The potentially toxic diatom genus Pseudo-nitzschia and particulate domoic acid (pDA) concentrations were investigated weekly to bi-weekly from October 2013 to September 2014 in the Golden Horn Estuary. Pseudo-nitzschia species were detected in 324 of the 666 samples (approximate to 49%). Two species, Pseudo-nitzschia calliantha and Pseudo-nitzschia pungens, were identified based on the scanning electron microscopy examination. A bloom of P. calliantha occurred between May and June, average cell density, 1.1x10(6) cellsL(-1) in May. Pseudo-nitzschia calliantha was commoner than P. pungens almost throughout the study period. They occurred over a wide temperature range (7.4-26.3 degrees C) and moderate salinity (5.9-19.9). The highest cell density occurred at 14.7-19.6 degrees C and 18.5-19.0 salinity, in the lower and middle estuary in winter and late spring. Pseudo-nitzschia species were found in very low densities in the upper estuary, which has variable salinity and low light availability due to high concentrations of suspended material. The pDA concentration in seawater was 0.03-1.0 mu gL(-1). The distribution pattern of pDA coincided with the presence of Pseudo-nitzschia. Bu çalışmanın ana amacı Haliç' te var olan Pseudo-nitzschia türlerinin domoik asit (DA) üretip üretmediğinin belirlenmesi ve domoik asitin çevresel faktörler ile ilişkisinin belirlenmesidir. Bu amaç doğrultusunda; Amnezik Kabuklu Zehirlenmesine (ASP) neden olan, bazı Pseudo-nitzschia türlerinin salgıladığı önemli bir biyotoksin olan Domoik Asit (DA) dağılımı, İstanbul Haliç'i (Altın Boynuz) üst tabaka suyunda 1 yıl boyunca (Ağustos 2011 – Temmuz 2012) partiküler maddede ve fitoplanktonda Yüksek Performanslı Sıvı Kromatografisi-Floresans Dedeksiyonu (HPLC-FLD) yöntemiyle belirlenmiştir. Örnekleme noktalarına ait fiziksel, kimyasal ve biyolojik faktörler ile domoik asit miktarları arasındaki ilişki korelasyon analiziyle, DA miktarı ve dağılımını etkileyen nedenler ise Temel Bileşenler Analizi (PCA) ile değerlendirilmiştir. Çalışma periyodu içerisinde domoik asit miktarları, partiküler maddede 0,21-58,17 µg/L ve fitoplanktonda 0,76-94,34 µg/L konsantrasyonları arasında değişim göstermiştir. En yüksek biyotoksin miktarları Mayıs ayında 58,17 µg/L ve 94,34 µg/L olarak Haliç orta bölümde yer alan FD3 istasyonunun 10 m derinliğinde tespit edilmiştir. Domoik asitin varlığı Pseudo-nitzschia spp. bolluğu, sıcaklık ve tuzluluk değişkenleri ile ilişkilendirilmiştir. Ayrıca, ortamdaki silikat ve fosfatın sınırlayıcı etkisi ile yüksek azot konsantrasyonları domoik asit üretimini artırıcı etkide bulunmuştur. Amnezik Kabuklu Zehirlenmesine neden olan domoik asidin denizel canlılarda ve insanlarda ölüme sebebiyet vermesi dolayısıyla, düzenli veri setlerinin oluşturulabilmesi için bu yeni parametrenin su kalitesi izleme programlarına dahil edilmesi önerilmektedir. İstanbul Haliç' inde ilk kez gerçekleştirilen bu çalışma ile bölgesel ve global literatüre katkıda bulunulmuştur. The main goal of this study is to determine the relationship between domoic acid (DA) and environmental factors in Golden Horn Estuary. For this purpose; high performance liquid chromatography-fluoresence detection method (HPLC-FLD) was used to determine the distribution of particulate and plankton net concentrations of Domoic Acid (DA), which is an important biotoxin excreted by Pseudo-nitzschia spp. and causes Amnesic Shellfish Poisoning (ASP), in the upper layer water of the Golden Horn Estuary, Istanbul, between August 2011 and July 2012. The relationship between DA concentrations and environmental factors (physical, chemical and biological) was analysed with Spearman correlation and the forcing factors of the DA distribution were evaluated with Principal Component Analysis (PCA). In the study period, domoic acid concentrations ranged from 0.21 to 58.17 µg/L in the particulate matter and 0.76 - 94.34 µg/L in plankton net samples.. The highest biotoxin concentrations 58.17 µg/L and 94.34 µg/L were found in 10 m depth of FD3 station, in the middle section of the estuary, in May. Toxin presence may be related to the Pseudo-nitzschia spp. abundance, salinity and temperature. The limiting effect of the silicate and phosphate with high nitrogen concentrations evaluated as associated promoting factors with DA production.In order to consider ASP poisoning caused by domoic acid has lethal effect on human and marine animals, it is recommended that domoic acid should be included in water pollution monitoring programs, to constitute permanent data sets, as a new parameter. This study provided the first detection of domoic acid in the Golden Horn Estuary and contributes to the knowledge at regional and global scales. 134

Biyosensör teknolojileri Çevre Mühendisliğinde kullanılan izleme parametreleri arasında da hızla yerini almaktadır. Son yıllarda özellikle fenol ve türevlerinin daha güvenli, seçici, hassas ve hızlı ölçülmesi amacıyla fenol biyosensörlerinin geliştirilmesine yönelik literatürde çok sayıda çalışmaya rastlanmaktadır. Bu biyosensörler çalışma elektrotlarının ölçüm performansını artıracak şekilde çeşitli parametrelerin düzenlenmesiyle yeniden tasarlanmakta ve sonuçlar birbiriyle kıyaslanmaktadır.Amperometrik fenol biyosensörleri için gelinen noktada özellikle enzimin elektrot yüzeyine tutuklanmasını ve stabil bir çalışma elektrodu elde edilmesini sağlayan polimerik kompozit filmin tasarlanması başlıca çalışma alanıdır.Bu tez çalışmasında medyatör bağlı (vinilferrosen) 2 farklı polimer kullanılarak çalışma elektrotları tasarlanarak fenol ve türevlerinin ölçümünde denenmiştir.Kullanılan polimerler Poly(GMA-co-VFc) ve Poly(MTM-co-VFc)dir.1. tip çalışma elektrodunda tyrosinase enzimi polimer üzerine kimyasal olarak bağlanmıştır.2. tip elektrotta ise tyrosinase enzimi tiyofen ve polipirol arasında oluşturulan kopolimer aracılığı ile tuzağa düşürülerek tutuklanmıştır. . Hazırlanmış olan çalışma elektrotları kullanılarak geliştirilen biyosensörler kesikli sistemle işletilmiştir. Çalışma şartları optimize edilerek (pH, Potansiyel), çalışma elektrotları fenol ve türevleri için denenmiştir. Çalışma kapsamında kullanılan fenol ve türevleri için gerekli kalibrasyon eğrileri çizilmiş, bu kalibrasyon eğrileri kullanılarak ölçüm analitik parametreleri olan hassasiyet, lineer aralık ve alt belirleme limitleri hesaplanmıştır. Analitik parametrelere göre biyosensörlerin performansı, ölçüm güvenilirliği,tartışılmıştır. Biosensor technologies rapidly take place at following parameters which is used in Environmental Engineering applications. Recently a lot of study are found on literature to detect phenol and phenol derivatives more confident, selective, sensitive and fast. These biosensors are re-designed with restoring various parameters to get more detection performance of working electrodes and the results are comparised. At this point the goal is for amperometric phenol biosensors designing polymeric composite film which is supportive to get stabile working electrode and entrap the enzyme (tyrosinase) on the surface of the electrode. At this study, phenol and phenol derivatives were detected with using two different mediator (vinylferrocene) bounded polymers. The polymers are Poly(GMA-co-VFc) and Poly(MTM-co-VFc). At the first working electrode tyrosinase was chemically bounded on Poly(GMA-co-VFc). On the other hand, at the second working electrode, tyrosinase was entrapped with a thiophene and polypyrol copolymer. Working electrodes were operated at batch system. Working conditions (pH, potential) were optimized and the electrodes were tried for phenol and phenol derivatives? detection. For phenol and phenol derivatives , calibration curves were constructed and the sensitivity, lineer range and detection limit which are the analytical parameters of biosensor were calculated from these calibration curves. The performance of biosensors and the reliability of the measurement were discussed. 112

Particulate domoic acid (pDA) concentrations, Pseudo-nitzschia abundances and their relationship with environmental variables were investigated from August 2011 to July 2012 in the Golden Horn Estuary (GHE), Turkey. To determine the distribution of pDA concentrations, the high performance liquid chromatography - fluoresence detection (HPLC-FLD) method was used (detection limit 0.2 mu g mL(-1)). pDA concentrations ranged from 0.24 to 21.03 mu g mL(-1) and the highest value was measured at FD3 station, located in the middle estuary, in May 2012. pDA levels were undetectable between Aug. 2011 and March 2012 period in all stations, except in Nov. 2011 (0.83 mu g mL(-1), at FD3 station). pDA values reached the maximum value when Pseudo-nitzschia abundance was counted at the highest level (1968103 cells L-1, at FD3 station). The relationship between pDA and environmental parameters was investigated by Spearman correlation analysis and the influencing factors of the pDA distribution were evaluated with Principal Component Analysis (PCA). According to the results, pDA production is mostly controlled by temperature, SiO2 and NO3+NO2. This study reports the first results of DA in the GHE and these results may be used to consider the probability of fmding similar conditions in the Turkish seas.
