1. Living porous ceramics for bacteria-regulated gas sensing and carbon capture

Abstract

Microorganisms hosted in abiotic structures have led to engineered living materials that can grow, sense and adapt in ways that mimic biological systems. Although porous structures should favor colonization by microorganisms, they have not yet been exploited as abiotic scaffolds for the development of living materials. Here, we report porous ceramics that are colonized by bacteria to form an engineered living material with self-regulated and genetically programmable carbon capture and gas-sensing functionalities. The carbon capture capability is achieved using wild-type photosynthetic cyanobacteria, whereas the gas-sensing function is generated utilizing genetically engineered E. coli. Hierarchical porous clay is used as ceramic scaffold and evaluated in terms of bacterial growth, water uptake and mechanical properties. Using state-of-the-art chemical analysis techniques, we demonstrate the ability of the living porous ceramics to capture CO2 directly from the air and to metabolically turn minute amounts of a toxic gas into a benign scent detectable by humans.

Abstract

Visual Place Recognition (VPR) aims to estimate the location of an image by treating it as a retrieval problem. VPR uses a database of geo-tagged images and leverages deep neural networks to extract a global representation, called descriptor, from each image. While the training data for VPR models often originates from diverse, geographically scattered sources (geo-tagged images), the training process itself is typically assumed to be centralized. This research revisits the task of VPR through the lens of Federated Learning (FL), addressing several key challenges associated with this adaptation. VPR data inherently lacks well-defined classes, and models are typically trained using contrastive learning, which necessitates a data mining step on a centralized database. Additionally, client devices in federated systems can be highly heterogeneous in terms of their processing capabilities. Abstract

The classical Pell equation can be extended to the cubic case considering the elements of norm one in $Z[\sqrt[3]{r}]$, which satisfy $x^3 + r y^3 + r^2 z^3 - 3 r x y z = 1$. The solution of the cubic Pell equation is harder than the classical case, indeed a method for solving it as Diophantine equation is still missing. In this paper, we study the cubic Pell equation over finite fields, extending the results that hold for the classical one. In particular, we provide a novel method for counting the number of solutions in all possible cases depending on the value of r. Moreover, we are also able to provide a method for generating all the solutions.

Abstract Background Risk of biochemical recurrence (BCR) in localised prostate cancer can be stratified using the 5‐tier Cambridge Prognostic Group (CPG) or 3‐tier European Association of Urology (EAU) model. Active surveillance is the current recommendation if CPG1 or EAU low‐risk criteria are met. We aimed to determine the contemporary rates of upgrading, upstaging and BCR after radical prostatectomy for CPG1 or EAU low‐risk disease. Methods A database of all robotic‐assisted laparoscopic prostatectomies (RALPs) performed in Glasgow between 12/2015 and 05/2022 was analysed. Rates of upgrading, upstaging and BCR post‐RALP for CPG1 or EAU low‐risk disease were defined. Univariate and multivariate analysis were performed to assess the relationship between patient factors and outcomes. Results A total of 1223 RALP cases were identified. A total of 12.6% met CPG1 criteria with 70.1% and 25.3% upgraded and upstaged to extraprostatic disease post‐operatively respectively. Abstract

We present a study on the use of Pell hyperbolas in cryptosystems with security based on the discrete logarithm problem. Specifically, after introducing the group's structure over generalized Pell conics (and also giving the explicit isomorphisms with the classical Pell hyperbolas), we provide a parameterization with both an algebraic and a geometrical approach. The particular parameterization that we propose appears to be useful from a cryptographic point of view because the product that arises over the set of parameters is connected to the R\'edei rational functions, which can be evaluated in a fast way. Thus, we exploit these constructions for defining three different public key cryptosystems based on the ElGamal scheme. Abstract. Background. To evaluate the predictive values of Prostate Imaging Reporting and Data System version 2 (PI-RADS v2), prostate-specific antigen (PSA) level, PSA density (PSAD), digital rectal examination findings, and prostate volume, individually and in combination, for the detection of prostate cancer (PCa) in biopsy-naive patients. Methods. We retrospectively analyzed 630 patients who underwent transrectal systematic prostate biopsy following prostate multiparametric magnetic resonance imaging. A standard 12-core biopsy procedure was performed. Univariate and multivariate analyses were performed to determine the significant predictors of clinically significant cancer but not PCa. Results. The median age, PSA level, and PSAD were 70 years, 8.6 ng/mL, and 0.18 ng/mL/mL, respectively. A total of 374 (59.4%) of 630 patients were biopsy-positive for PCa, and 241 (64.4%) of 374 were diagnosed with clinically significant PCa (csPCa). Abstract

We study new primality tests based on linear recurrent sequences of degree two exploiting a matricial approach. The classical Lucas test arises as a particular case and we see how it can be easily improved. Moreover, this approach shows clearly how the Lucas pseudoprimes are connected to the Pell equation and the Brahamagupta product. We also introduce a new specific primality test, which we will call generalized Pell test. We perform some numerical computations on the new primality tests and, for the generalized Pell test, we do not any pseudoprime up to $10^{10}$.

Abstract

Biological cells are able to generate intricate structures and respond to external stimuli, sculpting their membrane from within. Simplified biomimetic systems can aid in understanding the principles which govern these shape changes and elucidate the response of the cell membrane under strong deformations. Here, a combined experimental and simulation approach is used to identify the conditions under which different non-equilibrium shapes and distinct active shape fluctuations can be obtained by enclosing self-propelled particles in giant vesicles. Interestingly, the most pronounced shape changes are observed at relatively low particle loadings, starting with the formation of tether-like protrusions to highly branched, dendritic structures. At high volume fractions, globally deformed vesicle shapes are observed. Abstract

ObjectiveThis study was undertaken to assess the impact of immunosuppressive and immunomodulatory therapies on the severity of coronavirus disease 2019 (COVID-19) in people with multiple sclerosis (PwMS).MethodsWe retrospectively collected data of PwMS with suspected or confirmed COVID-19. All the patients had complete follow-up to death or recovery. Severe COVID-19 was defined by a 3-level variable: mild disease not requiring hospitalization versus pneumonia or hospitalization versus intensive care unit (ICU) admission or death. We evaluated baseline characteristics and MS therapies associated with severe COVID-19 by multivariate and propensity score (PS)-weighted ordinal logistic models. Sensitivity analyses were run to confirm the results.ResultsOf 844 PwMS with suspected (n = 565) or confirmed (n = 279) COVID-19, 13 (1.54%) died; 11 of them were in a progressive MS phase, and 8 were without any therapy. Thirty-eight (4.5%) were admitted to an ICU; 99 (11.7%) had radiologically documented pneumonia; 96 (11.4%) were hospitalized. After adjusting for region, age, sex, progressive MS course, Expanded Disability Status Scale, disease duration, body mass index, comorbidities, and recent methylprednisolone use, therapy with an anti-CD20 agent (ocrelizumab or rituximab) was significantly associated (odds ratio [OR] = 2.37, 95% confidence interval [CI] = 1.18-4.74, p = 0.015) with increased risk of severe COVID-19. Abstract

This work examines cellular immunity against SARS-CoV-2 in patients from Córdoba, Argentina, during two major waves characterized by different circulating viral variants and different social behavior. Using flow cytometry, we evaluated the main lymphocyte populations of peripheral blood from hospitalized patients with moderate and severe COVID-19 disease. Our results show disturbances in the cellular immune compartment, as previously reported in different cohorts worldwide. We observed an increased frequency of B cells and a significant decrease in the frequency of CD3+ T cells in COVID-19 patients compared to healthy donors (HD). We also found a reduction in Tregs, which was more pronounced in severe patients. During the first wave, the frequency of GZMB, CD107a, CD39, and PD-1-expressing conventional CD4+ T (T conv) cells was significantly higher in moderate and severe patients than in HD. During the second wave, only the GZMB+ T conv cells of moderate and severe patients increased significantly. In addition, these patients showed a decreased frequency in IL-2-producing T conv cells. Interestingly, we identified two subsets of circulating CD8+ T cells with low and high CD8 surface expression in both HD and COVID-19 patients. While the percentages of CD8hi and CD8lo T cells within the CD8+ population in HD are similar, a significant increase was observed in CD8lo T cell frequency in COVID-19 patients. CD8lo T cell populations from HD as well as from SARS-CoV-2 infected patients exhibited lower frequencies of the effector cytokine-producing cells, TNF, IL-2, and IFN-γ, than CD8hi T cells. Interestingly, the frequency of CD8lo T cells increased with disease severity, suggesting that this parameter could be a potential marker for disease progression. Indeed, the CD8hi/CD8lo index helped to significantly improve the patient’s clinical stratification and disease outcome prediction. Our data support the addition of, at least, a CD8hi/CD8lo index into the panel of biomarkers commonly used in clinical labs, since its determination may be a useful tool with impact on the therapeutic management of the patients.

Mario Ridolfi uses drawing to explore constructive themes and also as a hallmark of his personal poetics. The point of convergence between technique and poetics is manifested, in particular, in the use of two tools: the abacus and the node. The abacus concerns the combinatorial variations between quantitative parameters affecting buildings and urban design. The node establishes analogical relationships between building elements of different sizes. The abacus and the node are multiscalar tools that merge architectural design and technical investigation.

Abstract

Abstract Background Robotic radical prostatectomy (RARP) is a first-line curative treatment option for localized prostate cancer. Postoperative erectile dysfunction and urinary incontinence are common associated adverse side effects that can negatively impact patients’ quality of life. Preserving the lateral neurovascular bundles (NS) during RARP improves functional outcomes. However, selecting men for NS may be difficult when there is concern about incurring in positive surgical margin (PSM) which in turn risks adverse oncological outcomes. The NeuroSAFE technique (intra-operative frozen section examination of the neurovascular structure adjacent prostate margin) can provide real-time pathological consult to promote optimal NS whilst avoiding PSM. Methods NeuroSAFE PROOF is a single-blinded, multi-centre, randomised controlled trial (RCT) in which men are randomly allocated 1:1 to either NeuroSAFE RARP or standard RARP. Men electing for RARP as primary treatment, who are continent and have good baseline erectile function (EF), defined by International Index of Erectile Function (IIEF-5) score > 21, are eligible. NS in the intervention arm is guided by the NeuroSAFE technique. NS in the standard arm is based on standard of care, i.e. a pre-operative image-based planning meeting, patient-specific clinical information, and digital rectal examination. The primary outcome is assessment of EF at 12 months. The primary endpoint is the proportion of men who achieve IIEF-5 score ≥ 21. A sample size of 404 was calculated to give a power of 90% to detect a difference of 14% between groups based on a feasibility study. Oncological outcomes are continuously monitored by an independent Data Monitoring Committee. Key secondary outcomes include urinary continence at 3 months assessed by the international consultation on incontinence questionnaire, rate of biochemical recurrence, EF recovery at 24 months, and difference in quality of life. Discussion NeuroSAFE PROOF is the first RCT of intra-operative frozen section during radical prostatectomy in the world. It is properly powered to evaluate a difference in the recovery of EF for men undergoing RARP assessed by patient-reported outcome measures. It will provide evidence to guide the use of the NeuroSAFE technique around the world. Trial registration NCT03317990 (23 October 2017). Regional Ethics Committee; reference 17/LO/1978.

Abstract

BackgroundCOVID-19 severity has been linked to an increased production of inflammatory mediators called “cytokine storm”. Available data is mainly restricted to the first international outbreak and reports highly variable results. This study compares demographic and clinical features of patients with COVID-19 from Córdoba, Argentina, during the first two waves of the pandemic and analyzes association between comorbidities and disease outcome with the “cytokine storm”, offering added value to the field.MethodsWe investigated serum concentration of thirteen soluble mediators, including cytokines and chemokines, in hospitalized patients with moderate and severe COVID-19, without previous rheumatic and autoimmune diseases, from the central region of Argentina during the first and second infection waves. Samples from healthy controls were also assayed. Clinical and biochemical parameters were collected.ResultsComparison between the two first COVID-19 waves in Argentina highlighted that patients recruited during the second wave were younger and showed less concurrent comorbidities than those from the first outbreak. We also recognized particularities in the signatures of systemic cytokines and chemokines in patients from both infection waves. We determined that concurrent pre-existing comorbidities did not have contribution to serum concentration of systemic cytokines and chemokines in COVID-19 patients. We also identified immunological and biochemical parameters associated to inflammation which can be used as prognostic markers. Thus, IL-6 concentration, C reactive protein level and platelet count allowed to discriminate between death and discharge in patients hospitalized with severe COVID-19 only during the first but not the second wave.ConclusionsOur data provide information that deepens our understanding of COVID-19 pathogenesis linking demographic features of a COVID-19 cohort with cytokines and chemokines systemic concentration, presence of comorbidities and different disease outcomes. Altogether, our findings provide information not only at local level by delineating inflammatory/anti-inflammatory response of patients but also at international level addressing the impact of comorbidities and the infection wave in the variability of cytokine and chemokine production upon SARS-CoV-2 infection.
