Melanoma antigen recognized by T cells 1 (MART-1) and tyrosinase-related protein-2 (TRP-2) are two useful markers for immunohistochemical detection of melanocytic tumors. However, these markers may be passively acquired (phagocytosed) rather than actively synthesized. Reverse transcriptase in situ polymerase chain reaction (RT in situ PCR) can amplify even small amounts of specific mRNA in cells and therefore confirm the cellular source of a marker. We developed a one-step RT in situ PCR procedure in which Thermus thermophilus DNA polymerase synthesizes and amplifies cDNA from mRNA in a single reaction mixture. To examine its practicability and feasibility with formalin-fixed, paraffin-embedded (FFPE) tissue, we compared the results of one-step RT in situ PCR with those of immunohistochemistry (IHC). MART-1 mRNA was identified in the cytoplasm of lesional cells from 23/26 primary melanomas (92%), 9/9 metastatic melanomas (100%) and 5/6 nevi (83%). MART-1 epitope was detected by IHC in 23/24 primary melanomas (96%), 9/9 metastatic melanomas (100%) and 5/6 nevi (83%). TRP-2 mRNA was identified in the cytoplasm of lesional cells from 17/26 primary melanomas (65%), 6/9 metastatic melanomas (67%) and 4/6 nevi (67%). TRP-2 epitope was detected by IHC in 20/24 primary melanomas (83%), 9/9 metastatic melanomas (100%) and 4/6 nevi (67%). Both techniques detected MART-1 and TRP-2 in FFPE melanoma cell lines. Neither marker was detected in squamous cell carcinomas or basal cell carcinomas by RT in situ PCR or IHC. We conclude that the RT in situ PCR technique can be successfully applied to FFPE tissue to determine the cellular sources of gene expression observed by conventional PCR approaches.

Berichtet wird uber eine 42-jahrige Patientin mit einer Kortikoidkontaktdermatitis. Die Patientin erhielt aufgrund eines jahrzehntelang bestehenden endogenen Ekzems entsprechend haufig externe Steroide. Nach einer intraartikularen Kortisonapplikation (Predni H-injekt 10®, Prednisolonacetat) entwickelte sie ausgepragte Rotung und Odeme im Bereich der Injektionsstelle. Die allergologische Testung erbrachte eine Typ-IV-Sensibilisierung gegenuber der Gruppe D2 der Glukokortikoide (keine Methylsubstitution an C16, Ester-Gruppe an C17, Seitenkette an C21 moglich; eingeteilt nach der neuesten Klassifikation). Die Inzidenz von Kontaktallergien gegenuber Glukokortikoiden betragt in Deutschland zwischen 1,1% und 3,1%.

Die Helicobacter-pylori-Infektion wird immer haufiger als Ursache einer Reihe von dermatologischen Erkrankungen, u. a. auch der chronischen Urtikaria, diskutiert. Um diesen Zusammenhang zu untersuchen, wurde bei 55 Patienten mit chronischer Urtikaria eine Befragung bezuglich der Intensitat ihrer Erkrankung ca. 10 (Zeitpunkt I, Median) und 16 Monate (Zeitpunkt II) nach stationarem Aufenthalt durchgefuhrt und somit die Abheilungsrate bestimmt. Es zeigte sich kein Zusammenhang zwischen der Abheilungsrate der Urtikaria und der Helicobacter-pylori-Besiedlung (χ2-Test). Ebenso zeigte sich keine statistische Signifikanz in der Abheilungsrate von Urtikariapatienten nach Tripel-Therapie gegenuber nichttherapierten Patienten (exakter Fisher-Test).

Uber einen Zeitraum von 16 Monaten, von Februar 1992 bis Ende Mai 1993, wurden in der Stadt Ansbach/Bayern (40.000 Einwohner) in Zusammenarbeit mit den niedergelassenen Dermatologen, Padiatern sowie Allgemein- bzw. Praktischen Arzten alle vom Varizella-Zoster-Virus ausgelosten Erkrankungen registriert. Aufgrund klinischer Befunde wurde bei 152 Patienten ein Zoster und bei 437 Patienten eine Varizellen-Erkrankung diagnostiziert. Fur den Zoster errechnete sich eine bevolkerungsbezogene Inzidenz von 22,6 Erkrankungen pro 10.000 Einwohner und Jahr, wahrend die Varizellen-Inzidenz 42,4 betrug. Bei Zoster zeigte sich eine leichte Bevorzugung des mannlichen Geschlechtes und eine deutliche Abhangigkeit der Lokalisation der befallenen Nervensegmente vom Alter und dem Geschlecht. Zoster trat in jedem Lebensalter auf, bei einer deutlich hoheren Erkrankungsrate alterer Menschen. Der Erkrankungsgipfel lag in unseren Fallen in der 8. Lebensdekade. Bei den Varizellen war die Geschlechtsverteilung vollig ausgeglichen. Als typische Kinderkrankheit wurden ganz uberwiegend Patienten der ersten Lebensdekade befallen, wahrend Varizellen bei alteren Menschen nur noch sporadisch vorkamen. Im Jahresverlauf trat die Zostererkrankung relativ gleichmasig zu allen Jahreszeiten auf. Bei Varizellen waren jedoch Erkrankungsgipfel zu beobachten. Moglicherweise waren Beginn einer Erkrankungswelle und deren Ende von meteorologischen Daten abhangig.

Down-regulation of the immune system facilitates tumor progression at different stages of cutaneous melanoma. Sentinel nodes, the first lymph nodes on lymphatics draining directly from a primary melanoma are immune down-regulated by tumor-generated immune-suppressive cytokines including interleukin-10. To better understand the kinetics of sentinel node suppression, we investigated interleukin-10 expression by melanoma cells and tumor-associated macrophages and lymphocytes at different stages of primary melanoma evolution. We used reverse transcriptase in situ polymerase chain reaction to identify cellular sources of interleukin-10 mRNA in 39 melanomas. Interleukin-10 mRNA was identified in tumor cells of 2/6 melanomas in situ (33%), 17/21 invasive melanomas (81%) and 11/12 metastatic melanomas (92%). Higher interleukin-10 expression correlates with tumor progression, with differences between melanoma in situ, invasive melanoma and metastatic melanoma. In primary melanomas, the interleukin-10 mRNA content of tumor cells correlates with Clark level. There was significantly more interleukin-10 mRNA in vertical growth phase melanoma cells than in radial growth phase cells. In a logistic regression model, moderate to high interleukin-10 mRNA expression by tumor cells was significantly associated with vertical growth phase melanoma. Interleukin-10 mRNA was detected in melanoma-associated macrophages and lymphocytes. In invasive melanomas, the interleukin-10 mRNA reactivity of macrophages declined as Clark level increased. Alterations of immunity by interleukin-10 derived from melanoma cells and melanoma-associated macrophages and lymphocytes potentially facilitate evolution of the primary melanoma and render regional lymph nodes susceptible to metastases.

It has been debated whether malignant transformation of trichoblastoma occurs. The concept was recently forwarded that basal cell carcinoma is as a malignant neoplasm of follicular germinative cells and should be named trichoblastic carcinoma to show its relationship to trichoblastoma. Almost all basal cell carcinomas are low-grade malignant neoplasms and develop metastases only very rarely, and if so, only after very long duration and untreated growth. Only rare basal cell carcinomas arise in trichoblastomas. Up to now there have only been two reports of high-grade trichoblastic carcinoma arising in trichoblastoma, showing systemic metastatic spread and death. We add two further cases of trichoblastic carcinoma with anaplastic nuclei, arising in trichoblastoma. One of the tumors arose in a small nodular trichoblastoma on the right forearm of an 84-year-old male patient. The other one was a trichoblastic carcinoma at the base of a trichoepithelioma on the right thigh of an 87-year-old woman with Brooke-Spiegler syndrome. Our cases emphasize that high-grade trichoblastic carcinoma develops via malignant transformation of trichoblastoma, and is very rare.

A 39-year-old female patient presented with an intense allergic reaction and shock after ingesting sunflower seeds and simultaneously acetylsalicylic acid (ASA). Skin tests and CAP specific. IgE demonstrated an IgE-mediated sensibilization to sunflower seeds. When sunflower seeds were eaten alone, only discrete paresthesia of the oral mucosa occurred. Surprisingly, an oral challenge with ASA was well tolerated. The supplementary contribution of ASA to the allergic reaction was dose-dependent. The quantity of the allergen also modified the intensity of the symptoms. This surprising effect of ASA may be attributed to increased gastric resorption of the allergens. During the course of this reaction, eosinophil-cationic-protein was released.

Intralesional injection of beta-interferon can lead to remission of metastatic melanomas. Simultaneous radiotherapy may augment the effect of these injections. The effect of intralesional beta-interferon injections combined with radiotherapy was examined in patients with metastatic malignant melanoma. A total of 20 patients with inoperable, incapacitating and disfiguring malignant melanoma metastases were treated with simultaneous external beam radiotherapy (electrons and/or photons) and intralesional injection of beta-interferon. The total radiation dosage ranged from 40-50 Gy, fractionated as 1.8 Gy five times per week. beta-Interferon (Fiblaferon) 3-5 million units per injection was administered three times weekly until symptoms disappeared or until termination of therapy. One patient was treated with beta-interferon alone; the area harbouring the tumour had been previously irradiated. Five patients treated in this way showed partial remission after combined therapy and 12 showed complete remission, with permanent regression of metastases. The latter group included five patients with lengthy survival times, including one patient who has been free of symptoms for 7 years following treatment. Injected metastases showed complete regression in the patient who was treated exclusively with beta-interferon post-irradiation. In conclusion, combined treatment with radiotherapy and intratumoral injection of beta-interferon controlled local tumour growth in inoperable metastatic malignant melanomas.

New surgical oncology techniques, such as lymphatic mapping and sentinel node biopsy, require precise identification of the presence of even very small numbers of tumor cells. The gold standard for such analysis remains microscopic assessment of tissue sections, stained conventionally or by immunohistochemistry for appropriate tumor markers. This approach is limited by sampling constraints and requires a high degree of expertise from the microscopist. Recent studies have demonstrated a subgroup of patients whose sentinel nodes are negative on microscopy, but whose nodes yield an enhanced signal for melanoma markers when evaluated by RT-PCR. These enhanced signals reflect a mixture of signal sources, including small numbers of melanoma cells and cells other than melanoma cells that express the relevant markers(s). Because the preparative techniques for RT-PCR destroy the structural integrity of the tissues and disrupt individual cells, the exact cellular source of enhanced signal from a tissue cannot be demonstrated by conventional RT-PCR. RT in situ PCR, in which the RT-PCR technique is applied on a tissue section, does identify the cells that are the source of signal. We have attempted to optimize this interesting approach and have applied it to the detection of relevant melanoma markers in tissue culture lines.

A case of eosinophilic granuloma faciale is presented in which the lesions have been successfully treated by cryotherapy, healing with slight pigmentation. Since this disease appears to be difficult to treat, a great many forms of therapy have been proposed in the literature. These will be discussed with the results presented.

21. Katamnestische Untersuchungen an Patienten mit chronischer Urtikaria und ASS-Intoleranz

Of a total of 131 patients suffering from chronic urticaria, 15 were cautiously reexposed to ASA after an initial provocative exposure during an urticaria test programme 2 – 11 years before. Only 1 of these patients, who had undergone the initial provocative test 7 years earlier, reacted at the same intensity; 1 other patient reacted with much less intense symptoms 4 years after the original test. Among 3 other patients, who merely reacted to ASA intake with urticarial eruptions and did not suffer from chronic urticaria, only 1 presented 4 years after the initial exposure with oedema of the skin and itching. The tolerance threshold was markedly higher. These results suggest that the sensitivity to intolerance-inducing agents is reduced relatively quickly and may subside completely in most cases.

Diagnose und sekundare Pravention von Nahrungsmitteluberempfindlichkeiten gehoren zu den schwierigsten allergologischen Aufgaben. Dabei wurden in den letzten Jahren auf dem Gebiet der Nahrungsmitteluberempfindlichkeit sehr bedeutsame Fortschritte erzielt. So ist es inzwischen gesichertes Wissen, dass nicht nur allergische Reaktionen vom Soforttyp oder gastrointestinale Symptome durch eine Nahrungsmitteluberempfindlichkeit ausgelwerden, sondern auch atopisches Ekzem oder kontaktallergisches Ekzem durch Nahrungsmittelinhaltsstoffe provoziert werden konnen. Erst in Ansatzen erkennbar sind die molekularen Pathomechanismen, die diesen Reaktionen zugrunde liegen.

Fruher herrschte die irrige Vorstellung, das sich Melanome rasch entwickeln und — haben sie erst einmal gejuckt oder geblutet — der operativen Entfernung des Primartumors auch bald die Metastasen in den regionaren Lymphknoten oder in anderen Organen folgen. Wir wissen heute, das solche Verlaufe allenfalls nur bei verpaster Fruherkennung des Primartumors vorkommen und eher als Ausnahmen die „Regel“ bestatigen. Die wissenschaftlichen Untersuchungen der letzten Jahre haben diese Vorstellung vollig widerlegt und ein realistisches Bild der Tumorwachstumsdynamik ergeben. Das Wachstum der Melanome last sich namlich wie bei keinem anderen bosartigen Tumor des Menschen retrospektiv anhand von zufallig gemachten privaten Photos der Patienten und statistisch rechnerisch verfolgen. Diese einzigartige Moglichkeit der photokatamnestischen Studie von malignem Tumorwachstum besteht nur beim pigmentierten malignen Melanom der Haut [11, 12, 13, 14, 15].

We examined the frequency, distribution, and immunophenotype (S-100 protein, CD1) of epidermal Langerhans cells (LC) in the epidermis overlying primary melanoma, and dermal dendritic cells (DC) in the dermis deep to melanoma, and in adjacent normal skin. There is a substantial reduction in S-100+ LC and a lesser decline in CD1+ LC in the epidermis over melanoma. There is a simultaneous increase in the frequency of cells expressing these phenotypes in the dermis deep to tumor. Double-staining studies in progress show coexpression of S-100 and CD1 in most of these cells. The depletion of S-100+/CD1+ DC from the peritumoral epidermis is maximum in deeper (Clark level III-V) and thicker (> 0.76 mm) tumors. The reduction in total DC in peritumoral epidermis is, however, proportionally less than the reduction in S-100+/CD1+ DC owing to an increase in cells that are S-100-/CD1+. The proportion of S-100+/CD1+ dermal DC deep to tumor is similar to that in normal epidermis and dermis, suggesting that there is no increased migration of S-100-/CD1+ DC from tumor-associated epidermis to subjacent dermis. Non-dendritic leukocytes in the dermis deep to tumor were increased in frequency, maximally in the dermis deep to thinner and more superficial melanomas. Most such cells were T lymphocytes with helper/inducer cells predominating. The data presented show substantial alterations in the frequency, distribution, and phenotype of LC/DC as melanoma evolves, alterations that may be critical for development of effective tumor-directed immunity.

28. Efficiency of Acitretin in Combination With UV-B in the Treatment of Severe Psoriasis

• Compared with the antipsoriatic retinoid etretinate, the new aromatic retinoid acitretin represents an important advance due to its rapid elimination kinetics. Since in psoriasis vulgaris retinoids are used predominantly in combination regimens, we investigated the therapeutic efficacy of acitretin and UV-B compared with placebo and UV-B in a double-blind, randomized multicenter trial in 82 patients with severe psoriasis. They were treated with 35 mg of the study medication during the first 4 weeks of therapy and 25 mg thereafter, concomitantly with UV-B irradiation in increasing energy doses. Forty patients who underwent therapy with acitretin and UV-B and 38 patients who underwent therapy with placebo and UV-B were evaluated for efficacy. The target variables—psoriasis severity index and total UV-B dose—were reported at intervals of 2 weeks over a maximum period of 8 weeks. At the end of treatment, the psoriasis severity index decrease was 79% in the acitretin and UV-B group and 35% in the placebo and UV-B group. The response rate, defined as greater than or equal to a 75% decrease of the psoriasis severity index, was 60% for the combination treatment and only 24% for the control treatment. This treatment response was achieved with markedly lower cumulative UV-B energy. The median cumulative UV-B energy applied to reach 75% clinical improvement was 11.8 J/cm 2 vs 6.9 J/ cm 2 . Side effects showed a similar pattern in both groups. Our data show that the acitretin dramatically improves the results of UV-B treatment in patients with severe psoriasis. In addition, it markedly decreases the effective cumulative UV-B dose, thereby reducing the potential long-term hazards of UV irradiation. We conclude that the acitretin plus UV-B combination treatment represents a highly effective therapeutic regimen in severe psoriasis. ( Arch Dermatol . 1990;126:482-486)

Vierzig papillomatose Naevuszellnaevi, grostenteils noch mit junktionaler Aktivitat, wurden im Hautniveau so abgetragen, das am Rande ein schmaler Restnaevussaum stehenblieb und die tiefen Naevuszellpartien den Wundgrund bildeten. Nach erfolgter Wundheilung trennte eine dunne Schicht Narbengewebe die neu gebildete Epidermis von den in der Dermis verbliebenen Restnaevuszellen. Nach einem sehr unterschiedlichen Zeitraum, fruhestens jedoch 30 Tage post operationem, konnten in der Epidermis des Narbenbereiches typische Veranderungen beobachtet werden. Am haufigsten kam es zu einer Hyperpigmentierung mit einer Vermehrung basaler, fluorescierender Dendritenzellen, die auch auf die oberen Abschnitte der Hautanhangsgebilde ubergriffen. Auserdem fanden sich nestformige Zusammenballungen pigmentbildender Zellen zu junktionalen Naevuszellnestern. Etwa 2–3 Monate nach der traumatischen Irritation waren in einigen Fallen “Abtropfungsvorgange” im Gange, die eine Naevusregeneration anzeigten. Fluorescenzhistochemische und enzymhistochemische Befunde sprechen dafur, das starke Veranderungen nicht nur im neu gebildeten Epithel der narbe, sondern auch in den morphologisch wenig veranderten Restnaevuszellen am Rande der Narbe und unterhalb des Narbengewebes vor sich gehen.

With fluorescence-histochemical methods (formalin-induced fluorescence), the different stadia of development of lentigo maligna (Morbus Dubreuih) and lentigo maligna melanoma were investigated. In this way, the special stadia can be clearly characterized using the fluorescence-microscope. In the earliest stadium of malignancy, only the pigment-cells of the basal part of the epidermis seem to be numerous. At this time, these cells are mainly arranged as palisades in the basal layer, and their strongest dendrites are usually directed towards the corneal layer. As the malignancy progresses, the pigment-cells are arranged in several layers in the basal epidermis. In this case, polymorphism of the cells is obvious and their dendrites spread in all directions. A further stadium shows pathological alterations resembling a lentigo with long rete ridges. The atypical cells cluster together into the so-called pseudonests, predominantly at the tips of these rete ridges. With the fluorescence-microscope, dendrites are seldom visible here. In the stadium of tumorous growth, all above-described alterations of the epidermis in lentigo maligna are no longer detectable. Malignant cells in the dermis, which form as a tumor mode, seldom show dendrites. These cells are mainly round or oval-shaped; at best there are some spindle cells in certain areas of the tumor. The epidermis covering the tumor node is infiltrated by some tumor cells but the characteristic alterations as described above for the various stadia of lentigo maligna are no longer visible. Even fluorescence-microscopically, a tumor node of lentigo maligna does not seem to be different from a primary nodular melanoma or a tumor node of a superficial spreading melanoma, if the flat parts of the tumors are not considered in the diagnosis.

• Series of "private" photographs taken of patients over many years can greatly aid in following the course of development of malignant melanomas. In one case of superficial spreading melanoma, the pigmented lesion displayed spread around the entire circumference, maintaining its round shape throughout its course. The other tumors enlarged by focused spreading at various points along the border, changing in shape over several years of development. The temporary appearance of two closely adjacent pigmented lesions frequently occurred during the early development of malignant melanomas, followed by fusion after progressive spread. Lentigo maligna melanoma showed the greatest continuous alteration of shape. Different tumor types observed over similar periods of time showed different dynamics of growth with reference to shape (horizontal spreading growth) and depth of invasion (vertical growth). Mit neurohistologischen Techniken (Nauta-Verfahren) wurde der Anteil des Stirnorgans von Rana temporaria und Rana esculenta an der zentralnervosen Projektion des lichtempfindlichen Pinealkomplexes gepruft. Nach operativer Unterbrechung des Nervus pinealis im dorsalen Lymphsack lassen sich degenerierende Nervenfasern sowohl im Tractus pinealis als auch im stirnorgannahen Stumpf des Nervus pinealis nachweisen. Die ersteren werden als cerebropetale (afferente), die letzteren als zum Stirnorgan ziehende (efferente) Faserelemente gedeutet. Es ist gelungen, die hirnwarts gerichteten Nervenfasern des Stirnorgans bis in die unmittelbare Umgebung des sekretorischen Subcommissuralorgans zu verfolgen; zerfallende Faserfragmente liegen dicht der Basis des Subcommissuralorgans an. Anders als nach Durchtrennung des Tractus pinealis (vgl. Paul, Hartwig und Oksche, 1971) liesen sich nach Unterbrechung des Nervus pinealis keine Degenerationszeichen im mesencephalen „Zentralen Grau“ darstellen.

Die Epithelzellen derPlexus chorioidei ventr. III undIV vonRana temporaria L. sind sehr glykogenreich. DiesesGlykogen hat offenbar den Charakter eines innerhalb des Gehirns gelegenen Kohlenhydratdepots. Die Glykogenpartikeln des Plexusepithels (Durchmesser etwa 300 A) bilden Rosetten oder Aggregate von einer weniger regelmasigen Beschaffenheit als die α-Partikeln, die in der Leberzelle beschrieben worden sind. Nach Verabfolgung vonAdrenalin ist mit histochemischer, elektronenmikroskopischer und quantitativer Methodik eine Abnahme des Plexusglykogens zu beobachten. Die f unktionelle Bedeutung des Glykogendepots im Anurengehirn wird auf der Basis dieser Befunde diskutiert. Weiterhin werden einige Aspekte des Stoffwechsels und der Gefasversorgung der Plexus chorioidei vonRana temporaria, besprochen. Die Ultrastruktur des Glykogens im Plexusepithel wird unter normalen und Versuchsbedingungen mit dem elektronenmikroskopischen Bild des Leberglykogens verglichen.

Der Einflus des lichtempfindlichen Pinealkomplexes (Stirnorgan und Epiphysis cerebri) von Rana temporaria auf dorsale diencephale und mesencephale Hirnzentren breitet sich uber ein groseres Gebiet aus als man bisher angenommen hat. Nach operativer Unterbrechung des Nervus- und Tractus pinealis wurde mit Punktzahlverfahren die Dichte der Zell- und Zellkernvolumina sowie der Kapillaroberflache im dorsalen Thalamus, mesencephalen (postthalamischen) „zentralen Grau“, Ganglion habenulae, Tectum opticum, in der Area praetectalis und im Subcommissuralorgan (Sco) bestimmt. Die morphometrischen Mesergebnisse lassen sich wie folgt interpretieren: 1. Der Pinealkomplex wirkt auf das Sco, das „zentrale Grau“ und die Area praetectalis ein (vgl. Paul u. Mitarb., 1971); das Sco wird in erster Linie uber das Stirnorgan beeinflust. 2. Weitere Kerngebiete des Zwischenhirns (Ganglion habenulae und dorsaler Thalamus) gehoren funktionell zu dem vom Pinealkomplex kontrollierten System. 3. Die von Kemali und Braitenberg (1970) beschriebene Asymmetrie der Ganglia habenularum zeigt sich auch im funktionellen Zusammenspiel dieser Kerngebiete mit dem Pinealkomplex, dem dorsalen Thalamus und dem dorsalen Abschnitt des „zentralen Graus“. Die in Stirnorgan und Epiphysis cerebri transformierten Lichtreize werden in Hirnabschnitte weitergeleitet, die olfaktorische und retinale Signalmuster verarbeiten. Uber die biologische Bedeutung der nervosen Beziehungen des Stirnorgans zum sekretorischen Sco konnen noch keine Angaben gemacht werden.

Die Epithelzellen der Plexus chorioidei ventriculi III und IV und der Paraphyse von Rana temporaria L. zeigen histochemisch eine deutliche Aktivitat der Phosphorylase, Glucose-6-Phosphat-Dehydrogenase und Aldolase. Die Aktivitat der Uridindiphosphoglucose-Glycogen-Transferase ist im Plexusepithel der Frosche gering. Glucose-6-Phosphatase last sich in den Plexus chorioidei und in der Paraphyse von Rana temporaria nicht darstellen. Phosphorylase, Glucose-6-Phosphat-Dehydrogenase und Aldolase zeichnen sich durch ein charakteristisches, verschiedenartiges Verteilungsmuster aus. Der Einflus von Adrenalininjektionen auf die Aktivitat der obengenannten Enzyme des Plexus-und Paraphysenepithels und funktionelle Zusammenhange mit der gleichzeitig eintretenden Entspeicherung ihres Glykogenvorrats werden erortert. In fluoreszenzmikroskopischen Untersuchungen mit der Falck-Hillarp Methode last sich weiterhin beobachten, das nach Adrenalingaben im Plexusepithel der Frosche ein Fluorophor auftritt, das bei Kontrolltieren vollstandig fehlt. Im Plexusstroma finden sich nur vereinzelt adrenerge Nervenfasern.

Das Problem der Zwischenneurone nimmt bei der Deutung der chromatischen und achromatischen Antworten der pinealen Sinnesorgane eine Schlusselstellung ein. Fruhere Ergebnisse schienen darauf hinzuweisen, das der nervose Apparat des Pinealorgans (Epiphysis cerebri) vonRana temporaria undRana esculenta bineuronal organisiert ist. Mit modifizierten Methylenblau- und Golgi-Methoden gelang es jetzt, die nervosen Strukturen der pinealen Rezeptoren und Nervenzellen praziser darzustellen. Das neurohistologische Bild der Epiphysis cerebri enthalt auch Nervenzellen, die an Zwischenneurone denken lassen. Diese diffus verstreuten kleinen Elemente unterscheiden sich von den klassischen Typen der retinalen Bipolar-, Horizontal- und Amakrinzellen. Der Tractus pinealis vonR. temporaria undR. esculenta wird von Axonen groser multipolarer und kleiner, wenig verzweigter Ganglienzellen gebildet. Die zentrale Projektion dieser pinealofugalen (afferenten) Bahn ist von grosem funktionellem Interesse. Nach vollstandiger Unterbrechung des Tr. pinealis finden sich degenerierende Faserelemente innerhalb und am unteren Rand der Comm. posterior, in der Area praetectalis und in den Kernarealen des sog. „Zentralen Graus“. Keine degenerierenden Tractusfasern sind in der subependymalen Schicht und in den aminergen Kerngebieten des Mesencephalon zu beobachten. Die beschriebenen Verbindungen des Tr. pinealis konnten die anatomische Basis einiger lichtabhangiger (phototaktischer) Reflexe darstellen. Das Gehirn adulter Grasfrosche (Rana temporaria L.) zeigt ein charakteristisches regionales Verteilungsbild seiner Ependym- und Gliazellen. Es wurden die folgenden Ependym- und Gliatypen identifiziert: unipolare und bipolare ependymale Tanycyten, Furchenependym, freie Tanycytenformen, regressiv modifiziertes Ependym und Matrix-ependym. Sternformig verzweigte astrocytare Elemente sind auserst selten. Aufmerksamkeit verdienen die im Vordergund stehenden ependymalen Tanycyten, die eigenartige Kontakte mit den Blutgefasen und Nervenzellen bilden. Die regionale Verteilung der verschiedenen Ependymtypen wird in vier Karten dargestellt. Diese Karten sind eine Basis fur weitere histochemische Studien und Modellversuche zur Frage der trophischen Rolle und der Stoffwechselfunktion der Neuroglia.

Die Epithelzellen der Plexus chor. ventr. III und IV, das Paraphysenepithel und das Ventrikelependym von Rana temporaria L. zeigen eine deutliche — wenn auch quantitativ unterschiedliche — Aktivitat von SDH, β-HBDH und LDH. Der Nachweis einer UDPGGT-Aktivitat fallt dagegen nur im Plexusepithel positiv aus. In der Plexusepithelzelle sind die Formazangranula gleichmasig uber das gesamte Plasma verteilt, wahrend sie in den Ependymzellen vorwiegend im apikalen, lumenwartigen Teil der Zelle angeordnet sind. Die Ependymzellen lassen im Reaktionsausfall Typenunterschiede erkennen. Bei einigen Tieren wird die Auswirkung einer Adrenalininjektion auf das Enzymmuster der Plexusepithelzellen untersucht; die funktionellen Zusammenhange mit einer gleichzeitigen Entspeicherung ihres Glykogenvorrats werden erortert. Die Verteilung und der Lamellierungstyp der Mitochondrien im Plexus- und Paraphysenepithel werden mit elektronenmikroskopischen Aufnahmen illustriert. Einige Komentare sind dem histochemischen Verhalten des Wundrandes gewidmet.

Mit der Methode von Falck-Hillarp lassen sich im Ruckenmark der Vogel Bundel fluoreszierender Nervenfasern nachweisen, die in Hohe des Glykogenkorpers (Lumbalwulst) seitlich an dieses Organ grenzen, von wo aus zahlreiche Fasern in den Glykogenkorper eindringen. Vergleichend-anatomisch gehoren die fluoreszierenden Areale des Ruckenmarks zu den autonomen Zentren. Es wurde versucht, mit Pharmaka, die das autonome Nervensystem beeinflussen oder Krampfe erzeugen, das Glykogen des Lumbalwulstes zu mobilisieren (Quantitative Glykogenbestimmung nach Krisman, 1962). Besondere Beachtung verdient die elektronenmikroskopische Feinstruktur der Zwickelraume zwischen den grosen Glykogenkorperzellen, wo Auslaufer dieser Zellen und Nervenfasern eng beieinander liegen. Die Kapillaren des Glykogenkorpers werden fast vollstandig von Auslaufern seiner Zellen umscheidet.

Mit Versilberung und mit der fluoreszenzmikroskopischen Technik von Falck-Hillarp wurden im Glykogenkorper des Vogelruckenmarks Nervenfasern dargestellt. Diese Nervenfasern stammen aus Kerngebieten, die den Glykogenkorper flankieren. Die starkste Fluoreszenz dieser lumbalen Kernareale findet sich an den beiden Polen des Glykogenkorpers. Es wird angenommen, das das beschriebene aminerge Neuronensystem einen funktionellen Einflus auf den Glykogenkorper ausubt. Der Glykogenkorper der Vogel wird mit anderen zentralnervosen Glykogendepots verglichen.

Embryonic skin and eyes, and melanomas of xiphophorine fish were investigated by formaldehyde-induced fluorescence in order to test whether the pigment cells in these tissues may be identified by a specific green- yellow fluorescence. Skin of pigmented fish embryos showed no fluorescence in the black pigment cells (melanocytes and melanophores), while skin of albino embryos showed a green-yellow fluorescence in all cells which correspond to the black pigment cells of pigmented embryos. The skin of both pigmented and albino embryos showed a bright orange fluorescence in the red pigment cells (pterinophores). No fluorescence was observed in the retinal pigment epithelium of pigmented embryos, while a green-yellow fluorescence was observed in the pigment epithelium of albino embryos. Neither the melanotic melanomas of pigmented fish nor the amelanotic melanomas of albino fish showed any specific fluorescence.

The regeneration of nevus cells after shave biopsy was investigated by means of fluorescence-histochemical and electron microscopic methods. A prerequisite for the regeneration of pigmented nevi seems to be the retention of their deepest part. It appears that the remaining nevus cells primarily induce proliferation of pigment-producing dendritic cells in the epidermis. These dendritic cells form nest-like conglomerations at the dermal-epidermal junction, which eventually separate from the epidermis during a process called "dropping off". The fluorescence-histochemical and electron-microscopic results lead to the conclusion that under these circumstances melanocytes are transformed into nevus cells.

Fifty-two congenital melanocytic nevi (CMN) as precursors to melanoma or severe melanocytic dysplasia were reviewed macroscopically and microscopically. Forty-eight "small" CMN measured less than 10 cm in diameter. Histologically, only five reached to the lower third of the dermis or subcutis ("deep type"); the remaining 47 were limited to the upper two thirds of the corium ("superficial type"). There were 47 invasive melanomas, two in situ melanomas, and three severe focal melanocytic dysplasias. All melanomas were of "epidermal" origin and primarily of the superficial spreading type. The age at diagnosis ranged from 18 to 79 years. Prepubertal melanomas were not observed. Melanoma may also arise on small CMN. The most frequent origin of the melanomas in small CMN and the preferred age at manifestation seem to be different from that for giant nevi.
