The family Hypoxylaceae has a cosmopolitan distribution with greater diversity in tropical regions, its growth habit is saprotrophic, endophytic and potentially phytopathogenic. From the revision of herbarium specimens and field collections from the Yucatan Peninsula, two new species were described: Annulohypoxylon fusisporum, characterized by having fusiform spores and Hypoxylon xmatkuilense which is distinguished by having stromata vinaceous and dark brown KOH-extractable pigments. The species are described based on morphological characters and Bayesian Inference analyses of four molecular markers (ITS, LSU, RPB2 and TUB2). In addition, one new record from Mexico is presented: Hypoxylon bellicolor. The presence of Daldinia eschscholtzii, H. lenormandii, H. lividipigmentum and Entonaema liquescens is confirmed with molecular data.

Abstract Background Latinos are more likely than non-Latino Whites to develop dementia and be prescribed antipsychotics for dementia-related behavioral symptoms. Antipsychotics have significant risks yet are often overprescribed. Our understanding of how Latino caregivers of Latino older adults living with dementia perceive and address behavioral issues is limited, impeding our ability to address the root causes of antipsychotic overprescribing. Methods We interviewed Latino older adults’ caregivers and community-based organization workers serving older adults with cognitive impairment (key informants), focusing on the management of behavioral symptoms and experiences with health services. Results We interviewed 8 caregivers and 2 key informants. Caregivers were the spouses, children, or grandchildren of the older adult living with cognitive impairment; their ages ranged from 30 to 95. We identified three categories of how caregivers learned about, managed, and coped with behavioral symptoms: caregivers often faced shortcomings with dementia care in the medical system, receiving limited guidance and support; caregivers found community organizations and senior day centers to be lifelines, as they received relevant, timely advice and support, caregivers often devised their own creative strategies to manage behavioral symptoms. Conclusion In-depth interviews suggest that the healthcare system is failing to provide support for behavioral symptoms from dementia; caregivers of Latino older adults rely on community organizations instead.

In this work, we study the feeling of membership in undergraduate students who interact with their higher education institution through a virtual environment. We explore the "Centro de Investigaciones en Diseño Industrial" (CIDI) case, in which a virtual environment was built using the Roblox platform to enhance the online experience that students were having during the COVID-19 pandemic. Through the application of the "Psychological Sense of School Membership" scale, we show that it is possible to evoke a sense of belonging in a physical space with a virtual representation. We achieved this by virtually replicating the physicality of the original CIDI and by promoting social behaviors using three modalities of social interaction.

Abnormal placental invasion is a condition related to multiple conditions including previous elective cesarean delivery and its increased frequency. For the obstetrician, it is also increasingly common to face this clinical entity, which has a high morbidity due to organ injury both directly and indirectly during the surgical procedure and not being attended by a highly qualified medical staff. In this paper, we focussed mainly on the surgical approach and its multidisciplinary management and a clinical case on placental percretism attended at the HMPMPS using a complementary diagnostic method (CT angiography) to plan the vascular approach during surgery.

The Anderson localization of light in one-dimensional disordered photonic superlattices is theoretically studied. The system is considered to be made of alternating dispersive and nondispersive layers of different randomthickness. Dispersive slabs of the heterostructure are characterized by Drude-like frequency-dependent electric permittivities and magnetic permeabilities. Numerical results for the localization length are obtained via an analytical model, only valid in the case of weak disorder, and also through its general definition involving the transmissivity of the multilayered system. Anomalous λ4- and λ-4-dependencies of the localization length in positive-negative disordered photonic superlattices are obtained, in certain cases, in the long and short wavelength limits, respectively. Antecedentes: El síndrome postimplantación (SPI) es una complicación aguda que puede surgir tras colocar endoprótesis para reparación de aneurismas aórticos abdominales (AAA). Su incidencia oscila entre 11.2-60%. Objetivo: Describir frecuencia de PIS y su asociación con factores de riesgo en pacientes sometidos a reparación endovascular (EVAR) de AAA. Método: Estudio observacional, analítico y ambispectivo. Se analizaron los registros clínicos de pacientes con AAA sometidos a EVAR; teniendo seguimiento a corto plazo (72 horas) y mediano plazo (un mes). Resultados: Se incluyeron 50 pacientes, 43 varones y 7 mujeres (edad media 74 ± 8 años). El 97.9% presentó aneurismas fusiformes (diámetro medio 66.2 ± 17.5 mm). Se empleó dacrón en 54.2% (n = 26/50) y politetrafluoroetileno en el resto de las prótesis. Siete casos (14.6%) presentaron PIS durante las primeras 24 horas. Dos pacientes fallecieron (sin desarrollar SPI). Se observó asociación entre dislipidemia y el desarrollo de PIS (odds ratio [OR]: 9.500; IC95%: 1.416-63.724; p = 0.033). Conclusión: La frecuencia de SPI fue similar a otras series y en el presente estudio encontramos que la dislipidemia incrementa el riesgo de SPI (OR: 9.5).

Abstract Ferroelectrics are a class of polar and switchable functional materials with diverse applications, from microelectronics to energy conversion. Computational searches for new ferroelectric materials have been constrained by accurate prediction of the polarization and switchability with electric field, properties that, in principle, require a comparison with a nonpolar phase whose atomic-scale unit cell is continuously deformable from the polar ground state. For most polar materials, such a higher-symmetry nonpolar phase does not exist or is unknown. Here, we introduce a general high-throughput workflow that screens polar materials as potential ferroelectrics. We demonstrate our workflow on 1978 polar structures in the Materials Project database, for which we automatically generate a nonpolar reference structure using pseudosymmetries, and then compute the polarization difference and energy barrier between polar and nonpolar phases, comparing the predicted values to known ferroelectrics. Focusing on a subset of 182 potential ferroelectrics, we implement a systematic ranking strategy that prioritizes candidates with large polarization and small polar-nonpolar energy differences. To assess stability and synthesizability, we combine information including the computed formation energy above the convex hull, the Inorganic Crystal Structure Database id number, a previously reported machine learning-based synthesizability score, and ab initio phonon band structures. To distinguish between previously reported ferroelectrics, materials known for alternative applications, and lesser-known materials, we combine this ranking with a survey of the existing literature on these candidates through Google Scholar and Scopus databases, revealing ~130 promising materials uninvestigated as ferroelectric. Our workflow and large-scale high-throughput screening lays the groundwork for the discovery of novel ferroelectrics, revealing numerous candidates materials for future experimental and theoretical endeavors.

Background: As the COVID-19 pandemic persists, infections continue to surge globally. Presently, the most effective strategies to curb the disease and prevent outbreaks involve fostering immunity, promptly identifying positive cases, and ensuring their timely isolation. Notably, there are instances where the SARS-CoV-2 virus remains infectious even after patients have completed their quarantine. Objective: Understanding viral persistence post-quarantine is crucial as it could account for localized infection outbreaks. Therefore, studying and documenting such instances is vital for shaping future public health policies. Design: This study delves into a unique case of SARS-CoV-2 persistence in a 60-year-old female healthcare worker with a medical history of hypertension and hypothyroidism. The research spans 55 days, marking the duration between her initial and subsequent diagnosis during Chile's first COVID-19 wave, with the analysis conducted using RT-qPCR. Results: Genomic sequencing-based phylogenetic analysis revealed that the SARS-CoV-2 detected in both Nasopharyngeal swab samples (NPSs) was consistent with the 20B clade of the Nextstrain classification, even after a 55-day interval. Conclusion: This research underscores the need for heightened vigilance concerning cases of viral persistence. Such instances, albeit rare, might be pivotal in understanding sporadic infection outbreaks that occur post-quarantine.

The first case of cerebral venous thrombosis, described in the past century, was identified in obstetric patients in 1825. The clinical manifestations represent a real diagnostic challenge due to the few recorded cases, the wide variety of possible clinical symptoms, and the diversity of medical conditions that cause it. In the past century, the etiology was mainly associated with septic processes; however, due to the widespread use of antibiotics, this cause has been considerably reduced. In the context of the puerperium, several conditions make this group of patients more vulnerable, such as dehydration, hypercoagulable state, iron deficiency, puerperal sepsis, and preeclampsia, the latter is particularly known for an endothelial lesion with different degrees of associated severity, according to the clinical phenotype present. This review aims to highlight the most relevant aspects of cerebral thrombosis in the context of a puerperal patient. It will also present clinical cases reports treated successfully in this health unit.

The skin of bony fish is the first physical barrier and is responsible for maintaining the integrity of the fish. Lesions make the skin vulnerable to potential infection by pathogens present in the aquatic environment. In this way, wound repair has barely been studied in gilthead sea bream. Thus, this study investigated the modulation of peripheral neuro-endocrine and tissue repair markers at the transcriptional level in the skin of teleost fish subjected to mechanical damage above or below the lateral line (dorsal and ventral lesions, respectively). Samples were evaluated using RT-qPCR at 2-, 4-, and 20-days post-injury. Fish with a ventral lesion presented a trend of progressive increase in the expressions of corticotropin-releasing hormone (crh), pro-opiomelanocortin-A (pomca), proenkephalin-B (penkb), cholecystokinin (cck), oxytocin (oxt), angiotensinogen (agt), and (less pronounced) somatostatin-1B (sst1b). By contrast, fish with a dorsal lesion registered no significant increase or biological trend for the genes evaluated at the different sampling times. Collectively, the results show a rapid and more robust response of neuro-endocrine and tissue repair markers in the injuries below than above the lateral line, which could be attributable to their proximity to vital organs.

Vaccination is the most effective tool for paratuberculosis control. Currently, available vaccines prevent the progression of clinical disease in most animals but do not fully protect them against infection and induce the formation of an injection site granuloma. The precise mechanisms that operate in response to vaccination and granuloma development, as well as the effect that adjuvants could trigger, have not been fully investigated. Therefore, this study aimed to investigate the injection site granulomas induced by two inactivated paratuberculosis vaccines, which differ in the adjuvant employed. Two groups of 45-day-old lambs were immunized with two commercially available vaccines—one (n = 4) with Gudair® and the other (n = 4) with Silirum®. A third group (n = 4) was not vaccinated and served as control. The peripheral humoral response was assessed throughout the study by a commercial anti-Mycobacterium avium subspecies paratuberculosis (Map) antibody indirect ELISA, and the cellular immune response was assessed similarly by the IFN-γ release and comparative intradermal tests. The injection site granulomas were measured during the experiment and sampled at 75 days post-vaccination (dpv) when the animals were euthanized. The tissue damage, antigen and adjuvant distribution, and the presence and amount of immune cells were then determined and assessed by immunohistochemical methods. Antibodies against Map antigens; a general macrophage marker (Iba1), M1 (iNOS), and M2 (CD204) macrophages; T (CD3), B (CD20), and γδ T lymphocytes, proteins MHC-II and NRAMP1, and cytokines IL-4, IL-10, TNF, and IFN-γ were employed. Silirum® elicited a stronger peripheral cellular immune response than Gudair®, while the latter induced larger granulomas and more tissue damage at the site of injection. Additionally, adjuvant and Map antigen distribution throughout the granulomatous inflammatory infiltrate, as well as the NRAMP1 cell expression, which is linked to antigen phagocytosis, were highly irregular. In Silirum® induced granulomas, a higher number of MHC-II and TNF-expressing cells and a lower number of M2 macrophages suggested an improved antigen presentation, which could be due to the better antigen distribution and reduced tissue damage induced by this vaccine.

Introducción: Los cambios físicos, neuroendocrinos, sociales y psicológicos en el puerperio favorecen la depresión posparto, condición que impacta en la relación madre e hijo, incluyendo la lactancia materna. Objetivo: Determinar la probable depresión posparto como factor asociado al abandono de la lactancia materna exclusiva. Método: Diseño transversal analítico en mujeres puérperas; se integraron dos grupos, mujeres con probable depresión posparto y sin probable depresión posparto, determinado por la Escala de depresión posparto de Edimburgo. El tamaño de la muestra fue de 63 mujeres con depresión y 153 sin depresión. La lactancia materna exclusiva se definió como la alimentación de la madre, sin ningún otro suplemento sólido o líquido como el agua, al menos durante un mes de vida del recién nacido. El plan de análisis estadístico incluyó prueba de chi cuadrada, razón de momios e intervalo de confianza para razón de momios. Resultados: Por cada 5,10 (IC95%: 2,71-9,52) madres con probable depresión posparto que no tienen lactancia materna exclusiva existe una madre sin probable depresión posparto que tampoco tiene lactancia materna exclusiva. En el primer grupo la prevalencia de no lactancia es del 65,1% y en el segundo grupo del 26,8% (p = 0,013). Conclusión: Se puede afirmar que la probable depresión posparto es un factor asociado a la suspensión de la lactancia materna exclusiva en el primer mes posterior al nacimiento.

Piscirickettsia salmonis is the most important health problem facing Chilean Aquaculture. Previous reports suggest that P. salmonis can survive in salmonid macrophages by interfering with the host immune response. However, the relevant aspects of the molecular pathogenesis of P. salmonis have been poorly characterized. In this work, we evaluated the transcriptomic changes in macrophage-like cell line SHK-1 infected with P. salmonis at 24- and 48-hours post-infection (hpi) and generated network models of the macrophage response to the infection using co-expression analysis and regulatory transcription factor-target gene information. Transcriptomic analysis showed that 635 genes were differentially expressed after 24- and/or 48-hpi. The pattern of expression of these genes was analyzed by weighted co-expression network analysis (WGCNA), which classified genes into 4 modules of expression, comprising early responses to the bacterium. Induced genes included genes involved in metabolism and cell differentiation, intracellular transportation, and cytoskeleton reorganization, while repressed genes included genes involved in extracellular matrix organization and RNA metabolism. To understand how these expression changes are orchestrated and to pinpoint relevant transcription factors (TFs) controlling the response, we established a curated database of TF-target gene regulatory interactions in Salmo salar, SalSaDB. Using this resource, together with co-expression module data, we generated infection context-specific networks that were analyzed to determine highly connected TF nodes. We found that the most connected TF of the 24- and 48-hpi response networks is KLF17, an ortholog of the KLF4 TF involved in the polarization of macrophages to an M2-phenotype in mammals. Interestingly, while KLF17 is induced by P. salmonis infection, other TFs, such as NOTCH3 and NFATC1, whose orthologs in mammals are related to M1-like macrophages, are repressed. In sum, our results suggest the induction of early regulatory events associated with an M2-like phenotype of macrophages that drives effectors related to the lysosome, RNA metabolism, cytoskeleton organization, and extracellular matrix remodeling. Moreover, the M1-like response seems delayed in generating an effective response, suggesting a polarization towards M2-like macrophages that allows the survival of P. salmonis. This work also contributes to SalSaDB, a curated database of TF-target gene interactions that is freely available for the Atlantic salmon community.

The COVID-19 pandemic continues to affect several countries. One of the best ways to control its spread is the timely identification of infected patients for isolation and quarantine. While an episode of infection lasts an average of 8–10 days from the onset of symptoms, there is literature describing long-lasting viral persistence events. Here, we report a case of persistence of SARS-CoV-2 for 386 days in a health worker from Santiago de Chile. Our study could be one of the longest reported viral persistence events. RNA sequencing analyses indicated that the first positive diagnosis (8 June 2020) corresponded to a SARS-CoV-2 variant belonging to Clade Nextstrain 20A. Three hundred eighty-six days later (23 September 2021), the second positive result reached the same viral variant (Clade 20A) but without presence or circulation in Chile since May 2021. Both sequencing coverages showed an identity of 99.21%, with some mutations related to the severity of the disease (ORF1b:P314L) and more infectivity (S:D614G). This work reinforces the idea of implementing an RT-qPCR or rapid antigen test once the quarantine is fulfilled to ensure viral absence, identify potential persistence, and, consequently, minimize the risk of local outbreaks of SARS-CoV-2 infection.

Paralichthys adspersus is a native species from the Pacific coast of South America that is of great economic importance for Peruvian aquaculture. Even though establishing sustainable farming depends on avoiding irreversible damage to the population gene pool due to processes such as inbreeding, studies focusing on genetically characterizing farmed stocks are yet to be documented. By using ten microsatellite loci on captive and wild individuals of P. adspersus, we successfully characterized the only commercial broodstock of this species in Peru by means of determining the genetic diversity and inferring relatedness. Although most microsatellite loci showed deviations from Hardy-Weinberg equilibrium, the genetic diversity levels in the commercial broodstock were considered healthy compared to those obtained for a wild population, with an average number of alleles of 17.40, an effective number of alleles of 9.14, and an observed and expected heterozygosity of 0.49 and 0.87. No significant differences between the broodstock and the wild population in terms of genetic diversity were observed. The fixation index and analysis of molecular variance also indicated a low rate of differentiation between both populations. In relatedness estimation, the analysis based on five method-of-moment estimators and a maximum-likelihood estimator showed the category “unrelated” as being the most probable relationship among the individuals within the commercial broodstock. Our findings reveal the conservation of genetic diversity in this population and outline potential breeding strategies that hatchery managers could use to minimize the loss of genetic diversity and long-term inbreeding. This could help establish proper genetic management in captive populations of P. adspersus and should apply to other captive stocks where pedigree information is lacking.

For decades, mangroves have been exposed to various pressures, resulting in the loss of large swathes around the world. For this reason, ecological restoration actions are presented as alternatives to recover mangroves that can restore their ecosystem services while helping to mitigate climate change’s effects. Mangroves are crucial, as they capture and sequester carbon in biomass and soil, highlighting their importance in environmental conservation and in the fight against climate change. In this research, the amount of carbon sequestered in a mangrove area restored eight years ago and its relationship with soil physicochemical parameters were evaluated and compared to those of a reference forest. Soil cores were collected at a depth of 30 cm from both sites, and in situ measurements of physical chemistry were made at different depths. In addition, soil salinity, bulk density, and carbon concentration were determined. The results revealed a similar amount of carbon sequestered both in the reference forest (BR) (470.17 ± 67.14 Mg C/ha) and in the restoration area (RA) (444.53 ± 86.11 Mg C/ha) (p > 0.05). A direct relationship was observed between carbon sequestration and soil depth. In the case of the RA, a direct relationship was found between carbon sequestration and soil salinity. In conclusion, the results of this study indicate that the behavior of carbon sequestration in soil is determined by the physicochemical conditions in both the BR and the RA, as well as by the presence or absence of vegetation.

Liver rupture secondary to preeclampsia represents a complication that, if is not recognized early, can be fatal. Its presentation is rare, but very complex to treat, having options that are not clearly protocolized and usually derive from the experience of the surgeon who faces it. We present a case of liver rupture of a patient from Toluca, State of Mexico who was admitted by emergency air transfer with immediate puerperium complicated by severe preeclampsia, postpartum hemorrhage, liver rupture with hemodynamic instability, it was necessary to intervene surgically for a total of seven occasions to control the hemorrhage, as well as pharmacological, medical measures and multiple transfusions; however, she died on the eighteenth day of his hospitalization for septic shock.

Abstract Objective The advancement of molecular techniques in an era in which high-throughput sequencing has revolutionized biology renders old-fashioned alternatives to high-throughput methods obsolete. Such advanced molecular techniques, however, are not yet accessible to economically disadvantaged region-based laboratories that still obtain DNA profiles using gel-based techniques. To explore whether cost-efficient techniques can produce results that are as robust as those obtained using high-throughput methods, we compared the performance of polyacrylamide gel electrophoresis (PAGE)- and capillary electrophoresis (CE)-derived genomic data in estimating genetic diversity and inferring relatedness using 70 individuals of fine flounder (Paralichthys adspersus) selected from a hatchery population and genotyped for five microsatellite loci. Results Here, we show that PAGE- and CE-derived genomic datasets yield comparable genetic diversity levels regarding allelic diversity measures and heterozygosity. However, relatedness inferred from each dataset showed that the categorization of dyads in the different relationship categories strongly differed. This suggests that while scientists can reliably use PAGE-derived genomic data to estimate genetic diversity, they cannot use the same for parentage testing. The findings could help laboratories committed to population research not be discouraged from using the PAGE system if high-throughput technologies are unavailable and the method is adequate to address the biological question.

Abstract The main objective of this study is to propose a theoretical model to understand the importance of disruption—in three of its dimensions, thinking, creativity, and innovation—developed and applied in the thinking and exercise of entrepreneurship and, at the same time, highlight the relationship of the disruption with competitiveness. Being competitive is one of the most demanding goals that entrepreneurs can and should set themselves; however, unlike established or traditional companies, usually the entrepreneur cannot promote competitiveness in the same way that they do. Taking these into account, it is necessary to use unconventional tools, or a different perspective methodology, for promoting creativity and innovation beyond the entrepreneurial activity itself. Through a qualitative methodology, with an emphasis on documentary research and inferential and deductive reasoning, a theoretical model is proposed. The model seeks to illustrate how disruptive currents can help entrepreneurs to be more competitive and, at the same time, boost their entrepreneurial spirit.

Air pollution in urban environments has been shown to have a negative impact on air quality and human health, particularly in megacities. Over recent decades, Delhi, India, has suffered high atmospheric pollution, with significant particulate matter (PM) concentrations as a result of anthropogenic activities. Organic aerosols (OAs) are composed of thousands of different chemical species and are one of the main constituents of submicron particles. However, quantitative knowledge of OA composition, their sources and their processes in urban environments is still limited. This is important particularly in India, as Delhi is a massive, inhomogeneous conurbation, where we would expect the apportionment and concentrations to vary depending on where in Delhi the measurements/source apportionment is performed, indicating the need for multisite measurements. This study presents the first multisite analysis carried out in India over different seasons, with a focus on identifying OA sources. The measurements were taken during 2018 at two sites in Delhi, India. One site was located at the India Meteorological Department, New Delhi (ND). The other site was located at the Indira Gandhi Delhi Technical University for Women, Old Delhi (OD). Non-refractory submicron aerosol (NR-PM1) concentrations (ammonium, nitrate, sulfate, chloride and organic aerosols) of four aerosol mass spectrometers were analysed. Collocated measurements of volatile organic compounds, black carbon, NOx and CO were performed. Positive matrix factorisation (PMF) analysis was performed to separate the organic fraction, identifying a number of conventional factors: hydrocarbon-like OAs (HOAs) related to traffic emissions, biomass burning OAs (BBOAs), cooking OAs (COAs) and secondary OAs (SOAs). A composition-based estimate of PM1 is defined by combining black carbon (BC) and NR-PM1 (C-PM1= BC + NR-PM1). No significant difference was observed in C-PM1 concentrations between sites, OD (142 ± 117 µg m−3) compared to ND (123 ± 71 µg m3), from post-monsoon measurements. A wider variability was observed between seasons, where pre-monsoon and monsoon showed C-PM1 concentrations lower than 60 µg m−3. A seasonal variation in C-PM1 composition was observed; SO42- showed a high contribution over pre-monsoon and monsoon seasons, while NO3- and Cl− had a higher contribution in winter and post-monsoon. The main primary aerosol source was from traffic, which is consistent with the PMF analysis and Aethalometer model analysis. Thus, in order to reduce PM1 concentrations in Delhi through local emission controls, traffic emission control offers the greatest opportunity. PMF–aerosol mass spectrometer (AMS) mass spectra will help to improve future aerosol source apportionment studies. The information generated in this study increases our understanding of PM1 composition and OA sources in Delhi, India. Furthermore, the scientific findings provide significant information to strengthen legislation that aims to improve air quality in India.

Atmospheric aerosol particles are known to have detrimental effects on human health and climate. Black carbon is an important constituent of atmospheric aerosol particulate matter (PM), emitted from incomplete combustion. Source apportionment of BC is very important, to evaluate the influence of different sources. The high-resolution soot particle aerosol mass spectrometer (HR-SP-AMS) instrument uses a laser vaporiser, which allows the real-time detection and characterisation of refractory black carbon (rBC) and its internally mixed particles such as metals, coating species, and rBC subcomponents in the form of HOA + fullerene. In this case study, the soot data were collected by using HR-SP-AMS during Guy Fawkes Night on 5 November 2014. Positive matrix factorisation was applied to positively discriminate between different wood-burning and bonfire sources for the first time, which no existing black carbon source apportionment technique is currently able to do. Along with this, the use of the fullerene signals in differentiating between soot sources and the use of metals as a tracer for fireworks has also been investigated, which did not significantly contribute to the rBC concentrations. The addition of fullerene signals and successful positive matrix factorisation (PMF) application to HR-SP-AMS data apportioned rBC into more than two sources. These bonfire sources are HOA + fullerene, biomass burning organic aerosol, more oxidised oxygenated organic aerosol (MO-OOA), and non-bonfire sources such as hydrocarbon-like OA and domestic burning. The result of correlation analysis between HR-SP-AMS data and previously published Aethalometer, MAAP, and CIMS data provides an effective way of gaining insights into the relationships between the variables and provide a quantitative estimate of the source contributions to the BC budget during this period. This research study is an important demonstration of using HR-SP-AMS for the purpose of BC source apportionment.

Abstract Background SARS-CoV-2 is a recently emerged pandemic coronavirus (CoV) capable of causing severe respiratory illness. However, a significant number of infected people present as asymptomatic or pauci-symptomatic. In this prospective assessment of at-risk healthcare workers (HCWs) we seek to determine whether pre-existing antibody or T cell responses to previous seasonal human coronavirus (HCoV) infections affect immunological or clinical responses to SARS-CoV-2 infection or vaccination. Methods A cohort of 300 healthcare workers, confirmed negative for SARS-CoV-2 exposure upon study entry, will be followed for up to 1 year with monthly serology analysis of IgM and IgG antibodies against the spike proteins of SARS-CoV-2 and the four major seasonal human coronavirus - HCoV-OC43, HCoV-HKU1, HCoV-229E, and HCoV-NL63. Participants will complete monthly questionnaires that ask about Coronavirus Disease 2019 (COVID-19) exposure risks, and a standardized, validated symptom questionnaire (scoring viral respiratory disease symptoms, intensity and severity) at least twice monthly and any day when any symptoms manifest. SARS-CoV-2 PCR testing will be performed any time participants develop symptoms consistent with COVID-19. For those individuals that seroconvert and/or test positive by SARS-CoV-2 PCR, or receive the SARS-CoV-2 vaccine, additional studies of T cell activation and cytokine production in response to SARS-CoV-2 peptide pools and analysis of Natural Killer cell numbers and function will be conducted on that participant’s cryopreserved baseline peripheral blood mononuclear cells (PBMCs). Following the first year of this study we will further analyze those participants having tested positive for COVID-19, and/or having received an authorized/licensed SARS-CoV-2 vaccine, quarterly (year 2) and semi-annually (years 3 and 4) to investigate immune response longevity. Discussion This study will determine the frequency of asymptomatic and pauci-symptomatic SARS-CoV-2 infection in a cohort of at-risk healthcare workers. Baseline and longitudinal assays will determine the frequency and magnitude of anti-spike glycoprotein antibodies to the seasonal HCoV-OC43, HCoV-HKU1, HCoV-229E, and HCoV-NL63, and may inform whether pre-existing antibodies to these human coronaviruses are associated with altered COVID-19 disease course. Finally, this study will evaluate whether pre-existing immune responses to seasonal HCoVs affect the magnitude and duration of antibody and T cell responses to SARS-CoV-2 vaccination, adjusting for demographic covariates.

In this study, the seismic responses of moment-resisting steel frames (MRSFs) with welded and post-tensioned connections under 28 artificial seismic sequences (mainshock–aftershock) are compared. For this aim, the mainshock are scaled at different ground motion intensity levels as a function of the spectral pseudo-acceleration corresponding to the fundamental period of vibration of the structure Sa(T1), whereas different intensity levels of the aftershocks are used for a percentage of the peak maximum acceleration of the mainshock. The seismic performance comparison of both structural systems is computed for the maximum, residual inter-story drift and hysteretic energy demands. The results show that post-tensioned frames significantly reduce the structural demands, especially in the case of residual inter-story drifts and hysteretic energy in comparison with moment-resisting steel frames with welded connections. The reductions in the structural response tend to be larger as the intensity of the aftershock tends to increase. Therefore, it is concluded that the use of post-tensioned connections is a great alternative to mitigate the seismic response of buildings subjected to seismic sequences.

Gastric cancer is a challenging public health concern worldwide and remains a leading cause of cancer-related mortality. The primary risk factor implicated in gastric cancer development is infection with Helicobacter pylori. H. pylori induces chronic inflammation affecting the gastric epithelium, which can lead to DNA damage and the promotion of precancerous lesions. Disease manifestations associated with H. pylori are attributed to virulence factors with multiple activities, and its capacity to subvert host immunity. One of the most significant H. pylori virulence determinants is the cagPAI gene cluster, which encodes a type IV secretion system and the CagA toxin. This secretion system allows H. pylori to inject the CagA oncoprotein into host cells, causing multiple cellular perturbations. Despite the high prevalence of H. pylori infection, only a small percentage of affected individuals develop significant clinical outcomes, while most remain asymptomatic. Therefore, understanding how H. pylori triggers carcinogenesis and its immune evasion mechanisms is critical in preventing gastric cancer and mitigating the burden of this life-threatening disease. This review aims to provide an overview of our current understanding of H. pylori infection, its association with gastric cancer and other gastric diseases, and how it subverts the host immune system to establish persistent infection.

Abstract The dense Miocene record of cetaceans is known from localities along the coasts of all continents, mostly in the northern Atlantic or the eastern Pacific regions, but Antarctica. Fossils from the Caribbean region are few and include of a couple of findings from Panama and Venezuela. Here, we report a partly complete skull from the Caujarao Formation (middle Miocene), Falcon State, Caribbean region of Venezuela. Our phylogenetic analyses indicate that the Caujarao specimen is a ‘stem delphinidan’, a group that includes several taxa of early diverging odontocetes whose phylogenetic affinities remain a matter of debate. The fossil record has shown that this group of stem delphinidans was taxonomically diverse, but displayed a somewhat homogeneous cranial patterning, with most of the variations being found within the mandible or tympanoperiotic characters. As other stem delphinidans the Caujarao odontocete displays an enlarged temporal fossa and a fairly symmetrical cranium. Because the skull is missing several key diagnostic characters due to the preservation state of the specimen, a more precise taxonomic identification is not possible. Despite this, the finding of this specimen highlights the importance of the fossil record from the Neogene of Venezuela, and the importance of the area to understand cetacean evolution in the proto-Caribbean.

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous pollutants in air, soil, and water and are known to have harmful effects on human health and the environment. The diurnal and nocturnal variations of 17 PAHs in ambient particle-bound PAHs were measured in urban Beijing (China) and Delhi (India) during the summer season using gas-chromatography–quadrupole time-of-flight mass spectrometry (GC-Q-TOF-MS). The mean concentration of particles less than 2.5 µm (PM2.5) observed in Delhi was 3.6 times higher than in Beijing during the measurement period in both the daytime and night-time. In Beijing, the mean concentration of the sum of the 17 PAHs (∑17 PAHs) was 8.2 ± 5.1 ng m−3 in daytime, with the highest contribution from indeno[1,2,3-cd]pyrene (12 %), while at night-time the total PAHs was 7.2 ± 2.0 ng m−3, with the largest contribution from benzo[b]fluoranthene (14 %). In Delhi, the mean ∑17 PAHs was 13.6 ± 5.9 ng m−3 in daytime and 22.7 ± 9.4 ng m−3 at night-time, with the largest contribution from indeno[1,2,3-cd]pyrene in both the day (17 %) and night (20 %). Elevated mean concentrations of total PAHs in Delhi observed at night were attributed to emissions from vehicles and biomass burning and to meteorological conditions leading to their accumulation from a stable and low atmospheric boundary layer. Local emission sources were typically identified as the major contributors to total measured PAHs in both cities. Major emission sources were characterized based on the contribution from each class of PAHs, with the four-, five- and six-ring PAHs accounting ∼ 95 % of the total PM2.5-bound PAHs mass in both locations. The high contribution of five-ring PAHs to total PAH concentration in summer Beijing and Delhi suggests a high contribution from petroleum combustion. In Delhi, a high contribution from six-ring PAHs was observed at night, suggesting a potential emission source from the combustion of fuel and oil in power generators, widely used in Delhi. The lifetime excess lung cancer risk (LECR) was calculated for Beijing and Delhi, with the highest estimated risk attributed to Delhi (LECR = 155 per million people), which is 2.2 times higher than the Beijing risk assessment value (LECR = 70 per million people). Finally, we have assessed the emission control policies in each city and identified those major sectors that could be subject to mitigation measures.

Little is known about how cardiac metabolism remodels following cardiac injury. Here, the authors show that mitochondrial CaMKII plays an important role in remodeling cardiac metabolism after injury and that replacement of mitochondrial creatine kinase improves energetics and protects against adverse remodeling.

Nicotinamide adenine dinucleotide and coenzyme A serve as a 5′-cap of prokaryotic RNA. Here the authors report that methylated and non-methylated dinucleoside polyphosphates (Np n Ns) exist as Escherichia coli RNA caps which can be cleaved by 5′-pyrophosphohydrolase (RppH) and bis(5′-nucleosyl)-tetraphosphatase (ApaH).

The stress response in fish is characterized by the activation of the HPI axis resulting in the release of cortisol. Previous studies in rainbow trout (Oncorhynchus mykiss) and gilthead sea bream (Sparus aurata) have shown that an acute stressor modulates the expression of immune- and stress-related genes in mucosal-associated lymphoid tissues (MALTs), particularly in the skin (SALT), gills (GIALT), and gut (GALT). However, there are no antecedents on whether the modulation on the mucosal transcriptomic profile is coordinated through the local presence of cortisol in the mucosal tissue surface. Thus, the aim of this study was to evaluate the effect of cortisol upon the modulation of a set of immune- (il-1β, il-6, tnf-α, and cox-2) and stress-related (hsp70, gr1) genes. For this purpose, tissue explants cultures were incubated with cortisol (100 ng/100 mg tissue) for 2-, 4-, and 24 h and the gene expression profile was evaluated at each time-point by real-time PCR. No differences were found in the gene expression between cortisol-incubated tissue explants and mock-incubated tissues in any of the time-points tested for both species. These results suggest that the quick modulation of the gene expression during the first 24 h after the exposure to stressor challenge reported in previous studies, is probably coordinated and mediated through a systemic-dependent mechanism but not through a peripheral/local response on mucosal tissue surfaces.

The spray-dried porcine plasma (SDPP) is an abattoir by-product used in animal nutrition with beneficial effects reported in livestock and commercial aquatic species. Previous results have found that the dietary inclusion of SDPP in gilthead sea bream (Sparus aurata) increased the density of intestinal goblet cells, and it did not result in significant changes in the autochthonous microbiota. However, there is no comprehensive data on the mechanisms that could take place on the intestine of gilthead sea bream fed with an SDPP-supplemented diet. For this reason, this study aimed to unveil the biological mechanisms modulated in response to the dietary administration of SDPP in the gilthead sea bream gut. To achieve this goal, we made a microarrays-based transcriptomic approach in gut samples from gilthead sea bream fed with an SDPP-supplemented diet for 95 days. As control diet, we used a protein-rich commercial feed (51% crude protein, 17% crude fat, and 20.6 MJ/kg gross energy) which was supplemented with 3% SDPP at the expense of LT70 fishmeal. The microarray analyses showed a total of 803 (468 up- and 335 down-regulated) differential expressed genes (DEGs). The functional network analysis revealed that dietary inclusion of SDPP induced sustained changes in 120 biological processes, grouped in 12-clusters. Among them, the metabolic-related process (cellular catabolic process, organic substance catabolic process, protein metabolism process), protein transport, and leukocyte mediated immunity interacted in the leading interactome network. This evidence confirms the previous evidence of the enhancement of the mucosal health status in response to the dietary administration of SDPP and provides further understanding of the mode of action of this ingredient in aquafeeds.

The transcriptomic response of the head kidney, the main lymphohematopoietic tissue of the body, was evaluated in Atlantic salmon (Salmo salar) smolts fed a functional feed containing a phytogenic rich in verbascoside and triterpenic compounds like ursolic acid. Fish (initial body weight = 55.0 ± 0.1 g) were fed two experimental diets (40% crude protein, 22% crude fat; 21.6 MJ/kg gross energy) that only differed in the phytogenic content (0.1% inclusion). Each diet has six replicates and was tested over a period of 133 days. The tested zootechnical feed additive a medicinal plant leaf extract (MPLE) obtained from sage (Salvia officinalis) and lemon verbena (Lippia citriodora). At the end of the trial, smolts fed the MPLE diet were heavier than their congeners from the control group (271.5 ± 7.9 g vs. 240.2 ± 19.3 g, respectively; P < 0.05). Feed conversion ratio (FCR) values in fish fed the control diet were higher than those in fish fed the MPLE diet (FCRcontrol diet = 1.27 ± 0.08 vs. FCR0.1% MPLE diet = 1.08 ± 0.05; P < 0.05). The immunomodulatory properties of the functional diet were evaluated by means of an in vivo challenge with Aeromonas salmonicida subsp. salmonicida (1 × 107 CFU mL–1). The microarray analysis of head kidney samples from both dietary groups revealed 1,178 differentially expressed genes (802 upregulated and 376 downregulated). Among them, several biological processes related to immunity were identified in fish fed the MPLE diet (i.e., interferon-gamma-mediated signaling pathway, antigen processing and presentation of peptide antigen via MHC class II, autophagy, regulation of i-kappaB kinase/NF-kappaB signaling, and leukocyte activation). Results from the bacterial challenge showed that survival rates were higher in smolts from the MPLE group (90.6 ± 6.4%) in comparison to the control group (60.7 ± 13.5%), confirming the functional benefits of the phytogenic in terms of host’s immunity and disease resistance. Biological processes such as cytoskeleton organization and regulation of cellular protein metabolic process detected in fish fed the MPLE diet supported the metabolic changes related to increased somatic growth promoted. The present findings showed that the inclusion at 0.1% of the tested MPLE obtained from sage and lemon verbena in diets for Atlantic salmon smolts promoted somatic growth, and enhanced their systemic immune response and reduced mortality when fish were challenged with A. salmonicida cumulative, the causative agent of furunculosis in salmonids.

The productivity of mangrove ecosystems is associated with litterfall production, which continuously contributes large quantities of organic matter, in the form of detritus, to the food web via adjacent ecosystems. However, the degree of deterioration of mangrove ecosystems worldwide has been increasing due to anthropogenic activities, leading to the loss of vegetation cover and changes in hydrological patterns, the chemical conditions of interstitial water and soil, and the litterfall degradation rate and, thus, the integration of organic matter into the ecosystem. In this study, we investigate the relationship between leaf degradation and interstitial water chemistry in two mangrove forests located in Oaxaca, Mexico, that are characterized by differences in environmental conditions, species, and anthropogenic activity. Forty-two 10 cm × 20 cm nylon mesh bags were installed in the Rhizophora mangle forest along two flood-associated lines (21 per line), and twenty-one bags were installed centrally in the Avicennia germinans forest because of the flood conditions in this area. Three bags per line were collected each month. This material was then dried and calcined for determination of the decomposition rate (k). The in situ redox potential and interstitial water salinity of mangrove forests were measured using a HACH HQ40d multiparametric probe and A&O refractometer, and the sulfate concentration was determined by ion chromatography. The results show that daily average decomposition rates were higher in the Salina lagoon (k = 0.01 g·day−1) than in the Chacahua lagoon (k = 0.004 g·day−1). The degradation model was Y = 66.054e−0.010t, R2 = 0.89, p ≤ 0.05, for the Salina lagoon and Y = 67.75e−0.004t, R2 = 0.76, p ≤ 0.05, for the Chacahua lagoon. Leaf decomposition rates differed between the Salina and Chacahua lagoons (F1,206 = 4.8, p < 0.03). In the Salina lagoon, dominated by A. germinans, an inverse relationship was established between the percentage of degraded biomass with respect to salinity concentration (R2 = 0.82, p < 0.013) and redox potential (R2 = 0.89, p < 0. 015), and for the Chacahua lagoon, dominated by R. mangle, the percentage of degraded litter biomass was found to be inversely correlated with redox potential (R2 = 0.94, p < 0.005) and sulfate concentration (R2 = 0.88, p < 0.017). Based on the results obtained in this study, we conclude that variations in the chemical conditions of interstitial water and hydrological patterns can affect the process of mangrove leaf degradation based on species and the integration of organic matter in the soil and in adjacent ecosystems. These findings are potentially useful for mangrove management because they advance understanding of the dynamics of organic matter in mangroves and the importance of maintaining the health of these ecosystems, which is necessary for the maintenance of coastal fishing production.

Timely detection of severe acute respiratory syndrome due to coronavirus 2 (SARS-CoV-2) by reverse transcription quantitative polymerase chain reaction (RT-qPCR) has been the gold- strategy for identifying positive cases during the current pandemic. However, faster and less expensive methodologies are also applied for the massive diagnosis of COVID-19. In this way, the rapid antigen test (RAT) is widely used. However, it is necessary to evaluate its detection efficiency considering the current pandemic context with the circulation of new viral variants. In this study, we evaluated the sensitivity and specificity of RAT (SD BIOSENSOR, South Korea), widely used for testing and SARS-CoV-2 diagnosis in Santiago of Chile. The RAT showed a 90% (amplification range of 20 ≤ Cq 30. In SARS-CoV-2 variant detection, RAT had a 42.8% detection sensitivity in samples with RT-qPCR amplification range 20 ≤ Cq

The early detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) using the real-time quantitative polymerase chain reaction (RT-qPCR) as a gold-standard molecular tool has allowed to test and trace the viral spread and the isolation of COVID-19-infected patients. The detection capacity of viral and internal genes is an essential parameter to consider and analyze during the assay. In this study, we analyze the performance of the two commercial RT-qPCR kits used in Chile, TaqMan™ 2019-nCoV Control Kit v1 (Thermo Fisher) and MaxCov19 (TAAG Genetics), for the COVID-19 diagnosis from nasopharyngeal swab samples (NPSs). Our results show a lower sensitivity of the TAAG kit compared to the Thermo Fisher kit, even in the detection of SARS-CoV-2 mutations associated with its variants. This study reinforces the relevance of evaluating the performance of RT-qPCR kits before being used massively since those with lower sensitivity can generate false negatives and produce outbreaks of local infections.

This paper addresses the importance of engineering asset management decisions and control. For this purpose, a Life-Cycle Cost (LCC) analysis is conducted for typical reinforced concrete (R/C) buildings located in Mexico City. The objective of this study is to develop an artificial neural network (ANN) model that can estimate the total expected cost of R/C buildings by using LCC functions. The total cost includes the initial cost and the cost of the damage caused by future possible ground motions at the site of interest. The present value of the cost includes: initial cost, repair or reconstruction cost, cost of damage to the contents, costs associated with the loss of life or injuries and economic losses. The structural performance is evaluated using probabilistic models, artificial neural networks models are used to obtain the seismic response of the buildings. The methodology is applied to a set of reinforced concrete buildings with 4, 8, and 12 stories which are located at the soft soil of Mexico City. Finally, it is concluded that the life-cycle cost is efficiently obtained using artificial neural network models for estimating the structural reliability of reinforced concrete buildings, in such a way that it can be used as an excellent planning tool that covers long spans of time.

Blood by-products are an untapped source of high-quality ingredients for aquafeeds, containing a broad variety of cytokines, hormones, growth factors, proteins, bioactive peptides, and amino acids. The effects of the spray-dried porcine plasma (SDPP), a type of processed animal protein on several immune parameters, were evaluated in sea bream using ex vivo and in vitro assays. In this study, fish were fed with two isoproteic, isolipidic, and isoenergetic diets: control diet (7% fish meal, FM) and SDPP diet (2% FM and 5% SDPP). At the end of the 92-days trial, those fed the SDPP diet were larger in body weight (p < 0.05) without differences in feed conversion ratio (p > 0.05). The ex vivo immune stimulation of splenocytes indicated that SDPP had a beneficial effect in promoting systemic immunity, since the surface cell marker (cd4), pro- (il-1β), and anti-inflammatory (tgf-β1) cytokines, and genes involved in humoral immunity (IgM) were up-regulated. The co-culture assays of skin mucus corroborated that SDPP enhanced the antibacterial capacity of mucus against V. anguillarum. In addition, main mucus biomarkers did not show significant differences, except for cortisol levels which were lower in the SDPP diet. The present study indicated that SDPP may be considered a functional ingredient in aquafeeds formulated with low FM levels.

La región, en medio de las dificultades recurrentes a las que se enfrenta, tiene también el desafío de fortalecer el conjunto de instituciones, puesto que la educación, juega un papel por demás determinante en los procesos de desarrollo. En este sentido, el principal objetivo de este trabajo es analizar el comportamiento de la inversión en educación, por parte de los Países Andinos, Bolivia, Colombia, Ecuador, Perú y Venezuela, en comparación con América Latina y el Caribe, a fin de considerar los elementos centrales de su incidencia en la responsabilidad social, en el período de 1995 a 2015. El estudio es descriptivo-documental, basado en la información de reconocidas bases de datos. Entre los resultados destaca que los Países Andinos muestran mayores niveles de inversión en educación que el total de América Latina y el Caribe. Asimismo, la educación, en especial en Bolivia, Ecuador y Venezuela, durante el período estudiado tendió a enfatizar, en la política social y económica general. En conclusión, la concepción de educación como un bien público accesible a la población, fortalece los mecanismos de la oferta, por tanto en el nivel universitario, las instancias públicas pueden jugar un papel primordial en la formación del capital o talento humano.

Resumen: Introducción: La falla cardiaca asociada a deficiencia de hierro se relaciona directamente con disminución de parámetros funcionales y, por ende, con deterioro de la calidad de vida y pobre pronóstico de los pacientes que la padecen. Se ha encontrado que la corrección de la deficiencia de hierro mejora a corto plazo la clase funcional y otros parámetros de la enfermedad. Objetivo: Evaluar la eficacia y la seguridad del uso de hierro en el tratamiento de la falla cardiaca. Métodos: De seleccionaron ensayos clínicos que evaluaran el uso de hierro en administración intravenosa u oral vs. placebo en el contexto de pacientes con falla cardíaca y a su vez se eligieron pacientes que concomitantemente sufrieran anemia ferropénica. En la búsqueda se incluyeron bases de datos como MEDLINE (PubMed), Embase, Cochrane Central Register of Clinical trials (CENTRAL), LILACS y WPRIM. Resultados: De 10.729 títulos obtenidos 6 fueron elegibles con 835 pacientes, de los cuales 520 fueron tratados con terapia férrica y 315 con placebo o terapia convencional de falla cardiaca. Debido a la heterogeneidad de los resultados no fue posible hacer un metaanálisis. Conclusión: Se identificó mejoría significativa en múltiples parámetros evaluados, tales como el test de caminata de 6 minutos, clase funcional de la New York Heart Association, consumo máximo de oxígeno (VO2 máx.) y valoración global subjetiva. Por consiguiente, la terapia férrica es una opción segura y eficaz en el manejo de pacientes con falla cardiaca ya que mejora significativamente su capacidad funcional, calidad de vida y múltiples parámetros paraclínicos. Abstract: Introduction: Heart failure combined with iron deficiency is directly related with the reduction in functional parameters and, as consequence, with a deterioration in the quality of life and poor prognosis in the patients that suffer from it. It has been found that correction of the iron deficiency improves the functional class and other parameters of the disease in the short-term. Objective: To evaluate the efficacy and safety of the use of iron in the treatment of heart failure. Methods: Clinical trials were selected that evaluated the use of administering intravenous or oral iron vs. placebo in the context of patients with cardiac failure and at the same time, patients were selected that concomitantly suffered from iron deficiency anaemia. The search was made in data bases such as MEDLINE (PubMed), Embase, Cochrane Central Register of Clinical trials (CENTRAL), LILACS and WPRIM. Results: Of the 10,729 articles obtained, only 6 were eligible with 835 patients, of which 520 were treated with iron therapy and 315 with a placebo or conventional heart failure therapy. Due to the heterogeneity of the results, it was not possible to perform a meta-analysis. Conclusion: A significant improvement was identified in several of the parameters evaluated, such as the 6-minute walk test, the New York Heart Association functional class, maximum oxygen uptake (VO2 max.), and an overall subjective evaluation. This showed that iron therapy is a safe and effective option in the management of patients with heart failure since there was a significant improvement in their functional capacity, quality of life, and several para-clinical parameters. At the end of 2015, a CO2/CH4/CO cavity ring-down spectrometer (CRDS) was installed at the Izaña Global Atmosphere Watch (GAW) station (Tenerife, Spain) to improve the Izaña Greenhouse Gases GAW Measurement Programme, and to guarantee the renewal of the instrumentation and the long-term maintenance of this program. We present the results of the CRDS acceptance tests, the raw data processing scheme applied, and the response functions used. Also, the calibration results, the implemented water vapor correction, the target gas injection statistics, the ambient measurements performed from December 2015 to July 2017, and their comparison with other continuous in situ measurements are described. The agreement with other in situ continuous measurements is good most of the time for CO2 and CH4, but for CO it is just outside the GAW 2 ppb objective. It seems the disagreement is not produced by significant drifts in the CRDS CO World Meteorological Organization (WMO) tertiary standards. The more relevant contributions of the present article are (1) determination of linear relationships between flow rate, CRDS inlet pressure, and CRDS outlet valve aperture; (2) determination of a slight CO2 correction that takes into account changes in the inlet pressure/flow rate (as well as its stability over the years), and attributing it to the existence of a small spatial inhomogeneity in the pressure field inside the CRDS cavity due to the gas dynamics; (3) drift rate determination for the pressure and temperature sensors located inside the CRDS cavity from the CO2 and CH4 response function drift trends; (4) the determination of the H2O correction for CO has been performed using raw spectral peak data instead of the raw CO provided by the CRDS and using a running mean to smooth random noise in a long water-droplet test (12 h) before performing the least square fit; and (5) the existence of a small H2O dependence in the CRDS flow and of a small spatial inhomogeneity in the temperature field inside the CRDS cavity are pointed out and their origin discussed.

Aquaculture growth will unavoidably involve the implementation of innovative and sustainable production strategies, being functional feeds among the most promising ones. A wide spectrum of phytogenics, particularly those containing terpenes and organosulfur compounds, are increasingly studied in aquafeeds, due to their growth promoting, antimicrobial, immunostimulant, antioxidant, anti-inflammatory and sedative properties. This trend relies on the importance of the mucosal barrier in the fish defense. Establishing the phytogenics’ mode of action in mucosal tissues is of importance for further use and safe administration. Although the impact of phytogenics upon fish mucosal immunity has been extensively approached, most of the studies fail in addressing the mechanisms underlying their pharmacological effects. Unstandardized testing as an extended practice also questions the reproducibility and safety of such studies, limiting the use of phytogenics at commercial scale. The information presented herein provides insight on the fish mucosal immune responses to phytogenics, suggesting their mode of action, and ultimately encouraging the practice of reliable and reproducible research for novel feed additives for aquafeeds. For proper screening, characterization and optimization of their mode of action, we encourage the evaluation of purified compounds using in vitro systems before moving forward to in vivo trials. The formulation of additives with combinations of compounds previously characterized is recommended to avoid bacterial resistance. To improve the delivery of phytogenics and overcome limitations associated to compounds volatility and susceptibility to degradation, the use of encapsulation is advisable. Besides, newer approaches and dedicated methodologies are needed to elucidate the phytogenics pharmacokinetics and mode of action in depth.

One of the main targets for the use of phytogenics in aquafeeds is the mucosal tissues as they constitute a physical and biochemical shield against environmental and pathogenic threats, comprising elements from both the innate and acquired immunity. In the present study, the modulation of the skin transcriptional immune response, the bacterial growth capacity in skin mucus, and the overall health condition of gilthead seabream (Sparus aurata) juveniles fed a dietary supplementation of garlic essential oil, carvacrol, and thymol were assessed. The enrichment analysis of the skin transcriptional profile of fish fed the phytogenic-supplemented diet revealed the regulation of genes associated to cellular components involved in the secretory pathway, suggesting the stimulation, and recruitment of phagocytic cells. Genes recognized by their involvement in non-specific immune response were also identified in the analysis. The promotion of the secretion of non-specific immune molecules into the skin mucus was proposed to be involved in the in vitro decreased growth capacity of pathogenic bacteria in the mucus of fish fed the phytogenic-supplemented diet. Although the mucus antioxidant capacity was not affected by the phytogenics supplementation, the regulation of genes coding for oxidative stress enzymes suggested the reduction of the skin oxidative stress. Additionally, the decreased levels of cortisol in mucus indicated a reduction in the fish allostatic load due to the properties of the tested additive. Altogether, the dietary garlic, carvacrol, and thymol appear to promote the gilthead seabream skin innate immunity and the mucus protective capacity, decreasing its susceptibility to be colonized by pathogenic bacteria.

Essential oils (EOs) are promising alternatives to chemotherapeutics in animal production due to their immunostimulant, antimicrobial, and antioxidant properties, without associated environmental or hazardous side effects. In the present study, the modulation of the transcriptional immune response (microarray analysis) and microbiota [16S Ribosomal RNA (rRNA) sequencing] in the intestine of the euryhaline fish gilthead seabream (Sparus aurata) fed a dietary supplementation of garlic, carvacrol, and thymol EOs was evaluated. The transcriptomic functional analysis showed the regulation of genes related to processes of proteolysis and inflammatory modulation, immunity, transport and secretion, response to cyclic compounds, symbiosis, and RNA metabolism in fish fed the EOs-supplemented diet. Particularly, the activation of leukocytes, such as acidophilic granulocytes, was suggested to be the primary actors of the innate immune response promoted by the tested functional feed additive in the gut. Fish growth performance and gut microbiota alpha diversity indices were not affected, while dietary EOs promoted alterations in bacterial abundances in terms of phylum, class, and genus. Subtle, but significant alterations in microbiota composition, such as the decrease in Bacteroidia and Clostridia classes, were suggested to participate in the modulation of the intestine transcriptional immune profile observed in fish fed the EOs diet. Moreover, regarding microbiota functionality, increased bacterial sequences associated with glutathione and lipid metabolisms, among others, detected in fish fed the EOs supported the metabolic alterations suggested to potentially affect the observed immune-related transcriptional response. The overall results indicated that the tested dietary EOs may promote intestinal local immunity through the impact of the EOs on the host-microbial co-metabolism and consequent regulation of significant biological processes, evidencing the crosstalk between gut and microbiota in the inflammatory regulation upon administration of immunostimulant feed additives.
