12. The Schmidt reaction of some alphabeta-unsaturated ketones

Abstract

Benzylideneacetone has been subjected to the Schmidt reaction in sulphuric acid under various conditions, and the only amide isolated was N-methylcinnamamide. Benzylideneacetons p-methoxy-, p-nitro- and m-nitro-benzylideneacetones were subjected to the Schmidt reaction in hydrochloric acid and in polyphosphoric acid. In hydrochloric acid each ketone gave a good yield of the N-methylcinnamamide. In polyphosphoric acid the nitrobenzylideneacetones gave a mixture of the N-styrylacetamide and the N-methylcinnamamide, benzylideneacetone gave mainly N-methylcinnamamide, only a trace of N-styrylacetamide being isolated in some runs of shortened duration, and the N-methylcinnamamide was the only amide isolated from p-methoxybenzyl-ideneacetone. The formation of aniline and p-anisidine during the reactions of benzylideneacetone and p-methoxybenzylidene-acetone in polyphosphoric acid has been explained. A mechanism has been proposed to explain the exclusive production of the N-methylcinnamamides in hydrochloric acid, and the novel variation of the amide ratio with solvent observed in the reactions with the nitrobenzylideneacetones. The Beckmann rearrangements of the alpha-oximes of benzylideneacetone, p-nitro- and m-nitro-benzylideneacetones have been re-examined. Methyl vinyl ketone, methyl isopropenyl ketone and mesityl oxide were subjected to Schmidt reaction conditions in hydrochloric acid. Ammonia was isolated from each reaction and, in addition methyl vinyl ketone gave diacetyl, methyl isopropenyl ketone gave methyl ethyl ketone, and mesityl oxide gave methylamine and acetone. Addition of hydrazoic acid to the olefinic bond of the ketones yielded azidoketones, decomposition of which in acids gave ammonia in each case, diacetyl and ethylenediamine from the azide of methyl vinyl ketone, shown to be 1-azidobutan-3-one, and methylamine from the azide of mesityl oxide. A mechanism for the decomposition of 1-azidobutan-3-one has been proposed.

Abstract

Blood-group-specific glycoproteins obtained from ovarian cyst fluids of A1 and A2 persons were degraded with NaOH/NaBH4. The oligosaccharides released were de-N-acetylated with Ba(OH)2 and then hydrolysed with dilute H2SO4. The products were fractionated on columns of ion-exchange resin and the components isolated were re-N-acetylated with 14C-labelled acetic anhydride; further purification was effected by paper chromatography. The following trisaccharides: type 1, GaINAc(αl - 3)Gal(βl - 3)G1cNAc; type 2, Ga1NAc(αl - 3)- Gal(βl -4)GlcNAc; type 3 (reduced), GalNAc(αl - 3)Gal(β1 - 3)GalNAcOH (where Gal is galactose, GalNAc is N-acetylgalactosamine, G1cNAc is N-acetylglucosamine and Ga1NAcOH is N-acetylgalactosaminitol) were isolated and characterised from both the A1 and A2 materials. The type 3 (reduced) trisaccharide has not previously been obtained from human glycoproteins. Chromatographic evidence indicated that the three trie saccharide structures were also present in other A1, A2, A1B and A2B ovarian cyst glycoproteins and in A1 and A2 salivary glycoproteins. Abstract

The tetrasaccharides GalNAcß1-4[NeuAcα2-3]Galß1-4Glc and GalNAcß1-4[NeuAcα2-3]Galß1-4GlcNAc were synthesised by enzymic transfer of GalNAc from UDP-GalNAc to 3′-sialyllactose (NeuAcα2-3Galß1-4Glc) and 3′-sialyl-N-acetyllactosamine (NeuAcα2-3Galß1-4GlcNAc). The structures of the products were established by methylation and1H-500 MHz NMR spectroscopy. In Sdaserological tests the product formed with 3′-sialyl-N-acetyllactosamine was highly active whereas that formed with 3′-sialyllactose had only weak activity.

42. Purification of the blood groupHgene associated α-2-l-fucosyltransferase from human plasma

Abstract

The α-2-l-fucosyltransferase in human plasma has been freed from α-3-l-fucosyltransferase activity and purified approximately 200,000-fold by a series of steps involving ammonium sulphate precipitation, hydrophobic chromatography on Phenyl Sepharose 4B and affinity chromatography first on GDP-adipate-Sepharose and then on GDP-hexanolamine-Sepharose. The purified α-2-l-fucosyltransferase had a Mron gel filtration HPLC of 158,000 and showed optimal activity in the pH range 6.5–7.0. The enzyme transferred fucose equally well to Type 1 (Galβ1-3GlcNAc) and Type 2 (Galβ1-4GlcNAc) substrates but Type 3 (Galβ1-3GalNAc) structures were less efficient acceptors. Competition experiments indicated that a single enzyme species in the purified preparation was responsible for reactivity with the Type 1 and Type 2 structures. Thus the differences in conformation between the Type 1 and Type 2 disaccharides do not appear to influence the capacities of their terminal non-reducing β-d-galactosyl residues to function as acceptor substrates for the α-2-l-fucosyltransferase expressed by the blood groupHgene in haemopoietic tissue.

Abstract

1. The glycoprotein components of a human ovarian-cyst fluid were isolated by a solvent [95% (w/w) phenol]-extraction procedure; the phenol-insoluble water-soluble glycoprotein was further fractionated by (NH4)2SO4 and by ethanol to yield eight fractions. 2. The fractions were analysed in terms of amino acids, fucose, galactose, N-acetylglucosamine, N-acetylgalactosamine and sialic acid. Variations occurred, particularly in the proportion of peptide; these were partly correlated with varying extent of serological activity. 3. The fractions were characterized physicochemically in terms of buoyant density and degree of spreading in a density gradient, sedimentation velocity and molecular weight; their partial specific volumes and specific refraction increments were also determined. 4. The fractions showed wide variations in their sedimentation-velocity and density-gradient patterns, and gave evidence of pauci-dispersity in density. The fraction regarded as the most typical blood-group-specific glycoprotein sedimented as a single rapidly spreading peak and was of high molecular weight. 5. Significant correlations were observed between the physical properties of the glycoprotein fractions and the amount of their peptide component. The buoyant densities and sedimentation coefficients varied in a manner that suggested the existence of two families of glycoproteins. 6. It is suggested that variability in the extent of glycosylation, or in the degree of cross-linking, might account for the two families of glycoproteins, and that the extent of cross-linkage might also be a factor determining the solubility of these glycoproteins in hot saturated (NH4)2SO4.

45. The Schmidt reaction of some alphabeta-unsaturated ketones

Abstract

Benzylideneacetone has been subjected to the Schmidt reaction in sulphuric acid under various conditions, and the only amide isolated was N-methylcinnamamide. Benzylideneacetons p-methoxy-, p-nitro- and m-nitro-benzylideneacetones were subjected to the Schmidt reaction in hydrochloric acid and in polyphosphoric acid. In hydrochloric acid each ketone gave a good yield of the N-methylcinnamamide. In polyphosphoric acid the nitrobenzylideneacetones gave a mixture of the N-styrylacetamide and the N-methylcinnamamide, benzylideneacetone gave mainly N-methylcinnamamide, only a trace of N-styrylacetamide being isolated in some runs of shortened duration, and the N-methylcinnamamide was the only amide isolated from p-methoxybenzyl-ideneacetone. The formation of aniline and p-anisidine during the reactions of benzylideneacetone and p-methoxybenzylidene-acetone in polyphosphoric acid has been explained. A mechanism has been proposed to explain the exclusive production of the N-methylcinnamamides in hydrochloric acid, and the novel variation of the amide ratio with solvent observed in the reactions with the nitrobenzylideneacetones. The Beckmann rearrangements of the alpha-oximes of benzylideneacetone, p-nitro- and m-nitro-benzylideneacetones have been re-examined. Methyl vinyl ketone, methyl isopropenyl ketone and mesityl oxide were subjected to Schmidt reaction conditions in hydrochloric acid. Ammonia was isolated from each reaction and, in addition methyl vinyl ketone gave diacetyl, methyl isopropenyl ketone gave methyl ethyl ketone, and mesityl oxide gave methylamine and acetone. Addition of hydrazoic acid to the olefinic bond of the ketones yielded azidoketones, decomposition of which in acids gave ammonia in each case, diacetyl and ethylenediamine from the azide of methyl vinyl ketone, shown to be 1-azidobutan-3-one, and methylamine from the azide of mesityl oxide. A mechanism for the decomposition of 1-azidobutan-3-one has been proposed.

A method is given by which a-differential equation with initial conditions can be converted into an integral equation. This procedure is used to derive the Multiplication Theorems for Bessel functions, and to obtain an expansion of the confluent hyper geometric function in terms, of Bessel functions. The method is adapted to find approximate eigenvalues: and eigenfunctions of bounded quantum mechanical problems, and to obtain an approximate solution of a non-linear differential equation.
