Abstract

Foodborne illnesses caused by Salmonellarepresent a significant public health problem worldwide. The aim of this study was to establish multiplex PCR (mPCR) for the rapid identification of Salmonellaserogroups B, C1, C2, D, and E as well as for the serovars enteritidisand typhimurium. Employing pan-genome analysis and PCR verification, B-rfbJ, C1-9679, C2-pimB, D-rfbJ, E-rfbC, and four genes (SE18636, SE16574, SE2599, and SE13329) were identified as specific target genes for Salmonellaserogroups B, C1, C2, D, E, and S. enteritidis, respectively. Thereafter, three novel mPCR assays (one of 3-mPCR and two of 2-mPCR) were successfully developed to identify these bacteria based on the target genes and another S. typhimurium-specific STM4495gene. The primers targeting C1-9679, C2-pimB, and E-rfbCgenes specific to the serogroups C1, C2, and E, respectively, constituted a 3-mPCR, while the other two 2-mPCRs, respectively, consisting primers specific to serogroup D and S. enteritidis(D-rfbJand SE16574), and serogroup B and S. typhimurium-specific primers (B-rfbJand STM4495), were also designed. The specificity of each mPCR was further evaluated by using non-target strains. The detection limits of mPCRs were approximately 103–104CFU mL−1in pure culture and 104–105CFU g−1in spiked chicken meat. In addition, mPCR assays could correctly detect target Salmonellain food samples. These results suggest that specific targets could be mined efficiently through a pan-genome analysis tool, and the novel mPCR assays developed in this study offer a promising technique for rapid and accurate detection of five serogroups of Salmonella(B, C1, C2, D, and E) and two serovars (S. enteritidisand S. typhimurium).

Abstract

Many studies have obtained high-quality plant point clouds but it remains difficult to extract the midrib from a leaf point cloud. In this study, a fast and accurate approach to the extraction of leaf midrib from point clouds was developed. The leaf was converted into its principal component analysis (PCA) coordinates to find the tip and base points of the leaf. A new search algorithm was proposed, which quickly searched for the approximate shortest curve between tip and base points on the leaf point cloud. The curve then was projected and fitted to eliminate the deviation between the curve and the leaf midrib. Two types of point cloud, generated using the structure-from-motion method and using a laser scanner, were obtained to verify our approach. As a result, the extracted curves were both in good agreement with the leaf midrib for the two kinds of point clouds. Compared with manual measurements of the leaf length, the root-mean-square error (RMSE) of the lengths of the two types of the extracted curve were 2.55 mm and 1.38 mm, respectively. 37. Assessing Potential Climate Change Impacts and Adaptive Measures on Rice Yields: The Case of Zhejiang Province in China.

Abstract

Increasing temperatures, greater carbon dioxide concentrations, and changes in related climatic variables will continue to affect the growth and yields of agricultural crops. Rice (Oryza sativa L.) is extremely vulnerable to these climatic changes. Therefore, investigating the degree to which climate changes could influence rice yields and what effective adaptive strategies could be taken to mitigate the potential adverse impacts is of vital importance. In this article, the impacts of climate change on rice yields in Zhejiang province, China, were simulated under the Representative Concentration Pathway (RCP) 4.5 and 8.5 scenarios. The impacts of climate change, with and without CO2 fertilization effects, were evaluated and the three most effective adaptive measures were examined. Compared with the yield for the baseline time of 1981–2010, the simulated average yields of all cultivars were inevitably projected to decrease under both RCPs when the CO2 fertilization effects were not considered during the three periods of the 2020s (2011–2040), 2050s (2041–2070), and 2080s (2071–2099), respectively. Declines in rice yields were able to be alleviated when the CO2 fertilization effects were accounted for, but the yields were still lower than those of the baseline. Therefore, the three adaptive measures of advancing planting dates, switching to high-temperature-tolerant cultivars, and breeding new cultivars were simulated. The results indicated that adaptive measures could effectively mitigate the adverse effects of climate change. Although the simulation had uncertainties and limitations, the results provide useful insights into the potential impacts of climate change in Zhejiang province while also proposing adaptive measures. 38. Corporate Governance and Organization Performance in Non-profit Organizations: Evidence from the UK Charities

Abstract

This paper explores the relationship between the internal corporate governance mechanism and the charity performance in the UK. Based on agency theory and resource dependency theory, this study quantifies five characteristics of the board, including the board size, board tenure, board activity, board committees, and board leadership. The charitable commitment, fundraising expense percentage, and charitable income reliance are efficiency indicators. This study employs 121 charities out of top 300 charities in the UK and selects related data from 2013-2017. Descriptive and OLS regression methods are used to analyse the statistics. The findings suggest the board size and tenure are positively related to the charitable income reliance ratio showing that the large board with long tenure could generate more resources and have positive impacts on the efficiency. Besides, the results present that the separation of leadership roles improves efficiency by mitigating agency problem in the charity. In regard to functional committees including the audit committee and the finance committee, significant inverse relationships are presented with the charity performance. Together with the same results of the board meetings, the ineffectiveness of the board of trustees may be presented. Alternatively, the board activity and functional committees may be used to respond to the poor performance of charities. The presence of the nomination committee is not significantly related to the charity performance. Due to inadequate observations and insufficient corporate governance variables, future studies are suggested employing a larger sample size and more representative variables for deeper investigation.
