On-site surveys of weed populations provide information on the relative occurrence and density of weeds that can be useful to growers in that region. Data generated by weed surveys can aid in the management of weed issues by monitoring the movement of problem weeds and forecasting areas susceptible to infestations. Currently, on-site surveys are often performed on a small scale, within single fields or counties. Questionnaire surveys are helpful for assessing relative abundance but do not always provide detailed information on weed distribution in time or space. A survey was conducted annually in Ohio from 2013 through 2017 in 49 counties with soybean [Glycine max (L.) Merr.] production to assess the late-season occurrence of horseweed [Conyza canadensis (L.) Cronquist]. The objectives of this research were to: (1) determine the frequency, level of infestation, and distribution of C. canadensis in soybean fields in the primary soybean-producing Ohio counties over 5 yr; and (2) identify significant spatial clusters or movement trends over time. Conyza canadensis was encountered in each county from 2013 through 2017. Spatial cores of interest, or counties identified as having significant levels of C. canadensis infestations or a lack thereof relative to surrounding counties, were identified in all years except 2017. The lowest frequency of C. canadensis encountered at all rating levels occurred in 2017, which coincided with second-highest frequency of infestations (highest density level) among years. There was no distinct distribution or pattern of C. canadensis movement within the state from year to year, but there was an increase in counties with infestations over time compared with the early years of the survey when many counties had few to no infestations. These results suggest that C. canadensis persists as a common and troublesome threat to Ohio soybean producers and that growers should continue making C. canadensis management a priority when developing weed control programs.

Epidermal necrolysis (EN, including Stevens-Johnson syndrome and toxic epidermal necrolysis (TEN)) is mostly induced by drugs. Prevalence of lupus erythematosus (LE) is higher in EN than in general population (1.2 to 2%).1,2 Furthermore, flares of LE may present as EN (TEN-like LE).3-5 We aimed to describe EN in patients with clinical or biological LE to better characterize the relation between both diseases.

International audience; Ambrosia trifida L. (giant ragweed, Asteraceae) is native to the North American continent and was introduced into Europe and Asia at the end of the 19th century. In its native range, this tall annual species is common in riparian and ruderal habitats and is also a major weed in annual cropping systems. For nearly a century, A. trifida has also been of great concern in the U.S. for its highly allergenic pollen, necessitating targeted control measures to reduce its impact on human populations. Based on the distribution of A. trifida in North America and in its introduced range, riparian systems in the rest of the world may be particularly at risk to invasion, with potential negative consequences for their biodiversity. Currently, A. trifida has invaded Asia more widely than Europe, likely due to the more favourable local conditions in Asia. Throughout its introduced range, A. trifida is host to a limited number of invertebrates and pathogens and only a few biological agents are available for its control. The main impacts of A. trifida at a global level are on crop yield and human health, resulting in significant socio-economic impacts. The success of A. trifida invasion in areas in which it has been introduced is still unclear, but climate change may increase climate suitability, increasing the potential for A. trifida to spread. While effective management in cultivated fields seems potentially possible, the development and control of A. trifida in natural riparian habitats is of great concern due to the difficulty of management in these areas.

Current recommendations for the control of glyphosate-resistant horseweed [Conyza canadensis (L.) Cronquist var. canadensis] in soybeans [Glycine max (L.) Merr.] consist of comprehensive herbicide programs, which often include herbicide applications outside the soybean growing season. Integration of cover crops with herbicides could potentially improve C. canadensis control and allow for a reduction in herbicide inputs. Two separate field studies were conducted from 2016 through 2018 with the objectives of: (1) determining the effect of planting date and seeding rate of a cereal rye (Secale cereale L.) cover crop on C. canadensis population density and control in the subsequent soybean crop; and (2) determining whether the cover crop could replace a fall herbicide treatment or allow for a reduction in the use of spring-applied residual herbicides. There was no effect of rye planting date, late September versus late October, on C. canadensis density in either study. In 2016 to 2017, C. canadensis density was greater in the absence of a rye cover crop in both studies, but otherwise not affected by seeding rates of 50 versus 100 kg ha−1. In the 2017 to 2018 season, the presence of rye resulted in an increased C. canadensis density in the spring residual herbicide study (Study I), and had no effect in the fall herbicide study (Study II). Conyza canadensis densities were lowest in the treatments where a comprehensive spring residual or fall herbicide treatment had been applied, averaged over rye planting date and seeding rate. Earlier-planted rye at a higher seeding rate produced the most biomass but did not result in lower C. canadensis densities. These results suggest that cereal rye planted at a density of 50 kg ha−1 as a cover crop before no-till soybeans may be sufficient to reduce glyphosate-resistant C. canadensis plant density, but cannot be relied upon to reduce the need for fall herbicide treatments and spring residual programs.

Introduced seed dispersers can shift seed predation dynamics in native species, impacting native plant establishment and spread. We studied the effect of the non-native earthworm, Lumbricus terrestris, on seed loss dynamics in the large-seeded native annual, Ambrosia trifida (giant ragweed), an expanding agricultural weed whose seeds are heavily predated by mice. Lumbricus terrestris may protect seeds against rodent predation by caching them in its burrows from which seedlings can emerge unharmed; however, little is known about the ability of earthworms to compete with rodents for seeds. We investigated this interaction and how environmental factors affected the competitive outcome. In a 2-year field study, we analysed relative rates of seed removal by earthworms and mice for seeds dispersed at various times in habitats that varied in vegetative cover. Species-specific responses to environmental conditions drove variation in the share of seeds taken by earthworms versus mice, with earthworms gaining relatively more seeds under warmer, wetter conditions and in low plant cover habitats, and mice obtaining more seeds under colder, drier conditions and in high cover habitats. Environmental factors also determined which competitor accessed seeds first, and this conferred a competitive advantage that was compounded over time. Earthworms cached some seeds under all experimental conditions, suggesting that L. terrestris can act mutualistically with giant ragweed in diverse environments. Synthesis and applications. Our results support the hypothesis that the non-native earthworm Lumbricus terrestris behaves as a seed dispersal mutualist for the native annual giant ragweed by caching its seeds in its burrows, thereby reducing their availability to rodent seed predators. The data also support the view that interactions among the environment and competing seed predators determine the fate of seed pools in response to species-specific environmental preferences. In crop fields where L. terrestris is abundant, additional efforts to prevent giant ragweed seed return may be warranted due to the likelihood that earthworms will increase giant ragweed soil seed pools. Lumbricus terrestris is expanding its range throughout temperate regions and may similarly affect secondary seed dispersal and seed predation dynamics of large-seeded species in other plant communities. Integrated pest management (IPM) for weeds is considered to be in the early stages of development, especially when compared with insect and disease management. The need to develop IPM principles for weed management will increase as weed population shifts, species adaptation, and environmental impacts of weed control methods threaten the sustainability of agricultural systems. In this paper, we propose a framework for discussion and development of the weed component of IPM. We describe five levels of weed IPM that encompass progressively larger spatial scales and longer time periods, with corresponding changes in goals and complexity of supporting technology and information. The first level consists of the use of a 240single weed management tool at the field level, such as the use of a herbicide as the sole means for weed control. Level II integration incorporates tactical use of multiple tools for weed management, including combinations of herbicides, crop rotation, and mechanical and biological controls, as well as other cultural practices designed to reduce the damage caused by weeds. Level III represents the threshold of much current weed science research, and involves the design of entire cropping systems at the farm and landscape level as a strategy to resist weed invasion, tolerate weed presence, and decrease weed population survival and persistence. Levels IV and V are more speculative because the ecological basis for them is in development. Level IV addresses the management of habitats and landscapes at the ecosystem level to address large scale problems related to weed spread, dispersal, and invasion. At Level V, national trade and environmental policies are considered with respect to their impact on weed populations at a global scale. Approaches that might be taken at the various levels to management of herbicide resistance are described. Research has tended to focus on the first two levels of integration; however, development of IPM principles for weeds will require that attention be given to all levels of integration so that agricultural systems can be designed to more effectively deter and withstand the inevitable presence of weeds. Spatial patterns of trait variation across a species' range have implications for population success and evolutionary change potential, particularly in range-expanding and weedy species that encounter distinct selective pressures at large and small spatial scales simultaneously. We investigated intraspecific trait variation in a common garden experiment with giant ragweed (Ambrosia trifida), a highly variable agricultural weed with an expanding geographic range and broad ecological amplitude. Our study included paired populations from agricultural and natural riparian habitats in each of seven regions ranging east to west from the core of the species' distribution in central Ohio to southeastern Minnesota, which is nearer the current invasion front. We observed trait variation across both large- and small-scale putative selective gradients. At large scales, giant ragweed populations from the westernmost locations were nearly four times more fecund and had a nearly 50% increase in reproductive allocation compared to populations from the core. The degree of surface texture on fruits also declined from east to west. Greater fecundity in the west represents a putative trade-off between fruit size and fruit number across the study region, although no such trade-off was found across individual plants. This pattern may effectively result in greater propagule pressure closer to the invasion front. At smaller spatial scales, plants from agricultural populations emerged later and were smaller than plants from riparian populations. However, because plants from agricultural populations allocated more biomass to reproduction, total fecundity did not differ across habitats. Our emergence data are consistent with previous observations showing delayed emergence in agricultural compared to natural populations; thus evolutionary change may be predictable as giant ragweed continues spreading into agricultural fields throughout North America. These shifts in life-history strategy apparently bear no fecundity cost, suggesting that giant ragweed's success can be attributed at least in part to its substantial adaptive potential.

Classic Kaposi's sarcoma (CKS) occurs predominantly among elderly men and is associated with Kaposi's sarcoma-associated herpesvirus (KSHV). In low-endemic countries, KSHV infects predominantly men having sex with men (MSM).To describe a cohort of classic Kaposi sarcoma in a low-endemic area for KSHV, to highlight the features of CKS in MSM and identify prognostic factors.Retrospective single-centre study of CKS cases. We compared MSM to heterosexual patients. Then, we divided the patients into two subgroups, those requiring a systemic treatment and the others, and we performed univariate and multivariate analyses to determine aggressiveness of CKS.Between 2006 and 2015, seventy-four patients were included. Mean age at diagnosis was 68.9 years; sex ratio (M/F) was 6.4, and 28% were MSM; MSM patients were younger (P = 0.02), less often originated from endemic areas (P0.0001). KS was less severe (P = 0.04), required more often a local treatment than a systemic one (P = 0.03). On multivariate analysis, CD4 T-cell count500/mmFirst CKS cohort in low-endemic zone. We describe a fifth subtype of KS: KS in MSM. The CD4 T-cell count was found to correlate with prognosis.

The objective in routine analyses is generally to determine a small number of analytes. With samples containing ~103 or more components there will be insufficient peak capacity to resolve analytes from nonanalytes. This issue was addressed herein through a new type of separation mechanism in which small groups of targeted analytes are bound with high affinity to a soluble analyte-sequestering transport phase (ASTP) composed of a ~25 nm Stokes radius hydrophilic polymer core (HPC). When introduced into a 30 nm pore diameter size-exclusion chromatography (SEC) column, ASTP/analyte complexes elute within minutes, together, unretained, and relatively pure in the first chromatographic peak. Nonanalytes, in contrast, enter pore matrices of the packing material, are retarded in elution velocity, and are eluted later, separated from analytes. Fabrication of ASTPs was achieved by covalently coupling an antibody or some other affinity selector to a high molecular weight HPC. Beyond sequestering analytes, the function of ASTPs is to act as a molecular weight shifting agent, conveying an effective molecular weight to analytes that is much larger than that of nonanalytes and causing them to elute in the SEC void volume. This mode of separation is referred to as mobile affinity sorbent chromatography (MASC). Subsequent to their purification, ASTP/analyte complexes were detected by fluorescence spectrometry.

Introduction Les maladies immunologiques (lupus, dermatomyosite) sont des facteurs de risque de necrolyse epidermique (NE) medicamenteuse. D’autre part, en poussee aigue, elles peuvent mimer une NE. Cependant, le cadre nosologique des NEs avec auto-immunite peut etre difficile a statuer. Materiel et methodes Cette etude retrospective multicentrique a inclus les adultes avec NE de 07/2014 a 03/2020, avec antecedent de connectivite et/ou auto-anticorps (Ac) decouverts au moment de la NE (AAN, anti-DNA, anti-ECT, dot-myosite), qu’il y ait ou non un medicament (mdt) suspect. Les histologies ont ete relues en aveugle. Resultats Dix-huit patients ont ete inclus (17 F, âge median 47 ans), dont 4 avec lupus connu, 2 avec syndrome de Sjogren primaire, 2 avec photosensibilite. Sept (39 %) ont ete vus au printemps/ete. Onze (61 %) avaient un mdt suspect (ALDEN entre 3, possible et 6, certain). Le decollement median etait de 28 % (2-60 %). Une atteinte cephalique nette etait notee dans 12 cas. L’atteinte muqueuse etait constante. Trois patients sont decedes, 5 ont eu des sequelles severes. Trois situations ont ete observees : – Dans 5 cas : lesions annulaires, necrotiques, franche predominance cephalique, rare atteinte oculaire, connectivite connue, pas de mdt suspect. Les Ac etaient des AAN (n = 3, titre median 1/640), anti-ADN (n = 2), anti-SSA (n = 5), anti-MDA5 (n = 1). Ils ont recu un traitement specifique de la connectivite en phase aigue de la NE ; – Dans 10 cas : NE sans particularite clinique, frequente atteinte oculaire, mdt suspect dans 8 cas (80 %, ALDEN 4 a 6). Les Ac etaient des AAN (n = 8 titre median 1/800), anti-ADN (n = 1), anti-SSA (n = 6), anti-RNP (n = 1), anti-TIFF1γ (n = 1). Aucun n’a recu de traitement specifique en phase aigue. A distance, les Ac ont persiste sans manifestation clinique ; – Pour 3 patientes : NE sans particularite, au cours d’une poussee de lupus, avec mdt suspect : cotrimoxazole (n = 2, ALDEN 6), AINS (ALDEN 3). La nosologie a ete difficile a trancher. Au plan histologique, il n’y avait pas de difference significative entre les groupes, meme si une tendance a une mucinose dermique et moins de lesions lichenoides pilaires a ete observee dans le groupe 1. Un patient (groupe 1) avait une bande lupique en IFD. Discussion La presence d’une auto-immunite au cours d’une NE fait discuter 3 situations : – une NE immunologique, « non toxique », sur connectivite connue ou non (« lupus-Lyell »), melant au decollement des lesions annulaires, necrotiques, peu d’atteinte oculaire, justifiant un traitement specifique de la connectivite ; – une NE medicamenteuse, ou les Ac semblent fortuits (precessifs ? epitope spreading ?), n’imposant pas de traitement specifique de connectivite ; – une situation difficile a trancher (« lupus-Lyell » ou NE medicamenteuse favorisee par le lupus) chez des patients severes. L’indication d’un traitement immunomodulateur d’une NE avec auto-immunite doit etre pesee selon le cadre nosologique.

Field studies were conducted in 2011 and 2012 to compare mulch treatments of shredded newspaper, a combination of shredded newspaper plus turfgrass clippings (NP + grass), hardwood bark chips, black polyethylene plastic, and bare soil on weeds, insects, soil moisture, and soil temperature in pumpkins. Newspaper mulch or black plastic reduced total weed biomass ≥90%, and woodchip or NP + grass mulch each reduced total weed biomass 78% compared with bare soil under high rainfall conditions in 2011. In 2012, under low rainfall, all mulches reduced weed biomass 97% or more compared with bare soil. In both years, all mulches resulted in higher squash bug infestations than bare soil. The woodchip, newspaper, and NP + grass mulches retained higher soil moistures than bare soil or black plastic over the course of each growing season, and the woodchip and NP + grass mulches caused greatest fluctuations in soil temperature. Pumpkin yields were abnormally low in 2011 and did not differ among treatments. In 2012, all mulches produced greater total marketable pumpkin fruit weights compared with bare soil, but only black plastic, newspaper, and NP + grass mulches resulted in greater total numbers of marketable pumpkins. Overall results indicate that shredded newspaper or NP + grass mulches may be useful for organic and/or small-scale urban crop producers as sustainable alternatives to black plastic mulch; however, their weed suppression efficacy may require higher application rates with increasing moisture conditions, and they may require greater squash bug control measures than under bare soil conditions.

Giant ragweed has been increasing as a major weed of row crops in the last 30 yr, but quantitative data regarding its pattern and mechanisms of spread in crop fields are lacking. To address this gap, we conducted a Web-based survey of certified crop advisors in the U.S. Corn Belt and Ontario, Canada. Participants were asked questions regarding giant ragweed and crop production practices for the county of their choice. Responses were mapped and correlation analyses were conducted among the responses to determine factors associated with giant ragweed populations. Respondents rated giant ragweed as the most or one of the most difficult weeds to manage in 45% of 421 U.S. counties responding, and 57% of responding counties reported giant ragweed populations with herbicide resistance to acetolactate synthase inhibitors, glyphosate, or both herbicides. Results suggest that giant ragweed is increasing in crop fields outward from the east-central U.S. Corn Belt in most directions. Crop production practices associated with giant ragweed populations included minimum tillage, continuous soybean, and multiple-application herbicide programs; ecological factors included giant ragweed presence in noncrop edge habitats, early and prolonged emergence, and presence of the seed-burying common earthworm in crop fields. Managing giant ragweed in noncrop areas could reduce giant ragweed migration from noncrop habitats into crop fields and slow its spread. Where giant ragweed is already established in crop fields, including a more diverse combination of crop species, tillage practices, and herbicide sites of action will be critical to reduce populations, disrupt emergence patterns, and select against herbicide-resistant giant ragweed genotypes. Incorporation of a cereal grain into the crop rotation may help suppress early giant ragweed emergence and provide chemical or mechanical control options for late-emerging giant ragweed.

Background:Regulatory T cells (Tregs) prevent autoimmunity and control inflammation. As low-dose interleukin-2 (ld-IL2) expands and activates Tregs, it has a broad therapeutic potential for any autoimmune or inflammatory disease (AIID). We performed a disease-finding “basket trial” (TRANSREGNCT01988506) in patients affected by one of 11 different AIID and reported the outcome of the first 46 patients (Rosenzwajg et al, ARD 2019).Objectives:Here we analyzed and discussed results from deep immunophenotyping, of 78 patients, to comprehensively study the effect of ld-IL2 on the immune system of patients affected by various AIIDMethods:We performed a prospective, open label, phase I-IIa study in 78 patients with a mild to moderate form of one of 13 selected AIID. All patients received ld-IL2 (1 million IU/day) for 5 days, followed by fortnightly injections for 6 months. Deep immunophenotyping was performed before and after 5 days of ld-IL2.Results:ld-IL2 significantly expands both memory Tregs as well as naïve Tregs, including recent thymic emigrant Tregs. It also activates Tregs as demonstrated by the significantly increased expression of HLA-DR, CD39, CD73, GITR, CTLA-4. Zucchini squash (Cucurbita pepo) has a high pollination demand, and the native, ground-nesting squash bee (Peponapis pruinosa) provides the majority of the crop’s pollination requirement in some environments. Squash bees nest directly in crop fields, and nests can be disturbed by tillage and other management operations. Mulches that use municipal waste materials may provide a weed control strategy for squash plantings that is more benign to squash bees than cultivation. Field and greenhouse studies were conducted in 2011 and 2012 to compare the effects of nontillage weed control methods including polyethylene black plastic, woodchips, shredded newspaper, a combination of shredded newspaper plus grass clippings (NP + grass), and bare soil (control) on soil characteristics, squash pollination and fruit production, and squash bee nesting. Woodchips, shredded newspaper, and NP + grass mulch decreased soil temperature, while soils beneath newspaper mulch retained more moisture. Unmarketable, misshapen fruit occurred more frequently in plastic than in the other mulch treatments. No measurable differences in floral resource production or crop pollination were found among treatments, suggesting that misshapen fruit resulted from high soil temperatures in black plastic plots rather than poor pollinator attraction. Squash bee nests were located within bare soil, newspaper, and NP + grass plots, indicating that these mulches did not prevent nesting. NP + grass mulch had a positive effect on plant growth and fruit production, possibly from an addition of plant-available nitrogen or the presence of preferable nesting ground. Shredded newspaper when combined with grass clippings performed as an effective mulch material that improved crop performance with no apparent negative impacts on squash bee nesting or on squash floral resources and pollination.

Summary We report an imported case of Histoplasma capsulatum var. duboisii ( H. duboisii ) infection in a white French woman revealed by cutaneous lesions of the scalp, 18 years after her last stay in West and Central Africa. Asymptomatic bilateral pulmonary infiltrates were discovered on thoracic computed tomography. Skin biopsy allowed the positive diagnosis showing the typical yeasts; culture of biopsy specimens was positive for H. capsulatum . In the absence of criteria of severity, the patient was treated for one year with oral itraconazole 400 mg/day. The outcome was favourable, skin and pulmonary lesions resolved slowly. The follow up is 5 years without relapse after the end of treatment. This case illustrates the possibility of late occurrence of H. duboisii infection, many years after exposure and the major importance of asking any patient for travelling or residency in tropical countries.

BACKGROUND Linear IgA bullous dermatosis (LABD) is a clinically and immunologically heterogeneous, subepidermal, autoimmune bullous disease (AIBD), for which the long-term evolution is poorly described. OBJECTIVES To investigate the clinical and immunological characteristics, follow-up and prognostic factors of adult idiopathic LABD. METHODS This retrospective study, conducted in our AIBD referral centre, included adults, diagnosed between 1995 and 2012, with idiopathic LABD, defined as pure or predominant IgA deposits by direct immunofluorescence. Clinical, histological and immunological findings were collected from charts. Standard histology was systematically reviewed, and indirect immunofluorescence (IIF) on salt-split skin (SSS) and immunoblots (IBs) on amniotic membrane extracts using anti-IgA secondary antibodies were performed, when biopsies and sera obtained at diagnosis were available. Prognostic factors for complete remission (CR) were identified using univariate and multivariate analyses. RESULTS Of the 72 patients included (median age 54 years), 60% had mucous membrane (MM) involvement. IgA IIF on SSS was positive for 21 of 35 patients tested; 15 had epidermal and dermal labellings. Immunoelectron microscopy performed on the biopsies of 31 patients labelled lamina lucida (LL) (26%), lamina densa (23%), anchoring-fibril zone (AFz) (19%) and LL+AFz (23%). Of the 34 IgA IBs, 22 were positive, mostly for LAD-1/LABD97 (44%) and full-length BP180 (33%). The median follow-up was 39 months. Overall, 24 patients (36%) achieved sustained CR, 19 (29%) relapsed and 35% had chronic disease. CR was significantly associated with age > 70 years or no MM involvement. No prognostic immunological factor was identified. CONCLUSIONS Patients with LABD who are < 70 years old and have MM involvement are at risk for chronic evolution.

Giant ragweed (Ambrosia trifida L.) has become one of the most persistent and problematic weeds to farmers and allergy sufferers in North America over the last decades. This study aims to identify and assess the scholarly literature using a bibliometric analysis approach. Based on bibliographic records retrieved from the Web of Science database, the study describes the periodic growth of literature, scientific fields and journals, author productivity and collaborations, institutions and countries, and topical focus and uses article citation analysis to identify the most influential works in this research field during the period of 1903 to 2012.

Trypsin digestion is a major component of preparing proteins for peptide based identification and quantification by mass spectral (MS) analysis. Surprisingly proteolysis is the slowest part of the proteomics process by an order of magnitude. Numerous recent efforts to reduce protein digestion to a few minutes have centered on the use of an immobilized enzyme reactor (IMER) to minimize both trypsin autolysis and vastly increase the trypsin to protein ratio. A central question in this approach is whether proteolysis with an IMER produces the same peptide cleavage products as derived from solution based digestion. The studies reported here examined this question with transferrin; a model protein of known resistance to trypsin digestion. Results from these studies confirmed that a trypsin-IMER can in fact digest transferrin in a few minutes; providing tryptic peptides that subsequent to MS analysis allow sequence identification equivalent to solution digestion. Although many of the peptides obtained from these two trypsin digestion systems were identical, many were not. The greatest difference was that the trypsin- IMER produces (i) numerous peptides bearing multiple lysine and/or arginine residues and (ii) identical portions of the protein sequence were found in multiple peptides. Most of these peptides were derived from five regions in transferrin. These results were interpreted to mean that proteolysis in the case of transferrin occurred faster than the rate at which buried lysine and arginine residues were unmasked in the five regions providing peptides that were only partially digested.

Treatments with atmospheric pressure non-thermal plasma are easy to implement and inexpensive. Among them gliding arc (GlidArc) remains rarely used in surface treatment of polymers. However, it offers economic and flexible way to treat quickly large areas. In addition the choice of carrier gas makes it possible to bring the active species and other radicals allowing different types of grafting and functionalization of the treated surfaces, for example in order to apply for anti-biofouling prevention. This preliminary work includes analysis of the surface of epoxy resins by infrared spectroscopy: the different affected chemical bonds were studied depending on the duration of treatment. The degree of oxidation (the C/O ratio) is obtained by X-ray microanalysis and contact angle analysis have been performed to determinate the wettability properties of the treated surface. A spectroscopic study of the plasma allows to determine the possible active species in the different zones of the discharge.

We used a lectin chromatography/MS-based approach to screen conditioned medium from a panel of luminal (less aggressive) and triple negative (more aggressive) breast cancer cell lines (n = 5/subtype). The samples were fractionated using the lectins Aleuria aurantia (AAL) and Sambucus nigra agglutinin (SNA), which recognize fucose and sialic acid, respectively. The bound fractions were enzymatically N-deglycosylated and analyzed by LC-MS/MS. In total, we identified 533 glycoproteins, ~90% of which were components of the cell surface or extracellular matrix. We observed 1011 glycosites, 100 of which were solely detected in ≥3 triple negative lines. Statistical analyses suggested that a number of these glycosites were triple negative-specific and thus potential biomarkers for this tumor subtype. An analysis of RNAseq data revealed that approximately half of the mRNAs encoding the protein scaffolds that carried potential biomarker glycosites were upregulated in triple negative vs. luminal cell lines, and that a number of genes encoding fucosyl- or sialyltransferases were differentially expressed between the two subtypes, suggesting that alterations in glycosylation may also drive candidate identification. Notably, the glycoproteins from which these putative biomarker candidates were derived are involved in cancer-related processes. Thus, they may represent novel therapeutic targets for this aggressive tumor subtype.

Giant ragweed germination is delayed by both a physiological dormancy of the embryo (embryo dormancy) and an inhibitory influence of embryo-covering structures (covering structure-enforced [CSE] dormancy). To clarify the roles of embryo and CSE dormancy in giant ragweed seedling emergence timing, we conducted two experiments to address the following objectives: (1) determine changes in germinability for giant ragweed dispersal units (hereafter “involucres”) and their components under natural burial conditions, and (2) compare embryo and CSE dormancy alleviation and emergence periodicity between successional and agricultural populations. In Experiment 1, involucres were buried in crop fields at Columbus, OH, periodically excavated, and brought to the laboratory for dissection. Involucres, achenes, and embryos were then subjected to germination assays at 20 C. In Experiment 2, temporal patterns of seedling emergence were determined at a common burial site. Reductions in embryo and CSE dormancy were compared with controlled-environment stratification followed by germination assays at 12 and 20 C, temperatures representative of soil conditions in spring and summer. Results indicated that overwinter dormancy loss involved sequential reductions in embryo and CSE dormancy. CSE dormancy, which may limit potential for fatal germination during fall, was caused by the pericarp and/or embryo-covering structures within the pericarp. In Experiment 2, successional populations emerged synchronously in early spring, whereas agricultural populations emerged throughout the growing season. Levels of embryo dormancy were greater in the agricultural populations than the successional populations, but CSE dormancy levels were similar among populations. In 12 C germination assays, embryo dormancy levels were positively correlated with time required to reach 95% cumulative emergence (run 1:r= 0.81, P = 0.03; run 2:r= 0.76, P = 0.05). These results suggest that late-season emergence in giant ragweed involves high levels of embryo dormancy that prevent germination at low temperatures in spring.

Background: The incidence of severe infections is increased under biologic therapies and the skin is the second localization. Objective: To appraise the factors associated with severe skin infections (SSI) in patients under biologic therapies for inflammatory rheumatic diseases (IRD). Methods: We performed a case-control (ratio 1:3) study nested in a prospective cohort of patients with IRD. SSI was defined as requiring hospitalization or intravenous anti-infectious therapy. We defined two imbedded periods: period A was the time window between the first biologic therapy and the SSI; period B was the last 3 or 12 months (for tumor necrosis factor blockers or rituximab, respectively) before the SSI. Results: Among 4,361 patients with IRD, 29 had a SSI under biologic therapy. In multivariate analyses, SSI were significantly associated with smoking, baseline C-reactive protein and gammaglobulinemia, non-steroidal anti-inflammatory drugs before biologic therapy, cumulative dose of steroids, concomitant steroids during period A, number of different biologic therapies during period A, treatment with infliximab during period A, period B or as first biologic therapy and treatment at high dose during period B. Conclusion: In patients under biologic therapies for IRD, the risk of SSI is associated with several factors including tobacco, treatment with infliximab or high dose range.

The focus of this study was on the assessment of technology that might be of clinical utility in identification, quantification, characterization of carbonylation in human plasma proteins. Carbonylation is widely associated with oxidative stress diseases. Breast cancer patient samples were chosen as a stress positive case based on the fact that oxidative stress has been reported to be elevated in this disease. Measurements of 8-isoprostane in plasma confirmed that breast cancer patients in this study were indeed experiencing significant oxidative stress. Carbonyl groups in proteins from freshly drawn blood were derivatized with biotin hydrazide after which the samples were dialyzed and the biotinylated proteins subsequently selected, digested and labeled with iTRAQ™ heavy isotope coding reagent(s). Four hundred sixty proteins were identified and quantified, 95 of which changed 1.5 fold or more in concentration. Beyond confirming the utility of the analytical method, association of protein carbonylation was examined as well. Nearly one fourth of the selected proteins were of cytoplasmic, nuclear, or membrane origin. Analysis of the data by unbiased knowledge assembly methods indicated the most likely disease associated with the proteins was breast neoplasm. Pathway analysis showed the proteins which changed in carbonylation were strongly associated with Brca1, the breast cancer type-1 susceptibility protein. Pathway analysis indicated the major molecular functions of these proteins are defense, immunity and nucleic acid binding.

Cyclin dependent kinase-5 (Cdk5) activity is deregulated in Alzheimer's disease (AD) and contributes to all three hallmarks: neurotoxic β-amyloid formation, neurofibrillary tangles, and neuronal death. However, the mechanism leading to Cdk5 deregulation remains controversial. Cdk5 deregulation in AD is usually linked to the formation of p25, a proteolysis product of Cdk5 activator p35, which leads to Cdk5 mislocalization and hyperactivation. A few studies have indeed shown increased p25 levels in AD brains; however, others have refuted this observation. These contradictory findings suggest that additional factors contribute to Cdk5 deregulation. This study identified glutathione-S-transferase pi 1 (GSTP1) as a novel Cdk5 regulatory protein. We demonstrate that it is a critical determinant of Cdk5 activity in human AD brains and various cancer and neuronal cells. Increased GSTP1 levels were consistently associated with reduced Cdk5 activity. GSTP1 directly inhibits Cdk5 by dislodging p25/p35, and indirectly by eliminating oxidative stress. Cdk5 promotes and is activated by oxidative stress, thereby engaging a feedback loop which ultimately leads to cell death. Not surprisingly, GSTP1 transduction conferred a high degree of neuroprotection under neurotoxic conditions. Given the critical role of oxidative stress in AD pathogenesis, an increase in GSTP1 level may be an alternative way to modulate Cdk5 signaling, eliminate oxidative stress, and prevent neurodegeneration.

Glycans are cell-type-specific, posttranslational protein modifications that are modulated during developmental and disease processes. As such, glycoproteins are attractive biomarker candidates. Here, we describe a mass spectrometry-based workflow that incorporates lectin affinity chromatography to enrich for proteins that carry specific glycan structures. As increases in sialylation and fucosylation are prominent among cancer-associated modifications, we focused on Sambucus nigra agglutinin (SNA) and Aleuria aurantia lectin (AAL), lectins which bind sialic acid- and fucose-containing structures, respectively. Fucosylated and sialylated glycopeptides from human lactoferrin served as positive controls, and high-mannose structures from yeast invertase served as negative controls. The standards were spiked into Multiple Affinity Removal System (MARS) 14-depleted, trypsin-digested human plasma from healthy donors. Samples were loaded onto lectin columns, separated by HPLC into flow-through and bound fractions, and treated with peptide: N-glycosidase F to remove N-linked glycans. The deglycosylated peptide fractions were interrogated by ESI HPLC-MS/MS. We identified a total of 122 human plasma glycoproteins containing 247 unique glycosites. Importantly, several of the observed glycoproteins (e.g., cadherin 5 and neutrophil gelatinase-associated lipocalin) typically circulate in plasma at low nanogram per milliliter levels. Together, these results provide mass spectrometry-based evidence of the utility of incorporating lectin-separation platforms into cancer biomarker discovery pipelines.

Recent studies have shown that antibodies targeting Lewis x (Le(x)) antigen are a valuable tool in the isolation and identification of glycoproteins in plasma. A focus of this study was to determine whether sialylated Lewis x (sLe(x)) antigen carrying glycoproteins occur in human plasma and whether an antibody targeting this antigen could be used to isolate and identify glycoproteins bearing this antigen. An additional objective was to determine the degree to which proteins conjugated to Le(x) and sLe(x) antigens are similar in structure. A specific anti-sLe(x) antibody (anti-sLe(x)Ab), CHO-131, immobilized in an immunoaffinity column was used to select a set of specific sLe(x) bearing proteins from human plasma, after which they were identified by either of two analytical strategies. One approach was to further resolve the affinity selected proteins by reversed phase chromatography (RPC), tryptic digest the RPC fractions, and identify peptide fragments by MALDI-MS/MS. The second was to tryptic digest the affinity selected protein fraction, further resolve the tryptic fragments by RPC, and identify peptides from RPC fractions by MALDI-MS/MS. Histidine-rich glycoprotein, plasminogen, apolipoprotein A-I, vitronectin, proteoglycan-4, clusterin, Ig gamma-2 chain C region, Ig mu chain C region, and interalpha-trypsin inhibitor heavy chain H4 were found to change three folds or more in association with breast cancer. Fifty percent of the glycoproteins carrying either sLe(x) antigen from CHO-131 selection, Le(x) antigen from selection with TG-1 antibody, or both were found to be changed three folds or more in concentration in breast cancer plasma relative to controls.

With four kinds of mobile phases, methanol-water, ethanol-water, 2-propanol and acetonitrile-water (all containing 0.1% trifluroacetic acid), the displacement between solute and solvent in RPLC was proved to be universal in frontal analysis (FA). Based on the measured Z value in usual RPLC to be a constant and the quantitative determination of methanol increment in mobile phase in FA, the stoichiometric displacement (SD) between insulin and methanol was directly proved by the experiment. The SD was also proved to occur only on about the one-fourth of the maximum amount of adsorbed methanol in the bonded phase layer (BPL) without any dynamic problem of mass transfer, while in FA, the SD firstly occurs on the surface of the BPL and then gradually sinks into the deeper sites companied with a dynamic problem. Although the displaced solvent by the same solute is less in the former case, the SD is independent of how deep of the solute enters the BPL. In addition, the adsorbed amount of solute on an adsorbent not only depends on the numbers of the adsorbed layer on the adsorbent surface, but also on the extent of me complete removal of the displace-able solvent in the BPL. The physical fundamental of the SD and the methodology for investigation were also discussed.

With insulin methanol-water, and the ion-pairing agent, hydrochloric acid and trifluroacetic acid (TFA), the character of the first plateau (FP) on the elution curve of frontal analysis in reversed phase liquid chromatography (RPLC) was investigated by on-line UV-spectrometry and identified with nuclear magnetic resonance (NMR) spectrometry and mass spectrometry. The profile of the FP is the same as that of a usual elution curve of methanol in frontal analysis (FA). When the insulin concentration was limited to a certain range, the height of the FP was found to be proportional to the insulin concentration in mobile phase and its length companying to shorten. The FP profile on the intersection of two tangents reflects the components of the microstructure in the depth direction of the bonded stationary phase layer and the desorption dynamics of the displaced components. The displaced methanol was quantitatively determined by NMR and on-line UV spectrometries. TFA with high UV absorbance can not be used as an ion-pairing agent for the investigation of the FP in RPLC, but it can be used as a good marker to investigate the complicated transfer process of components in the stationary phase in RPLC. A stoichiometric displacement process between solute and solvent was proved to be valid in both usual and FA in RPLC. From the point of view of dynamics of mass transfer, the solutes can only contact to the surface of stationary phase in usual RPLC, while solute can penetrate into it in FA of RPLC. The solvation of insulin in methanol and water solution as an example indicating the usage of the FP in the FA was also investigated in this paper.

An on-line UV spectrometric method for the quantitative determination of methanol increment of methanol-water in the mobile phase (i.e., of greater concentration than that of the mobile phase) by frontal analysis (FA) of insulin in reversed phase liquid chromatography (RPLC) was presented. When the methanol increment concentration ranged from 0 05% to 0 50%, V(CH 3OH)/V(H 2O), a set of elution curves could be obtained at 198 nm by a diode-array detector in the presence of 47% methanol, V(CH 3OH)/V(H 2O) containing 0 03% hydrochloric acid, V(CH 3OH-H 2O)/V(HCl) in the mobile phase. The plateau height of the elution curves of the methanol increment was found to be proportional to the methanol increment in the mobile phase. The methanol increment could be determined on a quantitative basis. When the method was used to investigate the elution curve of insulin by FA in RPLC, a small plateau, being the methanol increment, was detected before the usual insulin plateau of each elution curve. In this case the methanol increment was found to vary with insulin concentration in the mobile phase. When that concentration was between 0 025 mg/mL and 0 30 mg/mL, the methanol increment could be determined in the range from 0 03% to 0 19% with a deviation of ±0 02% and a relative deviation of ±10%. A nuclear magnetic resonance spectrometer (NMR) was also employed to confirm the obtained result. A methodology with a very rigorous experimental procedure for obtaining results of such accuracy is also included. The presented result may be used to prove that a displacement process definitely occurs as insulin is adsorbed by the RPLC stationary phase inFA.

The earthworm, Lumbricus terrestris L., caches seeds of the agricultural weed Ambrosia trifida L. in its burrow, providing seeds with a protected overwintering site. Seedlings subsequently emerge from the burrows, resulting in an association of the two species (hereafter “LtAt association”). Although populations of these species frequently co-exist in no-till agricultural fields in the eastern U.S. Corn Belt, an association is not always evident. To identify environmental influences on the LtAt association, 30 no-till agricultural fields were surveyed across the eastern U.S. Corn Belt during spring 2007, 2008 and 2009. The LtAt association occurred across states and soil types, but the strength of the association varied with climate differences during the previous September through March. The strongest environmental driver of LtAt association was frequency of “moderate rain day” (MRD; day that received 12.8–25.3 mm of precipitation), with a 1-day increase in MRD frequency increasing the odds of LtAt association by a factor of 1.42. Thus, the potential for L. terrestris to cache seeds and facilitate seedling recruitment is increased by precipitation frequency and amount during September through March. These results highlight the importance of climate variation within a region in driving trophic interactions that regulate weed population dynamics.

Excessive oxidative stress leaves a protein carbonylation fingerprint in biological systems. Carbonylation is an irreversible post-translational modification (PTM) that often leads to the loss of protein function and can be a component of multiple diseases. Protein carbonyl groups can be generated directly (by amino acids oxidation and the alpha-amidation pathway) or indirectly by forming adducts with lipid peroxidation products or glycation and advanced glycation end-products. Studies of oxidative stress are complicated by the low concentration of oxidation products and a wide array of routes by which proteins are carbonylated. The development of new selection and enrichment techniques coupled with advances in mass spectrometry are allowing the identification of hundreds of new carbonylated protein products from a broad range of proteins located at many sites in biological systems. The focus of this review is on the use of proteomics tools and methods to identify oxidized proteins along with specific sites of oxidative damage and the consequences of protein oxidation.
