Abstract

International audience; A transparent fire protective sol-gel coating deposited on wood has been evaluated using different fire tests, such as mass loss cone, mini single burning item and furnace tests. The heat release rate and total heat release during combustion, but also the flame spread behavior and the thermal protection performances of the coatings have been studied. Two processes to apply the sol-gel coating on wood have been assessed to show the thickness dependence of the fire performance. When deposited as a thick coating (800 μm thick) on wood, the sol-gel coating lowers the total heat release, decreases the flame spread and greatly improves the thermal protection of the substrate.

Abstract

Abstract Background The aim of environmental risk assessment (ERA) for pesticides is to protect ecosystems by ensuring that specific protection goals (SPGs) are met. The ERA follows a prospective tiered approach, starting with the most conservative and simple step in risk assessment (RA) (so-called tier 1) using the lowest available appropriate endpoint derived from ecotoxicological tests. In 2015, for the tier 1 RA of aquatic primary producers, the recommendation was changed from using the lowest of the 50% inhibition (EC50) values based on biomass (area under the curve—EbC50), increase in biomass (yield- EyC50) or growth rate (ErC50) to only using the growth rate inhibition endpoint (ErC50) because it is independent of the test design and thus more robust. This study examines the implications of this such on the level of conservatism provided by the tier 1 RA and evaluates whether it ensures a suitable minimum protection level. Results Our analysis shows that replacing the lowest endpoint with the growth rate inhibition endpoint while maintaining the assessment factor (AF) of 10 significantly reduces the conservatism in the tier 1 RA. Comparing protection levels achieved with different endpoints reveals that the current assessment is less protective. To maintain the previous level of protection, and since the protection goals have not changed, we recommend to multiply the default AF of 10 by an extra factor of minimum 2.4 in the tier 1 RA based on ErC50. Independently of the endpoint selected in tier 1 RA, several issues in the general RA of pesticides contribute to uncertainties when assessing the protection levels, e.g., lack of appropriate comparison of the higher tier experimental studies (i.e., best achievable approximation of field situation, so-called surrogate reference tier) with field conditions or the regulatory framework's failure to consider realistic conditions in agricultural landscapes with multiple stressors and pesticide mixtures. Conclusions We advise to consider adjusting the risk assessment in order to reach at least the previous protection level for aquatic primary producers. Indeed continuing using an endpoint with a higher value and without adjustment of the assessment factor is likely to jeopardize the need of halting biodiversity loss in surface waters.

Abstract

In this study, the poly-(L-lactide) (PLLA)-degrading enzyme from the thermophilic filamentous bacterium, Laceyella sacchari LP175 was characterized for application in biological recycling process. The gene encoding the PLLA-degrading gene (plla_lp175) was cloned and expressed in Escherichia coli. The native protein comprises 383 amino acids with a molecular mass and pI of 39.45 kDa and 8.26, respectively. The recombinant protein was purified by one-step purification, and was found to have a molecular weight of 28 kDa. Functional expression of PLLA_LP175 in E. coli with a pMAL-c5X vector enhanced the expression of PLLA-degrading enzyme up to 756 U/mg of protein, which is a 2.3-fold increase compared to the native strain. The purified recombinant protein is active in the range of pH 7.0-9.0 and 45-60 degrees C, with optimum activity at pH 9.0 and 60 degrees C. The recombinant enzyme could hydrolyse PLLA objects to a different extent depending on PLLA composition, at pH 9.0 and 50 degrees C within 24 h. Monomeric lactic acid was detected as the product from enzymatic degradation of PLLA objects. A scanning electron micrograph, showing a rough surface with holes of all PLLA objects after treatments with the recombinant enzyme, confirmed the ability of this enzyme to degrade PLLA objects.

Abstract

Chromatographic techniques isolate two isoforms of metal-binding proteins from two species of cadmium-injected flatfish, Limanda limanda and Microstomus kitt. Purification of the amino acid composition of these proteins reveals the presence of metallathioneins. The injection path has no effect on protein nature and one of the protein isoforms from each species exhibits homologous ionic characteristics.

Abstract

Raw starch degrading enzyme (RSDE) produced by Laceyella sacchari LP175 was purified 14.7 fold to a 40.5 % yield. The first 15 N-terminal amino acids were sequenced and showed 100% homology with alpha-amylase from Laceyella sp. DS3 and Thermoactinomyces vulgaris. The RSDE gene was functionally annotated with the Laceyella sacchari strain GS1-1 available genome, which showed the presence of a putative gene of 1362 bp encoding 453 amino acids. The RSDE gene was amplified from Laceyella sacchari LP175 genomic DNA and cloned for expression in Escherichia coli, which showed the highest activity on raw cassava starch at pEl 6.5 and a temperature of 50 degrees C. Homology structure analysis revealed the presence of three domains that are conserved among the structures of GH13 alpha-amylases, where the active and binding sites both play an important role in starch hydrolysis. The recombinant LsA175 could hydrolyze raw cassava starch at below gelatinization temperature, and showed higher efficiency for hydrolysis than commercial alpha-amylase (Termamyl((R))) at 50 degrees C. This show's the possibility for application of recombinant LsA175 at an industrial level, particularly in terms of energy consumption savings.

It deals with vertical fire resistance test (fire protection of steel according to UL 1709 and ISO 834), test for building products (single burning item or SBI according to EN 13823) and the environmental test procedure for airborne equipment (resistance to fire of materials in designated fire zones according to ISO 2685). It covers different types of material (composites, steel and wood) and several applications (wooden panel, composite and steel for building and carbon fiber reinforced composite for aviation).
