extended this in vitro findings to in vivo settings in order to investigate the effects of this specific siRNA on the tumor growth and metastasis development of PCa in vivo., Materials and Methods: The siRNA-PSCA and ectopically overexpressed-PSCA vector were constructed and transfected into human PCa PC-3M and LNCaP cells, respectively, and were subcutaneously inoculated into the male SCID mice. Tumor growth was measured with a caliper, and formation of metastasis in mice bearing xenograft tumors was studied by magnetic resonance imaging (MRI) and autopsy analysis. Western blot and immunohistochemistry were used to assess the expression levels of PSCA protein in tumor tissues from xenograft and distant metastases., Results: Consistent with our previous in vitro findings, the subcutaneous xenografts of PC-3M-siPSCA exhibited the almost completely inhibited expression of PSCA protein in their tumors tissues (P < 0.001 and P < 0.001, respectively), and consequently had a significant reduction in tumor growth volumes (P < 0.05 for all), and metastasis onset and sites (P < 0.001 for all) compared to those of PC-3M and PC-3M-siScrm. Conversely, LNCaP-PSCA showed significantly enhanced primary tumor growth and metastasis formation of xenografts compared to LNCaP-vehicle and LNCaP cells (P < 0.001 for all). Moreover, the up-regulated expression of PSCA protein was detected in the distant metastases of xenograft tumors from all groups., Conclusions: Taken together, these observations suggest that PSCA has a promoting role in the growth and metastasis of PCa and siRNA-PSCA may be a potential therapeutic strategy for PSCA-positive PCa., (© 2015 Wiley Periodicals, Inc.)
