CLAAS-3, the third edition of the Cloud property dAtAset using SEVIRI (Spinning Enhanced Visible and InfraRed Imager), was released in December 2022. It is based on observations from SEVIRI, on board geostationary satellites Meteosat-8, 9, 10 and 11, which are operated by the European Organisation for the Exploitation of Meteorological Satellites (EUMETSAT). CLAAS-3 was produced and released by the EUMETSAT Satellite Application Facility on Climate Monitoring (CM SAF), which aims to provide high-quality satellite-based data records suitable for climate monitoring applications. Compared to previous CLAAS releases, CLAAS-3 is expanded in terms of both temporal extent and cloud properties included, and it is based on partly updated retrieval algorithms. The available data span the period from 2004 to present, covering Europe; Africa; the Atlantic Ocean; and parts of South America, the Middle East and the Indian Ocean. They include cloud fractional coverage, cloud-top height, phase (liquid or ice) and optical and microphysical properties (water path, optical thickness, effective radius and droplet number concentration), from instantaneous data (every 15 min) to monthly averages. In this study we present an extensive evaluation of CLAAS-3 cloud properties, based on independent reference data sets. These include satellite-based retrievals from active and passive sensors, ground-based observations and in situ measurements from flight campaigns. Overall results show very good agreement, with small biases attributable to different sensor characteristics, retrieval/sampling approaches and viewing/illumination conditions. These findings demonstrate the fitness of CLAAS-3 to support the intended applications, which include evaluation of climate models, cloud characterisation and process studies focusing especially on the diurnal cycle and cloud filtering for other applications. This paper presents the third edition of The Satellite Application Facility on Climate Monitoring's (CM SAF) cloud, albedo, and surface radiation dataset from advanced very-high-resolution radiometer (AVHRR) data, CLARA-A3. The content of earlier CLARA editions, namely cloud, surface albedo, and surface radiation products, has been extended with two additional surface albedo products (blue- and white-sky albedo), three additional surface radiation products (net shortwave and longwave radiation, and surface radiation budget), and two top of atmosphere radiation budget products (reflected solar flux and outgoing longwave radiation). The record length is extended to 42 years (1979–2020) by also incorporating results from the first version of the advanced very high resolution radiometer imager (AVHRR/1). A continuous extension of the climate data record (CDR) has also been implemented by processing an interim climate data record (ICDR) based on the same set of algorithms but with slightly changed ancillary input data. All products are briefly described together with validation results and intercomparisons with currently existing similar CDRs. The extension of the product portfolio and the temporal coverage of the data record, together with product improvements, is expected to enlarge the potential of using CLARA-A3 for climate change studies and, in particular, studies of potential feedback effects between clouds, surface albedo, and radiation. Understanding the coupling of clouds to large-scale circulation is one of the grand challenges for the global climate research community. In this context, realistically modelling the vertical structure of cloud radiative heating (CRH) and/or cooling in Earth system models is a key premise to understand this coupling. Here, we evaluate CRH in two versions of the European Community Earth System Model (EC-Earth) using retrievals derived from the combined radar and lidar data from the CloudSat and Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observation (CALIPSO) satellites. One model version is also used with two different horizontal resolutions. Our study evaluates large-scale intraseasonal variability in the vertical structure of CRH and cloud properties and investigates the changes in CRH during different phases of the El Niño–Southern Oscillation (ENSO), a process that dominates the interannual climate variability in the tropics. EC-Earth generally captures both the intraseasonal and meridional pattern of variability in CRH over the convectively active and stratocumulus regions and the CRH during the positive and negative phases of ENSO. However, two key differences between model simulations and satellite retrievals emerge. First, the magnitude of CRH, in the upper troposphere, over the convectively active zones is up to twice as large in the models compared to the satellite data. Further dissection of net CRH into its shortwave and longwave components reveals noticeable differences in their vertical structure. The shortwave component of the radiative heating is overestimated by all model versions in the lowermost troposphere and underestimated in the middle troposphere. These over- and underestimates of shortwave heating are partly compensated by an overestimate of longwave cooling in the lowermost troposphere and heating in the middle troposphere. The biases in CRH can be traced back to disagreement in cloud amount and cloud water content. There is no noticeable improvement of CRH by increasing the horizontal resolution in the model alone. Our findings highlight the importance of evaluating models with satellite observations that resolve the vertical structure of clouds and cloud properties.

Fetal and adult hematopoietic stem and progenitor cells (HSPCs) are characterized by distinct redox homeostasis that may influence their differential cellular behavior in normal and malignant hematopoiesis. In this work, we have applied a quantitative mass spectrometry-based redox proteomic approach to comprehensively describe reversible cysteine modifications in primary mouse fetal and adult HSPCs. We defined the redox state of 4,438 cysteines in fetal and adult HSPCs and demonstrated a higher susceptibility to oxidation of protein thiols in fetal HSPCs. Our data identified ontogenic changes to oxidation state of thiols in proteins with a pronounced role in metabolism and protein homeostasis. Additional redox proteomic analysis identified oxidation changes to thiols acting in mitochondrial respiration as well as protein homeostasis to be triggered during onset of MLL-ENL leukemogenesis in fetal HSPCs. Our data has demonstrated that redox signaling contributes to the regulation of fundamental processes of developmental hematopoiesis and has pinpointed potential targetable redox-sensitive proteins in in utero-initiated MLL-rearranged leukemia.

Antisecretory factor (AF) is a protein which regulates fluid transport in the intestine and other organs. Extensive malting of wheat releases certain chemicals which induce AF and inhibit enterotoxic diarrhoea. The aim of this study was to identify AF-inducing substances in wheat malt. The active fractions of the malt leachate contained guaiacol, ferulic acid, and vanillic acid. These three 2-methyl-catechols gave >50% inhibition of cholera toxin-induced secretion in the gut. In contrast, fully methylated catechol had the opposite action; that is, they were secretory rather than antisecretory. Guaiacol and ferulic acid were further shown to induce AF in blood. Since the 2-methoxyphenol structure is present in substances binding to the vanilloid receptor, TRPV1, the specific blocker of this receptor—capsazepine—was tested. This substance exerted >50% inhibition of cholera secretion. Thus 2-methyl-catechols in wheat malt induce AF, and probably exert their effect via the TRPV1 receptor.

The geometries of a catchment constitute the basis for distributed physically based numerical modeling of different geoscientific disciplines. In this paper results from ground-penetrating radar (GPR) measurements, in terms of a 3-D model of total sediment thickness and active layer thickness in a periglacial catchment in western Greenland, are presented. Using the topography, the thickness and distribution of sediments are calculated. Vegetation classification and GPR measurements are used to scale active layer thickness from local measurements to catchment-scale models. Annual maximum active layer thickness varies from 0.3 m in wetlands to 2.0 m in barren areas and areas of exposed bedrock. Maximum sediment thickness is estimated to be 12.3 m in the major valleys of the catchment. A method to correlate surface vegetation with active layer thickness is also presented. By using relatively simple methods, such as probing and vegetation classification, it is possible to upscale local point measurements to catchment-scale models, in areas where the upper subsurface is relatively homogeneous. The resulting spatial model of active layer thickness can be used in combination with the sediment model as a geometrical input to further studies of subsurface mass transport and hydrological flow paths in the periglacial catchment through numerical modeling. Global warming is expected to be most pronounced in the Arctic where permafrost thaw and release of old carbon may provide an important feedback mechanism to the climate system. To better understand and predict climate effects and feedbacks on the cycling of elements within and between ecosystems in northern latitude landscapes, a thorough understanding of the processes related to transport and cycling of elements is required. A fundamental requirement to reach a better process understanding is to have access to high-quality empirical data on chemical concentrations and biotic properties for a wide range of ecosystem domains and functional units (abiotic and biotic pools). The aim of this study is therefore to make one of the most extensive field data sets from a periglacial catchment readily available that can be used both to describe present-day periglacial processes and to improve predictions of the future. Here we present the sampling and analytical methods, field and laboratory equipment and the resulting biogeochemical data from a state-of-the-art whole-ecosystem investigation of the terrestrial and aquatic parts of a lake catchment in the Kangerlussuaq region, West Greenland. This data set allows for the calculation of whole-ecosystem mass balance budgets for a long list of elements, including carbon, nutrients and major and trace metals. Clouds forming during the summer monsoon over the Indian subcontinent affect its evolution through their radiative impact as well as the release of latent heat. While the latter is previously studied to some extent, comparatively little is known about the radiative impact of different cloud types and the vertical structure of their radiative heating/cooling effects. Therefore, the main aim of this study is to partly fill this knowledge gap by investigating and documenting the vertical distributions of the different cloud types associated with the Indian monsoon and their radiative heating/cooling using the active radar and lidar sensors onboard CloudSat and CALIPSO. The intraseasonal evolution of clouds from May to October is also investigated to understand pre-to-post monsoon transitioning of their radiative heating/cooling effects. The vertical structure of cloud radiative heating (CRH) follows the northward migration and retreat of the monsoon from May to October. Throughout this time period, stratiform clouds radiatively warm the middle troposphere and cool the upper troposphere by more than ±0.2 K day−1 (after weighing by cloud fraction), with the largest impacts observed in June, July and August. During these months, the fraction of high thin cloud remains high in the tropical tropopause layer (TTL). Deep convective towers cause considerable radiative warming in the middle and upper troposphere, but strongly cool the base and inside of the TTL. This cooling is stronger during active (−1.23 K day−1) monsoon periods compared to break periods (−0.36 K day−1). The contrasting radiative warming effect of high clouds in the TTL is twice as large during active periods than in break periods. These results highlight the increasing importance of CRH with altitude, especially in the TTL. Stratiform (made up of alto- and nimbostratus clouds) and deep convection clouds radiatively cool the surface by approximately −100 and −400 W m−2 respectively while warming the atmosphere radiatively by about 40 to 150 W m−2. While the cooling at the surface induced by deep convection and stratiform clouds is largest during active periods of monsoon, the importance of stratiform clouds further increases during break periods. The contrasting CREs (cloud radiative effects) in the atmosphere and at surface, and during active and break periods, should have direct implications for the monsoonal circulation.

Few hydrological studies have been conducted in Greenland, other than on glacial hydrology associated with the ice sheet. Understanding permafrost hydrology and hydroclimatic change and variability, however, provides key information for understanding climate change effects and feedbacks in the Arctic landscape. This paper presents a new, extensive, and detailed hydrological and meteorological open access data set, with high temporal resolution from a 1.56 km2 permafrost catchment, with a lake underlain by a through-talik close to the ice sheet in the Kangerlussuaq region, western Greenland. The paper describes the hydrological site investigations and utilized equipment, as well as the data collection and processing. The investigations were performed between 2010 and 2013. The high spatial resolution, within the investigated area, of the data set makes it highly suitable for various detailed hydrological and ecological studies on catchment scale. Gonorrhoea is a sexually transmitted infection with major public health implications and Neisseria gonorrhoeae has developed resistance to all antimicrobials introduced for treatment. Enhanced surveillance of antimicrobial resistance in N. gonorrhoeae is crucial globally. This is the first internationally reported antimicrobial resistance data for N. gonorrhoeae from Estonia (44 isolates cultured in 2009–2013). A high prevalence of resistance was observed for azithromycin, ciprofloxacin and tetracycline. One and two isolates with resistance and decreased susceptibility to the last remaining first-line treatment option ceftriaxone, respectively, were identified. It is crucial to implement surveillance of gonococcal antimicrobial resistance (ideally also treatment failures) in Estonia.

A method for detailed evaluation of a new satellite-derived global 28 yr cloud and radiation climatology (Climate Monitoring SAF Clouds, Albedo and Radiation from AVHRR data, named CLARA-A1) from polar-orbiting NOAA and Metop satellites is presented. The method combines 1 km and 5 km resolution cloud datasets from the CALIPSO-CALIOP (Cloud-Aerosol Lidar and Infrared Pathfinder Satellite Observation – Cloud-Aerosol Lidar with Orthogonal Polarization) cloud lidar for estimating cloud detection limitations and the accuracy of cloud top height estimations. Cloud detection is shown to work efficiently for clouds with optical thicknesses above 0.30 except for at twilight conditions when this value increases to 0.45. Some misclassifications of cloud-free surfaces during daytime were revealed for semi-arid land areas in the sub-tropical and tropical regions leading to up to 20% overestimated cloud amounts. In addition, a substantial fraction (at least 20–30%) of all clouds remains undetected in the polar regions during the polar winter season due to the lack of or an inverted temperature contrast between Earth surfaces and clouds. Subsequent cloud top height evaluation took into account the derived information about the cloud detection limits. It was shown that this has fundamental importance for the achieved results. An overall bias of −274 m was achieved compared to a bias of −2762 m when no measures were taken to compensate for cloud detection limitations. Despite this improvement it was concluded that high-level clouds still suffer from substantial height underestimations, while the opposite is true for low-level (boundary layer) clouds. The validation method and the specifically collected satellite dataset with optimal matching in time and space are suggested for a wider use in the future for evaluation of other cloud retrieval methods based on passive satellite imagery.

This paper investigates two options for integration of biomass-based olefin production with a fossil-based steam cracker plant at the heart of a chemical cluster. The work was conducted in the form of a case study considering the possible future partial replacement of a fraction of the cracker olefins with approx. 220 kt/y of biomass-based olefins (ethylene, propylene, and butylene) (approx. 25 % of total capacity) produced via gasification, methanol synthesis, and the methanol-to-olefins (MTO) process. Two options were compared with base case operation with fossil-only feedstock: (i) purchase of methanol produced off-site, and (ii) on- site methanol production. In both cases, the MTO section was assumed to be located at the cracker site, making use of existing olefin separation equipment. Consequences of such partial feedstock substitution for the steam, fuel gas, and electric power balances of the cracker plant were investigated. Potentials for generation of steam and electric power were estimated by assuming integration with a heat recovery steam cycle. Greenhouse gas (GHG) emission balances of the proposed options were estimated by applying a system boundary expansion approach. The GHG emission reduction potentials are shown to be between 50 % and 70 %, compared with the base case. The reduction potential depends on the choice of reference grid electricity generation technology but the major contribution comes from the introduction of renewable feedstock.

Bread-making quality of wheat flour is influenced by wheat storage proteins. The percentage of sodium dodecyl sulphate (SDS)-unextracted polymeric proteins in total polymeric proteins (% UPP), as one important factors determining bread-making quality, is largely influenced by the individual and interactive effect of genetical background and environmental conditions. Four spring wheat cultivars were grown in green house. Two of the cultivars have high molecular weight glutenin subunits 5+10 while the other two have 2+12. Four different nitrogen regimes and two different temperature levels (distinguished as low and high temperature) were used. The results highlighted the importance of interactive influence of cultivars, nitrogen regimes and temperature levels for creating differences in the amount of % UPP during grain maturation period (GMP) and at maturity. The shift to end up with either high or low % UPP at maturity happened early during GMP (around 12 days after anthesis). Influences of cultivars on % UPP was found early during the GMP while influences of temperature and nitrogen regimes were seen much later during the GMP. The results will help in understanding the relationship between quality parameters of wheat flour and how these are influenced by different environmental and genetic factors.
