Pseudomonas syringae pv. apii, causal agent of bacterial leaf spot (BLS) of celery, was first identified in California in 1989. By 1991, BLS was apparent in all celery-growing areas of the state. Greenhouse-produced transplants were affected most severely, and disease incidence approached 100% in some greenhouses. In this study, sources of inoculum and factors contributing to disease development were investigated in three Salinas Valley greenhouse operations during the 1991, 1992, and 1993 celery transplant seasons (January to August). Epiphytic P. syringae pv. apii was not detected on celery transplants until April or May of each year. Increased epiphytic populations preceded BLS outbreaks, and high-pressure, overhead irrigation favored bacterial infiltration and disease development. In seed-wash assays, P. syringae pv. apii was recovered from 5 of 24 commercial celery seed lots. In field tests, epiphytic P. syringae pv. apii was found on umbels of inoculated celery plants, and seeds from these plants were heavily contaminated with P. syringae pv. apii. Contaminated seed produced seedlings with large epiphytic P. syringae pv. apii populations. Hot-water treatment (50°C for 25 min) eliminated >99.9% of seed contamination. Based on these results, disease management techniques are proposed.

Bacterial blight is a new disease of broccoli raab or rappini (Brassica rapa subsp. rapa) that has developed on commercially grown crops in the Salinas Valley (Monterey County) in California. Symptoms consist of small, angular, water-soaked flecks on lower foliage that are visible from both adaxial and abaxial sides of the leaves. These flecks expand and become surrounded by bright yellow borders. With time, multiple leaf spots coalesce and result in large, irregular necrotic areas, extensive leaf yellowing, and eventual leaf death. If symptoms develop on the uppermost leaves attached to the inflorescence, the shoot loses market quality and will not be harvested. Pseudomonas syringae was consistently isolated from symptomatic plants, and selected strains caused similar symptoms when inoculated onto broccoli raab test plants. Broccoli raab strains caused leaf spot symptoms on nine other Cruciferous plants, as well as on three grass species (California brome, oat, and common timothy). Conversely, broccoli raab was not infected by P. syringae pathovars coronafaciens, maculicola, and tomato. Broccoli raab strains were positive for coronatine toxin production. Fatty acid analyses indicated that the P. syringae from broccoli raab was most closely related to P. syringae pvs. coronafaciens and maculicola, but its distinct host range suggests that it may be considered a separate pathovar.

The effects of 11 different auxins and one cytokinin-like compound were tested at four concentrations for their ability to induce primary and repetitive somatic embryos from mature, dry peanut (Arachis hypogaea L.) epicotyls of genotype AT120. Treatment with picloram and centrophenoxine at 83.0 and 124.4 μM resulted in the greatest number of embryos per explant and the highest percentage of explants responding. In a follow-up experiment, picloram, centrophenoxine, and dicamba were tested at 83.0 and 124.4 μM on four peanut genotypes (AT120, 59-4144, GK7, and VC1). Picloram and centrophenoxine induced similar numbers of globular-stage and total embryos from each genotype, while dicamba was less effective. Similar results were observed with percentage of responding axes. Genotypes AT120 and VC1 yielded more clusters of repetitive embryos than GK7 and 59-4144. After 5 months, embryos derived from repetitive embryogenic cultures were converted into mature plants.

Cucurbit yellow vine disease (CYVD), caused by the squash bug (Anasa tristis)-transmitted bacterium Serratia marcescens, was first identified in Oklahoma and Texas in 1988 and in Georgia in 2012. S. marcescens is a highly diverse species found in many ecological niches. In previous studies, CYVD strains of S. marcescens formed a closely related group separate from non-CYVD strains based on biological and molecular characterization techniques. Multilocus sequence analysis (MLSA) of six housekeeping genes and repetitive elements-based polymerase chain reaction (rep-PCR) using the BOX and ERIC primers were used to assess the genetic diversity of CYVD strains of S. marcescens collected in Georgia together with a strain from Texas and seven non-CYVD strains of S. marcescens. rep-PCR results revealed genetic diversity among CYVD strains while MLSA results showed a 100% similarity across the six loci for all but one of the CYVD strains, which differed at the icd locus by five polymorphisms. For both methods, CYVD strains clustered separately from nonplant-pathogenic S. marcescens strains and were most similar to a rice endophyte strain. One CYVD strain isolated from a squash bug shared genetic similarities with non-CYVD strains, and may be the result of a recombination event between CYVD and non-CYVD strains.

Bacterial canker is one of the most economically important diseases of stone fruit trees, including ‘French’ prune (Prunus domestica). Field trials were conducted to evaluate the effect of rootstock selection and budding height on the incidence and severity of bacterial canker in four orchards with low to high disease pressure. Treatments included French prune scions low-grafted on ‘Lovell’ peach (Prunus persica) rootstocks as well as Myrobalan 29C (Prunus cerasifera) plum rootstocks grafted at 15, 50, and 90 cm above the rootstock crown. Another treatment consisted of growing Myrobalan 29C plum rootstocks in the field for one growing season, then field-grafting French prune buds onto rootstock scaffolds. Lovell peach rootstock provided the greatest protection from bacterial canker as measured by disease incidence and tree mortality in all orchards. Field-budded rootstocks and rootstocks grafted at the highest budding height provided moderate levels of resistance to bacterial canker. These treatments reduced the incidence but not the severity of disease.

A bacterial disease of celery, caused by Pseudomonas syringae pv. apii, has been identified for the first time in California. Bacterial blight develops on greenhouse-grown transplants and has spread into production fields throughout coastal regions of the state. Although this disease can be confused with fungal celery blights caused by Septoria apiicola and Cercospora apii, it has several distinguishing characteristics.

Strains of Pseudomonas syringae pv. syringae were isolated from healthy and diseased stone fruit tissues sampled from 43 orchard sites in California in 1995 and 1996. These strains, together with P. syringae strains from other hosts and pathovars, were tested for pathogenicity and the presence of the syrB and syrC genes and were genetically characterized by using enterobacterial repetitive intergenic consensus (ERIC) primers and PCR. All 89 strains of P. syringae pv. syringae tested were moderately to highly pathogenic on Lovell peach seedlings regardless of the host of origin, while strains of other pathovars exhibited low or no pathogenicity. The 19 strains of P. syringae pv. syringae examined by restriction fragment length polymorphism analysis contained the syrB and syrC genes, whereas no hybridization occurred with 4 strains of other P. syringae pathovars. The P. syringae pv. syringae strains from stone fruit, except for a strain from New Zealand, generated ERIC genomic fingerprints which shared four fragments of similar mobility. Of the P. syringae pv. syringae strains tested from other hosts, only strains from rose, kiwi, and pear generated genomic fingerprints that had the same four fragments as the stone fruit strains. Analysis of the ERIC fingerprints from P. syringae pv. syringae strains showed that the strains isolated from stone fruits formed a distinct cluster separate from most of the strains isolated from other hosts. These results provide evidence of host specialization within the diverse pathovar P. syringae pv. syringae.

In this study, we examined the relationship among various strains of Pseudomonas syringae pv. tomato (Pst), P. syringae pv. apii (Psa), and P. syringae pv. macuilcoia (Psm). The potential for cross-pathogenicity among Psa, Psm, and Pst strains was evaluated using pathogenicity tests. Genetic diversity was examined using restriction fragment length polymorphisms (RFLPs) and random amplified polymorphic DNA (RAPDs) analyses. Our results suggest that classification of these bacteria as distinct pathovars is valid.
