Human cytomegalovirus (HCMV) is a ubiquitous herpesvirus and the leading cause of infectious disease related birth defects worldwide. How the immune response modulates the risk of intrauterine transmission of HCMV after maternal infection remains poorly understood. Maternal T cells likely play a critical role in preventing infection at the maternal-fetal interface and limiting spread across the placenta, but concerns exist that immune responses to infection may also cause placental dysfunction and adverse pregnancy outcomes. This study investigated the role of CD4+ and CD8+ T cells in a guinea pig model of primary cytomegalovirus infection. Monoclonal antibodies specific to guinea pig CD4 and CD8 were used to deplete T cells in non-pregnant and in pregnant guinea pigs after mid-gestation. CD4+ T cell depletion increased the severity of illness, caused significantly elevated viral loads, and increased the rate of congenital guinea pig cytomegalovirus (GPCMV) infection relative to animals treated with control antibody. CD8+ T cell depletion was comparably well tolerated and did not significantly affect the weight of infected guinea pigs or viral loads in their blood or tissue. However, significantly more viral genomes and transcripts were detected in the placenta and decidua of CD8+ T cell depleted dams post-infection. This study corroborates earlier findings made in nonhuman primates that maternal CD4+ T cells play a critical role in limiting the severity of primary CMV infection during pregnancy while also revealing that other innate and adaptive immune responses can compensate for an absent CD8+ T cell response in α-CD8-treated guinea pigs.

BackgroundAcute kidney injury (AKI) and multiple organ failure (MOF) are leading causes of mortality in trauma injuries. Early diagnosis of AKI and MOF is vital to improve outcomes, but current diagnostic criteria rely on laboratory markers that are delayed or unreliable. In this study, we investigated whether damage associated molecular patterns such as high-mobility group box 1 (HMGB1), syndecan-1 (SDC-1) and C3a correlate with the development of trauma-induced AKI and MOF. MethodsThirty-nine swine underwent smoke inhalation and severe burns, then received critical care for 72 hours or until death. AKI was defined by the KDIGO (Kidney Disease: Improving Global Outcomes) criteria, which labels AKI when a 1.5-fold increase in blood creatinine levels from baseline or a urine output < 0.5 mL/kg/h for 6 hours or more occurs. Abstract Ice penetrating radar sounding is the primary geophysical technique for imaging the subsurface of planetary ice shells and has the potential to directly detect the ice–ocean interface. However, many sounding measurements may lack laterally extensive features that would aid their physical interpretation. In this scenario, the detection of sparse echoes can also provide rich information on ice shell properties. To explore and demonstrate this possibility, we consider three cases of isolated radar signatures: pore‐curing, eutectic melt, and unattributed echoes. We show that through detection of unattributed sparse echoes, the thickness of the conductive portion of Europa's ice shell can be constrained. These constraints can be improved by attributing sparse echoes to thermally diagnostic signatures such as pore‐curing and eutectic melt. Notably, this approach to radar sounding echo analysis is particularly compatible with joint inversions with other planetary geophysical observations such as tidal deformation, magnetic induction, and rotation state.

Abstract Background Newer antiretrivirals (ART) have shifted the metabolic experiences of people with HIV (PWH) from those of wasting syndrome to increases in body mass index (BMI). This study sought to examine the relationship between BMI and ART use among youth with HIV (YWH). Methods Charts from YWH ages 10–24 with at least two documented BMIs at least 6 months apart between 2017 and 2020 were included (N = 44). Statistical analyses were conducted in SAS 9.4. Results Clients were predominately African American (66%) males (73%) aged 19–24 years (64%), with men having sex with men (48%) being the most common mode of transmission. YWH on non-integrase inhibitor (INSTI) regimens had greater absolute increases in BMI compared to those on INSTI regimens (p = 0.03). Fourteen percent of clients using INSTI experienced an increase in BMI class from normal to overweight or overweight to obese; no non-INSTI users changed BMI class. Time since diagnosis and BMI change due to weight gain were positively associated (p = 0.03) among behaviorally-acquired YWH. Conclusions Increasing BMI and changing BMI classes may be more likely among YWH using INSTI. More longitudinal studies inclusive of diet and exercise profiles are needed to understand the relationship between INSTI and YWH BMI.

Abstract Adélie‐George V Land in East Antarctica, encompassing the vast Wilkes Subglacial Basin, has a configuration that could be prone to ice sheet instability: the basin's retrograde bed slope could make its marine terminating glaciers vulnerable to warm seawater intrusion and irreversible retreat under predicted climate forcing. However, future projections are uncertain, due in part to limited subglacial observations near the grounding zone. Here, we develop a novel statistical approach to characterize subglacial conditions from radar sounding observations. Our method reveals intermixed frozen and thawed bed within 100 km of the grounding‐zone near the Wilkes Subglacial Basin outflow, and enables comparisons to ice sheet model‐inferred thermal states. The signs of intermixed or near thawed conditions raises the possibility that changes in basal thermal state could impact the stability of Adélie‐George V Land, adding to the region's potentially vulnerable topographic configuration and sensitivity to ocean forcing driven grounding line retreat.

Abstract Background Achieving early and sustained viral suppression (VS) following diagnosis of HIV infection is critical to improving outcomes for persons with HIV (PWH). The Deep South of the United States (US) is a region that is disproportionately impacted by the domestic HIV epidemic. Time to VS, defined as time from diagnosis to initial VS, is substantially longer in the South than other regions of the US. We describe the development and implementation of a distributed data network between an academic institution and state health departments to investigate variation in time to VS in the Deep South. Methods Representatives of state health departments, the Centers for Disease Control and Prevention (CDC), and the academic partner met to establish core objectives and procedures at the beginning of the project. Importantly, this project used the CDC-developed Enhanced HIV/AIDS Reporting System (eHARS) through a distributed data network model that maintained the confidentiality and integrity of the data. Software programs to build datasets and calculate time to VS were written by the academic partner and shared with each public health partner. To develop spatial elements of the eHARS data, health departments geocoded residential addresses of each newly diagnosed individual in eHARS between 2012–2019, supported by the academic partner. Health departments conducted all analyses within their own systems. Aggregate results were combined across states using meta-analysis techniques. Additionally, we created a synthetic eHARS data set for code development and testing. Results The collaborative structure and distributed data network have allowed us to refine the study questions and analytic plans to conduct investigations into variation in time to VS for both research and public health practice. Additionally, a synthetic eHARS data set has been created and is publicly available for researchers and public health practitioners. Conclusions These efforts have leveraged the practice expertise and surveillance data within state health departments and the analytic and methodologic expertise of the academic partner. This study could serve as an illustrative example of effective collaboration between academic institutions and public health agencies and provides resources to facilitate future use of the US HIV surveillance system for research and public health practice.

Nickel titanium (NiTi, or Nitinol) alloy is used in several biomedical applications, including cardiac, peripheral vascular, and fallopian tube stents. There are significant biocompatibility issues of metallic implants to nickel ions and nano-/micro-sized alloy particles. Our laboratories have recently shown that microscale CoCr wear particles from metal-on-metal hips crosslink with the innate immune signaling Toll-like receptor 4 (TLR4), prompting downstream signaling that results in interleukin (IL)-1β and IL-8 gene expression. In vivo, NiTi alloy can also generate wear particles on the nanoscale (NP) that have thus far not been studied for their potential to induce inflammation and angiogenesis that can, in turn, contribute to implant (e.g. stent) failure. Earlier studies by others demonstrated that nickel could induce contact hypersensitivity by crosslinking the human, but not the mouse, TLR4. In the present work, it is demonstrated that NiCl2 ions and NiTi nanoparticles induce pro-inflammatory and pro-angiogenic cytokine/chemokine expression in human endothelial and monocyte cell lines in vitro. These observations prompt concerns about potential mechanisms for stent failure. The data here showed a direct correlation between intracellular uptake of Ni2+ and generation of reactive oxygen species. To determine a role for nickel and NiTi nanoparticles in inducing angiogenesis in vivo, 1-cm silicone angioreactors were implanted subcutaneously into athymic (T-cell-deficient) nude mice. The angioreactors contained Matrigel (a gelatinous protein mixture that resembles extracellular matrix) in addition to one of the following: PBS (negative control), VEGF/FGF-2 (positive control), NiCl2, or NiTi NP. The implantation of angioreactors represents a potential tool for quantification of angiogenic potentials of medical device-derived particles and ions in vivo. By this approach, NiTi NP were found to be markedly angiogenic, while Ni2+ was less-so. The angioreactors may provide a powerful tool to examine if debris shed from medical devices may promote untoward biological effects.

Abstract Objective: Gaps in sexually transmitted infection (STI) testing can lead to poor health outcomes due to untreated illness among youth living with HIV (YLHIV). Thus, the objective of this study is to examine STI testing behavior and outcomes among a sample of YLHIV in the southern United States. Clinical records of 139 YLHIV who received HIV care in Alabama (2017–2020) were evaluated for receipt of STI testing (gonorrhea, chlamydia, syphilis), prevalence of positive test results, and factors associated with testing outcomes (933 clinical visits). Results: Nearly 80% of our sample identified as African American, most were 20–24 years, and about 60% reported detectable viral load at first visit during the study period. Just under 60% of cisgender male and transgender female clients reported receipt of at least one STI test, compared to less than 40% of cisgender females. Identifying as a cisgender male and having been diagnosed with HIV related to sex with men were associated with greater likelihood receiving STI testing. Cisgender males reported higher rates of positive syphilis test results than cisgender females; the highest rates of positive STI tests were among transgender females. Results underscore need for providers to promote routine STI testing to YLHIV.

This study uses ice sheet modeling experiments to show that thawing portions of the Antarctic ice sheet bed can increase century-scale mass loss, particularly in the Wilkes and Enderby Land regions of East Antarctica.

Ice-penetrating radar sounding is a powerful geophysical tool for studying terrestrial and planetary ice with a rich glaciological heritage reaching back over half a century. Recent years have also seen rapid growth in both the radioglaciological community itself and in the scope and sophistication of its analysis of ice-penetrating radar data. This has been spurred by a combination of growing datasets and improvements in computational resources as well as advances in radar sounding instrumentation and platforms. Together, these developments are transforming the field and highlight exciting paths forward for future innovation and investigation.

Despite the availability of seasonal vaccines and antiviral medications, influenza virus continues to be a major health concern and pandemic threat due to the continually changing antigenic regions of the major surface glycoprotein, hemagglutinin (HA). One emerging strategy for the development of more efficacious seasonal and universal influenza vaccines is structure-guided design of nanoparticles that display conserved regions of HA, such as the stem. Using the H1 HA subtype to establish proof of concept, we found that tandem copies of an alpha-helical fragment from the conserved stem region (helix-A) can be displayed on the protruding spikes structures of a capsid scaffold. The stem region of HA on these designed chimeric nanoparticles is immunogenic and the nanoparticles are biochemically robust in that heat exposure did not destroy the particles and immunogenicity was retained. Furthermore, mice vaccinated with H1-nanoparticles were protected from lethal challenge with H1N1 influenza virus. By using a nanoparticle library approach with this helix-A nanoparticle design, we show that this vaccine nanoparticle construct design could be applicable to different influenza HA subtypes. Importantly, antibodies elicited by H1, H5, and H7 nanoparticles demonstrated homosubtypic and heterosubtypic cross-reactivity binding to different HA subtypes. Also, helix-A nanoparticle immunizations were used to isolate mouse monoclonal antibodies that demonstrated heterosubtypic cross-reactivity and provided protection to mice from viral challenge via passive-transfer. This tandem helix-A nanoparticle construct represents a novel design to display several hundred copies of non-trimeric conserved HA stem epitopes on vaccine nanoparticles. This design concept provides a new approach to universal influenza vaccine development strategies and opens opportunities for the development of nanoparticles with broad coverage over many antigenically diverse influenza HA subtypes.

The earliest airborne geophysical campaigns over Antarctica and Greenland in the 1960s and 1970s collected ice penetrating radar data on 35 mm optical film. Early subglacial topographic and englacial stratigraphic analyses of these data were foundational to the field of radioglaciology. Recent efforts to digitize and release these data have resulted in geometric and ice-thickness analysis that constrain subsurface change over multiple decades but stop short of radiometric interpretation. The primary challenge for radiometric analysis is the poorly-characterized compression applied to Z-scope records and the sparse sampling of A-scope records. Here, we demonstrate the information richness and radiometric interpretability of Z-scope records. Z-scope pixels have uncalibrated fast-time, slow-time, and intensity scales. We develop approaches for mapping each of these scales to physical units (microseconds, seconds, and signal to noise ratio). We then demonstrate the application of this calibration and analysis approach to a flight in the interior of East Antarctica with subglacial lakes and to a reflight of an East Antarctic ice shelf that was observed by both archival and modern radar. These results demonstrate the potential use of Z-scope signals to extend the baseline of radiometric observations of the subsurface by decades.

Farmed mink are one of few animals in which infection with SARS-CoV-2 has resulted in sustained transmission among a population and spillback from mink to people. In September 2020, mink on a Michigan farm exhibited increased morbidity and mortality rates due to confirmed SARS-CoV-2 infection. We conducted an epidemiologic investigation to identify the source of initial mink exposure, assess the degree of spread within the facility’s overall mink population, and evaluate the risk of further viral spread on the farm and in surrounding wildlife habitats. Three farm employees reported symptoms consistent with COVID-19 the same day that increased mortality rates were observed among the mink herd. One of these individuals, and another asymptomatic employee, tested positive for SARS-CoV-2 by real-time reverse transcription PCR (RT-qPCR) 9 days later. All but one mink sampled on the farm were positive for SARS-CoV-2 based on nucleic acid detection from at least one oral, nasal, or rectal swab tested by RT-qPCR (99%). Sequence analysis showed high degrees of similarity between sequences from mink and the two positive farm employees. Epidemiologic and genomic data, including the presence of F486L and N501T mutations believed to arise through mink adaptation, support the hypothesis that the two employees with SARS-CoV-2 nucleic acid detection contracted COVID-19 from mink. However, the specific source of virus introduction onto the farm was not identified. Three companion animals living with mink farm employees and 31 wild animals of six species sampled in the surrounding area were negative for SARS-CoV-2 by RT-qPCR. Results from this investigation support the necessity of a One Health approach to manage the zoonotic spread of SARS-CoV-2 and underscores the critical need for multifaceted public health approaches to prevent the introduction and spread of respiratory viruses on mink farms.

The formation of double ridges on Europa is poorly understood. Here the authors analyze airborne radar observations of an analog feature on the Greenland Ice Sheet to show that the refreezing of shallow water sills may produce such ridges.

Worldwide, Human T-lymphtropic virus-1 and 2 (HTLV-1 and 2) infects approximately more than 10 million people, mostly occurring in hyperendemic areas such as the region of Latin America and the Caribbean (LAC). A comprehensive bibliographic exploration of original articles published on the Web of Science Core Collection database over the last 10 years was done. A bibliometric analysis was performed using the bibliometrix package in RStudio and VOSviewer. A total of 519 articles published in 194 journals were identified along the 10 years studied. In 2012 the peak number of publications was identified and the average number of citations per document was 1.33. Galvao-Castro B was the author with the greatest number of publications. Aids Research and Human Retroviruses was the most productive journal, and the study by Bangham CRM was the most cited. Brazil was the country with most corresponding authors that had the most publications and the most significant number of total citations. Infections and HTLV-1 were the most used keywords. In conclusion, according to the current quantitative analysis, there is a need for more significant promotion of research on HTLV-1 and 2 among the scientific community of LAC.

BackgroundPre-exposure prophylaxis (PrEP) is a highly effective biomedical prevention intervention and a major strategy for reducing the HIV burden in the United States. However, PrEP provision and uptake remain lower than estimated needs, and in ways that may exacerbate HIV disparities among Black adolescent girls and young women in the southern United States. Data suggest that gaps in provider knowledge of HIV epidemiology and PrEP and skills assessing sexual health practices are important barriers to provision and uptake, with limited evidence-based interventions to address these gaps. ObjectiveThis paper describes the “PrEP-Pro” intervention, a multicomponent intervention to train and support family medicine (FM) trainees to promote PrEP for adolescent girls and young women in Alabama. MethodsThe PrEP-Pro intervention comprises 3 main components guided by the Capability-Opportunity-Motivation-Behavior (COM-B) model for behavioral change and the Consolidated Framework for Implementation Research (CFIR): (1) provider HIV epidemiology and PrEP education, (2) sexual history taking, and (3) PrEP Champions. In phase 1, we will work with community advisory boards (providers and clients) and then conduct focus groups with FM trainees to adapt content to train FM residents on HIV epidemiology and PrEP and develop implementation strategies, including provider-facing tools and client-facing educational materials. In phase 2, we will pretest and then pilot-test the initially adapted PrEP-Pro intervention with FM trainees. FM trainees will complete baseline, 3-, and 6-month questionnaires post PrEP-Pro intervention. We will also conduct in-depth interviews (IDIs) with FM pilot participants, adolescent girls and young women who accessed care after the PrEP-Pro pilot, and key stakeholders. The primary outcomes are PrEP-Pro acceptability and feasibility, which would be assessed using validated instruments at months 3 (among pretest participants) and 6 (among pilot participants). Secondary outcomes will also be assessed, including PrEP knowledge, sexual history-taking attitudes and practices, PrEP prescriptions among adolescent girls and young women encounters, and sexually transmitted infections (STIs) and HIV testing among adolescent girls and young women encounters in 6 months. ResultsStudy results will be disseminated to practices, state health officials, and other key stakeholders to solicit feedback on implementation opportunities and challenges to inform a hybrid effectiveness implementation trial. Our results will also be presented at local and national conferences and submitted to peer-reviewed journals. ConclusionsAs PrEP grows, there is a pressing need to train FM providers and develop appropriate, contextually relevant tools to support PrEP implementation. The PrEP-Pro intervention is a multicomponent intervention to train FM residents across Alabama on sexual history-taking, PrEP provision for adolescent girls and young women, and supporting practice-based PrEP Champions. The PrEP-Pro intervention is anticipated to increase PrEP prescriptions for adolescent girls and young women and expand comprehensive sexual and reproductive health care for adolescent girls and young women in rural and urban Alabama. Objective: Single-cell RNA sequencing (scRNA-seq) is a powerful technology that can be applied to the cells populating the whole knee in the study of joint pathology. The knee contains cells embedded in hard structural tissues, cells in softer tissues and membranes, and immune cells. This creates a technical challenge in preparing a viable and representative cell suspension suitable for use in scRNA-seq in minimal time, where under-digestion may exclude cells in hard tissues, over-digestion may damage soft tissue cells, and prolonged digestion may induce phenotypic drift. We developed a rapid two-stage digestion protocol to overcome these difficulties. Design: A two-stage digest consisting of first collagenase IV, an intermediate cell recovery, then collagenase II on the remaining hard tissue. Cells were sequenced on the 10x Genomics platform. Results: We observed consistent cell numbers and viable single cell suspensions suitable for scRNA-seq analysis. Comparison of contralateral knees and separate mice showed reproducible cell yields and gene expression patterns by similar cell-types. A diverse collection of structural and immune cells were captured with a majority from immune origins. Two digestions were necessary to capture all cell-types. Conclusions: The knee contains a diverse mixture of stromal and immune cells that may be crucial for the study of osteoarthritis. The two-stage digestion presented here reproducibly generated highly viable and representative single-cell suspension for sequencing from the whole knee. This protocol facilitates transcriptomic studies of the joint as a complete organ.

Sunn hemp (Crotalaria juncea L.), a warm season leguminous cover crop, is commonly used in rotation with organic strawberry production in Florida’s subtropical environment. This study was conducted to explore the impacts of sunn hemp on growth and yield performance of the subsequent organic strawberry crop in sandy soils, taking into consideration the nutrient contribution from soil incorporation of sunn hemp biomass. Sunn hemp was seeded during the summer off-season and terminated before flowering, three weeks prior to the fall planting of two strawberry cultivars (‘Strawberry Festival’ and ‘Camino Real’). With sunn hemp residues incorporated into the soil, two application rates of nitrogen (N) through pre-plant organic fertilization for the strawberry season were used, including N at a rate of 84 kg/ha, without consideration of the N credit from sunn hemp, and N at a rate of 19.8 kg/ha, with consideration of the estimated N credit from sunn hemp. A summer fallow without cover crop and with a pre-plant organic fertilizer application at the N rate of 84 kg/ha was included as the control. Overall, the sunn hemp incorporation at three weeks after termination did not benefit the strawberry plant growth or fruit yield in this study, with rather low levels of soilborne pathogen and nematode infestations. Both sunn hemp treatments exhibited a significantly lower level of total soil N compared to the summer fallow plots at the end of the strawberry season. The reduction in the pre-plant N fertilization resulted in lower above-ground plant dry weight and accumulation of N, phosphorus (P), and potassium (K) at the end of the strawberry season, along with fewer leaves and smaller crowns of the strawberry plants during the early season. Both sunn hemp treatments decreased early fruit yields, while the sunn hemp treatment with the reduced N fertilization also led to a significant reduction in the total fruit number and weight, although no significant differences in the whole-season marketable fruit yield were observed among the nutrient management treatments. Overall, ‘Strawberry Festival’ yielded higher than ‘Camino Real’, but the effects of nutrient management did not vary with the strawberry cultivars. Further studies are needed to enhance organic strawberry nutrient management involving rotational cover crops.

Higher education is facing the need for its leaders to adapt to an ever-increasingly virtual landscape. More than ever before, this became evident during the COVID-19 pandemic, which began in 2020. Previously-utilized leadership strategies proved less than effective tools for managing virtual faculty teams, and new, creative methods of leading people working remotely across wide geographical areas were launched almost overnight. Some worked; some did not. The purpose of this work is to explore the best practices for academic leaders to mentor, motivate, and guide their teams in virtual environments. The themes of creating a culture of trust, team-building and collaboration, and communication emerged in the literature as traits of effective leaders. Here, we apply those traits to personal experiences between March 2020 and May 2021 to propose a model for leadership in a virtual space.

Background: This protocol details the adaptation and pilot testing of the Finding Respect and Ending Stigma around HIV (FRESH) intervention in Dominican Republic. FRESH is a healthcare setting stigma-reduction intervention designed to reduce stigmas affecting people living with HIV (PLHIV), focusing on HIV and intersectional stigmas experienced by sexual and gender minority (SGM) people living with HIV. After the successful adaptation of the FRESH intervention, it will be pilot-tested through the conduct of a pilot stepped wedge cluster randomized controlled trial. Methods: Three aims are included in this study; Aim 1 includes exploratory qualitative assessment, specifically the conduct four focus groups with men who have sex with men (MSM) living with HIV (n = 24–32) and in-depth interviews with transgender women living with HIV to explore their experiences with stigma in clinics (n = 9–12). In-depth interviews will also be held with HIV healthcare workers to elucidate their perceptions and behaviors towards their SGM clients (n = 9–12). In Aim 2, informed by Aim 1 data, we will use the sequential phases of the ADAPT-ITT framework to iteratively adapt the FRESH intervention for the Dominican Republic. In Aim 3, the adapted intervention will be pilot-tested via a cluster stepped wedge randomized controlled trial to assess feasibility and acceptability of the intervention and study protocols. Conclusions: If this pilot trial is successful, next steps will include testing the adapted intervention across Dominican Republic or in similar Spanish-speaking Caribbean nations in a larger trial to assess effectiveness in reducing stigma in clinical settings towards PLHIV.

The long-term impact of extreme surface melt on the Greenland Ice Sheet is poorly constrained. Here the authors use airborne radar to characterize a subsurface refrozen melt layer that formed following extreme melt in 2012, showing that it likely reduced drainage pathways for subsequent melt.

ABSTRACT Lateral gene transfer plays an important role in the evolution of genetic diversity in prokaryotes. DNA transfer via natural transformation depends on the ability of recipient cells to actively transport DNA from the environment into the cytoplasm, termed natural competence, which relies on the presence of type IV pili and other competence proteins. Natural competence has been described in cyanobacteria for several organisms, including unicellular and filamentous species. However, natural competence in cyanobacteria that differentiate specialized cells for N2-fixation (heterocysts) and form branching or multiseriate cell filaments (termed subsection V) remains unknown. Here, we show that genes essential for natural competence are conserved in subsection V cyanobacteria. Furthermore, using the replicating plasmid pRL25C, we experimentally demonstrate natural competence in a subsection V organism: Chlorogloeopsis fritschii PCC 6912. Our results suggest that natural competence is a common trait in cyanobacteria forming complex cell filament morphologies. IMPORTANCE Cyanobacteria are crucial players in the global biogeochemical cycles, where they contribute to CO2- and N2-fixation. Their main ecological significance is the primary biomass production owing to oxygenic photosynthesis. Cyanobacteria are a diverse phylum, in which the most complex species differentiate specialized cell types and form true-branching or multiseriate cell filament structures (termed subsection V cyanobacteria). These bacteria are considered a peak in the evolution of prokaryotic multicellularity. Among others, species in that group inhabit fresh and marine water habitats, soil, and extreme habitats such as thermal springs. Here, we show that the core genes required for natural competence are frequent in subsection V cyanobacteria and demonstrate for the first time natural transformation in a member of subsection V. The prevalence of natural competence has implications for the role of DNA acquisition in the genome evolution of cyanobacteria. Furthermore, the presence of mechanisms for natural transformation opens up new possibilities for the genetic modification of subsection V cyanobacteria.

Subglacial topography is an important feature in numerous ice-sheet analyses and can drive the routing of water at the bed. Bed topography is primarily measured with ice-penetrating radar. Significant gaps, however, remain in data coverage that require interpolation. Topographic interpolations are typically made with kriging, as well as with mass conservation, where ice flow dynamics are used to constrain bed geometry. However, these techniques generate bed topography that is unrealistically smooth at small scales, which biases subglacial water flowpath models and makes it difficult to rigorously quantify uncertainty in subglacial drainage patterns. To address this challenge, we adapt a geostatistical simulation method with probabilistic modeling to stochastically simulate bed topography such that the interpolated topography retains the spatial statistics of the ice-penetrating radar data. We use this method to simulate subglacial topography using mass conservation topography as a secondary constraint. We apply a water routing model to each of these realizations. Our results show that many of the flowpaths significantly change with each topographic realization, demonstrating that geostatistical simulation can be useful for assessing confidence in subglacial flowpaths.

Abstract Objective Our aim was to conduct a pilot randomized controlled trial of a novel cognitive behaviorally based intervention for pediatric PNES called Retraining and Control Therapy (ReACT). Methods Participants were randomized to receive either eight sessions of ReACT or supportive therapy, and participants completed follow‐up visits at 7‐ and 60‐days posttreatment. The primary outcome measure was PNES frequency at 7‐days posttreatment. Eligibility criteria included children with video‐EEG confirmed PNES and participant/parent or guardian willingness to participate in treatment. Exclusion criteria included substance use, psychosis, and severe intellectual disability. Forty‐two patients were assessed for eligibility and 32 were randomized. ReACT aimed to retrain classically conditioned, involuntary PNES by targeting catastrophic symptom expectations and a low sense of control over symptoms using principles of habit reversal. Supportive therapy was based on the assumption that relief from stress or problems can be achieved by discussion with a therapist. Results Twenty‐nine participants (Mage = 15.1 years, SDage = 2.5; 72.2% female; 57.1% Caucasian, 28.6% African American) completed 7‐days postprocedures. Although grower interest in high tunnel tomato (Solanum lycopersicum) production has increased in recent years, systematic high tunnel research conducted in humid, subtropical regions has been limited. The potential of tomato grafting to mitigate biotic and abiotic stresses makes it complementary to high-value production systems in high tunnels. In this 2-year study, grafted vs. nongrafted organic tomato production in high tunnels and open fields was investigated to determine possible synergistic effects of these two technologies. In 2016, high tunnels resulted in a significant increase of total and marketable yields, by 43% and 87%, respectively, over open field production. Grafting also significantly increased total and marketable yields over nongrafted plants by 34% and 42%, respectively. Cultivar effects demonstrated greater benefits with the implementation of high tunnel and grafting technologies for ‘Tribute’ (a beefsteak-type tomato) than for ‘Garden Gem’ (a plum-type tomato), as the increase in marketable yield was 33% greater for ‘Tribute’ in high tunnels and 45% greater for ‘Tribute’ with grafting. In 2017, a delayed effective transplanting date and the lack of high tunnel summer season extension produced results that were generally cultivar specific. While grafting increased the total yield of both cultivars (by 18%), marketable yield was increased by grafting only for ‘Tribute’ in high tunnels (by 42%). Additionally, high tunnels improved marketable yield of ‘Tribute’ by 129% but had no effect on ‘Garden Gem’. This demonstrated the consistent trend of the beefsteak-type tomato benefiting more from the combination of high tunnel and grafting technologies than the plum-type tomato. High tunnels reduced fruit decay and cracking by up to 71% compared with open field production. Stink bug (Pentatomidae) damage had the greatest impact on marketable yields each season, reaching 13% and 34% of total yields in 2016 and 2017, respectively, and was unaffected by high tunnel production or grafting. This study revealed the benefits of integrating high tunnel and grafting technologies for enhancing organic production of fresh-market tomato in the humid subtropics, and demonstrated more research is warranted to establish regional planting dates and further optimize this high-value cropping system.

The Antarctic Impulsive Transient Antenna (ANITA) balloon experiment was designed to detect radio signals initiated by high-energy neutrinos and cosmic ray (CR) air showers. These signals are typically discriminated by the polarization and phase inversions of the radio signal. The reflected signal from CRs suffer phase inversion compared to a direct ‘tau neutrino’ event. In this paper, we study subsurface reflection, which can occur without phase inversion, in the context of the two anomalous up-going events reported by ANITA. It is found that subsurface layers and firn density inversions may plausibly account for the events, while ice fabric layers and wind ablation crusts could also play a role. This hypothesis can be tested with radar surveying of the Antarctic region in the vicinity of the anomalous ANITA events. Future experiments should not use phase inversion as a sole criterion to discriminate between down-going and up-going events, unless the subsurface reflection properties are well understood.

Airborne radio-echo sounding (RES) surveys are widely used to measure ice-sheet bed topography. Measuring bed topography as accurately and widely as possible is of critical importance to modelling ice dynamics and hence to constraining better future ice response to climate change. Measurement accuracy of RES surveys is influenced both by the geometry of bed topography and the survey design. Here we develop a novel approach for simulating RES surveys over glaciated terrain, to quantify the sensitivity of derived bed elevation to topographic geometry. Furthermore, we investigate how measurement errors influence the quantification of glacial valley geometry. We find a negative bias across RES measurements, where off-nadir return measurement error is typically −1.8 ± 11.6 m. Topographic highlands are under-measured an order of magnitude more than lowlands. Consequently, valley depth and cross-sectional area are largely under-estimated. While overall estimates of ice thickness are likely too high, we find large glacier valley cross-sectional area to be under-estimated by −2.8 ± 18.1%. Therefore, estimates of ice flux through large outlet glaciers are likely too low when this effect is not taken into account. Additionally, bed mismeasurements potentially impact our appreciation of outlet-glacier stability.

Radar surveys across ice sheets typically measure numerous englacial layers that can often be regarded as isochrones. Such layers are valuable for extrapolating age–depth relationships away from ice-core locations, reconstructing palaeoaccumulation variability, and investigating past ice-sheet dynamics. However, the use of englacial layers in Antarctica has been hampered by underdeveloped techniques for characterising layer continuity and geometry over large distances, with techniques developed independently and little opportunity for inter-comparison of results. In this paper, we present a methodology to assess the performance of automated layer-tracking and layer-dip-estimation algorithms through their ability to propagate a correct age–depth model. We use this to assess isochrone-tracking techniques applied to two test case datasets, selected from CreSIS MCoRDS data over Antarctica from a range of environments including low-dip, continuous layers and layers with terminations. We find that dip-estimation techniques are generally successful in tracking englacial dip but break down in the upper and lower regions of the ice sheet. The results of testing two previously published layer-tracking algorithms show that further development is required to attain a good constraint of age–depth relationships away from dated ice cores. We recommend that auto-tracking techniques focus on improved linking of picked stratigraphy across signal disruptions to enable accurate determination of the Antarctic-wide age–depth structure.

Fast ice flow on the Antarctic continent constitutes much of the mass loss from the ice sheet. However, geophysical methods struggle to constrain ice flow history at depth, or separate the signatures of topography, ice dynamics and basal conditions on layer structure. We develop and demonstrate a methodology to compare layer signatures in multiple airborne radar transects in order to characterize ice flow at depth, or improve coverage of existing radar surveys. We apply this technique to generate synthetic, along-flow radargrams and compare different deformation regimes to observed radargram structure. Specifically, we investigate flow around the central sticky spot of Whillans Ice Stream, West Antarctica. Our study suggests that present-day velocity flowlines are insufficient to characterize flow at depth as expressed in layer geometry, and streaklines provide a better characterization of flow around a basal sticky spot. For Whillans Ice Stream, this suggests that ice flow wraps around the central sticky spot, supported by idealized flow simulations. While tracking isochrone translation and rotation across survey lines is complex, we demonstrate that our approach to combine radargram interpretation and modeling can reveal critical details of past ice flow.

Here we use polarimetric measurements from an Autonomous phase-sensitive Radio-Echo Sounder (ApRES) to investigate ice fabric within Whillans Ice Stream, West Antarctica. The survey traverse is bounded at one end by the suture zone with the Mercer Ice Stream and at the other end by a basal ‘sticky spot’. Our data analysis employs a phase-based polarimetric coherence method to estimate horizontal ice fabric properties: the fabric orientation and the magnitude of the horizontal fabric asymmetry. We infer an azimuthal rotation in the prevailing horizontal c-axis between the near-surface (z ≈ 10–50 m) and deeper ice (z ≈ 170–360 m), with the near-surface orientated closer to perpendicular to flow and deeper ice closer to parallel. In the near-surface, the fabric asymmetry increases toward the center of Whillans Ice Stream which is consistent with the surface compression direction. By contrast, the fabric orientation in deeper ice is not aligned with the surface compression direction but is consistent with englacial ice reacting to longitudinal compression associated with basal resistance from the nearby sticky spot.

Radar sounding is a powerful geophysical approach for characterizing the subsurface conditions of terrestrial and planetary ice masses at local to global scales. As a result, a wide array of orbital, airborne, ground-based, and in situ instruments, platforms and data analysis approaches for radioglaciology have been developed, applied or proposed. Terrestrially, airborne radar sounding has been used in glaciology to observe ice thickness, basal topography and englacial layers for five decades. More recently, radar sounding data have also been exploited to estimate the extent and configuration of subglacial water, the geometry of subglacial bedforms and the subglacial and englacial thermal states of ice sheets. Planetary radar sounders have observed, or are planned to observe, the subsurfaces and near-surfaces of Mars, Earth's Moon, comets and the icy moons of Jupiter. In this review paper, and the thematic issue of the Annals of Glaciology on ‘Five decades of radioglaciology’ to which it belongs, we present recent advances in the fields of radar systems, missions, signal processing, data analysis, modeling and scientific interpretation. Our review presents progress in these fields since the last radio-glaciological Annals of Glaciology issue of 2014, the context of their history and future prospects.

Basal units – visibly distinct englacial structures near the ice-bed interface – warrant investigation for a number of reasons. Many are of unknown composition and origin, characteristics that could provide substantial insight into subglacial processes and ice-sheet history. Their significance, moreover, is not limited to near-bed depths; these units appear to dramatically influence the flow of surrounding ice. In order to enable improved characterization of these features, we develop and apply an algorithm that allows for the automatic detection of basal units. We use a tunable layer-optimized SAR processor to distinguish these structures from the bed, isochronous englacial layers and the ice-sheet surface, presenting a conceptual framework for the use of radio-echo character in the identification of ice-sheet features. We also outline a method by which our processor could be used to place observational constraints on basal units’ configuration, composition and provenance.

ObjectivesTo investigate the prevalence, associated factors and socioeconomic inequalities in chronic disease relapses (CDR) during 2020 in Peru.MethodsA secondary analysis was made of the National Household Survey on Living Conditions and Poverty (ENAHO) 2020. Participants older than 18 years who suffered from a chronic disease and with information about the occurrence of a CDR in the last 4 weeks prior to the survey were included. Adjusted prevalence ratios (aPRs) were estimated to determine the associated factors. Socioeconomic inequality in CDR was estimated using concentration curves (CC) and the Erreygers concentration index (ECI).ResultsData from 38,662 participants were analyzed; the prevalence of CDR in the last 4 weeks prior to the survey was 16.5% (95% CI: 15.8-17.2). Being female (aPR 1.29; 95% CI: 1.21-1.37), with regards to being male; being 30-39 (aPR 1.22; 95% CI: 1.05-1.42), 40-49 (aPR 1.29; 95% CI: 1.12-1.48), 50-59 (aPR 1.60; 95% CI: 1.41-1.82), and 60 years or older (aPR 1.80; 95% CI: 1.58-2.04), compared to 18-29; reaching up to primary (aPR 1.18; 95% CI: 1.07-1.31), or secondary education (aPR 1.13; 95% CI: 1.02-1.24), in contrast to tertiary education; presenting some physical, psychological or cognitive limitation (aPR 1.33; 95% CI: 1.21-1.46), with respect to experiencing no limitations; and being affiliated to a health insurance (aPR 1.18; CI 95%: 1.09-1.29), opposed to not having health insurance; were associated with a higher probability of CDR. Residing in the natural region of the coastal area (aPR 0.83; 95% CI: 0.74-0.92) was associated with a lower probability of relapse compared to residing in the jungle area. In people with limitations and residents of the jungle areas, the prevalence of CDR was concentrated in those with higher per capita spending.ConclusionsApproximately 1 in 6 Peruvians with chronic diseases had a relapse within the last 4 weeks prior to the survey of 2020 and certain geographic and sociodemographic factors were found to be associated with CDR. It was also found that a higher concentration of CDR was observed in the population with the highest per capita spending with some limitations, as well as in residents of the jungle, implying the need for appropriate policy interventions that address CDR with a special focus on these populations.

Abstract The U.S. fresh-market tomato industry faces increasing competition from Mexico, which achieves greater productivity and quality due to the use of protected structures. Protected agriculture is limited in humid, subtropical regions of the United States. Although grower interest in high tunnel production has increased in recent years, systematic high tunnel research has not yet been conducted in subtropical Florida. Additionally, although tomato grafting has shown the potential to overcome biotic and abiotic stresses, research of high-tunnel, grafted tomato production in subtropical conditions is lacking. During this 2-year study (Citra, FL), a side-by-side comparison of open field and high tunnel organic tomato production was conducted using a split-split plot design. The most significant benefit of high tunnel production was season extension achieved through the reduction of foliar disease severity, which reduced the area under the disease progress curve by 64% across two seasons. This may be largely attributed to the pronounced reduction in the duration of leaf wetness during the wet months of the growing cycle. Grafting with ‘Multifort’ rootstock reduced the root-knot nematode soil population density by 88% as well as root galling severity, both of which demonstrated the potential for increased levels in the high tunnel production system compared with open field production. The more severe root-knot nematode infestation in high tunnels was likely due to the modification of soil temperatures, which were 2 °C greater during the early part of the season but were reduced after shadecloth application. Compared with the open field, solar radiation was reduced by 23% in the high tunnel before shadecloth application and by 51% after shadecloth application; however, due to the high radiation levels in subtropical Florida, daily light integral levels indicated that light was not limiting for high-quality tomato production. The average wind speed was reduced by 57% in the high tunnel and, together with the reduction in solar radiation, indicated the potential reduction in summer abiotic stress and evapotranspiration within high tunnels. These results revealed that the integrated use of high tunnel and grafting technologies may be important for enhancing fresh-market tomato production in the humid subtropics, especially in organic systems.

Late Holocene sediment deposits in Pine Island Bay, West Antarctica, are hypothesized to be linked to intensive meltwater drainage during the retreat of the paleo-Pine Island Ice Stream after the Last Glacial Maximum. The uppermost sediment units show an abrupt transition from ice-proximal debris to a draped silt during the late Holocene, which is interpreted to coincide with rapid deglaciation. The small scale and fine sorting of the upper unit could be attributed to origins in subglacial meltwater; however the thickness and deposition rate for this unit imply punctuated- rather than continuous-deposition. This, combined with the deposit's location seaward of large, bedrock basins, has led to the interpretation of this unit as the result of subglacial lake outbursts in these basins. However, the fine-scale sorting of the silt unit is problematic for this energetic interpretation, which should mobilize and deposit a wider range of sediment sizes. To resolve this discrepancy, we present an alternative mechanism in which the silt was sorted by a distributed subglacial water system, stored in bedrock basins far inland of the grounding line, and subsequently eroded at higher flow speeds during retreat. We demonstrate that this mechanism is physically plausible given the subglacial conditions during the late Holocene. We hypothesize that similar silt units observed elsewhere in Antarctica downstream of bedrock basins could be the result of the same mechanism.

Englacial layers in Antarctica and Greenland are indicators of the dynamic, rheological and subglacial configuration of the ice sheets. Airborne radar sounder data is the primary remote sensing solution for directly observing englacial layers and structures at the glacier-catchment to ice-sheet scale. However, when traditional along-track synthetic aperture radar (SAR) processing is applied, steep layers can disappear, limiting the detectability and interpretability of englacial layer geometry. This study provides a reconstruction algorithm to address the problem of destructive phase interference during the radargram formation. We develop and apply a novel SAR processor optimized for layer detection that enhances the Signal-to-Noise ratio (SNR) of specular reflectors. The algorithm also enables the automatic estimation of layer slope. We demonstrate the algorithm using data acquired at the Institute Ice Stream, West Antarctica.

A subset of COVID-19 patients exhibit post-acute sequelae of COVID-19 (PASC), but little is known about the immune signatures associated with these syndromes. We investigated longitudinal peripheral blood samples in 50 individuals with previously confirmed SARS-CoV-2 infection, including 20 who experienced prolonged duration of COVID-19 symptoms (lasting more than 30 days; median = 74 days) compared with 30 who had symptom resolution within 20 days. Individuals with prolonged symptom duration maintained antigen-specific T cell response magnitudes to SARS-CoV-2 spike protein in CD4+ and circulating T follicular helper cell populations during late convalescence, while those without persistent symptoms demonstrated an expected decline. The prolonged group also displayed increased IgG avidity to SARS-CoV-2 spike protein. Significant correlations between symptom duration and both SARS-CoV-2–specific T cells and antibodies were observed. Activation and exhaustion markers were evaluated in multiple immune cell types, revealing few phenotypic differences between prolonged and recovered groups, suggesting that prolonged symptom duration is not due to persistent systemic inflammation. These findings demonstrate that SARS-CoV-2–specific immune responses are maintained in patients suffering from prolonged post–COVID-19 symptom duration in contrast to those with resolved symptoms and may suggest the persistence of viral antigens as an underlying etiology.

Traumatic brain injury (TBI) results in complex pathological reactions, where the initial lesion is followed by secondary inflammation and edema. Our laboratory and others have reported that angiotensin receptor blockers (ARBs) have efficacy in improving recovery from traumatic brain injury in mice. Treatment of mice with a subhypotensive dose of the ARB candesartan results in improved functional recovery, and reduced pathology (lesion volume, inflammation and gliosis). In order to gain a better understanding of the molecular mechanisms through which candesartan improves recovery after controlled cortical impact injury (CCI), we performed transcriptomic profiling on brain regions after injury and drug treatment. We examined RNA expression in the ipsilateral hippocampus, thalamus and hypothalamus at 3 or 29 days post injury (dpi) treated with either candesartan (0.1 mg/kg) or vehicle. RNA was isolated and analyzed by bulk mRNA-seq. Gene expression in injured and/or candesartan treated brain region was compared to that in sham vehicle treated mice in the same brain region to identify genes that were differentially expressed (DEGs) between groups. The most DEGs were expressed in the hippocampus at 3 dpi, and the number of DEGs reduced with distance and time from the lesion. Among pathways that were differentially expressed at 3 dpi after CCI, candesartan treatment altered genes involved in angiogenesis, interferon signaling, extracellular matrix regulation including integrins and chromosome maintenance and DNA replication. At 29 dpi, candesartan treatment reduced the expression of genes involved in the inflammatory response. Some changes in gene expression were confirmed in a separate cohort of animals by qPCR. Fewer DEGs were found in the thalamus, and only one in the hypothalamus at 3 dpi. Additionally, in the hippocampi of sham injured mice, 3 days of candesartan treatment led to the differential expression of 384 genes showing that candesartan in the absence of injury had a powerful impact on gene expression specifically in the hippocampus. Our results suggest that candesartan has broad actions in the brain after injury and affects different processes at acute and chronic times after injury. These data should assist in elucidating the beneficial effect of candesartan on recovery from TBI.

Paracrine signaling in the tissue microenvironment is a central mediator of morphogenesis, and modeling this dynamic intercellular activity in vitro is critical to understanding normal and abnormal development. For example, Sonic Hedgehog (Shh) signaling is a conserved mechanism involved in multiple developmental processes and strongly linked to human birth defects including orofacial clefts of the lip and palate. SHH ligand produced, processed, and secreted from the epithelial ectoderm is shuttled through the extracellular matrix where it binds mesenchymal receptors, establishing a gradient of transcriptional response that drives orofacial morphogenesis. In humans, complex interactions of genetic predispositions and environmental insults acting on diverse molecular targets are thought to underlie orofacial cleft etiology. Consequently, there is a need for tractable in vitro approaches that model this complex cellular and environmental interplay and are sensitive to disruption across the multistep signaling cascade. We developed a microplate-based device that supports an epithelium directly overlaid onto an extracellular matrix-embedded mesenchyme, mimicking the basic tissue architecture of developing orofacial tissues. SHH ligand produced from the epithelium generated a gradient of SHH-driven transcription in the adjacent mesenchyme, recapitulating the gradient of pathway activity observed in vivo. Shh pathway activation was antagonized by small molecule inhibitors of epithelial secretory, extracellular matrix transport, and mesenchymal sensing targets, supporting the use of this approach in high-content chemical screening of the complete Shh pathway. Together, these findings demonstrate a novel and practical microphysiological model with broad utility for investigating epithelial-mesenchymal interactions and environmental signaling disruptions in development.

Recent preclinical data suggest that alterations in the gut microbiota may be an important factor linking obesity to vascular dysfunction, an early sign of cardiovascular disease. The purpose of this study was to begin translation of these preclinical data by examining whether vascular phenotypes in humans are transmissible through the gut microbiota. We hypothesized that germ-free mice colonized with gut microbiota from obese individuals would display diminished vascular function compared to germ-free mice receiving microbiota from lean individuals. We transplanted fecal material from obese and lean age-and sex-matched participants with disparate vascular function to germ-free mice. Using Principle Component Analysis, the microbiota of colonized mice separated by donor group along the first principle component, accounting for between 70–93% of the total variability in the dataset. The microbiota of mice receiving transplants from lean individuals was also characterized by increased alpha diversity, as well as increased relative abundance of potentially beneficial bacteria, including Bifidobacterium, Lactobacillus, and Bacteroides ovatis. Endothelium-dependent dilation, aortic pulse wave velocity and glucose tolerance were significantly altered in mice receiving microbiota from the obese donor relative to those receiving microbiota from the lean donor or those remaining germ-free. These data indicate that the obesity-associated human gut microbiota is sufficient to alter the vascular phenotype in germ-free mice in the absence of differences in body weight or dietary manipulation, and provide justification for future clinical trials to test the efficacy of microbiota-targeted therapies in the prevention or treatment of cardiovascular disease.

BackgroundData on the prevalence and correlates of restless legs syndrome (RLS) in people with HIV are limited. This study sought to determine the prevalence of RLS, associated clinical correlates, and characterize sleep-related differences in men with and without HIV.MethodsSleep-related data were collected in men who have sex with men participating in the Multicenter AIDS Cohort Study (MACS). Demographic, health behaviors, HIV status, comorbidities, and serological data were obtained from the MACS visit coinciding with sleep assessments. Participants completed questionnaires, home polysomnography, and wrist actigraphy. RLS status was determined with the Cambridge-Hopkins RLS questionnaire. RLS prevalence was compared in men with and without HIV. Multinomial logistic regression was used to examine correlates of RLS among all participants and men with HIV alone. Sleep-related differences were examined in men with and without HIV by RLS status.ResultsThe sample consisted of 942 men (56% HIV+; mean age 57 years; 69% white). The prevalence of definite RLS was comparable in men with and without HIV (9.1% vs 8.7%). In multinomial regression, HIV status was not associated with RLS prevalence. However, white race, anemia, depression, and antidepressant use were each independently associated with RLS. HIV disease duration was also associated with RLS. Men with HIV and RLS reported poorer sleep quality, greater sleepiness, and had worse objective sleep efficiency/fragmentation than men without HIV/RLS.ConclusionsThe prevalence of RLS in men with and without HIV was similar. Screening for RLS may be considered among people with HIV with insomnia and with long-standing disease.

Sonic hedgehog (Shh) pathway disruption causes craniofacial malformations including orofacial clefts (OFCs) of the lip and palate. In normal craniofacial morphogenesis, Shh signals to multipotent cranial neural crest cells (cNCCs) and was recently discovered to regulate the angiogenic transcriptome, including expression markers of perivascular cells and pericytes. The mural cells of microvasculature, pericytes in the brain and face differentiate from cNCCs, but their role in facial development is not known. Here, we examined microvascular morphogenesis in a mouse model of Shh pathway antagonist-induced cleft lip and the impact of cNCC-specific Shh pathway activation in a cNCC-endothelial cell co-culture system. During cleft pathogenesis in vivo, disrupted microvascular morphogenesis localized with attenuated tissue outgrowth in the medial nasal processes that form the upper lip. In vitro, we found that human umbilical vein endothelial cell (HUVEC) cord formation was not affected by direct Shh pathway perturbation. However, in a co-culture system in which cNCCs directly interact with endothelial cells, cNCC-autonomous Shh pathway activity significantly prolonged endothelial cord network stability. Taken together, these findings support the premise that Shh pathway activation in cNCCs promotes pericyte-like function and microvascular stability. In addition to suggesting a previously unrecognized role for Shh signaling in facial development, these studies also identify perivascular differentiation and microvascular morphogenesis as new focuses for understanding normal and abnormal craniofacial development.

Abstract Cytosolic Malic Enzyme (ME1) provides reduced NADP for anabolism and maintenance of redox status. To examine the role of ME1 in tumor genesis of the gastrointestinal tract, we crossed mice having augmented intestinal epithelial expression of ME1 (ME1-Tg mice) with ApcMin/+ mice to obtain male ApcMin/+/ME1-Tg mice. ME1 protein levels were significantly greater within gut epithelium and adenomas of male ApcMin/+/ME1-Tg than ApcMin/+ mice. Male ApcMin/+/ME1-Tg mice had larger and greater numbers of adenomas in the small intestine (jejunum and ileum) than male ApcMin/+ mice. Male ApcMin/+/ME1-Tg mice exhibited greater small intestine crypt depth and villus length in non-adenoma regions, correspondent with increased KLF9 protein abundance in crypts and lamina propria. Small intestines of male ApcMin/+/ME1-Tg mice also had enhanced levels of Sp5 mRNA, suggesting Wnt/β-catenin pathway activation. A small molecule inhibitor of ME1 suppressed growth of human CRC cells in vitro, but had little effect on normal rat intestinal epithelial cells. Targeting of ME1 may add to the armentarium of therapies for cancers of the gastrointestinal tract.

The phase-sensitive radio-echo sounder (pRES) is a powerful new instrument that can measure the depth of internal layers and the glacier bed to millimetre accuracy. We use a stationary 16-antenna pRES array on Store Glacier in West Greenland to measure the three-dimensional orientation of dipping internal reflectors, extending the capabilities of pRES beyond conventional depth sounding. This novel technique portrays the effectiveness of pRES in deriving the orientation of dipping internal layers that may complement profiles obtained through other geophysical surveying methods. Deriving ice vertical strain rates from changes in layer depth as measured by a sequence of pRES observations assumes that the internal reflections come from vertically beneath the antenna. By revealing the orientation of internal reflectors and the potential deviation from nadir of their associated reflections, the use of an antenna array can correct this assumption. While the array configuration was able to resolve the geometry of englacial layers, the same configuration could not be used to accurately image the glacier bed. Here, we use simulations of the performance of different array geometries to identify configurations that can be tailored to study different types of basal geometry for future deployments.

The catchments of Pine Island Glacier and Thwaites Glacier in the Amundsen Sea Embayment are two of the largest, most rapidly changing, and potentially unstable sectors of the West Antarctic Ice Sheet. They are also neighboring outlets, separated by the topographically unconfined eastern shear margin of Thwaites Glacier and the southwest tributary of Pine Island Glacier. This tributary begins just downstream of the eastern shear margin and flows into the Pine Island ice shelf. As a result, it is a potential locus of interaction between the two glaciers and could result in cross-catchment feedback during the retreat of either. Here, we analyze relative basal reflectivity profiles from three radar sounding survey lines collected using the UTIG HiCARS radar system in 2004 and CReSIS MCoRDS radar system in 2012 and 2014 to investigate the extent and character of ocean access beneath the southwest tributary. These profiles provide evidence of ocean access ~12 km inland of the 1992–2011 InSAR-derived grounding line by 2014, suggesting either retreat since 2011 or the intrusion of ocean water kilometers inland of the grounding line.

Abstract Background The evolutionarily conserved Sonic Hedgehog (Shh) signaling pathway is essential for embryogenesis and orofacial development. SHH ligand secreted from the surface ectoderm activates pathway activity in the underlying cranial neural crest cell (cNCC)-derived mesenchyme of the developing upper lip and palate. Disruption of Shh signaling causes orofacial clefts, but the biological action of Shh signaling and the full set of Shh target genes that mediate normal and abnormal orofacial morphogenesis have not been described. Results Using comparative transcriptional profiling, we have defined the Shh-regulated genes of the cNCC-derived mesenchyme. Enrichment analysis demonstrated that in cultured cNCCs, Shh-regulated genes are involved in smooth muscle and chondrocyte differentiation, as well as regulation of the Forkhead family of transcription factors, G1/S cell cycle transition, and angiogenesis. Next, this gene set from Shh-activated cNCCs in vitro was compared to the set of genes dysregulated in the facial primordia in vivo during the initial pathogenesis of Shh pathway inhibitor-induced orofacial clefting. Functional gene annotation enrichment analysis of the 112 Shh-regulated genes with concordant expression changes linked Shh signaling to interdependent and unique biological processes including mesenchyme development, cell adhesion, cell proliferation, cell migration, angiogenesis, perivascular cell markers, and orofacial clefting. Conclusions We defined the Shh-regulated transcriptome of the cNCC-derived mesenchyme by comparing the expression signatures of Shh-activated cNCCs in vitro to primordial midfacial tissues exposed to the Shh pathway inhibitor in vivo. In addition to improving our understanding of cNCC biology by determining the identity and possible roles of cNCC-specific Shh target genes, this study presents novel candidate genes whose examination in the context of human orofacial clefting etiology is warranted.

Abstract Background Type 2 diabetes (T2D) is associated with generalized vascular dysfunction characterized by increases in large artery stiffness, endothelial dysfunction, and vascular smooth muscle dysfunction. Sodium glucose cotransporter 2 inhibitors (SGLT2i) represent the most recently approved class of oral medications for the treatment of T2D, and have been shown to reduce cardiovascular and overall mortality. Although it is currently unclear how SGLT2i decrease cardiovascular risk, an improvement in vascular function is one potential mechanism. The aim of the current study was to examine if dapagliflozin, a widely prescribed STLT2i, improves generalized vascular dysfunction in type 2 diabetic mice. In light of several studies demonstrating a bi-directional relation between orally ingested medications and the gut microbiota, a secondary aim was to determine the effects of dapagliflozin on the gut microbiota. Methods Male diabetic mice (Db, n = 24) and control littermates (Con; n = 23) were randomized to receive either a standard diet or a standard diet containing dapagliflozin (60 mg dapagliflozin/kg diet; 0.006%) for 8 weeks. Arterial stiffness was assessed by aortic pulse wave velocity; endothelial function and vascular smooth muscle dysfunction were assessed by dilatory responses to acetylcholine and sodium nitroprusside, respectively.
