3. Children’s understanding of well-being related questions: results of cognitive interviews in four European countries

Abstract

This paper presents the results of cognitive interviews with 8-year-old children from four European countries–Croatia, France, Finland, and Ireland. The aim of the interviews was to pre-test a selection of well-being-related questions as a part of questionnaire development for the first European multinational birth cohort study–Growing up in Digital Europe (GUIDE)/EuroCohort. Unlike most previous studies, we focused on a younger and more age-homogenous sample, as well as a more diverse set of well-known questionnaires. A total of 68 children participated in the study. The main suggestion for the interviewing procedure is to create a safe environment yet minimize the parents’ interference in answering. The questionnaires should use child-friendly vocabulary, tangible examples, avoid complex sentence structure and negative statements. The use of timeframes in questions should be minimal. The children can use Likert-type scales, but the number of different scales in the questionnaire should be limited.

Abstract

The Breast Surgery theoretical and practical knowledge curriculum comprehensively describes the knowledge and skills expected of a fully trained surgeon practicing in the European Union and European Economic Area (EEA). It forms part of a range of factors that contribute to the delivery of high quality cancer care. It has been developed by a panel of experts from across Europe and has been validated by professional breast surgery societies in Europe. The curriculum maps closely to the syllabus of the Union of European Medical Specialists (UEMS) Breast Surgery Exam, the UK FRCS (breast specialist interest) curriculum and other professional standards across Europe and globally (USA Society of Surgical Oncology, SSO). It is envisioned that this will serve as the basis for breast surgery training, examination and accreditation across Europe to harmonise and raise standards as breast surgery develops as a separate discipline from its parent specialties (general surgery, gynaecology, surgical oncology and plastic surgery).\ud \ud \ud \ud The curriculum is not static but will be revised and updated by the curriculum development group of the European Breast Surgical Oncology Certification group (BRESO) every 2 years.

14. Optimisation of noise equivalent count rates for brain and body FDG imaging using gamma camera PET

Abstract

This paper describes the use of Noise Equivalent Count rates to optimise the clinical use of a modified dual headed gamma camera for positron emission tomography. Phantoms were used to simulate the distribution and uptake for brain and body 18F-fluorodeoxyglucose imaging, including the presence of activity outside the imaging field of view. Camera count rates were recorded as a function of activity in the field of view in the presence and absence of lead septa. The use of variable number and width of energy windows was also investigated. Results indicate that acquisitions without axial collimators is optimal for brain imaging while a restriction of 12 [degrees] in the axial acceptance angle leads to superior NEC rates for torso imaging. The most significant factor affecting the NEC rates for body imaging with no axial collimation is the detection of random coincidences from out of field activity. Under most imaging conditions the use of a Compton-photopeak energy window in addition to the traditional photopeak-photopeak appears to lead to increased NEC rates, although an accurate scatter correction would be necessary to compensate for the associated increase in the scatter fraction.

Abstract

Analysis of DNA methylation patterns relies increasingly on sequencing-based profiling methods. The four most frequently used sequencing-based technologies are the bisulfite-based methods MethylC-seq and reduced representation bisulfite sequencing (RRBS), and the enrichment-based techniques methylated DNA immunoprecipitation sequencing (MeDIP-seq) and methylated DNA binding domain sequencing (MBD-seq). We applied all four methods to biological replicates of human embryonic stem cells to assess their genome-wide CpG coverage, resolution, cost, concordance and the influence of CpG density and genomic context. The methylation levels assessed by the two bisulfite methods were concordant (their difference did not exceed a given threshold) for 82% for CpGs and 99% of the non-CpG cytosines. Using binary methylation calls, the two enrichment methods were 99% concordant and regions assessed by all four methods were 97% concordant. We combined MeDIP-seq with methylation-sensitive restriction enzyme (MRE-seq) sequencing for comprehensive methylome coverage at lower cost. This, along with RNA-seq and ChIP-seq of the ES cells enabled us to detect regions with allele-specific epigenetic states, identifying most known imprinted regions and new loci with monoallelic epigenetic marks and monoallelic expression.
