12. Morphology of PP/COC composites: effects of shear rate and organoclay partitioning

Abstract

The blends and hybrid nanocomposites of polypropylene - cycloolefin copolymer were prepared by a twin-screw extruder followed by microfibril formation using a single screw extruder. The effects of shear rate and organoclay on the morphology of polypropylene - cycloolefin copolymer (PP/COC - 80/20 wt. %) blends were studied by using a combination of rheological measurements, X-ray diffraction (XRD) and scanning electron microscopy (SEM). It was found that although the viscosity ratio of PP/COC blend was unfavorable for COC droplet deformation, the COC phase was converted to finely dispersed fibrils in PP matrix. This could be asserted to the high elasticity of the COC droplets that suppressed the droplet breakup in the favor of fibrillation process. The high glass transition temperature of COC (140 degree centigrade) could also assist restoring the generated microfibril morphology upon cooling. The experimental results also depicted that the droplet deformation and microfibril formation of PP/COC/organoclay nanocomposites mainly depend on the organoclay partitioning, which could be controlled predominantly by kinetic parameters. While the localization of organoclay in the PP matrix resulted in smaller COC droplets and fibrils with smaller diameter, the presence of organoclay inside COC droplets reduced the droplet deformability, leading to fibrils with larger diameter., Comment: 24 pages, 11 figures

Abstract

Doxorubicin (DOX) is a chemotherapeutic with considerable efficacy, but its application is limited due to cardiotoxicity. Nanoparticles can improve DOX efficacy and prevent its adverse effects. Herein, DOX-loaded extracellular vesicles (DOX-EVs) were prepared using different loading methods including incubation, electroporation, and sonication in different hydration buffers to permeabilize nanostructures or desalinize DOX for improved entrapment. Different protein:drug (µg:µg) ratios of 1:10, 1:5, and 1:2, and incubation parameters were also investigated. The optimal formulation was characterized by western blotting, electron microscopy, Zetasizer, infrared spectroscopy, and release study. The cellular uptake and efficacy were investigated in MCF-7 spheroids via MTS assay, spheroid formation assay (SFA), confocal microscopy, and flow cytometry. The percentage of entrapment efficiency (EE) of formulations was improved from 1.0 ± 0.1 to 22.0 ± 1.4 using a protein:drug ratio of 1:2 and sonication in Tween 80 (0.1%w/v) containing buffer. Characterization studies verified the vesicles' identity, spherical morphology, and controlled drug release properties. Cellular studies revealed the accumulation and cytotoxicity of DOX-EVs in the spheroids, and SFA and confocal microscopy confirmed the efficacy and cellular localization. Flow cytometry results revealed a comparable and amplified efficacy for DOX-EV formulations with different cell origins. Abstract

Introduction: Distinguishing the cellular origin of lung cancer is essential for tailored patient care. This pioneering pilot study explores the synergy of ¹HMagnetic Resonance Spectroscopy (¹H-MRS) and 2-[18F] fluoro-2-deoxy-D-glucose positron emission tomography/computed tomography ([18F]FDG PET/CT) in the differentiation of primary squamous cell carcinoma (SCC) of the lung from Hodgkin's lymphoma (HL) metastases. Methods: Ethically approved, the study enrolled 21 participants with confirmed lung lesions (10 SCC, 11 HL). [18F]FDG PET/CT and ¹H-MRS were conducted, and analyses were performed to assess diagnostic potential. Results: Significant differences in [18F]FDG PET/CT parameters (SUVmax BSA, SUVmax LBM, and ID%) between SCC and HL were observed. Metabolite concentrations (Cho, Lac, Cr) from ¹H-MRS also exhibited distinctions. Correlations between PET values and metabolite concentrations hinted at links between glucose metabolism and molecular composition. Conclusion: This study presents an innovative approach, integrating ¹H-MRS and [18F]FDG PET/CT to distinguish primary from metastatic lung lesions. The results hold promise for improving non-invasive diagnostic accuracy and guiding targeted therapies. Abstract

Introduction: Diagnosing Juvenile Idiopathic Arthritis (JIA) presents challenges due to symptom variations, clinical-radiologic delays, and the absence of definitive diagnostic tools. This study aimed to evaluate the diagnostic capability of radiomic features derived from blood pool phase images obtained through bone scintigraphy in JIA. Methods: A cohort of 190 patients was included, utilizing the area between knee growth plates as the region of interest (ROI) for extracting image features. After preprocessing, quantitative features were extracted from original and filtered images. A recursive feature elimination (RFE) algorithm identified significant features, subsequently employed in training a random forest classifier. Results: In the validation phase, our radiomic model, comprising 14 features (4 original and 10 filtered image features), achieved an area under the receiver operating characteristic curve (AUC) of 0.89 (95% CI: 0.88-0.92). This robust performance confirmed the efficacy of radiomics in identifying active knee arthritis using technetium--99m-methyl diphosphonate blood pool images in JIA patients. Conclusion: This study highlights the diagnostic accuracy of radiomics in discerning arthritic joints, suggesting its potential as an alternative to conventional quantification techniques. Abstract

Objective: The clinical studies of acute myeloid leukaemia (AML) revealed that antigen escaping variants cause cancer recurrence even after treatment with chimeric antigen receptor (CAR)-T cells that target a single tumour antigen. Due to the heterogeneous expression of antigens on leukaemia blasts, we hypothesized that a novel bispecific CAR, directed to the folate receptor beta (FRβ)-binding single-chain variable fragment (scFv) and an IL3α-binding receptor (CD123) that has more expression in AML blasts, can decrease CAR-T cell exhaustion and increase the efficacy of CAR-T cells to prevent antigen escaping and consequent recurrence of AML. Materials and Methods: In this experimental study, the survival, proliferation, and cytolysis of CAR-T cells remains suboptimal even with a costimulatory endodomain. Hence, we designed and constructed a tandem CAR that joins an FRβ and CD123 in the second generation retroviral vector to generate a bispecific tandem CAR (TanCAR-T cell). Results: TanCAR FRβ-CD123 T cells showed distinct binding to FRβ or CD123 expressing cells. They could lyse the leukaemia cell lines (66.1 ± 11%) comparable to the single CAR-T cells against these determinants. TanCAR FRβ-CD123 T cells simultaneously engaged FRβ and CD123, which promoted T cell activation in targeting and lysis of the examined leukaemia cell lines. TanCAR-T cell significantly induced interferon gamma (IFNγ) and interleukin 2 (IL-2) production more than single CAR-T cells, which produced a synergistic enhancement of TanCAR FRβ-CD123 T cell function when dual antigens faced simultaneously. Conclusion: Dual-specific TanCAR FRβ-CD123 T cells showed therapeutic potential to improve AML control by coengaging FRβ and CD123 molecules in a robust, divalent immune system. This strategy may be a useful therapeutic approach in patients with relapsed B-cell malignancies.
