Dipeptidyl peptidase 4 (DPP4; CD26) is involved in the regulation of various metabolic, immunological, and neurobiological processes in healthy individuals. Observations based on epidemiological data indicate that DPP4 inhibition by gliptins, typically used in patients with diabetes, may reduce the risk for cerebral ischemia and may also improve related outcomes. However, as DPP4 inhibitor application is neither complete nor specific for suppression of DPP4 enzymatic activity and DPP4 has non-enzymatic functions as well, the variety of consequences is a matter of debate. Therefore, we here used DPP4 knock-out (KO) mice to analyze the specific contribution of DPP4 to cellular, immunological, and functional consequences of experimental focal cerebral ischemia.We observed a significantly higher survival rate of DPP4 KO mice after ischemia, which was accompanied by a lower abundance of the pro-inflammatory chemokine CCL2 and reduced activation of Iba1-positive microglia cells in brain tissue of DPP4 KO mice. In addition, after ischemia for 24 h to 72 h, decreased concentrations of CCL5 and CCL12 in plasma and of CCL17 in brain tissue of DPP4 KO mice were observed when compared to wild type mice. Other aspects analyzed, such as the functional Menzies score, astrocyte activation and chemokine levels in plasma and brain tissue were affected by ischemia but appeared to be unaffected by the DPP4 KO genotype.Taken together, experimental ablation of DPP4 functions in mice improves survival and ameliorates aspects of cellular and molecular inflammation after focal cerebral ischemia.

This Indigenous-led project aims to better understand historical and contemporary ways in which Alaska Native Peoples steward salmon and the values connected to salmon stewardship. Indigenous Peoples have stewarded Alaska lands and waters for thousands of years yet have been largely excluded from western science and management systems. In this project, we utilize a participatory approach that allows for the equitable valuing of ideas and knowledges to document the breadth and depth of Yup'ik and Athabascan knowledge and governance systems in southwestern Alaska. We reshape research methodologies by centering Indigenous frameworks and methodologies, including circle dialogues and multi-generational interviews led by Indigenous scholars and students in their home communities and regions. In this paper, we share the Yup'ik and Athabascan values, knowledge, management, and governance mechanisms that can improve the long-term sustainability and equity of Alaska salmon systems. This research elevates the voices of Alaska Native salmon stewards and experts from the Kuskokwim Bay and the Kuskokwim River. We elaborate on five key themes that emerged from this research, including traditional Indigenous ways of life, Indigenous stewardship, self-determination, food and livelihood sovereignty, and ecosystem changes, and identify a more equitable and sustainable path forward for salmon and people in Alaska.

This article provides perspectives on the graying of the fleet and research efforts to understand it in Alaska fisheries, discusses recent abrupt multifaceted challenges faced by Alaskan fishermen, and compares divergent public perceptions and support programs for farmers and fishermen in the United States. Equity concerns have seen a resurgence in academic and policy realms and the graying of the fleet is now a common example of distributional inequity, yet despite in-depth research attention on the issue, few substantive programmatic modifications have been made to address it in Alaska. Aging trends in United States fisheries have typically been framed as one of changing aspirational preferences. Research suggests coastal youth do desire and highly value fishing careers, but structural and financial barriers limit opportunities for new generations. The precarity of the fishing industry was highlighted in 2023 with sudden global seafood market instability, rising interest rates and operating costs, destabilized geopolitical tensions, and hastening climate change impacts. This recent industry turmoil disproportionately impacts those on the margins and adds an additional layer of complexity to addressing graying of the fleet trends. Unlike in fisheries, there are numerous federal programs to support young farmers and the agricultural industry. This paper calls for proactive and comprehensive measures to stabilize and support fishing communities and the viability of next generation pathways amidst recent fishing industry market turmoil and longer term inequities in fishery governance.

Glutaminyl cyclase (QC) and its isoenzyme (isoQC) catalyze the formation of N-terminal pyroglutamate (pGlu) from glutamine on a number of neuropeptides, peptide hormones and chemokines. Chemokines of the C-C ligand (CCL) motif family are known to contribute to inflammation in neurodegenerative conditions. Here, we used a model of transient focal cerebral ischemia to explore functional, cellular and molecular responses to ischemia in mice lacking genes for QC, isoQC and their substrate CCL2. Mice of the different genotypes were evaluated for functional consequences of stroke, infarct volume, activation of glia cells, and for QC, isoQC and CCL2 expression. The number of QC-immunoreactive, but not of isoQC-immunoreactive, neurons increased robustly in the infarct area at 24 and 72 h after ischemia. In parallel, immunohistochemical signals for the QC substrate CCL2 increased from 24 to 72 h after ischemia induction without differences between genotypes analyzed. The increase in CCL2 was accompanied by morphological activation of Iba1-immunoreactive microglia and recruitment of MHC-II-positive cells at 72 h after ischemia. Among other chemokines quantified in the brain tissue, CCL17 showed higher concentrations at 72 h compared to 24 h after ischemia. Collectively, these data suggest a critical role for QC in inflammatory processes in the stroke-affected brain.

Large and grand IT projects seem to fail worldwide. Several studies researched the indicators of success and failure on a national level, but there is little international comparative research. What if governments could apply international lessons learned when starting their IT projects? We assessed the factors of success of 110 projects in the Netherlands by the project benchmark resolution method of the Standish Group and compared large governmental and small projects to an international sample of the Standish database. This study supports the relation between the Dutch IT Projects and the international sample of the Standish Group, thus offering an opportunity to learn from international projects when applied to the large and grand governmental IT-projects and small IT-projects in the Netherlands. Acknowledging international post-mortem lessons learned could raise awareness of applying an international project resolution benchmark in the prenatal phase of IT-projects

Indigenous Peoples and salmon in the lands now called Alaska have been closely entwined for at least 12,000 years. Salmon continue to be central to the ways of life of Alaska Natives, contributing to physical, social, economic, cultural, spiritual, psychological, and emotional well-being. Salmon have also become important to Alaskan settlers. Our research and advisory team conducted a synthesis of what is known about these diverse human-salmon relationships, drawing on 865 published scientific studies; Indigenous knowledge; state, federal, and tribal data; archival materials; oral histories; and cross-cultural dialogs at working group meetings. Two important socio-cultural dimensions of salmon-people systems emerged from this synthesis as fundamentally important but largely invisible outside of Indigenous communities and the social science disciplines that work closely with these communities: (1) the deep relationships between Indigenous Peoples and salmon and (2) the pronounced inequities that threaten these relationships and stewardship systems. These deep relationships are evident in the spiritual, cultural, social, and economic centrality of salmon across time and cultures in Alaska. We describe Indigenous salmon stewardship systems for the Tlingit, Ahtna, and Central Yup'ik. The inequities in Alaska's salmon systems are evident in the criminalization and limitation of traditional fishing ways of life and the dramatic alienation of Indigenous fishing rights. The loss of fish camps and legal battles over traditional hunting and fishing rights through time has caused deep hardship and stress. Statewide, the commodification and marketization of commercial fishing rights has dispossessed Indigenous communities from their human and cultural rights to fishing ways of life; as a result, many rural and Indigenous youth struggle to gain access to fishing livelihoods, leaving many fishing communities in a precarious state. These deep relationships and relatively recent fractures have motivated a concerted effort by a group of committed Indigenous and western scholars to better understand the root causes and opportunities for redress, as well as to document the breadth of research that has already been conducted, in an effort to improve the visibility of these often-overlooked dimensions of our salmon systems.

Salmon are intrinsic to health and well-being in Alaska, and sit at the center of myriad social, cultural, and spiritual practices, norms, and values. These practices and values are essential to living and being well in many communities in Alaska, but often remain invisible and unaccounted for in management contexts. This paper stems from the collective efforts of a cross-disciplinary, cross-cultural project team brought together as part of the State of Alaska's Salmon and People (SASAP) knowledge synthesis project. In this paper, we assess the sustainability and equity of Alaska salmon systems through a well-being framework. Key objectives include (1) defining and conceptualizing well-being in the context of Alaska salmon systems; (2) developing and assessing well-being indicators for Alaska salmon systems; and (3) evaluating how well-being concepts are currently incorporated into Alaska salmon management and suggesting improvements. We draw on specific examples to evaluate the application of well-being indicators as a tool to more effectively measure and evaluate social considerations, and discuss how to better integrate well-being concepts into governance and management to improve data collection and decision making. As part of this effort, we discuss trends and inequities in Alaska fisheries and communities that impact well-being, and tensions between equality and equity in the context of Alaska salmon management.

Purpose: The authors tested associations between (a) parent-reported temporary vs. persistent vocabulary delay and (b) parent-reported behavioral/emotional problems in a sample of 5,497 young Dutch children participating in a prospective population-based study. Method: Mothers completed the MacArthur Communicative Development Inventory--Netherlands (Zink & Lejaegere, 2003) at age 18 months and the Language Development Survey (Rescorla, 1989) at age 30 months, with expressive vocabulary delay defined as scores in the lowest 15th age and gender-specific percentiles. The Child Behavior Checklist (Achenbach & Rescorla, 2000) was completed by mothers when their children were age 18 months and by both parents when their children were age 36 months, from which Internalizing Problems and Externalizing Problems scores were analyzed. Results: All analyses were adjusted for covariates. Expressive vocabulary delay at age 18 months was weakly related to Internalizing Problems scores at age 18 months as well as mother-reported Externalizing and Internalizing Problems scores at age 36 months (the latter for boys only). Expressive vocabulary delay at age 30 months was weakly associated with mother-reported Externalizing and Internalizing Problems scores (the latter for boys only) and father-reported Internalizing Problems scores. Persistent expressive vocabulary delay predicted the highest risk of mother-reported internalizing and externalizing problems at age 36 months. Conclusion: This population-based study showed modest associations between vocabulary delay and behavioral/emotional problems detectable from 18 months onward. Objective: We sought to better understand what defines a critical incident experience for the surgical trainee. Summary Background Data: Critical incidents are formative moments stamped indelibly on one's memory that shape professional identity. The critical incident technique—using participants' narratives to identify patterns and learn from their perceptions—has been explored in some healthcare settings, but there has been no inquiry within surgery. Methods: Surgical residents at 5 residency programs (1 community, 1 university-affiliated, 3 university) were surveyed using an online questionnaire from November to December 2020. Convenience sampling was used to identify the study population. Participants were invited to write about formative, impactful experiences in training. Interpretive description was the qualitative methodology used to locate information, analyze, and record patterns in the data. Individual responses were categorized and assessed for overlying themes. Results: Overall, 28 narratives were collected from surgery residents in 3 specialties (general surgery, plastic surgery, and urology), with postgraduate year representation of post-graduate years 1 to 6. Respondents were 40% female. Nineteen of the narratives reported a negative experience. Four themes were identified from responses: 1) growth through personal self-reflection, 2) difficult interpersonal interactions, 3) positive team dynamics as a psychological safety net, and 4) supportive program cultures that promote learning. Conclusions: Critical incident narratives among surgical residents indicate that unforgettable and formative experiences—both positive and negative— occur in 4 domains: within the individual, within a relationship, among a team, and within a program. The rise in attention to equity as a goal in fisheries management has revealed a dearth of data related to how certain communities and groups participate in, rely on, and may be impacted by fishery management decisions. Lack of data on impacts to Tribal and Indigenous participants and other underserved communities have been identified as a primary barrier to assessing and improving equity considerations in fishery governance. Data on fishery participation trends, including who benefits from fisheries, is essential to determining if and how fishery management and decision-making are achieving desired management objectives such as providing for intergenerational access and preserving a way of life. Understanding changes in participation trends is also critical for developing effective conservation solutions. In Alaska salmon fisheries, shifts in permit holdings are generally categorized by the residency of permit holders. This means that little is known about how Alaska Native participation, as well as women's participation, in Alaska commercial fisheries has changed over time. This paper presents a case study on local salmon permit holdings in the Bristol Bay region of southwest Alaska. We discuss the consequences of conflating rural with Alaska Native in data-driven decision-making, and highlight the ways in which official fishery statistics can mask past and ongoing harms to Indigenous communities and peoples. This paper reports on the "de-segregation" of students with disabilities, focusing on processes occurring before pupils actually enter the mainstream. De-segregation involves identifying target children, discussing the situation among staff members, evaluation by educational psychologist, discussing with parents and the child, selecting a mainstream school, and reviewing after six weeks of mainstreaming. Abstract This paper presents results from a study that explored a key threat to fisheries sustainability in Alaska – the graying of the commercial fishing fleet. In the Kodiak Archipelago region this research utilized a political ecology framework and mixed methods ethnography, including 70 semi-structured interviews and participant observation, to further understand this problem. Study results suggest that opportunities for young rural fishermen are increasingly constrained by interrelated socioeconomic and cultural barriers, which have created systemic equity and sustainability concerns. Furthermore, research indicates that the privatization paradigm of fisheries access is a major catalyst of change that has created and amplified barriers, transformed opportunity, and generated lasting inequities and social conflict. Foucault's concept of governmentality is used to describe how some fishermen are internalizing and normalizing privatization discourses to advance further regulatory change. Pauly's concept of shifting baseline syndrome is evoked to argue that current structures of degraded access and equity in the human fishery system are mistakenly assumed to be a natural state, rather than a result of a specific history of public policy choices. Due to the suite of challenges facing fishing people and communities, it is increasingly important to acknowledge the privatization of access as a key threat to sustainable coastal fishing futures. Engaging other health care providers in oral health-related activities and interprofessional care (IPC) could increase access to oral health care for underserved populations in the U.S. The aims of this study were to assess dental hygiene, dental, and medical students' intra- and interprofessional and oral and maxillofacial surgery (OMFS)/hospital dentistry-related knowledge/skills, attitudes, and behavior; determine whether first and second year vs. third and fourth year cohorts' responses differed; and explore how intra- and interprofessional knowledge was related to interprofessional education (IPE) and interprofessional attitudes and behavior. Data were collected between April 2014 and May 2015 from 69 dental hygiene, 316 dental, and 187 medical students. Response rates across classes for the dental hygiene students ranged from 85% to 100%; 24% to 100% for the dental students; and 13% to 35% for the medical students. The results showed that the medical students had lower oral health-related and interprofessional knowledge and less positive attitudes about oral health-related behavior, IPE, and interprofessional teamwork than the dental hygiene and dental students. While third- and fourth-year medical students' interprofessional knowledge/skills and behavior were higher than those of first- and second-year students, the two groups' IPE-related and interprofessional attitudes did not differ. The students' knowledge correlated with their IPE and interprofessional communication-related skills and behavior, but not with their interprofessional attitudes. These dental hygiene, dental, and medical students' OMFS/hospital dentistry-related knowledge/skills and behavior increased over the course of their academic programs, while their IPE-related and intra- and interprofessional attitudes, especially for medical students, did not improve over time. OMFS and hospital dentistry units in medical centers offer distinctive opportunities for IPE and IPC. Utilizing these units may be one way to ensure that graduating providers are motivated to engage in IPC in their practice, thus contributing to reducing oral health disparities and increasing access to oral care for underserved populations. In 2012, the North Pacific Fishery Management Council initiated the process of designing a new management structure for the Gulf of Alaska trawl groundfish fleet. The new program is currently structured as a catch share program and driven by the need to end the ‘race for fish’ and provide the trawl fleet with the tools to reduce bycatch. To date much of the discussion among policy makers and stakeholders has centered on community protection measures and how best to avoid the negative social and economic impacts of catch share programs including impacts to crew, consolidation and the flight of quota and resource wealth from Gulf of Alaska fishery dependent communities. This paper examines how community protection measures related to the distribution of benefits and access to fishery resources are considered and challenged in the North Pacific today. Special attention is given to the ongoing debate surrounding the potential inclusion of an initial allocation to place-based Gulf of Alaska communities via a Community Fishing Association. As part of this effort, this paper examines the political space and underlying power dynamics in which consideration of alternatives to the commodification of fishing rights occurs. The challenge of designing institutions and resource policy for ecosystem and social resilience in rural and small-scale fisheries is receiving renewed attention in Alaska and elsewhere. Many rural and Indigenous fishing communities have been negatively impacted by modern resource allocation and management regimes that restrict and privatize fishery access through the creation of individual property rights. This article draws on ethnographic and interview data from a multi-sited study to improve policy considerations for rural and small-scale fisheries access. The Bristol Bay region of southwest Alaska is a site of concerning social trends including the 'graying of the fleet' and a rise in nonlocal ownership of fishing rights. Since the state began limiting entry into salmon fisheries in 1975, local permit holdings in Bristol Bay communities have declined by roughly 50%. This paper examines the ways in which assumptions and norms operating within state, regional, and local institutions support and/or constrain local fishing practices and participation in the region. A central objective is to challenge dominant and universalist assumptions of fishermen as dis-embedded, profit-maximizing, self-interested actors that do not fit well with small-scale, rural, and Indigenous fisheries. This paper identifies social relationships and interdependencies as central to rural fishing communities and livelihoods and absent from the rational choice/individual economic actor assumptions of modern resource allocation and management regimes.
