The Fermi paradox is the conflict between an expectation of a high {\em ex ante} probability of intelligent life elsewhere in the universe and the apparently lifeless universe we in fact observe. The expectation that the universe should be teeming with intelligent life is linked to models like the Drake equation, which suggest that even if the probability of intelligent life developing at a given site is small, the sheer multitude of possible sites should nonetheless yield a large number of potentially observable civilizations. We show that this conflict arises from the use of Drake-like equations, which implicitly assume certainty regarding highly uncertain parameters. We examine these parameters, incorporating models of chemical and genetic transitions on paths to the origin of life, and show that extant scientific knowledge corresponds to uncertainties that span multiple orders of magnitude. This makes a stark difference. When the model is recast to represent realistic distributions of uncertainty, we find a substantial {\em ex ante} probability of there being no other intelligent life in our observable universe, and thus that there should be little surprise when we fail to detect any signs of it. This result dissolves the Fermi paradox, and in doing so removes any need to invoke speculative mechanisms by which civilizations would inevitably fail to have observable effects upon the universe., Comment: Submitted to Proceedings of the Royal Society of London A; 4 supplements

Item does not contain fulltext, BACKGROUND: Mild Cognitive Impairment (MCI) is a risk factor for Alzheimer's disease (AD) and other forms of dementia. However, much heterogeneity concerning neuropsychological measures, prevalence and progression rates impedes distinct diagnosis and treatment implications. OBJECTIVE: Aim of the present study was the identification of specific tests providing a high certainty for stable MCI and factors that precipitate instability of MCI in a community based sample examined at three measurement points. METHOD: 130 participants were tested annually with an extensive test battery including measures of memory, language, executive functions, intelligence and dementia screening tests. Exclusion criteria at baseline comprised, severe cognitive deficits (e.g. diagnosis of dementia, psychiatric or neurological disease). Possible predictors for stability or instability of MCI-diagnosis were analyzed using Regression and Receiver Operating Characteristic (ROC) curve analysis. Age, IQ and APOE status were tested for moderating effects on the interaction of test performances and group membership. RESULTS: A high prevalence of MCI (49%) was observed at baseline with a reversion rate of 18% after two years. Stability of MCI was related to performances in four measures (VLMT: delayed recall, CERAD: recall drawings, CERAD: Boston Naming Test, Benton Visual Retention Test: number of mistakes). Conversion to MCI is associated with language functions. Reversion to 'normal' was primarily predicted by single domain impairment. There was no significant influence of demographic, medical or genetic variables. CONCLUSION: The results highlight the role of repeated measurements for a reliable identification of functional neuropsychological predictors and better diagnostic reliability. In cases of high uncertainty close monitoring over time is needed in order of estimating outcome.

- Abstract
Advances in biological engineering are likely to have substantial impacts on global society. To explore these potential impacts we ran a horizon scanning exercise to capture a range of perspectives on the opportunities and risks presented by biological engineering. We first identified 70 potential issues, and then used an iterative process to prioritise 20 issues that we considered to be emerging, to have potential global impact, and to be relatively unknown outside the field of biological engineering. The issues identified may be of interest to researchers, businesses and policy makers in sectors such as health, energy, agriculture and the environment.

The SENTINEL study showed that the addition of natalizumab improved outcomes for patients with relapsing multiple sclerosis (MS) who had experienced disease activity while receiving interferon beta-1a (IFNbeta-1a) alone. Previously unreported secondary and tertiary magnetic resonance imaging (MRI) measures are presented here. Patients received natalizumab 300 mg (n=589) or placebo (n=582) intravenously every 4 weeks plus IFNbeta-1a 30 microg intramuscularly once weekly. Annual MRI scans allowed comparison of a range of MRI end points versus baseline. Over 2 years, 67% of patients receiving natalizumab plus IFNbeta-1a remained free of new or enlarging T2-lesions compared with 30% of patients receiving IFNbeta-1a alone. The mean change from baseline in T2 lesion volume over 2 years decreased in patients receiving natalizumab plus IFNbeta-1a and increased in those receiving IFNbeta-1a alone (-277.5mm(3) versus 525.6mm(3); p0.001). Compared with IFNbeta-1a alone, add-on natalizumab therapy resulted in a smaller increase in mean T1-hypointense lesion volume after 2 years (1821.3mm(3) versus 2210.5mm(3); p0.001), a smaller mean number of new T1-hypointense lesions over 2 years (2.3 versus 4.1; p0.001), and a slower rate of brain atrophy during the second year of therapy (-0.31% versus -0.40%; p=0.020). Natalizumab add-on therapy reduced gadolinium-enhancing, T1-hypointense, and T2 MRI lesion activity and slowed brain atrophy progression in patients with relapsing MS who experienced disease activity despite treatment with IFNbeta-1a alone.

Objective: To determine the incidence and clinical effects of antibodies that develop during treatment with natalizumab. Methods: In two randomized, double-blind, placebo-controlled studies (natalizumab safety and efficacy in relapsing remitting multiple sclerosis [MS, AFFIRM] and safety and efficacy of natalizumab in combination with interferon β-1a [INFβ1a] in patients with relapsing remitting MS [SENTINEL]) of patients with relapsing multiple sclerosis, blood samples were obtained at baseline and every 12 weeks to determine the presence of antibodies against natalizumab. Antibodies to natalizumab were measured using an ELISA. Patients were categorized as “transiently positive” if they had detectable antibodies (≥0.5 μg/mL) at a single time point or “persistently positive” if they had antibodies at two or more time points ≥6 weeks apart. Results: In the AFFIRM study, antibodies were detected in 57 of 625 (9%) of natalizumab-treated patients: Twenty (3%) were transiently positive and 37 (6%) were persistently positive. Persistently positive patients showed a loss of clinical efficacy as measured by disability progression ( p ≤ 0.05), relapse rate ( p = 0.009), and MRI ( p ≤ 0.05) compared with antibody-negative patients. In transiently positive patients, full efficacy was achieved after approximately 6 months of treatment, the time when patients were becoming antibody negative. The incidence of infusion-related adverse events was significantly higher in persistently positive patients. Results of SENTINEL were similar to AFFIRM, except with regard to sustained disability progression; differences between persistently positive and antibody-negative patients were not statistically significant. Conclusions: The incidence of persistent antibody positivity associated with natalizumab is 6%. Reduced clinical efficacy is apparent in persistently positive patients. Patients with a suboptimal clinical response or persistent infusion-related adverse events should be considered for antibody testing. Objective To report the relationship between disease activity and health-related quality of life (HRQoL) in relapsing multiple sclerosis, and the impact of natalizumab. Methods HRQoL data were available from 2,113 multiple sclerosis patients in natalizumab clinical studies. In the Natalizumab Safety and Efficacy in Relapsing Remitting Multiple Sclerosis (AFFIRM) study, patients received natalizumab 300mg (n = 627) or placebo (n = 315); in the Safety and Efficacy of Natalizumab in Combination with Interferon Beta-1a in Patients with Relapsing Remitting Multiple Sclerosis (SENTINEL) study, patients received interferon beta-1a (IFN-β-1a) plus natalizumab 300mg (n = 589), or IFN-β-1a plus placebo (n = 582). The Short Form-36 (SF-36) and a subject global assessment visual analog scale were administered at baseline and weeks 24, 52, and 104. Prespecified analyses included changes from baseline to week 104 in SF-36 and visual analog scale scores. Odds ratios for clinically meaningful improvement or worsening on the SF-36 Physical Component Summary (PCS) and Mental Component Summary were calculated. Results Mean baseline SF-36 scores were significantly less than the general US population and correlated with Expanded Disability Status Scale scores, sustained disability progression, relapse number, and increased volume of brain magnetic resonance imaging lesions. Natalizumab significantly improved SF-36 PCS and Mental Component Summary scores at week 104 in AFFIRM. PCS changes were significantly improved by week 24 and at all subsequent time points. Natalizumab-treated patients in both studies were more likely to experience clinically important improvement and less likely to experience clinically important deterioration on the SF-36 PCS. The visual analog scale also showed significantly improved HRQoL with natalizumab. Interpretation HRQoL was impaired in relapsing multiple sclerosis patients, correlated with severity of disease as measured by neurological ratings or magnetic resonance imaging, and improved significantly with natalizumab. Background Interferon beta is used to modify the course of relapsing multiple sclerosis. Despite interferon beta therapy, many patients have relapses. Natalizumab, an α 4 integrin antagonist, appeared to be safe and effective alone and when added to interferon beta-1a in preliminary studies. Methods We randomly assigned 1171 patients who, despite interferon beta-1a therapy, had had at least one relapse during the 12-month period before randomization to receive continued interferon beta-1a in combination with 300 mg of natalizumab (589 patients) or placebo (582 patients) intravenously every 4 weeks for up to 116 weeks. The primary end points were the rate of clinical relapse at 1 year and the cumulative probability of disability progression sustained for 12 weeks, as measured by the Expanded Disability Status Scale, at 2 years. Results Combination therapy resulted in a 24 percent reduction in the relative risk of sustained disability progression (hazard ratio, 0.76; 95 percent confidence interval, 0.61 to 0.96; P = 0.02). Kaplan–Meier estimates of the cumulative probability of progression at two years were 23 percent with combination therapy and 29 percent with interferon beta-1a alone. Lunar surface materials offer a source of raw materials for space processing to produce structural metals, oxygen, silicon, glass, and ceramic products. Significant difference exist, however, between lunar surface materials in the highlands and those in the maria. In the highlands the soil depth is at least an order of magnitude greater, the Al:Fe ratio is ten times greater, the content of plagioclase as a source of clear glass is three times as great, and the content of Ti is at least an order of magnitude lower. Evaluation of the extractive metallurgy and chemical operations associated with carbothermic and silicothermic refinement of lunar regolith suggests that Fe, Al, Si, Mg and probably Ti, Cr and Mn can be recovered, while oxygen is produced as a by-product. A conservative plant design yields its own weight in oxygen, silicon, and structural metals in less than six days. Power requirements for a throughput of 300,000 tons per year is less than 500 megawatts. The processing is done more economically in space than on the lunar surface.

ABSTRACT Hydrogen is known to have a deleterious effect on most engineering alloys. It has been shown repeatedly that the strength of steels is inversely related to the ductility of the material in hydrogen gas. However, the fatigue properties with respect to strength are not as well documented or understood. Here, we present the results of tests of the fatigue crack growth rate (FCGR) on API X70 from two sources. The two materials were tested in air, 5.5 and 34 MPa pressurized hydrogen gas, and at both 1 and 0.1 Hz. At these hydrogen pressures, the FCGR increases above that of air for all values of the stress intensity factor range (Δ K) greater than ~7 MPa · m1/2. The effect of hydrogen is particularly sensitive at values of Δ K below ~15 MPa · m1/2. That is, for values of Δ K between 7 and 15 MPa · m1/2, the FCGR rapidly increases from approximately that found in air to as much as two orders of magnitude above that in air. Above 15 MPa · m1/2, the FCGR remains approximately one to two orders of magnitude higher than that of air. [ABSTRACT FROM AUTHOR]

- Abstract
The review of cryogenic mechanical and thermal properties presented here is part of a broader NIST program to assess new high-strength Al-Li alloys for use in the cryogenic tankage of the Advanced Launch System. This program is sponsored by the Air Force Systems Command, Astronautics Laboratory, Edwards Air Force Base, with Bao Nguyen, Task Manager. It is part of the Materials and Process Validation (3101) of the Structures, Materials, and Manufacturing (3000) portion of the ALS Advanced Development Program. Since the purpose of the NIST program has been to assess the relative suitability of high-strength Al-Li alloys and alloy 2219 for us in ALS cryogenic tanks, data on Al-Li alloys 8090, 2090, WeldaliteTM 049, and Al alloy 2219 have been included in the survey. Properties covered in this survey are tensile strength, yield strength, elongation, fracture toughness, elastic constants, specific heat, thermal conductivity, and thermal expansion.

- Abstract
We found Cop 1 to be effective and relatively safe in a previous (exacerbating-remitting) clinical trial. This current trial involves 106 chronic-progressive patients. The major end point, confirmed progression of 1.0 or 1.5 units (depending on baseline disability) on the Kurtzke Expanded Disability Status Scale, was observed in nine (17.6) treated and 14 (25.5) control patients. The differences between the overall survival curves were not significant. Progression rates at 12 and 24 months were higher for the placebo group (p0.088) with 2-year probabilities of progressing of 20.4 for Cop 1 and 29.5 for placebo. We found a significant difference at 24 months between placebo and Cop 1 at one but not the other center. Two-year progression rates for two secondary end points, unconfirmed progression, and progression of 0.5 EDSS units, (p0.03) are significant.
