This study investigated the use of endophyte-assisted Tillandsia brachycaulos to enhance formaldehyde removal in indoor environments. A formaldehyde-degrading endophyte from the root of Epipremnum aureum , Pseudomonas plecoglossicida , was identified and used for inoculation. Among the inoculation methods, spraying proved to be the most effective, resulting in a significant 35 % increase in formaldehyde removal after 36 hours. The results of the light exposure experiment (3000 Lux) demonstrate that an increase in light intensity reduces the efficiency of the Tillandsia brachycaulos -microbial system in degrading formaldehyde. In a 15-day formaldehyde fumigation experiment at 2 ppm in a normal indoor environment, the inoculated Tillandsia brachycaulos exhibited removal efficiency ranging from 42.53 % to 66.13 %, while the uninoculated declined from 31.62 % to 3.17 %. The Pseudomonas plecoglossicida (referred to as PP-1) became the predominant bacteria within the Tillandsia brachycaulos after fumigation. Moreover, the endophytic inoculation effectively increased the resistance and tolerance of Tillandsia brachycaulos to formaldehyde, as evidenced by lower levels of hydroxyl radical, malondialdehyde (MDA), free protein, and peroxidase activity (POD), as well as higher chlorophyll content compared to uninoculated Tillandsia brachycaulos. These findings indicate that the combination of endophytic bacteria and Tillandsia brachycaulos has significant potential for improving indoor air quality. Potato Verticillium wilt (PVW) caused by Verticillium dahliae is a vascular disease, that seriously affects potato (Solanum tuberosum L.) yield and quality worldwide. V. dahliae occupies the vascular bundle and therefore it cannot efficiently be treated with fungicides. Further, the application of these pesticides causes serious environmental problems. Therefore, it is of great importance to find environmentally friendly biological control methods. In this study, bacterial strains were isolated from agricultural lands on which potato had been cultured for 5 years. Five strains with a broad-spectrum antagonistic activity were selected. Among these five strains, Bacillus velezensis XS142 showed the highest antagonistic activity. To study the mechanism of XS142, by which this strain might confer tolerance to V. dahliae in potato, the genome of strain XS142 was sequenced. This showed that its genome has a high level of sequence identity with the model strain B. velezensis FZB42 as the OrthoANI (Average Nucleotide Identity by Orthology) value is 98%. The fungal suppressing mechanisms of this model strain are well studied. Based on the genome comparison it can be predicted that XS142 has the potential to suppress the growth of V. dahliae by production of bacillomycin D, fengycin, and chitinase. Further, the transcriptomes of potatoes treated with XS142 were analyzed and this showed that XS142 does not induce ISR, but the expression of genes encoding peptides with antifungal activity. Here we showed that XS142 is an endophyte. Further, it is isolated from a field where potato had been cultured for several years. Introduction: Phosphate-solubilizing bacteria that function through acidification (organic acid synthesis) or mineralization (production of enzymes such as phytase and phosphatases) have been explored as a biotechnological alternative to enhance plant access to phosphorus (P) retained in organic and inorganic forms in agricultural soils. This study tested the hypothesis that applying a biofertilizer composed of a recognized phosphate-solubilizing bacterium(Bacillus velezensis - endophytic strain BVPS01) and an underexplored plant growth-promoting bacterium (Lysinibacillus fusiformis - endophytic strain BVPS02) would improve the growth and grain yield of Glycine max L. plants. Methods: Initial in vitro tests assessed the functional traits of these bacteria, and a mix of strains BVPS01 and BVPS02 was produced and tested under field conditions to evaluate its agronomic efficiency. Results: The results confirmed the hypothesis that the tested biofertilizer enhances the agronomic performance of G. max plants in the field. The B. velezensis strain (BVPS01) was found to be more effective than the L. fusiformis strain (BVPS02) in solubilizing phosphates via the phosphatase enzyme production pathway, indicated by the expression of the phoC and phoD genes. In contrast, L. fusiformis was more effective in solubilizing phosphates through organic acid and phytase-related pathways, in addition to synthesizing indole-3-acetic acid and increasing the mitotic index in the root meristem of G. max plants. These strains exhibited biological compatibility, and the formulated product based on these rhizobacteria enhanced root development and increased the number of nodules and flowers, positively affecting 1000-grain weight, grain yield, and grain P content. Discussion: Thus, the tested biofertilizer demonstrated potential to improve root growth and increase both the yield and quality of soybean crops, making it a sustainable and low-cost strategy. Endophytic bacteria have not been reported widely for their lipolytic abilities, so they are not used for large‐scale lipase production. The purpose of this study was to explore an endophytic bacterium for the production of lipase implementing cost‐effective techniques, including the use of Cassia fistula seeds, as a substrate in the production medium and stem pieces of Phragmites karka as the immobilization matrix. The endophytic strain Bacillus sp. E4 was originally isolated from the halophytic plant Arthrocnemum macrostachyum. Bacillus sp. E4 produced 6.05 IU mL−1 lipase in the presence of powdered seeds of Cassia fistula (golden shower tree). Initial trial experiments using a one‐factor‐at‐a‐time approach led to an improvement in lipase titers to 10.05 IU mL−1. Consequently, investigations using the Plackett–Burman design suggested the influence of three significant factors – incubation period, inoculum size, and substrate concentration – on lipase production. They were optimized using the Box–Behnken design (BBD). In the response optimization experiment, strain E4 yielded 52.35 IU mL−1 lipase, which was in accordance with the predicted yield and indicated an overall 8.65 fold improvement in lipase production. To investigate the use of free cells, strain E4 was immobilized on the stem pieces of a halophytic plant, Phragmites karka, which was used for the first time as an immobilization matrix. The immobilized cells retained lipase production ability for up to six cycles with the highest yield of 110 IU mL−1, which corresponded to an improvement of more than eighteenfold. Scanning electron micrographs confirmed the colonization of E4 cells in the matrix and demonstrated the utilization of C. fistula seeds. Fourier transform infrared spectroscopy affirmed the utilization of components including fatty acids by the immobilized E4 cells. Introduction: The antibacterial protein PAG14 was extracted from a metabolite of Bacillus G14 isolated from mangrove plants. Methods: In this study, Pseudomonas aeruginosa, Pasteurell multocide, Enterobacter aerogenes, and Enterococcus faecalis were used as indicator bacteria to screen endophytes that exhibited antibacterial activity. The endophyte culture conditions were optimized to enhance productivity. Subsequently, the culture supernatant was salted using ammonium sulfate, followed by purification using dextran gel chromatography and ion exchange column techniques. Finally, the structures of antibacterial proteins were identified using mass spectrometry. Results and Discussion: The optimal culture conditions for Bacillus G14 were 2% mannitol, 0.5% fish peptone, 0.05% KH2PO4 + 0.05% K2HPO4 + 0.025% MnSO4·H2O. The antibacterial substances exhibited stability within the temperature range of 30-40°C and pH range of 5.0-7.0, while displaying sensitivity toward enzymes. The antibacterial activity decreased as the duration of UV irradiation increased. The antibacterial protein PAG14, isolated from the culture broth of Bacillus G14 through purification using dextran gel and ion-exchange columns, was identified as a class III bacteriocin using LC-MS/MS, similar to Lysozyme C. Living in diverse environmentally harsh conditions, the plant exhibits a unique survival mechanism. As a result, the endophytes residing within the plant produce specific compounds that promote the plant's growth and defend it against pathogens. Plants and algae symbiotically harbor endophytes, i.e., microbes and microorganisms living within them. The objective of this study is to isolate endophytic fungi, specifically strains of Aspergillus terreus, from the leaves of the salt-tolerant plant Tetraena qatarensis and to explore the salt tolerance, antagonistic activity, and growth promotion properties. Strain C A. terreus (ON117337.1) was screened for salt tolerance and antagonistic effects. Regarding salt tolerance, the isolate demonstrated the ability to thrive in a concentration of up to 10% NaCl. A. terreus showed inhibitory activity against four fungal phytopathogens, namely Fusarium oxysporum, Alternaria alternata, Colletotrichum gloeosporioides, and Botrytis cinerea. The GC-MS investigation of the fungal (strain C Aspergillus terreus) extract showed the presence of about 66 compounds (secondary metabolites). Secondary metabolites (SMs) are produced, like Hexadecanoic acid, which aids in controlling phytopathogens. Also produced is lovastatin, which is used to treat hypercholesterolemia. Strain C, which showed salinity tolerance and the highest inhibitory activity, was further analyzed for its effect on tomato seed germination under pathogen stress from Fusarium oxysporum. The greenhouse experiment indicated that the fungi increased the length of tomato seedlings and the plant biomass. Therefore, the selected endophytes derived from Tetraena qatarensis were scrutinized for their potential as biocontrol agents, aiming to thwart fungal pathogens and stimulate plant growth. The in vitro and in vivo assessments of strain C (Aspergillus terreus) against Fusarium oxysporum in this investigation indicate the promising role of endophytes as effective biological control agents. Introduction: Cistanche salsa (C.A.Mey.) G. Beck is a perennial holoparasitic herb recognized for its medicinal properties, particularly in kidney-tonifying and laxative treatments. Despite its therapeutic potential, little is known about the endophyte communities inhabiting C. salsa and its host plants, and how these microorganisms may impact the production and accumulation of metabolites in C. salsa. Methods: We conducted a dual analysis focusing on metabolomics of wild C. salsa and microbiome characterization of both C. salsa and its host plant, Kalidium foliatum (Pall.) Moq. The metabolomics analysis revealed variations in metabolite composition across different parts of C. salsa. Additionally, the microbiome analysis involved studying endophytic bacteria and fungi, comparing their community structures between parasitic C. salsa and its host plant. Results: Significant variations in metabolite composition were observed through metabolomic profiling, which identified 93 secondary metabolites and 398 primary metabolites across various parts of C. salsa. Emphasis was placed on differences in metabolite composition within the flowers. Microbiome analysis revealed differential community compositions of endophytic bacteria between the parasitic and host plants, whereas differences in endophytic fungi were less pronounced. Certain endophytes, such as Bacteroidota, Proteobacteria, Ascomycota, and Basidiomycota, were associated with the production of specific secondary metabolites in C. salsa, including the plant-specific compound salsaside. Discussion: Our findings highlight the intricate relationship between C. salsa and its endophytic microbiota, suggesting a potential role of these microorganisms in modulating the biosynthesis of bioactive compounds. The differential preferences of endophytic bacteria and fungi across various microenvironments within the parasitic plant system underscore the complexity of these interactions. There is an enormous diversity of life forms present in the extremely intricate marine environment. The growth and development of seaweeds in this particular environment are controlled by the bacteria that settle on their surfaces and generate a diverse range of inorganic and organic chemicals. The purpose of this work was to identify epiphytic and endophytic bacterial populations associated with ten common marine macroalgae from various areas along the Mediterranean Sea coast in Alexandria. This was done to target their distribution and possible functional aspects. Examine the effects of the algal habitat on the counting and phenotypic characterization of bacteria, which involves grouping bacteria based on characteristics such as shape, colour, mucoid nature, type of Gram stain, and their ability to generate spores. Furthermore, studying the physiological traits of the isolates under exploration provides insight into the optimum environmental circumstances for bacteria associated with the formation of algae. The majority of the bacterial isolates exhibited a wide range of enzyme activities, with cellulase, alginase, and caseinase being the most prevalent, according to the data. Nevertheless, 26% of the isolates displayed amylolytic activity, while certain isolates from Miami, Eastern Harbor, and Montaza lacked catalase activity. Geographical variations with the addition of algal extract may impact on the enumeration of the bacterial population, and this might have a relationship with host phylogeny. The most significant observation was that endophytic bacteria associated with green algae increased in all sites, while those associated with red algae increased in Abu Qir and Miami sites and decreased in Eastern Harbor. At the species level, the addition of algal extract led to a ninefold increase in the estimated number of epiphytic bacteria for Cladophora pellucida in Montaza. Notably, after adding algal extract, the number of presented endophytic bacteria associated with Codium sp. increased in Abu Qir while decreasing with the same species in Montaza. Introduction: Constructed wetlands (CWs) are nature-based solutions for wastewater treatment where the root system microbiome plays a key role in terms of nutrient and pollutant removal. Nonetheless, little is known on plant-microbe interactions and bacterial population selection in CWs, which are mostly characterized in terms of engineering aspects. Methods: Here, cultivation-independent and cultivation-based analyses were applied to study the bacterial communities associated to the root systems of Phragmites australis and Typha domingensis co-occurring in the same cell of a CW receiving primary treated wastewaters. Results and discussion: Two endophytic bacteria collections (n = 156) were established aiming to find novel strains for microbial-assisted phytodepuration, however basing on their taxonomy the possible use of these strains was limited by their low degrading potential and/or for risks related to the One-Health concept. A sharp differentiation arose between the P. australis and T. domingensis collections, mainly represented by lactic acid bacteria (98%) and Enterobacteriaceae (69%), respectively. Hence, 16S rRNA amplicon sequencing was used to disentangle the microbiome composition in the root system fractions collected at increasing distance from the root surface. Both the fraction type and the plant species were recognized as drivers of the bacterial community structure. Moreover, differential abundance analysis revealed that, in all fractions, several bacteria families were significantly and differentially enriched in P. australis or in T. domingensis. CWs have been also reported as interesting options for the removal of emerging contaminants (e.g, antibiotic resistance genes, ARGs). In this study, ARGs were mostly present in the rhizosphere of both plant species, compared to the other analyzed fractions. Notably, qPCR data showed that ARGs (i.e., ermB, blaTEM, tetA) and intl1 gene (integrase gene of the class 1 integrons) were significantly higher in Phragmites than Typha rhizospheres, suggesting that macrophyte species growing in CWs can display a different ability to remove ARGs from wastewater. Overall, the results suggest the importance to consider the plant-microbiome interactions, besides engineering aspects, to select the most suitable species when designing phytodepuration systems. Some plant-associated microorganisms could improve host plants biotic and abiotic stress tolerance. Imperata cylindrica is a dominant pioneer plant in some abandoned mine lands with higher concentrations of heavy metal (HM). To discover the specific microbiome of I. cylindrica in this extreme environment and evaluate its role, the microbiome of I. cylindrica’s seeds and rhizosphere soils from HM heavily contaminated (H) and lightly contaminated (L) sites were studied. It was found that HM-contamination significantly reduced the richness of endophytic bacteria in seeds, but increased the abundance of resistant species, such as Massilia sp. and Duganella sp. Spearman’s rank correlation coefficient analysis showed that both Massilia sp. and Duganella sp. showed a significant positive correlation with Zn concentration, indicating that it may have a strong tolerance to Zn. A comparison of the microbiome of rhizosphere soils (RS) and adjacent bare soils (BS) of site H showed that I. cylindrica colonization significantly increased the diversity of fungi in rhizosphere soil and the abundance of Ascomycota associated with soil nutrient cycling. Spearman’s rank correlation coefficient analysis showed that Ascomycota was positively correlated with the total nitrogen. Combined with the fact that the total nitrogen content of RS was significantly higher than that of BS, we suppose that Ascomycota may enhance the nitrogen fixation of I. cylindrica, thereby promoting its growth in such an extreme environment. In conclusion, the concentration of HM and nutrient contents in the soil significantly affected the microbial community of rhizosphere soils and seeds of I. cylindrica, in turn, the different microbiomes further affected soil HM concentration and nutrient contents. [Objective] To identify siderophilic endophytic bacteria that efficiently promote iron absorption in rice, and to investigate the siderophore-producing capacity and iron absorption promoting effect of different genera of endophytic bacteria from four wild rice. [Method] This study analyzed the siderophore contents of 198 endophytic bacteria isolated from four types of wild rice including Oryza longistaminata, O. rufipogon, O. officinalis, and O. minuta. The 82 strains with highly efficient siderophore-producing ability were screened from 128 strains with siderophore-producing ability. Analysis of the 16S rRNA gene sequences revealed that these 82 strains belonged to 18 different genera or species. Based on their siderophore production abilities, 18 candidate strains were selected to assess their abilities of promoting iron absorption in rice. [Result] The results demonstrated that different genera had varying effects on promoting rice iron absorption, with Bacillus (class), Pseudomonas, and Saccharomycetes showing significant effects. Five high-efficient siderophore producing strains were selected from 18 strains and further confirmed that endophytic bacteria partially restored the iron deficiency phenotype of a rice iron-absorption-deficient mutant (osbhlh156). [Conclusion] This study elucidates that siderophore produced by plant-interacting microorganisms can be utilized by rice, thereby enriching the microbial resources associated with iron absorption in rice. Cocoa (Theobroma cacao L.) is one of the leading commodities in Indonesia. Cocoa pod husk is part of the mesocarp or part of the cacao pod husk that wraps the cocoa beans. Endophytic bacteria are microbes that live in host plant tissues and do not interfere with host plant growth. This study aimed to determine characteristics of endophytic bacterial isolates contained in endocarp cocoa pod husk and to determine the antioxidant power of endophytic bacterial isolates contained in cocoa pod husk. Isolation of endophytic bacteria from the inner pod of cocoa which be expected to produce antioxidants has been carried out. Isolation of bacteria using surface sterilization method. Antioxidant test using DPPH method (1,1-diphenyl-2-picrylhydrazyl) with UV-Vis spectrophotometer at 600 nm. Two isolates out of four endophytic bacteria were obtained from the results of the preliminary test of phenolic tests, namely isolates of bacteria KK2 and KK3 which had different colony morphology. The results of the secondary metabolite test showed that the bacterial isolates KK2 and KK3 contained flavonoids and phenolics. Based on the test results, the value of IC50 of bacterial isolate KK2 was 2104.58 ppm and KK3 bacterial isolate was 1123.04 ppm. Terpenoid indole alkaloids (TIAs) are natural compounds found in medicinal plants that exhibit various therapeutic activities, such as antimicrobial, anti-inflammatory, antioxidant, anti-diabetic, anti-helminthic, and anti-tumor properties. However, the production of these alkaloids in plants is limited, and there is a high demand for them due to the increasing incidence of cancer cases. To address this research gap, researchers have focused on optimizing culture media, eliciting metabolic pathways, overexpressing genes, and searching for potential sources of TIAs in organisms other than plants. The insufficient number of essential genes and enzymes in the biosynthesis pathway is the reason behind the limited production of TIAs. As the field of natural product discovery from biological species continues to grow, endophytes are being investigated more and more as potential sources of bioactive metabolites with a variety of chemical structures. Endophytes are microorganisms (fungi, bacteria, archaea, and actinomycetes), that exert a significant influence on the metabolic pathways of both the host plants and the endophytic cells. Bio-prospection of fungal endophytes has shown the discovery of novel, high-value bioactive compounds of commercial significance. The discovery of therapeutically significant secondary metabolites has been made easier by endophytic entities' abundant but understudied diversity. It has been observed that fungal endophytes have better intermediate processing ability due to cellular compartmentation. This paper focuses on fungal endophytes and their metabolic ability to produce complex TIAs, recent advancements in this area, and addressing the limitations and future perspectives related to TIA production. Previously it was shown that endophytic bacteria had the ability to move out of the roots of pea plant seedlings (Pisum sativum L.) into the rhizosphere. In this study, six distinct bacterial strains were isolated from the root growth medium during the cultivation of seedlings in an aqueous medium. By analyzing the nucleotide sequence of 16S rRNA genes, the taxonomic position of these strains was established. Their morphological and cultural parameters were assessed, and the activity of hydrolytic enzymes (pectinase, cellulase, protease) and the IAA-producing capability were examined. It has been observed that the quantity of endophytic bacteria that appears on the root surface during the growth of pea seedlings significantly surpasses the quantity present in the root tissues. It is assumed that hydrolytic enzymes such as pectinase and cellulase are involved in the release of bacteria into the external environment, causing the destruction of carbohydrate structures in plant cell walls. The metabolic parameters established in the studied strains, and the significance of these endophytic bacteria for the host plant after their exit from the roots into the rhizosphere are under discussion. Sake, which is produced mainly from japonica rice (Oryza sativa subsp. japonica), is one of the most important alcohol products in Japan. In this study, we aimed to investigate a hypothesis that the early root endophytic bacterial communities in Japanese sake rice cultivars would be distinct from those in table rice cultivars, comparing four sake rice cultivars and two table rice cultivars. Rice roots in the vegetative stage were collected 0, 3, and 6 weeks after transplanting, and 16S rRNA gene amplicon sequencing revealed significant differences in bacterial community composition diversity between the sake and table rice cultivars. The root endophytic bacterial communities at the transplanting differed significantly between the rice cultivars, indicating differences in each seed-derived endophytic community. After an overall dominance of Pantoea and Methylobacterium-Methylorubrum at the transplanting, the endophytic community was gradually replaced by soil-derived bacteria that varied by the rice cultivars. Notably, PERMANOVA results showed that the rice endophytic bacterial community composition differed significantly between the sake and table rice cultivars (p < 0.001). These results highlight the distinct root endophytic bacterial composition in the sake rice cultivars compared to those in the table rice cultivars, supporting our hypothesis. [ABSTRACT FROM AUTHOR]

We studied the occurrence of endophytic fungi and bacteria in the roots of F. gigantea, a woodland perennial grass common throughout Europe and temperate Asia. The taxonomic assignment was carried out according to the isolate's colony and cytological species morphotype characteristics and confirmed by the assessment of the standard DNA sequences, ITS, RPB2, SSU, and TEF1-a for fungi and 16S rDNA for bacteria. Our study has shown that F. gigantea roots are the habitat to a wide range of fungi and bacteria. The occurrence of fungal structures was determined in ~40% of the roots examined by Trypan Blue staining. In a surface-sterile root-cutting culture on PDA medium, we obtained isolates of six endophytic fungi species: four members of Ascomycota—Alternaria alternata, Cadophora fastigiata, Chaetomium funicola, and Microdochium bolleyi—and two of Basidiomycota—Coprinellus sp. and Sistotrema brinkmannii. In addition, we report bacteria co-occurring endophytically in the roots of this grass. The Firmicutes group was the most prevalent, consisting of four Gram-positive, endospore-forming bacteria taxa. The isolates were identified as Bacillus pumilus, Bacillus sp., Lysinibacillus sp., and Priestia aryabhattai. Moreover, two Gram-negative bacteria were detected—Kosakonia sp. (Proteobacteria) and Pedobacter sp. (Bacteroidetes). Thus, applying the isolate-culture approach, we identified a set of microorganisms in the roots of a typical grass native to the deciduous forest floor. The functional roles of these endophytes are diverse, and many of them, saprotrophs and decomposers of wood and plant debris, are linked to the decomposition of organic matter. This is the first detailed report on fungal and bacterial endophytes inhabiting the roots of F. gigantea. This study fills in a research gap on endophytes associated with the below-ground parts of Festuca spp., hitherto extensively studied for Epichloë/Neotyphodium associations in their foliar parts. Here, we report on a Cordyceps species entering into a multi-trophic, multi-kingdom association. Cordyceps cateniannulata , isolated from the stem of wild Coffea arabica in Ethiopia, is shown to function as an endophyte, a mycoparasite and an entomopathogen. A detailed polyphasic taxonomic study, including a multilocus phylogenetic analysis, confirmed its identity. An emended description of C. cateniannulata is provided herein. Previously, this species was known as a pathogen of various insect hosts in both the Old and New World. The endophytic status of C. cateniannulata was confirmed by re-isolating it from inoculated coffee plants. Inoculation studies have further shown that C. cateniannulata is a mycoparasite of Hemileia vastatrix , as well as an entomopathogen of major coffee pests; infecting and killing Hypothenemus hampei and Leucoptera coffeella. This is the first record of C. cateniannulata from Africa, as well as an endophyte and a mycoparasite. Introduction: Vernonia amygdalina (V. amygdalina ) (Del.) has been reported to have medicinal values, and the phytochemical constituents which is present in different parts of the plant have been found to be responsible for the antimicrobial efficacy of the plant. This study focuses on the antibacterial activity of the stem bark extracts of V. amygdalina against multiple antibiotic-resistant (MAR) bacteria isolated from wound samples and characterize the bioactive compounds present in the plant. Methods: Bacteria were isolated from wound samples using standard microbiological techniques and disc diffusion method was used to determine the sensitivity patterns of the isolated bacteria to conventional antibiotics and to the various extracts of V. amygdalina stem bark. Ciprofloxacin was used as control during the antibacterial assay. GC-MS analysis was carried out to identify the bioactive compounds in the stem bark extracts of the plant. Results: Pseudomonas aeruginosa (36%), Staphylococcus aureus (29%), Staphylococcus epidermidis (14%), Bacillus subtilis (6%), Escherichia coli (6%), Klebsiella pneumoniae (5%), and Proteus mirabilis (4%) were the bacteria isolated from the wound swabs sampled in this study. The ethyl acetate extract of V. amygdalina (300 mg/ml) inhibited the growth of these organisms with the greatest effect on S. aureus with inhibition zone of 23.17±0.60 mm. The GC-MS analysis of the plant extracts revealed the presence of bioactive compounds such as coumarin and oleic acid, among others. Conclusion: The findings from this study have further established that V. amygdalina is a promising candidate and effective alternative treatment means for MAR bacteria that are associated with wound infections. Organic and natural sources of bio-stimulant have a great expectancy to boost green agriculture practices for sustainable, safe, and smart cultivation of crops. In that regard, beneficial endophytic bacteria have great potential. They have unique features in promoting plant growth by colonizing and establishing well in plant roots. In this study, endophytes isolated from the roots of moringa, neem, sesbania, and chilli were screened for crop's growth-enhancing activities, such as phosphorus (P) solubilization, 1-aminocyclopropane-1-carboxylic-acid deaminase (ACC deaminase) production, and indole-acetic acid (IAA) production. The phosphorus solubilization, indole-acetic acid production, and ACC deaminase production values fall in the range of 55–88 ppm, 20–164 ppm, and 0.317–0.375 mM, respectively. Chilli seeds' three highest vigor index (VI) values were attained by MR10 (12,457 VI), MR3 (9450 VI), and MR13 (8730 VI). MR13 showed the highest seed germination energy (221%), followed by MR1 (178%) and MR3 (156%). The promising endophytes were tested on chilli seedlings as single and mixed inoculum treatments to study the efficiency of root colonization. Mixed cultures containing CKR8 and MR13 exhibited the highest seedling height (17.0 cm), followed by MR13, MR10, and MR13 (16.8 cm) compared to the control (12.6 cm). A single culture of MR10 (109.0 g and 13.53 cm2) and a mixed culture of MR10 and MR13 (100.0 g and 13.09 cm2) showed the maximum root length and surface area, respectively. The highest relative chlorophyll content was recorded by MR10 and MR13 (40.3 SPAD value), followed by MR13, MR3, and CKR8 (36.8 SPAD value). The most serious problems for cultivated grapes are pathogenic microorganisms, which reduce the yield and quality of fruit. One of the most widespread disease of grapes is "gray mold", caused by the fungus Botrytis cinerea. Some strains of Bacillus, such as Bacillus halotolerans, Bacillus amyloliquefaciens, and Bacillus velezensis, are known to be active against major post-harvest plant rots. In this study, we showed that the endophytic bacteria B. velezensis strain AMR25 isolated from the leaves of wild grapes Vitis amurensis Rupr. exhibited antimicrobial activity against grape pathogens, including B. cinerea. The genome of B. velezensis AMR25 has one circular chromosome with a length of 3,909,646 bp. with 3689 open reading frames. Genomic analysis identified ten gene clusters involved in the nonribosomal synthesis of polyketides (macrolactin, bacillene, and difficidin), lipopeptides (surfactin, fengycin, and bacillizin), and bacteriocins (difficidin). Also, the genome under study contains a number of genes involved in root colonization, biofilm formation, and biosynthesis of phytohormones. Background: Studies have shown that plant endophytic microbial communities are ubiquitous and closely related to plant growth and health. To clarify the mechanism of the melon varieties with high resistant to wilt, the endophytic microbial compositions and metabolites in roots of melon varieties with high resistant ability to wilt were analyzed. Results: The results showed that the abundances of Firmicutes, Ascomycota, Bacillus, Bradyrhizobium, Amycolatopsis, Actinospica, and Catenulispora all increased in roots of wilt high resistant melon varieties (MT) which compared to wilt susceptible melon varieties (MS). Meanwhile, Ochrobactrum, Bordetella, Roseateles, Staphylococcus, Acidovorax, Amycolatopsis, Catenulispora, Promicromonospora, and Gymnopilus were the unique endophytic microbes in roots of MT. Moreover, in comparison with the MS varieties, the functions of Defense mechanisms, Secondary metabolites biosynthesis, transport and catabolism, Nucleotide transport and metabolism, Signal transduction mechanisms, Coenzyme transport and metabolism, Carbohydrate transport and metabolism and Amino acid transport and metabolism all increased in roots of MT varieties. Additionally, the nucleotide metabolism and biosynthesis of cofactors metabolic pathways were also significantly increased in roots of MT varieties. On the other hand, the untargeted metabolome results showed that Biosynthesis of various plant secondary metabolites, Nucleotide metabolism and Biosynthesis of cofactors metabolic pathways were significantly increased in the expression of MT varieties; and the content of metabolic compounds such as flavonoids, Cinnamic acid compounds, Organic acid compounds, and Nucleotides were increased. In addition, the correlation between microbiome and metabolome indicates a significant correlation between the two. The isolation and identification of plant growth-promoting endophytic bacteria (PGPEB) from Achyranthes bidentata roots have profound theoretical and practical implications in ecological agriculture, particularly as bio-inoculants to address challenges associated with continuous monoculture. Our research revealed a significant increase in the abundance of these beneficial bacteria in A. bidentata rhizosphere soil under prolonged monoculture conditions, as shown by bioinformatics analysis. Subsequently, we isolated 563 strains of endophytic bacteria from A. bidentata roots. Functional characterization highlighted diverse plant growth-promoting traits among these bacteria, including the secretion of indole-3-acetic acid (IAA) ranging from 68.01 to 73.25 mg/L, phosphorus and potassium solubilization capacities, and antagonistic activity against pathogenic fungi (21.54%-50.81%). Through 16S rDNA sequencing, we identified nine strains exhibiting biocontrol and growth-promoting potential. Introduction of a synthetic microbial consortium (SMC) in pot experiments significantly increased root biomass by 48.19% in A. bidentata and 27.01% in replanted Rehmannia glutinosa. These findings provide innovative insights and strategies for addressing continuous cropping challenges, highlighting the practical promise of PGPEB from A. bidentata in ecological agriculture to overcome replanting obstacles for non-host plants like R. glutinosa, thereby promoting robust growth in medicinal plants. Aflatoxins (AFs) are hazardous carcinogens and mutagens produced by some molds, particularly Aspergillus spp. Therefore, the purpose of this study was to isolate and identify endophytic bacteria, extract and characterize their bioactive metabolites, and evaluate their antifungal, antiaflatoxigenic, and cytotoxic efficacy against brine shrimp (Artemia salina) and hepatocellular carcinoma (HepG2). Among the 36 bacterial strains isolated, ten bacterial isolates showed high antifungal activity, and thus were identified using biochemical parameters and MALDI-TOF MS. Bioactive metabolites were extracted from two bacterial isolates, and studied for their antifungal activity. The bioactive metabolites (No. 4, and 5) extracted from Bacillus cereus DSM 31T DSM, exhibited strong antifungal capabilities, and generated volatile organic compounds (VOCs) and polyphenols. The major VOCs were butanoic acid, 2-methyl, and 9,12-Octadecadienoic acid (Z,Z) in extracts No. 4, and 5 respectively. Cinnamic acid and 3,4-dihydroxybenzoic acid were the most abundant phenolic acids in extracts No. 4, and 5 respectively. These bioactive metabolites had antifungal efficiency against A. flavus and caused morphological alterations in fungal conidiophores and conidiospores. Data also indicated that both extracts No. 4, and 5 reduced AFB1 production by 99.98%. On assessing the toxicity of bioactive metabolites on A. salina the IC50 recorded 275 and 300 µg/mL, for extracts No. 4, and 5 respectively. Meanwhile, the effect of these extracts on HepG2 revealed that the IC50 of extract No. 5 recorded 79.4 µg/mL, whereas No. 4 showed no cytotoxic activity. It could be concluded that bioactive metabolites derived from Bacillus species showed antifungal and anti-aflatoxigenic activities, indicating their potential use in food safety. Introduction: The importance of plant rhizodeposition to sustain microbial growth and induce xenobiotic degradation in polluted environments is increasingly recognized. Methods: Here the "cry-for-help" hypothesis, consisting in root chemistry remodeling upon stress, was investigated in the presence of polychlorinated biphenyls (PCBs), highly recalcitrant and phytotoxic compounds, highlighting its role in reshaping the nutritional and signaling features of the root niche to accommodate PCB-degrading microorganisms. Results: Arabidopsis exposure to 70 µM PCB-18 triggered plant-detrimental effects, stress-related traits, and PCB-responsive gene expression, reproducing PCB phytotoxicity. The root exudates of plantlets exposed for 2 days to the pollutant were collected and characterized through untargeted metabolomics analysis by liquid chromatography-mass spectrometry. Principal component analysis disclosed a different root exudation fingerprint in PCB-18-exposed plants, potentially contributing to the "cry-for-help" event. To investigate this aspect, the five compounds identified in the exudate metabolomic analysis (i.e., scopoletin, N-hydroxyethyl-b-alanine, hypoxanthine, L-arginyl-L-valine, and Lseryl-L-phenylalanine) were assayed for their influence on the physiology and functionality of the PCB-degrading strains Pseudomonas alcaliphila JAB1, Paraburkholderia xenovorans LB400, and Acinetobacter calcoaceticus P320. Scopoletin, whose relative abundance decreased in PCB-18-stressed plant exudates, hampered the growth and proliferation of strains JAB1 and P320, presumably due to its antimicrobial activity, and reduced the beneficial effect of Acinetobacter P320, which showed a higher degree of growth promotion in the scopoletin-depleted mutant f6'h1 compared to Arabidopsis WT plants exposed to PCB. Nevertheless, scopoletin induced the expression of the bph catabolic operon in strains JAB1 and LB400. The primary metabolites hypoxanthine, Larginyl-L-valine, and L-seryl-L-phenylalanine, which increased in relative abundance upon PCB-18 stress, were preferentially used as nutrients and growth-stimulating factors by the three degrading strains and showed a variable ability to affect rhizocompetence traits like motility and biofilm formation. Abiotic stresses, especially drought stress and salt stress in crop plants are accelerating due to climate change. The combined impact of drought and salt is anticipated to lead to the loss of up to 50% of arable land globally, resulting in diminished growth and substantial yield losses threatening food security. Addressing the challenges, agriculture through sustainable practices emerges as a potential solution to achieve Zero Hunger, one of the sustainable development goals set by the IUCN. Plants deploy a myriad of mechanisms to effectively address drought and salt stress with phytohormones playing pivotal roles as crucial signaling molecules for stress tolerance. The phytohormone auxin, particularly indole acetic acid (IAA) emerges as a paramount regulator integral to numerous aspects of plant growth and development. During both drought and salt stress conditions, auxin plays crucial roles for tolerance, but stress-induced processes lead to decreased levels of endogenous free auxin in the plant, leading to an urgent need for auxin production. With an aim to augment this auxin deficiency, several researchers have extensively investigated auxin production, particularly IAA by plant-associated microorganisms, including endophytic bacteria. These endophytic bacteria have been introduced into various crop plants subjected to drought or salt stress and potential isolates promoting plant growth have been identified. However, post-identification, essential studies on translational research to advance these potential isolates from the laboratory to the field are lacking. Plant-parasitic nematodes (PPNs) are among the most damaging pathogens to host plants. Plants can modulate their associated bacteria to cope with nematode infections. The tritrophic plant-nematode-microbe interactions are highly taxadependent, resulting in the effectiveness of nematode agents being variable among different host plants. Ficus tikoua is a versatile plant with high application potential for fruits or medicines. In recent years, a few farmers have attempted to cultivate this species in Sichuan, China, where parasitic nematodes are present. We used 16S rRNA genes to explore the effects of nematode parasitism on rootassociated bacteria in this species. Our results revealed that nematode infection had effects on both endophytic bacterial communities and rhizosphere communities in F. tikoua roots, but on different levels. The species richness increased in the rhizosphere bacterial communities of infected individuals, but the community composition remained similar as compared with that of healthy individuals. Nematode infection induces a deterministic assembly process in the endophytic bacterial communities of parasitized organs. Significant taxonomic and functional changes were observed in the endophytic communities of root knots. These changes were characterized by the enrichment of nitrogen-fixing bacteria, including Bradyrhizobium, Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium, and nematode-antagonistic bacteria, such as Pseudonocardia, Pseudomonas, Steroidobacter, Rhizobacter, and Ferrovibrio. Our results would help the understanding of the tritrophic plant-nematode-bacterium interactions in host plants other than dominant crops and vegetables and would provide essential information for successful nematode management when F. tikoua were cultivated on large scales. The formation of symbionts by using different combinations of endophytic bacteria, microalgae, and fungi to purify antibiotics-containing wastewater is an effective and promising biomaterial technology. As it enhances the mixed antibiotics removal performance of the bio-system, this technology is currently extensively studied. Using exogenous supplementation of various low concentrations of the phytohormone strigolactone analogue GR24, the removal of various antibiotics from simulated wastewater was examined. The performances of Chlorella vulgaris monoculture, activated sludge–C. vulgaris–Clonostachys rosea, Bacillus licheniformis–C. vulgaris–C. rosea, and endophytic bacteria (S395-2)–C. vulgaris–C. rosea co-culture systems were systematically compared. Their removal capacities for tetracycline, oxytetracycline, and chlortetracycline antibiotics from simulated wastewater were assessed. Chlorella vulgaris–endophytic bacteria–C. rosea co-cultures achieved the best performance under 0.25 mg L−1 antibiotics, which could be further enhanced by GR24 supplementation. This result demonstrates that the combination of endophytic bacteria with microalgae and fungi is superior to activated sludge–B. licheniformis–microalgae–fungi systems. As an indispensable part of insects, intestinal symbiotic bacteria play a vital role in the growth and development of insects and their adaptability. Rhoptroceros cyatheae, the main pest of the relict plant Alsophila spinulosa, poses a serious threat to the development of the A. spinulosa population. In the present study, 16S rDNA and internal transcribed spacer high-throughput sequencing techniques were used to analyze the structure of intestinal microbes and the diversity of the insect feeding on two different plants, as well as the similarities between the intestinal microorganisms of R. cyatheae. The dominant bacteria of leaf endophytes were also compared based on the sequencing data. The results showed that Proteobacteria, Firmicutes, and Actinobacteria were the dominant phyla of intestinal bacteria, and Ascomycota was the dominant phylum of intestinal fungi. Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium, Methylobacterium-Methylorubrum, and Enterococcus were the dominant genera in the intestine of R. cyatheae feeding on two plants, and the relative abundance was significantly different between the two groups. Candida was the common dominant genus of intestinal fungi in the two groups, and no significant difference was observed in its abundance between the two groups. This showed that compared with the intestinal fungi of R. cyatheae, the abundance of the intestinal bacteria was greatly affected by food. The common core microbiota between the microorganisms in A. spinulosa leaves and the insect gut indicated the presence of a microbial exchange between the two. The network correlation diagram showed that the gut microbes of R. cyatheae feeding on Gymnosphaera metteniana were more closely related to each other, which could help the host to better cope with the adverse external environment. Meloidogyne enterolobii is regarded as one of the most severe and threatening pathogens worldwide. Guava production in India is quenched by the invasion of root-knot nematode, Meloidogyne enterolobii. Bacterial bio-control agents confer resistance against nematodes by producing various bioactive molecules. Exploiting these biomolecules will lead to the development of novel nematicidal formulations for managing plant parasitic nematodes. Considering this, our present investigation was focused on the isolation and identification of endophytic bacterial communities from wild guava Psidium eugeniaefolia. Amongst various endophytes, Bacillus subtilis GEB-1 exhibited a 90% reduction in the population of M. enterolobii. Cell-free supernatant extracts of B. subtilis GEB-1 revealed the presence of seventeen unique biomolecules. Attempts were made to dock these biomolecules against major protein targets of M. enterolobii such as E3 ubiquitin ligase, Neurotransmitter-gated ion-channel, G protein-coupled receptor kinase, Peptidase M12B domain-containing protein, Hyaluronidase through molecular docking techniques. The results of in-silico studies revealed that all seventeen biomolecules had a significant impact on protein targets of M. enterolobii. Among other bacterial metabolites, 2-heptanone and 2,3-butanediol were found to have a greater binding affinity with protein targets of M. enterolobii. Furthermore, the in-vitro evaluation of 2-heptanone and 2,3-butanediol at different concentrations confirmed their nematicidal activity against eggs and second-stage juveniles of M. enterolobii. Hence our present study revealed that bacterial metabolites, 2 heptanone, and 2,3-butanediol can be explored as novel nematicidal molecules against guava root-knot nematodes, M. enterolobii. To obtain effective biocontrol strains for downy mildew of grape, 38 endophytic bacteria were isolated from fruits, seeds, and old stems of six grape varieties. Using spot inoculation mixtures of sporangial suspensions of Plasmopara viticola and biocontrol bacterial suspension, this screen yielded three strains (G1, G5, and G9) with good antagonistic effects against P. viticola. The growth inhibition rate was 100%, which was comparable to the effect of the positive control Bacillus subtilis strain CN181. The enzyme activity and the metabolites of strain G1 were examined on casein hydrolysate medium, sodium carboxymethyl cellulose agar plates, and chrome azurol sulfonate (CAS) agar plates. The antifungal protein component was identified by liquid chromatography–mass spectrometry (LC–MS). The results showed that strain G1 was more effective against Plasmopara viticola after two field trials, and the inhibition rates of strain G1 on the seventh day of the two field trials were 47.5% and 36.9%, respectively. Strain G1 was identified as Bacillus amyloliquefaciens based on morphological examination and 16S rDNA sequence analysis. It produced proteases, cellulases, and siderophores. Endophytes play an important role in plant development, survival, and establishment, but their temporal dynamics in young conifer plants are still largely unknown. In this study, the bacterial community was determined by metabarcoding of the 16S rRNA gene in the rhizoplane, roots, and aerial parts of 1- and 5-month-old seedlings of natural populations of Abies religiosa (Kunth) Schltdl. & Cham. In 1-month-old seedlings, Pseudomonas dominated aerial parts (relative abundance 71.6%) and roots (37.9%). However, the roots exhibited significantly higher bacterial species richness than the aerial parts, with the dissimilarity between these plant sections mostly explained by the loss of bacterial amplification sequence variants. After 5 months, Mucilaginibacter dominated in the rhizoplane (9.0%), Streptomyces in the roots (12.2%), and Pseudomonas in the aerial parts (18.1%). The bacterial richness and community structure differed significantly between the plant sections, and these variations were explained mostly by 1-for-1 substitution. The relative abundance of putative metabolic pathways significantly differed between the plant sections at both 1 and 5 months. All the dominant bacterial genera (e.g., Pseudomonas and Burkholderia-Caballeronia-Paraburkholderia) have been reported to have plant growth-promoting capacities and/or antagonism against pathogens, but what defines their role for plant development has still to be determined. The phyllosphere is an important but underestimated habitat for a variety of microorganisms, with limited knowledge about leaf endophytes as a crucial component of the phyllosphere microbiome. In this study, we investigated the mechanisms of communities and co-occurrence networks of leaf endophytes in response to forest thinning in a temperate forest. As we expected, contrasting responses of fungal and bacterial endophytes were observed. Specifically, the diversity of leaf endophytic fungi and the complexity of their co-occurrence networks increased significantly with thinning intensity, whereas the complexity of endophytic bacterial co-occurrence networks decreased. In particular, microbiota inhabiting damaged leaves seem to be more intensively interacting, showing an evident fungi–bacteria trade-off under forest thinning. In damaged leaves, besides the direct effects of thinning, thinning-induced changes in neighbor tree diversity indirectly altered the diversity of leaf fungal and bacterial endophytes via modifying leaf functional traits such as leaf dry matter content and specific leaf area. These findings provide new experimental evidence for the trade-offs between leaf endophytic fungi and bacteria under the different magnitudes of deforestation, highlighting their dependence on the presence or absence of leaf damage. Aims and background: The resurrection plant Myrothamnus flabellifolia tolerates complete desiccation and is a great model for studying how plants cope with extreme drought. Root-associated microbes play a major role in stress tolerance and are an attractive target for enhancing drought tolerance in staple crops. However, how these dynamics play out under the most extreme water limitation remains underexplored. This study aimed to identify bacterial and fungal communities that tolerate extreme drought stress in the bulk soil, rhizosphere, and endosphere of M. flabellifolia. Methods: High-throughput amplicon sequencing was used to characterise the microbial communities associated with M. flabellifolia. Results: The bacterial phyla that were most abundant across all compartments were Acidobacteriota, Actinobacteriota, Chloroflexota, Planctomycetota, and Pseudomonadota, while the most abundant fungal phyla were Ascomycota and Basidiomycota. Although the bulk soil hosted multiple beneficial root-associated microbes, the rhizosphere compartment showed the highest functional diversity of bacteria and fungi. In contrast, the endosphere exhibited a low abundance and diversity of microbes. These findings share consistent with the theory that M. flabellifolia recruits soil microbes from the bulk to the rhizosphere and finally to the endosphere. It is possible that these microbes could promote drought tolerance in associated plant tissues. Conclusion: We find that compartments act as the major driver of microbial diversity, but the soil physicochemical factors also influence microbial composition. Aims: Milpas are traditional agroecosystems found in rural areas of Mexico, where low input practices favor the presence of microbial associates with greater abundance and diversity than modern crops. Milpas can be found in a wide range of climates, but the beneficial functions of microbes are yet to be explored across biotic and abiotic conditions. In semi-arid regions of Hidalgo, Mexico, maize landraces have been selected to grow in milpas despite low rainfall and we hypothesized that associated bacteria contribute to the drought tolerance of these plants. Methods: We collected maize seeds from arid and tropical milpas, as well as hybrid varieties from modern crops to test their responses to drought during germination and early vegetative growth; next, we studied the role of seed endophytic bacteria for this trait. Results: Landraces from arid milpas displayed decreased phenotypic responses to drought (i.e. increased tolerance), and elimination of seed-endophytic bacteria from these landraces reduced germination in drought. Next, 16 S amplicon sequencing revealed that drought-tolerant landraces harbor Pseudomonas spp., Brachybacterium spp., and strains from the Bacilli class that were not found in drought-sensitive varieties; likewise, culturing methods allowed the isolation of these bacterial groups. Finally, inoculation of Pseudomonas spp. and Brachybacterium sp. strains improved the germination of a drought-sensitive variety. Conclusions: Our results indicate that seed-endophytic bacteria associated to maize landraces from arid milpas contribute to drought tolerance in early developmental stages and support that ancestral practices manifest in the microbial ecology of milpas, selecting beneficial microbes to cope with local conditions. Simple Summary: The emerald ash borer (EAB, Agrilus planipennis Fairmaire) is currently a highly destructive forest pest, causing substantial economic losses. Ash species (Fraxinus spp.) are economically invaluable components of natural forests and urban environments. However, these trees have been severely damaged by EAB. Endophytes, which are prevalent in plants, are hypothesized to contribute to the complex relationships between insects and plants. However, research on the effect of EAB infestation on the community changes in endophytic fungi and bacteria in phloem of Fraxinus is scarce. To bridge this gap, we compare changes in fungi and bacteria diversity, community makeup, and the potential roles of various endophytic fungi and bacteria in both EAB-infested and uninfected Fraxinus bungeana trees. We observed an elevation in bacterial richness, without notable changes in diversity, whereas fungal richness and diversity remained unaffected. Furthermore, we identified four key microbial genera undergoing substantial shifts post-infestation. The functional roles of endophytic fungi and bacteria also exhibited changes, with a decline in beneficial activities and an emergence of potentially detrimental functions. Network analyses have shown elevated negative correlations and core genera shift in EAB-infected phloem. The findings of our study contribute to a better understanding of the complex interactions between plants, insects, and endophytic microorganisms. Endophytes, prevalent in plants, mediate plant–insect interactions. Nevertheless, our understanding of the key members of endophyte communities involved in inhibiting or assisting EAB infestation remains limited. Employing ITS and 16S rRNA high-throughput sequencing, along with network analysis techniques, we conducted a comprehensive investigation into the reaction of endophytic fungi and bacteria within F. bungeana phloem by comparing EAB-infested and uninfected samples. Our findings reveal that EAB infestation significantly impacts the endophytic communities, altering both their diversity and overall structure. Interestingly, both endophytic fungi and bacteria exhibited distinct patterns in response to the infestation. For instance, in the EAB-infested phloem, the fungi abundance remained unchanged, but diversity decreased significantly. Conversely, bacterial abundance increased, without significant diversity changes. The fungi community structure altered significantly, which was not observed in bacteria. The bacterial composition in the infested phloem underwent significant changes, characterized by a substantial decrease in beneficial species abundance, whereas the fungal composition remained largely unaffected. In network analysis, the endophytes in infested phloem exhibited a modular topology, demonstrating greater complexity due to an augmented number of network nodes, elevated negative correlations, and a core genera shift compared to those observed in healthy phloem. One of the challenges of agromining is the adoption of more environmentally‐friendly solutions to improve plant biomass yields and Ni concentrations in plants. Here, we focused on four sustainable solutions for optimizing nickel phytoextraction by the hyperaccumulator Odontarrhena chalcidica: a biostimulant, another biostimulant/plant defense stimulator, artificial root exudates, and a biodegradable metal chelator. Their effects on the growth and physiology of O. chalcidica, on Ni phytoextraction capacity, on physicochemical soil characteristics, and on the diversity of rhizosphere and endophytic bacteria were compared to a conventional mineral fertilizer. A 5‐month pot experiment was carried out with O. chalcidica growing on an ultramafic soil. Element concentrations in both soil and plant were measured. Moreover, numerous compounds were analyzed (photosynthetic pigments, malondialdehyde, flavonoids, free amino acids, and starch). We also characterized rhizosphere and endophytic bacterial communities associated with this hyperaccumulator. Biostimulants appeared to be a promising way of improving Ni concentration in shoots and plant biomass production, and showed a positive effect on bacterial richness and diversity. In contrast, our experiments did not show that artificial exudates and mineral fertilizer had a positive effect on Ni phytoextraction. Finally, the biodegradable chelator had no significant effect. The use of sustainable amendments into a Ni agromining system improved both plant biomass and Ni yields, in comparison to mineral fertilization. The biomass filtrate of the endophytic bacterial strain Bacillus amyloliquefaciens Fa.2 was utilized for the eco-friendly production of silver nanoparticles (Ag-NPs). The yellowish-brown color's optical properties showed a maximum surface plasmon resonance at 415 nm. The morphological and elemental composition analysis reveals the formation of spherical shapes with sizes of 5–40 nm, and the Ag ion comprises the major component of the produced Ag-NPs. X-ray diffraction confirmed the crystalline structure, whereas dynamic light scattering reveals the high stability of synthesized Ag-NPs with a polydispersity index of 0.413 and a negative zeta potential value. The photocatalytic experiment showed the efficacy of Ag-NPs to degrade methylene blue with maximum percentages of 73.9 ± 0.5 and 87.4 ± 0.9% under sunshine and UV irradiation, respectively, compared with 39.8% under dark conditions after 210 min. Additionally, the reusability of Ag-NPs was still more active for the fifth run, with a percentage decrease of 11.6% compared with the first run. Interestingly, the biogenic Ag-NPs showed superior antimicrobial activity against different pathogenic Gram-negative bacteria (MIC = 6.25 µg mL−1), Gram-positive bacteria (MIC = 12.5 µg mL−1), and uni- and multicellular fungi (MIC = 12.5 µg mL−1). Moreover, the biosynthesized Ag-NPs could target cancer cells (Pc3 and Mcf7) at low concentrations compared with normal cell (Vero) lines. The IC50 of normal cells is 383.7 ± 4.1 µg mL−1 compared with IC50 Pc3 (2.5 ± 3.5 µg mL−1) and McF7 (156.1 ± 6.8 µg mL−1). The effects of different nitrogen (N) application levels on seed endophytes and grain nutritional quality are not yet clear. The impact of four N application levels on endophytes and amino acid accumulation in purple rice seeds was examined using 16S rRNA and ITS amplicon sequencing technology. This study integrates 16S rRNA, ITS amplicon sequencing technology and amino acid‐targeted detection to explore the effects of four different nitrogen application levels (0 kg hm−2, Y1N0; 180 kg hm−2, Y1N1; 270 kg hm−2, Y1N2; 360 kg hm−2, Y1N3) on the accumulation of endophytic bacteria, fungi and amino acid content in purple rice seeds and their interaction mechanisms. The findings indicated an increase in the contents of most amino acids with increasing N application. The dominant bacterial species in the community were mainly from the phyla Proteobacteria and Actinobacteriota, while the dominant fungal species were from the phyla Ascomycota and Basidiomycota. There was a significant difference in the richness of endophytic fungal communities between Y1N0 and Y1N2. Y1N1 showed significant differences in Mucoromycota compared to Y1N3. The quantity of operational taxonomic units (OTUs) in the bacterial and fungal community co‐occurrence network increased with increasing N fertilizer, showing strong correlations with Sporidiobolus, Chaetomium, Humicola, Botryotrichum, Ophiosphaeria and Dioszegia for most amino acids. These findings indicate that a high amount of N fertilizer greatly increases amino acid contents in purple rice seeds and improves the diversity and stability of endophytic fungal populations. The importance of microorganisms residing within the host plant for their growth and health is increasingly acknowledged, yet the significance of microbes associated with seeds, particularly seed endophytic bacteria, remains underestimated. Seeds harbor a wide range of bacteria that can boost the growth and resilience of their host plants against environmental challenges. These endophytic associations also offer advantages for germination and seedling establishment, as seed endophytic bacteria are present during the initial stages of plant growth and development. Furthermore, plants can selectively choose bacteria possessing beneficial traits, which are subsequently transmitted through seeds to confer benefits to future generations. Interestingly, even with the ongoing discovery of endophytes in seeds through high-throughput sequencing methods, certain endophytes remain challenging to isolate and culture from seeds, despite their high abundance. These challenges pose difficulties in studying seed endophytes, making many of their effects on plants unclear. In this article, a framework for understanding the assembly and function of seed endophytes, including their sources and colonization processes was outlined in detail and available research on bacterial endophytes discovered within the seeds of various plant species has also been explored. Thus, this current review aims to provide valuable insights into the mechanism of underlying seed endophytic bacteria-host plant interactions and offers significant recommendations for utilizing the seed endophytic bacteria in sustainable agriculture as plant growth promoters and enhancers of environmental stress tolerance. Jojoba shrubs are wild plants cultivated in arid and semiarid lands and characterized by tolerance to drought, salinity, and high temperatures. Fungi associated with such plants may be attributed to the tolerance of host plants against biotic stress in addition to the promotion of plant growth. Previous studies showed the importance of jojoba as jojoba oil in the agricultural field; however, no prior study discussed the role of jojoba-associated fungi (JAF) in reflecting plant health and the possibility of using JAF in biocontrol. Here, the culture-independent and culture-dependent approaches were performed to study the diversity of the jojoba-associated fungi. Then, the cultivable fungi were evaluated for in-vitro antagonistic activity and in vitro plant growth promotion assays. The metagenome analysis revealed the existence of four fungal phyla: Ascomycota, Aphelidiomycota, Basidiomycota, and Mortierellomycota. The phylum Ascomycota was the most common and had the highest relative abundance in soil, root, branch, and fruit samples (59.7%, 50.7%, 49.8%, and 52.4%, respectively). Alternaria was the most abundant genus in aboveground tissues: branch (43.7%) and fruit (32.1%), while the genus Discosia had the highest abundance in the underground samples: soil (24%) and root (30.7%). For the culture-dependent method, a total of 14 fungi were isolated, identified, and screened for their chitinolytic and antagonist activity against three phytopathogenic fungi (Fusarium oxysporum, Alternaria alternata and Rhizoctonia solani) as well as their in vitro plant growth promotion (PGP) activity. Based on ITS sequence analysis, the selected potent isolates were identified as Aspergillus stellatusEJ-JFF3, Aspergillus flavus EJ-JFF4, Stilbocrea sp. EJ-JLF1, Fusarium solani EJ-JRF3, and Amesia atrobrunneaEJ-JSF4. The endophyte strain A. flavus EJ-JFF4 exhibited the highest chitinolytic activity (9 Enzyme Index) and antagonistic potential against Fusarium oxysporum, Alternaria alternata, and Rhizoctonia solani phytopathogens with inhibitory percentages of 72, 70, and 80 respectively. Also, A. flavus EJ-JFF4 had significant multiple PGP properties, including siderophore production (69.3%), phosphate solubilization (95.4 µg ml−1). The greatest production of Indol-3-Acetic Acid was belonged to A. atrobrunnea EJ-JSF4 (114.5 µg ml−1). The analysis of FUNGuild revealed the abundance of symbiotrophs over other trophic modes, and the guild of endophytes was commonly assigned in all samples. For the first time, this study uncovered fungal diversity associated with jojoba plants using a culture-independent approach and in-vitro assessed the roles of cultivable fungal strains in promoting plant growth and biocontrol. Gluconacetobacter diazotrophicus is a diazotrophic endophytic bacterium that promotes the growth and development of several plant species. However, the molecular mechanisms activated during plant response to this bacterium remain unclear. Here, we used the RNA-seq approach to understand better the effect of G. diazotrophicus PAL5 on the transcriptome of shoot and root tissues of Arabidopsis thaliana. G. diazotrophicus colonized A. thaliana roots and promoted growth, increasing leaf area and biomass. The transcriptomic analysis revealed several differentially expressed genes (DEGs) between inoculated and non-inoculated plants in the shoot and root tissues. A higher number of DEGs were up-regulated in roots compared to shoots. Genes up-regulated in both shoot and root tissues were associated with nitrogen metabolism, production of glucosinolates and flavonoids, receptor kinases, and transcription factors. In contrast, the main groups of down-regulated genes were associated with pathogenesis-related proteins and heat-shock proteins in both shoot and root tissues. Genes encoding enzymes involved in cell wall biogenesis and modification were down-regulated in shoots and up-regulated in roots. In contrast, genes associated with ROS detoxification were up-regulated in shoots and down-regulated in roots. Drunken horse grass (Achnatherum inebrians (Hance) Keng) is a toxic perennial bunchgrass native to Northwestern China. Epichloë endophytic fungi infection could enhance the stress tolerance of drunken horse grass. However, there is a scarcity of literature regarding the effects of intraspecific competition. As a result, we anticipated that the intraspecific competitive dynamics between endophyte-infected (EI) and endophyte-free (EF) plants would become more transparent for four years by planted as the proportions of 2: 1, 1: 1, and 1: 2. The results showed the EI plants exhibited more biomass, seed yields, and survival rates than EF plants. Endophyte infection also facilitated a competitive advantage by enhancing photosynthesis and soil nutrition. Our findings constituted the inaugural investigation into the influence of the intraspecific competitive ability of grass infected with Epichloë endophyte fungi. EI plants caused them to become stronger and stronger, while EF became weaker and weaker by timing and planting proportion increasing, and EF drunken horse grass could be replaced by EI. These conclusions were instrumental in elucidating why the endophytic fungal infection rate of drunken horse grass is 100% observed in natural wilderness. Epichloë endophyte could reduce plant diversity and enhance the dominance of EI plants in intraspecific competition; drunken horse grass may be threatening the persistence of native plant species. Bacterial endophytes from plants harbor diverse metabolites that play major roles in biocontrol and improve plant growth. In this study, a total of 12 endophytic bacteria were isolated from the ginger rhizome. The strain K3 was highly effective in preventing mycelia growth of Pythium myriotylum (78.5 ± 1.5% inhibition) in dual culture. The cell‐free extract (2.5%) of endophyte K3 inhibited 76.3 ± 4.8% mycelia growth, and 92.4 ± 4.2% inhibition was observed at a 5% sample concentration. The secondary metabolites produced by Bacillus licheniformis K3 showed maximum activity against Pseudomonas syringae (24 ± 1 mm zone of inhibition) and Xanthomonas campestris (28 ± 3 mm zone of inhibition). The strain K3 produced 28.3 ± 1.7 IU mL−1 protease, 28.3 ± 1.7 IU mL−1 cellulase, and 2.04 ± 0.13 IU mL−1 chitinase, respectively. The ginger rhizome treated with K3 in the greenhouse registered 53.8 ± 1.4% soft rot incidence, and the streptomycin‐treated pot registered 78.3 ± 1.7% disease incidence. The selected endophyte K3 improved ascorbate peroxidase (1.37 ± 0.009 µmole ASC min−1 mg−1 protein), catalase (8.7 ± 0.28 µmole min−1 mg−1 protein), and phenylalanine ammonia‐lyase (26.2 ± 0.99 Umg−1) in the greenhouse. In addition, K3 treatment in the field trial improved rhizome yield (730 ± 18.4 g) after 180 days (p < 0.01). The shoot length was 46 ± 8.3 cm in K3‐treated plants, and it was about 31% higher than the control treatment (p < 0.01). Seed endophytes in maize, which facilitate the transmission of microorganisms from one plant generation to the next, may play a crucial role in plant protection and growth promotion. This study aimed to investigate the effects of various maize varieties on the communities of endophytic bacteria in seeds and germinating roots. This study utilized Illumina high-throughput sequencing technology to examine the structural and diversity differences of endophytic bacterial communities within seed maize (BY1507), silage maize (QQ446), and wild maize (Teosinte) in both seeds and germinating roots. The results showed that 416 bacterial genera were detected, with Pantoea, Lachnospiraceae, Pararhizobium, Enterobacteriaceae, Stenotrophomonas, and Pseudonocardia being the most prevalent (relative abundance > 10%) at the genus level. No significant difference was observed in diversity indices (Chao1, ACE, Shannon, and Simpson) of seed endophytes among BY1507, QQ446, and Teosinte. The Shannon and Simpson indices for the germinating root endophyte from the wild variety (Teosinte) were significantly higher than the domesticated varieties (BY1507 and QQ446). PCoA revealed a notable overlap in the endophytic bacterial communities from the seeds of BY1507, QQ446, and Teosinte. Yet, clustering patterns were found. Co-occurrence network analysis showed that BY1507, QQ446, and Teosinte share a notable proportion of shared endophytic bacteria (>30%) between the seeds and germinating roots. This investigation elucidates the characteristics of endophytic microbial communities of seeds and germinating roots with seed maize, silage maize, and wild maize, offering data for future research on the physiological ecological adaptation of these endophytic microbial communities. Magnaporthe oryzae is one of the most important fungal pathogens of rice. Chitin and avirulent strains can induce two layers of immunity response, pathogen-associated molecular pattern (PAMP)-triggered immunity (PTI) and effector-triggered immunity (ETI), in rice with cognate R genes. However, little is known about the assembly of the rice microbiome induced by PTI and ETI in rice. In this study, we investigate the impact of continuous treatment of the avirulent M. oryzae strain with AvrPi9 and chitin on the bacterial endophytic community of rice varieties harboring resistant gene Pi9 and their antagonistic activity against rice blast fungus. Analysis of the 16S rRNA showed a significant increase in the diversity and microbial co-occurrence network complexity and the number of beneficial taxa—Bacillus, Pseudomonas, Microbacterium, and Stenotrophomonas spp.—following the chitin and avirulent strain treatments. The antifungal assay with bacterial endophytes recovered from the leaves showed few bacteria with antagonistic potential in rice treated with avirulent strains, suggesting that the sequential treatment of the avirulent strain decreased the antagonistic bacteria against M. oryzae. Moreover, we identified Bacillus safensis Ch_66 and Bacillus altitudinis Nc_68 with overall antagonistic activities in vivo and in vitro. Xylella fastidiosa is the causal agent of several plant diseases affecting fruit and nut crops. Methylobacterium mesophilicum strain SR1.6/6 was isolated from Citrus sinensis and shown to promote plant growth by producing phytohormones, providing nutrients, inhibiting X. fastidiosa, and preventing Citrus Variegated Chlorosis. However, the molecular mechanisms involved in the interaction among these microbes are still unclear. The present work aimed to analyze physiological and molecular aspects of M. mesophilicum SR1.6/6 and X. fastidiosa 9a5c in co-culture. The transcriptome and secretome analyses indicated that X. fastidiosa down-regulates cell division and transport genes and up-regulates stress via induction of chaperones and pathogenicity-related genes including, the lipase-esterase LesA, a protease, as well as an oligopeptidase in response to M. mesophilicum competition. On the other hand, M. mesophilicum also down-regulated transport genes, except for iron uptake, which was up-regulated. Secretome analysis identified four proteins in M. mesophilicum exclusively produced in co-culture with X. fastidiosa, among these, three are related to phosphorous uptake. These results suggest that M. mesophilicum inhibits X. fastidiosa growth mainly due to nutrient competition for iron and phosphorous, thus promoting X. fastidiosa starvation, besides producing enzymes that degrade X. fastidiosa cell wall, mainly hydrolases. The understanding of these interactions provides a direction for control and management of the phytopathogen X. fastidiosa, and consequently, helps to improve citrus growth and productivity.

Abstract There is an enormous diversity of life forms present in the extremely intricate marine environment. The growth and development of seaweeds in this particular environment are controlled by the bacteria that settle on their surfaces and generate a diverse range of inorganic and organic chemicals. The purpose of this work was to identify epiphytic and endophytic bacterial populations associated with ten common marine macroalgae from various areas along the Mediterranean Sea coast in Alexandria. This was done to target their distribution and possible functional aspects. Examine the effects of the algal habitat on the counting and phenotypic characterization of bacteria, which involves grouping bacteria based on characteristics such as shape, colour, mucoid nature, type of Gram stain, and their ability to generate spores. Furthermore, studying the physiological traits of the isolates under exploration provides insight into the optimum environmental circumstances for bacteria associated with the formation of algae. The majority of the bacterial isolates exhibited a wide range of enzyme activities, with cellulase, alginase, and caseinase being the most prevalent, according to the data. Nevertheless, 26% of the isolates displayed amylolytic activity, while certain isolates from Miami, Eastern Harbor, and Montaza lacked catalase activity. Geographical variations with the addition of algal extract may impact on the enumeration of the bacterial population, and this might have a relationship with host phylogeny. The most significant observation was that endophytic bacteria associated with green algae increased in all sites, while those associated with red algae increased in Abu Qir and Miami sites and decreased in Eastern Harbor. At the species level, the addition of algal extract led to a ninefold increase in the estimated number of epiphytic bacteria for Cladophora pellucida in Montaza. Notably, after adding algal extract, the number of presented endophytic bacteria associated with Codium sp. increased in Abu Qir while decreasing with the same species in Montaza. In addition to having the most different varieties of algae, Abu Qir has the most different bacterial isolates.

Abstract Background Biotechnology provides a cost-effective way to produce nanomaterials such as silver oxide nanoparticles (Ag2ONPs), which have emerged as versatile entities with diverse applications. This study investigated the ability of endophytic bacteria to biosynthesize Ag2ONPs. Results A novel endophytic bacterial strain, Neobacillus niacini AUMC-B524, was isolated from Lycium shawii Roem. & Schult leaves and used to synthesize Ag2ONPS extracellularly. Plackett–Burman design and response surface approach was carried out to optimize the biosynthesis of Ag2ONPs (Bio-Ag2ONPs). Comprehensive characterization techniques, including UV–vis spectral analysis, Fourier transform infrared spectroscopy, transmission electron microscopy, X-ray diffraction, dynamic light scattering analysis, Raman microscopy, and energy dispersive X-ray analysis, confirmed the precise composition of the Ag2ONPS. Bio-Ag2ONPs were effective against multidrug-resistant wound pathogens, with minimum inhibitory concentrations (1–25 µg mL−1). Notably, Bio-Ag2ONPs demonstrated no cytotoxic effects on human skin fibroblasts (HSF) in vitro, while effectively suppressing the proliferation of human epidermoid skin carcinoma (A-431) cells, inducing apoptosis and modulating the key apoptotic genes including Bcl-2 associated X protein (Bax), B-cell lymphoma 2 (Bcl-2), Caspase-3 (Cas-3), and guardian of the genome (P53). Conclusions These findings highlight the therapeutic potential of Bio-Ag2ONPs synthesized by endophytic N. niacini AUMC-B524, underscoring their antibacterial efficacy, anticancer activity, and biocompatibility, paving the way for novel therapeutic strategies.

Abstract The formation of symbionts by using different combinations of endophytic bacteria, microalgae, and fungi to purify antibiotics-containing wastewater is an effective and promising biomaterial technology. As it enhances the mixed antibiotics removal performance of the bio-system, this technology is currently extensively studied. Using exogenous supplementation of various low concentrations of the phytohormone strigolactone analogue GR24, the removal of various antibiotics from simulated wastewater was examined. The performances of Chlorella vulgaris monoculture, activated sludge–C. vulgaris–Clonostachys rosea, Bacillus licheniformis–C. vulgaris–C. rosea, and endophytic bacteria (S395-2)–C. vulgaris–C. rosea co-culture systems were systematically compared. Their removal capacities for tetracycline, oxytetracycline, and chlortetracycline antibiotics from simulated wastewater were assessed. Chlorella vulgaris–endophytic bacteria–C. rosea co-cultures achieved the best performance under 0.25 mg L−1 antibiotics, which could be further enhanced by GR24 supplementation. This result demonstrates that the combination of endophytic bacteria with microalgae and fungi is superior to activated sludge–B. licheniformis–microalgae–fungi systems. Exogenous supplementation of GR24 is an effective strategy to improve the performance of antibiotics removal from wastewater.
