Polyacrylamide gel (PAAG) as a novel embedding medium enhances the in situ detection and imaging of metabolites in biological fragile tissues (e.g., eyeballs) via MALDI-MSI.

Michler's ethylketone (MEK, 4,4’-bis(diethylamino)benzophenone), commonly-known as an intermediate in the synthesis of dyes and pigments, was successfully screened and optimized as a novel matrix for the enhancement of lipid in situ detection and imaging in tissues by MALDI-MSI. The results show several properties of MEK as a powerful MALDI matrix, including strong UV absorption, μm-sized crystals and uniform matrix-coating, super high vacuum chemical stability, low matrix-related ion interference, super soft ionization, as well as high lipid ionization efficiency.

BACKGROUND: Lower urinary tract symptoms (LUTSs) are the most common problem in older men with benign prostatic hyperplasia (BPH). Physical activity and sedentariness may be important LUTS risk factors and suitable targets for intervention. However, what is the most effective way to engage in physical activity? There is still no consensus among academics. OBJECTIVE: To observe which of the two different physical activity modalities, square dancing (SD) or walking, is more effective in delaying the effects of LUTSs/BPH in older men and to provide a basis for promoting active participation in appropriate physical activity for BPH patients. DESIGN: This study was a chain design study with cross-sectional (June 2018), retrospective (from June 2014 to June 2018) and follow-up (June 2018 to June 2020) components for LUTSs. PARTICIPANTS: Men aged 60 and over who could perform physical activity alone and lived in the same community. MAIN MEASURES: The effect of different forms of physical activity on the alleviation of LUTSs in BPH patients was observed retrospectively; the main physical activity patterns of older adults in the town were walking and SD. Therefore, the two physical activity modalities of walking and SD were used as the main study factors, and the propensity score 1:1 matching method was applied to control for confounding factors in the retrospective cohort, which was divided into walking and SD groups to compare the clinical progression of LUTSs in the two groups. The time between study entry into the cohort and LUTS progression (appearance or exacerbation of symptoms) was used as the outcome to determine whether LUTS progression occurred in the two groups. The Cox proportional hazards model was applied to analyze the effect of walking and SD on the delay in the clinical progression of LUTSs/BPH in older men. KEY RESULT: Multifactorial Cox regression analysis showed that patients in the square dancing group had a 0.41-fold lower risk of LUTSs than those in the walking group (HR=0.41, 95% CI: 0.19-0.88, p=0.02). CONCLUSIONS: SD can more effectively delay the clinical progression of LUTSs in BPH patients, and the results of this study provide a good reference for the selection of self-guided physical activity intervention modalities for BPH patients.

To our knowledge, this was the first study in which caffeic acid (CA) was successfully evaluated as a matrix to enhance the in situ detection and imaging of endogenous proteins in three biological tissue sections (i.e., a rat brain and Capparis masaikai and germinating soybean seeds) by matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). Our results show several properties of CA, including strong ultraviolet absorption, a super-wide MS detection mass range close to 200,000 Da, micrometer-sized matrix crystals, uniform matrix deposition, and high ionization efficiency. More high-molecular-weight (HMW) protein ion signals (m/z > 30,000) could be clearly detected in biological tissues with the use of CA, compared to two commonly used MALDI matrices, i.e., sinapinic acid (SA) and ferulic acid (FA). Notably, CA shows excellent performance for HMW protein in situ detection from biological tissues in the mass range m/z > 80,000, compared to the use of SA and FA. Furthermore, the use of a CA matrix also significantly enhanced the imaging of proteins on the surface of selected biological tissue sections. Three HMW protein ion signals (m/z 50,419, m/z 65,874, and m/z 191,872) from a rat brain, two sweet proteins (mabinlin-2 and mabinlin-4) from a Capparis masaikai seed, and three HMW protein ion signals (m/z 94,838, m/z 134,204, and m/z 198,738) from a germinating soybean seed were successfully imaged for the first time. Our study proves that CA has the potential to become a standard organic acid matrix for enhanced tissue imaging of HMW proteins by MALDI-MSI in both animal and plant tissues.

This study used constitutive CD226 gene knockout (KO) mice as a model to investigate the functions and mechanisms of CD226 in megakaryocyte (MK) maturation and platelet activation. Although CD226 deficiency did not cause MK polyploidization or platelet granule abnormalities, increased MK counts were detected in the femora bone marrow (BM) and spleen of CD226 KO mice. Particularly, CD226 KO mice have a more extensive membrane system in MKs and platelets than wild-type (WT) mice. We also demonstrated that CD226 KO mice displayed increased platelet counts, shortened bleeding time, and enhanced platelet aggregation. CD226 KO platelets had an increased mature platelet ratio compared to the control platelets. In addition, the observed reduction in bleeding time may be due to decreased nitric oxide (NO) production in the platelets. Platelet-specific CD226-deficient mice showed similar increased MK counts, shortened bleeding time, enhanced platelet aggregation, and decreased NO production in platelets. Furthermore, we performed middle cerebral artery occlusion-reperfusion surgery on WT and CD226 KO mice to explore the potential effect of CD226 on acute ischemia-reperfusion injury; the results revealed that CD226 deficiency led to significantly increased infarct area. Thus, CD226 is a promising candidate for the treatment of thrombotic disorders.

Collective motion is one of the most fascinating phenomena and mainly caused by the interactions between individuals. Physical-barriers, as the particular facilities which divide the crowd into different lanes, greatly affect the measurement of such interactions. In this paper we propose the physical-barrier detection based collective motion analysis (PDCMA) approach. The main idea is that the interaction between spatially adjacent pedestrians actually does not exist if they are separated by the physical-barrier. Firstly, the physical-barriers are extracted by two-stage clustering. The scene is automatically divided into several motion regions. Secondly, local region collectiveness is calculated to represent the interactions between pedestrians in each region. Finally, extensive evaluations use the three typical methods, i.e., the PDCMA, the Collectiveness, and the average normalized Velocity, to show the efficiency and efficacy of our approach in the scenes with and without physical barriers. Moreover, several escalator scenes are selected as the typical physical-barrier test scenes to demonstrate the performance of our approach. Compared with the current collectivemotion analysis methods, our approach better adapts to the scenes with physical barriers.

The fast proliferation of cloud computing promotes the rapid growth of datacenters. More and more cloud service providers use geo-distributed green datacenters to support the expanding scale of cloud applications as well as minimize the carbon footprint. In such a geo-distributed green datacenter system, a basic and urgent demand is inter-datacenter bulk data transfer that is usually used for periodic data backup, software distribution, virtual machines cloning, etc. Though many existing research efforts have been made to build green datacenters or provide optimal scheduling for inter-datacenter bulk data transfers separately, still the goal for optimal scheduling of inter-green-datacenter bulk data transfers is being underachieved. This is an important problem, especially when an increasing number of geo-distributed datacenters are powered by renewable energy for reducing energy cost and protecting environment. In this paper, we study the problem of maximizing renewable energy use and minimizing grid energy cost for bulk data transfers between sustainable and green datacenters. We model this problem and propose a heuristic method to solve it. The proposed method is the first to explicitly address the green energy use maximization and grid energy cost minimization problem of inter-green-datacenter bulk data transfers for green and sustainable datacenters in the multi-electricity market environment. Extensive evaluations with real-life network topology, available wind power, and electricity prices show that our method can maximize renewable energy use and bring more energy cost savings over existing bulk data transfer strategies.

BackgroundOsteosarcoma (OS) is a highly malignant and aggressive bone tumor. This study was performed to explore the mechanisms of HuR (human antigen R) in the progression of OS.MethodsHuR expression levels in OS tissues and cells were detected by immunohistochemistry and western blotting. HuR siRNA was transfected into SJSA-1 OS cells to downregulate HuR expression, and then cell proliferation, migration, and epithelial-mesenchymal transition (EMT) were evaluated. RNA immunoprecipitation was performed to determine the association of the long non-coding RNA (lncRNA) XIST and argonaute RISC catalytic component (AGO) 2 with HuR. Fluorescence in situ hybridization analysis was performed to detect the expression of lncRNA XIST. Western blotting and immunofluorescence assays were performed to observe AGO2 expression after HuR or/and lncRNA XIST knockdown.ResultsKnockdown of HuR repressed OS cell migration and EMT. AGO2 was identified as a target of HuR and silencing of HuR decreased AGO2 expression. The lncRNA XIST was associated with HuR-mediated AGO2 suppression. Moreover, knockdown of AGO2 significantly inhibited cell proliferation, migration, and EMT in OS.ConclusionOur findings indicate that HuR knockdown suppresses OS cell EMT by regulating lncRNA XIST/AGO2 signaling.

Background: Leukocyte-associated immunoglobulin-like receptor-1 (LAIR-1) is a collagen receptor belonging to the immunoglobulin superfamily. Although prior studies have evaluated the biological role of LAIR in solid tumors, the precise mechanisms underlying LAIR-1 functions as a regulator of tumor biological functions remains unclear. Methods: LAIR-1 expression was evaluated using an osteosarcoma (OS) tissue microarray by immunohistochemical analysis. Wound healing and Transwell assays were performed to evaluate tumor cell migration. Quantitative PCR and western blotting were conducted to detect the expression of epithelial-mesenchymal transition (EMT)-related molecules. RNA-sequencing (RNA-seq) was conducted to evaluate the mRNA expression profiles after overexpressing LAIR-1 in OS cells. Glucose uptake and glucose transporter (Glut) 1 expression in OS cells in vitro were evaluated by flow cytometry and western blotting. Results: LAIR-1 expression significantly differed between the T1 and T2 stages of OS tumors, and LAIR-1 overexpression inhibited OS cell migration. LAIR-1 expression was inversely correlated with the expression of EMT-associated transcription factors via the Forkhead box O1/Twist1 signal transduction pathway. Furthermore, RNA-seq and quantitative PCR demonstrated that EMT energy metabolism-related molecules were significantly reduced after LAIR-1 overexpression. Conclusions: Notably, overexpression of LAIR-1 in OS cells decreased Glut1 expression. These findings provide insight into the molecular mechanism underlying OS progression.

Gemin5 acts as a U1 small nuclear RNA (snRNA)-binding protein in U1 small nuclear ribonucleic protein (snRNP) biogenesis. Here, we report a role for Gemin5 in unassembled U1 snRNP disposal under survival of motor neuron (SMN) protein-deficient conditions. We demonstrate that non-Sm protein-associated U1 snRNA and U1A are enriched in cytoplasmic granules and colocalize to P bodies in SMN-deficient cells. Immunoprecipitation assays show increased associations of the U1 snRNP component U1A with P body components and Gemin5 in SMN-deficient cells. More importantly, Gemin5 knockdown eliminates the unassembled U1 snRNP granules and rescues U1 snRNA levels in SMN-deficient cells. Taken together, our study provides direct evidence that Gemin5 is involved in unassembled-U1 snRNA disposal under conditions of SMN deficiency.

Crowd behavior simulation has become one of the key support technologies for evacuation drills and safety monitoring. However, complex psychology, personality traits and interactions among individuals increase the challenges of creating realistic crowd behavioral simulations. In this paper, we propose a crowd behavior simulation method to simulate crowd behaviors by combining the shadow obstacle model (SOM) and optimal reciprocal collision avoidance (ORCA) model. First, the SOM is converted to the expected speed, as well as the half plane of ORCA. Then, a manual generation method of SOM is proposed to simulate the crowd behaviors in corners or beside a non-closed wall. Finally, we combine behavior simulation and physical simulation techniques, and propose a velocity-based crowd simulation framework. To test the effectiveness of our proposed method, several simulation methods are implemented in a Unity3D engine-based crowd simulation in subway station environment, and the comparison results demonstrate that our proposed method outperforms force-based simulation methods.

Counting pedestrians in surveillance videos has become an urgent safety concern in critical areas. However, surveillance videos of subway spaces suffer from severe crowd occlusion and perspective distortion. In this paper, a novel double-region learning algorithm is presented to overcome these challenges. The main idea of this algorithm is to identify the best two-region boundary and then design a reasonable pedestrian-counting method in each separated region. First, a separate line is obtained via possibility learning, and each frame is divided into a nearby region and a distant region to eliminate the influence of perspective distortion. Second, in the nearby region, we apply the improved aggregate channel feature detection to count the number of pedestrians N 1 . In the distant region, we employ the Extreme Learning Machine and Gaussian Process regression methods to estimate the number of pedestrians N 2 . Finally, the total number of pedestrians in each frame can be obtained with high accuracy according to N 1 and N 2 . We establish a subway pedestrian video dataset about several typical subway stations in Shanghai to validate the algorithm performance. Various experimental results demonstrate that the accuracy of the proposed approach surpasses that of compared methods, which means that our algorithm can meet the management requirements of subway stations.

In this study, we observed that deletion of CD226 on regulatory T cells (Tregs) precedes renal fibrosis in a mouse unilateral ureteral obstruction (UUO) model. First, we generated Treg-specific CD226 gene knockout mice (CD226fl/fl Foxp3YFP-Cre). Next, CD226fl/fl Foxp3YFP-Cre mice and Foxp3YFP-Cre control mice were subjected to UUO surgery. Pathologic analysis and Sirius red and Masson's trichrome staining showed that the kidneys of CD226fl/fl Foxp3YFP-Cre mice following UUO showed much more severe interstitial fibrosis than Foxp3YFP-Cre control mice at days 10 and 20. Additionally, CD226fl/fl Foxp3YFP-Cre mice showed increased fibronectin expression, as demonstrated by immunohistochemistry (IHC) staining. Although Treg cell-restricted CD226 deficiency showed increased Foxp3+ expression, expression of the cell surface functional molecule CD103 was significantly reduced, indicating impaired homeostasis in the Tregs of CD226fl/fl Foxp3YFP-Cre mice. To better understand CD226 function, RNA sequencing (RNA-Seq) analysis was conducted in Tregs isolated from CD226fl/fl Foxp3YFP-Cre and Foxp3YFP-Cre mice. RNA-Seq data showed that the helper T cell (Th) 2-related cytokines IL-4 and IL-10 were significantly up-regulated in CD226 deficient Tregs. In addition, mRNA analysis of kidney samples from the mice following UUO by qPCR also showed increased IL-4 and IL-10 expression in CD226fl/fl Foxp3YFP-Cre mice, as well as elevated TGF-β1 levels, indicating that CD226 deficiency in Tregs resulted in the acquisition of the ability to produce Th2 cytokines. Finally, we found that microRNA-340 (miR-340), which was down-regulated in Tregs isolated from CD226fl/fl Foxp3YFP-Cre mice, directly regulated IL-4 gene expression in vitro. These data suggest that the promotion of CD226 signaling on Tregs is a therapeutic target for renal disease.

The aging property of poly(1-butene) in which different antioxidant systems were added was characterized by its physical and mechanical properties before and after aging for 300 h in a hot air aging box. In addition, the oxidation induction time (OIT) of all designed samples was tested. The result demonstrated that the primary antioxidant pentaerythritol tetrakys 3-(3,5-ditert-butyl-4-hydroxyphenyl) propionate performed better than n-octadecyl-β-(4-hydroxy-3,5-di-tert -butyl-phenyl)-propionate. The anti-aging effect of the secondary antioxidant was ranked in the order bis-(2,4-di-tert-butyl-pheny)-phosphiterythritol diphosphite better than tns-(2.4-di-tert-butyl)-phosphite, with both better than distearyl thiodipropionic acid. Confirmation of oxidation of the polymer was made by IR analysis and a general oxidation mechanism scheme for poly(1-butene) is presented.

This study was conducted to investigate the effect of CD226 on the differentiation, activation, and polyploidization of megakaryocytes (MKs) and explore the potential mechanism. Dami (megakaryocyte line) cell maturation was induced by phorbol 12-myristate 13-acetate. CD226 was silenced by infection with a CD226-specific shRNA lentiviral vector. The mRNA level of CD226 was detected by qRT-PCR. The expressions of Dami cells surface CD226, MK specific markers CD41 and CD62P, and DNA ploidy in Dami cells and CD226 knockdown (KD) cells were evaluated by flow cytometry. The effect of CD226 on the expression of megakaryocyte-associated transcription factors was measured by western blot and confocal analysis. Transfection with CD226 shRNA lentivirus dramatically decreased the level of CD226 and expression of CD62 P in Dami cells. Silencing of CD226 caused morphological changes and differentiation retardation in low-ploidy MK. Furthermore, CD226 knockout (KO) mice exhibited increased 2N–4N low-ploidy MK and decreased ≥8N polyploidy. Interestingly, silencing of CD226 in megakaryocytic cells down-regulated the expression of early stage transcription factors includes GATA‐binding factor 1 (GATA-1) and friend leukemia integration 1 (FLI-1), but not late-stage nuclear factor, erythroid 2 (NF-E2). CD226 is involved in MKs activation and polyploidy cell cycle control.

The high penetration of third-party intellectual property (3PIP) brings a high risk of malicious inclusions and data leakage in products due to the planted hardware Trojans, and system level security constraints have recently been proposed for MPSoCs protection against hardware Trojans. However, secret communication still can be established in the context of the proposed security constraints, and thus, another type of security constraints is also introduced to fully prevent such malicious inclusions. In addition, fulfilling the security constraints incurs serious overhead of schedule length, and a two-stage performance-constrained task scheduling algorithm is then proposed to maintain most of the security constraints. In the first stage, the schedule length is iteratively reduced by assigning sets of adjacent tasks into the same core after calculating the maximum weight independent set of a graph consisting of all timing critical paths. In the second stage, tasks are assigned to proper IP vendors and scheduled to time periods with a minimization of cores required. The experimental results show that our work reduces the schedule length of a task graph, while only a small number of security constraints are violated.

Bacterial epigenetic modifications play key roles in cellular processes such as stress responses, DNA replication, segregation, antimicrobial resistance, etc. In recent years, emerging new sequencing technologies, including single-molecule real-time (SMRT) sequencing, and nanopore sequencing, have enabled the directed reading of epigenetic modifications without pre-treatment of DNA or DNA amplification. The applications of SMRT and nanopore sequencing open the door for the identification of more diverse epigenetic modifications of DNA bases and backbones and potentially facilitate the understanding of the novel functions of these epigenetic markers in cell physiology. With ongoing improvements in throughput and accuracy, new-generation sequencing has become a contender as an alternative to second-generation sequencing technologies. Here, the authors review recent advances in bacterial epigenetic analysis using SMRT sequencing and nanopore sequencing to provide insights regarding the detection and analysis of DNA epigenetic modifications in bacterial genomes.

In situ FTIR testing demonstrated that crystallization of novel poly(1-butene)/poly(propylene-co-butene) in-reactor alloys from melt shows thermodynamic stable Form I directly rather than general reported unstable crystal Form II. In order to make clear this phenomenon, the microstructure and monomer sequence distribution of the as-obtained poly(1-butene)/poly(propylene-co-butene) in-reactor alloys was determined by 13C-NMR. The raw alloy was separated by fractional dissolution to five grades. The characterization of raw alloys and different grades demonstrates that propylene monomer unit distributes in a form of isolated or segmented along poly(1-butene) molecular chains. The fractional dissolution of the selected alloy indicates that chain structure changes gradually with fractional solvent. The number average sequence length of propylene and 1-butene unit has been calculated. The number average sequence length and distribution could help us to study the crystallization and transformation clearly. From the results of in situ FTIR and NMR, the random distribution of the propylene unit with certain content in the as-obtained alloys play role to accelerate the crystallization transformation.

Objective: Systems engineering theory, hand-hold fundus camera(FC) and internet cooperative platform were applied to help community staff, long distance specialists' organizations and teams to cooperate on the screening for diabetic fundus pathological change(or diabetic retinopathy, DR) in regional residents. It aims at promoting the ability to prevent and control (PC) the important non-infectious chronic diseases (NCD) to design a proper value-chain for mutually beneficial mode of interactions among regional health services, and the other corresponding social roles via technological and organizational innovation.

To assess the relationship between nodular goiter and hepatitis C virus infection.Ninety-seven cases of early treatment in patients with chronic hepatitis C were collected for analysis.Data on patient age,sex,hepatitis duration and other general information were collected.In addition, data on clinical measures of thyroid function (including T3, t4, tSH) and thyroid autoantibodies (thyroid peroxidase antibody TPO-Ab, thyroglobulin antibody Tg-Ab), as well as findings from thyroid dimensional ultrasonography were collected. One hundred and eleven cases of early treatment in patients with chronic hepatitis B and 106 eases of females 40 years old or older with high risk of nodular goiter were collected for use as controls.The relationship between nodular goiter with thyroid function, thyroid autoantibodies levels,sex,age,and hepatitis C virus infection were statistically analyzed.The prevalence rates of nodular goiter in the chronic hepatitis C group, the chronic hepatitis B group and the more than or equal to 40 year-old women with high risk of nodular goiter were 53.6%,36.9% and 59.4% respectively.The prevalence rates of nodular goiter in the chronic hepatitis C group and the more than or equal to 40 year-old women with high risk of nodular goiter were significantly higber than that in the chronic hepatitis B group (x² values: 5.820 and 10.996, P0.05). The average age of patients with chronic hepatitis C combined with nodular goiter was significantly higher than their counterparts without goiter (F=6.408, P0.05),and the prevalence rate in the more than or equal to 40 year-old women with high risk of nodular goiter was significantly higher than that of their counterparts who were less than 40 years-old (60.0% vs. 23.5%; x² =7.499, P less than 0.05). The prevalence of nodular goiter in patients with chronic hepatitis C was significantly greater for females than for males (62.1% vs. 41.0%; x 2 =4.152, P0.05).The prevalence of nodular goiter in patients with chronic hepatitis C was also significantly higher for females more than or equal to 40 years old than for males (70.2%, 33/47 vs. 45.5%,15/33; x² = 4.952, P0.05).The duration of hepatitis, thyroid function and thyroid autoantibodies were similar between the patients in the chronic hepatitis C group with or without nodular goiter.The patients with chronic hepatitis C had a higher prevalence of nodular goiter,with an average of up to 53.6%, than the patients with chronic hepatitis B,and the women the more than or equal to 40 years old had even higher prevalence, at 70.2%, suggesting that patients with chronic hepatitis C should be routinely examined by thyroid ultrasound. Thyroid function and thyroid autoantibodies were not correlated with prevalence of goiter among the chronic hepatitis C patients.

Tumor necrosis factor receptor-associated factor 1 (TRAF1), an adapter in signal transduction, is involved in immunity and in apoptotic processes in various cell types. However, little is known about its function and the molecular mechanism of its activation during liver injury. This study tested the hypothesis that TRAF1 is a mediator of cell injury after hepatic ischemia/reperfusion injury (I/R). In a mouse hepatic I/R injury model, we found that TRAF1 expression was highly induced. TRAF1 deficiency was liver protective, whereas sustained TRAF1 overexpression aggravated liver injury in response to hepatic I/R injury. Mechanistic studies demonstrated that a deficiency of TRAF1 in cultured hepatocytes led to the inhibition of NF-κB-mediated inflammatory responses, suppression of the ASK/JNK pro-death pathway and promotion of cellular regeneration capacity. In contrast, the converse occurred in hepatocyte-specific TRAF1 transgenic mice. TRAF1 activated the ASK1/JNK pathway and promoted hepatic injury. Our study demonstrates that TRAF1 is a crucial early mediator of hepatic I/R injury and suggests that TRAF1 may be a potential gene therapy target for the treatment of liver injury.

Crystallization and polymorphic transformation of poly(1-butene)/(1-butene-co-propylene) in-reactor alloy (iPB/B-co-P) with different chain structure prepared by two stage sequential polymerization was investigated by DSC, FTIR and POM. As can be concluded from DSC results, the crystal form II to I transformation rate is enhanced with the increase of propylene unit content for alloy with the chain structure of isolated propylene unit distribution. However, the alloy which has the chain structure of predominant segmented propylene co-unit distribution, although with the highest propylene unit content and lowestmolecular weight, the crystal form II to I transformation rate still lower than two of the three alloys with isolated propylene unit and lower propylene unit content. This conclusion was also demonstrated by the subsequent in-situ FT-IR characterization. The crystallization process and morphology was investigated through polarized optical microscopy.

The polymorphic transformations (from form II to form I) of three kinds of isotactic poly(1-butene) were characterized by FTIR and by changes in their macroscopic physical and mechanical properties, including density and tensile characteristics. The kinetics of the form II to form I transformation were followed by performing FTIR measurements in real time. At 25 °C, as the crystallization time increased, the characteristic FTIR peak of form I of the poly(1-butene) increased, while that of form II diminished and finally disappeared. The polymorphic transformation from form II to form I of the poly(1-butene) obtained by the solution method (S-PB) occurred the most rapidly, followed by the corresponding transformation for the poly(1-butene) obtained from Mitsui Chemicals (M-PB) and then that for the poly(1-butene) produced using the high-pressure bulk polymerization method (B-PB). This transformation behavior is due to the higher isotacticity of the S-PB, as also demonstrated by the results of the density and tensile tests. The density of S-PB is higher than the densities of M-PB and B-PB due to its greater crystallinity.

To screen the anti-tumor active parts from Tripterygium hypoglaucum by anti-tumor experimental model in vivo and in vitro.Ethanol extraction was separated and purified by column chromatography of ion polymeric adsorbent and macroporous adsorptive resins. MTT assay and the inhibition effect to S180 solid tumor were used to detect anti-tumor activity of each separation.There are anti-tumor activities in the ethanol extraction, total alkaloids, and macroporous resin absorption in vivo and vitro. Minimum IC50 of Total alkaloids was 29.90 mg L(-1), and S180 solid tumor inhibition ratio of different dose of 25, 50, 100 mg kg(-1) were 38.10%, 50.60%, 60.71% respectively. Minimum IC50 of macroporous resin absorption was 98.56 mg L(-1), and S180 solid tumor inhibition ratio of different dose of 100, 200, 400 mg kg(-1) were 42.96%, 53.57%, 63.79% respectively. Water solubility position had no effect in vivo and vitro.T. hypoglaucum has fine anti-tumor activity in vivo and in vitro, and total alkaloids are the main part of anti-tumor active part.
