The present study was designed to investigate the effects ofRheum ribesL. plant root extracts on DNA damage, biochemical and antioxidant parameters in rats with experimental obesity induced with a high-calorie diet. The study groups were divided as "normal control(NC)", "obese control(OC)", "obese +Rheum ribes(OR1)(200 mg/kg)" and "obese +Rheum ribes(OR2)(400 mg/kg)". At the end of the application, rats were sacrificed and blood and tissue samples were obtained. According to the results obtained, the marker of DNA damage in tissues of 8-OHdG was determined to be significantly reduced in brain tissue of the OR1 and OR2 groups compared to the NC group. However, fluctuations were identified in the MDA activity, antioxidant defense system elements and serum biomarkers in tissues. In conclusion,Rheum ribesplant root extract ensured improvements in DNA damage in brain tissues and MDA levels and showed positive effects on antioxidant parameter activities in different tissues.

The therapeutic potential and antioxidant capacity of Ferula elaeochytris extract (FE) in the liver, kidney and pancreas of rats with diabetes induced by streptozotocin (STZ) was assessed using biochemistry, histopathology and immunohistochemistry. Forty adult Wistar albino male rats were divided randomly into five groups of eight rats each. The normal control (NC) group was untreated. The diabetes control (DC) group was treated with STZ to induce diabetes. The diabetes + acarbose group (DAC) was treated with STZ, then with acarbose daily for 28 days. The diabetes + FE (DFE) group was treated with STZ, then FE daily for 28 days. DC rats had inflammatory cell infiltration, hydropic degeneration and necrosis, whereas the DFE rats exhibited nearly normal histology. Insulin immunostaining in the pancreatic beta cells was decreased in the DC group compared to the NC group, whereas the DFE group was similar to the NC group. Many serum biomarkers of damage to liver, kidneys or pancreas were elevated in the DC group compared to the NC group; these biomarkers were decreased in the DFE group. The DC group exhibited increased malondialdehyde levels and decreased levels of the antioxidant defense system constituents compared to the NC group. The level of biomarkers the DFE group was close to the NC group. FE exhibited a protective effect against tissue damage owing to its antioxidant activities and to its ability to effect regeneration of beta-cells in STZ induced diabetic rats.

Bu çalışma yüksek yağlı diyetle indüklenmiş sıçanlardaGundelia tournefortii L. (Kenger)bitki ekstrelerinin hematolojik parametreler üzerine etkilerinin araştırılmasıamacıyla gerçekleştirilmiştir. Bu amaçla canlı ağırlıkları 150-200 gr olantoplam 24 adet Wistar-albino ırkı erkek sıçan, her grupta 6 sıçan olacakşekilde toplam 4 gruba ayrıldı. Gruplar ''Normal Kontrol (NK)'', ''Obez Kontrol(OK)'', ''Obez+Gundelia tournefortii (200mg/kg)(OG1)''ve ''Obez+ Gundelia tournefortii (400mg/kg)(OG2)''olacak şekilde oluşturuldu. 12 hafta boyunca devam eden uygulama sonundasıçanlar sakrifiye edilerek, kan örnekleri alındı. Tam kanda total lökosit(WBC), Eritrosit (RBC), Hemoglobin (HGB), Hematokrit (HCT), Ortalama AlyuvarHacmi (MCV), Ortalama Alyuvar Hemoglobin Konsantrasyonu (MCHC), OrtalamaAlyuvar Hemoglobini (MCH), Trombosit (PLT), Trombosit Dağılım Genişliği (PDW), BüyükHücreli Trombosit Oranı (PLCR), Prokalsitonin (PCT), Eritrosit DağılımGenişliği (RDW) ve Ortalama Trombosit Hacmi (MPV) ölçümleri yapıldı. Analiz sonuçlarınagöre; PDW, HCT ve WBC parametre değerleri için OK grubu ile kıyaslandığında hemOG1 hem de OG2 gruplarında istatiksel olarak anlamlı bir farklılık belirlenmişiken PLCR ve MPV parametre değerleri için ise OK grubu ile kıyaslandığında OG1grubunda istatiksel olarak anlamlı bir farklılık belirlenmiştir. Buparametreler dışında WBC, RBC, HGB, MCV, MCH, MCHC, RDW, PLT, PCT ve RDWparametre değerleri için OG1 ve OG2 gruplarında hem NK hem de OK gruplarıylayapılan karşılaştırmalarda dalgalanmalar görülmesine rağmen, bu değişimler istatistikselolarak anlamlı bulunmamıştır.Sonuçolarak; Gundelia tournefortii bitkiekstresinin, yüksek yağlı diyet ile obezite oluşturulan sıçanlarda ortaya çıkanhiperkolesterolemik şartlarda hematolojik değerler üzerinde olumsuz sayılabilecekbazı etkilere karşı etkili olabileceği söylenilebilir.

The aim of this study was to investigate the protective role of Urtica dioica seed (UDS) extract against azoxymethane (AOM)-induced colon carcinogenesis in rats. Thirty-two male Wistar albino rats were divided into four groups: Control, AOM, AOM + UDS, and UDS. The AOM and AOM + UDS groups were induced by AOM (15 mg/kg body weight) subcutaneously once a week for 10 weeks. AOM + UDS and UDS groups additionally received fed with pellets included 30 ml/kg UDS extract. At the end of the trial, blood and colon tissue samples were taken from the rats following necropsy. The gross and histopathological findings revealed that the administration of UDS extract significantly decreased lesions including aberrant cript foci, adenoma, and adenocarcinoma formation both numerically and dimensionally. Immunohistochemically, slight CEA and COX-2, strong Caspase-3 immune-expressions were detected in the group AOM + UDS compared to AOM group. Biochemical examinations indicated that a markedly increase in the malondialdehyde and fluctuated antioxidant defense system constituents levels such as reduced glutathione, glutathione s-transferase, glutathione peroxidase, superoxide dismutase were restored in AOM + UDS group. These results reveal that the UDS may act as a chemopreventive dietary agent, inducing apoptosis, resulting in a significant reduction of colon carcinogenesis.

Objective: In this study, the ameliorative potential and antioxidant capacity of treated with different doses of Bryonia multiflora extract (BE) was investigated using histopathological and immunohistochemical changes in pancreatic beta cells, liver and kidney tissues of streptozotocin (STZ)-induced diabetic rats. Material and Methods: A total of forty-two healthy adult Wistar albino male rats were divided randomly into six groups as Control (C); Diabetes mellitus (DM); DM+Akarboz 20 mg/kg; DM+100 mg/kg BE extract (BE1); DM+200 mg/kg BE extract (BE2); DM+400 mg/kg BE extract (BE3). Experimental diabetes was established by a single-dose [45 mg/kg, intra-peritoneal (i.p)] STZ injection. Essential dosages of BE extracts and Akarboz were applied with gastric gavage for 21 day. Blood glucose levels were recorded throughout the all experiment period. Results: Histopathological studies showed that hepatorenal and pancreatic protection by depending on the dose level of BE extracts was further supported by the almost normal histology in DM+BE extract-treated group as compared to the degenerative changes such as disorder of architectural structure, inflammatory cell infiltration, hydropic degeneration and necrosis in pancreas, liver and kidney tissues of STZ-treated rats. Immunohistochemical investigation revealed that STZ-induced degenerative changes in beta-cells in the pancreas of the diabetic rats has reduced insulin immunoreactivity. On the other hand, insulin immunoreactivity in the beta cells of pancreas of BE -treated diabetic rats has significantly increased. Additionally, Glutathione peroxidase 1 (GPx1) immunoreactivity was lower in the tissues of diabetic rats (DM group) compared to the other groups. Conclusion: In conclusion, BE extract has a protective effect on tissue damage probably due to its antioxidant activity and possess the ability to regenerate beta-cell in STZ-induced diabetic rats.

Objective: This study aims to investigate the antioxidant properties and protective effects of Heracleum persicum (HP) extract in streptozotocin (STZ)-induced diabetic rats. Material and Methods: Forty-two Wistar albino male rats were divided into six groups including Control (C); Diabetes mellitus (DM); DM+Akarboz 20 mg/kg; DM+100 mg/kg HP extract (HP1); DM+200 mg/kg HP extract (HP2) and DM+400 mg/kg HP extract (HP3). Experimental diabetes was established by a single-dose [45 mg/kg, intra-peritoneal (i.p)] STZ injection. Essential dosages of HP extracts and Akarboz were applied with gastric gavage for 21 day. Results: In histopathological evaluation of the stained liver and kidney sections of diabetic rats showed degeneration and necrosis of hepatocytes, inflammatory cell infiltration and hydropic degeneration and necrosis in tubulus epithelial cells, disorder of glomerular structure and lymphocyte infiltration. These histopathological changes were ameliorated in the HP-treated rats depending on the dose level. Glutathione peroxidase 1 (GPx-1) immunoreactivity was detected in hepatocytes of liver and tubule epithelial cells of kidney. We have shown that treatment with extracts of HP modulates GPx-1 expression in HP-treated rats. STZ-induced degenerative changes in beta-cells caused decreases in the number of functioning beta-cells and insulin immunoreactivity in the pancreas of the diabetic rats. The pancreas of HP-treated rats were improved and the number of immunoreactive beta cells were significantly increased. Conclusion: Our data suggests that the STZ-induced immunohistochemical and histopathological alterations could be prevented by HP extract probably due to possess the ability to regenerate beta-cells.

Aberrant vessels is the most common cause of extrinsic uretero-pelvic junction (UPJ) obstruction. Due to the left flank pain, 18-year-old male patient with UPJ obstruction due to aberrant vessels in left malrotated kidney and right renal cross ectopia without fusion, had been operated. Ureter was reconstructed and anastomosed anterior to the aberrant vessels after the Double-J-Stent has been placed. At the postoperative period there were improvements in the clinical symptoms and renal function. So, even in the later childhood surgical choice is still the acceptable treatment modality in such cases. To the best of our knowledge, although there are some papers about aberrant vessels which cause UPJ obstruction, there is no such a case with ipsilateral renal hydronephrosis due to aberrant vessels associated with contralateral renal cross ectopia without fusion.
