The ability to form precise, episodic memories develops with age, with young children only able to form gist-like memories that lack precision. The cellular and molecular events in the developing hippocampus that underlie the emergence of precise, episodic-like memory are unclear. In mice, the absence of a competitive neuronal engram allocation process in the immature hippocampus precluded the formation of sparse engrams and precise memories until the fourth postnatal week, when inhibitory circuits in the hippocampus mature. This age-dependent shift in precision of episodic-like memories involved the functional maturation of parvalbumin-expressing interneurons in subfield CA1 through assembly of extracellular perineuronal nets, which is necessary and sufficient for the onset of competitive neuronal allocation, sparse engram formation, and memory precision.

Brucella melitensis (B. melitensis) is an important facultative intracellular bacterium that causes global zoonotic diseases. Continuous intracellular survival and replication are the main obstruction responsible for the accessibility of prevention and treatment of brucellosis. Bacteria respond to complex environment by regulating gene expression. Many regulatory factors function at loci where RNA polymerase initiates messenger RNA synthesis. However, limited gene annotation is a current obstacle for the research on expression regulation in bacteria. To improve annotation and explore potential functional sites, we proposed a novel genome-wide method called Capping-seq for transcription start site (TSS) mapping in B. melitensis. This technique combines capture of capped primary transcripts with Single Molecule Real-Time (SMRT) sequencing technology. We identified 2,369 TSSs at single nucleotide resolution by Capping-seq. TSSs analysis of Brucella transcripts showed a preference of purine on the TSS positions. Our results revealed that -35 and -10 elements of promoter contained consensus sequences of TTGNNN and TATNNN, respectively. The 5' ends analysis showed that 57% genes are associated with more than one TSS and 47% genes contain long leader regions, suggested potential complex regulation at the 5' ends of genes in B. melitensis. Moreover, we identified 52 leaderless genes that are mainly involved in the metabolic processes. Overall, Capping-seq technology provides a unique solution for TSS determination in prokaryotes. Our findings develop a systematic insight into the primary transcriptome characterization of B. melitensis. Biodiversity conservation, plant growth and spatial distribution of plant species are the central issues in contemporary community ecology. Ephemeral stream may influence soil properties, which in turn may determine biodiversity and function of an ecosystem in alluvial fan of arid desert region. Ephemeral stream is one of the most common natural disturbances, yet the effects of the ephemeral stream on plant communities in terms of species diversity and plant species distribution remain poorly studied. In this study, the information of species distribution, ephemeral stream beds ('washes'), and the characteristics of plant growth, i.e. height, crown area, were interpreted at different heights using the images of low altitude unmanned aerial vehicle (UAV). After that, soil properties such as soil texture (sand, silt and clay), soil water content, pH, soil organic matter, soil electric conductivity, soil bulk density and the percentage of gravel content, and their relationships with UAV data were assessed in order to explore the influences of ephemeral stream on species diversity, plant growth characteristics and species distribution in an alluvial fan of arid desert region. The results showed that deep-rooted plants were only distributed in washes whereas shallow-rooted plants were distributed in both washes and the outside of washes ('non-washes'). Species richness was significantly higher in washes than that in non-washes whereas the opposite pattern was true for abundance. Soil properties, plant height and crown area were higher in washes than that in non-washes. Plant height, crown area and the total number of individual plants increased with increasing wash width and per unit length of stream flow. This study highlights that the coupling factors of ephemeral stream, such as soil erosion, particle transport and sedimentation, can dramatically cause changes in soil properties and total number of individual plants, and hence, can influence species diversity, plant growth characteristics and spatial distribution of plant species in an alluvial fan of arid desert regions., Competing Interests: The authors have declared that no competing interests exist.

Aberrant microRNA expression is involved in the regulation of various cellular processes, such as proliferation and metastasis in multiple diseases including cancers. MicroRNA-30e-5p (miR-30e) was previously reported as an oncogenic or tumour suppressing miRNA in some malignancies, but its function in lung adenocarcinoma (LAC) remains largely undefined. In this study, we found that the expression of miR-30e was increased in LAC tissues and cell lines, associated with tumour size and represented an independent prognostic factor for overall survival and recurrence of LAC patients. Further functional experiments showed that knockdown of miR-30e suppressed cell growth while its overexpression promoted growth of LAC cells and xenografts in vitro and in vivo. Mechanistically, PTPN13 was identified as the direct target of miR-30e in LAC, in which PTPN13 expression was down-regulated in LAC tissues and showed the inverse correlation with miR-30e expression. Overexpression of PTPN13 inhibited cell growth and rescued the proliferation-promoting effect of miR-30e through inhibition of the EGFR signalling. Altogether, our findings suggest that miR-30e could function as an oncogene in LAC via targeting PTPN13 and act as a potential therapeutic target for treating LAC., (© 2017 The Authors. Effects of ciprofloxacin hydrochloride (CIP) initial concentration, Pb (NO3) 2, Cd (NO3) 2, PbCl2 and CdCl2 on the photodegradation of CIP using UV irradiation were investigated. The experiments results showed that UV irradiation could lead to effective removal of CIP, but there was no CIP degradation in dark. The photodegradation rate of CIP reduced with increasing initial concentration under UV irradiation. Pb(NO3)2 and Cd(NO3)2 (except for the experiment group of 0.006 mmol x L(-1)) could enhance CIP photodegradation, and the half-life of CIP gradually increased with increasing molar ratio between heavy metal and CIP ( with the decreasing concentration of nitrate). With increasing molar ratio (with the decreasing concentration of chlorate), PbCl2 and CdCl2 first promoted and then inhibited the CIP photodegradation.

In this study, a central venous catheter (CVC)-associated infection model was established in rats to investigate and evaluate the effect of biofilms on the virulence of the pathogens. Twenty-four adult SD rats were randomly divided into biofilm positive (BF+) and biofilm negative (BF-) groups to be challenged with strains of S.epidermidis. Serum levels of inflammatory cytokines were measured and the infection rate and counts of bacteria cells were studied. Compared to rats of BF- group, the serum level of TNF and IL-6 significantly increased in rats of BF+ group (P < 0.01) and the level of IL-10 and IFN-γ significantly decreased (P < 0.01), striking the balance of pro-inflammatory/anti-inflammatory cytokines. The infection rate and bacterial counts in tissues and blood of rats of BF + group were significantly higher than those of rats of BF- group (P < 0.05).Inflammatory cell infiltration in vital organs (heart, lung, liver and kidneys) was more significant in rats of BF+ group than that of rats of BF- group. CVC-associated infection model can be successfully reproduced in rats by injecting 5 × 10(6) CFU of S.epidermidis. Biofilm formation can significantly enhance the virulence of the bacteria, leading to uncontrolled infection. The serum level of inflammatory cytokines, infection rate and the extent of inflammatory cell infiltration are important markers for evaluating the virulence of biofilm.

Tetracycline (TC) has been widely used in veterinary medicines and disease treatments, and has been discharged into nature system due to manure application. To know the harmfulness of TC residues, and to investigate the natural attenuation of TC under different environmental conditions, simulated attenuation experiments in the presence of light, sediment, Cd (NO3) 2 and Pb(NO3)2 were performed. Results showed that the natural attenuation of TC was not obvious under the sunlight, and the natural attenuation rate of TC in water with unsterilized sediment was greater than that with sterilized sediment. The natural attenuation of TC in water with Pb(NO3)2 was slow at the first stage of the experiment, and progressively speeded up as time went on. However, the natural attenuation of TC in water with Cd(NO3)2 was more rapid at the beginning, and significantly faster than that of Pb(NO3)2. The natural attenuation rates of 0.08 mmol x L(-1) TC under different conditions follow the sequence: unsterilized sediment (87.2%) > sterilized sediment (70.37%) > Cd(NO3)2 (64.2%) > Pb(NO3)2 (32.3%) > blank (6.6%), suggesting that all these factors can promote the natural attenuation of TC. Though the natural attenuation trend under different conditions in the dark is similar to that under the sunlight, the natural attenuation rate is less than that under the sunlight, which implicates that sunlight may promote the natural attenuation of TC.

A large-mode-area polymer photonic crystal fiber made of polymethyl methacrylate with the cladding having only one layer of air holes near the edge of the fiber is designed and proposed to be used in surface plasmon resonance sensors. In such sensor, a nanoscale metal film and analyte can be deposited on the outer side of the fiber instead of coating or filling in the holes of the conventional PCF, which make the real time detection with high sensitivity easily to realize. Moreover, it is relatively stable to changes of the amount and the diameter of air holes, which is very beneficial for sensor fabrication and sensing applications. Numerical simulation results show that under the conditions of the similar spectral and intensity sensitivity of 8.3 × 10(-5)-9.4 × 10(-5) RIU, the confinement loss can be increased dramatically.
