Summary A new species of the dagger nematode, genus Xiphinema, was recovered from the rhizospheric soil samples collected from rangelands in Semnan province, north-central Iran, and described based upon both morphological and molecular data. Xiphinema sangesarense n. sp. is characterised by 3.7-4.8 mm long females having 136-154 μm long odontostyle, 74-87 μm long odontophore, two equally developed genital tracts with crystalloid bodies in the tubular part of the uterus, short dorsally convex, ventrally slightly convex tail with a mucron at the end, four juvenile developmental stages and males present, functional. The new species most closely resembles X. azarbaijanense and is regarded as its tentative cryptic form, but was separated from it by reproductive mode, and morphological and molecular differences. Compared to X. afratakhtehense, the second similar species, it could be separated based on morphological differences, especially characteristics of juveniles and molecular data. Furthermore, the new species has close morphology with five know species, namely: X. aequum, X. horvatovicae, X. illyricum, X. macedonicum and X. vuittenezi, in terms of morphological characteristics (mainly structure of uterus and similar female tail), and their differences are discussed. In phylogenetic analyses of the new species using several sequences of two partial large subunit (LSU) D2-D3, and internal transcribed spacer (ITS) sequences, the new species formed a clade with Xiphinema sp. in the LSU tree, and in the ITS tree it formed a clade with X. afratakhtehense. In the cytochrome c oxidase I (COI) gene tree, sequences of the new species occupied a placement inside the clade of several sequences of X. afratakhtehense.

Summary The family Haliplectidae is reported from Iran for the first time and Haliplectus iranicus sp. n., from a mangrove forest in the Persian Gulf, is described. The new species is characterised by one ventromedian supplement near to cloacal aperture and a group of three supplements, 10-15 μm anterior to cloacal opening, absence of protuberant caudal papillae and tail sexual dimorphism. A tabular key containing morphometric data and morphological information of 26 valid Haliplectus species is presented. Near full length SSU rDNA sequences are provided for H. iranicus sp. n. Our SSU analysis strongly indicates that Haliplectus does not belong to the order Plectida as in the current classification, and confirms the monophyly of Haliplectus based upon currently available data, but phylogenetic relationships of the genus remain uncertain. In fact, the classification of Haliplectidae is considered incerta sedis within the class Chromadorea.

A new species from the family Tylenchidae was recovered in the grasslands of Isfahan province, Iran, and is herein described based on morphological and molecular data. Ottolenchus isfahanicus n. sp. is mainly characterized by having a faintly annulated cuticle; elongated, slightly sigmoid amphidial apertures developed metacorpus with distinct valve under LM; vulva located at 69.4-72.3% of body length; large spermatheca about 2.75 times of corresponding body width; and elongated conoid tail with broadly rounded tip. SEM observations showed that the lip region is smooth; the amphidial apertures are elongated, slightly sigmoid slits; and the lateral field is a simple band. It is further characterized by 477-to-515-μm-long females with delicate 5.7-to-6.9-μm-long stylets with small, slightly posteriorly sloping knobs, as well as functional males, in the population. The new species closely resembles O. facultativus, but is separated from it based on morphological and molecular data. It was further morphologically compared with O. discrepans, O. fungivorus, and O. sinipersici. The phylogenetic relationships of the new species with other relevant genera and species were reconstructed using near-full-length sequences of small subunit and D2-D3 expansion segments of large subunit (SSU and LSU D2-D3). In the inferred SSU phylogeny, the newly generated sequence of Ottolenchus isfahanicus n. sp. formed a clade with two sequences of O. sinipersici and sequences assigned to O. facultativus and O. fungivorus. In the inferred LSU phylogeny, the three newly generated sequences of the new species and LSU sequences of O. sinipersici and O. discrepans formed a clade., The financial support of Tarbiat Modares University, Iran, and University of Jaén, Spain, through the research program “PAIUJA 2021/2022: EI_RNM02_2021” are appreciated. Summary Ditylenchus acantholimonis n. sp. is described based on morphological, morphometric and molecular characters. It was isolated from the rhizosphere soil of Acantholimon sp. in Golestan province, Iran, and is mainly characterised by having four lines in the lateral field, a pyriform to bottle-shaped offset pharyngeal bulb, post-vulval uterine sac 36.6-56.1% of the vulva to anus distance long, and a subcylindrical to conical tail with widely rounded tip. It is further characterised by short to medium-sized females, 480-617 μm long, with a fine stylet having small rounded knobs, V = 80.8-83.6, c = 11.0-13.8, c′ = 3.3-4.6, and males with 16.0-17.0 μm long spicules. The new species was morphologically compared with six species having four lines in their lateral field, rounded tail tip and comparable morphometric data namely: D. dipsacoideus, D. emus, D. exilis, D. paraparvus, D. sturhani, and D. solani. It was also compared with two species, D. ferepolitor and D. angustus, forming a maximally supported clade in the 18S tree. The phylogenetic analyses using the maximal number of Anguinidae and several Sphaerularioidea genera based upon partial 18S and 28S rDNA D2-D3 sequences revealed that Ditylenchus is polyphyletic. In the 18S tree, the new species formed a clade with D. ferepolitor (KJ636374) and D. angustus (AJ966483); in the 28S tree it formed a poorly supported clade with D. phyllobios (KT192618) and Ditylenchus sp. (MG865719).

Summary The Desmodoridae is a diverse and widespread family of free-living nematodes. Here, we provide the first record of the group in the Persian Gulf and describe three new species: Metachromadoroides sinuspersici sp. n., Zalonema iranicum sp. n. and Z. supplementorum sp. n. Metachromadoroides sinuspersici sp. n. is characterised by finely annulated cuticle, short and stout cephalic sensilla, amphidial fovea on cuticular thickening, pharyngeal bulb well developed and partitioned into three sections, absence of precloacal supplements, and presence of 6-8 pairs of rounded postcloacal papillae. Zalonema iranicum sp. n. is characterised by papilliform subcephalic sensilla (best observed with SEM), convex cephalic capsule, large multispiral amphidial fovea with 4-5 turns in both males and females, buccal cavity with one ventrosublateral and two dorsal teeth and posterior body of males with lateral alae extending from the last third of the body to the cloacal aperture and ventral alae extending 1395-2250 μm anterior to the cloacal aperture, and no precloacal supplements. Zalonema supplementorum sp. n. is characterised by four subcephalic sensilla 1-2 μm long, multispiral amphidial fovea with three turns in both males and females, buccal cavity with one dorsal and two ventrosublateral teeth, males with lateral alae present on each side of body from posterior half of body to cloacal aperture, ventral alae extending 942-1257 μm anterior to cloacal aperture, strongly cuticularised spicules 41-43 μm long, and 12-16 precloacal supplements. Near full length SSU and partial D2-D3 LSU sequences are provided for M. sinuspersici sp. n. and Z. iranicum sp. n., and the COI sequence is provided for Z. iranicum. The SSU phylogeny suggests a close relationship between M. sinuspersici sp. n. and Metachromadora and Metachromadoroides species and the monophyly of Zalonema (after currently available data). The LSU phylogeny suggests an affinity between Metachromadoroides and Zalonema with Spirinia and Acanthopharynx, respectively.

Five populations of a new dagger nematode species were recovered from natural grasslands and forests of north and northwest Iran, and described based upon morphological and molecular data in present paper. Xiphinema hyrcaniense n. sp. is characterized by 3.9–5.5 mm long females, having 102–142 μm long odontostyle, 64–88 μm long odontophore, guiding ring located at 115–147 μm distance from anterior end, two equally developed genital branches having crystalloids in tubular part of uteri and pseudo-Z-organ at their junction with pars dilatata uteri, short, rounded to dorsally more convex tail with a mucro or in few specimens, without it. Common males with 72–95 μm long spicules and four juvenile developmental stages. The new species is similar to six known species belonging to artificial morphospecies group 5, especially looking closest to X. montenegrinum in its general morphology, but could be separated by ontogenesis of tail shape and morphometric indices. In molecular phylogenetic analyses using partial large subunit, and internal transcribe spacer 1 ribosomal DNA (LSU D2-D3 and ITS1 rDNA) sequences, the new species formed a clade with X. cretense and Xiphinema sp. in LSU; and in ITS1 tree, with X. dentatum and X. paradentatum. © 2021, Koninklijke Nederlandse Planteziektenkundige Vereniging.

Summary Synonchium iranicum sp. n. and Richtersia bispinata sp. n. are described from the Persian Gulf and their relationships investigated using SSU phylogenetic analyses. Synonchium iranicum sp. n. is characterised by a buccal cavity with three equally sized mandibles, each with five small teeth, a rounded tail with three distinct caudal glands and distinct duct, males without gubernaculum and supplements, and short spicules. Richtersia bispinata sp. n. is characterised by multispiral amphids with 3.0-3.5 turns in males and 2.0-2.5 turns in females, cuticle with numerous, irregularly arranged, simple spines in the cervical region, 20-25 longitudinal rows of double spines in the mid-body region, 15-25 longitudinal rows of simple spines in the tail region, and the left spicule markedly longer than the right one. We provided the first near-full-length small subunit ribosomal DNA (SSU rDNA) sequences for both newly described species, as well as cytochrome c oxidase I (COI), and provided D2-D3 expansion segments of large subunit ribosomal DNA (LSU rDNA) sequence for the new species of Richtersia. Our SSU phylogenetic analyses confirmed the placement of Synonchium iranicum sp. n. within the Selachinematidae, and suggested that the selachinematids may form a distinct clade from other chromadorids Our phylogenetic analyses indicated that Richtersia should not be classified within the Selachinematidae as suggested by some authors. Instead, we propose that Richtersia should be placed within the Desmodorida. Although Richtersia differs from Desmodorida in the arrangement of cephalic sensilla, it shares the golden body colouration characteristic of desmodorids in glycerin preparations, the same structure of genital branches in both males and females, and similar cuticular ornamentation to some desmodorid genera (notably Desmodorella).

Summary Members of the family Leptolaimidae are reported for the first time from Iran with description of two new species of Leptolaimus from the Persian Gulf in the south of the country. Leptolaimus hormozganensis sp. n. is described based upon three populations. It differs from other Leptolaimus species in the position of the pharyngeal supplement and the number of precloacal tubular supplements and has a single sickle-shaped tubular pharyngeal supplement at mid-pharynx level in both sexes, 9-12 male precloacal supplements and one female preanal supplement. Leptolaimus dimorphus sp. n. is characterised by the presence of a pharyngeal tubular supplement only in some adults, one female preanal supplement and four male precloacal supplements. It differs from other species in that the pharyngeal supplement is only present in some specimens, presence of four male precloacal supplements, tail 50-59 μm long and ventrosublateral position of vulva on either the right or left side of the body. Near full length SSU rDNA sequences are provided for L. hormozganensis sp. n. and two different males (one with, and one without, a pharyngeal supplement) of L. dimorphus sp. n. COI sequences for three populations of L. hormozganensis sp. n. from different regions of the Persian Gulf are also obtained. New sequences of two Leptolaimus species formed a clade with other sequenced Leptolaimus spp. and representatives of two genera of Leptolaimidae, i.e., Aphanolaimus and Paraplectonema in a phylogeny based on SSU rDNA gene with 1.00/81 Bayesian posterior probability/maximum likelihood bootstrap (BPP/BS) values. Molecular phylogeny confirmed the synonymy of Leptolaimus and Halaphanolaimus that was formerly proposed based only on morphology. Based on SSU sequences and morphological characters, the presence of a pharyngeal supplement may not be a species-specific character and should not be used to differentiate genera.

Summary Ektaphelenchoides shiroodensis n. sp. is described and illustrated based upon morphological, morphometric and molecular data. It was recovered from the bark samples of a dead alder tree (Alnus sp.) from countryside around Shirood city, Mazandaran province, in the north of Iran. The new species is characterised by 768-985 μm long females, its lip region separated from the body contour by constriction, lateral field with three barely visible lines, forming two weak bands, 26-29 μm long stylet with wide lumen without conophore and basal swellings, excretory pore always at the level with median bulb, post-vulval uterine sac (PUS) 43-76 μm long, elongate conoid posterior body region ending to a long filiform part with pointed tip, males common with dorsally convex conical tail ending to a short narrower region with pointed tip, seven precloacal + cloacal + caudal papillae and arcuate spicules with wide condylus, blunt rostrum and simple end. Based upon the similar posterior body region (‘tail’) and comparable PUS length, the new species was compared to three known species, viz., E. attenuata, E. hunti and E. pini. Comparisons with the aforementioned species and similar species under the genus Seinura are discussed. The phylogenetic affinities of the new species, based upon two partial small and large subunit ribosomal DNA (SSU and LSU rDNA D2-D3) sequences, are discussed. Furthermore, several ektaphelenchid and seinurid species previously described from Iran were recovered in the present study, sequenced for their aforementioned genomic markers, and included in the phylogenetic analyses.

Summary Two populations of an aphelenchoidid nematode were recovered from natural forests in Golestan province, Iran. Both populations resembled each other morphologically. The females of the first population, from rotten wood of Fagus orientalis in Golestan National Park, measured 294-338 μm long, and females of the second population, from forests of Ramian county, measured 365-464 μm long. A reverse taxonomic approach, based upon SSU and LSU phylogenies using a maximal number of Aphelenchoididae genera, revealed the two isolates to be conspecific and representatives of a new species of Basilaphelenchus. Basilaphelenchus magnabulbus n. sp. is described and illustrated herein and is mainly characterised by the unique form of the metacorpus, this being elongate and filling the corresponding body region similar to the common form in other aphelenchoidids (vs smaller with well posteriorly located small valve in previously described species of the genus). The new species has an elevated lip region, fine stylet with three elongate posteriorly directed knobs, female tail elongate conoid and ventrally bent, and male tail short conical with three pairs of caudal papillae. It was morphologically compared with four previously known species of the genus, viz., B. persicus, B. brevicaudatus, B. gorganensis and B. grosmannae, and three species of Aphelenchoides having a broadly similar morphology.

A new species of the genus Boleodorus, recovered from southern Iran, is described and illustrated based upon morphological and molecular data. B. bushehrensis n. sp. is mainly characterized by having a wide and low cephalic region (which is continuous with the adjacent body), the oral aperture in a depression in side view under a light microscope, four lines in the lateral field, weak metacorpus with a vestigial-to-invisible valve, and short (26–38 mm long) hooked tail with rounded tip. The females are 334–464 mm long and have a spherical spermatheca filled with spheroid sperm; males have 11.5- to 12.0-mm-long spicules and weakly developed bursa. The new species has an annulated low cephalic region, four large cephalic papillae, and small crescent-shaped amphidial openings when observed by scanning electron microscopy (SEM). Its morphological and morphometric differences with seven known species are discussed. The phylogenetic relationships of the new species with other relevant genera and species have been studied using partial sequences of small and large subunit ribosomal DNA (SSU and LSU rDNA). In both the SSU and LSU phylogenies, the sequences of B. bushehrensis n. sp. and other Boleodorus spp. formed a clade. A second species, B. thylactus, when studied under SEM, has a raised, smooth cephalic region, four large cephalic papillae, and oblique amphidial slits, with the oral opening in a depression.

A new species of the genus

A new species of the dagger nematode genus Xiphinema was recovered from three different points in West Azarbaijan province, northwestern Iran, from the rhizosphere of grasses and described based on both morphological and molecular data. Xiphinema azarbaijanense n. sp. belongs to the Xiphinema non-americanum group and is characterized by having 3.8-5.3 mm long females with long odontostyle (129-151 μm), two equally developed genital tracts with crystalloid bodies in the tubular part of uterus, short tail, four juvenile developmental stages and males absent. It is similar to 14 known species belonging to morphospecies group 6, and is almost indistinguishable from most of them using traditional taxonomy; thus, is part of a complex of cryptic species. Molecular phylogenetic data using two genomic structural (18S, 28S D2-D3) and one ITS1 ribosomal DNA (rDNA) loci were used to separate the new species from close taxa. The new species formed an independent lineage in three reconstructed trees based upon the resolved topologies, and was also morphologically compared with phylogenetically related species.

This contribution provides the morphological and molecular identification of a new species of the genus Longidorella (Saevadorella). L. (S.) caspica n. sp., was recovered from the rhizospheric soil of grasses in Mazandaran province in the seashore of the Caspian Sea. It is characterized by females with a length of 788 to 874 μm and a cephalic region with prominent papillae; and separated from the rest of the body by a remarkable constriction, an odontostyle of 32 to 33 μm, vulva at 52.5 to 59.0%, and a tail of 33 to 38 μm with a rounded tip. Males have 32 to 35 μm long spicules of dorylaimoid form and five to seven ventral supplements ending at 48 to 55 μm distance from cloacal pair. The new species was morphologically compared with seven nominal species viz. L. (S.) arenicola, L. (S.) cuspidata, L. (S.) magna, L. (S.) perplexa, L. (S.) saadi, L. (S.) saevae, and L. (S.) tharensis. It was further compared with similar species under the subgenus Longidorella (Enchodorella) viz. L. (E.) deliblatica and L. (E.) murithi. Besides morphological studies, molecular phylogenetic studies using partial sequences of D2 to D3 expansion fragments of 28S rDNA were performed for the new species and its phylogenetic relationships with other species and genera were discussed.

Summary Sigmolenchus n. gen., is proposed as a new member of the Tylenchidae. The new genus is proposed based upon light and scanning electron microscopic (LM and SEM) observations and phylogenetic analyses based upon small, large and internal transcribed spacer sequences of ribosomal DNA (SSU, LSU D2-D3, ITS rDNA) and mitochondrial cytochrome c oxidase I gene (COI mtDNA) sequences. It is characterised partly by its slender body, but mainly by having a dorso-ventrally flattened, smooth and elevated cephalic region, unique elongate sinuous amphidial openings, each appearing as a wavy slit with three peaks, originating from the vicinity of the labial plate, extending posteriorly and reaching and passing the first body annulus, and a spacious amphidial fovea. It has an anteriorly rounded outline under LM, knobbed stylet, its conus comprising about one-third of the total length, lateral field with a single band, metacorpus with a distinct valve, vulva with small lateral flaps, and males common with tylenchoidiform spicules. The new genus is morphologically compared with seven tylenchid genera, viz., Chilenchus, Ecphyadophoroides, Filenchus, Labrys, Lelenchus, Sakia, and Tenunemellus, mainly by having small slender body and dorso-ventrally flat cephalic region, and is placed under the subfamily Ecphyadophorinae because of its slender body, elevated smooth cephalic region, long amphidial openings and spacious amphidial fovea. It was recovered from marine muddy sediments taken 2 m deep in mangrove forests off southern Iran. Currently S. sinuosus n. gen., n. sp. is monotypic. In SSU phylogeny, the new genus formed an unsupported sister relationship with Filenchus discrepans, and in LSU phylogeny it occupied a placement inside a major clade including several Tylenchinae genera.

Summary Extracts of nine agricultural wastes prepared with five different solvents were assessed for their potential nematicidal activity against three nematode species, Aphelenchus avenae, Meloidogyne incognita and Pratylenchus neglectus, in in vitro condition. The 50% v/v hydro-ethanolic extracts showed the highest performance for two tested plant wastes of cabbage leaves and faba bean pods. These two extracts were tested on nematodes in three different concentrations. The highest in vitro nematistatic activity was recorded for 3000 and 1500 ppm of cabbage leaf extracts by 100% paralysis of all three nematode species after 48 h, and the highest nematicidal activity was recorded for the above-mentioned extract by 25-100% mortality depending on nematode species and extract concentration. A 14-94% mortality was recorded for all three species of nematodes after treatment with faba bean pod hydro-ethanolic extract in in vitro conditions. Hatching inhibition and repellent activity of cabbage leaf and faba bean pod extracts were observed in P. neglectus and M. incognita. In vivo assays confirmed the in vitro results when both of the extracts showed moderate to high inhibition of nematode population development and nematode infection parameters on tomato root system in pot experiments.

Summary During nematological surveys in grasslands and natural forests of north and north-western Iran, three species of Miculenchus, including two new and one known species, were recovered and characterised based upon morphological and molecular approaches. Miculenchus brevisalvus n. sp., the first new species, is mainly characterised by its short females 334-388 μm long and with a short 6.0-7.5 μm long stylet, pyriform to pyriform-elongate pharyngeal bulb, 4-8 μm long post-uterine sac (PUS), offset rounded spermatheca filled with small spheroid sperm, elongate conoid tail 62-83 μm long with a sharp tip, and males with simple cloacal lips. Miculenchus muscus n. sp., the second new species, is characterised by a combination of the following features: body 401-467 μm long, well-developed protuberant labial plate at the anterior end under light microscopy, stylet 7-9 μm long, pyriform pharyngeal bulb, PUS 4-9 μm long, gradually narrowing conical tail 62-74 μm long with a finely pointed or sharp end and bearing several fine bristles at tip, and a male with projecting cloacal lips. Both newly described species were morphologically compared with four currently known species of the genus, viz., M. elegans, M. salmae, M. salvus, and M. tesselatus. Miculenchus salmae was also recovered and reported from Iran for the first time. It is mainly characterised by lacking a PUS and the characteristic vagina shape. Miculenchus muscus n. sp. and M. salmae were both characterised using scanning electron microscopic images, yielding new morphological observations for the genus. All three species are studied for their molecular phylogenetic characters using sequences of near-full length fragments of the small subunit ribosomal RNA gene (SSU rDNA) and the D2-D3 expansion segments of the large subunit ribosomal RNA gene (LSU rDNA D2-D3). In both SSU and LSU phylogenies, all currently sequenced species of Miculenchus formed a monophyletic group with maximal clade support in both Bayesian inference and maximum likelihood analysis.

SummaryCryptaphelenchus baujardin. sp. is described and illustrated based on two populations from Golestan province, northern Iran. The new species is characterised by female and male body length of 224 (190-261) and 215 (195-229)μm, respectively, offset cephalic region with a shallow constriction, delicate stylet 6.6 (6.0-7.0)μm long, four incisures in the lateral field, presence of a rudimentary post-uterine sac in the female gonoduct, elongate-conoid female tail ending in an acute to finely rounded terminus, male tail conical, male with seven caudal papillae and delicate spicules. The new species shows a strong resemblance toC. varicaudatusandC. iranicus, but has a sclerotised mass near the tip of the spicule and an indistinct spermatheca. Phylogenetic analyses based on both partial SSU and LSU rDNA sequences confirm its status as a new taxon.

Summary Ektaphelenchoides tonekabonensis n. sp. is described and illustrated based upon morphological, morphometric and molecular characters. The new species is characterised by 476-659 μm long females, lip region broad and continuous with body contour, two weak lateral bands barely visible in lateral view and cross section, needle-like stylet with its conus forming ca 50% of the total length and with a very narrow lumen in the shaft, excretory pore located just opposite the metacorpus base or slightly posterior, a dense (sclerotised) mass just anterior to the vagina, a short post-uterine sac 5-7 μm long, elongate conoid distal body region ending in a long filiform part, males common with a dorsally convex conical tail ending to a filiform tip, 7-8 μm long spicules with a small projection at the capitulum close to the condylus, its tip posteriorly bent, and five precloacal + caudal papillae. Based upon morphological similarities and phylogenetic affinities, it is similar to six known species of the genus, namely: E. attenuata, E. caspiensis, E. fuchsi, E. kelardashtensis, E. musae, and E. ruehmi. Ektaphelenchoides caspiensis is typologically most similar to the new species and can be separated based on vulval position, excretory pore position and male tail characters. The next most similar species, E. kelardashtensis, has a longer stylet and posteriorly located excretory pore. Comparisons with the other aforementioned species and typologically similar species under Seinura are also discussed. Molecular phylogenetic analyses were performed using Bayesian inference (BI) based upon two partial small and large subunit ribosomal DNA (SSU and LSU rDNA D2-D3) sequences and the phylogenetic affinities of the new species in both phylogenies were discussed.

Summary A population of Neolobocriconema serratum was recovered from the rhizosphere of Hosta sp., northern Iran, for the first time. It was studied using morphological and molecular data and new morphological observations were made. The Iranian population is characterised by females having 37-43 body annuli, their outer margin ornamented with 12-14 small lobe-like outgrowths under light microscopy, anastomoses occur sparsely in some females and the succeeding annulus is usually anteriorly bent opposite to the vulva. The cephalic region has a single smooth wavy annulus with grooves and is separated from the first, wider body annulus by a very short neck. Four prominent discoid submedian lobes and two lateral pseudolips present, appearing two layered (i.e., one above the other) in scanning electron microscopy (SEM). Body 487-607 μm long, stylet 85-93 μm long with rounded to anchor-shaped knobs, ovary very long, vulva generally closed, but open in some specimens under SEM. Tail conoid with bluntly rounded end and males absent. The recovered population agreed well with the type population based upon the morphometric data and female morphology. In having an almost identical morphology and morphometrics, N. allantoideum is proposed as a junior synonym of N. serratum. The molecular phylogenetic analyses were done by using near-full-length sequences of the small subunit ribosomal DNA (SSU rDNA), the internal transcribed spacer 1 (ITS1 rDNA) and the mitochondrial cytochrome c oxidase subunit I (COI mtDNA). The Iranian population of this species formed a clade with three other populations of the species in the SSU rDNA, and with one population in both the ITS1 rDNA and the COI mtDNA trees using Bayesian inference.

Summary Two populations of Lobocriconema were recovered from the rhizosphere of Parrotia persica in Rango forest, Gorgan, and Nargesi forest, Nokandeh, Golestan province, northern Iran. Both of the recovered populations were characterised using morphological (light and scanning electron microscopy), morphometric and molecular data based upon small, partial large and internal transcribed spacer 1 ribosomal RNA (SSU, LSU D2-D3, ITS1 rDNA) and the mitochondrial cytochrome c subunit oxidase I (COI mtDNA) gene sequences. The first examined population, recovered from Gorgan, was mainly characterised by having 34-39 retrorse body annuli 11-16 μm thick at mid-body with crenate, dentate and lobulate margins, longitudinal cuticular scratches on entire body on both lateral regions, sometimes one or two anastomoses along the body, a single labial annulus with smooth margin and smaller than the first body annulus, four distinct submedian lobes, simple open vulva with no overhang, and a terminal anus. This population was almost identical to Neolobocriconema iranense, a close examination of a topotype population recovered in this study confirming that they were conspecific. The second population, L. nokandense n. sp., was recovered from the Nokandeh and looked similar to the first population but was morphologically separated from it by a slightly crenate labial annulus under SEM (vs smooth), longer tail (13-21 vs 6-10 μm) and smaller ratio c (23.6-32.4 vs 44.7-86.0). In molecular phylogenetic analyses, the two populations formed a clade in both SSU and LSU phylogenies. In ITS1 and COI phylogenies, the Nokandeh population (the new species) formed a separate sister clade to the N. iranense clade. Detailed observations using SEM data revealed a small annulus between the submedian lobes and the single labial annulus, an open vulva and a slightly ornamented cuticle in all three studied populations (the newly recovered populations from Golestan and the topotype population of N. iranense from Mazandaran province and the new species), corroborating their placement under Lobocriconema. As a result, N. iranense was transferred to Lobocriconema as L. iranense n. comb. (= N. iranense). The new species was morphologically compared with its close relatives L. iranense n. comb., L. pauperum, L. incrassatum, and L. nasuense, all of which having the aforementioned small annulus between the submedian lobes and the first labial annulus. The phylogeny of the genus is discussed.

Summary Basilaphelenchus brevicaudatus n. sp., the third species of this apparently rare genus, is described and illustrated. It was recovered from wood and bark samples from a dead forest tree in the Golestan province of northern Iran. It is typologically characterised by female body length (448 (365-492) μm), three lines in the lateral fields, raised cephalic region having a sclerotised vestibule and cephalic framework, stylet thin with delicate conus and thicker shaft, both parts lacking a visible lumen, and with three elongate, backwardly directed knobs, small, spherical to spade-shaped metacorpus with small, posteriorly located valve (at 72 (58-74)% of metacorpus length), simple vulva without flap at 72.5 (69-78)% of body length, post-vulval uterine sac 32.4 (29.0-37.0) μm long, functional rectum and anus, female tail conical, short (c′ = 2.6 (1.9-3.3) in female, and 2.5 (2.3-2.8) in male), dorsally convex and ventrally concave with blunt end or having a small mucron, both forms with a hyaline-like tip. Males common, with well-curved 9.2 (9.0-10.5) μm long spicules measured along the mid-line, three pairs of small caudal papillae (lacking the single P1 ventral papilla) and no bursa at tail tip, but with hyaline region, similar to females. Basilaphelenchus brevicaudatus n. sp. is compared with the two currently known species of the genus, the type species, B. persicus, and B. grosmannae. Molecular phylogenetic inferences using partial sequences of small and large subunit ribosomal RNA genes (SSU and LSU) from different isolates of the new species revealed that it belongs to the Tylaphelenchinae clade.

This contribution provides the morphological and molecular identification of a new species of the genus Longidorella (Saevadorella). L. (S.) caspica n. sp., was recovered from the rhizospheric soil of grasses in Mazandaran province in the seashore of the Caspian Sea. It is characterized by females with a length of 788 to 874 μm and a cephalic region with prominent papillae; and separated from the rest of the body by a remarkable constriction, an odontostyle of 32 to 33 μm, vulva at 52.5 to 59.0%, and a tail of 33 to 38 μm with a rounded tip. Males have 32 to 35 μm long spicules of dorylaimoid form and five to seven ventral supplements ending at 48 to 55 μm distance from cloacal pair. The new species was morphologically compared with seven nominal species viz. L. (S.) arenicola, L. (S.) cuspidata, L. (S.) magna, L. (S.) perplexa, L. (S.) saadi, L. (S.) saevae, and L. (S.) tharensis. It was further compared with similar species under the subgenus Longidorella (Enchodorella) viz. L. (E.) deliblatica and L. (E.) murithi. Besides morphological studies, molecular phylogenetic studies using partial sequences of D2 to D3 expansion fragments of 28S rDNA were performed for the new species and its phylogenetic relationships with other species and genera were discussed.

Antagonistic fungi are well-known as viable alternatives to chemical control of root-knot nematodes. In this paper, serine protease Ac1 as an important pathogenicity factor was used to enhance the antagonistic activity of Arthrobotrys conoides and A. oligospora against Meloidogyne javanica J2 (second stage juveniles). Ac1 gene was extracted from A. conoides and cloned in pCAMBIA1304 vector. The recombinant plasmid was then transferred to these fungi using two strains of Agrobacterium tumefaciens (LBA4404 and AGL1). Transgenic isolates were confirmed by PCR amplification of Hygromycin B resistance gene (hph) as selectable marker, protease assay using casein substrate and in vitro bioassay. Transferring of Ac1 gene by homologous recombination to A. conoides, increased protease activity. Our results showed that production of protease in both transgenic species was increased compared to wild type. Bioassay results indicated that pathogenicity rates of transformants, in both the number of traps formed and the number of trapped J2, increased compared to the wild type. The results showed that inhibition of root-knot nematode, M. javanica, was markedly increased by transgenic isolates of Arthrobotrys spp. compared to the wild types.

Longidorus azarbaijanensis n. sp. is described and illustrated using morphological and molecular data. It was recovered in West Azarbaijan province, northwestern Iran, from the rhizospheric soil of foxtail weed. The new species is characterized by having 5.4 to 6.8 mm long females, offset, anteriorly flat lip region and separated from the rest of the body by a shallow constriction, funnel-shaped amphidial pouches, the guiding ring at 23 to 27 µm from the anterior end, 73 to 81 and 44 to 50 µm long odontostyle and odontophore, respectively, 95.0 to 113.5 µm long pharyngeal bulb, didelphic-amphidelphic reproductive system with long tubular uteri lacking sperm cells, vulva located at 52.0% to 56.4%, conical tail dorsally convex, ventrally flat, with bluntly rounded wide tip, for juvenile developmental stages and absence of males. The general body shape of the new species is similar to that of five known species of the genus namely L. euonymus, L. perangustus, L. persicus, L. protae and L. sturhani. The morphological differences of the new species with the aforementioned species are discussed. For all the aforementioned species (except L. protae, currently lacking molecular data) the differences of the new species was also confirmed with differences in molecular sequences of D2-D3 expansion domains of 28S rDNA and the corresponding phylogenetic analyses. The partial sequence of the internal transcribed spacer 1 (ITS1) of the new species was also used in phylogenetic analyses. In partial 28S tree, the clade including the new species and six other species (L. attenuatus, Longidorus sp. and four above-mentioned species having molecular data for this fragment) was well supported in Bayesian inference. In the ITS1 tree, the new species formed a clade with L. euonymus, L. perangustus and L. persicus, as in 28S tree. This is one of the cases from which the morphologically similar species are separated using molecular sequences.

Summary Basilaphelenchus persicus n. gen., n. sp. is described and illustrated based on morphological and molecular phylogenetic studies. The new genus belongs to the family Aphelenchoididae and is characterised mainly by the small body size of females (297-393 μm) and males (304-331 μm), high cephalic region in both sexes, fine 5.5-7.8 μm long stylet that is barely visible in mounted specimens yet clear in fresh individuals, stylet with a linear conus and weak shaft ending in three elongate, posteriorly directed, knobs, small spherical metacorpus with posteriorly located (at 72-75% of metacorpus) weak but distinct valve, monodelphic-prodelphic reproductive system with a long post-vulval uterine sac containing large rounded sperm cells, a vulva lacking any flap apparatus, conical tail of female usually ventrally bent at tip and male tail ventrally bent as in the female, lacking the P1 supplement, but with three pairs of caudal papillae, and small arcuate spicules with rounded condylus and small pointed rostrum. The new genus is similar to both Tylaphelenchus and Pseudaphelenchus in the possession of a raised cephalic region and a small spherical metacorpus, placing it in the subfamily Tylaphelenchinae. It is also typologically similar to Aphelenchoides. Molecular phylogenetic studies using both SSU and LSU rRNA gene sequences (both partial) revealed that the new genus formed a robustly supported sister clade with Pseudaphelenchus spp. and had a basal placement to this clade and, probably, to all other aphelenchoidids. Tylaphelenchus grosmannae is transferred to the new genus as B. grosmannae n. comb. and A. christinae n. comb. (= T. christinae) is proposed. The taxonomic status of Tylaphelenchus is discussed and T. sinodendroni n. comb. (= A. sinodendroni) proposed.

Labrys filiformis n. sp., the second species of the rare genus Labrys, was recovered from natural forests of Gilan province and is described based upon morphological and molecular characters. The new species is characterized by its smooth cuticle under light microscopy, lateral field with two incisures forming a single plain band, lip region continuous with body contour, dorso-ventrally flattened and forming four poorly prominent lobes, having a dorso-ventrally narrower protuberant labial plate laterally extended to the amphidial margins, oral area (oral plate) dorso-ventrally elongated and embedded in the labial plate with six small labial sensilla surrounding the slightly prominent oral aperture, amphidial apertures as longitudinally lemniscatic slits bordered by the labial plate extensions which are overlapped at the middle length of amphids, stylet delicate, 6 to 7 µm long, elongate weakly developed fusiform median bulb with weak valve, wide excretory pore with long and heavily sclerotized duct, offset spermatheca filled with small spheroid sperm cells, 106 to 127 µm long elongate-conoid tail with filiform distal region and finely rounded tip. Molecular phylogenetic analyses were performed using a near-full length fragment of the 18S rDNA and the D2–D3 expansion segments of the 28S rDNA using Bayesian inference and maximum likelihood methods. In the inferred phylogenetic tree with 18S rDNA, the new species has a close affinity with several isolates of the type species, Labrys chinensis. The reconstructed phylogenetic tree using partial 28S rDNA, revealed the new species is nested inside the putative monophyletic group of several populations of L. chinensis.

Summary The genus Sakia is recorded from Iran for the first time and Sakia arboris n. sp., recovered from two geographical points in northern Iran, is described. The new species is characterised by its smooth cuticle under light microscopy (LM), absence of lateral fields, head continuous with body contour, flattened lip region, 9.3 (9.0-10.0) μm long stylet, vulva at 60.2 (59.3-61.3)%, bilobed spermatheca, 117 (102-128) μm long tail, and 14.3 (14-15) μm long spicules. By lacking lateral fields, the new species comes close to three known species of the genus, namely: S. alii, S. castori and S. indica. Molecular phylogenetic analyses using sequences of the near full-length fragment of the small subunit of ribosomal DNA (18S rDNA) and the D2-D3 expansion segments of the large subunit ribosomal DNA (28S rDNA) were performed using Bayesian inference (BI) and maximum likelihood (ML) methods. In the reconstructed Bayesian tree using the 18S rDNA sequence of the type population, the new species occupied a position in a clade including two isolates of Sakia sp. and some species of Lelenchus, with maximal BPP and high ML BS values (1.00/99%). In the reconstructed 28S rDNA phylogenetic tree, two newly sequenced isolates of S. arboris n. sp. formed a well-supported clade with Lelenchus spp.

Aphelenchoides primadentusn. sp. is described and illustrated based on its morphological, morphometric and molecular characters. It was recovered from natural forests of northern Iran in association with mosses. The new species is characterised by its body length of 502-613μm in females and 461-564μm in males, presence of three lines in the lateral fields, a low lip region slightly offset by a shallow depression, 11.1-13.8μm long stylet in females, its conus being longer than the shaft (m = 53.0-68.5) and bearing well-developed, rounded basal knobs, the excretory pore located atcaone body diam. posterior to the metacorpus, a conical tail with a multipapillate terminus or with a single, warty, wide mucron-like structure at the tip, and males with 19-22μm long arcuate spicules that lack a well-developed condylus and rostrum. The new species was morphologically compared with species of the genus having three lines in the lateral field and a multipapillate tail tip or tail with warty mucron, namelyA. iranicusandA. heidelbergi. Compared toA. ensete,A. huntensisandA. gorganensis, three species with a multipapillate tail tip or warty mucron at the tail tip, the new species has a basic difference in the number of lines in the lateral field and, compared to two species,A. composticolaandA. petersi, which have three lines in the lateral field and a similar overall morphology, the new species has basic differences in tail tip morphology. The results of phylogenetic analyses using partial sequences of ribosomal small subunit RNA (SSU) and mitochondrial cytochrome c oxidase 1 (COImtDNA) genes revealed there is currently no sequenced species of the genus phylogenetically close to the new species being sequenced for these two genomic and non-genomic regions. The new species forms a clade withA. ritzemabosiin SSU, and three other species,A. besseyi,A. ritzemabosiandA. fujianensis, in theCOItree.
