The Marine Ice Sheet–Ocean Model Intercomparison Project – phase 2 (MISOMIP2) is a natural progression of previous and ongoing model intercomparison exercises that have focused on the simulation of ice-sheet and ocean processes in Antarctica. The previous exercises motivate the move towards realistic configurations, as well as more diverse model parameters and resolutions. The main objective of MISOMIP2 is to investigate the performance of existing ocean and coupled ice-sheet–ocean models in a range of Antarctic environments through comparisons to observational data. We will assess the status of ice-sheet–ocean modelling as a community and identify common characteristics of models that are best able to capture observed features. As models are highly tuned based on present-day data, we will also compare their sensitivity to prescribed abrupt atmospheric perturbations leading to either very warm or slightly warmer ocean conditions compared to the present day. The approach of MISOMIP2 is to welcome contributions of models as they are, including global and regional configurations, but we request standardized variables and common grids for the outputs. We target the analysis at two specific regions, the Amundsen Sea and the Weddell Sea, since they describe two different ocean environments and have been relatively well observed compared to other areas of Antarctica. An observational “MIPkit” synthesizing existing ocean and ice-sheet observations for a common period is provided to evaluate ocean and ice-sheet models in these two regions.

Abstract We present an exploration of the unpolarized isovector proton generalized parton distributions (GPDs) H u−d (x, ξ, t) and E u−d (x, ξ, t) in the pseudo-distribution formalism using distillation. Taking advantage of the large kinematic coverage made possible by this approach, we present results on the moments of GPDs up to the order x 3 — including their skewness dependence — at a pion mass m π = 358 MeV and a lattice spacing a = 0.094 fm.

The Pine Island and Thwaites glaciers are the two largest contributors to sea level rise from Antarctica. Here we examine the influence of basal friction and ice shelf basal melt in determining projected losses. We examine both Weertman and Coulomb friction laws with explicit weakening as the ice thins to flotation, which many friction laws include implicitly via the effective pressure. We find relatively small differences with the choice of friction law (Weertman or Coulomb) but find losses to be highly sensitive to the rate at which the basal traction is reduced as the area upstream of the grounding line thins. Consistent with earlier work on Pine Island Glacier, we find sea level contributions from both glaciers to vary linearly with the melt volume averaged over time and space, with little influence from the spatial or temporal distribution of melt. Based on recent estimates of melt from other studies, our simulations suggest that the combined melt-driven and sea level rise contribution from both glaciers may not exceed 10 cm by 2200, although the uncertainty in model parameters allows for larger increases. We do not include other factors, such as ice shelf breakup, that might increase loss, or factors such as increased accumulation and isostatic uplift that may mitigate loss.

Abstract Lattice QCD offers the possibility of computing parton distributions from first principles, although not in the usual MS ¯ $$ \overline{MS} $$ factorization scheme. Calculations are therefore matched to MS ¯ $$ \overline{MS} $$ using a perturbative procedure which is the source of significant uncertainty within the currently accessible kinematics. We present the possibility of computing the z 2 evolution of non-singlet pseudo-parton distribution functions within the short factorization scheme in a numerically improvable way. The goal is to have tools to evolve a calculation to a scale where perturbative uncertainties are less pronounced. We compare a numerical extraction of the evolution operator from lattice data to the computation of z 2 dependence in perturbation theory. Finally, we discuss how this numerical work may be extended to address the two-scale problem that arises when the Ioffe time range must be made large to extend the reach of the calculation of the pseudo-PDF to smaller values of the momentum fraction.

Abstract Chronic obstructive pulmonary disease (COPD) affects over 250 million individuals globally and stands as the third leading cause of mortality. Respiratory viral infections serve as the primary drivers of acute exacerbations, hastening the decline in lung function and worsening the prognosis. Notably, Human Parainfluenza Virus type 3 (HPIV-3) is responsible for COPD exacerbations with a frequency comparable to that of Respiratory Syncytial Virus and Influenza viruses. However, the impact of HPIV-3 on respiratory epithelium within the context of COPD remains uncharacterized. In this study, we employed in vitro reconstitution of lower airway epithelia from lung tissues sourced from healthy donors (n = 4) and COPD patients (n = 5), maintained under air–liquid interface conditions. Through a next-generation sequencing-based transcriptome analysis, we compared the cellular response to HPIV-3 infection. Prior to infection, COPD respiratory epithelia exhibited a pro-inflammatory profile, notably enriched in canonical pathways linked to antiviral response, B cell signaling, IL-17 signaling, and epithelial-mesenchymal transition, in contrast to non-COPD epithelia. Intriguingly, post HPIV-3 infection, only non-COPD epithelia exhibited significant enrichment in interferon signaling, pattern recognition receptors of viruses and bacteria, and other pathways involved in antiviral responses. This deficiency could potentially hinder immune cell recruitment essential for controlling viral infections, thus fostering prolonged viral presence and persistent inflammation.

Abstract We present a systematic study demonstrating the impact of lattice QCD data on the extraction of generalised parton distributions (GPDs). For this purpose, we use a previously developed modelling of GPDs based on machine learning techniques fulfilling the theoretical requirements of polynomiality, a form of positivity constraint and known reduction limits. A special care is given to estimate the uncertainty stemming from the ill-posed character of the connection between GPDs and the experimental processes usually considered to constrain them, like deeply virtual Compton scattering (DVCS). Moke lattice QCD data inputs are included in a Bayesian framework to a prior model based on an Artificial Neural Network. This prior model is fitted to reproduce the most experimentally accessible information of a phenomenological extraction by Goloskokov and Kroll. We highlight the impact of the precision, correlation and kinematic coverage of lattice data on GPD extraction at moderate $$\xi $$ ξ which has only been brushed in the literature so far, paving the way for a joint extraction of GPDs.

Glaciers in the Amundsen Sea Embayment of West Antarctica are rapidly retreating and contributing to sea level rise. Ice loss is occurring primarily via exposure to warm ocean water, which varies in response to local wind variability. There is evidence that retreat was initiated in the mid-20th century, but the perturbation that may have triggered retreat remains unknown. A leading hypothesis is that large pressure and wind anomalies in the 1940s drove exceptionally strong oceanic ice-shelf melting. However, the characteristics, drivers, and rarity of the atmospheric event remain poorly constrained. We investigate the 1940s atmospheric event using paleoclimate reconstructions and climate model simulations. The reconstructions show that large westerly wind anomalies occurred from ∼1938–1942, a combined response to the very large El Niño event from 1940–1942 and other variability beginning years earlier. Climate model simulations provide evidence that events of similar magnitude and duration may occur tens to hundreds of times per 10 kyr of internal climate variability (∼0.2 to 2.5 occurrences per century). Our results suggest that the 1940s westerly event is unlikely to have been exceptional enough to be the sole explanation for the initiation of Amundsen Sea glacier retreat. Additional factors are likely needed to explain the onset of retreat in West Antarctica, such as naturally arising variability in ocean conditions prior to the 1940s or anthropogenically driven trends since the 1940s.

Positron beams, both polarized and unpolarized, are identified as important ingredients for the experimental programs at the next generation of lepton accelerators. In the context of the hadronic physics program at Jefferson Lab (JLab), positron beams are complementary, even essential, tools for a precise understanding of the electromagnetic structure of nucleons and nuclei, in both the elastic and deep-inelastic regimes. For instance, elastic scattering of polarized and unpolarized electrons and positrons from the nucleon enables a model independent determination of its electromagnetic form factors. Also, the deeply-virtual scattering of polarized and unpolarized electrons and positrons allows unambiguous separation of the different contributions to the cross section of the lepto-production of photons and of lepton-pairs, enabling an accurate determination of the nucleons and nuclei generalized parton distributions, and providing an access to the gravitational form factors of the energy-momentum tensor. Furthermore, positron beams offer the possibility of alternative tests of the Standard Model of particle physics through the search of a dark photon, the precise measurement of electroweak couplings, and the investigation of charged lepton flavor violation. This document discusses the perspectives of an experimental program with high duty-cycle positron beams at JLab.

Multiple myeloma (MM) is a hematological malignancy characterized by an abnormal clonal proliferation of malignant plasma cells. Despite the introduction of novel agents that have significantly improved clinical outcome, most patients relapse and develop drug resistance. MM is characterized by genomic instability and a high level of replicative stress. In response to replicative and DNA damage stress, MM cells activate various DNA damage signaling pathways. In this study, we reported that high CHK1 and WEE1 expression is associated with poor outcome in independent cohorts of MM patients treated with high dose melphalan chemotherapy or anti-CD38 immunotherapy. Combined targeting of Chk1 and Wee1 demonstrates synergistic toxicities on MM cells and was associated with higher DNA double-strand break induction, as evidenced by an increased percentage of γH2AX positive cells subsequently leading to apoptosis. The therapeutic interest of Chk1/Wee1 inhibitors’ combination was validated on primary MM cells of patients. The toxicity was specific of MM cells since normal bone marrow cells were not significantly affected. Using deconvolution approach, MM patients with high CHK1 expression exhibited a significant lower percentage of NK cells whereas patients with high WEE1 expression displayed a significant higher percentage of regulatory T cells in the bone marrow. These data emphasize that MM cell adaptation to replicative stress through Wee1 and Chk1 upregulation may decrease the activation of the cell-intrinsic innate immune response. Our study suggests that association of Chk1 and Wee1 inhibitors may represent a promising therapeutic approach in high-risk MM patients characterized by high CHK1 and WEE1 expression.

The authors combine measurements of ice loss from West Antarctica with climate modelling to show that periods of drought or extremely heavy precipitation can significantly increase or decrease rates of mass loss for periods lasting several years.

ABSTRACT Infection by retroviruses as HIV-1 requires the stable integration of their genome into the host cells. This process needs the formation of integrase (IN)-viral DNA complexes, called intasomes, and their interaction with the target DNA wrapped around nucleosomes within cell chromatin. To provide new tools to analyze this association and select drugs, we applied the AlphaLISA technology to the complex formed between the prototype foamy virus (PFV) intasome and nucleosome reconstituted on 601 Widom sequence. This system allowed us to monitor the association between both partners and select small molecules that could modulate the intasome/nucleosome association. Using this approach, drugs acting either on the DNA topology within the nucleosome or on the IN/histone tail interactions have been selected. Within these compounds, doxorubicin and histone binders calixarenes were characterized using biochemical, in silico molecular simulations and cellular approaches. These drugs were shown to inhibit both PFV and HIV-1 integration in vitro. Treatment of HIV-1-infected PBMCs with the selected molecules induces a decrease in viral infectivity and blocks the integration process. Thus, in addition to providing new information about intasome-nucleosome interaction determinants, our work also paves the way for further unedited antiviral strategies that target the final step of intasome/chromatin anchoring. IMPORTANCE In this work, we report the first monitoring of retroviral intasome/nucleosome interaction by AlphaLISA. This is the first description of the AlphaLISA application for large nucleoprotein complexes (>200 kDa) proving that this technology is suitable for molecular characterization and bimolecular inhibitor screening assays using such large complexes. Using this system, we have identified new drugs disrupting or preventing the intasome/nucleosome complex and inhibiting HIV-1 integration both in vitro and in infected cells. This first monitoring of the retroviral/intasome complex should allow the development of multiple applications including the analyses of the influence of cellular partners, the study of additional retroviral intasomes, and the determination of specific interfaces. Our work also provides the technical bases for the screening of larger libraries of drugs targeting specifically these functional nucleoprotein complexes, or additional nucleosome-partner complexes, as well as for their characterization.

In the Amundsen Sea, modified Circumpolar Deep Water (mCDW) intrudes into ice shelf cavities, causing high ice shelf melting near the ice sheet grounding lines, accelerating ice flow, and controlling the pace of future Antarctic contributions to global sea level. The pathways of mCDW towards grounding lines are crucial as they directly control the heat reaching the ice. A realistic representation of mCDW circulation, however, remains challenging due to the sparsity of in-situ observations and the difficulty of ocean models to reproduce the available observations. In this study, we use an unprecedentedly high-resolution (200 m horizontal and 10 m vertical grid spacing) ocean model that resolves shelf-sea and sub-ice-shelf environments in qualitative agreement with existing observations during austral summer conditions. We demonstrate that the waters reaching the Pine Island and Thwaites grounding lines follow specific, topographically-constrained routes, all passing through a relatively small area located around 104°W and 74.3°S. The temporal and spatial variabilities of ice shelf melt rates are dominantly controlled by the sub-ice shelf ocean current. Our findings highlight the importance of accurate and high-resolution ocean bathymetry and subglacial topography for determining mCDW pathways and ice shelf melt rates.

Ocean-driven ice loss from the West Antarctic Ice Sheet is a significant contributor to sea-level rise. Recent ocean variability in the Amundsen Sea is controlled by near-surface winds. We combine palaeoclimate reconstructions and climate model simulations to understand past and future influences on Amundsen Sea winds from anthropogenic forcing and internal climate variability. The reconstructions show strong historical wind trends. External forcing from greenhouse gases and stratospheric ozone depletion drove zonally uniform westerly wind trends centred over the deep Southern Ocean. Internally generated trends resemble a South Pacific Rossby wave train and were highly influential over the Amundsen Sea continental shelf. There was strong interannual and interdecadal variability over the Amundsen Sea, with periods of anticyclonic wind anomalies in the 1940s and 1990s, when rapid ice-sheet loss was initiated. Similar anticyclonic anomalies probably occurred prior to the 20th century but without causing the present ice loss. This suggests that ice loss may have been triggered naturally in the 1940s but failed to recover subsequently due to the increasing importance of anthropogenic forcing from greenhouse gases (since the 1960s) and ozone depletion (since the 1980s). Future projections also feature strong wind trends. Emissions mitigation influences wind trends over the deep Southern Ocean but has less influence on winds over the Amundsen Sea shelf, where internal variability creates a large and irreducible uncertainty. This suggests that strong emissions mitigation is needed to minimise ice loss this century but that the uncontrollable future influence of internal climate variability could be equally important.

Abstract We revisit the evolution of generalised parton distributions (GPDs) in momentum space. We formulate the evolution kernels at one loop in perturbative quantum chromodynamics (pQCD) in a form that is suitable for numerical implementation and that allows for an accurate study of their properties. This leads to the first open-source implementation of GPD evolution equations able to cover the entire kinematic region and allowing for heavy-quark-threshold crossings. The numerical implementation of the GPD evolution equations is publicly accessible through the APFEL++ evolution library and is available within the PARTONS framework. Our formulation makes use of the operator definition of GPDs in light-cone gauge renormalised in the $$\overline{\text{ MS }}$$ MS ¯ scheme. For the sake of clarity, we recompute the evolution kernels at one loop in pQCD, confirming previous calculations. We obtain general conditions on the evolution kernels derived from the GPD sum rules and show that our formulation obeys these conditions. We analytically show that our calculation reproduces the DGLAP and the ERBL equations in the appropriate limits and that it guarantees the continuity of GPDs. We numerically check that the evolved GPDs fulfil DGLAP and ERBL limits, continuity, and polynomiality. We benchmark our numerical implementation against analytical evolution in conformal space. Finally, we perform a numerical comparison to an existing implementation of GPD evolution, finding general good agreement on the kinematic region accessible to the latter. This work provides a pedagogical description of GPD evolution equations which benefits from a renewed interest as future colliders, such as the electron-ion colliders in the United States and in China, are being designed. It also paves the way for the extension of GPD evolution codes to higher accuracies in pQCD desirable for precision phenomenology at these facilities.

Sea level rise contributions from the Pine Island Glacier (PIG) are strongly modulated by the backstress that its floating extension – Pine Island Ice Shelf (PIIS) – exerts on the adjoining grounded ice. The front of PIIS has recently retreated significantly via calving, and satellite and theoretical analyses have suggested further retreat is inevitable. As well as inducing an instantaneous increase in ice flow, retreat of the PIIS front may result in increased ocean melting, by relaxing the topographic barrier to warm ocean water that is currently provided by a prominent seabed ridge. Recently published research (Bradley and others, 2022a) has shown that PIIS may exhibit a strong melting response to calving, with melting close to the PIG grounding line always increasing with ice front retreat. Here, we summarise this research and, additionally, place the results in a glaciological context by comparing the impact of melt-induced and ice-dynamical changes in the ice shelf thinning rate. We find that while PIG is expected to experience rapid acceleration in response to further ice front retreat, the mean instantaneous thinning response is set primarily by changes in melting, rather than ice dynamics. Overall, further ice front retreat is expected to lead to enhanced ice-shelf thinning, with potentially detrimental consequences for ice shelf stability.

Abstract At the Trans‐Atlantic Geotraverse hydrothermal field, metalliferous sediments cover extinct hydrothermal mounds and the surrounding seafloor. Here, we report the morphological, mineralogical and geochemical processes that deposit these sediments, remobilize their metals, and affect their preservation. We found that the initial sediment metal tenor is controlled by physical transport of hydrothermal material from its source, followed by diagenetic redistribution and potentially diffuse fluid flow after high‐temperature hydrothermal activity has ceased. We distinguished three different environments: (a) proximal metalliferous sediments on top of extinct mounds are mainly derived from oxidative weathering of primary sulfide structures and are predominantly composed of Fe oxyhydroxides with low contents of Cu, Co, and Zn; metal enrichments in specific layers are likely related to upward flow of low‐temperature hydrothermal fluids; (b) medial distant metalliferous sediments found at the base of the mounds, deposited by mass transport, contain cm‐thick layers of unsorted sulfide sands with high base metal contents (e.g., up to 28% Cu); these buried sulfides continue to undergo dissolution, resulting in metal release into porewaters; (c) distal metalliferous sediments, found in depositional basins a few hundreds of meters from the extinct mounds, include fining‐upwards sequences of thin sulfide sand layers with Fe oxyhydroxides and were deposited by recurrent turbiditic flows. Dissolved metals (e.g., Cu2+ and Mn2+) diffuse upwards under reducing conditions and precipitate within the sediment. Hence, when using hydrothermal sediments to construct reliable geochronological records of hydrothermal activity, distance from source, local seafloor morphology, mass‐transport and depositional, and diagenetic modification should all be considered.

Old-growth tropical forests harbor an immense diversity of tree species but are rapidly being cleared, while secondary forests that regrow on abandoned agricultural lands increase in extent. We assess how tree species richness and composition recover during secondary succession across gradients in environmental conditions and anthropogenic disturbance in an unprecedented multisite analysis for the Neotropics. Secondary forests recover remarkably fast in species richness but slowly in species composition. Secondary forests take a median time of five decades to recover the species richness of old-growth forest (80% recovery after 20 years) based on rarefaction analysis. Full recovery of species composition takes centuries (only 34% recovery after 20 years). A dual strategy that maintains both old-growth forests and species-rich secondary forests is therefore crucial for biodiversity conservation in human-modified tropical landscapes.

Abstract We use United Kingdom Earth System Model simulations from the Coupled Model Intercomparison Project 6 to analyze the Ross Gyre (RG) dynamics during the historical 1850–2014 period and under two contrasting future climate‐change scenarios. The modeled RG is relatively stable, with an extent and strength that agree with observations. The projections exhibit an eastward gyre expansion into the Amundsen‐Bellingshausen Seas that starts during the 2040s. The associated cyclonic ocean circulation enhances the onshore transport of warm Circumpolar Deep Water into the inner regional shelf, a regime change that increases the local subsurface shelf temperatures by up to 1.2°C and is independent of future forcing scenario. The RG expansion is generated by a regional ocean surface stress curl intensification associated with anthropogenic sea ice loss. If realised in reality, such a warming would strongly influence the future stability of the West Antarctic Ice Sheet.

The intrusion of relatively warm water is causing the recent rapid thinning of the Dotson ice shelf, West Antarctica. Here, the authors analyzed two-years of mooring data from the Dotson ice shelf front and found that seasonal variability of the ocean circulation and ocean surface stress are the main causes of variability in heat transport.

Abstract We discuss the use of machine learning techniques in effectively nonparametric modelling of generalised parton distributions (GPDs) in view of their future extraction from experimental data. Current parameterisations of GPDs suffer from model dependency that lessens their impact on phenomenology and brings unknown systematics to the estimation of quantities like Mellin moments. The new strategy presented in this study allows to describe GPDs in a way fulfilling theory-driven constraints, keeping model dependency to a minimum. Getting a better grip on the control of systematic effects, our work will help the GPD phenomenology to achieve its maturity in the precision era commenced by the new generation of experiments.

Glacial melt can modify heat transport, and therefore ocean processes, associated with ice front retreat, as revealed by direct observations from the Pine Island Bay region of Antarctica.

Multiple myeloma (MM) is a hematologic cancer characterized by accumulation of malignant plasma cells in the bone marrow. To date, no definitive cure exists for MM and resistance to current treatments is one of the major challenges of this disease. The DNA helicase BLM, whose depletion or mutation causes the cancer-prone Bloom’s syndrome (BS), is a central factor of DNA damage repair by homologous recombination (HR) and genomic stability maintenance. Using independent cohorts of MM patients, we identified that high expression of BLM is associated with a poor outcome with a significant enrichment in replication stress signature. We provide evidence that chemical inhibition of BLM by the small molecule ML216 in HMCLs (human myeloma cell lines) leads to cell cycle arrest and increases apoptosis, likely by accumulation of DNA damage. BLM inhibition synergizes with the alkylating agent melphalan to efficiently inhibit growth and promote cell death in HMCLs. Moreover, ML216 treatment re-sensitizes melphalan-resistant cell lines to this conventional therapeutic agent. Altogether, these data suggest that inhibition of BLM in combination with DNA damaging agents could be of therapeutic interest in the treatment of MM, especially in those patients with high BLM expression and/or resistance to melphalan.

The monitoring of the N’Kossa offshore oil and gas fields in the Republic of Congo allowed us to assess the ecological traits of two polychaete species belonging to Sigambra (Annelida, Pilargidae). Sigambra parva occur in very low densities in all bottoms, except the most impacted, where it is totally absent; it is an undescribed species that reached >4,000 ind/m2 in hydrocarbon-enriched sediments. Their distribution patterns are compared with those of other polychaetes, showing a range of affinities for hydrocarbon-enriched sediments in the N’Kossa region. Our results suggest that S. parva would be a representative of the original local fauna, while the species associated with artificial hydrocarbon-enriched sediments, including the other Sigambra, six more polychaetes and a bivalve, could be natively associated with natural hydrocarbon-enriched sediments, using the former as alternative habitats and as dispersal stepping stones. This ecological segregation, together with a careful morphological and morphometric analyses led us to describe the latter as a new species, namely Sigambra nkossa sp. nov. Moreover, morphometric analysis allowed us to discuss on the taxonomic robustness of the key morphological characters of S. nkossa sp. nov., as well as to emend the generic diagnosis of Sigambra to accommodate the new species.

We employ airborne gravity data from NASA's Operation IceBridge collected in 2009–2014 to infer the bathymetry of sub–ice shelf cavities in front of Pine Island, Thwaites, Smith, and Kohler glaciers, West Antarctica. We use a three-dimensional inversion constrained by multibeam echo sounding data offshore and bed topography from a mass conservation reconstruction on land. The seamless bed elevation data refine details of the Pine Island sub–ice shelf cavity, a slightly thinner cavity beneath Thwaites, and previously unknown deep (>1200 m) channels beneath the Crosson and Dotson ice shelves that shallow (500 m and 750 m, respectively) near the ice shelf fronts. These sub–ice shelf channels define the natural pathways for warm, circumpolar deep water to reach the glacier grounding lines, melt the ice shelves from below, and constrain the pattern of past and future glacial retreat.

Abstract A unique feature of generalised parton distributions is their relation to the QCD energy–momentum tensor. In particular, they provide access to the mechanical properties of the proton i.e. the distributions of pressure and shear stress induced by its quark and gluon structure. In principle the pressure distribution can be experimentally determined in a model-independent way from a dispersive analysis of deeply virtual Compton scattering data through the measurement of the subtraction constant. In practice the kinematic coverage and accuracy of existing experimental data make this endeavour a challenge. Elaborating on recent global fits of deeply virtual Compton scattering measurements using artificial neural networks, our analysis presents the current knowledge on this subtraction constant and assesses the impact of the most frequent systematic assumptions made in this field of research. This study will pave the way for future works when more precise data will become available, e.g. obtained in the foreseen electron-ion colliders EIC and EIcC.

The Getz region of West Antarctica is losing ice at an increasing rate; however, the forcing mechanisms remain unclear. Here we show for the first time that since 1994, widespread speedup has occurred on the majority of glaciers in the Getz drainage basin, with some glaciers speeding up by over 44 %.
