In the present study, follicles were classified according to the morphology of their granulosa cells. Type B follicles contained only flattened granulosa cells; type B/C follicles had a mixture of flattened and cuboidal granulosa cells in a single layer, and type C follicles had a single layer of cuboidal granulosa cells. The primary objectives of the study were to determine whether 5-bromo-2-deoxyuridine incorporation into type B/C follicles was a marker for initiation of growth and how long type B/C follicles could remain at the same stage before transformation to type C follicles. Female Holtzman rats received bromo-deoxyuridine for 7 days. After the infusion (day minipumps were removed = day 0), rats were ovariectomized on days 0 (n = 9), 30 (n = 8), 90 (n = 8) and 150 (n = 9). The numbers of type B, B/C and C follicles within one ovary were determined using modified fractionator counting. Analysis over all times demonstrated that there were more (P < 0.0001) type B/C (941 +/- 61 per ovary) than type C (140 +/- 18 per ovary) or type B (159 +/- 19 per ovary) follicles. The numbers of type B and type C follicles did not differ from each other at any time. Only one of 34 rats evaluated had bromo-deoxyuridine-labelled type B follicles. On day 150, 57% of the bromo-deoxyuridine-labelled type B/C follicles remained from day 0. It is concluded that (1) DNA synthesis in granulosa cells of type B/C follicles is not a reliable indicator of impending growth; and (2) type B and type B/C follicles are both components of the pool of primordial follicles.

Unlabelled: The number of primordial follicles within an ovary is frequently determined by counting 5, 7 or 10 microns thick sections and multiplying by the fraction of sections counted and a correction factor to adjust for duplicate counts. The objectives of the present study were: (i) to evaluate the accuracy of the correction factor developed by Abercrombie (1946); (ii) to evaluate the accuracy of the classification of primordial follicles from single tissue sections; and (iii) to determine the incorporation rate of 5-bromo-2-deoxyuridine into primordial follicles. In Expt 1, rat ovaries were sectioned at a thickness of 5, 7 or 10 microns. Primordial follicles were counted and classified across ten adjacent ovarian sections. The percentage of primordial follicles from single sections that were counted twice was 10, 9 and 2% in 5, 7 and 10 microns sections, respectively. This was lower than predicted by Abercrombie's method. The major error in counting from single sections was classification of early primary follicles as primordial follicles (55, 33 and 3% in 5, 7 and 10 microns sections, respectively). In Expt 2, a mean of 12 +/- 7% of primordial follicles incorporated 5-bromo-2-deoxyuridine after infusion for 7 days (four of seven rats had no labelled primordial follicles)., In Conclusion: (i) Abercrombie's correction factor should not be used for adjusting counts of follicles; (ii) evaluation of primordial follicles from single sections gives inaccurate counts and incorrect classification is of greater importance than duplicate counting, particularly in thinner sections; (iii) for evaluation of the number of follicles, 10 microns is the optimal thickness; and (iv) primordial follicles incorporated 5-bromo-2-deoxyuridine infrequently.

The primary objective of these studies was to determine whether unilateral ovariectomy (ULO) would affect rate of loss of primordial follicles. In experiment 1, retired breeder rats, unilaterally ovariectomized and maintained on the experiment for 90 days after surgery, had fewer (p less than 0.01) primordial follicles per ovary than sham-operated controls of the same age. The purpose of experiment 2 was to determine whether time after ULO or age of rats was the critical factor necessary for increased loss of primordial follicles found after ULO in experiment 1. It was found that age was more important than time: when ULO was performed at 30 days of age, the number of primordial follicles did not decrease in ULO rats compared to controls (p greater than 0.05) before 250 days of age. Concentrations of FSH during metestrus were not greater (p greater than 0.05) in ULO rats than in controls until rats were 250 days old. There were also fewer (p less than 0.05) growing follicles per ovary in ULO than in sham-operated rats at 250 days of age. It is concluded that ULO can increase the loss of primordial follicles, but only in old rats (greater than or equal to 250 days of age).

The purpose of this study was to determine whether or not there was a decrease in skeletal muscle fibre number in the rat from the early postnatal period to adulthood. Twelve male Sprague-Dawley rats were used in the study. At 25 days of age the animals underwent surgical removal of the soleus and tibialis anterior muscles from one hindlimb. At 180 days of age, 6 of the animals were killed and the soleus and tibialis anterior muscles from the contralateral leg were removed. The same procedure was carried out on the remaining 6 animals at 365 days of age. Fibre number was determined by the nitric acid digestion method for the soleus muscles and the mean fibre dry weight method for the tibialis anterior muscles. There was no difference in fibre number for either muscle between 25 days of age and 180 days of age or between 25 days of age and 365 days of age. The results of this study indicate that there is no change in skeletal muscle fibre number from the early postnatal period until adulthood in the rat.

The purpose of this study was to describe the soleus muscle fiber number variation within litter and within strain of three lines of laboratory rodents in an effort to determine the value of these lines in muscle fiber number research. The three lines of laboratory rodents used for the study were inbred mice (18 litters, 113 animals), outbred mice (8 litters, 97 animals), and inbred rats (14 litters, 126 animals). The animals were sacrificed at approximately 7 weeks of age and the soleus muscle from the right leg was removed for fiber number determination. Fiber number was determined by the nitric acid digestion method. The mean percent difference between the animals with the highest and lowest fiber number within litters was 14.4%, 13.0%, and 18.3% for inbred mice, outbred mice, and inbred rats, respectively. This was substantially less than the 38.2%, 27.8%, and 49.1% differences, respectively, that existed within strain. The results of this study indicate that fiber number variation within litter and within strain in the three lines of laboratory rodents used in this study would make comparison of fiber number between animals difficult to interpret.

The effect of increased growth rate during the suckling period on subsequent body growth rate and muscle weight to body weight (MW/BW) ratio was examined in inbred and outbred male and female mice. Growth rate during the suckling period was increased by reducing litter size to 4 pups within six hours of birth. Body weight, MW/BW ratio, and dry weight to wet weight (DW/WW) ratio for the soleus, tibialis anterior, extensor digitorum longus, biceps brachii, and heart muscles were measured at 4, 8, 16, and 24 weeks of age. The results indicate that increasing growth rate during the suckling period results in an increased body weight at 24 weeks of age in outbred male and female mice; whereas, in inbred male and female mice body weight is greater at 4 weeks of age but by 8 weeks of age the mice raised in normal litters have "caught up" with those raised in small litters. MW/BW ratio is increased in several muscles during the suckling period but returns to normal during the post weaning period.
