Abstract Recent advances in DNA nanotechnology allow for the assembly of nanocomponents with nanoscale precision, leading to the emergence of DNA-based material fabrication approaches. Yet, transferring these nano- and micron-scale structural arrangements to the macroscale morphologies remains a challenge, which limits the development of materials and devices based on DNA nanotechnology. Here, we demonstrate a materials fabrication approach that combines DNA-programmable assembly with actively driven processes controlled by acoustic fields. This combination provides a prescribed nanoscale order, as dictated by equilibrium assembly through DNA-encoded interactions, and field-shaped macroscale morphology, as regulated by out-of-equilibrium materials formation through specific acoustic stimulation. Using optical and electron microscopy imaging and x-ray scattering, we further revealed the nucleation processes, domain fusion, and crystal growth under different acoustically stimulated conditions. The developed approach provides a pathway for the fabrication of complexly shaped macroscale morphologies for DNA-programmable nanomaterials by controlling spatiotemporal characteristics of the acoustic fields.

Abstract Recent attention has focused on accelerated glacial losses along the Amundsen Sea coast that result from changes in atmosphere and ocean circulation, with sea ice playing a mediating but not well-understood role. Here, we investigated how sea ice has changed in the Amundsen Sea over the period of 1979 to 2014, focusing on spatio-temporal changes in ice edge advance/retreat and percent sea ice cover in relation to changes in winds. In contrast to the widespread sea ice decreases to the east and increases to the west of the Amundsen Sea, sea ice changes in the Amundsen Sea were confined to three areas: (i) offshore of the shelf break, (ii) the southern Pine Island Polynya, and (iii) the eastern Amundsen Sea Polynya. Offshore, a 2-month decrease in ice season duration coincided with seasonal shifts in wind speed and direction from March to May (relating to later ice advance) and from September to August (relating to earlier retreat), consistent with reported changes in the depth/location of the Amundsen Sea Low. In contrast, sea ice decreases in the polynya areas corresponded to episodic or step changes in spring ice retreat (earlier by 1–2 months) and were coincident with changes to Thwaites Iceberg Tongue (located between the two polynyas) and increased southeasterly winds. Temporal correlations among these three areas were weak, indicating different local forcing and/or differential response to large-scale forcing. Although our analysis has shown that part of the variability can be explained by changes in winds or to the coastal icescape, an additional but unknown factor is how sea ice has responded to changes in ocean heat and freshwater inputs. Unraveling cause and effect, critical for predicting changes to this rapidly evolving ocean-ice shelf-sea ice system, will require in situ observations, along with improved remote sensing capabilities and ocean modeling.

Rain gardens installed as green infrastructure to divert storm runoff from entering combined sewers also collect dissolved constituents and particulates. An urban rain garden in northwestern Indiana, USA, was continuously monitored from November 2019 to May 2021 to evaluate the fate of dissolved constituents entering the rain garden in runoff. Physical and chemical properties of soils in the rain garden were also monitored, along with underlying groundwater. Linear regression models relating specific conductance to chloride concentration indicated that the 0.0371-ha (3998 square feet) rain garden collected approximately 1490 kg (3285 pounds) of road salt from the surrounding 0.2228 ha (24,500 square feet) of impervious surfaces. Soils and groundwater were seasonally affected by road salt application but carryover from year to year was not indicated. Rain garden soil permeability (5.20 × 10−5 to 9.72 × 10−5 m/s) remained unchanged during the study period and soil organic carbon generally increased under native vegetation. The results suggest that a rain garden built on sandy soil can divert substantial quantities of runoff and dissolved constituents from combined sewers; however, chloride is transported to sub-infrastructure groundwater that eventually discharges to adjacent waterways with concentrations lower than those observed in runoff. The mesospheric polar vortex (MPV) plays a critical role in coupling the atmosphere-ionosphere system, so its accurate simulation is imperative for robust predictions of the thermosphere and ionosphere. While the stratospheric polar vortex is widely understood and characterized, the mesospheric polar vortex is much less well-known and observed, a short-coming that must be addressed to improve predictability of the ionosphere. The winter MPV facilitates top-down coupling via the communication of high energy particle precipitation effects from the thermosphere down to the stratosphere, though the details of this mechanism are poorly understood. Coupling from the bottom-up involves gravity waves (GWs), planetary waves (PWs), and tidal interactions that are distinctly different and important during weak vs. strong vortex states, and yet remain poorly understood as well. Moreover, generation and modulation of GWs by the large wind shears at the vortex edge contribute to the generation of traveling atmospheric disturbances and traveling ionospheric disturbances. Unfortunately, representation of the MPV is generally not accurate in state-of-the-art general circulation models, even when compared to the limited observational data available. Models substantially underestimate eastward momentum at the top of the MPV, which limits the ability to predict upward effects in the thermosphere. The zonal wind bias responsible for this missing momentum in models has been attributed to deficiencies in the treatment of GWs and to an inaccurate representation of the high-latitude dynamics. In the coming decade, simulations of the MPV must be improved.

PurposeJanus kinase-1 (JAK1) tyrosine kinase mediates signaling from multiple cytokine receptors, including interferon alpha/beta and gamma (IFN-α/β and IFN-γ), which are important for viral and mycobacterial protection respectively. We previously reported autosomal recessive (AR) hypomorphic JAK1 mutations in a patient with recurrent atypical mycobacterial infections and relatively minor viral infections. This study tests the impact of partial JAK1 deficiency on cellular responses to IFNs and pathogen control.MethodsWe investigated the role of partial JAK1 deficiency using patient cells and cell models generated with lentiviral vectors expressing shRNA.ResultsPartial JAK1 deficiency impairs IFN-γ-dependent responses in multiple cell types including THP-1 macrophages, Epstein-Barr Virus (EBV)-transformed B cells and primary dermal fibroblasts. In THP-1 myeloid cells, partial JAK1 deficiency reduced phagosome acidification and apoptosis and resulted in defective control of mycobacterial infection with enhanced intracellular survival. Although both EBV-B cells and primary dermal fibroblasts with partial JAK1 deficiency demonstrate reduced IFN-α responses, control of viral infection was impaired only in patient EBV-B cells and surprisingly intact in patient primary dermal fibroblasts.ConclusionOur data suggests that partial JAK1 deficiency predominantly affects susceptibility to mycobacterial infection through impact on the IFN-γ responsive pathway in myeloid cells. Susceptibility to viral infections as a result of reduced IFN-α responses is variable depending on cell type. Description of additional patients with inherited JAK1 deficiency will further clarify the spectrum of bacterial and viral susceptibility in this condition. Our results have broader relevance for anticipating infectious complications from the increasing use of selective JAK1 inhibitors.

Summary: Health inequities and decreasing median American lifespan, potentiated by the worldwide COVID 19 crisis, have taken centre stage in the public consciousness. Specifically, for this discourse, rural radiation oncology challenges external to the pandemic and unique to the rural American radiation oncology care delivery result from a confluence of the following: a) increased incidence of cancer in the United States;1 b) recent legislative emphasis on rural healthcare equity initiatives;2 c) pandemic-associated delays in cancer screening, diagnosis, and treatment3,4 with resultant presentation of advanced oncologic stages; d) social spotlight on healthcare equity and inclusion for disenfranchised populations.5We will attempt to delineate these issues and propose widely applicable common-sense solutions. We will review what has transpired at the University of Kentucky over the last two decades, specifically at radiation oncology centre in Morehead, a clinic in eastern Kentucky in the Appalachian foothills. While much more work remains ahead, this clinic has successfully applied many of the initiatives discussed. Abstract Background Terms used to describe government-led resource withdrawal from ineffective and unsafe medical services, including ‘rationing’ and ‘disinvestment’, have tended to be used interchangeably, despite having distinct characteristics. This lack of descriptive precision for arguably distinct terms contributes to the obscurity that hinders effective communication and the achievement of evidence-based decision-making. The objectives of this study are to (1) identify the various terms used to describe resource withdrawal and (2) propose definitions for the key or foundational terms, which includes a clear description of the unique characteristics of each. Methods This is a systematic qualitative synthesis of characteristics and terms found through a search of the academic and grey literature. This approach involved identifying commonly used resource withdrawal terms, extracting data about resource withdrawal characteristics associated with each term and conducting a comparative analysis by categorising elements as antecedents, attributes or outcomes. Results Findings from an analysis of 106 documents demonstrated that terms used to describe resource withdrawal are inconsistently defined and applied. The characteristics associated with these terms, mainly antecedents and attributes, are used interchangeably by many authors but are differentiated by others. Our analysis resulted in the development of a framework that organises these characteristics to demonstrate the unique attributes associated with each term. To enhance precision, these terms were classified as either policy options or patient health outcomes and refined definitions for rationing and disinvestment were developed. Rationing was defined as resource withdrawal that denies, on average, patient health benefits. Disinvestment was defined as resource withdrawal that results in, on average, improved or no change in health benefits. Conclusion Agreement on the definition of various resource withdrawal terms and their key characteristics is required for transparent government decision-making regarding medical service withdrawal. This systematic qualitative synthesis presents the proposed definitions of resource withdrawal terms that will promote consistency, benefit public policy dialogue and enhance the policy-making process for health systems.

Two important approaches for satellite studies of polar mesospheric clouds (PMCs) are nadir measurements adapting phase function analysis and limb measurements adapting spectroscopic analysis. Combining both approaches enables new studies of cloud structures and microphysical processes but is complicated by differences in scattering conditions, observation geometry and sensitivity. In this study, we compare common volume PMC observations from the nadir-viewing Cloud Imaging and Particle Size (CIPS) instrument on the Aeronomy of Ice in the Mesosphere (AIM) satellite and a special set of tomographic limb observations from the Optical Spectrograph and InfraRed Imager System (OSIRIS) on the Odin satellite performed over 18 d for the years 2010 and 2011 and the latitude range 78 to 80∘ N. While CIPS provides preeminent horizontal resolution, the OSIRIS tomographic analysis provides combined horizontal and vertical PMC information. This first direct comparison is an important step towards co-analysing CIPS and OSIRIS data, aiming at unprecedented insights into horizontal and vertical cloud processes. Important scientific questions on how the PMC life cycle is affected by changes in humidity and temperature due to atmospheric gravity waves, planetary waves and tides can be addressed by combining PMC observations in multiple dimensions. Two- and three-dimensional cloud structures simultaneously observed by CIPS and tomographic OSIRIS provide a useful tool for studies of cloud growth and sublimation. Moreover, the combined CIPS/tomographic OSIRIS dataset can be used for studies of even more fundamental character, such as the question of the assumption of the PMC particle size distribution. We perform the first thorough error characterization of OSIRIS tomographic cloud brightness and cloud ice water content (IWC). We establish a consistent method for comparing cloud properties from limb tomography and nadir observations, accounting for differences in scattering conditions, resolution and sensitivity. Based on an extensive common volume and a temporal coincidence criterion of only 5 min, our method enables a detailed comparison of PMC regions of varying brightness and IWC. However, since the dataset is limited to 18 d of observations this study does not include a comparison of cloud frequency. The cloud properties of the OSIRIS tomographic dataset are vertically resolved, while the cloud properties of the CIPS dataset is vertically integrated. To make these different quantities comparable, the OSIRIS tomographic cloud properties cloud scattering coefficient and ice mass density (IMD) have been integrated over the vertical extent of the cloud to form cloud albedo and IWC of the same quantity as CIPS cloud products. We find that the OSIRIS albedo (obtained from the vertical integration of the primary OSIRIS tomography product, cloud scattering coefficient) shows very good agreement with the primary CIPS product, cloud albedo, with a correlation coefficient of 0.96. However, OSIRIS systematically reports brighter clouds than CIPS and the bias between the instruments (OSIRIS – CIPS) is 3.4×10-6 sr−1 (±2.9×10-6 sr−1) on average. The OSIRIS tomography IWC (obtained from the vertical integration of IMD) agrees well with the CIPS IWC, with a correlation coefficient of 0.91. However, the IWC reported by OSIRIS is lower than CIPS, and we quantify the bias to −22 g km−2 (±14 g km−2) on average.

A new species of the butterflyfish genus Prognathodes (Chaetodontidae) is described from two specimens collected at a depth of 116 m off Ngemelis Island, Palau. Prognathodes geminus sp. n. is similar to P. basabei Pyle & Kosaki, 2016 from the Hawaiian archipelago, and P. guezei (Maugé & Bauchot, 1976) from the western Indian Ocean, but differs from these species in the number of soft dorsal-fin rays, size of head, body width, and body depth. There are also subtle differences in life color, and substantial differences in the mtDNA cytochrome oxidase I sequence (d ≈ 0.08). Although genetic comparisons with P. guezei are unavailable, it is expected that the genetic divergence between P. guezei and P. geminus will be even greater than that between P. geminus and P. basabei. It is named for the strikingly similar color pattern it shares with P. basabei.

The new species Tosanoides annepatrice sp. n. is described from four specimens collected at depths of 115–148 m near Palau and Pohnpei in Micronesia. It differs from the other three species of this genus in life color and in certain morphological characters, such as body depth, snout length, anterior three dorsal-fin spine lengths, caudal-fin length, and other characters. There are also genetic differences from the other four species of Tosanoides (d ≈ 0.04–0.12 in mtDNA cytochrome oxidase I). This species is presently known only from Palau and Pohnpei within Micronesia, but it likely occurs elsewhere throughout the tropical western Pacific.

Abstract Background Reducing monies spent on unnecessary medical care is one possible target to improve value in healthcare systems. Regional variation in the provision of medical care suggests physician behaviour and patient demands influence the provision of unnecessary medical care. Recently, Choosing Wisely campaigns began using ‘top 5 do-not-do’ lists to target unnecessary medical care by encouraging greater physician and patient dialogue at the point of care. The present study aims to examine the rationale for Choosing Wisely Canada’s (CWC) design and to analyse physician perceptions regarding the features of CWC aimed to reduce unnecessary medical care. Methods The study involved semi-structured interviews with 19 key informant physicians with CWC experience and the application of procedures of grounded theory to analyse interview transcripts and develop explanations addressing the objectives. Results Participants reported that the CWC was the medical community’s response to three pressures, namely (1) demand for unnecessary medical care from patients during the clinical encounter; (2) public perception that physicians do not always prioritise patients’ needs; and (3) ‘blunt’ government tools aimed to reduce costs rather than improving patient care. Respondents stated that involving the patient in decision-making would help alleviate these pressures by promoting the clinical encounter as the paramount decision-point in achieving necessary care. However, CWC does not address several of the key reasons, from a physician perspective, for providing unnecessary medical care, including time pressures in the clinical encounter, uncertainty about the optimal care pathway and fear of litigation. Conclusion This study contributes to our understanding of the perceptions of physicians regarding the CWC campaign. Specifically, physicians believe that CWC does little to address the clinical reasons for unnecessary medical care. Ultimately, because CWC has limited impact on physician behaviour or patient expectations, it is unlikely to have a major influence on unnecessary medical care.

Using data from the Aeronomy of Ice in the Mesosphere (AIM) and Aura satellites, we have categorized the interannual variability of winter- and springtime upper stratospheric methane (CH4). We further show the effects of this variability on the chemistry of the upper stratosphere throughout the following summer. Years with strong wintertime mesospheric descent followed by dynamically quiet springs, such as 2009, lead to the lowest summertime CH4. Years with relatively weak wintertime descent, but strong springtime planetary wave activity, such as 2011, have the highest summertime CH4. By sampling the Aura Microwave Limb Sounder (MLS) according to the occultation pattern of the AIM Solar Occultation for Ice Experiment (SOFIE), we show that summertime upper stratospheric chlorine monoxide (ClO) almost perfectly anticorrelates with the CH4. This is consistent with the reaction of atomic chlorine with CH4 to form the reservoir species, hydrochloric acid (HCl). The summertime ClO for years with strong, uninterrupted mesospheric descent is about 50 % greater than in years with strong horizontal transport and mixing of high CH4 air from lower latitudes. Small, but persistent effects on ozone are also seen such that between 1 and 2 hPa, ozone is about 4–5 % higher in summer for the years with the highest CH4 relative to the lowest. This is consistent with the role of the chlorine catalytic cycle on ozone. These dependencies may offer a means to monitor dynamical effects on the high-latitude upper stratosphere using summertime ClO measurements as a proxy. Additionally, these chlorine-controlled ozone decreases, which are seen to maximize after years with strong uninterrupted wintertime descent, represent a new mechanism by which mesospheric descent can affect polar ozone. Finally, given that the effects on ozone appear to persist much of the rest of the year, the consideration of winter/spring dynamical variability may also be relevant in studies of ozone trends.

Johnston Atoll is isolated in the Central Pacific Ocean (16°45′ N 169°31′ W) about 1287 km (800 miles) southwest of Honolulu, Hawaii and 1440 km (900 miles) north of the equatorial Line Islands, Kiribati. The labrid species, Thalassoma lutescens, has a wide range of distribution in the equatorial Pacific. The related species, Thalassoma duperrey, is endemic to the Hawaiian Islands. The pelagic larvae of both species dispersed to Johnston Atoll, where we found a mix of adult phenotypes representing a range of hybridization events over generations. A hybrid acanthurid was also documented. In addition, the arrival and colonization of two pomacentrid (damselfish) species to the atoll was observed in 1999. These pomacentrid sister-species, Abudefduf abdominalis and A. vaigiensis, have become established populations with subsequent hybridization. The biogeography of the Johnston Atoll coral reef fish population shows some degree of local population retention. It is also evident that this biogeographic isolation is periodically compromised by large ocean current oscillations in the equatorial and central Pacific Ocean that bring larval fishes from either Hawaii or the Line Islands, and may distribute Johnston Atoll originating larvae elsewhere as well. The reef fauna and oceanography of this atoll provides the circumstances for improving scientific insight into marine fish speciation and island biogeography.
