Source at https://doi.org/10.1371/journal.pone.0209273. We provide the first detailed identification of Barents Sea cold seep frenulate hosts and their symbionts. Mitochondrial COI sequence analysis, in combination with detailed morphological investigations through both light and electron microscopy was used for identifying frenulate hosts, and comparing them to Oligobrachia haakonmosbiensis and Oligobrachia webbi, two morphologically similar species known from the Norwegian Sea. Specimens from sites previously assumed to host O. haakonmosbiensis were included in our molecular analysis, which allowed us to provide new insight on the debate regarding species identity of these Oligobrachia worms. Our results indicate that high Arctic seeps are inhabited by a species that though closely related to Oligobrachia haakonmosbiensis, is nonetheless distinct. We refer to this group as the Oligobrachia sp. CPL-clade, based on the colloquial names of the sites they are currently known to inhabit. Since members of the Oligobrachia sp. CPL-clade cannot be distinguished from O. haakonmosbiensis or O. webbi based on morphology, we suggest that a complex of cryptic Oligobrachia species inhabit seeps in the Norwegian Sea and the Arctic. The symbionts of the Oligobrachia sp. CPL-clade were also found to be closely related to O. haakonmosbiensis symbionts, but genetically distinct. Fluorescent in situ hybridization and transmission electron micrographs revealed extremely dense populations of bacteria within the trophosome of members of the Oligobrachia sp. CPL-clade, which is unusual for frenulates. Bacterial genes for sulfur oxidation were detected and small rod shaped bacteria (round in cross section), typical of siboglinid-associated sulfur-oxidizing bacteria, were seen on electron micrographs of trophosome bacteriocytes, suggesting that sulfide constitutes the main energy source. We hypothesize that specific, local geochemical conditions, in particular, high sulfide fluxes and concentrations could account for the unusually high symbiont densities in members of the Oligrobrachia sp. CPL-clade.

Within the Sea of Marmara, the highly active North Anatolian Fault (NAF) is responsible for major earthquakes (Mw>=7), and acts as a pathway for fluid migration from deep sources to the seafloor. This work reports on pore water geochemistry from three sediment cores collected in the Gulfs of Izmit and Gemlik, along the Northern and the Middle strands of the NAF, respectively. The resulting data set shows that anaerobic oxidation of methane (AOM) is the major process responsible for sulfate depletion in the shallow sediment. In the Gulf of Gemlik, depth concentration profiles of both sulfate and alkalinity exhibit a kink-type profile. The Sulfate Methane Transition Zone (SMTZ) is located at moderate depth in the area. In the Gulf of Izmit, the low concentrations observed near the seawater-sediment interface for sulfate, calcium, strontium, and magnesium result from rapid geochemical processes, AOM, and carbonate precipitation, occurring in the uppermost part of the sedimentary column and sustained by free methane accumulation. Barite dissolution and carbonate recrystallization have also been identified at deeper depth at the easternmost basin of the Gulf of Izmit. This is supported by the profile of the strontium isotope ratios (87Sr/86Sr) as a function of depth which exhibits negative anomalies compared to the modern seawater value. The strontium isotopic signature also shows that these carbonates had precipitated during the reconnection of the Sea of Marmara with the Mediterranean Sea. Finally, a first attempt to interpret the sulfate profiles observed in the light of the seismic activity at both sites is presented. We propose the hypothesis that seismic activity in the areas is responsible for the transient sulfate profile, and that the very shallow SMTZ depths observed in the Gulf of Izmit is likely due to episodic release of significant amount of methane.

As a part of the Dutch–French MEDINAUT diving expedition in 1998, cold seeps and mud volcanoes were studied and sampled in two distinctive tectonic settings in the eastern Mediterranean Sea. The first setting was the Olimpi Mud Volcano field (OMV area), including Napoli, Milano, Maidstone and Moscow mud volcanoes, south of Crete on the Mediterranean ridge. The second setting was the Anaximander Mountains (AM area), southwestern Turkey, where Amsterdam, Kazan and Kula mud volcanoes were explored. Large methane concentrations (45–892 nmol/kg) were measured in the water column not only above mud volcanoes but also in seeps and vents along related fault systems, indicating intense degassing related to fluid circulation in sediments. The tracer results show that there is considerable variability in terms of gas seepage and matter flux between these mud volcanoes. Brine accumulations found as shallow pools on Napoli or associated with deep faults (Nadir Lake) outside mud volcanoes exhibit variable chlorinity, mineral and gas composition. The brines are significantly enriched in δ 18 O relative to ambient seawater and are consistent with evaporated seawater. In the Nadir Brine Lake, the level of methane is as high as 5.93 mmol/kg, lower than the methane saturation level of 120 mmol/kg theoretically found at the salinity (120), pressure (200 bar), and temperature (13.6°C) conditions of Nadir lake. In contrast, the shallow brine pools on Napoli mud volcano (also OMV area) have methane levels of only 4.45 μmol/kg. In all brines, helium data show a clear radiogenic isotopic ratio ( R =0.06× R a ), in excellent agreement with recently published data for the Urania basin. Methane to ethane ratios (>1000) and δ 13 C(CH 4 ) values (−65.6‰PDB) indicate that the CH 4 is microbially produced. Unlike mid-ocean ridges, where abiogenic methane and helium have a common origin in the brines, the large variation in the CH 4 /He ratio indicates that CH 4 and helium sources are unrelated, a fact that adds further support to the biogenic origin of methane. These results show that mud volcanoes in the eastern Mediterranean Sea are important sites of extensive biogenic methane fluxes, which are probably related to widespread occurrences of gas hydrates.
