Abstract

Edible fungi are a valuable resource in the search for sustainable solutions to environmental pollution. Their ability to degrade organic pollutants, extract heavy metals, and restore ecological balance has a huge potential for bioremediation. They are also sustainable food resources. Edible fungi (basidiomycetes or fungi from other divisions) represent an underutilized resource in the field of bioremediation. By maximizing their unique capabilities, it is possible to develop innovative approaches for addressing environmental contamination. The aim of the present study was to find selective chemical agents suppressing the growth of microfungi and bacteria, but not suppressing white-rot fungi, in order to perform large-scale cultivation of white-rot fungi in natural unsterile substrates and use it for different purposes. One application could be the preparation of a matrix composed of wooden sleeper (contaminated with PAHs) and soil for further hazardous waste bioremediation using white-rot fungi. In vitro microbiological methods were applied, such as, firstly, compatibility tests between bacteria and white-rot fungi or microfungi, allowing us to evaluate the interaction between different organisms, and secondly, the addition of chemicals on the surface of a Petri dish with a test strain of microorganisms of white-rot fungi, allowing us to determine the impact of chemicals on the growth of organisms. This study shows that white-rot fungi are not compatible to grow with several rhizobacteria or bacteria isolated from soil and bioremediated waste. Therefore, the impact of several inorganic materials, such as lime (hydrated form), charcoal, dolomite powder, ash, gypsum, phosphogypsum, hydrogen peroxide, potassium permanganate, and sodium hydroxide, was evaluated on the growth of microfungi (sixteen strains), white-rot fungi (three strains), and bacteria (nine strains) in vitro. Charcoal, dolomite powder, gypsum, and phosphogypsum did not suppress the growth either of microfungi or of bacteria in the tested substrate, and even acted as promoters of their growth. The effects of the other agents tested were strain dependent. Potassium permanganate could be used for bacteria and Candida spp. growth suppression, but not for other microfungi. Lime showed promising results by suppressing the growth of microfungi and bacteria, but it also suppressed the growth of white-rot fungi. Hydrogen peroxide showed strong suppression of microfungi, and even had a bactericidal effect on some bacteria, but did not have an impact on white-rot fungi. The study highlights the practical utility of using hydrogen peroxide up to 3% as an effective biota-suppressing chemical agent prior to inoculating white-rot fungi in the large-scale bioremediation of polluted substrates, or in the large-scale cultivation for mushroom production as a foodstuff. Abstract

In capillary electrophoresis (CE), analyte identification is primarily based on migration time, which is a function of the analyte's electrophoretic mobility and the electro-osmotic flow (EOF). The migration time can be impacted by the presence of parasitic flow from changes in temperature or pressure during the run. Presented here is a high-voltage-compatible flow sensor capable of monitoring the volumetric flow inside the capillary during a separation with nL/min resolution. The direct measurement of both flow and time allows for compensation of flow instabilities. Abstract

The methodology described in this article will significantly reduce the time required for understanding the relations between chromatographic data and bioactivity assays. The methodology is a hybrid of hypothesis-based and data driven scientific approaches. In this work, a novel chromatographic data segmentation method is proposed, which demonstrates the capability of finding what volatile substances are responsible for antiviral and cytotoxic effects in the medicinal plant extracts. Up until now, the full potential of the separation methods has not been exploited in the life sciences. This was due to the lack of data ordering methods capable of adequately preparing the chromatographic information. Furthermore, the data analysis methods suffer from multidimensionality, requiring a large number of investigated data points. A new method is described for processing any chromatographic information into a vector. The obtained vectors of highly complex and different origin samples can be compared mathematically. The proposed method, efficient with relatively small sized data sets, does not suffer from multidimensionality. In this novel analytical approach, the samples did not need fractionation and purification, which is typically used in hypothesis-based scientific research. All investigations were performed using crude extracts possessing hundreds of phyto-substances. The antiviral properties of medicinal plant extracts were investigated using gas chromatography-mass spectrometry, antiviral tests, and proposed data analysis methods. The findings suggested that (i) beta-cis-caryophyllene, linalool, and eucalyptol possess antiviral activity, while (ii) thujones do not, and (iii) alpha-thujone, beta-thujone, cis-p-menthan-3-one, and estragole show cytotoxic effects.

Abstract

5-aminolevulino rūgštis (ALA) – provaistas naudojamas fotodinaminėje (FDT) terapijoje, protoporfirino IX prekursorius hemo biosintezės kelyje. Manoma, kad FDT vienas iš selektyviausių odos vėžio gydymo metodų. Nors ALA naudojimas FDT yra daug žadantis tiek terapiniu tiek komerciniais požiūriais, gydytojai vis dar sunkiai priimą šį gydymo metodą. Perkutaninės vaistinės formos dažnai kritikuojamos dėl nepakankamos skvarbos per raginį sluoksnį į odą ir dėl nepakankamo stabilumo. Skvarbai pagerinti yra kelios priemonės: skvarbos stiprikliai (dimetilsulfoksidas (DMSO), oleino rūgštis (OR) ir kt.), ALA derivatizacija į esterius, raginio sluoksnio pašalinimas ir kt. Dideli kiekiai DMSO, ar OR žymiai pagerina skvarbą, tačiau oda gali būti negrįžtamai pažeista. ALA esterių naudojimas vienas iš sėkmingesnių būdų įvesti ALA į odą, tačiau irgi nevisada sėkmingas. Kai kurios ligos pasireiškia jau su pažeistu raginiu sluoksniu (pvz. aktininė keratozė), šiuo atvejų gerai skvarbai užtikrinti nebūtina atlikti jokių papildomų farmacinės formos ruošimo, ar terapinių procedūrų. Viena iš buvusių opiausių ALA farmacinių formų problemų – stabilumas tiek farmacinių formų saugojimo metu, tiek atliekant ekstrakciją biocheminių tyrimų metų. Daugumos autorių nuomone ALA stabilumas užtikrinamas sumažinant pH bent iki 5. Tačiau per daug sumažinus galima sukelti odos paraudimus, ar net pažeidimus, be to mažas pH slopina PpIX biosintezę. Penetration enhancers such as dimethylsulfoxide (DMSO) or oleic acid (OA) could be used in order to improve penetration to skin. Some skin lessions provide disrupted SC barrier (actinic keratosis). In this case, ALA penetration to skin is often sufficient to obtain therapeutical effect. Various authors state that ALA to be stable at pH 5 or lower, but too low pH can result in skin erythema, or more seriuos damage, moreover low pH inhibits PpIX biosynthesis. Nowadays there are numerous articles about ALA in skin penetration studies. Mostly, the HPLC method method might be epmloyed with a requirement of common derivatization reactions for amino acid determination. Penetration enhancers such as dimethylsulfoxide (DMSO) or oleic acid (OA) could be used in order to improve penetration to skin. ALA derivatization to obtain more lipopfilic esters and stratum corneum disruption procedures could also be used for penetration improvement. Some skin lessions provide disrupted SC barrier (actinic keratosis). In this case, ALA penetration to skin is often sufficient to obtain therapeutical effect. Various authors state that ALA to be stable at pH 5 or lower, but too low pH can result in skin erythema, or more seriuos damage, moreover low pH inhibits PpIX biosynthesis. Nowadays there are numerous articles about ALA in skin penetration studies. Mostly, the HPLC method method might be epmloyed with a requirement of common derivatization reactions for amino acid determination. Liquid scintillation may also be employed in penetration studies, but it is quite expensive and require radioactive substances for quantitative determination. There are only few methods available for direct analysis. Ion pairing, ion exchange chromathography and capillary electrophoresis (CE) may be used for these purposes. CE offers cheap, rapid, low running buffer and sample volumes requiring method, suitable for microanalysis. High selectivity is provided by CE method, which has been validated according to ICH guidelines. Mathematical and statistical calculations ensures analytical procedure‘s suitability for ALA penetration studies.After applying cream, determined concentrations in derma were: after 4 h – 14,7 µg/cm3 (±40,1%), after 8 h – 34,1 µg/cm3 (±23,6%), after 12 h – 51,7 µg/cm3 (±28,6%). After lipid extraction from SC employment, penetration dramatically increased. Determined concentrations in derma were: after 4 h – 60,4 µg/cm3 (±19,3%); 8 h – 96,7 µg/cm3 (±12,0%); 12 h – 137,2 µg/cm3 (±3,2%).

Abstract

Medicinal plants are reported to possess antiviral activity, but finding the substances that are responsible for antiviral activity in the complex mixture of the plant extract is an extremely difficult task. In this paper a methodology related to the determination of the antiviral properties of medicinal plant extracts and based on phytochemical analysis, antiviral tests and machine learning methods is described. 16 potentially antiviral medicinal plants were selected, and their chemometric characteristics and antiviral properties were investigated. Three different analytical methods were used for chemical analysis: (i) spectrophotometry, (ii) capillary electrophoresis with contactless conductivity detection, and (iii) gas chromatography-mass spectrometry. 14 attributes were obtained describing the composition of the plant extracts. Viral growth inhibition properties were investigated and 8 candidate plant extracts were selected as being active against viruses. Infectious bronchitis virus was used as a model virus. Machine learning techniques including deep neural network classification, classification and regression tree induction and hierarchical clusterization were used for mining the factors that are responsible for antiviral effects. It was determined that (i) phenolic compounds providing high radical scavenging activity and fractions containing high content of phenolic compounds are positively related to antiviral activity in plant extracts, (ii) hydrophilic compounds that are positively charged (pKa> 4.7) in acidic media and possess medium and low electrophoretic mobility properties are negatively related to antiviral activity in medicinal plants, (iii) phenolic acids with pKalower than 4.7 are not related to antiviral activity in the extracts, and (iv) volatile compounds in the extracts, including diversity, quantity and different volatility properties, do not affect the antiviral activity of plant extracts. Following the proposed methodological approach, it is possible to confirm which chemometric attributes are responsible for antiviral activity in medicinal plant extracts.

Abstract

A group of 36 fungal strains, belonging to the Lithuanian mycobiota, was collected and isolated from different locations, habitats, and matrices, including creosote-treated wood in storage yards for crosstie wastes. The eight most perspective strains selected according to preliminary assessment of tolerance to coal tar were subsequently identified combining taxonomical evaluation and molecular techniques. The tolerance of the eight identified fungal species (five basidiomycetes and three ascomycetes) to the presence of various concentrations of coal tar, and for the four most perspective fungal strains (Pleurotussp.,Schizophyllumsp.,Irpex lacteus,Bjerkandera adusta) to polycyclic aromatic hydrocarbons was evaluated. The ligninolytic enzymatic activity assay of the isolated strains resulted in a good correspondence between the tolerance to pollutants and the capability to produce ligninolytic enzymes indicating that this group of white-rot fungi is perspective for further investigation and eventual usage for mycoremediation of polycyclic aromatic hydrocarbons polluted substrates. Abstract

Construction of a contactless conductivity detector with a separate small dimension detection cell and integration into an automated capillary electrophoresis device is described. An optimized separation procedure of the pressurized hot water extract of Hibiscus sabdariffa L. and implementation of L-ascorbic acid as a novel background electrolyte in capillary zone electrophoresis with contactless conductivity detection are described. Optimized conditions allowed a rapid separation in less than 7 minutes with complementary data of UV-VIS absorbance and contactless conductivity detectors recorded simultaneously. The pressurized hot water extraction showed superior recovery to classical cold water extraction with increased peak areas by 9.4 times ± 9.7%, 2.8 times ± 9.7% and 4.1 times ± 7.2% for pigments, inorganic cations and total peak area, respectively.
