After two years of experience in overseeing groups of undergraduate and graduate criminal justice students in an Innocence Case Review course at the University of Central Missouri (UCM), the authors reviewed the structure of the course and the attitudes of the participating students. An examination of Innocence Case Review current procedures in undergraduate institutions and law schools provides background for course design. This article summarizes the findings of the students’ experiences, reactions, and attitudes of their yearlong case screening efforts, and to the criminal justice system in general. The first part of this article examines the literature on innocence case review classes at both undergraduate institutions and law schools exploring different learning styles and strategies inherent in this type of class. Two primary entities of the UN regime address the challenges of global climate change (GCC), the UN Framework Convention on Climate Change (UNFCCC) and the UN Security Council (UNSC). This chapter provides an overview of the efforts undertaken pursuant to the UNFCCC for developing the GCC strategies of mitigation, adaptation, and capacity building. The 2015 Paris Agreement provided much needed reinvigoration of the multilateral approach for addressing GCC. A continued lack of unanimity in the international community on the urgency to take concerted action presents significant challenges to the promise of the 2015 Paris Agreement. The UNSC’s perspective on dealing with threats to international peace and security has been expanding in recent years; however, in the formal sessions undertaken on the explicit issue of GCC, there was pronounced opposition, by developing nations along with China and Russia, for the UNSC to identify explicitly GCC as such a threat. This chapter examines the expanding role for the UNSC addressing threats to international peace and security where formally securitized issues are encompassing discrete challenges that could arise from GCC.

In Iraq and Syria, as well as in neighbouring states, the rise of ISIL has raised serious state and human security challenges. This study explores the relationships between the Global-Regional Partnership, the United Nations and nine organizations in their attempt to deal with the challenges presented by ISIL. Each organization is analyzed in terms of how it has responded in the past and how it is now responding to the ISIL threat based on three perspectives; resource capacities (military, political, economic, technological, normative); willingness and readiness; and impediments to capacity and abilities. The overall aim is to discern what capacities and abilities international organizations have to protect state and human security and prevent civilians from mass atrocities inflicted by ISIL forces. The study addresses the role of international organizations when the UNSC is unable or unwilling to uphold the most fundamental norms and values in the UN Charter. This approach acknowledges that within the international community there is an overall acceptance on security for a partnership between the UN and regional organizations, but that there is also a contested call for a renegotiated international contract on state and human security. In this article we present a theory of the prevalence of female-female non-partner violence in the African-American inner city. We employ `ghetto' (economic) and `internal colony' (political) models to illuminate dimensions of the African-American inner city which we view as proximal determinants of female-female interpersonal violence. These cultural and structural elements include conflict within the separate female-gendered cultural milieu centered in the female-headed household system, and the `rule of force' imposed by agencies of social control as the cultural model for dominance, oppression, and interpersonal violence. Verbatim accounts provided by respondents injured in female-female violent interactions are offered as illustrations of the theory.

The pathogenesis of human coronary artery disease (CAD) is likely to require the transcription of many different genes. We report here the differential gene expression profiling of human CAD using copy DNA (cDNA)/nylon array hybridization techniques.Human coronary arteries were obtained at the time of cardiac transplantation. Ten patients were transplanted for ischemic heart disease (IHD) and 5 for dilated cardiomyopathy (DCM). We generated a customized cDNA array containing 9206 clones and after hybridization of patient samples, data reduction, and refinement, identified 515 sequence-verified, differentially expressed clones. These clones represented 361 genes that were differentially expressed at significant levels between IHD and DCM arteries (t test, P.05). Of these clones, 70% were defined genes of known function and 30% were genes of unknown function. Of the differentially expressed genes, 53.6% were up-regulated and 46.4% were down-regulated. Hierarchical clustering was performed and several distinct functional clusters were identified, including a cluster of genes related to inflammatory mechanisms. Validation by real-time polymerase chain reaction was undertaken with 2 genes known to be up-regulated in atherosclerosis (interleukin 1beta [IL-1beta] and IL-8) and 2 novel genes identified by the array analysis (signal transducer and activator of transcription 6 [STAT6] and IL-1 receptor-associated kinase [IRAK]). Differential expression of IL-1beta, IL-8, and STAT6 were confirmed by this method. Immunohistochemistry of STAT6 demonstrated increased expression in vascular smooth muscle cells of IHD coronary arteries.These data support the inflammatory basis of human atherosclerotic CAD and identify novel genes in atherosclerosis.

Helicobacter pyloristrains containing thecagpathogenicity island (PAI) induce NF-κB activation and interleukin-8 secretion in gastric epithelial cells. The aim of this study was to investigate changes in epithelial gene expression induced bycagPAI-positive and -negative strains ofH. pyloriusing high-density cDNA array hybridization technology. Radio-labeled cDNA prepared fromH. pylori-infected Kato 3 gastric epithelial cells was hybridized to high-density cDNA arrays to identify changes in epithelial gene expression compared to noninfected controls. In vivo expression of selected, differentially expressed genes was examined by reverse transcription-PCR analysis ofH. pylori-positive and -negative gastric mucosa. Screening of ca. 57,800 cDNAs identified 208 known genes and 48 novel genes and/or expressed sequence tags of unknown function to be differentially expressed in Kato 3 cells followingH. pyloriinfection. Marked differences in gene expression profiles were observed followingcagPAI-positive andcagPAI-negative infection with 15 novel cDNAs and 92 known genes being differentially expressed.H. pyloriwas found to change the expression of genes encoding growth factors and cytokine/chemokines and their receptors, apoptosis proteins, transcription factors and metalloprotease-disintegrin proteins (ADAMs), and tissue inhibitors of metalloproteinases. Gastric differential expression of selected known genes (amphiregulin and ADAM 10) and a novel gene (HPYR1) was confirmed in vivo in patients withH. pyloriinfection. Confirmation of the in vivo expression of selected genes demonstrates the usefulness of this approach for investigating pathogen-induced changes in host gene expression.

BACKGROUND: The majority of patients with tuberculosis who comply with appropriate treatment are cured. However, approximately 5% subsequently have a repeat disease episode, usually within 2 years of successful combination therapy. Presently, there is no way of predicting which patients will experience a relapse. METHODS: We identified 10 subjects who had previously experienced recurrent tuberculosis and carefully matched them to cured subjects who had had only 1 episode of tuberculosis, to patients with active tuberculosis, and to latently infected healthy subjects. We compared their ex vivo whole-blood gene-expression profiles by use of DNA array technology and confirmed the results by use of quantitative reverse-transcriptase polymerase chain reaction (qRT-PCR). RESULTS: The 4 clinical tuberculosis groups exhibited distinct patterns of gene expression. The gene-transcript profiles of the patients with recurrent tuberculosis were more similar to those of the patients with active tuberculosis than to those of the cured or latently infected subjects. Discriminant analysis of a training data set showed that 9 genes were sufficient to classify the subjects. We confirmed that measurement of the expression of these genes by qRT-PCR can accurately discriminate between subjects in a test set of samples. CONCLUSIONS: A simple test based on gene-expression patterns may be used as a biomarker of cure while identifying patients who are at risk for relapse. This would facilitate the introduction of new tuberculosis drugs.

SIMSTAT for Windows is a full-featured statistical analysis package, as is its DOS ancestor previously reviewed in these pages (Wallace, 1994), upgraded to include a number of enhancements to the working environment. An important feature of the SIMSTAT software "family" is the bootstrap (resampling) module (hence the "SIM," denoting "simu lation," in the program's name), which offers an effective teaching tool for inferential statistics. Including all add-on modules, SIMSTAT for Windows is a cost-effective competi tor with the $695 institutional version of SPSS for Windows or similar statistical analysis software.

The analysis of gene changes associated with exposure to cannabinoids is critical due to the multiple possible signaling pathways potentially affected by cannabinoid receptor activation. A comparison of altered gene profiles under two different conditions, one in vivo (chronic exposure to delta-9-THC) and the other in vitro (neuroprotection mediated by WIN55212-2), was made to determine whether it was possible to identify common genes that were affected. Up and down-regulated sets of genes are described. Genes affected in one or the other circumstance include alterations in a 14-3-3 regulator protein of PKC, CREB, BDNF and GABA receptor subunit proteins, as well as several genes associated with known cannabinoid receptor-coupled signaling pathways. Unexpectedly, several genes that were altered in both circumstances were associated with synaptic and membrane structure, motility and neuron growth. These included, neuronal cell adhesion molecule (NCAM), hyloronidan motility receptor, and myelin proteolipid protein. While the basis for involvement of these particular genes in cannabinoid receptor activated functional processes within the cell is still not well understood, awareness that significant numbers of genes and presumably proteins are changed following either acute or long-term exposure may provide new insight into their effects.

Large-scale analysis of gene expression changes during acute and chronic exposure to Δ9-THC in rats. Physiol Genomics 3: 175–185, 2000.—Large-scale cDNA microarrays were employed to assess transient changes in gene expression levels following acute and chronic exposure to cannabinoids in rats. A total of 24,456 cDNA clones were randomly selected from a rat brain cDNA library, amplified by PCR, and arrayed at high density to investigate differential gene expression profiles following acute (24 h), intermediate (7 days), and chronic (21 days) exposure to Δ9-tetrahydrocannabinol (Δ9-THC), the psychoactive ingredient of marijuana. Hippocampal mRNA probes labeled with 33P obtained from both vehicle and Δ9-THC-treated animals were hybridized with identical cDNA microarrays. Results revealed a total of 49 different genes altered by Δ9-THC exposure; of these, 28 were identified, 10 had homologies to expressed sequence tags (ESTs), and 11 had no homology to known sequences in the GenBank database. Chronic or acute cannabinoid receptor activation altered expression of several genes (i.e., prostaglandin D synthase, calmodulin) involved in biochemical cascades of cannabinoid synthesis or cannabinoid effector systems. Other genes [i.e., neural cell adhesion molecule (NCAM), myelin basic protein], whose relation to cannabinoid system function was not immediately obvious, were also significantly altered. Verification of the changes obtained with the large-scale screen was determined by RNA dot blots in different groups of animals treated the same as those in the large-scale screen. Results are discussed in terms of the different types of genes affected at different times during chronic Δ9-THC exposure.

In order to investigate the effect of the Oct-2 POU family transcription factor on the regulation of genes encoding synaptic proteins, we have used cell lines in which the level of Oct-2 has been greatly reduced using an antisense approach. The reduced Oct-2 level results in enhanced expression of SNAP-25 and synapsin I, indicating that the genes encoding these proteins are normally repressed by Oct-2 in neuronal cells. In contrast, no alteration was observed in the levels of the synaptic proteins, synaptophysin and synaptotagmin. Although the neuronal forms of Oct-2 can repress the synapsin I promoter in co-transfection experiments, indicating that they have a direct effect on the expression of this gene, they have no effect on the activity of the SNAP-25 promoter, indicating that the effect of Oct-2 on this gene is likely to be indirect. These effects are discussed in terms of the differential effect of Oct-2 and the related POU family transcription factor Brn-3a, on the promoters of genes encoding different synaptic proteins.

Identification of helicobacter pylori epithelial cell response genes by screening high-density cDNA arrays
