Abstract

National audience; Le site d’Erbrée « La Huperie » en Ille-et-Vilaine, a fait l’objet d’une fouille durant l’hiver 2014-2015 à l’emplacement d’une future plate-forme logistique d’Inter-Marché. Elle fait suite à un diagnostic réalisé en 2014 (Aubry, 2014). Le projet initial portant sur 24 ha, ce sont 6,25 ha qui ont été décapés dans le cadre de cette fouille. L’ensemble est situé sur un plateau à une altitude moyenne de 133 m NGF, dominant un réseau hydrologique assez fourni et reposant sur des formations limoneuses éoliennes venant recouvrir des altérites du Briovérien. Il se présente sous la forme d’une ferme indigène isolée, dont l’occupation est attestée du Hallstatt final jusqu’à la fin de La Tène finale. Quelques rares indices laissent penser à une présence humaine à l’époque antique et médiévale, sans qu’il soit possible de parler de perduration et sans que le diagnostic ait permis de localiser leur emplacement.

Abstract

Adaptive behaviour entails the capacity to select actions as a function of their energy cost and expected value and the disruption of this faculty is now viewed as a possible cause of the symptoms of Parkinson's disease. Indirect evidence points to the involvement of the subthalamic nucleus-the most common target for deep brain stimulation in Parkinson's disease-in cost-benefit computation. However, this putative function appears at odds with the current view that the subthalamic nucleus is important for adjusting behaviour to conflict. Here we tested these contrasting hypotheses by recording the neuronal activity of the subthalamic nucleus of patients with Parkinson's disease during an effort-based decision task. Local field potentials were recorded from the subthalamic nucleus of 12 patients with advanced Parkinson's disease (mean age 63.8 years ± 6.8; mean disease duration 9.4 years ± 2.5) both OFF and ON levodopa while they had to decide whether to engage in an effort task based on the level of effort required and the value of the reward promised in return. The data were analysed using generalized linear mixed models and cluster-based permutation methods. Behaviourally, the probability of trial acceptance increased with the reward value and decreased with the required effort level. Dopamine replacement therapy increased the rate of acceptance for efforts associated with low rewards. When recording the subthalamic nucleus activity, we found a clear neural response to both reward and effort cues in the 1-10 Hz range. In addition these responses were informative of the subjective value of reward and level of effort rather than their actual quantities, such that they were predictive of the participant's decisions. Abstract

Recent studies have suggested that the inadequate evaluation of the cost-benefit ratio of actions in Parkinson Disease (PD), leads to a suboptimal allocation of resources, which would be responsible for the bradykinesia. In addition there is evidence indicating that the Subthalamic Nucleus (STN) is involved in motivated behaviours and in reward-related processes. Starting from these considerations, we tried to determine the extent to which variables that are critical to cost-benefit computation can be identified in the neural activity of STN. We also evaluated how the dopamine level affects the representation of these variables and how these changes relate to the behavioral deficits in PD. To address these issues, we recorded local field potentials (LFP) in the STN of 7 PD patients having benefited from the implantation of deep brain stimulation (DBS) electrodes. During these recordings, the patients were asked to perform a task requiring to squeeze a dynamometer with variable force and with a promise of a variable virtual monetary reward. In each trial, the patients were allowed to choose to either perform or skip the effort proposed as a function of effort intensity and reward amount. These two pieces of information were provided by visual cues at the beginning of each trial. Each patient was tested twice, ON and OFF dopamine replacement therapy. Preliminary results indicate that robust responses to the different visual cues are present in STN and that these responses are strongly modulated by reward, effort and choice variables. The nature of these responses differs according to the frequency bands. Responses in the beta range are inversely proportional to the probability of choosing to execute the effort whereas gamma range signals show the opposite effect.
