In study I, plasma progesterone concentrations were evaluated in anoestrous mares that received an intravaginal progesterone release device (IPRD) for 10 days. Mares were divided into 3 groups based on the dosage of progesterone (0 g, n=3; 1.38 g, n=5; and 1.9 g, n=5). No statistical differences were found in plasma progesterone concentrations between the two doses tested. In study II, the effects of a protocol based on a short program of artificial light combined with an IPRD containing 1.38 g of progesterone on oestrous behaviour and onset of ovulation were evaluated. IPRDs were inserted into 31 late transitional mares (10 days of treatment). The mares were divided into a control group (n=9, IPRD with 0 g of progesterone) and two treatment groups (T1, n=10, IPRD with 0 g of progesterone and artificial light; T2, n=12, IPRD with 1.38 g of progesterone and artificial light). The percentages of mares in heat within the first 14 days after treatment were 100%, 70%, and 100% in the control, T1, and T2 groups, respectively (P=0.097), and their ovulation rates were 44%, 60%, and 100%, respectively (P≤0.01). In conclusion, a protocol based on artificial light and an IPRD containing 1.38 g of progesterone for 10 days could be considered to advance the first ovulation of the year in late transitional mares, as it ensures a higher rate of ovulation within the first 14 days after treatment., (©2022 The Japanese Society of Equine Science.)

The objective of this study was to assess the efficacy of a subcutaneous melatonin implant to suppress estrus in queens (felis catus). The hypothesis was that this implant would temporarily and reversibly suppress estrus in queens without producing any clinically detectable side effects. Fourteen adult queens were maintained in cages under artificial illumination (14h light:10h dark) for 45 d and then randomly assigned to one of two treatments. At interestrus, queens received a single subcutaneous melatonin implant (18mg; Melovine [CEVA Sante Animal, Libourne, France]; MEL: n=9), or a single subcutaneous placebo implant without melatonin (0mg; PLA; n=5). At the next estrus, all queens received a second MEL (n=9) or PLA (n=5) implant. Blood samples were taken when queens displayed estrous signs and during interestrus to measure estradiol (E(2)) and progesterone (P(4)), respectively, by radioimmunoassay. There were no significant differences in duration of the interestrus interval in PLA cats, regardless of whether the implants were placed during interestrus or estrus (6.0+/-9.7 d vs. 6.0+/-9.7 d, respectively; least square means [LSM]+/-SEM). However, when MEL implants were placed during interestrus, the duration of interestrus was approximately twice as long as that occurring when MEL implants were placed during estrus (113.3+/-6.1 d vs. 61.1+/-6.8 d, respectively; P<0.01). Serum E(2) and P(4) concentrations were similar in queens with PLA and MEL implants and in queens that received implants in estrus and interestrus. In conclusion, a subcutaneous MEL implant effectively and reversibly suppressed estrus in queens for approximately 2 to 4 mo with no clinically detectable side effects.

Endometrial mRNA expression of the pro-inflammatory cytokines interleukin-1beta (IL-1beta), interleukin-6 (IL-6), and tumor necrosis factor alpha (TNF-alpha) was assessed in mares resistant (RM) or susceptible (SM) to persistent post-breeding endometritis (PPBE). Eight RM and eight SM, were selected based on reproductive records and functional tests out of a herd of 2,000 light cross-type mares. Three experiments were done to study transcription patterns in (i) basal conditions; (ii) after artificial insemination (AI); and (iii) after administration of an immunomodulator at time of artificial insemination. Endometrial biopsies were taken during consecutive cycles: (i) at estrus, when follicles reached 35 mm and at diestrus (7 +/- 1 days after ovulation); (ii) at 24 h post-AI, with dead semen (estrus) and in diestrus; (iii) at 24 h after treatment with a Mycobacterium phlei cell-wall extract (MCWE) preparation and AI (with dead semen), and at diestrus. mRNA expression was quantitated by real time PCR. Under basal conditions, SM had significantly higher mRNA expression of all cytokines in estrus and of IL-1beta and TNF-alpha in diestrus, compared to RM. After AI, there were no differences between RM and SM in estrus; however, mRNA expression for all three pro-inflammatory cytokines was higher than under basal conditions. In diestrus, RM showed significantly lower IL-1beta and TNF-alpha mRNA expression than SM. When MCWE was administered at time of AI, no differences between cytokine induction from RM and SM were found. Globally, mRNA expression for all three cytokines correlated well among themselves when expression was high. The present study showed that (i) in basal conditions RM had lower mRNA expression of pro-inflammatory cytokines than SM with no effect of estrous cycle; (ii) AI upregulated mRNA expression for all three cytokines in both RM and SM, with persistance in diestrus in the latter; (iii) treatment with MCWE at time of AI down-regulated mRNA expression of IL-1 with significant effects in SM which behaved like RM. Immunomodulation with MCWE could be of help in restoring homeostatic local inflammatory mechanisms, thus assisting in the prophylaxis of post-breeding endometritis in mares.

The intraperitoneal transfusion of red cells in rats and dogs was evaluated as an alternative route for blood restoration. Erythrocytes labeled with 59Fe, equivalent to about 50% of the estimated total red cell volume, were injected in the abdominal cavity of 5 dogs. Three other dogs and 12 rats were injected with a volume proportional to the 9%. The maximal incorporation of tagged red cells in the circulation of the rats was of 96 +/- 7% at 48 h. One half of the erythrocytes disappeared from the circulation 29 days later. Computing data from all dogs, the mean highest value, 91.2 +/- 6%, was reached at 96 h and although random destruction of the cells occurred, the estimated T1/2 of survival was about 66 days. The injection was completed in about 15 seconds in the rats and in 2 to 4 minutes in the dogs. At the necropsy only minor serosal reactions were observed in the peritoneal cavity of the animals. Intraperitoneal transfusion can be recommended as a rapid, safe and efficient procedure for blood supply.
