Zebrafish (Danio rerio) are currently used for various biomedical and environmental studies. Due to the comprehensive ethological and genetical description, but also due to a significant similarity to humans, zebrafish are successfully used in modelling some traits associated with human pathology (both psychiatric and others) and determining the effects of environmental factors on behaviour and highly conserved molecular processes. Due to the fact that chronic exposure to stress is a major problem for contemporary society, any description of the phenomenon and finding solutions to minimize its harmful effects are important research directions. Therefore, through this study, we aimed to evaluate and describe some behavioural and oxidative changes that occur following exposure to chronic stress, in an adapted animal model. Twenty zebrafish were acclimatized to laboratory conditions and randomly divided into 2 study groups: a control group and a group that was exposed to a combination of stressors for 10 consecutive days. Following stress exposure, locomotor activity and affective state were assessed. Afterwards, biological samples were collected and used to determine some oxidative status parameters (the enzymatic activity of superoxide dismutase, of glutathione peroxidase, and the amount of malondialdehyde). We obtained significant variations of some behavioural parameters suggesting a significant harmful effect of chronic stress on the locomotor activity. Also, manifestations of anxious-depressive behaviours were observed, as compared to the control group. Biochemical analyses revealed significant changes in the activity of the antioxidant enzymes and increased lipid peroxidation which suggests that exposure to chronic stress could lead to the occurrence of oxidative stress. The exposure of adult fish to warm or high temperatures is known to impair reproduction, yet the long-term reproductive impacts for treatments at early life are not well clarified. This study aimed to evaluate the effects of warm temperature (WT) during juvenile stage on gonad maturation, gamete quality, and offspring thermotolerance in rainbow trout. While the comparison of basic reproductive parameters in WT females did not reveal any kind of impairment, many WT males showed an atrophied, undeveloped gonad, or a smaller testis with lower milt volume; sperm quality parameters in WT males and deformity rates in the respective progeny were also highly affected. However, despite of such negative effects, many of the remaining progeny presented better rates of survival and growth when exposed to the same conditions as those of parental fish (WT), suggesting that thermal stress in parr stage males elicited intergenerational thermotolerance after a single generation. The present results support that prolonged warming stress during early life stages can adversely affect key reproductive aspects, but contrastingly increase offspring performance at upper thermal ranges. The cumulative effects of non-lethal stressors on the health of biodiversity are a primary concern for conservation, yet difficulties remain regarding their quantification. In mammals, many stressors are processed through a common stress-response pathway, and therefore epigenetic changes in genes of this pathway may provide a powerful tool for quantifying cumulative effects. As a preliminary assessment of this approach, we investigated epigenetic manifestations of stress in two killer whale populations with different levels of exposure to anthropogenic stressors. We used bisulfite amplicon sequencing to compare patterns of DNA methylation at 25 CpG sites found in three genes involved in stress response and identified large differences in the level of methylation at two sites consistent with differential stress exposure between Northern and Southern Resident killer whale populations. Abstract In the study, zebrafish were exposed to 0 and 200 μg/L Zn for 8 weeks, and then both groups were transferred to water including 0, 100, and 200 μg/L Cd for 4 days, respectively. Acute Cd exposure caused negative effects on stress defense, immune, and metal transport systems by increasing lipid peroxidation, iNOS activity and mRNA levels of il-6 and inos , and decreasing Cu/Zn-SOD and HSP70 levels, and mRNA levels of sod1 , cat , hsp70 , p65 , mtf-1 , znt5 , zip7 , atp7a , and atp7b. Lipid peroxidation was significantly reduced by Zn pre-exposure under Cd exposure, which may be explained by the enhanced stress defense capacity and the weaken inflammatory response. Firstly, Zn pre-exposure increased MTs and HSP70 levels and CAT activity in Cd-free water, which may facilitate fish quick response to Cd. Secondly, Zn pre-exposure reduced Cd accumulation at 100 and 200 μg/L Cd, down-regulated il-6 and il-1β at 100 μg/L Cd and p65 at 200 μg/L Cd, and increased Cu/Zn-SOD and CAT activities at 200 μg/L Cd. Thirdly, Zn pre-exposure alone up-regulated transcription factors (hsf1 , hsf2 , and mtf-1 , and nrf2) and their target genes (sod1 , cat, hsp70 , and mt2) under Cd exposure in a dose-dependent manner. It should be noted that Zn pre-exposure down-regulated several metal transport genes dramatically at 0 and 100 μg/L Cd, which may be an important mechanism for reducing Cd import into livers. Overall, long-term and environmental Zn pre-exposure mitigated Cd toxicity by the enhanced stress defense capacity and the down-regulated metal transport and inflammatory responses. Fluoxetine (FLX) is a pharmaceutical used to treat affective disorders in humans, but as environmental contaminant also affects inadvertently exposed fish in urban watersheds. In humans and fish, acute FLX treatment and exposure are linked to endocrine disruption, including effects on the reproductive and stress axes. Using the zebrafish model, we build on the recent finding that developmental FLX exposure reduced cortisol production across generations, to determine possible parental and/or life-stage-dependent (age and/or breeding experience) contributions to this phenotype. Specifically, we combined control and developmentally FLX-exposed animals of both sexes (F0) into four distinct breeding groups mated at 5 and 9 months, and measured offspring (F1) basal cortisol at 12 dpf. Basal cortisol was lower in F1 descended from developmentally FLX-exposed F0 females bred at 5, but not 9 months, revealing a maternal, life-stage dependent effect. To investigate potential molecular contributions to this phenotype, we profiled maternally deposited transcripts involved in endocrine stress axis development and regulation, epigenetic (de novo DNA methyltransferases) and post-transcriptional (miRNA pathway components and specific miRNAs) regulation of gene expression in unfertilized eggs. Maternal FLX exposure resulted in decreased transcript abundance of glucocorticoid receptor, dnmt3 paralogues and miRNA pathway components in eggs collected at 5 months, and increased transcript abundance of miRNA pathway components at 9 months. Specific miRNAs predicted to target stress axis transcripts decreased (miR-740) or increased (miR-26, miR-30d, miR-92a, miR-103) in eggs collected from FLX females at 5 months. Increased abundance of miRNA-30d and miRNA-92a persisted in eggs collected from FLX females at 9 months. Clustering and principal component analyses of egg transcript profiles separated eggs collected from FLX-females at 5 months from other groups, suggesting that oocyte molecular signatures, and miRNAs in particular, may serve as predictive tools for the offspring phenotype of reduced basal cortisol in response to maternal FLX exposure. Abstract Environmental factors, including drugs and diet, have been shown to affect the microbiome composition. However, whether physical factors affect the gut microbiome composition of fish and whether this change affects fish growth are unknown. The response of gut microorganisms to physical factors such as crowding was assessed by examining the gut microbiome and metabolome of fish from a high-density group and a low-density group. During the 90-day growth period, nine genera of gut microbes were significantly changed between the two groups. In addition, 32 metabolites were significantly changed between the two groups. The correlation analysis of differentially abundant gut microbes and metabolites allowed the identification of eight gut microbes, 13 metabolites and enrichment of seven metabolic pathways. Among the enriched pathways, the pentose phosphate pathway played potential role in the adaption of crowding stress and regulation of glucose metabolism. The response of the pentose phosphate pathway to crowding stress was further determined by evaluating key metabolites, biochemical changes, and the expression of a rate-controlling enzyme in this pathway. Our results provided new evidence that the gut microbiome might be involved in the response to crowding and consequently to the adaptation of fish to environmental stressors via the pentose phosphate pathway. Juvenile olive flounders, Paralichthys olivaceus (mean weight 2.69 ± 0.31 g), were raised in bio-floc and seawater for six months, these P. olivaceus (mean weight 280.1 ± 10.5 g, mean length 28.37 ± 2.3 cm) were exposed to different concentrations of waterborne nitrite (0, 25, 50, 100, and 200 mg NO 2 − /L) for 7 days. None of the P. olivaceus individuals exposed to bio-floc and seawater containing waterborne nitrite concentrations of 200 mg/L for 7 days survived. Hematological parameters (hemoglobin and hematocrit) were significantly reduced by nitrite exposure. Regarding plasma components, the concentrations of glucose, glutamic oxalate transaminase (GOT), and glutamic pyruvate transaminase (GPT) increased significantly in response to nitrite exposure, whereas cholesterol concentrations significantly decreased. Stress indicators, including concentrations of plasma glucose, cortisol, and liver and gill concentrations of heat shock protein 70 (HSP70) were significantly increased by nitrite exposure. The results of the study indicate that nitrite exposure affected the hematological parameters and stress indicators of P. olivaceus raised in bio-floc and seawater, and these changes were more prominent in the P. olivaceus raised in seawater than those raised in bio-floc. Anesthesia plays a critical role in prevention of mechanical injuries and manipulation stress in fish. Commercially reared fish are subjected to confinement and handling including netting, weighing, sorting, vaccination, and transport. In research, fish may also undergo surgical procedures ranging from tagging, blood sampling, and small incisions for invasive procedures. In these situations, treatment with anesthetic agents may be necessary to ensure the welfare of the fish. In this review, we provide information on optimal dosage and effects on fish physiology of three widely-used anesthetics worldwide: clove oil, tricaine methane sulphate, and 2-phenoxyethanol. When fish perceive stressful scenarios, their hypothalamus-pituitary-interrenal axis is activated resulting in the release of corticotropin releasing hormone, adrenocorticotropic hormone (ACTH), and finally cortisol. The physiologic stress response of fish has most often been linked to the reduced performance of the immune system, with a few exceptions where the immune system is activated. In this report, we tested the hypothesis that oxidative burst activity levels in juvenile Chinook salmon ( Oncorhynchus tshawytscha ) are altered when the fish is presented with a stressor. Fish were subjected to a stressor for 3 h and then allowed to recover for 20 h following the stressor. Plasma and spleens were collected from euthanized fish before the stressor, at the end of a 3 h stressor, and 23 h after the start of the experiment. Plasma was held at −80 °C until cortisol radioimmunoassay analysis was performed to confirm stress. Spleens were held in Dulbecco's Modified Eagle Medium overnight and analyzed the day following collection. Oxidative burst activity was measured in splenic leukocytes after being stimulated with phorbol 12-myristate 13-acetate. We found a significant increase in activated oxidative burst from fish subjected to the stressor as compared to unstressed fish. Abstract: This study sought to determine the optimal temperature(s) for aquaculture of juvenile red‐spotted grouper Epinephelus akaara (Temminck & Schlegel) (mean initial BW: 3.1 g). Growth performance, insulin‐like growth factor 1 (IGF‐1) expression and thermal stress responses (plasma cortisol, glucose, and hepatic heat shock protein 60 expression) were evaluated at three constant temperatures (24°C, 26°C and 28°C) in a 2‐week trial. At the end of the trial, final BW was significantly higher at 26°C and 28°C than at 24°C (p < 0.05); a quadratic regression analysis of final BW showed the optimum temperature for growth was 27.5°C (p < 0.05, R2 = 0.806). The highest hepatic IGF‐1 expression was observed at 26°C (p < 0.05). On the other hand, hepatic heat shock protein 60 expression was highest at 28°C (p < 0.05), suggesting thermal stress. In conclusion, temperature optima, which support excellent growth but induce minimal thermal stress, was 26°C. This fine information within a narrow temperature range is expected to give empirical information for red‐spotted grouper farmers to sustain maximal production efficiency with avoiding thermal stress and to determine the future location of production, especially in consideration of arising seawater temperatures. Abstract: Stocking density is an important factor affecting growth in aquaculture. The main objectives of this study are to evaluate the effects of different stocking densities on Holothuria arguinensis and Holothuria mammata's growth and determine the optimal density. Four different stocking densities were selected for H. arguinensis (1, 3, 5 and 7 ind/0.2 m2) and three for H. mammata (6, 10 and 12 ind/0.5 m2). Growth (specific growth rate (SGR) and growth rate (GR)), weight change, coefficient of variation and productivity were estimated, additionally the feeding rate on H. mammata. Growth on H. arguinensis decreased as the stocking density increased, showing the higher growth at the stocking density of 1 ind/0.2 m2 (SGR: 1.24%/day ± 0.16 and GR: 0.94 g/day ± 0.05), with a 104.65% (±9.98) of weight change. The best H. mammata growth was registered at 10 ind/m2 (SGR: 0.56% ± 0.04%/day and GR: 0.48 ± 0.04 g/day) and 25.48% (±1.52) of weight change. H. mammata feeding rate decreased as stocking density increased. Crowding stress could be considered the main factor affecting the sea cucumbers growth and performance on our study. The optimal stocking density for H. arguinensis and H. mammata under tank‐based conditions were established as 1 ind/0.2 m2 and 5 ind/0.5 m2 respectively. The critic biomass for H. arguinensis (471.65 g/m2) and for H. mammata (988.11 g/m2) were reached during the experiment at the fourth and fifth weeks respectively. Abstract: In the present study, protective effects of dietary eucalyptol were investigated against copper toxicity in common carp (Cyprinus carpio). The fish were fed with diets supplemented with 0 (control), 0.5% and 1% eucalyptol for 14 days before exposure to 0.25 mg/L ambient copper for 7 days. Blood samples were taken from the fish before and 1, 3 and 7 days after copper challenge. Before copper challenge, eucalyptol significantly decrease serum aspartate aminotransferase (AST) activity, but increase blood WBC, RBC, hematocrit and haemoglobin, serum lysozyme, total immunoglobulin (Ig) compared with the control. Copper exposure led to stress, tissue damage and immunosuppression. After the copper challenge, significant elevation in serum cortisol, glucose, alanine aminotransferase (ALT) and AST, but decrease in blood RBC and WBC, and serum lysozyme, complement (ACH50) and total immunoglobulin (Ig) was observed. Eucalyptol administration significantly mitigated the increase in serum cortisol, glucose, ALT and AST, and decrease in WBC, ACH50, lysozyme and total Ig. Dietary 1% eucalyptol had significantly higher protective effects than 0.5% in the case of serum AST, lysozyme, total Ig and ACH50. In conclusion, eucalyptol has beneficial health effects on common carp and enables to mitigate copper‐induced stress, tissue damage, and immunosuppression. Glucocorticoids are critical regulators of the cellular processes that allow animals to cope with stressors. In teleosts, cortisol is the primary circulating glucocorticoid and this hormone mediates a suite of physiological responses, most importantly energy substrate mobilization that is essential for acute stress adaptation. Cortisol signaling has been extensively studied and the majority of work has been on the activation of the glucocorticoid receptor (GR), a ligand-bound transcription factor, and the associated downstream transcriptional and protein responses. Despite the role of this hormone in acute stress adaptation, very few studies have examined the rapid effects of this hormone on cellular function. The nongenomic corticosteroid effects, which are rapid (seconds to minutes) and independent of transcription and translation, involve changes to second-messenger pathways and effector proteins, but the primary receptors involved in this pathway activation remain elusive. In teleosts, a few studies suggested the possibility that GR located on the membrane may be initiating a rapid response based on the abrogation of this effect with RU486, a GR antagonist. However, studies have also proposed other signaling mechanisms, including a putative novel membrane receptor and changes to membrane biophysical properties as initiators of rapid signaling in response to cortisol stimulation. Emerging evidence suggests that cortisol activates multiple signaling pathways in cells to bring about rapid effects, but the underlying physiological implications on acute stress adaptation are far from clear. The corticotropin-releasing factor (CRF) system is expressed in the earliest stages of zebrafish development, long before its canonical function in the endocrine stress response is realized, and yet its function during embryogenesis is unknown. We tested the hypothesis that CRF protects embryos from stress-induced apoptosis. Here we confirm that a 1 h heat shock applied at either 6 h post-fertilization (hpf) or 30 hpf elicits an increase in caspase-3 activity, a key effector of apoptosis. Temporal changes in the expression of crf and its binding protein ( crf-bp ) during recovery from heat shock indicate that the CRF system is responsive to stressors experienced as early as gastrulation. Next, we heat shocked embryos that were microinjected with crf mRNA, and showed that caspase-3 induction is significantly reduced in embryos that overexpress CRF relative to control embryos. In addition, incubating embryos in the presence of the CRF receptor type 1 (CRF-R1) antagonist, antalarmin, during recovery from heat shock significantly increased caspase-3 activity, suggesting that CRF regulates caspase-3 via a CRF-R1-dependent pathway. Finally, we show that most heat shock-induced mortality occurred during the first hour of recovery, long before a significant increase in caspase-3 activity was detected. Indeed, the delayed caspase-3 induction coincided with a mortality plateau, and neither CRF overexpression nor antalarmin treatment altered heat shock induced mortality, supporting previous in vitro evidence that CRF-mediated cytoprotection occurs through the slow and tightly controlled apoptotic pathway. This study provides novel in vivo evidence that CRF regulates stress-induced apoptosis in a vertebrate model species, and demonstrates for the first time a function for the CRF system in early development that precedes its role in the endocrine stress response. Abstract: Freshwater organisms are increasingly exposed to combinations of stressors. However, because it is time‐consuming and costly, research on the interaction of stressors, such as compound toxicity and global warming on vertebrates, is scarce. Studies on multigenerational effects of these combined stressors are almost nonexistent. In the present study, we tested the combined effects of 4 °C warming and cadmium (Cd) exposure on life‐history traits, biomarkers, bioaccumulation, and multigenerational tolerance in the turquoise killifish, Nothobranchius furzeri. The extremely short life cycle of this vertebrate model allows for assessment of sublethal and multigenerational effects within 4 mo. The applied Cd concentrations had only limited effects on the measured endpoints, which suggests that N. furzeri is more resistant to Cd than fathead minnow and rainbow trout. In contrast, the temperature increase of 4 °C was stressful: it delayed female maturation and lowered adult mass and fecundity. Finally, indications of synergistic effects were found on peak fecundity and embryonic survival. The present study aimed to compare effects of increasing chronic stress load on the stress response of European seabass (Dicentrarchus labrax) and gilthead seabream (Sparus aurata) to identify neuroendocrine functions that regulate this response. Fish were left undisturbed (controls) or exposed to three levels of chronic stress for 3 weeks and then subjected to an acute stress test (ACT). Chronic stress impeded growth and decreased feed consumption in seabass, not in seabream. In seabass basal cortisol levels are high and increase with stress load; the response to a subsequent ACT decreases with increasing (earlier) load. Basal cortisol levels in seabream increase with the stress load, whereas the ACT induced a similar response in all groups. In seabass and seabream plasma a-MSH levels and brain stem serotonergic activity and turnover were similar and not affected by chronic stress. Species-specific molecular neuro-regional differences were seen. In-situ hybridization analysis of the early immediate gene cfos in the preoptic area showed ACT-activation in seabream; in seabass the expression level was not affected by ACT and seems constitutively high. In seabream, expression levels of telencephalic crf, crfbp, gr1, and mr were downregulated; the seabass hypothalamic preoptic area showed increased expression of crf and gr1, and decreased expression of mr, and this increased the gr1/mr ratio considerably. We substantiate species-specific physiological differences to stress coping between seabream and seabass at an endocrine and neuroendocrine molecular level. Seabass appear less resilient to stress, which we conclude from high basal activities of stress-related parameters and poor, or absent, responses to ACT. Stress-related physiological changes and post-release survival of elephant fish ( Callorhinchus milii ) in response to fishing capture and handling were assessed by experimental simulation using several different fishing gears for various periods under controlled laboratory conditions and by repeated blood sampling during a subsequent 72-h period to monitor the animals’ condition. Capture and handling caused physiological changes in elephant fish indicated by elevated plasma lactate, potassium and osmolality and immediately depressed plasma glucose. Maximum values of plasma potassium and osmolality occurred mainly immediately after capture (0 h), and maximum levels of plasma lactate at 0 and 3 h. Elevated plasma lactate concentrations returned to baseline values after 12 h of capture. Minimum levels of plasma glucose were recorded 3 h after capture. Hypoglycaemia followed by hyperglycaemia after stress was recorded. Handling alone significantly increased plasma lactate levels. Gillnet capture caused the greatest physiological stress, followed by longline, and then angling. Immediate mortality was 0% for all capture treatments, but delayed mortality varied widely among the capture treatments. Post-release survival rates were 70% (i.e. 30% delayed mortality) for longline and gillnet depending on fishing duration and 100% (i.e. 0% delayed mortality) for hook and line (angling) provided the animals were returned to the water immediately following capture. Influence of temperature, salinity, and dissolved oxygen on the stress response of bull (Carcharhinus leucas) and bonnethead (Sphyrna tiburo) sharks after capture and handling. Capture and handling stress can induce acidosis in sharks. This response, endured during commercial bycatch and in catch-and-release recreational fisheries, could be exacerbated in certain environmental conditions. To assess environmental influence on stress response, changes in acid-base, blood gas, and metabolite analytes (pH, pCO2, and lactate) measured with the i-STAT portable clinical analyzer were evaluated immediately after capture and removal from gillnets among wild bull (Carcharhinus leucas) and bonnethead (Sphyrna tiburo) sharks caught in waters of differing temperature (T), salinity (Sal), and dissolved oxygen (DO). Time from capture to blood collection (C-BD) was also recorded. Effects of T, Sal, DO, and C-BD on acid-base physiology were evaluated by modeling their ability to predict pH, pCO2, and lactate concentrations using ordinal logistic regression (OLR). The OLR models suggest that C. leucas sharks experienced a mixed metabolic and respiratory acidosis in warmer waters and at the low end of their salinity tolerance, and that S. tiburo sharks experienced a metabolic acidosis in warmer waters with a potential for respiratory acidosis at the high end of their salinity tolerance. In S. tiburo, capture and handling time exacerbated acidosis. Based on these findings, it is recommended that commercial and catch-and-release fisheries conduct operations cautiously during times of the year when water temperatures are high and salinities are at either extreme, by decreasing soak times, using the strongest proper tackle gear to reduce fight times, and releasing sharks as soon as possible after capture and detection. Abstract: Aquaculture and fisheries products are expected to play a leading part in addressing the food necessities of the increasing human population. However, in intensive aquaculture, fish are exposed to stress conditions that deteriorate their immune systems, leading to increased disease vulnerability. These disease outbreaks have resulted in production losses and represent one of the key constraints in fish farming. Accordingly, aquaculture disease management efforts have concentrated on the establishment of environmental friendly and lasting methods. Recently, several studies have revealed that various bacteria (especially probiotics)‐derived cellular components might be used as promising immunostimulants in fish aquaculture. Such components curtail the risk concomitant with the use of antibiotics, chemotherapeutics, vaccines and probiotics. Among the various health advantages ascribed to bacterial cellular components, the modulation of specific and nonspecific immune responses, cytokine gene expression and anti‐pathogenic activities are most anticipated benefits. Their potential as vaccine adjuvants for fish culture has also been demonstrated. However, as an emerging practice, limited knowledge is available regarding the detailed interactions with fish immune systems. Therefore, the underlying mechanisms involved in fish immune cell activation need to be explored further. In this context, the present article aspires to review the available studies on the use of bacterial cellular components in fish culture and their biological effects on fish such as growth promotion, immunostimulation and anti‐pathogenic activities as well as to evaluate the factors that control the optimal induction of immune responses in fish. The stress hormone cortisol, together with antioxidants, melatonin (Mel) and its biologically active metabolites, 5-methoxykynuramines, including AFMK, set up a local stress response system in mammalian skin. Our in vitro study of the European flounder ( Platichthys flesus ) was designed to examine whether Mel and AFMK would respond to cortisol while a glucocorticoid is added to the incubation medium. The concentrations of cortisol in the incubation medium mimic plasma cortisol levels seen in fish exposed to different types of stresses such as handling, confinement, high density, food-deprivation or air-exposure. We measured Mel and AFMK in skin explants and culture media using high-performance liquid chromatography (HPLC) with fluorescence detection. We also analysed melanosome response (dispersion/aggregation) in the explants subjected to the different treatments. Cortisol stimulated the release of Mel and AFMK from skin explants in a dose-dependent manner. Melanosome dispersion and a darkening of the skin explants were observed after incubation with cortisol. This study is the first to demonstrate the interrelationship between cortisol and Mel/AFMK in fish skin. Our data strongly suggest that the cutaneous stress response system (CSRS) is present in fish. The question remains whether Mel, AFMK or cortisol are synthetized locally in fish skin and/or transported by the bloodstream. This study investigated the anesthetic effect of the essential oils (EOs) from the peel of Citrus x aurantium (EOCA) and Citrus x latifolia (EOCL) on silver catfish Rhamdia quelen. Fish were exposed to different concentrations of EOCA and EOCL to determine time of anesthesia induction and recovery. Induction of anesthesia was observed in all fish exposed to 400, 600 or 800 μL L−1 EOCA and 300, 400 or 500 μL L−1 EOCL. Another group of fish were exposed for 8 h to 50, 100, or 200 μL L−1 of either EOs. Overall, fish exposed to ethanol and both EOs presented higher ventilatory frequencies (VF) than the control group throughout the 8 h of exposure. Net ion (Na+, K+ and Cl-) effluxes and ammonia excretion were significantly lower in fish exposed to 50, 100 or 200 μL L−1 of either EOs compared to control fish. Mortality was 37% in fish exposed to 200 μL L−1 of either EOs after 8 h. These findings suggest that EOCA and EOCL are useful anesthetics and sedatives for Rhamdia quelen, but their usefulness as alternatives to reduce stress in fish transportation at the lower concentrations tested (50-100 μL L−1) deserves further study. Increases in carbon dioxide (CO 2 ) and other greenhouse gases emissions are changing ocean temperature and carbonate chemistry (warming and acidification, respectively). Moreover, the simultaneous occurrence of highly toxic and persistent contaminants, such as methylmercury, will play a key role in further shaping the ecophysiology of marine organisms. Despite recent studies reporting mostly additive interactions between contaminant and climate change effects, the consequences of multi-stressor exposure are still largely unknown. Here we disentangled how Argyrosomus regius physiology will be affected by future stressors, by analysing organ-dependent mercury (Hg) accumulation (gills, liver and muscle) within isolated/combined warming (ΔT = 4 °C) and acidification (ΔpCO 2 = 1100 μatm) scenarios, as well as direct deleterious effects and phenotypic stress response over multi-stressor contexts. After 30 days of exposure, although no mortalities were observed in any treatments, Hg concentration was enhanced under warming conditions, especially in the liver. On the other hand, elevated CO 2 decreased Hg accumulation and consistently elicited a dampening effect on warming and contamination-elicited oxidative stress (catalase, superoxide dismutase and glutathione- S -transferase activities) and heat shock responses. Thus, potentially unpinned on CO 2 -promoted protein removal and ionic equilibrium between hydrogen and reactive oxygen species, we found that co-occurring acidification decreased heavy metal accumulation and contributed to physiological homeostasis. Although this indicates that fish can be physiologically capable of withstanding future ocean conditions, additional experiments are needed to fully understand the biochemical repercussions of interactive stressors (additive, synergistic or antagonistic). The fish brain plays an important role in controlling growth, development, reproduction, and adaptation to environmental change. However, few studies stem from the perspective of whole transcriptome change in a fish brain and its response to long-term hypersaline stress. This study compares the differential transcriptomic responses of juvenile Nile tilapia (Oreochromis niloticus) maintained for 8 weeks in brackish water (16 practical salinity units, psu) and in freshwater. Fish brains from each treatment were collected for RNA-seq analysis to identify potential genes and pathways responding to hypersaline stress. A total of 27,089 genes were annotated, and 391 genes were expressed differently in the salinity treatment. Ten pathways containing 40 differentially expressed genes were identified in the tilapia brain. Antigen processing and presentation and phagosome were the two principally affected pathways in the immune system. Thirty-one of 40 genes were involved in various expressions associated with environmental information processing pathways such as neuroactive ligand-receptor interaction, cytokine-cytokine receptor interaction, the Jak-STAT signaling pathway, cell adhesion molecules (CAMs), and the PI3K-Akt signaling pathway, which are the upstream pathways for modulation of immunity and osmoregulation. The most-changed genes (>5-fold) were all down-regulated, including four growth hormone/prolactin gene families, i.e., prolactin precursor (-10.62), prolactin-1 (-11), somatotropin (-10.15), somatolactin-like (-6.18), and two other genes [thyrotropin subunit beta (-7.73) and gonadotropin subunit beta-2 (-5.06)] that stimulated prolactin release in tilapia. The downregulation pattern of these genes corroborates the decrease in tilapia immunity with increasing salinity and reveals an adaptive mechanism of tilapia to long-term hypersaline stress. Ovarian steroidogenesis, isoquinoline alkaloid biosynthesis, and phenylalanine metabolism are the three important pathways in the response of the fish to long-term hypersaline stress. This study has identified several pathways and relevant genes that are involved in salinity regulation in a euryhaline fish and provides insight into understanding regulatory mechanisms of fish to salinity change. This review analyzes what could be regarded as the “ clandestine organs ” of the endocrine system: the gut microbiome, the immune system, and the stress system. The immune system is very closely related to the endocrine system, with many intertwined processes and signals. Many researchers now consider the microbiome as an ‘organ’ that affects the organism at many different levels. While stress is certainly not an organ, it affects so many processes, including endocrine-related processes, that the stress response system deserved a special section in this review. Understanding the connections, effects, and feedback mechanisms between the different “ clandestine organs ” and the endocrine system will provide us with a better understanding of how an organism functions, as well as reinforce the idea that there are no independent organs or systems, but a complex, interacting network of molecules, cells, tissues, signaling pathways, and mechanisms that constitute an individual. Abstract: Physical stressors, such as man‐made obstacles, are considered one of the main causes that negatively affect freshwater fish. Even small weirs may impact fish populations, including potamodromous cyprinids, by partially or totally blocking upstream migratory movements. Some studies have addressed the effect of key hydraulic parameters on upstream movements past small weirs, but little is known on how these parameters interact to induce swimming or jumping behaviour in negotiating such obstacles. This study aims to evaluate the passage behaviour (swimming vs. jumping) of Iberian barbel (Luciobarbus bocagei), a potamodromous species, over an experimental small broad‐crested weir, considering the effect of different plunge pool depths (D), waterfall heights (H), and weir discharges (Q). Results revealed that passage behaviour was highly dependent on combinations of plunge pool depths and waterfall heights. Barbel navigated most configurations by swimming (overall outcomes: 81.5% swimming vs. 18.5% jumping), except the ones with the higher waterfall heights (25 cm) tested. Therefore, higher waterfalls proved to be preponderant in the switching of passage behaviour from swimming to jumping. Regarding the discharge over the 1‐m‐wide weir (overall outcomes: 85% swimming vs. 15% jumping), contrary to what was expected, there was no evidence that passage behaviour was discharge‐related, for the range of discharges tested (25–100 L/s). These results are useful to identify potential migration obstacles and should be taken into consideration in river restoration projects and when designing fishways for potamodromous cyprinids. Juvenile rockfish Sebastes schlegelii (mean length 10.8 ± 1.4 cm, and mean weight 31.7 ± 3.6 g) were exposed for 4 weeks to different levels of dietary chromium (Cr 6+ ) at 0, 120, and 240 mg/L and ascorbic acid (AsA) at 100, 200, and 400 mg/L. Growth performance of S. schlegelii was significantly decreased due to dietary Cr exposure, whereas lysozyme activity was notably increased. Exposure to dietary Cr resulted in substantial accumulation of Cr in the blood. Levels of two stress indicators, plasma cortisol and heat shock protein 70, of S. schlegelii were increased due to dietary Cr exposure. The results indicated that dietary Cr exposure affected growth performance, lysozyme activity, and stress responses of S. schlegelii , and high levels of AsA supplementation significantly attenuated dietary Cr-induced toxicity. Optimal feeding strategies improve fish growth and health but may be affected by ammonia stress in closed rearing systems such as tanks or ponds. This study aimed to evaluate the effects of feeding frequency and ammonia levels in rearing water on the enzymes and genes involved in oxidative stress of yellow catfish. Experiment (ammonia exposure) and control groups were randomly assigned to one of three feeding frequencies (1, 2 and 4 times daily) for 8 weeks. Weight gain increased as feeding frequency increased from 1 to 4 times daily, but feed conversion ratio values decreased. The highest survival in ammonia group was found when fish was fed 2 times daily. Glutathione peroxidase activity and total antioxidant capacity in liver and brain of fish exposed to ammonia increased as feeding frequency increased from 1 to 4 times daily. Liver malondialdehyde content in control group decreased as feeding frequency increased from 1 to 4 times daily. The lowest liver malondialdehyde content in ammonia group was observed when fish was fed 2 times daily. Liver mRNA expression of superoxide dismutase and catalase in control group increased as feeding frequency increased from 1 to 4 times daily, but the highest superoxide dismutase and catalase expression in ammonia group were observed when fish were fed 2 times daily. This study indicates that higher feeding frequency of yellow catfish exposed to ammonia could result in oxidative stress and poor survival. Winter can be a challenging period for fish in northern temperate rivers and streams, particularly in those that are channelized, structurally simple or regulated by, for instance, hydropower. In these systems, dynamic sub-surface ice formation commonly occurs and stable periods with ice cover may be short. Under these adverse conditions, access to shelters has been shown to be an important factor that influences overwinter survival, and exclusion from shelters by anchor ice may cause stress. Here, stress responses of juvenile brown trout under simulated winter conditions in an artificial stream were studied. Trout were subjected to three treatments in which the trout (1) were excluded from an instream wood shelter, simulating the effects of anchor ice, (2) had access to the shelter or (3) had surface ice cover in addition to the shelter. There was a positive correlation between ventilation frequency and plasma cortisol concentration. Trout without access to shelter had 30% higher ventilation frequency than trout with instream shelter and surface ice, but no differences in cortisol concentration or stress colour were found between the treatments. River regulation that reduces surface ice and increases anchor ice formation may lead to increased stress and consequently reduce overwinter survival rates. [ABSTRACT FROM AUTHOR]

Despite the threat of climate change, the physiological mechanisms responsible for reduced performance at high temperatures remain unclear for most species. Elevated but sublethal temperatures may act via endocrine and cellular stress responses to limit performance in important life-history traits such as growth. Here, brook trout (Salvelinus fontinalis) subjected to chronically elevated or daily oscillating temperatures were monitored for growth and physiological stress responses. Growth rate decreased at temperatures above 16°C and was negative at 24°C, with an estimated upper limit for positive growth of 23.4°C. Plasma cortisol increased with temperature and was 12- and 18-fold higher at 22 and 24°C, respectively, than at 16°C, whereas plasma glucose was unaffected by temperature. Abundance of heat shock protein 70 (HSP70) in the gill increased with temperature and was 11- and 56-fold higher at 22°C and 24°C, respectively, than at 16°C. Therewas no relationship between temperature and plasma Cl-, but there was a 53% and 80% decrease in gill Na+/K+-ATPase activity and abundance at 24°C in comparison with 16°C. Daily temperature oscillations of 4°C or 8°C (19-23°C or 17-25°C) were compared with 21°C controls. Growth rate decreased with temperature and was 43% and 35% lower by length and mass, respectively, in the 8°C daily oscillation treatment than in the controls. There was no effect of temperature oscillation on plasma cortisol or glucose levels. In contrast, gill HSP70 abundance increased with increasing daily oscillation and was 40- and 700-fold greater at 4°C and 8°C daily oscillation, respectively, than in the constant temperature controls. In individuals exposed to 17-25°C diel oscillations for 4 days and then allowed to recover at 21°C, gill HSP70 abundance was still elevated after 4 days recovery, but not after 10 days. Our results demonstrate that elevated temperatures induce cellular and endocrine stress responses and provide a possible mechanism by which growth is limited at elevated temperatures. Tropical organisms are predicted to be among the most impacted by increasing sea surface temperatures, particularly those from intertidal habitats. In this study, a complete thermal biology assessment was conducted for two widespread tropical Atlantic shallow reef fish: Abudefduf saxatilis (damselfish) and Scartella cristata (blenny), which make extensive use of tide pools. The main objectives were to measure the time-course changes during one month in i) thermal and oxidative stress biomarkers (in gills, muscle and skin), ii) upper thermal limits, acclimation capacity and thermal safety margins and iii) body size, condition and energy reserves (total protein and lipid contents), under two temperature treatments (control – mean summer temperature, and elevated temperature − + 3 °C, as projected by climate warming scenarios for the end of this century). Results from biomarker analyses suggest that under increased temperature, both species displayed a typical response of physiological stress characterized by the activation of molecular chaperones and antioxidant protection. Both species presented a significant acclimation potential in the long term, as shown by increased critical thermal maxima values at higher temperature. However, these species may already be at risk during summer heat waves, as thermal safety margins for both species were low. Additionally, despite acclimation, some energetic tradeoffs may exist, since specimens from both species showed smaller body sizes at higher temperature (even though maintaining body condition). Finally, temperature treatments had a significant influence not only in the total amount of energy reserves (lipid contents) but also in their rate of deposition or depletion (total proteins and lipid contents). This is the first multi-end-point holistic approach to assess the impact of warming in shallow tropical water fish and it highlights the high risk that intertidal organisms are facing in both present and future sea surface temperature conditions. Stress is an event that most animals experience and that, induces a number of responses involving all three regulatory systems, neural, endocrine and immune. Fish cultures are especially at risk to the adverse effects of stress. Blood chemistry and hematological measurements can provide valuable physiological indices that may offer critical feedback on different stressors. Blood samples were collected from the caudal vein of Carassius carassius after subjected to stressors and parameters such as plasma cortisol and glucose levels were estimated. Also, immunological response through neutrophil/lymphocyte ratio were evaluated. The responses of C. carassius to stress were characterized by rapid and transient significant increases in glucose, hemoglobin, hematocrit, as well as an equally dramatic but delayed increase in cortisol levels. High ratio of neutrophils to lymphocytes (N: L) in blood fish were found, which reliably is related with high glucocorticoid levels. Our results strongly indicate the close relationship between stress hormones and neutrophil/lymphocyte ratio, concluding that N: L ratio and its relation with glucocorticoid hormones can provide a reliable method to study responses of fish to stress. The present study was conducted to determine the effect of stress in liver of Carassius carassius, a freshwater fish that normally dwell in the bottom layer of the waters of lakes, rivers and reservoirs. It is a sedentary fish that naturally feeds on zoo benthos and plant components and thanks to its sensitivity to the changes in surrounding medium is an ideal animal for indication of the health of freshwater aquatic ecosystems. It is susceptible for the teleost fish liver to be disturbed by numerous, stressful factors that influence in their health. Liver pieces of 30 individuals collected from Seferani Lake, in Dumrea region (Elbasan, Albania), were excised and processed for standard histopathological analysis. The result revealed pathological changes in liver tissue including heterogeneity of tissue parenchyma, irregular hepatocyte cells and their nuclei. Massive vacuolization of liver cells and their nuclei, necrotic foci, karyolysis and karyopicnosis were also observed. Our findings imply that histopathological evaluation can be used effectively as biomarker of fish physiological stress response and health status. [ABSTRACT FROM AUTHOR]

Stressful housing conditions like social isolation have been shown to profoundly affect the physiology and health of various organisms which is rarely addressed in fish species. In the present study, we used a shoaling species, zebrafish, to investigate the stress reactivity of grouped and individually housed fish. We also hypothesized if isolation is a stressful condition may disrupt growth performance and innate immune response of individuals. To this end, fish were housed individually (social isolated treatment) or in groups of five fish (control treatment) for 60 days. Growth indices of fish were not affected by social isolation. Sixty-day social isolation did insignificant effect on baseline cortisol levels of specimens; however, individually housed zebrafish showed lower plasma cortisol to chasing stress than the control grouped fish. On the contrary, exposure to predator caused higher cortisol levels in social isolated fish. Serum lysozyme activity of isolated individuals was significantly lower than control fish, but activity of serum complement remained unchanged. Our results represent evidences that zebrafish experienced social isolation showed broad changes in physiological and immunological functions which may affect the quality of life. The ornamental fish trade is estimated to handle up to 1·5 billion fishes. Transportation and handling of fishes imposes a range of stressors that can result in mortality at rates of up to 73%. These rates vary hugely, however, and can be as low as 2%, because they are generally estimated rather than based on experimental work. Given the numbers of ornamental fishes traded, any of the estimated mortality rates potentially incur significant financial losses and serious welfare issues. Industry bodies, such as the Ornamental Aquatic Trade Association (OATA), have established standards and codes of best practice for handling fishes, but little scientific research has been conducted to understand the links between stress, health and welfare in ornamental species. In aquaculture, many of the same stressors occur as those in the ornamental trade, including poor water quality, handling, transportation, confinement, poor social and physical environment and disease and in this sector directed research and some resulting interventions have resulted in improved welfare standards. This review considers the concept of welfare in fishes and evaluates reported rates of mortality in the ornamental trade. It assesses how the stress response can be quantified and used as a welfare indicator in fishes. It then analyses whether lessons from aquaculture can be usefully applied to the ornamental fish industry to improve welfare. Finally, this analysis is used to suggest how future research might be directed to help improve welfare in the ornamental trade. [ABSTRACT FROM AUTHOR]

Toll-like Receptors (TLRs) are the first innate receptors in recognizing pathogen-associated molecular patterns. In fish, upregulation of toll-like receptors during infection has been largely demonstrated while the effects of abiotic stressors on their expression remain poorly investigated. In this study, striped catfish were submitted during 20 days to three salinity profiles (freshwater, low saline water, saline water), followed by injection of a bacterial strain of Edwardsiella ictaluri . The expression of TLRs 1, 3, 4, 5, 7, 9, 19, 21, and 22 was measured in kidney at different time points in non infected and infected striped catfish. Infection induced overexpression of TLRs 1, 3, 4, 5, 7, 21 and 22. With elevated salinity, the expression of all TLRs, except TLR5, was severely decreased, particularly after bacterial infection. The TLRs responsiveness of striped catfish facing bacterial disease and salinity stress and possible consequences on striped catfish immune response's efficiency are discussed. Fish welfare is an important issue for growth of the aquaculture industry. Stress responses represent animal's natural reactions to challenging conditions and could be used as a welfare indicator. Cortisol level is relevant to fish welfare condition, and is a readily measured component of the primary stress response system. Generally, cortisol is measured by blood sampling. However, fish blood cortisol level could be instantly influenced by handling-stress at sampling. Fecal corticoid metabolites (FCM) are a mixture of several different metabolites with a wide range of polarities. Thus, feces could be promising alternative less handling-sensitive and non-invasive biological matrices for cortisol evaluation in Atlantic salmon. In this study we developed non-invasive method for determination of fecal corticoid metabolites in farmed Atlantic salmon ( Salmo salar L.) using enzyme-linked immunosorbent assay (ELISA). It was demonstrated that salmon FCM extracted from salmon feces is insoluble in non-polar solvents like diethyl ether, but well soluble in polar solvents like methanol. The proper extraction ratio could be one ml 100% methanol for 100 μL of the liquid part of salmon feces or 100 mg of the solid part. The FCM directly detected in unextracted liquid part of feces correlated well with the FCM extracted from both liquid and solid part of the corresponding samples, without significant difference. Thus, it is feasible to measure FCM directly in the liquid part of salmon feces without any extraction procedure. Then, we applied this assay for FCM analysis in the group of salmon that experienced salmon pancreas disease (PD) and amoebic gill disease (AGD). We demonstrated 1) both plasma cortisol and FCM increased significantly during the outbreak of inflammatory disease (P < 0.01). Plasma cortisol level was elevated from 28 ± 40 ng/ml to 164.4 ± 62.5 ng/ml, FCM from 14.4 ± 13.2 ng/ml to 170.7 ± 89.7 ng/ml 2) Growth and starvation has no significant impact on either cortisol or FCM level. 3) FCM correlated well with plasma cortisol level (P < 0.01). Furthermore, there seems more individual variation in plasma cortisol levels than in FCM levels. These results suggest FCM could be directly analyzed in liquid part of salmon feces without extraction. This directly detected FCM level could represent the total fecal FCM level and plasma cortisol level. This simple and non-invasive method makes FCM a proper indicator for salmon welfare. The reformed European Common Fisheries Policy introduced a discard ban, with a possibility of exempting species where a high discard survival can be demonstrated. This necessitates a validation of the methods used for estimating the discard mortality of candidate species. In this study, we assess whether reflex impairment can predict short-term mortality in commercially trawled European plaice upon landing and after air exposure of up to 90min. Sub-lethal stress was assessed by a suite of physiological variables. Over a 10-day period, mortality was monitored for a total of 199 plaice following trawl and air exposure of varying duration, and for 50 control fish scored for reflex impairment on board the vessel. Mortality was only observed in fish exposed to air for >60 min, and averaged 11.1% (95% CI = 7.1-16.3%). Reflex impairment was found to be a significant (P < 0.001) predictor of mortality in a generalized linear model, excluding other initially included variables by using a stepwise method. Plasma cortisol, haematocrit, and plasma osmolality all indicated a profound and increasing level of stress with air exposure, accompanied by a near depletion of muscle phosphocreatine and nucleotides. Fishing site had an unexpected, but significant (p < 0.05) effect on stress levels, which was also reflected in reflex impairment and mortality. The European Commission landing obligation, including species-specific "high survival" exemptions, has established a need for accurate dis- card survival estimates. This study presents the first discard survival estimates on-board Dutch commercial pulse trawlers. During seven, six, and one fishing trip(s), respectively, undersized plaice (Pleuronectes platessa), sole (Solea solea), and dab (Limanda limanda) were collected, assessed for vitality and subsequently monitored up to 21 days. Uncorrected for any potential impacts from predation, tagging, research-related handling, or holding conditions overall survival for plaice (n = 349), sole (n = 226), and dab (n = 187) was assessed as 15% [95% CI: 11-19%], 29% [95% CI: 24-35%], and 16% [95% CI: 10-26%] respectively. Survival was mainly effected by water temperature and factors linked to the fishing vessel. Fish length was not found to affect survival. Catch processing time and haul duration affected plaice survival but not sole. Vitality index, which averages reflex impairment and external damage scores, correlated with survival and may be developed as a proxy for discard survival. Compared to tickler-chain beam trawlers, pulse trawlers showed relatively higher discard survival under fishing conditions pertinent to these studies. [ABSTRACT FROM AUTHOR]

The aim of the present study was to assess the effects of the short-term live storage of Atlantic cod and haddock on residual blood in fillets. The fillet colour characteristics of fish sampled after 0, 1.5, 3 and 6 h of live storage were compared with fish subjected to current commercial processing procedures. Fish behaviour during live storage was also assessed, along with handling stress, by measuring the blood constituents (cortisol, glucose, lactate, pH and haematocrit), the initial white muscle pH, muscle twitches and length of time to the onset of rigor. The fillet colour in the CIE L*a*b* colour space was determined on fresh fillets (on-board) and the presence of discolouration was quantified by using a modified version of the Fillet Quality Index method. Fish behaviour analysis performed during live storage showed some signs of stress and that the condition of fish caught at greater depths was inferior to fish caught in shallower waters. The survival rate varied between the different trials (48.9–92.5%), and was likely impacted by the fishing depth. The blood chemistry data showed that the captured fish were somewhat stressed, but we were not able to clarify whether the fish were becoming gradually more stressed during the subsequent live storage, or whether the observed increase or lack of recovery were a result of a delayed response for the various stress parameters. The occurrence of blood spots and discolouration was low in fillets cut from both species of fish just after capture. Subsequent live storage did not change this scenario. Increases in carbon dioxide (CO2) and other greenhouse gases emissions are leading to changes in ocean temperature and carbonate chemistry, the so-called ocean warming and acidification phenomena, respectively. Methylmercury (MeHg) is the most abundant form of mercury (Hg), well-known for its toxic effects on biota and environmental persistency. Despite more than likely co-occurrence in future oceans, the interactive effects of these stressors are largely unknown. Here we assessed organ-dependent Hg accumulation (gills, liver and muscle) within a warming (▵ T = 4 °C) and acidification (▵ pCO2 = 1100 μatm) context, and the respective phenotypic responses of molecular chaperone and antioxidant enzymatic machineries, in a commercially important fish (the meagre Argyrosomus regius). After 30 days of exposure, although no mortalities were observed in any treatments, Hg concentration was significantly enhanced under warming conditions, significantly more so in the liver. On the other hand, increased CO2 decreased Hg accumulation and, despite negative effects prompted as a sole stressor, consistently elicited an antagonistic effect with temperature and contamination on oxidative stress (catalase, superoxide dismutase and glutathione-S-tranferase activities) and heat shock (Hsp70 levels) responses. We argue that the mechanistic interactions are grounded on simultaneous increase in excessive hydrogen (H+) and reactive oxygen species (e.g. O2-) free radicals, and subsequent chemical reaction equilibrium balancing. Additional multi-stressor experiments are needed to understand such biochemical mechanism and further disentangle interactive (additive, synergistic or antagonistic) stressor effects on fish ecophysiology in the oceans of tomorrow. The aim of the present study was to evaluate the effects of dietary vitamin E, nanoselenium (nanoSe), and their combination on growth and acute stress responses in rainbow trout previously subjected to chronic stress. The fish were divided into five groups according to stocking density and diet: Normal (20 kg m − 3 ; basal diet), Dense (80 kg m − 3 ; basal diet), Vit E (80 kg m − 3 ; 500 mg kg − 1 vitamin E-supplemented diet), NanoSe (80 kg m − 3 ; 1 mg kg − 1 nanoSe-supplemented diet), and Combination (80 kg m − 3 ; 500 mg kg − 1 vitamin E and 1 mg kg − 1 nanoSe-supplemented diet). The fish in each group were weighed after 60 days. The best growth under high density condition was observed in fish fed on vitamin E-supplemented diets (Vit E and Combination groups). Following high stocking rearing, fish were subjected to an acute confinement stress by decreasing water volume in the tanks (200 kg fish m − 3 ) for 45 min and sampled at 0, 2, 4, 8 and 24 h post-stress to evaluate the fish performance to additional stressors. The levels of serum cortisol, glucose, lactate, alanine aminotransferase, aspartate transaminase, and alkaline phosphatase were significantly higher in the Dense group at most of time points after initiation of acute stress. The results showed that rainbow trout can reach adaptation to chronic stress conditions, resulting in intensified stress responses to an acute stressor. The data also showed that vitamin E-supplemented diets can exert positive effects on the welfare of chronically stressed rainbow trout subjected to an additional acute stressor. The effects of rearing volume on on-growing European sea bass performance and stress parameters were investigated for the first time in a pilot aquaculture farm. Fish were held under the same initial stocking densities in triplicate net-pen cages of different sizes (1.4, 45 and 252 m3) for a period of 8 months. Results showed significant differences among the experimental groups in most parameters tested, with better performance in the two larger rearing volumes. In particular, growth rate showed a linear association with rearing volume, being 0.68 g day−1 for the large cage group, 0.56 g day−1 and 0.32 g day−1 for the medium and the small groups respectively. The feed conversion ratio and per cent survival (%) were also better in the large cage group. Fish reared in the small and medium-sized cages showed higher plasma cortisol concentrations than those reared in large cages, which showed low basal cortisol concentrations. Additionally, after an acute chasing stress challenge, fish in the large and small groups, but not the medium group, showed increased cortisol concentrations. Differences also occurred in the ratio of the expression of cortisol receptors, namely the mineralocorticoid ( mr) and glucocorticoid receptors ( gr). In specific, the ratio of mr to gr mRNA expression in the liver was higher in fish reared in the small cages. The health of freshwater biota is dependent on streamflow, yet identification of the flow regimes required to maintain ecological integrity remains challenging to states in the United States seeking to establish ecological flows. We tested the relationship between decreases in streamflow and Shannon-Weaver diversity index of fish species for four flow-based habitat guilds: riffle, riffle-run, pool-run, and pool in North Carolina. We found species that prefer shallow habitats, such as riffles and riffle-runs were the most sensitive to decreases in streamflow; whereas no significant relationships were found for pool or pool-run species. The sensitivity to decreases in streamflow was greatest during summer and fall, when streams are naturally lower. When all fish habitat guilds were included in the assessment of flow-biology relationships, there were no significant relationships to decreases in streamflow. As the sensitivity of fish to reductions in streamflow is not constant across habitat guilds, combining all fish species together for flow-biology analyses may greatly underestimate the response of fish species to decreases in flow and should be acknowledged when establishing ecological flows. This study aimed to evaluate different stunning methods [percussion (T1), immersion in ice/water slurry (T2), and gill cut (T3)] on quality and stress conditions of silver carp ( Hypophthalmichthys molitrix ) fillets stored at 4 °C in 72 h postmortem. Rigor index (RI%), behavioral analysis, levels of lactic acid and muscle glycogen were measured for stress level evaluation. Meanwhile, sensory assessment, texture properties, cooking loss, adenosine triphosphate (ATP) related compounds, adenosine monophosphate deaminase (ADA) activity, and acid phosphatase (ACP) activity were analyzed. The least stress condition, significantly ( P < 0.05) higher initial glycogen content was observed in T1. Ice/water stunning reduced the rate of ATP degradation, reflected in the lowest K value during 72 h. Aversive behaviors, significantly ( P < 0.05) higher cooking loss, hypoxanthine riboside (HxR) content, and lower sensory score were observed in T3. Eutrophication and climate change are increasing the incidence of severe hypoxia in fish nursery habitats, yet the programming effects of hypoxia on stress responsiveness in later life are poorly understood. In this study, to investigate whether early hypoxia alters the developmental trajectory of the stress response, zebrafish embryos were exposed to 4 h of anoxia at 36 h post-fertilization and reared to adults when the responses to secondary stressors were assessed. While embryonic anoxia did not affect basal cortisol levels or the cortisol response to hypoxia in later life, it had a marked effect on the responses to a social stressor. In dyadic social interactions, adults derived from embryonic anoxia initiated more chases, bit more often, entered fewer freezes and had lower cortisol levels. Adults derived from embryonic anoxia also performed more bites towards their mirror image, had lower gonadal aromatase gene expression and had higher testosterone levels. We conclude that acute embryonic anoxia has long-lasting consequences for the hormonal and behavioural responses to social interactions in zebrafish. Specifically, we demonstrate that acute embryonic anoxia favours the development of a dominant and aggressive phenotype, and that a disruption in sex steroid production may contribute to the programming effects of environmental hypoxia. As human populations continue to expand, increases in coastal development have led to the alteration of much of the world's mangrove habitat, creating problems for the multitude of species that inhabit these unique ecosystems. Habitat alteration often leads to changes in habitat complexity and predation risk, which may serve as additional stressors for those species that rely on mangroves for protection from predators. However, few studies have been conducted to date to assess the effects of these specific stressors on glucocorticoid (GC) stress hormone levels in wild fish populations. Using the checkered puffer as a model, our study sought to examine the effects of physical habitat complexity and predator environment on baseline and acute stress-induced GC levels. This was accomplished by examining changes in glucose and cortisol concentrations of fish placed in artificial environments for short periods (several hours) where substrate type and the presence of mangrove roots and predator cues were manipulated. Our results suggest that baseline and stress-induced GC levels are not significantly influenced by changes in physical habitat complexity or the predator environment using the experimental protocol that we applied. Although more research is required, the current study suggests that checkered puffers may be capable of withstanding changes in habitat complexity and increases in predation risk without experiencing adverse GC-mediated physiological effects, possibly as a result of the puffers' unique morphological and chemical defenses that help them to avoid predation in the wild.
