Apitherapy is defined as “the use of Apis mellifera L. products such as royal jelly, pollen, honey, propolis, beeswax, and bee venom in the treatment of ailments”. Although honey is the primary product acquired, other bee products are also obtained in Turkey. These commodities, in addition to being utilized as nutrition, have been employed to promote human health since ancient times owing to the biologically active compounds they contain. Bee venom is increasingly commonly used in apitherapy and has a wide range of biological effects including antiviral, antidiabetic, anticancer, antirheumatic, anticoagulant, antibacterial, anti-cancer, anti-aging, neuroprotective, analgesic, antioxidant, hepatoprotective, and anti-asthmatic properties. According to the literature, bee venom has promising biological implications for human health, which constitutes the topic of this review.

The approach proposed as the cholinergic hypothesis for Alzheimer's Disease contributed significantly to the explanation of cognitive decline observed in the elderly and Alzheimer's patients due to dysfunction of neurons containing acetylcholine in the brain. To date, this notion has formed the basis of most of the treatment strategies and drug development approaches for Alzheimer Disease. Within the scope of this study, in which the synthesis and biological evaluation of pyridazinones, which are designed for multi-enzyme targets and expected to act as AChE and BChE inhibitors, were performed, 12 new compounds were synthesized, their in vitro enzyme inhibitor activities were evaluated and molecular modeling studies were carried out. While the compounds did not show significant AChE inhibition, they showed a high degree of BChE inhibition. D2e was determined as the most potent BChE inhibitor and molecular modeling and molecular dynamics simulation studies were also carried out for D2e and tacrine. The obtained results suggest that new pyridazinone derivatives can act as BChE inhibitory agents. Although the synthesized compounds are less effective than the reference drugs, it has been concluded that it is possible to reach the precursor compounds as a result of suitable modifications.

The genera Geranium and Erodium belonging to the Geraniaceae family have been reportedto have various ethnopharmacological uses worldwide. In this study, the ethanol extractsobtained from Geranium and Erodium species were screened for their DPPH radicalscavenging and acetylcholinesterase enzyme inhibitory activity, which are associated withthe pathophysiology of Alzheimer's disease.Geranium and Erodium species were collectedthroughout Turkey. The antioxidant activity of the extracts prepared with 96% ethanol wasinvestigated by DPPH radical scavenging method and their in vitro inhibitory effect on AChEwere determined using ELISA microtiter assays. 24 extracts out of 47 extracts showedconcentration-dependent inhibitory effect towards AChE over 50% inhibition. The highestAChE inhibitory activity at 2000 µg/mL was found in G.macrostylum Boiss. collected fromAntalya 87.90 ± 2.15% (IC50= 4.73± 2.96 µg/mL). The inhibitory activity of galanthamine at1000 µg/mL, used as the reference drug, was found 97.11±1.26% (IC50= 0.68 ± 0.05µg/mL). All of extracts exhibited excellent antioxidant activity. The highest antioxidantactivity at 2000 µg/mL was found in G.purpureum Vill. collected from Antalya (85.80 ±0.26%). The radical scavenging activity of quercetin used as reference to compare in theantioxidant activity was found 85.51 ± 0.17 at 1000 µg/mL.Our findings revealed thatGeranium and Erodium species screened in this study display high radical scavengingactivity against DPPH and remarkable inhibitory activity against AChE.The phytochemicalanalysis of the active extracts to elucidate the compounds responsible for the mentionedactivities is in progress in our laboratory

Bu tez çalışmasında, Myrtaceae familyasından Myrtus communis L. bitkisinin, kültürü yapılan beyazmeyveli (Hambeles) ve siyah meyveli (Yakup) çeşitlerine ait bitki örneklerinin yaprak örnekleri 12 ayboyunca toplanmış, olgun meyve örnekleri de alınmıştır. Yaprak ve meyve örneklerinden hareketle etanolekstreleri hazırlanmış ve asetilkolinesteraz (AChE) ve bütirilkolinesterazdan (BChE) oluşan kolinesterazenzim ailesi ile tirozinaza (TYR) karşı in vitro inhibitör aktiviteleri yönünden ELISA yöntemiyle testedilmiştir. Polifenollerce zengin olduğu bilinen bitkinin 12 ay boyunca toplanan Hambeles ve Yakupçeşitlerinin yaprak ekstreleri ile iki çeşide ait meyve ekstrelerinin fenolik bileşiklerinin tespit edilmesiamacıyla sıvı kromatografisi-kütle spektrofotometresi (LC-MS) tekniği ile toplam 26 adet ekstre üzerindefitokimyasal analizler gerçekleştirilmiştir. Ekstrelerde toplam fenol ve flavonoit miktar tayini yapılmış,LC-Q-ToF-MS ile ekstrelerde ortak olan fenolik bileşiklerin; galloil glukoz izomeri 1 ve 2, kinik asitizomeri, galloil kinik asit izomeri 1, 2 ve 3, striktinin izomeri 1 ve 2, digalloil kinik asit izomeri,digalloil-hekzahidroksifenoil-glukoz izomeri 1 ve 2, trigalloil glukoz, mirisetin galloil hekzosit,mirisetin galaktozit, mirisetin galloil ramnopiranozit, mirisetin-3-O-arabinozit, mirisetin-3-Oramnozit,kemferol-O-galloil-hekzosit ve kersetin ramnozit olduğu tespit edilmiştir. En yüksekkolinesteraz inhibisyonuna yol açan Hambeles çeşidinin Ocak ayına ait yaprak ekstresinin IC50değeri 18,49 ± 3,50 g/mL olarak hesaplanmıştır. Referans olarak kullanılan galantamin, AChE veBChE’a karşı sırasıyla IC50 değerleri 1,33 ± 0,11 ve 37,69 ± 2,93 μg/mL olarak bulunmuştur.Ekstreler TRY’a karşı 7,14 ± 3,03 – 17,32 ± 1,98 μg/mL arasında olmak üzere düşük inhibisyonayol açmıştır. Ayrıca kolinesteraz enzim inhibitör aktivite deneylerimizde en yüksek inhibisyongösteren Hambeles çeşidinin Ocak ayında toplanan yaprak ekstresinin SHSY-5Y hücrelerinde 6-OHDA aracılığıyla oluşturulan nörotoksisiteye karşı etkisi incelenmiştir. Buna göre; hücre canlılıkanalizleri ile uyumlu olarak en yüksek sitotoksik değerin 24 saat 100 μM 6-OHDA’nın önuygulamasının ardından 24 saat süre ile uygulanan ekstrenin 100 μg/mL konsantrasyonunda olduğugözlenmiştir. Bu çalışma, ülkemizde kültürü yapılan M. communis’e ait yaprak ve meyve ekstrelerininkolinesteraz ve TYR enzim inhibisyonu ve hücre kültürü yöntemleri ile nöroprotektif etkisi ile fenolikbileşenleri açısından fitokimyasının yıllık bazdaki değişimlerinin incelendiği ilk çalışmayı teşkiletmektedir.Anahtar Kelimeler : Myrtus communis, Nöroprotektif Etki, Enzim İnhibisyonu, AlzheimerHastalığı, Hücre Kültürü, LC-Q-ToF-MS In the current thesis study, the leaf samples of the plant samples belonging to the cultivated whitefruit (Hambeles) and black fruit (Yakup) cultivars of Myrtus communis L. from the Myrtaceae familywere collected during 12 months and the mature fruit samples were also taken. The ethanol extractswere prepared from the leaf and fruit samples and tested by ELISA method for their in vitro inhibitoryactivity against cholinesterase enzyme family consisting of acetylcholinesterase (AChE) andbutyrylcholinesterase (BChE) as well as tyrosinase (TYR). In order to determine the phenoliccompounds of the leaf extracts of Hambeles and Yakup varieties and the fruit extracts of these twovarieties, which are known to be rich in polyphenols, collected during 12 months, phytochemicalanalyzes were performed on a total of 26 extracts using liquid chromatography-massspectrophotometer (LC-MS) technique. Total phenol and flavonoid amounts were determined in theextracts, while the phenolic compounds, e.g. galloyl glucose isomers 1 and 2, quinic acid isomer,galloyl quinic acid isomers 1, 2 and 3, strictin isomers 1 and 2, digalloyl quinic acid isomer, digalloylhexahydroxyphenol-glucose isomers 1 and 2, trigalloyl glucose, myricetin galloyl hexoside,myricetin galactoside, myricetin galloyl rhamnopyranoside, myricetin-3-O-arabinoside, myricetin-3-O-rhamnoside, kaempferol-O-galloyl-hexoside, and quercetin rhamnoside were determined toexist commonly in the extracts by LC-Q-ToF-MS. IC50 value of the leaf extract of the Hambelesvariety from January collection, which caused the highest cholinesterase inhibition, was calculatedas 18.49 ± 3.50 g/mL. IC50 values of galanthamine used as reference against AChE and BChE were1.33 ± 0.11 and 37.69 ± 2.93μg/mL, respectively. The extracts caused low inhibition against TRY,ranging from 7.14 ± 3.03 to 17.32 ± 1.98 μg/mL. In addition, in our cholinesterase enzyme inhibitoryactivity experiments, the effect of the leaf extract of Hambeles variety from January collection, whichshowed the highest cholinesterase inhibition, against the neurotoxicity induced by 6-OHDA inSHSY-5Y cells was investigated. According to this; consistent with cell viability analyzes, thehighest cytotoxic value was observed at 100 μg/mL concentration of the extract applied for 24 hoursafter pre-application of 100 μM 6-OHDA for 24 hours. This study constitutes the first study on M.communis leaf and fruit extracts cultivated in our country regarding annual changes in thephytochemistry relevant to its phenolic components as well as neuroprotective effect throughcholinesterase and TYR enzyme inhibition along with cell culture assays.Key Words : M yrtus communis, Neuroprotective Effect, Enzyme Inhibition,Alzheimer’s Disease, Cell Culture, LC-Q-ToF-MS

Bitkiler çok eski çağlardan bu yana sağlık, gıda ve bazı diğer amaçlarla kullanılarak insanlığa hizmet etmektedir. Dolayısıyla bitkilerin tedavi amacıyla kullanıldığı yüzyıllar boyunca hekimlik ve eczacılık birbirinden ayrılmayan iki meslek olagelmiştir. Tarih öncesi çağlardan itibaren bitkilerin tedavi amacıyla kullanımına dair pek çok yazılı kaynak mevcuttur. Tedavide kullanılan bitkilerin bilimsel süzgeçten geçirilerek kullanımlarının sağlanması toplum sağlığı açısından önem ihtiva etmektedir. Tıbbın ve bitkilerle tedavinin gelişim sürecine bakıldığında, bu tez çalışmasında başta Mezopotamya kökenli olmak üzere kadim medeniyetler (Sümer, Babil, Hitit, Yunan, vs) ile Hint, Çin, Japon ve İslam medeniyetlerinin katkıları “Genel Bilgiler” kısmında gözden geçirilmiştir. Tez kapsamında, Milli Kütüphane bünyesindeki Yazma Eserler Müdürlüğü koleksiyonunda bulunan Osmanlı dönemine ait, fitoterapi ile ilgili olan eski Türkçe ile yazılmış “Envâ-ı Emrâz” isimli el yazması eser günümüz Türkçesine çevrilmiş ve içindeki tıbbi bilgiler; hastalıkların ilgili oldukları sistemlere, tedavide kullanılan ilaçların hazırlanış yöntemlerine ve eserde ismi geçen bitkilerin literatürde yer alan karşılıklarına göre incelenmiştir. Envâ-ı Emrâz adlı eser 604 babdan oluşan 3 bölüm halindedir. 1. bölümde Galen hekimin, 2. bölüm Millet hekimin, 3. bölümde ise Hipokrat hekimin reçeteleri bulunmaktadır. Eserin her bölümünde sırasıyla tüm hastalıklar tekrar edilmiştir. 2. Bölümde farklı olarak özellikle savaş yaralarının tedavilerine yer verilmiştir. Terkipler genellikle bitkilerin yaprak, çiçek veya kök gibi kısımlarının karışımı veya tek bitki kısmının kaynatılması sonucu elde edilen şuruplar ve macunlar şeklindedir. Eserde detaylı tarifler çok nadiren yer almaktadır. Genellikle tek cümle ile hangi şikâyet için neyin iyi geleceği bahsedilmiş ve bitkilerin kullanımına dair bilgiler yüzeysel olarak verilmiştir. Aynı şikâyet için farklı reçeteler (varsa) aynı babda peş peşe verilmiştir. Tez çalışmamız neticesinde, tıbbi bitkilerin tedavide kullanılışlarına dair Envâ-ı Emrâz’da bulunan kadim bilgiler gün ışığına çıkartılmış olup, geleneksel bilgilerin modern fitoterapi araştırmalarında kullanılabilmesi için literatüre katkıda bulunulmuştur.Anahtar KelimelerFitoterapi, geleneksel tıp, el yazması eser, Osmanlı tıbbı, Envâ-ı Emrâz, Galen, Hipokrat Plants have served humanity for health, food, and other purposes since ancient times. Therefore, medicine and pharmacy have been two professions that are not separated from each other for centuries, when plants are used for therapeutic purposes. There are many written sources about the therapeutic use of plants since prehistoric times. Ensuring the use of the plants used in treatment by passing them through a scientific filter is important in terms of public health. In this thesis study, contributions of the ancient civilizations particularly in Mesopotamia (e.g. Sumerian, Babillion, Hittite, Greek) as well as Indian, Chinese, Japanese, and Islamic civilizations are briefly reviewed in “General Information” section, considering the development process of medicine and herbal therapy. The manuscript named “Envâ-ı Emrâz”, written in ancient Turkish and related to phytotherapy, belonging to the Ottoman period, which is preserved in the collection of the Directorate of Manuscripts in the National Library, has been translated into today's Turkish within the scope of the thesis and medical information it contains; the systems to which the diseases belong, the preparation methods of the drugs used in the treatment and the equivalents of the plants mentioned in the work in the literature. The book named Envâ-ı Emrâz consists of 3 chapters having 604 babs. The prescriptions of the physician Galen in the first part, the physician Millet in the second part, and the physician of Hippocrates in the third part were written. In each part of the work, all diseases are repeated respectively. In the second chapter, the treatment of war wounds is discussed in particular as being different from other two chapters. The compositions are generally in the form of syrups and pastes obtained by boiling a mixture of plant parts such as leaves, flowers, and roots or or single plant part. Detailed recipes are found very rarely in the manuscript. Generally, it was mentioned in one sentence what would be good for which complaint. Information on uses of the plants were given roughly. Different prescriptions (if any) for the same complaint were given in the same item one after the other. As a result of our thesis, ancient information about the use of medicinal plants in treatment was brought to light, and a contribution was made to the literature for the use of traditional information in modern phytotherapy research.Key WordsPhytotherapy, traditional medicine, ancient manuscript, Ottoman medicine, Envâ-ı Emrâz, Galenus, Hippocrates

Our microbiome consists of trillions of microorganisms. Probiotics are known to be favorable microorganisms (bacteria and yeast) to keep the gut healthy and other health effects. They have been shown to possess positive effects not only in digestion, but also on heart, brain, etc. Lately, many studies pointed out to promoting activities of probiotics in mental diseases through gut microbiota, which is so-called “psychobiome”. It is now clear that bacterial inhabitants of the intestines may impact neurons and the brain through some mechanisms. Consequently, psychobiotics have been described as a new class of probiotics. In the current mini review, the role of psychobiotics is scrutinized focusing on some neurological and mental diseases.

The pollen of Pinus brutia Ten., known as red pine (Pinaceae), is used against liver diseases in Anatolian folk medicine. Materials and Methods: In the current study, pollen ethanol extract of P. brutia was investigated for its possible hepatoprotective activity using a mouse model of CCl4-induced hepatotoxicity. Swiss albino mice were divided into five groups, and extract-treated groups were compared with a silymarin-treated group as the reference. The extract was tested at 100, 200, and 300 mg/kg (b.w.). Phenolic acids were analyzed using high-performance column chromatography (HPLC) in the extracts as pollens are usually known to be rich in phenolics. Results: Our data revealed that the extract displayed the best hepatoprotection at a dose of 100 mg/kg when compared with silymarin (Legalon®), the reference drug. HPLC analysis indicated presence of protocatechuic acid (0.176 mg/g extract), p-hydroxybenzoic acid (0.001 mg/g extract), vanillic acid (VA) (0.537 mg/g extract), syringic acid (0.050 mg/g extract), and tr-cinnamic acid (0.310 mg/g extract), while the major phenolic acid was VA. Conclusion: The outcomes of this study allow us to conclude that red pine pollen extract can serve as a promising hepatoprotective agent. Among the phenolic acids analyzed in the pollen extract, vanillic acid as the major one besides some other phenolic acids detected seems to be responsible for its remarkable hepatoprotective effect.

3-Hidroksi-3-metil-glutaril-koenzim A (HMG-KoA) redüktaz enzimi kolesterol biyosentezinde görev alan anahtar enzim olduğu için HMG-KoA redüktaz inhibisyonu hiperkolesterolemi tedavisi için en önemli hedeflerden biri olarak kabul edilmektedir. Diğer yandan, yeni ilaç keşfi araştırmalarında tıbbi bitkiler ve bunlardan izole edilen moleküller önemli bir yer tutmaktadır. Bu çalışmada, Türkiye’de yetişen ve daha önce bu enzime yönelik olarak üzerinde araştırma yapılmamış olan Lamiaceae familyasına ait 37 bitki taksonunun etanol ekstrelerinin in vitro HMG-KoA redüktaz enzim inhibisyon aktiviteleri ELISA mikroplak yöntemi kullanılarak araştırılmıştır. Taramamız sonucunda inhibitör etkili bulunan Salvia ekstreleri, Salvia multicaulis Vahl. (% 61,66±1,52) ve Salvia blepharochlaena Hedge & Hub.Mor. (% 55,21±1,12) olarak tespit edilmiştir.

Bu tez çalışmasında, Lamiaceae familyasından Salvia, Satureja, Origanum ve Nepeta cinsine ait 37 türünetanol ekstreleri 3-hidroksi-3-metil-glutaril Koenzim A (HMG-KoA) redüktaz enzimine karşı in vitroinhibitör aktiviteleri yönünden ELISA yöntemiyle test edilmiş ve en yüksek inhibitör aktiviteyi Salviamulticaulis Vahl. göstermiştir. Daha sonra bu türün değişik polaritede kök ve topraküstü ekstrelerihazırlanarak tekrar HMG-KoA redüktaza karşı inhibisyon kapasiteleri test edilmiş olup, bitkinindiklorometan:aseton (1:1) kök ekstresinin en yüksek inhibisyon (% 71,97) gösterdiği saptanmıştır. Dahasonra aktivite-yönlendirmeli fraksiyonlama (AYF) yöntemiyle etkiden sorumlu bileşiğin veya bileşiklerintespit edilmesi amacıyla ekstre üzerinde fitokimyasal çalışmalar gerçekleştirilmiştir. Bu kapsamda ekstreönce kolon kromatografisi ile fraksiyonlanmış olup, elde edilen fraksiyonlar ince tabaka kromatografisiile takip edilmiştir. Birbirine benzer olan fraksiyonlar birleştirilmiş ve toplam 15 adet fraksiyon eldeedilmiştir. Bunlardan 5 tanesi (M-3, M-6, M-7, M-8 ve M-10) HMG-KoA redüktaza karşı %50’ninüzerinde inhibitör aktivite göstermiştir. Daha ileri kromatografik çalışmalar aktif fraksiyonlar üzerindeyürütülmüştür. Bu fraksiyonlardan hareketle 10 adet bileşik (biri 1:1 karışım olmak üzere) izole edilmiştir.İzole edilen ve yapıları aydınlatılan bileşiklerin, referans ilaç olarak atorvastatin kullanılarak yapılanHMG-KoA redüktaz inhibitör aktivite sonuçlarına göre; 7-asetoksihorminon (SY-20) % 84,15 ± 0,10;horminon ve 7-asetoksihorminon (1:1) karışımı (SY17) % 76,26 ± 0,14 değerleri ile en yüksek inhibitöraktiviteyi göstermiştir. En aktif bileşik olarak aktiviteden sorumlu olduğu düşünülen iki bileşik olan 7-asetoksihorminon ile horminon ve 7-asetoksihorminon karışımının enzimin aktif bölgesindekiaminoasitlerle etkileşimi moleküler kenetlenme modeli ile incelenmiştir. Bu çalışma ülkemizde yetişenbazı Salvia, Satureja, Origanum ve Nepeta türlerinin HMG-KoA redüktaz enzim inhibitör aktivitesininve Salvia multicaulis’in inhibitör aktiviteden sorumlu olduğu bileşiklerin incelendiği ilk çalışmadır.Anahtar Kelimeler : HMG-KoA Redüktaz, Enzim İnhibisyonu, Lamiaceae, Salvia,Satureja, Origanum, Nepeta, Hiperkolesterolemi In this thesis study, the ethanolic extracts of 37 plants belonging to the genus Salvia, Satureja, Origanum,and Nepeta from the Lamiaceae family were tested in vitro by ELISA method for their inhibitory activityagainst 3-hydroxy-3-methyl-glutaryl Coenzyme A (HMG-CoA) reductase. Salvia multicaulis Vahl.showed the highest inhibitory activity among these plants. Afterwards, the extracts in different polarityof this species were prepared and their inhibition capacity against HMG-CoA reductase was tested again,and it was found that the dichloromethane:acetone (1:1) root extract of the plant exhibited the highestinhibition (71.97%). Later on, phytochemical studies were run on the extract in order to determine thecompound(s) responsible for the inhibitory effect by the activity-guided fractionation method. In thiscontext, the extract was firstly fractionated over column chromatography and the obtained fractions weremonitored by thin layer chromatography. The fractions that were similar to each other were combinedand a total of 15 fractions were obtained. 5 of them (M-3, M-6, M-7, M-8, and M-10) displayed over 50%inhibitory activity against HMG-CoA reductase. Further chromatographic studies were conducted on theactive fractions, which led to isolation of 10 compounds, one of which is a mixture of two compounds(1:1). According to our HMG-CoA reductase inhibitory activity results which atorvastatin was employedas the reference drug, chemical structures of the isolated compounds were elucidated as follows; 7-acetoxyhorminone (SY-20) and horminone and 7-acetoxyhorminone (1:1) mixture (SY17) which showedthe highest inhibitory activity with 84.15 ± 0.10% and 76.26 ± 0.14% inhibition values, respectively. Theinteractions of 7-acetoxyhorminone as well as horminone and 7-acetoxyhorminone, considered to beresponsible for the activity as the most active compounds, with amino acids in the active site of theenzyme were investigated by molecular docking simulations. 2019 yılı Aralık ayı sonunda Çin’in Wuhan şehrinde ortaya çıkarak, dünyaya yayılan ve 21. yüzyılın ilk pandemisine sebep olan yeni tip corona virüsün (SARS-CoV-2) neden olduğu COVID-19, Mayıs 2020 tarihi itibarıyla dünyada 4 milyondan fazla insanın enfekte olduğu küresel ölçekte bir sağlık sorunu haline gelmiştir. Bu süreçte hastalığın teşhisi ve tedavisi nedeniyle sağlık çalışanları (hekimler, hemşireler, eczacılar, hastabakıcılar, vs) COVID-19 hastalarıyla muhatap olmaları nedeniyle en büyük risk ile karşı karşıya kalan meslek grupları haline gelmiştir. Eczacılar, özellikle ülkemizde 1. basamak sağlık sunucusu statüsünde olan eczane eczacıları, halkın en kolay ulaşabildiği sağlık çalışanları olarak pandemi sürecinde hizmet vermeye devam etmiştir. Ancak damlacıklı temas yoluyla bulaşma riski çok yüksek olan yeni tip corona virüsten korunmak, günde yüzlerce hastanın ziyaret ettiği eczanelerde eczacı ve eczacı teknisyenleri için büyük bir sağlık sorunu teşkil etmesine rağmen, COVID-19 pandemi sürecinde eczaneler kişisel çapta hijyen önlemleri alarak ücretsiz maske dağıtımı görevini yüklenmiştir. Bu makalede, COVID-19 pandemi sürecinde ülkemizde eczacıların üstlendikleri görev ve sorumluluklar ile kamu hizmetleri tartışılacaktır.
