Die Pyranose-2-Oxidase aus Peniophora gigantea lag zu Beginn dieser Arbeit in E. coli kloniert vor und trug die Austausche E540K und K312E. Durch gerichtete Evolution in Kombination mit rationalem Design konnte die katalytische Effizienz dieser Variante durch Einführen der Austausche N71Y und S456N für das Substrat 2-Desoxy-D-glucose um das 30,3-fache erhöht werden. Ein rationales Design im aktiven Zentrum an Position T169 zur Verbesserung der Affinität zu D-Galactose hatte eine Variante mit 4,9-fach erhöhter katalytischer Effizienz zum Zielsubstrat zur Folge. Außerdem besaß die T169G eine 62-fach erhöhte katalytische Effizienz für LArabinose, welche ebenfalls ein sehr interessantes Substrat darstellt. Durch Austausch hydrophober Aminosäuren in der stark exponierten Kopfregion des Enzyms wurde die Bildung von inclusion bodies um 38% gesenkt und eine vielseitige und stabile Variante entwickelt (P2OxB2H1). Eine Coexpression mit dem Triggerfaktor aus E. coli bewirkte eine weitere Verbesserung auf 7,5%. Daneben konnte die Enzymausbeute durch Etablierung eines Verfahrens zur Hochzelldichtefermentation von E. coli um das 12,7-fache auf 6333 U/L gesteigert werden. Durch MALDI TOF-Analyse von Pepsin-fragmentierter P2Ox vor und nach Biokonversion wurde ein oxidationsanfälliges Methionin isoliert und per Sättigungsmutagenese durch Glutamin ersetzt. Die neue Variante wies eine höhere operative Stabilität auf und blieb auch bei Einsatz in mehreren aufeinanderfolgenden Konversionen aktiv. The Pyranose 2-Oxidase from Peniophora gigantea was cloned into E. coli prior to this work and mutated at positions E540K and K312E. The catalytic efficiency of this variant was optimized via directed evolution and rational design by introducing the substitutions N71Y and S456N and thus raised 30,3-fold for 2-Deoxy-D-glucose. Rational design at the active site at position T169 led to a variant (T169G) with 4,9- fold higher catalytic efficiency for D-Galactose and a 62-fold higher catalytic efficiency for L-Arabinose, another substrate of high interest. Substitution of hydrophobic amino acid residues in the highly exposed head domain by hydrophilic ones lowered the inclusion body formation by 38% and yielded a versatile and stable variant, P2OxB2H1. Coexpression of trigger factor from E. coli reduced the inclusion bodies further down to 7,5%. By establishing a method of high cell density fed batch fermentation of E. coli, enyzme yield of P2OxB2H1 could be raised 12,7-fold to 6333 U/L. By MALDI TOF analysis of P2Ox digested with pepsin prior to and after bioconversion, a crucial methionine for oxidation could be isolated and substituted by a glutamate via saturation mutagenesis. The new variant showed higher operational stability and remained active even after multible rounds of bioconversion.
