Abstract Excessive dietary fat intake increases plasma lipid levels and has been associated with reduced bone mineral density (BMD) and increased risk of osteoporotic fracture, especially in older postmenopausal women. The objective of this study was to investigate whether there are sex-related differences in lipid metabolism that could have an impact on large-scale bone regeneration. Because ribs provide a unique exception as the only bones capable of completely regenerating large-scale defects, we used a rib resection mouse model in which human features are recapitulated. After 10 days of exposure to a low-fat diet or high-fat diet (HFD), we performed large-scale rib resection surgeries on male and female mice (6–7 weeks old) with deletion of the low-density lipoprotein (LDL) receptor (Ldlr−/−) and age- and sex-matched wild-type (WT) mice were used as controls. Plasma analysis showed that short-term exposure to HFD significantly increases total cholesterol, LDL cholesterol, and triglycerides levels in Ldlr−/− mice but not in WT, with no differences between males and females. However, under HFD, callus bone volume was significantly reduced exclusively in male Ldlr−/− mice when compared to WT, although these differences were no longer apparent by 21 days after resection. Regardless of diet or genotype, BMD of regenerated ribs did not differ significantly between groups, although male mice typically had lower average BMD values. Together, these results suggest that short-term hyperlipidemia has transient effects on large-scale bone regeneration exclusively in male mice.

Abstract Background Inclusion bodies (IBs) are well-known subcellular structures in bacteria where protein aggregates are collected. Various methods have probed their structure, but single-cell spectroscopy remains challenging. Atomic Force Microscopy-based Infrared Spectroscopy (AFM-IR) is a novel technology with high potential for the characterisation of biomaterials such as IBs. Results We present a detailed investigation using AFM-IR, revealing the substructure of IBs and their variation at the single-cell level, including a rigorous optimisation of data collection parameters and addressing issues such as laser power, pulse frequency, and sample drift. An analysis pipeline was developed tailored to AFM-IR image data, allowing high-throughput, label-free imaging of more than 3500 IBs in 12,000 bacterial cells. We examined IBs generated in Escherichia coli under different stress conditions. Dimensionality reduction analysis of the resulting spectra suggested distinct clustering of stress conditions, aligning with the nature and severity of the applied stresses. Correlation analyses revealed intricate relationships between the physical and morphological properties of IBs. Conclusions Our study highlights the power and limitations of AFM-IR, revealing structural heterogeneity within and between IBs. We show that it is possible to perform quantitative analyses of AFM-IR maps over a large collection of different samples and determine how to control for various technical artefacts.

BackgroundGenome-wide studies of gene-environment interactions (G×E) may identify variants associated with disease risk in conjunction with lifestyle/environmental exposures. We conducted a genome-wide G×E analysis of ~ 7.6 million common variants and seven lifestyle/environmental risk factors for breast cancer risk overall and for estrogen receptor positive (ER +) breast cancer.MethodsAnalyses were conducted using 72,285 breast cancer cases and 80,354 controls of European ancestry from the Breast Cancer Association Consortium. Gene-environment interactions were evaluated using standard unconditional logistic regression models and likelihood ratio tests for breast cancer risk overall and for ER + breast cancer. The Upper Rhine Graben (URG), located in France and Germany, is bordered by north–south-trending faults, some of which are considered active, posing a potential threat to the dense population and infrastructures on the Alsace plain. The largest historical earthquake in the region was the M6.5±0.5 Basel earthquake in 1356. Current seismicity (M>2.5 since 1960) is mostly diffuse and located within the graben. We build upon previous seismic hazard studies of the URG by exploring uncertainties in greater detail and revisiting a number of assumptions. We first take into account the limited evidence of neotectonic activity and then explore tectonic scenarios that have not been taken into account previously, exploring uncertainties for Mmax, its recurrence time, the b value, and the moment released aseismically or through aftershocks. Uncertainties in faults' moment deficit rates, on the observed seismic events' magnitude–frequency distribution and on the moment–area scaling law of earthquakes, are also explored. Assuming a purely dip-slip normal faulting mechanism associated with a simplified model with three main faults, Mmax maximum probability is estimated at Mw 6.1. Considering this scenario, there would be a 99 % probability that Mmax is less than 7.3. In contrast, with a strike-slip assumption associated with a four-main-fault model, consistent with recent paleoseismological studies and the present-day stress field, Mmax is estimated at Mw 6.8. Based on this scenario, there would be a 99 % probability that Mmax is less than 7.6.

Digital adherence technologies are increasingly used to support tuberculosis (TB) treatment adherence. Using microcosting, we estimated healthcare system costs (in 2022 US dollars) of 2 digital adherence technologies, 99DOTS medication sleeves and video-observed therapy (VOT), implemented in demonstration projects during 2018–2021. We also obtained cost estimates for standard directly observed therapy (DOT). Estimated per-person costs of 99DOTS for drug-sensitive TB were $98 in Bangladesh (n = 719), $119 in the Philippines (n = 396), and $174 in Tanzania (n = 976). Estimated per-person costs of VOT were $1,154 in Haiti (87 drug-sensitive), $304 in Moldova (173 drug-sensitive), $452 in Moldova (135 drug-resistant), and $661 in the Philippines (110 drug-resistant). 99DOTS costs may be similar to or less expensive than standard DOT. VOT is more expensive, although in some settings, labor cost offsets or economies of scale may yield savings. 99DOTS and VOT may yield savings to local programs if donors cover infrastructure costs.

Wildfires are a critical driver of landscape transformation on Earth, representing a dynamic and ephemeral process that poses challenges for accurate early detection. To address this challenge, researchers have increasingly turned to deep learning techniques, which have demonstrated remarkable potential in enhancing the performance of wildfire detection systems. This paper provides a comprehensive review of fire detection using deep learning, spanning from 1990 to 2023. This study employed a comprehensive approach, combining bibliometric analysis, qualitative and quantitative methods, and systematic review techniques to examine the advancements in fire detection using deep learning in remote sensing. It unveils key trends in publication patterns, author collaborations, and thematic focuses, emphasizing the remarkable growth in fire detection using deep learning in remote sensing (FDDL) research, especially from the 2010s onward, fueled by advancements in computational power and remote sensing technologies. The review identifies “Remote Sensing” as the primary platform for FDDL research dissemination and highlights the field’s collaborative nature, with an average of 5.02 authors per paper. The co-occurrence network analysis reveals diverse research themes, spanning technical approaches and practical applications, with significant contributions from China, the United States, South Korea, Brazil, and Australia. Highly cited papers are explored, revealing their substantial influence on the field’s research focus. The analysis underscores the practical implications of integrating high-quality input data and advanced deep-learning techniques with remote sensing for effective fire detection. It provides actionable recommendations for future research, emphasizing interdisciplinary and international collaboration to propel FDDL technologies and applications. The study’s conclusions highlight the growing significance of FDDL technologies and the necessity for ongoing advancements in computational and remote sensing methodologies. The practical takeaway is clear: future research should prioritize enhancing the synergy between deep learning techniques and remote sensing technologies to develop more efficient and accurate fire detection systems, ultimately fostering groundbreaking innovations.

Takotsubo or stress cardiomyopathy is a syndrome of transient left ventricular systolic dysfunction seen in the absence of obstructive coronary artery disease. We describe a case of stress cardiomyopathy diagnosed in the emergency department (ED) using point of care ultrasound (POCUS) associated with traumatic hand amputation.  The patient suffered a near-complete amputation of the right hand while using a circular saw, subsequently complicated by brief cardiac arrest with rapid return of spontaneous circulation. Point-of-care ultrasonography in the ED revealed the classic findings of takotsubo cardiomyopathy, including apical ballooning of the left ventricle and hyperkinesis of the basal walls with a severely reduced ejection fraction. After formalization of the amputation and cardiovascular evaluation, the patient was discharged from the hospital in stable condition ten days later. Emergency physicians should be aware of the possibility of stress cardiomyopathy as a cause for acute decompensation, even in isolated extremity trauma.

El vertiginoso desarrollo de la revolución científico técnica y su incidencia en la esfera educacional exigen de la superación continúa de los directores de las instituciones educativas de la educación especial, pues a ellos les corresponde dirigir científicamente los procesos educativos, asesorar a docentes, familias y coordinar con los agentes socioeducativos de la red escolar para lograr en los educandos, el desarrollo de habilidades prácticas - sociales y un aprendizaje de calidad para toda la vida, de manera que egresen aptos y capaces de interactuar adecuadamente en su entorno socio familiar. De ahí, la necesidad de implementar acciones orientadas a la superación profesional del director. El artículo expone una estrategia pedagógica de superación dirigida a transformar los modos de actuación de los directivos de la educación especial para direccionar el desarrollo de habilidades de autodeterminación en los educandos en situación de discapacidad intelectual. Fueron empleados métodos del nivel teórico y empírico para la identificación y solución de las necesidades de superación de los directivos. La novedad de la estrategia radica en la concepción articulada de las diferentes formas de superación, establecidas en la Educación de Avanzada, desde el sistema de trabajo del municipio, con un carácter sistémico, planificado y consiente. Con su implementación se perfeccionó el desempeño profesional y personal de los directivos y los educandos quienes egresaron con el empoderamiento de habilidades para la toma de decisiones, la realización de elecciones, la resolución de problemas y el establecimiento de metas y objetivos.

IntroductionInterferon-gamma (IFN-γ) is pivotal in orchestrating immune responses during healthy pregnancy. However, its dysregulation, often due to autoimmunity, infections, or chronic inflammatory conditions, is implicated in adverse reproductive outcomes such as pregnancy failure or infertility. Additionally, the underlying immunological mechanisms remain elusive.MethodsHere, we explore the impact of systemic IFN-γ elevation on cytotoxic T cell responses in female reproduction utilizing a systemic lupus-prone mouse model with impaired IFN-γ degradation.ResultsOur findings reveal that heightened IFN-γ levels triggered the infiltration of CD8+T cells in the pituitary gland and female reproductive tract (FRT), resulting in prolactin deficiency and subsequent infertility. Furthermore, we demonstrate that chronic IFN-γ elevation increases effector memory CD8+T cells in the murine ovary and uterus.DiscussionThese insights broaden our understanding of the role of elevated IFN-γ in female reproductive dysfunction and suggest CD8+T cells as potential immunotherapeutic targets in female reproductive disorders associated with chronic systemic IFN-γ elevation.

Dryland regions around the world are facing intricate challenges due to climate change and human activities. The Caatinga biome in Brazil, an exceptional dryland ecosystem covering approximately 86.3 million hectares, is particularly impacted by human influence. We conducted an extensive study analyzing changes in land use and land cover within the Caatinga region over a span of 35 years, from 1985 to 2019. This study leverages collective knowledge and collaborative effort with the MapBiomas project to provide valuable insights into the biome’s landscape. It maps eight principal land cover classes using Landsat Collection 1 Tier 1 data normalized to top-of-atmosphere reflectance. All data processing was carried out within the Google Earth Engine platform, and the graphics were generate using R version 3.6.2. This study achieved an impressive 80% global accuracy in the time series of Caatinga land use and land cover (LULC) changes, with allocation and area discrepancies of 11.6% and 8.5%, respectively. The extensive 35-year LULC dataset reveals a substantial 11% reduction in natural vegetation in the Caatinga biome, translating to a loss of 6.57 million hectares. This decline is primarily attributed to the expansion of cattle ranching and agriculture; all types of natural vegetation have experienced decreases, with Savanna Formation (SF) areas declining by 11% and Forest Formation (FF) areas declining by 8%. In contrast, pasturelands expanded by 62% and agricultural land expanded by 284% during this period. With their urgent and significant real-world for informing social, economic, and environmental policy decisions within the Caatinga and other dryland regions globally, these findings underscore the importance and immediacy of our research.

Abstract Introduction Prostate cancer (PCa) is by far the most common type of cancer among men in western countries. However, relatively little is known about its etiology despite the high morbidity and mortality. It has been suggested that chronic inflammation may be involved in prostate carcinogenesis. We investigated the role of sexually and non‐sexually transmitted infections in prostate cancer risk with a specific interest in the aggressive types. Methods We used data from epidemiological study of prostate cancer (EPICAP), a population‐based case–control study. A total of 819 incident cases and 879 controls were interviewed face‐to‐face using a standardized questionnaire gathering information on known or suspected risk factors of prostate cancer and personal history of specific sexually and non‐sexually transmitted infections: gonorrhea, syphilis, trichomonas, herpes, mononucleosis, Epstein–Barr virus, varicella‐zoster, and dengue. Odds ratios (OR) and their 95% confidence interval were estimated using multivariate unconditional logistic regression. Results There was no significant association between gonorrhea (OR: 0.90, 95% CI: 0.61–1.33), trichomonas (OR: 0.74, 95% CI: 0.27–2.07), genital herpes (OR: 0.69, 95% CI: 0.38–1.27), and the risk of prostate cancer. No association emerged for overall sexually transmitted bacterial and viral infections (OR 1.05, 95% CI: 0.86–1.29) and overall non‐sexually transmitted viral infections (OR 1.11, 95% CI: 0.90–1.35) and the risk of prostate cancer. Conclusion Our results showed that sexually or non‐sexually transmitted infections, either bacterial or viral, were not associated to prostate cancer. Therefore, further investigation is needed to help advance our understanding of the role of chronic inflammation in the etiology of prostate cancer, with a particular focus on its most aggressive types.

La superación continua de los profesores de Educación Física constituye una necesidad que ha transcendido diferentes generaciones de docentes, determinada por el desarrollo creciente de la ciencia y la técnica, las exigencias del tercer perfeccionamiento educacional cubano, las demandas en la preparación y capacitación del profesorado y la necesidad de su incidencia en la formación de un egresado apto y capaz de desarrollar no solo habilidades y capacidades físicas, sino también para la vida, que les permitan actuar en el medio social, familiar y laboral. El objetivo de esta investigación consistió en diseñar una estrategia pedagógica de superación para transformar los modos de actuación de los profesores de Educación Física, en la dirección del proceso de enseñanza-aprendizaje, en función del desarrollo de habilidades de autodeterminación en los educandos en situación de discapacidad intelectual. Los métodos investigativos utilizados en el orden teórico y empírico fueron el análisis y síntesis, el histórico-lógico, el inductivo-deductivo, la modelación y el sistémico-estructural-funcional; además de la entrevista, la observación, la encuesta, el análisis de documentos, el criterio de expertos y la triangulación múltiple, los que permitieron revelar las causales que inciden en el normal desarrollo de las habilidades de autodeterminación, así como elaborar la estrategia. Con su implementación se perfeccionó el desempeño profesional y personal de los profesores y los educandos, quienes egresaron con el empoderamiento de habilidades para la toma de decisiones, la realización de elecciones, la resolución de problemas y el establecimiento de metas y objetivos.

BackgroundGenetic testing for breast cancer susceptibility is widely used, but for many genes, evidence of an association with breast cancer is weak, underlying risk estimates are imprecise, and reliable subtype-specific risk estimates are lacking.MethodsWe used a panel of 34 putative susceptibility genes to perform sequencing on samples from 60,466 women with breast cancer and 53,461 controls. In separate analyses for protein-truncating variants and rare missense variants in these genes, we estimated odds ratios for breast cancer overall and tumor subtypes. We evaluated missense-variant associations according to domain and classification of pathogenicity.ResultsProtein-truncating variants in 5 genes (ATM, BRCA1, BRCA2, CHEK2, and PALB2) were associated with a risk of breast cancer overall with a P value of less than 0.0001. Protein-truncating variants in 4 other genes (BARD1, RAD51C, RAD51D, and TP53) were associated with a risk of breast cancer overall with a P value of less than 0.05 and a Bayesian false-discovery probability of less than 0.05. For protein-truncating variants in 19 of the remaining 25 genes, the upper limit of the 95% confidence interval of the odds ratio for breast cancer overall was less than 2.0. For protein-truncating variants in ATM and CHEK2, odds ratios were higher for estrogen receptor (ER)-positive disease than for ER-negative disease; for protein-truncating variants in BARD1, BRCA1, BRCA2, PALB2, RAD51C, and RAD51D, odds ratios were higher for ER-negative disease than for ER-positive disease. Rare missense variants (in aggregate) in ATM, CHEK2, and TP53 were associated with a risk of breast cancer overall with a P value of less than 0.001. For BRCA1, BRCA2, and TP53, missense variants (in aggregate) that would be classified as pathogenic according to standard criteria were associated with a risk of breast cancer overall, with the risk being similar to that of protein-truncating variants.ConclusionsThe results of this study define the genes that are most clinically useful for inclusion on panels for the prediction of breast cancer risk, as well as provide estimates of the risks associated with protein-truncating variants, to guide genetic counseling. BackgroundPrevious gene-environment interaction studies of breast cancer risk have provided sparse evidence of interactions. Using the largest available dataset to date, we performed a comprehensive assessment of potential effect modification of 205 common susceptibility variants by 13 established breast cancer risk factors, including replication of previously reported interactions.MethodsAnalyses were performed using 28 176 cases and 32 209 controls genotyped with iCOGS array and 44 109 cases and 48 145 controls genotyped using OncoArray from the Breast Cancer Association Consortium (BCAC). Gene-environment interactions were assessed using unconditional logistic regression and likelihood ratio tests for breast cancer risk overall and by estrogen-receptor (ER) status. Bayesian false discovery probability was used to assess the noteworthiness of the meta-analysed array-specific interactions.ResultsNoteworthy evidence of interaction at ≤1% prior probability was observed for three single nucleotide polymorphism (SNP)-risk factor pairs. SNP rs4442975 was associated with a greater reduction of risk of ER-positive breast cancer [odds ratio (OR)int = 0.85 (0.78-0.93), Pint = 2.8 x 10-4] and overall breast cancer [ORint = 0.85 (0.78-0.92), Pint = 7.4 x 10-5) in current users of estrogen-progesterone therapy compared with non-users. This finding was supported by replication using OncoArray data of the previously reported interaction between rs13387042 (r2 = 0.93 with rs4442975) and current estrogen-progesterone therapy for overall disease (Pint = 0.004). The two other interactions suggested stronger associations between SNP rs6596100 and ER-negative breast cancer with increasing parity and younger age at first birth.ConclusionsOverall, our study does not suggest strong effect modification of common breast cancer susceptibility variants by established risk factors.

Conglomerate formation, where enantiomers within a racemic mixture self-segregate upon crystallization, is an advantageous property for obtaining chirally pure crystals and allows large-scale chiral resolution. However, the prevalence of conglomerates is low and difficult to predict. In this report, we describe our attempts to engineer conglomerates from racemate-forming compounds by integrating them into a conglomerate-forming matrix. In this regard, we found that Ni(II) and Fe(II) form molecular alloys with Zn(II) in [MxZn(1−x)(bpy)3](PF6)2 (where bpy = 2,2′-bipyridyl). Powder X-ray Diffraction (PXRD) and Energy-Dispersive X-ray spectroscopy (EDX) evidenced conglomerate crystallization with Ni(II) concentrations up to about 25%, while it was observed only for much lower concentrations of Fe(II). This can be attributed to the ability of [Ni(bpy)3](PF6)2 to access a metastable conglomerate phase, while no such phase has been detected in [Fe(bpy)3](PF6)2. Furthermore, the chiral phase appears to be favored in fast-growing precipitates, while the racemic phase is favored in slow re-crystallizations for both Ni(II) and Fe(II) molecular alloys. X-ray natural circular dichroism (XNCD) measurements on [Ni0.13Zn0.87(bpy)3](PF6)2 demonstrate the chirality of the nickel molecules within the zinc molecular matrix.
