Tea tree is a fluorine (F)-enriched plant, leading to much concern about the safety of drinking tea from tea tree (Camellia sinensis (L.) Kuntze). Tea tree is a perennial leaf-harvested crop, and tea production in China is generally categorized as spring tea, summer tea and autumn tea in its annual growth rounds. However, the seasonally dynamic changes of F content and accumulation in the leaves and its drinking safety are poorly understood. In this study, 85 tea varieties cultivated under the same conditions were investigated to analyze the seasonal variation of F content and it’s relationships with F accumulation, aluminum (Al), calcium (Ca) and manganese (Mn) and hazard quotient (HQ) in young leaves (one bud and two leaves, YL) and mature leaves (canopy leaves, ML). The average F contents and accumulations were 350 mg kg−1 and 203 g ha−1 in YL, and they were 2451 mg kg−1 and 2578 g ha−1 in ML, respectively, with F mainly accumulated in ML. As the growing season progresses, the F content showed a gradual increase in YL, while a decrease in ML, inferring that F may be redistributed from mature leaves to young leaves. Additionally, the F content was quite different among tea varieties which are suitable for processing oolong tea, green tea, and black tea, with higher F accumulation in oolong tea varieties than in green and black tea varieties. Moreover, F content and accumulation could be obviously affected by the geographical origin of the tea tree varieties, with significantly higher F content in the varieties from F rich fluorite belts than other regions. Furthermore, F content and accumulation showed a significant positive correlation with the content of Al and Mn (p < 0.05). Based on a daily tea consumption of 8.7 g, the HQ was investigated to show that the proportion of tea leaves with HQ

Ice sheet marine margins via outlet glaciers are susceptible to climate change and are expected to respond through retreat, steepening, and acceleration, although with significant spatial heterogeneity. However, research on ice–ocean interactions has continued to rely on decentralized, manual mapping of features at the ice–ocean interface, impeding progress in understanding the response of glaciers and ice sheets to climate change. The proliferation of remote-sensing images lays the foundation for a better understanding of ice–ocean interactions and also necessitates the automation of terminus delineation. While deep learning (DL) techniques have already been applied to automate the terminus delineation, none involve sufficient quality control and automation to enable DL applications to “big data” problems in glaciology. Here, we build on established methods to create a fully automated pipeline for terminus delineation that makes several advances over prior studies. First, we leverage existing manually picked terminus traces (16 440) as training data to significantly improve the generalization of the DL algorithm. Second, we employ a rigorous automated screening module to enhance the data product quality. Third, we perform a thoroughly automated uncertainty quantification on the resulting data. Finally, we automate several steps in the pipeline allowing data to be regularly delivered to public databases with increased frequency. The automation level of our method ensures the sustainability of terminus data production. Altogether, these improvements produce the most complete and high-quality record of terminus data that exists for the Greenland Ice Sheet (GrIS). Our pipeline has successfully picked 278 239 termini for 295 glaciers in Greenland from Landsat 5, 7, 8 and Sentinel-1 and Sentinel-2 images, spanning the period from 1984 to 2021. The pipeline has been tested on glaciers in Greenland with an error of 79 m. The high sampling frequency and the controlled quality of our terminus data will enable better quantification of ice sheet change and model-based parameterizations of ice–ocean interactions.

Many attentions have been previously focused to identify the multiple biochemical components related to tea quality and health benefits, however, the natural variation of biochemical components present in tea germplasm has not been adequately evaluated. In this study, the main biochemical components, leaf morphological and yield characteristics were evaluated for four rounds of tea leaves in a panel of 87 elite tea cultivars suitable for black, green, or oolong tea. Significant variations were observed among the tea cultivars, as well as seasonal differences in the levels of the free amino acid (FAA), caffeine (CAF), tea polyphenols (TP), water extract (WE) and TP to FAA ratio (TP/FAA). Results showed that the average levels of FAA showed a seasonal change, with the highest level of 4.0% in the 1st spring tea in the cultivars suitable for green tea and the lowest of 3.2% in summer tea in the cultivars suitable for black tea. The average CAF content was highest 3.2% in the cultivars suitable for oolong tea in the 1st spring and the lowest 2.5% in the cultivars suitable for green tea in summer. Limited seasonal and varietal variations were noticed in the average levels of WE among the three categories of tea. In addition, significant natural variation of the morphological characteristics, bud length varying from 2.5 cm to 8.7 cm, bud density from 190.3 buds · m-2 to 1 730.3 buds · m-2, mature leaves biomass from 128.4 kg · hm-2 to 2 888.4 kg · hm-2, and yield component traits of 100 buds (one bud with two leaves) dry weight from 3.7 g to 37.7 g, tea yield/round from 444.6 kg · hm-2 to 905.3 kg · hm-2, were observed. The aim of our evaluation was not only to identify the advantages of seasonal and clonal variations but also to provide a new viewpoint for their further application. Representative accessions were selected from the germplasm to promote the establishment of an inherent biochemical constituent expressing the quality of black, green, and oolong tea. The findings might be utilized to establish early selection criteria to enhance the tea breeding and production program.

The in vitro study demonstrates wirelessly controlled modulation of neural activity using magnetoelectric nanoparticles (MENPs), synchronized to magnetic field application with a sub-25-msec temporal response. Herein, MENPs are sub-30-nm CoFe2O4@BaTiO3 core-shell nanostructures. MENPs were added to E18 rat hippocampal cell cultures (0.5 μg of MENPs per 100,000 neurons) tagged with fluorescent Ca2+ sensitive indicator cal520. MENPs were shown to wirelessly induce calcium transients which were synchronized with application of 1200-Oe bipolar 25-msec magnetic pulses at a rate of 20 pulses/sec. The observed calcium transients were similar, in shape and magnitude, to those generated through the control electric field stimulation with a 50-μA current, and they were inhibited by the sodium channel blocker tetrodotoxin. The observed MENP-based magnetic excitation of neural activity is in agreement with the non-linear M − H hysteresis loop of the MENPs, wherein the MENPs’ coercivity value sets the threshold for the externally applied magnetic field.

Marine-terminating outlet glacier terminus traces, mapped from satellite and aerial imagery, have been used extensively in understanding how outlet glaciers adjust to climate change variability over a range of timescales. Numerous studies have digitized termini manually, but this process is labor intensive, and no consistent approach exists. A lack of coordination leads to duplication of efforts, particularly for Greenland, which is a major scientific research focus. At the same time, machine learning techniques are rapidly making progress in their ability to automate accurate extraction of glacier termini, with promising developments across a number of optical and synthetic aperture radar (SAR) satellite sensors. These techniques rely on high-quality, manually digitized terminus traces to be used as training data for robust automatic traces. Here we present a database of manually digitized terminus traces for machine learning and scientific applications. These data have been collected, cleaned, assigned with appropriate metadata including image scenes, and compiled so they can be easily accessed by scientists. The TermPicks data set includes 39 060 individual terminus traces for 278 glaciers with a mean of 136 ± 190 and median of 93 of traces per glacier. Across all glaciers, 32 567 dates have been digitized, of which 4467 have traces from more than one author, and there is a duplication rate of 17 %. We find a median error of ∼ 100 m among manually traced termini. Most traces are obtained after 1999, when Landsat 7 was launched. We also provide an overview of an updated version of the Google Earth Engine Digitization Tool (GEEDiT), which has been developed specifically for future manual picking of the Greenland Ice Sheet.

Chordoma is a rare malignant bone tumor that mainly occurs in the sacrum and the clivus/skull base. Surgical resection is the treatment of choice for chordoma, but the local recurrence rate is high with unsatisfactory prognosis. Compared with other common tumors, there is not much research and individualized treatment for chordoma, partly due to the rarity of the disease and the lack of appropriate disease models, which delay the discovery of therapeutic strategies. Recent advances in modern techniques have enabled gaining a better understanding of a number of rare diseases, including chordoma. Since the beginning of the 21st century, various chordoma cell lines and animal models have been reported, which have partially revealed the intrinsic mechanisms of tumor initiation and progression with the use of next-generation sequencing (NGS) techniques. In this study, we performed a systematic overview of the chordoma models and related sequencing studies in a chronological manner, from the first patient-derived chordoma cell line (U-CH1) to diverse preclinical models such as the patient-derived organoid-based xenograft (PDX) and patient-derived organoid (PDO) models. The use of modern sequencing techniques has discovered mutations and expression signatures that are considered potential treatment targets, such as the expression of Brachyury and overactivated receptor tyrosine kinases (RTKs). Moreover, computational and bioinformatics techniques have made drug repositioning/repurposing and individualized high-throughput drug screening available. These advantages facilitate the research and development of comprehensive and personalized treatment strategies for indicated patients and will dramatically improve their prognoses in the near feature.

Abstract Human blastocysts are comprised of the first three cell lineages of the embryo: trophectoderm, epiblast and primitive endoderm, all of which are essential for early development and organ formation. However, due to ethical concerns and restricted access to human blastocysts, a comprehensive understanding of early human embryogenesis is still lacking. To bridge this knowledge gap, a reliable model system that recapitulates early stages of human embryogenesis is needed. Here we developed a three-dimensional (3D), two-step induction protocol for generating blastocyst-like structures (EPS-blastoids) from human extended pluripotent stem (EPS) cells. Morphological and single-cell transcriptomic analyses revealed that EPS-blastoids contain key cell lineages and are transcriptionally similar to human blastocysts. Furthermore, EPS-blastoids are similar with human embryos that were cultured for 8 or 10 days in vitro, in terms of embryonic structures, cell lineages and transcriptomic profiles. In conclusion, we developed a scalable system to mimic human blastocyst development, which can potentially facilitate the study of early implantation failure that induced by developmental defects at early stage.

Chinese kale (Brassica oleracea var. alboglabra Bailey, CC) is a succulent stem vegetable in the Brassica family. Its allotetraploid (AACC) vegetable germplasm, which was synthesized via distant hybridization with the colloquially named ‘yellow turnip’ (B. rapa L. ssp. rapifera Matzg., AA), has a swelling stem similar to CC. To address the molecular mechanism of stem development for CC and AACC, RNA sequencing (RNA-seq) was used to investigate transcriptional regulation of their stem development at three key stages including 28 days, 42 days and the bolting stage (BS) after sowing. As a result, 32,642, 32,665, 33,816, 32,147, 32,293 and 32,275 genes were identified in six corresponding cDNA libraries. Among them, 25,459 genes were co-expressed, while 7,183, 7,206, 8,357, 6,688, 6,834 and 6,814 genes were specifically expressed. Additionally, a total of 29,222 differentially expressed genes (DEGs) were found for functional enrichment as well as many genes involved in plant hormones including gibberellin (GA), abscisic acid (ABA), cytokinin (CTK) and auxin (AUX). Based on gene expression consistency between CC and AACC, the gene families including DELLA, GID, PYR/PYL, PP2C, A-ARR and AUX/IAA might be related to stem development. Among these, eight genes including Bo00834s040, Bo5g093140, Bo6g086770, Bo9g070200, Bo7g116570, Bo3g054410, Bo7g093470 and Bo5g136600 may play important roles in stem development based on their remarkable expression levels as confirmed by qRT-PCR. These findings provide a new theoretical basis for understanding the molecular mechanism of stem development in Brassica vegetable stem breeding.

For tea plants, nitrogen (N) is a foundational element and large quantities of N are required during periods of roundly vigorous growth. However, the fluctuation of N in the tea garden could not always meet the dynamic demand of the tea plants. Autophagy, an intracellular degradation process for materials recycling in eukaryotes, plays an important role in nutrient remobilization upon stressful conditions and leaf senescence. Studies have proven that numerous autophagy-related genes (ATGs) are involved in N utilization efficiency in Arabidopsis thaliana and other species. Here, we identified an ATG gene, CsATG101, and characterized the potential functions in response to N in A. thaliana. The expression patterns of CsATG101 in four categories of aging gradient leaves among 24 tea cultivars indicated that autophagy mainly occurred in mature leaves at a relatively high level. Further, the in planta heterologous expression of CsATG101 in A. thaliana was employed to investigate the response of CsATG101 to low N stress. The results illustrated a delayed transition from vegetative to reproductive growth under normal N conditions, while premature senescence under N deficient conditions in transgenic plants vs. the wild type. The expression profiles of 12 AtATGs confirmed the autophagy process, especially in mature leaves of transgenic plants. Also, the relatively high expression levels for AtAAP1, AtLHT1, AtGLN1;1, and AtNIA1 in mature leaves illustrated that the mature leaves act as the source leaves in transgenic plants. Altogether, the findings demonstrated that CsATG101 is a candidate gene for improving annual fresh tea leaves yield under both deficient and sufficient N conditions via the autophagy process.

Over the past decades, paleoenvironmental studies in the Indian summer monsoon region have mainly focused on precipitation change, with few published terrestrial temperature records from the region. We analysed the distribution of isoprenoid glycerol dialkyl glycerol tetraethers (isoGDGTs) in the sediments of Lake Chenghai in southwest China across the Pleistocene–Holocene transition, to extract both regional hydrological and temperature signals for this important transition period. The lake level was reconstructed from the relative abundance of crenarchaeol in isoGDGTs (%cren) and the crenarchaeol′/crenarchaeol ratio. The %cren-inferred lake level identified a single lowstand (15.4–14.4 ka cal BP), while the crenarchaeol′/crenarchaeol ratio suggests a relatively lower lake level between 15.4–14.4 and 12.5–11.7 ka cal BP, corresponding to periods of weakened ISM during the Heinrich 1 and Younger Dryas cold event. A filtered TetraEther indeX consisting of 86 carbon atoms (TEX86 index) revealed that lake surface temperature was similar to present-day values during the last deglacial period and suggests a substantial warming of ∼4 ∘C from the early Holocene to the mid-Holocene. Our paleotemperature record is generally consistent with other records in southwest China, suggesting that the distribution of isoGDGTs in Lake Chenghai sediments has potential for quantitative paleotemperature reconstruction.

Understanding a country's scientific situation is essential for any country. Common indicators for measuring a country's scientific capability are the number of publications and citations. In this study, we design a new method, the citation induced scientific capability identification (CISCI) method, to measure a country's scientific capability from the perspective of the scientific roles the country undertakes in scientific research. Then, we employ this method to analyze countries' scientific capability based on downloaded citation data in the field of economics and business. Finally, we check the applicability of our designed method and explore countries' scientific capability in the accounting and physics disciplines. The results show that the scientific capability of 158 countries has been changing for many years. Every country has different scientific capabilities in different scientific roles, and China's scientific capability has improved dramatically. The CISCI method is applicable for analyzing scientific capability in scientific research.

The prognosis of patients with gastric cancer (GC) is still unsatisfying. Numerous markers of gastric cancer stem cells (GCSCs) have been identified and were thought to be related to cancer aggressiveness. However, the roles of GCSC markers in GC patients’ prognosis and immune infiltration remain unknown. Expression of GCSC markers was analyzed using Oncomine and Gene Expression Profiling Interactive Analysis (GEPIA). Their associations with clinicopathological parameters were analyzed using UALCAN and LinkedOmics. Alternations and protein expression of GCSC markers were analyzed by cBioPortal and the Human Protein Atlas databases, respectively. The prognostic significance of GCSC markers was evaluated using Kaplan-Meier plotter. Correlations between the expression of GCSC markers and immune infiltration along with biomarkers of tumor-infiltrating immune cells (TIICs) were assessed combined Tumor Immune Estimation Resource and GEPIA. GeneMANIA was used to discover the interactive genes of GCSC markers, and enrichment analysis was performed using Database for Annotation, Visualization, and Integrated Discovery server. We identified six GCSC markers significantly up-expressed in GC, compared with normal stomach tissues. Among them, the overexpression of ICAM1, THY1, and CXCR4 significantly indicated adverse, while EPCAM indicated beneficial clinicopathological features of GC patients. The up-regulation of CXCR4 showed unfavorable prognostic significance, whereas EPCAM and TFRC showed the opposite. The six GCSC markers were all correlated with the infiltration and activation of distinct TIICs. Especially, ICAM1, THY1, and CXCR4 showed strongly positive correlations with tumor-associated macrophages. Besides, chemokine, Toll-like receptor, NF-kappa B, and HIF-1 signaling pathways might be involved in the regulation of GCSC markers on cancer development. This study proposed that GCSC markers might be promising targets of GC treatment to weaken cancer stem-like properties and strengthen anticancer immunity.

The calving fronts of many tidewater glaciers in Greenland have been undergoing strong seasonal and interannual fluctuations. Conventionally, calving front positions have been manually delineated from remote sensing images. But manual practices can be labor-intensive and time-consuming, particularly when processing a large number of images taken over decades and covering large areas with many glaciers, such as Greenland. Applying U-Net, a deep learning architecture, to multitemporal synthetic aperture radar images taken by the TerraSAR-X satellite, we here automatically delineate the calving front positions of Jakobshavn Isbræ from 2009 to 2015. Our results are consistent with the manually delineated products generated by the Greenland Ice Sheet Climate Change Initiative project. We show that the calving fronts of Jakobshavn's two main branches retreated at mean rates of -117±1 and -157±1 m yr−1, respectively, during the years 2009 to 2015. The interannual calving front variations can be roughly divided into three phases for both branches. The retreat rates of the two branches tripled and doubled, respectively, from phase 1 (April 2009–January 2011) to phase 2 (January 2011–January 2013) and then stabilized to nearly zero in phase 3 (January 2013–December 2015). We suggest that the retreat of the calving front into an overdeepened basin whose bed is retrograde may have accelerated the retreat after 2011, while the inland–uphill bed slope behind the bottom of the overdeepened basin has prevented the glacier from retreating further after 2012. Demonstrating through this successful case study on Jakobshavn Isbræ and due to the transferable nature of deep learning, our methodology can be applied to many other tidewater glaciers both in Greenland and elsewhere in the world, using multitemporal and multisensor remote sensing imagery.

Molecular distributions and stable carbon isotopic (δ13C values) compositions of dicarboxylic acids and related secondary organic aerosols (SOA) in PM2.5 aerosols collected on a day/night basis at the summit of Mt. Tai (1534 m a.s.l.) in the summer of 2016 were analyzed to investigate the sources and photochemical aging process of organic aerosols in the forested highland region of the North China Plain. The molecular distributions of dicarboxylic acids and related SOA are characterized by the dominance of oxalic acid (C2), followed by malonic (C3), succinic (C4) and azelaic (C9) acids. The concentration ratios of C2 ∕ C4, diacid-C ∕ OC and C2 ∕ total diacids are larger in the daytime than in the nighttime, suggesting that the daytime aerosols are more photochemically aged than those in the nighttime due to the higher temperature and stronger solar radiation. Both ratios of C2 ∕ C4 (R2 > 0.5) and C3 ∕ C4 (R2 > 0.5) correlated strongly with the ambient temperatures, indicating that SOA in the mountaintop atmosphere are mainly derived from the photochemical oxidation of local emissions rather than long-range transport. The mass ratios of azelaic acid to adipic acid (C9 ∕ C6), azelaic acid to phthalic aid (C9 ∕ Ph) and glyoxal to methylglyoxal (Gly ∕ mGly) and the strong linear correlations of major dicarboxylic acids and related SOA (i.e., C2, C3, C4, ωC2, Pyr, Gly and mGly) with biogenic precursors (SOA tracers derived from isoprene, α/β-pinene and β-caryophyllene) further suggest that aerosols in this region are mainly originated from biogenic sources (i.e., tree emissions).C2 concentrations correlated well with aerosol pH, indicating that particle acidity favors the organic acid formation. The stable carbon isotopic compositions (δ13C) of the dicarboxylic acids are higher in the daytime than in the nighttime, with the highest value (−16.5±1.9 ‰) found for C2 and the lowest value (−25.2±2.7 ‰) found for C9. An increase in δ13C values of C2 along with increases in C2 ∕ Gly and C2 ∕ mGly ratios was observed, largely due to the isotopic fractionation effect during the precursor oxidation process.

Type 2 diabetes (T2D) is a heterogeneous disorder characterized by insulin resistance and impaired insulin secretion. Here Axelssonet al. show that Sox5, which is reduced in diabetes, regulates a set of differentially expressed genes in T2D and its genetic and pharmacological induction improves insulin secretion by diabetic islets.

A chironomid-based calibration training set comprised of 100 lakes from south-western China was established. Multivariate ordination analyses were used to investigate the relationship between the distribution and abundance of chironomid species and 18 environmental variables from these lakes. Canonical correspondence analyses (CCAs) and partial CCAs showed that mean July temperature is one of the independent and significant variables explaining the second-largest amount of variance after potassium ions (K+) in 100 south-western Chinese lakes. Quantitative transfer functions were created using the chironomid assemblages for this calibration data set. The second component of the weighted-average partial least squares (WA-PLS) model produced a coefficient of determination (r2bootstrap) of 0.63, maximum bias (bootstrap) of 5.16 and root-mean-square error of prediction (RMSEP) of 2.31 °C. We applied the transfer functions to a 150-year chironomid record from Tiancai Lake (26°38′3.8 N, 99°43′ E; 3898 m a.s.l.), Yunnan, China, to obtain mean July temperature inferences. We validated these results by applying several reconstruction diagnostics and comparing them to a 50-year instrumental record from the nearest weather station (26°51′29.22′′ N, 100°14′2.34′′ E; 2390 m a.s.l.). The transfer function performs well in this comparison. We argue that this 100-lake large training set is suitable for reconstruction work despite the low explanatory power of mean July temperature because it contains a complete range of modern temperature and environmental data for the chironomid taxa observed and is therefore robust.

This study explored the key volatile compounds in both male and female rabbit meat. Simultaneous distillation extraction with dichloromethane was adopted to extract the volatile compounds in Hyla rabbit meat. A total of 35 volatile compounds were identified by gas chromatography–mass spectrometry and quantified with 2, 4, 6-thimethylpyridine as internal standard. Seventeen volatile aldehydes, 4 alcohols, 2 ketones, 2 acids, 1 heterocyclic compound, 2 alkanes and 7 esters were detected. Hexanal, heptanal, octanal, nonanal, (E, E)-2, 4-decadienal, 1-octen-3-ol and (Z)-2-decenal were the key odorant compounds, with high relative odour activity value. Furthermore, the concentration of volatile compounds in male rabbit meat was higher than that in female rabbit meat.

We present the results of pollen analyses from a 1105 cm long sediment core from Wuxu Lake in southwestern China, which depict the variations of the East Asian winter monsoon (EAWM) and the Indian summer monsoon (ISM) during the last 12.3 ka. During the period of 12.3 to 11.3 cal ka BP, the dominance of Betula forest and open alpine shrub and meadow around Wuxu Lake indicates a climate with relatively cold winters and dry summers, corresponding to the Younger Dryas event. Between 11.3 and 10.4 cal ka BP, further expansion of Betula forest and the retreat of alpine shrubs and meadows reflect a greater seasonality with cold winters and gradually increasing summer precipitation. From 10.4 to 4.9 cal ka BP, the dense forest understory, together with the gradual decrease in Betula forest and increase in Tsuga forest, suggest that the winters became warmer and summer precipitation was at a maximum, corresponding to the Holocene climatic optimum. Between 4.9 and 2.6 cal ka BP, Tsuga forest and alpine shrubs and meadows expanded significantly, reflecting relatively warm winters and decreased summer precipitation. Since 2.6 cal ka BP, reforestation around Wuxu Lake indicates a renewed humid period in the late Holocene; however, the vegetation in the catchment may also have been affected by grazing activity during this period. The results of our study are generally consistent with previous findings; however, the timing and duration of the Holocene climatic optimum from different records are inconsistent, reflecting real contrast in local rainfall response to the ISM. Overall, the EAWM is broadly in-phase with the ISM on the orbital timescale, and both monsoons exhibit a trend of decreasing strength from the early to late Holocene, reflecting the interplay of solar insolation receipt between the winter and summer seasons and El Niño–Southern Oscillation strength in the tropical Pacific.

School-based telebehavioral health service is increasingly important in addressing the health disparities youth in rural communities face. The study examines barriers and successes of implementation and sustainability of a rural school-based telebehavioral health program from the perspective of site coordinators or school champions informed by the Consolidated Framework for Implementation Research framework. Six coordinators participated in semi-structured interviews and completed a self-report questionnaire (organizational readiness for implementing change) to describe their experiences with telebehavioral health. The authors used thematic analysis to extract themes. The overarching themes included (1) benefits, challenges, and opportunities; (2) resources and support; and (3) work processes and challenges. Site coordinators endorsed their workplaces' commitment to telebehavioral health but shared uncertainty about the telebehavioral health program's sustainability. The emphasis coordinators place on relationships, communication, continuity, and advocacy for families and students underscores their critical role in providing high-quality telehealth services in schools and the broader community. Their perspectives are vital to ensuring these services' continued successful implementation and sustainability. Site coordinators' focus on the need for school-based champions and increased awareness of telehealth services in their communities highlights a need for outreach efforts to continue engaging relevant stakeholders.

This paper is focused on an assessment of the state of the art of operational modal analysis (OMA) methodologies in estimating modal parameters from output responses of nonlinear structures. By means of the Volterra series, the nonlinear structure excited by random excitation is modeled as best linear approximation plus a term representing nonlinear distortions. As the nonlinear distortions are of stochastic nature and thus indistinguishable from the measurement noise, a protocol based on the use of the random phase multisine is proposed to reveal the accuracy and robustness of the linear OMA technique in the presence of the system nonlinearity. Several frequency- and time-domain based OMA techniques are examined for the modal identification of simulated and real nonlinear mechanical systems. Theoretical analyses are also provided to understand how the system nonlinearity degrades the performance of the OMA algorithms.

Recently, two-photon fluorescent probes based on anthocyanidin molecules have attracted extensive attention due to their outstanding photophysical properties. However, there are only a few two-photon excited fluorescent probes that really meet the requirements of relatively long emission wavelengths (4600 nm), large two-photon absorption (TPA) cross-sections (300 GM), significant Stokes shift (480 nm), and high fluorescence intensity. Herein, the photophysical properties of a series of anthocyanidins with the same substituents but different fluorophore skeletons are investigated in detail. Compared with b-series molecules, a-series molecules with a six-membered ring in the backbone have a slightly higher reorganization energy. This results in more energy loss upon light excitation, enabling the reaction products to detect NTR through a larger Stokes shift. More importantly, there is very little decrease in fluorescence intensity as the Stokes shift increases. These features are extremely valuable for high-resolution NTR detection. In light of this, novel 2a-n (n = 1–5) compounds are designed, which are accomplished by inhibiting the twisted intramolecular charge transfer (TICT) effect through alkyl cyclization, azetidine ring and extending π conjugation. Among them, 2a-3 gains a long emission spectrum (λem = 691.4 nm), noticeable TPA cross-section (957 GM), and large Stokes shift (110 nm), indicating that it serves as a promising candidate for two-photon fluorescent dyes. Introduction: Fuqi Guben Gao (FQGBG) is a botanical drug formulation composed of FuZi (FZ; Aconitum carmichaelii Debeaux [Ranunculaceae; Aconiti radix cocta]), Wolfberry (Lycium barbarum L. [Solanaceae; Lycii fructus]), and Cinnamon (Neolitsea cassia (L.) Kosterm. [Lauraceae; Cinnamomi cortex]). It has been used to clinically treat nocturia caused by kidney-yang deficiency syndrome (KYDS) for over 30 years and warms kidney yang. However, the pharmacological mechanism and the safety of FQGBG in humans require further exploration and evaluation. Methods: We investigated the efficacy of FQGBG in reducing urination and improving immune organ damage in two kinds of KYDS model rats (hydrocortisone-induced model and natural aging model), and evaluated the safety of different oral FQGBG doses through pharmacokinetic (PK) parameters, metabonomics, and occurrence of adverse reactions in healthy Chinese participants in a randomized, double-blind, placebo-controlled, single ascending dose clinical trial. Forty-two participants were allocated to six cohorts with FQGBG doses of 12.5, 25, 50, 75, 100, and 125 g. The PKs of FQGBG in plasma were determined using a fully validated LC-MS/MS method. Results: FQGBG significantly and rapidly improved the symptoms of increased urination in both two KYDS model rats and significantly resisted the adrenal atrophy in hydrocortisone-induced KYDS model rats. No apparent increase in adverse events was observed with dose escalation. Major adverse drug reactions included toothache, thirst, heat sensation, gum pain, diarrhea, abdominal distension, T-wave changes, and elevated creatinine levels. The PK results showed a higher exposure level of benzoylhypaconine (BHA) than benzoylmesaconine (BMA) and a shorter half-life of BMA than BHA. Toxic diester alkaloids, aconitine, mesaconitine, and hypaconitine were below the lower quantitative limit. Drug-induced metabolite markers primarily included lysophosphatidylcholines, fatty acids, phenylalanine, and arginine metabolites; no safety-related metabolite changes were observed. Conclusion: Under the investigated dosing regimen, FQGBG was safe. The efficacy mechanism of FQGBG in treating nocturia caused by KYDS may be related to the improvement of the hypothalamus-pituitary-adrenal axis function and increased energy metabolism. Steamed ginseng water (SGW) is a by-product of the repeated thermal processing of red ginseng, which is characterized by a high bioactive content, better skin care activity, and a large output. However, its value has been ignored, resulting in environmental pollution and resource waste. In this study, UHPLC-Q-Exactive-MS/MS liquid chromatography–mass spectrometry and multivariate statistical analysis were conducted to characterize the compositional features of the repeated thermaltreated SGW. The antioxidant activity (DPPH, ABTS, FRAP, and OH) and chemical composition (total sugars, total saponins, and reducing and non-reducing sugars) were comprehensively evaluated based on the entropy weighting method. Four comparison groups (groups 1 and 3, groups 1 and 5, groups 1 and 7, and groups 1 and 9) were screened for 37 important common difference markers using OPLS-DA analysis. The entropy weight method was used to analyze the weights of the indicators; the seventh SGW sample was reported to have a significant weight. The results of this study suggest that heat treatment time and frequency can be an important indicator value for the quality control of SGW cycling operations, which have great potential in antioxidant products. This paper discusses associative memories based on time-varying delayed fractional-order neural networks (DFNNs) with a type of piecewise nonlinear activation function from the perspective of multiple O(t−α) stability. Some sufficient conditions are gained to assure the existence of 5n equilibria for n-neuron DFNNs with the proposed piecewise nonlinear activation functions. Additionally, the criteria ensure the existence of at least 3n equilibria that are locally multiple O(t−α) stable. Furthermore, we apply these results to a more generic situation, revealing that DFNNs can attain (2k + 1)n equilibria, and among them, (k + 1)n equilibria are locally O(t−α) stable. Here, the parameter k is highly dependent on the sinusoidal function frequency in the expanded activation functions. Such DFNNs are well-suited to synthesize high-capacity associative memories; the design process is given via singular value decomposition. Ultimately, four illustrative examples, including applying neurodynamic associative memory to the explaining-lesson skills assessment of normal students, are supplied to validate the efficacy of the results.
