Background: In the last decade, a growing body of research has focused on the many aspects and challenges of combining parenthood with elite sport. Although the number of father-athletes is significantly higher than the number of mother-athletes, few studies to date have focused on male athletes' experiences in a parenting context. Aim: The aims of the present study were to explore how father-athlete challenges manifest among elite Nordic skiers in Norway, and to better understand how male athletes balance their priorities as they initiate, maintain, and/or discontinue their athletic career as a father-athlete. Methods: Qualitative data were collected through semi-structured interviews with 10 world-class male Nordic skiers in Norway (3 athletes without a child, 4 current father-athletes and 3 former father-athletes) and the content was analyzed using thematic analysis. Results: Four main stages were identified in the father-athlete transition: (a) Expecting incompatibility (b) Taking the step, (c), The first blow, and (d) Finding the optimal balance. Through these stages the informants expected/had experienced challenges such as performance decline, disturbed sleeping patterns, fear of sickness and role conflicts. To manage these challenges, the father-athletes had developed various strategies to balance their dual roles (e.g., adapting training and competition seasons). Among the benefits, the father-athletes mentioned that they had become more structured, time efficient and ruthless with their priorities, enhanced motivation to train and a better work-life balance. Challenges facing mother-athletes (MAs) have aroused research and media attention in recent years, with an increasing number of sportswomen attempting to successfully combine pregnancy and motherhood with an elite athletic career. The aims of this study were to explore how MA-specific challenges manifest in elite cross-country skiing in Scandinavia and to better understand how female athletes balance their priorities as they initiate, maintain, and/or discontinue their role as a MA. Qualitative data were collected through semistructured interviews with 13 female cross-country skiers from Norway and Sweden. Thematic analyses revealed four MA-specific challenges facing the athletes: (a) Biological clock versus peak performance, (b) Maintaining fitness versus training safely, (c) Receiving support versus facing deselection, and (d) Balancing competing MA demands. Many of the athletes felt pressured into prioritizing either motherhood or athletic excellence, particularly in their early to mid-30s when the window of opportunity for building a family was considered limited. Further, maintaining fitness and training safely during pregnancy were perceived as a challenge, as was balancing the MA role after childbirth. In many cases, athletes felt uncertain about whether they would receive support from their team or federation. Moreover, there were expectations of incompatibility surrounding the MA role. More research and educational efforts to promote MA-specific knowledge, as well as developing structured processes and providing policies to support female athletes, are identified as vital future steps. The chromatin-remodeler SPOC1 (PHF13) is a transcriptional co-regulator and has been identified as a restriction factor against various viruses, including human cytomegalovirus (HCMV). For HCMV, SPOC1 was shown to block the onset of immediate-early (IE) gene expression under low multiplicities of infection (MOI). Here, we demonstrate that SPOC1-mediated restriction of IE expression is neutralized by increasing viral titers. Interestingly, our study reveals that SPOC1 exerts an additional antiviral function beyond the IE phase of HCMV replication. Expression of SPOC1 under conditions of high MOI resulted in severely impaired viral DNA replication and viral particle release, which may be attributed to inefficient viral transcription. With the use of click chemistry, the localization of viral DNA was investigated at late time points after infection. Intriguingly, we detected a co-localization of SPOC1, RNA polymerase II S5P and polycomb repressor complex 2 (PRC2) components in close proximity to viral DNA in areas that are hypothesized to harbor viral transcription sites. We further identified the N-terminal domain of SPOC1 to be responsible for interaction with EZH2, a subunit of the PRC2 complex. With this study, we report a novel and potent antiviral function of SPOC1 against HCMV that is efficient even with unrestricted IE gene expression. [ABSTRACT FROM AUTHOR]

Abstract As aberrant alternative splicing by either dysregulation or mutations of splicing factors contributes to cancer initiation and progression, splicing factors are emerging as potential therapeutic targets for cancer therapy. Therefore, pharmacological modulators targeting splicing factors have been under development. Epithelial splicing regulatory protein 1 (ESRP1) is an epithelial cell-specific splicing factor, whose downregulation is associated with epithelial–mesenchymal transition (EMT) by regulating alternative splicing of multiple genes, such as CD44, CTNND1, ENAH, and FGFR2. Consistent with the downregulation of ESRP1 during EMT, it has been initially revealed that high ESRP1 expression is associated with favorable prognosis and ESRP1 plays a tumor-suppressive role in cancer progression. However, ESRP1 has been found to promote cancer progression in some cancers, such as breast and ovarian cancers, indicating that it plays a dual role in cancer progression depending on the type of cancer. Furthermore, recent studies have reported that ESRP1 affects tumor growth by regulating the metabolism of tumor cells or immune cell infiltration in the tumor microenvironment, suggesting the novel roles of ESRP1 in addition to EMT. ESRP1 expression was also associated with response to anticancer drugs. This review describes current understanding of the roles and mechanisms of ESRP1 in cancer progression, and further discusses the emerging novel roles of ESRP1 in cancer and recent attempts to target splicing factors for cancer therapy.

The dual operation of a chemical species in synthetic chemistry is an intriguing and relatively unexplored phenomenon. The application of such a species is expected to reduce the use of multiple reaction partners and catalysts/activators. Herein, we report a simple and easy‐to‐use protocol for the twin application of TiO(acac)2, as a reagent and an activator to synthesize β‐enamino ketones with amines in acetonitrile. The same early transition metal precursor when employed in N,N‐dimethylformamide with the amines, resulted in the formation of the substituted amides. Both reactions were explored with various substrates to check the viability of the present protocol. Moreover, experimental studies were conducted to understand the mechanism of both reactions. Nurse retention strategies are top of mind for nurse leaders as they face an unprecedented staffing crisis. A strategic approach that includes innovative models to enhance nurse satisfaction and nurse retention may include role enrichment strategies such as blended roles, alternative work arrangements, and shared staffing. Effective implementation requires authentic, transformational leadership, as well as structures and processes for replication, sustainability, and improved outcomes. This case study illustrates the potential of this strategy to positively influence key factors contributing to nurse retention, especially for millennial nurses. Nurse leaders in one critical care unit shared their experience with cross-training for blended roles, skill expansion for professional development for unit staff as well as float team members, leadership development opportunities, and shared staffing. Creating a staffing strategy that includes the leadership and infrastructure to support blended or dual roles is one promising element in a nurse leader tool kit for millennial nurse retention. The continuous increase in cancer-associated deaths despite the substantial improvement in diagnosis and treatment has sparked discussions on the need for novel biomarkers and therapeutic strategies for cancer. Although increasing evidence has demonstrated the pivotal role of relaxin-2 in multiple cancers, their role is a double-edged sword with both protumor and antitumor having been reported in various malignant tumors. Considering this dual role, it appears the biological mechanism underpinning the action of relaxin-2 in cancer is not clear and further studies to elucidate their potential as a preventive factor for cancers are of prime importance. Herein, a summarized up-to-date report on the role of relaxin-2 in human cancer including detailed clinical and experimental evidence supporting their tumor-promoting and inhibitory functions in cancer development and progression has been elucidated. Also, signaling pathways and other factors orchestrating the activities of relaxin-2 in the tumor microenvironment has been discussed. In organic synthesis, incorporating two functional groups into the carbon‐carbon double bond of α,β‐unsaturated amides is challenging due to the electron‐deficient nature of the olefin moiety. Although a few examples of dihydroxylation of α,β‐unsaturated amides have been demonstrated, producing cis‐1,2‐diols using either highly toxic OsO4 or other specialized metal reagents in organic solvents, they are limited to several specific amides. We describe herein a general and one‐pot direct synthesis of trans‐1,2‐diols from electron‐deficient α,β‐unsaturated amides through dihydroxylation using oxone as a dual‐role reagent in water. This reaction does not require any metal catalyst and produces non‐hazardous and nontoxic K2SO4 as the sole byproduct. Moreover, epoxidation products could also be selectively formed by adjusting the reaction conditions. By the strategy, the intermediates of Mcl‐1 inhibitor and antiallergic bioactive molecule can be synthesized in one pot. The gram‐scale synthesis of trans‐1,2‐diol which is isolated and purified by recrystallization further shows the potential applications of this new reaction in organic synthesis. We developed a novel and efficient method for the synthesis of various 3‐alkylated quinoxalin‐2(1H)‐ones (32 examples, 70%–96% yields) through electron donor‐acceptor complex enabled alkylation of quinoxalin‐2(1H)‐ones with N‐ hydroxyphthalimide esters as the alkyl source and Na2S as the catalytic electron donor under external photocatalyst‐, oxidant‐ and additive‐free conditions. Background: Nrf2 regulates oxidative stress, which is essential for cellular function. Fundamental initiation of Nrf2 in many malignancies increases prosurvival genes & endorses tumour cell propagation via metabolic reprogramming, suppression of tumour programmed cell death, & increased cancer stem cell self-renewal potential. More specifically, Nrf2 has been associated with cancer cell chemoresistance, radioresistance & inflammation-induced carcinogenesis. Methods and Results: Many Nrf2 inhibitors have been revealed for tumour treatment and targeting Nrf2 could be an effective cancer therapeutic method. Before spreading, cancer cells adapt to their surroundings. Cancer cells usually have mutations in tumor suppressor genes. In a variety of malignancies, somatic mutations & other anomalies in the Nrf2 genes, as well as renowned cancer suppressor genes including TP53, CDKN2A, PTEN & PIK3CA, have been found. In tumour cells, somatic mutations in the Nrf2 genes, as well as additional mechanisms that affect Nrf2 binding, and produce aberrant Nrf2 activation. Uncontrolled Nrf2 causes tumour cells to become resistant to antineoplastic drugs & reactive oxygen species (ROS), as well as guiding them toward metabolic reprogramming. Conclusions: As a result, Nrf2 has been studied as potential malignancy treatment target. We covered the pathways, mechanisms, and dual characteristics of Nrf2 in malignancy in this article. We study a dual-role game setting of locating facilities in a metric space where each agent can open a facility at her location or be a customer to receive the service, and an opening cost function is given to represent the cost of opening a facility at some specific location. We first show the existence of pure Nash equilibria (PNE) in such games by a polynomial-time algorithm, then use the price of anarchy (PoA) to measure the quality of PNE under social objectives of minimizing the maximum/social cost. For dual-role facility location games with general opening cost functions, we show the PoA under maximum/social cost can tend to be infinite. However, for games with L-Lipschitz conditioned opening cost functions where L ≥ 0 is a given parameter, the PoA under maximum cost is exactly L + 1 and the PoA under social cost is bounded by the interval (n + L) / 3 , n + max { L - 1 , 0 } . Sushi domain-containing protein 2 (SUSD2, also known as the complement control protein domain) is a representative and vital protein in the SUSD protein family involved in many physiological and pathological processes beyond complement regulation. Cancer is one of the leading causes of death worldwide. The complex role of SUSD2 in tumorigenesis and cancer progression has raised increasing concerns. Studies suggest that SUSD2 has different regulatory tendencies among different tumors and exerts its biological effects in a cancer type-specific manner; for instance, it has oncogenic effects on breast cancer, gastric cancer, and glioma and has tumor-suppression effects on lung cancer, bladder cancer, and colon cancer. Moreover, SUSD2 can be regulated by noncoding RNAs, its promoter methylation and other molecules, such as Galectin-1 (Gal-1), tropomyosin alpha-4 chain (TPM4), and p63. The therapeutic implications of targeting SUSD2 have already been preliminarily revealed in some malignancies, including melanoma, colon cancer, and breast cancer. This article reviews the role and regulatory mechanisms of SUSD2 in cancer development, as well as its structure and distribution. Chemoselective sulfonylation and isonitrilation reactions for the divergent synthesis of valuable diarylmethyl sulfones and isonitrile diarylmethanes starting from easy-to-synthesize para-quinone methides (p-QMs) and commercially abundant p-toluenesulfonylmethyl isocyanide (TosMIC) by using respectively zinc iodide and 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) as catalysts were developed. The distinguishing feature of this method is that TosMIC plays a dual role from the same substrates in the reaction: as a sulfonyl source or as an isonitrile source. The synthetic utility of this protocol was also demonstrated in the synthesis of difluoroalkylated diarylmethane 5 and diarylmethane ketone derivatives 6 and 7, which are important core structures in natural products and medicines.

Nanoparticles (NPs) bear unique properties that leads to its immense application in diverse domains. But excessive use of metal oxide nanoparticles (MONPs) in various industrial sectors may lead to detrimental impacts on our environment in the near future. Among NPs, applications of nanoiron in various industrial sectors are gradually increasing and its interaction with the soil-plant system is an area still to be explored. In this review, we have focused upon positive as well as the negative impact of nanoiron in plants. As per available literature, it is evident that higher concentration of nanoiron causes oxidative stress while lower concentration leads to plant growth promotion. In addition to this, review also highlights the uptake, translocation and accumulation of nanoiron in plant system which subsequently leads to alterations in the morpho-physiological, biochemical and molecular traits of plants. Moreover, various factors contributing towards the translocation of nanoiron in shoot and root system of plants has been duly discussed.

Introduction: Taurine is a naturally occurring sulfonic acid involved in various physiological and pathological processes, such as the regulation of calcium signaling, immune function, inflammatory response, and cellular aging. It has the potential to predict tumor malignant transformation and formation. Our previous work discovered the elevated taurine in lung cancer patients. However, the precise impact and mechanism of elevated serum taurine levels on lung cancer progression and the suitability of taurine or taurine-containing drinks for lung cancer patients remain unclear., Objectives: Our study aimed to systematically investigate the role of taurine in lung cancer, with the ultimate goal of contributing novel strategies for lung cancer treatment., Methods: Lung cancer C57 and nude mice models, RNA sequencing, and stable transfection were applied to explored the effects and mechanisms of taurine on lung cancer. Tissues of 129 non-small cell lung cancer (NSCLC) patients derived from 2014 to 2017 for immunohistochemistry were collected in Taihe Hospital., Results: Low doses of taurine, as well as taurine-infused beverages at equivalent doses, significantly enhanced lung tumor growth. Equally intriguing is that the promoting effect of taurine on lung cancer progression wanes as the dosage increases. The Nuclear factor erythroid 2-like 1 (Nfe2l1 or Nrf1)-reactive oxygen species (ROS)-PD-1 axis may be a potential mechanism for dual role of taurine in lung cancer progression. However, taurine's impacts on lung cancer progression and the anti-tumor function of Nfe2l1 were mainly determined by the immune competence. Taurine inhitited lung tumor growth probably by inhibiting NF-κB-mediated inflammatory responses in nude mice rather than by affecting Nfe2l1 function. As patients age increased, Nfe2l1 gene and protein gradually returned to the levels observed in healthy individuals, but lost its anti-lung cancer effects., Conclusions: Taurine emerges as a potential biomarker for lung cancer progression, predicting poor prognosis and unsuitability for specific patients. Lung cancer patients, especially young patients, should be conscious of potential effects of taurine-containing drinks. Abstract Background Mental health professionals (MHP) working in court-mandated treatment settings face ethical dilemmas due to their dual role in assuring their patient’s well-being while guaranteeing the security of the population. Clear practical guidelines to support these MHPs’ decision-making are lacking, amongst others, due to the ethical conflicts within this field. This qualitative interview study contributes to the much-needed empirical research on how MHPs resolve these ethical conflicts in daily clinical practice. Methods 31 MHPs working in court-mandated treatment settings were interviewed. The interviews were semi-structured and our in-depth analysis followed the thematic analysis approach. Results We first outline how mental health professionals perceive their dual loyalty conflict and how they describe their affiliations with the medical and the justice system. Our findings indicate that this positioning was influenced by situational factors, drawing the MHPs at times closer to the caring or controlling poles. Second, our results illustrate how participating MHPs solve their dual loyalty conflict. Participants considered central to motivate the patient, to see the benefits of treatment and its goals. Further, transparent communication with patients and representatives of the justice system was highlighted as key to develop a trustful relationship with the patient and to manage the influences from the different players involved. Conclusions Even though individual positioning and opinions towards dealing with the influences of the justice system varied, the results of our research show that, in spite of varying positions, the underlying practice is not very different across participating MHPs. Several techniques that allow developing a high-quality therapeutic alliance with the patient are key elements of general psychotherapy. Transparency appears as the crucial factor when communicating with the patient and with representatives of the justice system. More specifically, patients need to be informed since the beginning of therapy about the limits of medical confidentiality. It is also recommended to develop guidelines that define the level of detailed information that should be disclosed when communicating with the authorities of the justice system.

The COVID-19 pandemic lockdown disrupted the university sports season and had negative consequences on the academic and personal life of university student-athletes, resulting in several psychological challenges. The goal of this study is to document the symptoms of mental illness among university student-athletes during the second wave of the COVID-19 pandemic lockdown in Canada. It aims to (a) assess the prevalence of mental illness symptoms (anxiety, depression, disordered eating, and dangerous drinking) among university student-athletes and (b) identify which sociodemographic and sports characteristics, pandemic impacts, and levels of perceived stress most influence these symptoms. A total of 424 university student-athletes completed an online survey, which included questions on mental illness and the impact of the pandemic lockdown. The results revealed a notable prevalence of the symptoms of mental illness; depressive symptoms are reported by 37.9% of the participants, anxiety symptoms by 24.9%, dangerous drinking symptoms by 10.1%, and disordered eating by 8.6%. In addition, being female [OR = 0.56, 95% CI (0.33, 0.95)] or a member of a visible minority group [OR = 2.63, 95% CI (1.02, 6.78)] are significantly associated with the presence of depressive symptoms. Low academic motivation has a significant negative influence on the presence of depressive [OR = 3.37, 95% CI (1.82, 6.25)] and anxiety symptoms [OR = 2.75, 95% CI (1.35, 5.62)]. However, the presence of perceived stress was strongly associated with depressive [OR = 7.07, 95% CI (3.26, 15.35)], anxiety [OR = 6.51, 95% CI (3.30, 12.84)], and dangerous drinking symptoms [OR = 5.74, 95% CI (2.51, 13.14)]. This study advocates for specific mental illness prevention and treatment resources tailored to the unique needs of university student-athletes. Accordingly, partnerships and practical interventions to support university student-athletes' mental health are presented.

Purpose: This study sought to evaluate the attributes and characteristics of physician mentors, dual-roled physician assistant (PA) leaders, and how this relationship affects PA leaders and the PA leader-physician relationship. Dual-role PA leaders may possess additional formal job titles and duties as well as having direct reports. Method: We surveyed PA leaders at a single large academic medical center and its associated health system. The survey instrument evaluated perceptions, attitudes, and characteristics of the PA leader and influential physician and their relationship. Results: Of the 56 PA leaders surveyed, 34 responded, for a 60.7% response rate. Effective communication was the most prevalent relational characteristic ranked as most important for both PA leaders (30%) and physician mentors (36%). Additional self-identified positive attributes of the team included collaborative mindset, reliability/trustworthiness, work ethic, and delivery of patient care. Influential physician mentors may have a significant effect on PA leader job satisfaction and career advancement. Conclusions: This is a novel study of PA leaders examining perceptions, beliefs, and characteristics of the PA leader-physician relationship. PAs are integral healthcare team members and medical providers. As PAs work closely in physician-led teams, further understanding the PA leader-physician relationship could influence PA and physician professional development, career trajectory, and healthcare team outcomes. Many drugs can bind directly to proteins or be bioactivated by metabolizing enzymes to form reactive metabolites (RMs) that rapidly bind to proteins to form drug−protein conjugates or metabolite−protein conjugates (DMPCs). The close relationship between DMPCs and idiosyncratic adverse drug reactions (IADRs) has been recognized; drug discovery teams tend to avoid covalent interactions in drug discovery projects. Covalent interactions in DMPCs can provide high potency and long action duration and conquer the intractable targets, inspiring drug design, and development. This forms the dual role feature of DMPCs. Understanding the functional implications of DMPCs in IADR control and therapeutic applications requires precise identification of these conjugates from complex biological samples. While classical biochemical methods have contributed significantly to DMPC detection in the past decades, the low abundance and low coverage of DMPCs have become a bottleneck in this field. An emerging transformation toward shotgun proteomics is on the rise. The evolving shotgun proteomics techniques offer improved reproducibility, throughput, specificity, operability, and standardization. Here, we review recent progress in the systematic discovery of DMPCs using shotgun proteomics. Furthermore, the applications of shotgun proteomics supporting drug development, toxicity mechanism investigation, and drug repurposing processes are also reviewed and prospected. Endogenous indole and its derivatives (indoles), considered as promising N-substituted heterocyclic compounds, are tryptophan metabolites derived from intestinal microbiota and exhibit a range of biological activities. Recent studies indicate that indoles contribute to maintaining the biological barrier of the human intestine, which exert the anti-inflammatory activities mainly through activating AhR and PXR receptors to affect the immune system's function, significantly improving intestinal health (inflammatory bowel disease, hemorrhagic colitis, colorectal cancer) and further promote human health (diabetes mellitus, central system inflammation, and vascular regulation). However, the revealed toxic influences cannot be ignored. Indoxyl sulfate, an indole derivative, performs nephrotoxicity and cardiovascular toxicity. We addressed the interaction between indoles and intestinal microbiota and the indoles' effects on human health as double-edged swords. Endogenous indole and its derivatives (indoles), considered as promising N-substituted heterocyclic compounds, are tryptophan metabolites derived from intestinal microbiota and exhibit a range of biological activities. Recent studies indicate that indoles contribute to maintaining the biological barrier of the human intestine, which exert the anti-inflammatory activities mainly through activating AhR and PXR receptors to affect the immune system’s function, significantly improving intestinal health (inflammatory bowel disease, hemorrhagic colitis, colorectal cancer) and further promote human health (diabetes mellitus, central system inflammation, and vascular regulation). However, the revealed toxic influences cannot be ignored. Indoxyl sulfate, an indole derivative, performs nephrotoxicity and cardiovascular toxicity. We addressed the interaction between indoles and intestinal microbiota and the indoles’ effects on human health as double-edged swords. This review provides scientific bases for the correlation of indoles with diseases moreover highlights several directions for subsequent indoles-related studies.

• A novel ZnO/ZnFe 2 O 4 /CdS photoanode was designed for solar light PEC water splitting. • Interfacial charge transfer channels in photoanode facilitates electron transport. • 2D ZnFe 2 O 4 nanosheets as an interfacial bridge to build stepwise type II structure. • 2D ZnFe 2 O 4 nanosheets act as a supporting platform to load more CdS nanoparticles. • CdS NPs act as light energy absorber to remarkably improve light energy utilization. In this study, a novel ternary ZnO-based nanorod arrays (NRAs) photoanode is designed and constructed for the first time for unassisted solar-light-driven water splitting to generate H 2 using photoelectrochemical (PEC) cell. The ternary heterojunction photoanode is constructed by modifying ZnO NRAs with ZnFe 2 O 4 nanosheets (NSs) and CdS nanoparticles (NPs), successively. The band alignment study demonstrates that ZnFe 2 O 4 NSs as an interfacial modification layer not only enlarge the contact area with ZnO and CdS respectively to build a stair-step band alignment of conduction band levels for the efficient transport of electrons but also make the valence band of ZnFe 2 O 4 to be as the hole accumulation site in the ternary heterojunction. The electrochemical and photoluminescence measurements demonstrate the efficient separation of the carriers. Consequently, the ternary ZnO-based NRAs photoanode achieves a significant H 2 generation rate of 92.9 μmol/h, 6.23 and 2.97 times of ZnO NRAs and ZnO/ZnFe 2 O 4 NRAs/NSs photoanodes, respectively. Nitric oxide (NO) is a non-traditional regulator of the growth of plants and phytoplankton. Here, the growth of five marine phytoplankton species, Platymonas helgolandica var. tsingtaoensis, Platymonas subcordiformis, Skeletonema costatum, Gymnodinium sp., and Prorocentrum donghaiense, was monitored, and carbonate system parameters in the culture media were determined after adding different concentrations of NO and sodium nitroprusside (SNP, NO donor) solutions. The two dietary algae (P. helgolandica var. tsingtaoensis and P. subcordiformis) and three red tide algae (S. costatum, Gymnodinium sp., and P. donghaiense) showed different responses to the same NO concentration. The red tide algae were more sensitive to exogenous NO than the dietary algae. NO with a concentration of 1.4 × 10−6 mol L−1 had the optimal stimulatory effect on the growth of the microalgae and increased the cell density by 9.8–38.3%. SNP solution with a concentration of 100 μmol L−1 inhibited the growth of the two dietary algae, and the cell density decreased by 38.8–84.3%. The addition of 10 μmol L−1 SNP solution to the three red tide algae decreased the cell density by 95.3–99.9%. Low concentrations of SNP (0.1 μmol L−1 for the two dietary algae and 0.01 μmol L−1 for the red tide algae) slightly promoted the growth of microalgae. High concentrations of NO (100 μmol L−1 SNP) inhibited CO2 assimilation, whereas low concentrations of NO (0.01–1.0 μmol L−1 SNP) promoted CO2 assimilation, indicating that NO participates in the regulation of photosynthesis. The paper presents findings on primary teachers' and other school actors' constructions of the teacher and parental role. Specifically, it focuses on strategies for maintaining borders between the personal (parent) and professional (teacher) roles in school environments in Slovakia. We approached the concepts of role and identity from the perspective of social constructivism and symbolic interactionism. Thirty-one interviews and focus groups with school actors were analysed using critical discourse analysis. In the next step, discourses on managing the dual role were analysed using thematic analysis. Results show participants described regulating or restricting verbal, emotional and behavioural expression, and engaged in favouritism avoidance as strategies for separating out the roles. All point to the need to eliminate favouritism as a key meaning in the construction of the teacher role through impartialness and parental role as favourably inclined to create inter-role conflict. We stress the importance of exploring teacher and parental roles in various circumstances in order to obtain a complex picture of negotiating these roles in everyday life. [ABSTRACT FROM AUTHOR]

Adults under community corrections supervision and who have a mental illness (MI) are expected to comply with conditions of release which often include involvement with supportive social services. The rates of technical violation, arrest, and incarceration that result from failure to comply with these mandates are exceedingly high. Shared decision making among officer-supervisors and client-supervisees is a promising approach to promote engagement in community corrections services among supervisees who have MI. This paper reviews recent research on shared decision making and identifies three barriers to its implementation in this context: (1) a lack of role clarity, (2) a predilection for risk avoidance, and (3) stigma toward supervisees. Empirically supported recommendations are suggested to aid in overcoming these obstacles, facilitate shared decision making, and promote recovery among this population: (1) unification of supervisor rehabilitative and public safety roles, (2) maximizing opportunities for self-determination through low-stakes events and/or enhancement of supervisee strengths and capabilities, and (3) supervisor training in principles of mental health recovery. Social information is usually jointly utilized with rating information to help the traditional recommendation system providing more personalized services, while how to make full use of social information to build better recommendation models still faces lots of challenges. In this paper, we propose a novel social recommendation model taking advantage of both deep and shallow model, with deep auto-encoders acting as the nonlinear feature extractor and MF-based method being used to depict the user's preferences. Then, motivated by the idea of word2vec, we provide an appealing method that embeds users into latent space and meanwhile preserves the structural information of social networks during the embedding. Also, the embedded latent features of each user are corresponding to the dual roles the user plays in the recommendation. Furthermore, we design a loss function for the holistic training of the model, and our loss function is made up mainly of three parts which embody the effects of different factors on the rating predictions. Specifically, the loss function (i) captures the personal preference from user-item adoption matrix based on matrix factorization, (ii) discriminates the two different social functions of users and further evaluates the effects of interpersonal influence through user embedding and social influence matrix, (iii) and avoids overfitting by imposing a quadratic regularization penalty. As a result, our model can predict the missing ratings with the MF-based method by consuming the latent features of users and items extracted by the deep model. The experiments show that our method outperforms existing methods and performs well on cold start users.

Background: Mental health professionals (MHP) working in court-mandated treatment settings face ethical dilemmas due to their dual role in assuring their patient's well-being while guaranteeing the security of the population. Clear practical guidelines to support these MHPs' decision-making are lacking, amongst others, due to the ethical conflicts within this field. This qualitative interview study contributes to the much-needed empirical research on how MHPs resolve these ethical conflicts in daily clinical practice.Methods: 31 MHPs working in court-mandated treatment settings were interviewed. The interviews were semi-structured and our in-depth analysis followed the thematic analysis approach.Results: We first outline how mental health professionals perceive their dual loyalty conflict and how they describe their affiliations with the medical and the justice system. Our findings indicate that this positioning was influenced by situational factors, drawing the MHPs at times closer to the caring or controlling poles. Second, our results illustrate how participating MHPs solve their dual loyalty conflict. Participants considered central to motivate the patient, to see the benefits of treatment and its goals. Further, transparent communication with patients and representatives of the justice system was highlighted as key to develop a trustful relationship with the patient and to manage the influences from the different players involved.Conclusions: Even though individual positioning and opinions towards dealing with the influences of the justice system varied, the results of our research show that, in spite of varying positions, the underlying practice is not very different across participating MHPs. Several techniques that allow developing a high-quality therapeutic alliance with the patient are key elements of general psychotherapy. Transparency appears as the crucial factor when communicating with the patient and with representatives of the justice system. More specifically, patients need to be informed since the beginning of therapy about the limits of medical confidentiality. It is also recommended to develop guidelines that define the level of detailed information that should be disclosed when communicating with the authorities of the justice system. Autophagy, an essential biological process that affects immunity, is a powerful tool that host cells can use to defend against infections caused by pathogenic microorganisms. Autophagy can not only initiate innate immune responses but also degrade the cellular components that provide the conditions for removing the invaders. However, hyperactivated or inhibited autophagy leads to mitochondrial dysfunction, which is harmful to the host itself and is involved in many types of diseases. Mitochondria perform the functions of biological oxidation and energy exchange. In addition, mitochondrial functions are closely related to cell death, oxygen radical formation, and disease. Accumulation of mitochondrial metabolites affects survival of intracellular pathogens. In this mini-review, we focus on the crosstalk between autophagy and mitochondrial homeostasis during infection.

Hedgehog (Hh) signaling patterns embryonic tissues and contributes to homeostasis in adults. In Drosophila, Hh transport and signaling are thought to occur along a specialized class of actin-rich filopodia, termed cytonemes. Here, we report that Interference hedgehog (Ihog) not only forms a Hh receptor complex with Patched to mediate intracellular signaling, but Ihog also engages in trans-homophilic binding leading to cytoneme stabilization in a manner independent of its role as the Hh receptor. Both functions of Ihog (trans-homophilic binding for cytoneme stabilization and Hh binding for ligand sensing) involve a heparin-binding site on the first fibronectin repeat of the extracellular domain. Thus, the Ihog-Ihog interaction and the Hh-Ihog interaction cannot occur simultaneously for a single Ihog molecule. By combining experimental data and mathematical modeling, we determined that Hh-Ihog heterophilic interaction dominates and Hh can disrupt and displace Ihog molecules involved in trans-homophilic binding. Consequently, we proposed that the weaker Ihog-Ihog trans interaction promotes and stabilizes direct membrane contacts along cytonemes and that, as the cytoneme encounters secreted Hh ligands, the ligands trigger release of Ihog from trans Ihog-Ihog complex enabling transport or internalization of the Hh ligand-Ihog-Patched -receptor complex. Thus, the seemingly incompatible functions of Ihog in homophilic adhesion and ligand binding cooperate to assist Hh transport and reception along the cytonemes.

• Waste glass is used as a partial activator and as a mineral precursor in one-part AAMs. • Waste glass powder serves as an effective silica supplier in the solid activator. • Waste glass as a mineral precursor retards the early strength of one-part AAMs. • Waste glass-based one-part AAMs are 23% cheaper and 84% greener than OPC binders. The feasibility of the extensive recycling of waste glass in alkali-activated materials (AAMs) was evaluated. The waste glass was utilised in AAMs for two purposes: a partial activator and a mineral precursor. The waste glass was blended with commercial sodium hydroxide and then heated to produce the solid activator powder. The technical performance of waste glass-based activator was investigated to replace commercial sodium silicate, a common alkali-activator used in AAMs. The effect of waste glass using only as the activator (WGA) and using as both activator and precursor (WGAP) in fly ash/slag-based one-part AAMs was studied using strength and microstructure characterisations. A mass-cost and emission analysis of waste glass-based AAMs (WGA and WGAP) was conducted, comparing the results with ordinary Portland cement (OPC). Characterisation tests of waste glass-based activator showed the effective formation of sodium silicate minerals with the adequate dissolution of activator in water by releasing reactive alkali and silica. Both WGA and WGAP showed comparable strengths at 56 days with a denser microstructure under ambient curing. According to mass analysis, waste glass could be utilised up to 17% by mass of total binder. Based on the analysis of cost and CO 2 emissions, WGA and WGAP are around 23% and 15% cheaper and 84% and 82% greener than OPC. The dual role of waste glass in AAMs as an activator and as a precursor broadens the recycling of glass waste in the cement industry by favouring technical and environmental outcomes. Pekerjaan mempunyai beban kerja yang berat dikarenakan semua proses kerja dilakukan oleh pengrajin yang sama sehingga menimbulkan kelelahan disamping itu pekerjanya juga memiliki peran ganda. Tujuan penelitian ini adalah untuk mengetahui hubungan beban kerja dan peran ganda dengan perasaan lelah pada pengrajin batik mawar. Metode: Penelitian ini merupakan penelitian Kuantitatif dengan desain penelitian Cross Sectional. Populasi dalam penelitian ini adalah semua pengrajin di Batik Mawar. Pengambilan sampel pada penelitian ini menggunakan teknik total sampling dengan jumlah sampel penelitian sebanyak 40 responden. Analisis data yang digunakan Univariat dan Bivariat. Hasil: Hasil analisis data yang menggunakan uji Chi-Square untuk beban kerja didapatkan nilai p-Value = 0,001. BackgroundMental health professionals working in correctional contexts engage a double role to care and control. This dual loyalty conflict has repeatedly been criticized to impede the development of a high-quality alliance. As therapeutic alliance is a robust predictor of outcome measures of psychotherapy, it is essential to investigate the effects of this ethical dilemma.MethodsThis qualitative interview study investigates patients’ perceptions of their therapists’ dual role conflict in court-mandated treatment settings. We interviewed 41 older incarcerated persons using a semi-structured interview guide, the interviews were subsequently analyzed following thematic analysis.ResultsWe first present the patients’ perceptions of their treating psychotherapist’s dual loyalty conflict, which was linked to their overall treatment experience. In a second step, we outline the study participants’ reasons for this judgment, which were most commonly linked to feelings of trust or betrayal. More specifically, they named certain therapist characteristics and activities that enabled them to develop a trustful therapeutic alliance, which we grouped into four topics: (1) respecting the patient’s pace and perceived coercion; (2) patient health needs to be first priority; (3) clarity in roles and responsibilities; and (4) the art of communication – between transparency and unchecked information sharing.DiscussionDeveloping a high quality alliance in mandatory offender treatment is central due to its relationship with recovery and desistance. Our findings show that some therapists’ characteristics and activities attenuate the negative impact of their double role on the development and maintenance of the alliance. This review focuses on the thioredoxin domain containing 5 (TXNDC5), also known as endoplasmic reticulum protein 46 (ERp46), a member of the protein disulfide isomerase (PDI) family with a dual role in multiple diseases. TXNDC5 is highly expressed in endothelial cells, fibroblasts, pancreatic β-cells, liver cells, and hypoxic tissues, such as cancer endothelial cells and atherosclerotic plaques. TXNDC5 plays a crucial role in regulating cell proliferation, apoptosis, migration, and antioxidative stress. Its potential significance in cancer warrants further investigation, given the altered and highly adaptable metabolism of tumor cells. It has been reported that both high and low levels of TXNDC5 expression are associated with multiple diseases, such as arthritis, cancer, diabetes, brain diseases, and infections, as well as worse prognoses. TXNDC5 has been attributed to both oncogenic and tumor-suppressive features. It has been concluded that in cancer, TXNDC5 acts as a foe and responds to metabolic and cellular stress signals to promote the survival of tumor cells against apoptosis. Conversely, in normal cells, TXNDC5 acts as a friend to safeguard cells against oxidative and endoplasmic reticulum stress. Therefore, TXNDC5 could serve as a viable biomarker or even a potential pharmacological target.
