Abstract

Clostridium butyricum cells grown on glycerol show a higher carbon flow than those grown on glucose, as the NADH formed during metabolism is reoxidized by both the butyric and propanediol (PD) pathways. During growth on glycerol, glycerol dehydrogenase, 1,3-PD dehydrogenase and diol dehydratase regulate the flow of carbon to the pathways that form either acetate and butyrate, or 1,3-PD. During growth on glucose, the level of acetyl-CoA and NADH in the cells increases.

Isolated Clostridium butyricum DSM 5431 mutants have increased resistance to glycerol and 1,3-propanediol (1,3-PD) and produce more biomass compared to the wild type. The wild-type strain produces more acid than the mutant strain and releases more acetic acid than butyrate. In contrast to the wild type, the mutants use more NADH to form 1,3-PD. The mutant strains have low levels of hydrogenase and NADH-ferrodoxin activities and an increased level of ferrodoxin-NAD+ reductase activity.

8. Indium catalysts for low-pressure CO2/epoxide ring-opening copolymerization: Evidence for a mononuclear mechanism?

Abstract

The alternating copolymerization of CO2/epoxides is a useful means to incorporate high levels of carbon dioxide into polymers. The reaction is generally proposed to occur by bimetallic or bicomponent pathways. Here, the first indium catalysts are presented, which are proposed to operate by a distinct mononuclear pathway. The most active and selective catalysts are phosphasalen complexes, which feature ligands comprising two iminophosphoranes linked to sterically hindered ortho-phenolates. The catalysts are active at 1 bar pressure of carbon dioxide and are most effective without any cocatalyst. They show low-pressure activity (1 bar pressure) and yield polymer with high carbonate linkage selectivity (>99%) and isoselectivity ( Pm > 70%). Using these complexes, it is also possible to isolate and characterize key catalytic intermediates, including the propagating indium alkoxide and carbonate complexes that are rarely studied. The catalysts are mononuclear under polymerization conditions, and the key intermediates show different coordination geometries: the alkoxide complex is pentacoordinate, while the carbonate is hexacoordinate. Kinetic analyses reveal a first-order dependence on catalyst concentration and are zero-order in carbon dioxide pressure; these findings together with in situ spectroscopic studies underpin the mononuclear pathway. More generally, this research highlights the future opportunity for other homogeneous catalysts, featuring larger ionic radius metals and new ligands, to operate by mononuclear mechanisms.

11. SIMPLE: A Model for SIMulation of PLant Emergence Predicting the Effects of Soil Tillage and Sowing Operations

Abstract

A model predicting seedling emergence is described and applied to sugarbeet (Beta vulgaris L.). The input variables are the soil surface texture, soil temperature, rainfall, aggregate size distribution and position in the seedbed, sowing depths, characteristics of the seeds (initial seed mass distribution, germination time, and hypocotyi elongation distributions). A three-dimensional seedbed layer is created where the aggregates and seeds are placed. Soil water content is assumed not to limit sugarbeet emergence (sowing conditions in northern Europe). The time needed to reach the soil surface is calculated using germination thermal time, soil temperature, the presence or absence of aggregates, and the hypocotyl elongation function. The ability of seedlings to break through the soil surface is a function of crust development and moisture. The seedling growth after emergence is calculated with reference to seed mass distribution, emergence delay, and the presence or absence of mechanical obstacles. The emergence prediction was tested in field experiments with four seedbeds, from fine earth to cloddy structure, and a sowing depth of 17 to 35 mm. The predicted number and sizes of clods encountered by seedlings and the calculated hypocotyi length were not significantly different from measured ones. Predicted germination times were longer than the observed ones (differences [is less than]5 [degrees] C d); final rates were well predicted. Predicted vs. measured final emergence rates differed by less than 10%; changes with time differed from 15 to 30 [degrees] C. This was due to the hypocotyl elongation functions, which must be improved. Further improvements will be to predict soil water content variations and effects on emergence via water stress and soil strengthening.

Abstract

Little information is available on the distribution and shape of aggregates or their spatial arrangement within seedbeds, although it influences crop emergence. The aim of this study was to obtain quantitative information on seedbed structure by sampling soil layers, sieving, surface photographs, and stereological analysis of embedded sample sections. Seedbeds prepared with a combined implement (spring tine cultivator and rollers) in a silt loam soil (Typic Hapludalf) with two seeders (K and PI) on plowed (P), not plowed (NP), and not plowed and compacted (NPC) plots, were analyzed. Aggregates were described by measurements of L, l, h, the longest, intermediate and shortest axes and visually classified according to their surface roughness (smooth, 0; to rough, 4). The D fitting parameter of the power-law aggregate size distribution after seeding was significantly smaller (3.4) on NPC than on the other plots (3.8-4.0). The aggregate aspect ratios (l/L and h/L) did not vary with aggregate size or plot, their overall means were 0.77 and 0.55 respectively. Aggregates of roughness classes [is greater than] 2 were 28 to 32% on NPC and NP plots, compared with 74% on P plot. Seeders reduced the number of aggregates [is greater than] 10 mm by one-third compared with the values before seeding. Seeder K placed over 60% of the aggregates [is greater than] 20 mm on the soil surface. Aggregates were uniformly distributed across the row by K seeder and away from the center of the row by Pi seeder. The information obtained will be used in a computerized seedbed generator.

Abstract

A method is described for isolating epidermal protoplasts from the primary leaves of barley (Hordeum vulgare L.). Epidermal protoplasts are lighter than mesophyll protoplasts because of their smaller ratio of cytoplasm to vacuole, and can be separated from the latter by density-gradient centrifugation after complete digestion of the leaves. We have started a basic characterization of the epidermal protoplast fraction in comparison with mesophyll protoplasts. Epidermal protoplasts had a mean diameter of 63.5-mu-m, whereas that of mesophyll protoplasts was 35.7-mu-m. Their respiratory oxygen consumption was not influenced by light. They contained acid hydrolases and cytoplasmic enzymes in relative activities different from those of mesophyll protoplasts. Their polypeptide pattern as judged from two-dimensional separations was, in principle, similar to that of mesophyll cells after elimination of the plastids from the latter by the preparation of vacuoplasts. However, in addition, a considerable number of epidermis-specific polypeptides were observed. Isolated epidermal protoplasts were viable and efficiently incorporated [S-35]methionine into newly synthesized proteins. The results show that epidermal protoplasts are suitable for the investigation of the physiological and molecular properties of epidermal cells in leaves.

Abstract

The cation and anion distribution between the epidermis and mesophyll of primary leaves of 10-d-old barley (Hordeum vulgare L.) seedlings was studied in relation to growth conditions. A new method was employed to isolate epidermal protoplasts. The following observations were made: (i) Under standard hydroponic growth conditions, K+ was the dominant cation and NO3- the predominant anion, both in epidermal and mesophyll protoplasts. (ii) Levels of Cl- and particularly of Ca2+ were specifically increased in the epidermis when growth conditions stimulated uptake of these ions into the plants. (iii) Epidermal PO4(3-) was maintained at a very low level even in the presence of 50 mM phosphate in the rooting medium, whereas the phosphate concentration in the mesophyll was high, with phosphate being accumulated in the mesophyll vacuoles. (iv) Sulphate and NO3- accumulated to a similar degree in the mesophyll and the epidermis when K2SO4 or KNO3 in the rooting medium caused salt stress to the plants. (v) Epidermal protoplasts took up (SO4(2-))-S-35 and Cl-36- from the surrounding medium. A comparison of the uptake rates indicates that specific ion deposition into the epidermis may partially be the consequence of differential uptake of ions from the transpiration stream. The results indicate an important function of the epidermis for ion compartmentation in barley leaves.

Abstract

River corridors in urban environments provide areas of biodiversity which are important for both aesthetic and economic reasons. A physical habitat model, which was used to assess urban rivers in Birmingham, UK, was applied to pairs of selected reaches to represent differing levels of habitat diversity on three rivers. The results for different life-stages of dace, roach and chub suggest that the worst physical habitat occurs in highly modified channels and at the highest flows. Four scenarios, which were designed to represent alterations in flow regime caused by changes in management practices, were calculated using the hydrological model Changes in physical habitat created by changes to the flow regime were assessed using a consistent, replicable method. It was shown that an increase in runoff would have detrhymental effects in all cases, and that less engineered sites would benefit more from flow reductions. The lack of a suitable habitat for fry is shown to be a limiting factor for fish at all sites. [ABSTRACT FROM AUTHOR]
