Abstract

OBJECTIVE: Gastric cancer is one of the gastrointestinal tract tumors associated with a poor prognosis. Ficus carica L. is one of the most outstanding species among many traditional medicines. The aim of our study is to determine the antioxidant content of Ficus carica L., and its therapeutic effects on gastric cancer. PATIENTS AND METHODS: Ficus carica L. was extracted in a different solvent. The antioxidant parameters of the different concentrations of extracts were measured photometrically. The human gastric adenocarcinoma cells (AGS) were incubated with different concentrations of Ficus carica L. extract for 24 hours; cell viability by the ATP test, and intracellular reactive oxygen species levels were determined by H2DCF-DA fluorescent dye. Intracellular glutathione levels luminometrically, and mitochondrial membrane potential fluorometrically were detected. The apoptotic effect was analyzed with acridine orange/ ethidium bromide, and the genotoxic effect by the comet assay. Abstract

Objective:Irisin is a recently discovered protein and is defined as an adipomyokine. The relation of irisin with carbohydrate metabolism and other hormone parameters have been investigated. However, studies evaluating the relationship between irisin and puberty are limited and inconclusive. The aim was to evaluate serum concentrations of irisin during different pubertal stages in male adolescents.Methods:The study included normal weight pubertal male adolescents between the ages of 136/12-1411/12 who had entered puberty. Fasting serum irisin concentrations were evaluated, and bioelectrical impedance analysis was used to measure body fat ratio (BFR) and fat-free mass (FFM). BFR was also calculated by caliper measurement of subcutaneous fat at the triceps.Results:Sixty-eight adolescents were enrolled. The number of adolescents in pubertal stage 2, 3, 4 and 5 were n=17 (25%), n=13 (19.1%), n=21 (30.1%) and n=17 (25%), respectively. The median values of the irisin are 8.80, 8.20, 9.15 and 7.24 ng/mL according to the 2-5 pubertal stages, respectively. The levels of circulating irisin did not differ according to the pubertal stage. Additionally, there was no significant relationship between irisin levels and body fat percentage or FFM.Conclusion:Irisin levels do not differ after the onset of puberty or with progressing pubertal maturation. This study strengthens the evidence that there is no change in irisin concentration as puberty progresses. This may have important implications when using this adipomyokine in the future for diagnosis or treatment of obesity-related diseases.

Abstract

Cherry tomatoes inoculated with Escherichia coli O157:H7 and Salmonella Typhimurium were treated with vaporised ethyl pyruvate (EP). The changes of microbial and organoleptic properties of the samples during storage were investigated at two temperatures (4 and 10 °C) and four EP concentrations (0, 42, 105, and 420 ppm) for 7 days at 4 °C and for 5 days 10 °C. After 3 days, 4.3 log and 3.6 log reductions in E. coli O157:H7 numbers were detected in cherry tomatoes treated with 42 ppm EP at 4 °C and at 10 °C, respectively. The highest EP treatment (420 ppm) led to 5.7 log CFU g−1 E. coli O157:H7 reduction after 1 day at 4 and 10 °C. The reduction of Salmonella Typhimurium on samples treated with 420 ppm EP was 7.7 log CFU g–1 after 1 day at 4 °C, and 6.9 log after 1 day at 10 °C. The treatment of EP can be effective at decreasing pathogen populations and can protect the organoleptic and colour properties of fresh produce.

Abstract

The effects of nitrogen implantation conditions (ion energy, dose rate, and processing time) on the thickness and wear behavior of N-rich layers produced on 304 stainless-steel surfaces are examined. Surfaces implanted at elevated temperatures (≈400°C) with 0.4 to 2 keV nitrogen ions at high dose rates (1.5 to 3.8 mA/cm2) are compared to surfaces implanted at higher energies (30 to 60 keV) and lower current densities (0.1 to 0.25 mA/cm2). The most wear-resistant surfaces are observed when the implanted-ion energy is near 1 keV and the dose is very large (> 2 × 1019 ions/cm2). Typically, surfaces implanted under these optimum conditions exhibit load-bearing capabilities at least 1000 times that of the untreated material. Some comparisons are also made to surfaces processed using conventional plasma-nitriding. Samples treated using either process have wear-resistant surface layers in which the nitrogen is in solid solution in the fcc phase. It is argued that the deep N migration (> 1 μm) that occurs under low-energy implantation conditions is due to thermal diffusion that is enhanced by a mechanism other than radiation-induced vacancy production.

Dark current measurements have been done at the range from 6 K to 290 K temperature. Activation energies of the carriers have been obtained from the temperature dependence of the I-V measurements. The change of the activation energy with bias voltage has also been obtained. From the activation energy at zero bias and calculated quasi Fermi energy, barrier heights of the quantum wells and ground state energies were obtained. Abstract

This paper provides a general overview of solid waste data and management practices employed in Turkey during the last decade. Municipal solid waste statistics and management practices including waste recovery and recycling initiatives have been evaluated. Detailed data on solid waste management practices including collection, recovery and disposal, together with the results of cost analyses, have been presented. Based on these evaluations basic cost estimations on collection and sorting of recyclable solid waste in Turkey have been provided. The results indicate that the household solid waste generation in Turkey, per capita, is around 0.6 kg/year, whereas municipal solid waste generation is close to 1 kg/year. The major constituents of municipal solid waste are organic in nature and approximately 1/4 of municipal solid waste is recyclable. Separate collection programmes for recyclable household waste by more than 60 municipalities, continuing in excess of 3 years, demonstrate solid evidence for public acceptance and continuing support from the citizens. Opinion polls indicate that more than 80% of the population in the project regions is ready and willing to participate in separate collection programmes. The analysis of output data of the Material Recovery Facilities shows that, although paper, including cardboard, is the main constituent, the composition of recyclable waste varies strongly by the source or the type of collection point.

15. Outcome analysis of reoperations after lumbar discectomies: a report of 22 patients

Our aim was to analyse causes for persistence of pain after lumbar discectomy and outcome of reoperations. Out of 37 reoperated patients, 22 with a minimum follow-up period of one year were included in this retrospective study concerning the years 1993 to 2000. All patients had previously undergone laminotomy and discectomy and fusion was not required during second operations. Outcome was evaluated according to the modified criteria of Kawabata et al.. Overall incidence of reoperation was 6.5%. Reoperations were performed because of recurrent disc herniation in 9 patients, epidural fibrosis in 8, and de novo disc herniation at a different level in 5. Contrast enhanced computerized tomography was used in 17 patients and this might be the reason for misdiagnosis of recurrent disc herniation in the two patients with epidural fibrosis. In patients with de novo disc herniation, symptoms recurred earlier. Excellent results were obtained more in patients with recurrent disc herniation and poor outcomes correlated with long (1 year) time intervals for onset of recurrent sciatica (p0.05). However, patients with epidural fibrosis were also glad postoperatively for decreased pain severity. Only co-existence of epidural fibrosis and de novo disc herniation predicted an inferior outcome. Although recurrent disc herniation seemed to respond best to surgical treatment, we recommend reoperation when objective preoperative findings indicate the presence of surgically correctable compression regardless of its type.

Abstract

In order to clarify the formation and growth kinetics of iron nitrides, Fe2N (ζ), Fe2–3N (ɛ) and Fe4N (γ′), on the surface of iron during ion-nitriding, and their contribution to the mechanism of ion-nitriding coarse-grained specimens (2 to 6 mm) were ion-nitrided in pure nitrogen and nitrogen-hydrogen (20%–80%) plasma at temperatures between 500 and 600°C. Reflection electron diffraction (RED) showed immediate formation of the nitrides. Growth of these phases in latter stages of ion-nitriding was studied using optical microscopy and X-ray diffraction. The mechanism of the nitride layer formation is discussed and compared with the existing gas nitriding data. In the case region, Fe4N (γ′) and Fe16N2 (α″) precipitation was observed to occur under all experimental conditions studied. Case depth is seen to be parabolic with time and nitriding rate increases slightly when nitrogen-hydrogen plasma is used. Discussions are given to explain the difference in the nitriding efficiency under two different plasma compositions.

Abstract

STAT3 plays an important role in various complex and sometimes contradictory pathways such as proliferation, differentiation, migration, inflammation, and apoptosis. The transcriptional activity of the STAT3 gene is controlled by a transcription factor called ZNF341. There is insufficient data on radiation sensitivity and post-radiation DNA repair in STAT3- loss-of-function (LOF) patients. We aimed to investigate the radiosensitivity in patients with STAT3-LOF and ZNF341 deficiency. Twelve patients with STAT3-LOF and four ZNF341-deficiency patients were recruited from three clinical immunology centers in Turkey and evaluated for radiosensitivity by the Comet assay, comparing to 14 age- and sex-matched healthy controls. The tail length (TL) (μm), percentage of DNA in the tail (TDNA%), and olive tail moment (OTM) (arbitrary units) were evaluated at the same time for baseline (spontaneous), initial (immediately after 2 Gy irradiation), and recovery (2 h after irradiation) periods by using a computerized image-analysis system, estimating DNA damage. Except for a patient with ZNF341 deficiency who developed nasal cell primitive neuroendocrine tumor and papillary thyroid cancer during the follow-up, there was no cancer in both groups. During the recovery period of irradiation, TL, TDNA%, and OTM values of healthy controls decreased rapidly toward the baseline, while these values of patients with STAT3-LOF and ZNF341 deficiency continued to increase, implying impaired DNA repair mechanisms. Increased radiosensitivity and impaired DNA repair were demonstrated in patients diagnosed with STAT3-LOF and ZNF341 deficiency, potentially explaining the susceptibility to malignant transformation.

Abstract

© 2022 Paediatrics and Child Health Division (The Royal Australasian College of Physicians).Aim: Multisystem inflammatory syndrome in children (MIS-C) may cause shock and even death in children. The aim of this study is to describe the clinical features, laboratory characteristics and outcome of children diagnosed with MIS-C in 25 different hospitals in Turkey. Methods: The retrospective study was conducted between 8 April and 28 October 2020 in 25 different hospitals from 17 cities. Data were collected from patients' medical records using a standardised form. Clinical and laboratory characteristics and outcomes according to different age groups, gender and body mass index percentiles were compared using multivariate logistic regression analysis. Results: The study comprised 101 patients, median age 7 years (interquartile range (IQR) 4.6–9.3); 51 (50.5%) were boys. Reverse-transcriptase polymerase chain reaction (PCR) assay was positive in 21/100 (21%) patients; 62/83 (74.6%) patients had positive serology for SARS-CoV-2. The predominant complaints were fever (100%), fatigue (n = 90, 89.1%), and gastrointestinal symptoms (n = 81, 80.2%). Serum C-reactive protein (in 101 patients, median 165 mg/L; range 112–228), erythrocyte sedimentation rate (73/84, median 53 mm/s; IQR 30–84) and procalcitonin levels (86/89, median 5 μg/L; IQR 0.58–20.2) were elevated. Thirty-eight patients (37.6%) required admission to intensive care. Kawasaki disease (KD) was diagnosed in 70 (69.3%) patients, 40 of whom had classical KD. Most patients were treated with intravenous immunoglobulin (n = 92, 91%) and glucocorticoids (n = 59, 58.4%). Seven patients (6.9%) died. Conclusion: The clinical spectrum of MIS-C is broad, but clinicians should consider MIS-C in the differential diagnosis when persistent fever, fatigue and gastrointestinal symptoms are prominent. Most patients diagnosed with MIS-C were previously healthy. Immunomodulatory treatment and supportive intensive care are important in the management of cases with MIS-C. Glucocorticoids and intravenous immunoglobulins are the most common immunomodulatory treatment options for MIS-C. Prompt diagnosis and prompt treatment are essential for optimal management.

Yapmış olduğumuz çalışmada dış merkezde opere edilerek çeşitli yakınmalarla kliniğimize başvurmuş ve reopere edilerek pedikül vidaları çıkartılan hastalardan 12'si randomize olarak seçilerek pedikül vidalarının pediküle giren kısımlarının madde analizi yapılarak yerli ve yabancı birçok farklı kaynaktan elde edilmiş olan vidaların alaşımları (titanyum, alüminyum ve azot)arasında farklılık ortaya konarak özellikle kırılma nedeniyle çıkartılan vidaların alaşımının sağlam vidaların alaşımıyla farklılık gösterip göstermediği üzerinde duruldu.Çalışmanın amacı spinal enstrümantasyonun başarısız olduğu vakalarda pedikül vidalarının alaşımının bu yetersizlikte herhangi bir fonksiyonunun olup olmadığını belirlemekti.İstatistiksel Analiz Mann-Whitney ve Kruskal Vallis testleri yapılarak elde edildi.Yapmış olduğumuz istatistiksel analiz sonucunda üzerinde çalıştığımız örneklerde vida alaşım oranlarının dayanıklılık üzerinde istatistiksel olarak anlamlı bir etkisinin olmadığını gözlemledik. Bu veriden yola çıkarak, vida boyutu ve hasta ve endikasyonla ilgili faktörler daha ön plana çıkmaktadır. Enstrümanlı spinal füzyon kararı vermeden önce doğru hasta seçimi, doğru endikasyon ve özellikle postoperatif dönemde hastanın uyması gereken yaşam tarzı kuralları net biçimde ortaya konmalıdır İn this study we retrospectively analysed patients that were reoperated in our clinic due to failed spinal fusion with instrumentation. 12 of these patients were randomly selected and the pedicle screws were analysed to determine the compound of the screws.The screws were analysed for the amounts of titanium , aluminium and nitrogen We aimed to determine whether the compound of the screws played a role in failure of the surgery and especially whether there was a difference between compound of the broken screws and the intact ones.Statistical analysis was made usin Mann-Whitney and Kruskal Vallis tests.There was no statistically significant difference between the broken and intact screws in regard to compound.This result made us think about factors like screw size, patient selection and establishing the right indication.Before considerig spinal fusion with instrumentation the right patient, right indication , right materials and strict rules in the postoperative period should be regarded 58

71 7) ÖZET: Özellikle çocukluk yaş grubunun önemli tümörlerinden olan medulloblastomlar, nöroonkoloji literatüründe isimlendirmeden prognostik faktörleri ve sitogenetik özelliklerine kadar pek çok konuda görüş ayrılıklarının yaşandığı bir tümör türüdür. Bu çalışmada, medulloblastomların histolopatolojik özelliklerinden Kopelson skoru, p53 immünhistokimyasal boyanma özellikleri ve Kİ67 immünhistokimyasal boyanma özellikleri gerek klinik parametreler, gerekse mortalitenin yıllara göre dağılımı açısmdan incelenmiştir. Kopelson skoru, olguların Chang T evresi, cerrahi rezeksiyon derecesi ve postoperatif tümör rezidüsü açısından; p53 immünhistokimyasal boyanması, Chang M evresi açısmdan; Kİ67 immünhistokimyasal boyanması ise, preoperatif ve postoperatif ECOG skoru ile beraber postoperatif rezidü varlığı açısmdan farklı bulunmuşlardır. Mortalitenin yıllara göre dağılımı açısmdan ele alındığında ise, Kopelson skorunun anlamlı bir fark yaratmadığı, olumlu p53 immünhistokimyasal boyanma özelliklerinin ilk yıl içinde ölüm sayısını arttırmaya eğilimli olduğu, Kİ67 immünhistokimyasal boyanma indeksinin yüksek olduğu olgularda ise bu özelliğin ilk yıl içinde ölüm sayısıyla yakın ilişkili olduğu izlenmiştir. Abstract

ÖZET: Anatomik açıdan oldukça karışık ve girişimsel açıdan zorluklar içeren kafa tabanında, internal karotid arterin kafa tabanına giriş yeri ve katettiği kemik yapı olan temporal kemiğin petroz parçası, diğer çevre anatomik yapılar ile olan ilişkisi konu alınarak; var olan ve geliştirebileceğimiz cerrahi tekniklere veri olması planlandı ve çalışmaya alındı. Kuru temporal kemikteki yalın anatomik yapıdan sonra, plastik çamurlar ile temporal kemik üzerinde anatomik yapılar vizüalize olacak şekilde konuşlandırıldı. Ayrıca bir başka kafa tabanında foley sonda kullanılarak karotid kanal oluşturuldu ve opak madde ile internal karotid arterin intrakranial bölümünün trasesi gösterildi. Bu model yapılar, kadavra kafatası kullanılarak diseksiyon ile vizüalize edildi. Bilgisayarlı tomografide antropolojik taban hattı kullanılarak elde edilen kesitlerde basal yapıların en iyi görüldüğü kesitler kullanılarak belirlenen parametrelere göre ölçümler, EKG kağıdı, kampimetre, açı ölçer, cetvel ve negatoskop kullanılarak yapıldı. Elde edilen verilerin dökümleri yapılarak, birbirlerine göre değişkenlik gösterebileceği düşünülen bazı parametrelerin kombine grafik dökümleri yapıldı. Sonuçta bazı korelasyonlar gözlendi. Bunlardan en önemlisi ön fossa ve yapıları sabit kalmak üzere petroz kemiğin sagittal ve vertikal planda laterals açılanması ile orta fossanın lateral sının değişikliğine bağlı olarak santral ve perisantral anatomik yapıların dış tabulaya olan uzaklıklarının ve izdüşümlerinin değişmekte olduğu gözlendi. Yapılan çalışma ve alınan sonuçlar kaynaklardan elde edilen verilerle değerlendirildi; internal karotid arter ve temporal kemiğin petrozal parçasının embriyoloji, anatomi, radyoloji ve cerrahisi ile ilgili bilgiler sunuldu. 63

24. A Hydrogel-Based Multiplex Coculture Platform for Retinal Component Cells.

Abstract

There is growing interest in generating in vitro models of tissues and tissue-related diseases to mimic normal tissue organization and pathogenesis for different purposes. The retina is a highly complex multicellular tissue where the organization of the cellular components relative to each other is critical for retinal function. Many retinopathies arise due to the disruption of this order. In this study, we aimed to generate a coculture model of retina-derived cells, namely RPE and Müller cells, in multiplexed 3D hydrogels. Using methacrylated gelatin (GelMA)-based 3D hydrogels, we compared the behavior of RPE and Müller cells when they were cultured together. These patterned multiplex hydrogels containing cells were cultured for several days to reflect how cells would reorganize themselves in the presence of another cellular component derived from the same tissue. Here, we present a multicellular multiplex platform for the creation of cellular networks with cells of retinal tissue that can be easily adapted to create more complex tissue-like alternatives for large-scale tissue modeling and screening purposes. We also present an alternative method of coculture by generating spheroids from one of the components while keeping the other component free and motile in the hydrogel. The latter model predicts enhanced possibilities of cellular interactions by retarding the movement of one of the component cells.

25. Temperament and Voice Quality in Patients With Vocal Fold Nodules.

Abstract

Background: Vocal fold nodules are most common in women and patients with vocal fold nodules represent the largest group in voice clinics. The prevalence of vocal fold nodules is particularly high in professions where the voice is used on a regular basis. The quality of the voice is influenced by a number of factors, including temperament, stress, and emotional state. These factors can influence the physiological conditions of phonation. The objective of this study was to assess the acoustic parameters of voice in patients with vocal nodules in comparison to healthy controls, and to determine whether voice quality is influenced by emotional state and coping with stress., Methods: A total of 32 patients admitted to the ENT Department of the University Medical School with voice disorders between March and June 2007 constituted the study group. All patients were found to have a vocal nodule on physical and stroboscopic examination. The control group consisted of 30 healthy individuals who did not report any voice disorders. All subjects underwent voice recordings in the voice laboratory. Following the completion of the voice evaluation form, an aerodynamic assessment (a, s, and s/z-time), an index of vocal impairment, the Rosenbaum's Learned Resourcefulness Scale, and the Temperament and Characteristics Inventory (Temperament Evaluation of Memphis, Pisa, Paris, San Diego Autoquestionaire), all subjects underwent further assessment. Acoustic analysis was conducted using the CSL program in Multidimensional voice program analysis and the Vocal Assessment component of Dr. Speech., Results: The decrease in maximum phonation time in the study group was statistically significant. There were statistically significant differences in the parameters Mean Fundamental Frequence, Jitter, Relative Avarage Perturbation, Pitch Perturbation Quotient, Shimmer in dB, Shimmer, Amplitude Perturbation Quotient, Noise Hormonic Ratio, Soft Phonation Index from the Multidimensional voice program analysis, Jitter, Shimmer% from the voice assessment, and the perceptual rating (H, R, and B) from Dr. Speech's voice assessment analysis. The differences in the dimensions of anxious temperament and the examination of stress problem-solving strategies were significant between the study group and the control subjects. Differences in aerodynamic and acoustic parameters were found between disordered and healthy groups, as well as between individuals with different personalities. Overall, those with nodules were less likely to manage stress well than those without nodules., Conclusions: The study group and the control subjects showed significant differences in anxious temperament dimensions and stress problem-solving strategies. There were also differences in aerodynamic and acoustic parameters between the disordered and healthy groups, as well as between the groups with and without personality temperament differences. Overall, those with nodules were less likely to manage stress well than those without nodules. This finding indicates that stress management options are not effectively utilized in patients with vocal fold nodules. Abstract

The targeted delivery of pharmacologically active molecules, metabolites, and growth factors to the brain parenchyma has become one of the major challenges following the onset of neurodegeneration and pathological conditions. The therapeutic effect of active biomolecules is significantly impaired after systemic administration in the central nervous system (CNS) because of the blood-brain barrier (BBB). Therefore, the development of novel therapeutic approaches capable of overcoming these limitations is under discussion. Exosomes (Exo) are nano-sized vesicles of endosomal origin that have a high distribution rate in biofluids. Recent advances have introduced Exo as naturally suitable bio-shuttles for the delivery of neurotrophic factors to the brain parenchyma. In recent years, many researchers have attempted to regulate the delivery of Exo to target sites while reducing their removal from circulation. The encapsulation of Exo in natural and synthetic hydrogels offers a valuable strategy to address the limitations of Exo, maintaining their integrity and controlling their release at a desired site. Herein, we highlight the current and novel approaches related to the application of hydrogels for the encapsulation of Exo in the field of CNS tissue engineering.

Objective: The basis for current and future lung cancer immunotherapy depends on our knowledge of molecular mechanisms of interactions between tumor and immune system cells. Interactions that occur between different intratumoral populations of the same cells are important. In our study, we aimed to evaluate relationship between the clinical and prognostic features and T lymphocyte subgroups of patients with lung tumors after neoadjuvant treatment., Methods: A total of 72 patients were included in our study, including study group, 39 of whom received neoadjuvant chemotherapy. Clinical/radiological/pathological findings of patients and CD4/CD8 staining rates in peritumoral/intratumoral areas were recorded., Results: Our study revealed significantly lower intratumoral CD4 + T cell density and lower intratumoral CD4/CD8 ratio in primary tumor after neoadjuvant therapy (respectively, 0.012 and 0.016). Considering tumor types, when control-study groups were compared, inflammation was statistically significant only in adenocarcinoma subtype; intratumoral CD4/CD8 ratio was statistically significant only in squamous-cell carcinoma subtype (respectively, p = 0.0008 and p = 0.0139). When CD4 + T lymphocytes and CD8 + T lymphocytes and CD4/CD8 ratio were compared between control and study groups in low-stage patients according to clinical stages, only intratumoral CD4 + T lymphocyte values and intratumoral CD4/CD8 ratio were significant (respectively, p = 0.0291 ve p = 0.0154)., Conclusion: All cell types of innate and adaptive intratumoral immunity can affect lung cancer tissues simultaneously, and these interactions have a very complex structure. Abstract

Thymoquinone has antioxidant and anticancer effects. This study investigates the cytotoxic, genotoxic, and apoptotic effects of black seed and its active ingredient, thymoquinone on colorectal cancer cells. The antioxidant content of Black seed methanolic extracts (BSME) with different concentrations (50, 500 and 1000 μg/mL) were determined by the photometric methods. The reactive oxygen production (iROS) of BSME and thymoquinone on colorectal cancer cells (LoVo) and normal epithelial cells (CCD18Co) were analyzed by the fluorometric methods. A luminometric glutathione kit was employed to observe the changes in intracellular glutathione (GSH) levels. Cytotoxicity was determined by the ATP method, genotoxicity was determined by Comet Assay, and the apoptosis was identified by the Acridine Orange/Ethidium Bromide (AO/EB) double dye method. The cytotoxicity was increased by BSME and thymoquinone in LoVo cells in a dose-dependent manner (p<0.001). BSME and thymoquinone also increased iROS, and induced apoptosis and DNA damage (p<0.001). High doses of BSME and thymoquinone on cancer and healthy cells have cytotoxic, genotoxic and apoptotic effects with pro-oxidant effects. Abstract

Human induced pluripotent stem cells (hiPSCs) are a potential source of somatic cells for cell therapies due to their ability to self-renew and differentiate into various cells of the body. To date, the clinical application of hiPSCs has been limited due to safety issues. The present study aims to standardize the safety procedure of the derivation of GMP-compliant induced pluripotent stem cell (iPSC) lines from human fibroblasts. The hiPSC lines were generated using the nonintegrative Sendai virus method to incorporate Yamanaka reprogramming factors (OCT3/4, SOX2, KLF4 and c-MYC) into cells. A constant temperature was maintained during the cell culture, including all stages of the culture after transduction with Sendai virus. Pluripotency was proved in six independently generated hiPSC lines from adult female (47 years old) and male (57 years old) donors' derived fibroblasts via alkaline phosphatase live (ALP) staining, qPCR, and immunocytochemistry. The hiPSC lines showed a gradual decrease in the presence of the virus with each subsequent passage, and this reduction was specific to the hiPSC line. The frequency and probability of chromosomal aberrations in hiPSCs were dependent on both the iPSC clone identity and sex of the donor. In summary, the generation of hiPSC for clinical applications requires safety standards application (biosafety protocol, quality control of hiPSC lines, viral and genetic integrity screening) from the first stages of the clonal selection of hiPSC from the same donor.

Background: In this study, we aimed to compare the oxidative stress parameters, thiol-disulfide homeostasis, and plasma pro-inflammatory cytokines levels of patients with bipolar disorder (BD), BD patients' first-degree relatives (FDRs), and the healthy controls (HCs)., Methods: Thirty-five patients with BD, 35 FDRs of BD, and 35 healthy controls (HCs) were included. The individuals' ages varied from 28 to 58, and the groups were well-matched in terms of age and gender. The total thiol (TT), native thiol (NT), disulfide (DIS), total oxidant status (TOS), total antioxidant status (TAS), IL-1β, IL-6, and TNF-α concentrations were measured from serum samples. The oxidative stress index (OSI) was calculated using mathematical formulas., Results: TOS was significantly higher in both patients and FDRs than HCs (p < 0.01 for all pairwise comparisons). OSI, DIS, oxidized thiol, and the ratio of thiol oxidation-reduction levels were significantly higher in both patients with BD and FDRs than HCs (p < 0.01 for all pairwise comparisons). TAS, TT, NT, and reduced thiol levels were significantly lower in both patients with BD and FDRs than HCs (p < 0.01 for all pairwise comparisons). IL-1β, IL-6, and TNF-α were significantly higher in both patients and FDRs than HCs (p < 0.01 for all pairwise comparisons)., Limitations: Small sample size., Conclusions: Early diagnosis is important for treating of bipolar disorder. Abstract

The interaction of a multi-component system consisting of benzene-1,4-diyldimethanimine-bridged dimeric zinc-phthalocyanine groups (4OMPCZ) with calf thymus DNA (ct-DNA) was investigated using UV-Vis absorption, fluorescence emission spectroscopy methods, and viscosity measurements. The binding constant, K b , which is an important parameter to gain information about the binding mode, was found as 9.7 × 10 7 M -1 from the UV-Vis absorption studies. Another important spectrophotometric tool is competitive displacement assays with Ethidium bromide and Hoechst 33342. Through this experiment, a higher K SV value was obtained with Hoechst for the phthalocyanine derivative, 4OMPCZ, and the ct-DNA complex than with ethidium bromide. Additionally, molecular docking studies were conducted to calculate the theoretical binding constant and visualize the interactions of 4OMPCZ with a model DNA. According to docking results, although the interactions are mainly located in the major groove of the DNA helix, due to the wrapping, these interactions can also be extended to the minor groove of the DNA. Spectrophotometric, molecular docking, and viscosity studies revealed that the interaction of 4OMPCZ with DNA is likely to be via the major and minor grooves. The in vitro cytotoxic activity of 4OMPCZ was evaluated by MTT assay on human colon cancer cells (HT29) after 72 h of treatment. 4OMPCZ indicated significant cytotoxic activity when stimulated with UV light compared to the standard chemotherapy drugs, fluorouracil (5-FU), and cisplatin on HT29 colon cancer cells. Abstract

Dravet syndrome (DRVT) is a rare form of neurodevelopmental disorder with a high risk of sudden unexpected death in epilepsy (SUDEP), caused mainly (>80% cases) by mutations in the SCN1A gene, coding the Nav1.1 protein (alfa-subunit of voltage-sensitive sodium channel). Mutations in SCN1A are linked to heterogenous epileptic phenotypes of various types, severity, and patient prognosis. Here we generated iPSC lines from fibroblasts obtained from three individuals affected with DRVT carrying distinct mutations in the SCN1A gene (nonsense mutation p.Ser1516*, missense mutation p.Arg1596His, and splicing mutation c.2589+2dupT). The iPSC lines, generated with the non-integrative approach, retained the distinct SCN1A gene mutation of the donor fibroblasts and were characterized by confirming the expression of the pluripotency markers, the three-germ layer differentiation potential, the absence of exogenous vector expression, and a normal karyotype. The generated iPSC lines were used to establish ventral forebrain organoids, the most affected type of neurons in the pathology of DRVT. The DRVT organoid model will provide an additional resource for deciphering the pathology behind Nav1.1 haploinsufficiency and drug screening to remediate the functional deficits associated with the disease.

33. SARS-CoV-2 main protease targeting potent fluorescent inhibitors: Repurposing thioxanthones.

Abstract

The coronavirus disease, COVID-19, is the major focus of the whole world due to insufficient treatment options. It has spread all around the world and is responsible for the death of numerous human beings. The future consequences for the disease survivors are still unknown. Hence, all contributions to understand the disease and effectively inhibit the effects of the disease have great importance. In this study, different thioxanthone based molecules, which are known to be fluorescent compounds, were selectively chosen to study if they can inhibit the main protease of SARS-CoV-2 using various computational tools. All candidate ligands were optimized, molecular docking and adsorption, distribution, metabolism, excretion, and toxicity (ADMET) studies were conducted and subsequently, some were subjected to 100 ns molecular dynamics simulations in conjunction with the known antiviral drugs, favipiravir, and hydroxychloroquine. It was found that different functional groups containing thioxanthone based molecules are capable of different intermolecular interactions. Even though most of the studied ligands showed stable interactions with the main protease, para-oxygen-di-acetic acid functional group containing thioxanthone was found to be a more effective inhibitor due to the higher number of intermolecular interactions and higher stability during the simulations., (© TÜBİTAK.)

Abstract

The peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) is a well-known transcriptional coactivator involved in mitochondrial biogenesis. PGC-1α is implicated in the pathophysiology of many neurodegenerative disorders; therefore, a deep understanding of its functioning in the nervous system may lead to the development of new therapeutic strategies. The central nervous system (CNS)-specific isoforms of PGC-1α have been recently identified, and many functions of PGC-1α are assigned to the particular cell types of the central nervous system. In the mice CNS, deficiency of PGC-1α disturbed viability and functioning of interneurons and dopaminergic neurons, followed by alterations in inhibitory signaling and behavioral dysfunction. Furthermore, in the ALS rodent model, PGC-1α protects upper motoneurons from neurodegeneration. PGC-1α is engaged in the generation of neuromuscular junctions by lower motoneurons, protection of photoreceptors, and reduction in oxidative stress in sensory neurons. Furthermore, in the glial cells, PGC-1α is essential for the maturation and proliferation of astrocytes, myelination by oligodendrocytes, and mitophagy and autophagy of microglia. PGC-1α is also necessary for synaptogenesis in the developing brain and the generation and maintenance of synapses in postnatal life. This review provides an outlook of recent studies on the role of PGC-1α in various cells in the central nervous system.

35. Zooming their way through university: Which Big 5 traits facilitated students' adjustment to online courses during the COVID-19 pandemic.

Abstract

Many students struggled with the abrupt transition to online classes required by the COVID-19 pandemic. The present research used a self-determination theory framework to examine the role of the Big 5 personality traits in university students' adaptation to online learning, measured by their quality of motivation, subjective well-being, self-efficacy, online engagement, and online satisfaction. A longitudinal study was conducted across the 2020 fall semester and included a total of 350 university students who answered surveys in September and December. Results showed that conscientiousness and openness to experience were associated with higher self-efficacy and with different forms of autonomous motivation for online learning. Conscientiousness was related to identified motivation, whereas openness to experience was related to intrinsic motivation. In contrast, neuroticism was related to increases in controlled motivation. Only openness to experience was strongly related to engagement with online learning and higher levels of subjective well-being. These findings suggest that openness to experience may be an adaptive trait for students' transition to online classes during these times of uncertainty., (© 2021 Elsevier Ltd. All rights reserved.)

Objective: The aim of this study was to compare the efficacy of multiple antioxidant (Proxeed Plus (PP) with Carnitine, Selenium, Zinc, Coenzyme Q10, Vitamin C, Folic Acid, Vitamin B12) on local random skin flap healing with the hyperbaric oxygen (HBO) therapy., Methods: Fourty rats were equally divided into five groups (Control, PP, HBO, HBO + PP, PP + HBO + PP). Local random McFarlane skin flap was applied to all rats. Following the applications, evaluations were made biochemical (TAS, TOS, OSI, IL-1β, IL-6, TNF-α, TGF-β, VEGF) and histopathological parameters., Results: Necrosis percentage was found to be lower in the PP + HBO + PP group than all other groups whereas the necrosis percentages of PP and HBO groups were similar. Oxidative stress rates were significantly higher in the control group compared to the other groups whereas it was lower in the PP + HBO + PP group than all other groups. The inflammation parameters were the highest in the control group and the lowest in the PP + HBO + PP group. Growth factors were higher in the PP + HBO + PP group than all other groups. Epithelialization and wound healing were better in the HBO and PP groups than in the control group. The greatest healing, epithelialization and vascularization was seen in the PP + HBO + PP group. The histopathological findings in the PP + HBO + PP group were better in each inner region than in the other groups., Conclusion: Biochemical and histopathological parameters have shown that PP reduces ischemia and necrosis and increases oxygenation in flap healing by providing significant improvement thanks to the multiple molecular structures in its content., (Copyright © 2021 Tissue Viability Society. Abstract

Background: Although conventional chemotherapy is the most common method for cancer treatment, it has several side effects such as neuropathy, alopecia and cardiotoxicity. Since the drugs are given to body systemically, normal cells are also affected, just like cancer cells. However, in recent years, targeted drug delivery has been developed to overcome these drawbacks., Objective: The aim of this study was targeted co-delivery of doxorubicin (Dox) which is an anticancer agent and D-α-Tocopherol polyethylene glycol 1000 succinate (vitamin E TPGS or simply TPGS) to breast cancer cells. For this purpose, Magnetic Nanoparticles (MNPs) were synthesized and coated with Oleic Acid (OA). Coated nanoparticles were encapsulated in Poly Lactic-co-Glycolic Acid (PLGA) and TPGS polymers and loaded with Dox. The Nanoparticles (NPs) were characterized by Fourier Transform Infrared (FTIR) spectroscopy, zetapotential analysis, Dynamic Light Scattering (DLS) analysis, Thermal Gravimetric Analysis (TGA) and Scanning Electron Microscope (SEM) analysis., Results: The results showed that NPs were spherical, superparamagnetic and in the desired range for use in drug targeting. The targetability of NPs was confirmed. Moreover, TPGS and Dox loading was shown by TGA and FTIR analyses. NPs were internalized by cells and the cytotoxic effect of drug loaded NPs on sensitive (MCF-7) and drug-resistant (MCF-7/Dox) cells were examined. Abstract

Colour removal from industrial effluents, particularly from the textile industry, has become an important requirement as the adverse effects ofdyestuffs, such as toxicity, on the environment have been proven. Adsorption is a commonly used treatment method for colour removal. Although activated carbon is very effective for this purpose, a number of natural materials and waste materials, such as waste sludges generated from treatment systems, have been tested to reduce the cost of the process. In this paper, sludges arising from the operation of an electrocoagulation process that used stainless steel or aluminium electrodes were investigated as adsorbents for decolorization of reactive dyestuffs. Electrocoagulation waste sludges produced with the use of stainless steel electrodes provided higher than 90% or complete decolorization of Crimson HEXL, Yellow HE4R and RB5 dyestuffs. The sludge produced with aluminium electrodes yielded colour adsorption between 95% and 100% at a 1 g/L dose and pH 8.5-9.1 for two of the dyestuffs; the removal of RB5 did not exceed 60% up to a 4.76 g/L dose. FeCl3 coagulation and adsorption using freshly precipitated coagulation sludge resulted in poor colour removals of 10% for all three dyestuffs. The superior colour adsorption performance of electrocoagulation waste sludges was attributed to modification of the surface properties of the sludges during the electrocoagulation operation. A batch-wise kinetic study indicated that the adsorption of RB5 on to electrocoagulation waste sludges was well fitted by the pseudo-second-order kinetic model, suggesting the intra-particle diffusion process as the rate-limiting step of the adsorption process.

Abstract

Background: A foreign body in the urinary bladder can cause an acute abdomen in an adolescent., Case Report: A 14-year-old girl presented to the emergency department with severe suprapubic pain, dysuria, hematuria, and hypovolemic shock. An ultrasound examination revealed liquid in the abdomen. An X-ray study of the abdomen revealed an 8-cm-long metallic object located in the urinary bladder. The patient underwent an emergent laparotomy with the diagnosis of acute abdomen. At laparotomy, approximately 2000 cc of widely dispersed serohemorrhagic fluid was removed. Clearing of the abdomen exposed a nail; it had perforated the bladder and 5 cm of it was in the abdominal cavity. A vertical incision was made to the urinary bladder. A 10-penny caliber, 8.2-cm-long steel nail was gently removed and the perforation was closed primarily. The outcome was uneventful and the patient was discharged from the hospital on the seventh postoperative day., Conclusion: A foreign body can cause severe problems and should be kept in mind in the differential diagnosis of acute abdomen in an adolescent., (Copyright © 2011 Elsevier Inc. Abstract

Background: The aim of this study is to measure and compare the in vivo intraaneurysmal pressures of experimental lateral wall aneurysms, before and after onyx embolization. The data of this experiment will carry an important role in forming the scientific basis for the clinical endovascular applications., Materials and Methods: Five experimental lateral wall aneurysms were created by microsurgical techniques in 5 New Zealand rabbits' right common carotid arteries. Onyx embolization was applied to the aneurysms. Intraaneurysmal dome pressure and parent artery measurements before and after the procedure were recorded., Results: The mean arterial pressure recording in parent artery was 69.2 +/- 2.588 mm Hg under anesthesia. Mean heart rate was 131 beats per minute. The values were in physiologic limits. Meanwhile, aneurysm intradomal pressure recording showed a mean value of 59.2 +/- 5.069 mm Hg. Although there was incomplete occlusion of the aneurysm, intradomal mean pressure was recorded to be 24.4 +/- 8.876 mm Hg. After complete occlusion by onyx, mean intradomal aneurysm pressure was found to be 1.8 +/- 0.836 mm Hg., Discussion: This study is the first study reporting on intraaneurysmal pressure measurements before and after onyx embolization. The results in this experiment tend to show the adequate intraaneurysmal pressure control of onyx. This is important in the stabilization of the aneurysm to prevent rupture and rerupture. When compared with the findings of GDC coil, onyx embolization seems to be superior in intraaneurysmal pressure control., Conclusion: In this study, it has been shown that onyx embolization decreases the intradomal aneurysmal pressure effectively.

Abstract

Skin lesions are common in athletes. Athletic activities may lead to new skin lesions or aggravate existing ones. We aimed to determine the effects of sport type and participation length on the occurrence of sports-related dermatoses and to identify the localization characteristics of these lesions. A total of 121 licensed athletes (42 swimmers, 23 handball players, 33 soccer players and 23 wrestlers) and 121 sedentary controls were included in the study. A consultant dermatologist examined all subjects. Lesion types, duration, and localization characteristics were noted. The lesions were categorized as viral, bacterial, traumatic, and non-traumatic. Traumatic lesions were frequently seen in soccer players and wrestlers; fungal infections were more commonly seen in swimmers and in soccer players. Lesion types and localizations varied by sport type. There were no significant relationships between sport type and the incidence of viral and bacterial lesions. The results suggest that athletic activity seems to be a predisposing factor, especially for fungal infections and acute or chronic traumatic lesions. Thus, regular dermatological screening of athletes is critical for rapid identification and treatment of dermatoses disrupting sport performance.

42. Solid waste management practices and review of recovery and recycling operations in Turkey.

Abstract

This paper provides a general overview of solid waste data and management practices employed in Turkey during the last decade. Municipal solid waste statistics and management practices including waste recovery and recycling initiatives have been evaluated. Detailed data on solid waste management practices including collection, recovery and disposal, together with the results of cost analyses, have been presented. Based on these evaluations basic cost estimations on collection and sorting of recyclable solid waste in Turkey have been provided. The results indicate that the household solid waste generation in Turkey, per capita, is around 0.6 kg/year, whereas municipal solid waste generation is close to 1 kg/year. The major constituents of municipal solid waste are organic in nature and approximately 1/4 of municipal solid waste is recyclable. Separate collection programmes for recyclable household waste by more than 60 municipalities, continuing in excess of 3 years, demonstrate solid evidence for public acceptance and continuing support from the citizens. Opinion polls indicate that more than 80% of the population in the project regions is ready and willing to participate in separate collection programmes. The analysis of output data of the Material Recovery Facilities shows that, although paper, including cardboard, is the main constituent, the composition of recyclable waste varies strongly by the source or the type of collection point.

Abstract

Thallium-201 uptake patterns in the orbits are not widely known. Diffuse, bilateral orbital localization of TI-201 chloride may be an incidental finding because of orbital muscle or other soft tissue uptake. Differences in orbital blood flow and soft tissue mass may be the cause of the varying uptake intensities in most cases. Focal uptakes, even when bilateral, may indicate specific pathology and should be investigated. Sample cases are presented.
