Abstract

A pulse-chase technique was used to investigate the hydrolysis and oxidation of intramuscular lipid pools and the esterification and oxidation of palmitate in isolated soleus muscles at rest and during tetanic contractions. Intramuscular lipid pools were labeled by pulsing muscles with [14C]palmitate. Results show that oxidation rates of exogenous and intramuscular lipids increase in proportion to each and the concomitant palmitate is esterified in proportion to its oxidation during contraction.

Abstract

Studies were performed to examine the kinetics of palmitate transport into skeletal muscle vesicles, to study variations in the transport capacity in red and white muscles and to investigate the amount of putative long-chain fatty acid (LCFA) transporters in red and white muscles. LCFA transport was measured in the absence of LCFA metabolism using a procedure that measures palmitate transport in giant sarcolemmal vesicles. It was observed that palmitate transport occurs across sarcolemmal membranes in skeletal muscles. Also, red muscles not only contain large amount of LCFA transporters, its giant vesicles also provide greater transport than white muscles.

Abstract

The interaction between fatty acid oxidation and deposition with the muscle lipid oxidation was analyzed by pulse studies of incubated epitrochlearis, flexor digitorum and soleus muscles. Analysis of muscle lipid oxidation indicated the constant incorporation of pamitate into triacylglycerols (TGs) in skeletal muscles during lipid oxidation. Furthermore, the soleus muscle exhibited the highest degree of lipid potential while the flexor digitorum muscle was the least oxidative muscle. However, the incorporation of palmitate into TGs was highest in the flexor digitorum longus muscle.

Abstract

An analysis of the role of highly concentrated plasma free fatty acids during fat-carbohydrate interaction in skeletal muscle revealed the regulatory function of muscle glycogen phosphorylase during aerobic glucose fatty acid cycle. Measurement of muscle citrate, acetyl-CoA and pyruvate dehydrogenase activities enabled a comparison between experimentally analyzed fat-carbohydrate interaction and the theoretically postulated classic fatty acid cycle. Classic fatty acid suggested the role of muscle citrate and acetyl-CoA in attenuating glycogenolysis in the intralipid trial, which was contradicted by experimental observations.

Abstract

High-fat diet induced loss of adiponectin response occurs before development of insulin resistance in rodent skeletal muscle. Whether recovery of skeletal muscle adiponectin response is necessary for the exercise-induced recovery of insulin response is not known. Here, high-fat-fed insulin/adiponectin resistant rodents were treadmill trained or [beta]-GPA-supplemented (to reduce high-energy phosphagen stores) to determine if recovered adiponectin response corresponded with recovery of insulin response. Responses were assessed ' ex vivo' in solei via insulin-stimulated glucose uptake and adiponectin-stimulated palmitate oxidation. Insulin response was impaired following four weeks of high-fat feeding, but improved following one and two weeks of exercise, corresponding to partially restored Akt phosphorylation. Conversely, adiponectin response was not restored with exercise. Similar trends were observed following [beta]-GPA supplementation, suggesting that high-energy phosphagen perturbation suffices to recover skeletal muscle insulin response, but not adiponectin response. We conclude that the exercise-induced recovery of insulin response after high-fat feeding is not dependent on adiponectin response.

Abstract

This thesis is an investigation of the effects of recombinant resistin on glucose transport (GT) and fatty acid (FA) metabolism in mature rodent skeletal muscle. The purpose of the current study was to (1) verify that resistin impairs GT, (2) determine whether these effects are dependent on FA availability (ie. 0 (FF: fat free) vs. 2 mM (HF: high fat) palmitate) and, (3) determine resistin's effect on resting FA metabolism, using an isolated rat soleus muscle model. We also examined if (1) resistin impairs insulin signalling transduction at the level of Akt and AS160 phosphorylation and, (2) whether this inhibition can be improved by the addition of (a) AICAR, an activator of AMPK and FA oxidation, or (b) myriocin (M) and fumonisin B1 (F), inhibitors of ceramide synthesis. From our results, resistin impaired insulin-stimulated GT, but only in the presence of HF which coincided with impairments of Akt-ser473 and thr-308 phosphorylation. The addition of AICAR F and M were able to improve this impairment. Surprisingly, no effect of resistin was observed in FA metabolism per se. We conclude that resistin's mechanism of action in the impairment of GT and insulin-signalling with HF may involve early inhibition of AMPK or increased intramuscular accumulation of ceramides.
