Functional articular cartilage regeneration remains an unmet medical challenge, increasing the interest for innovative biomaterial-based tissue engineering (TE) strategies. Hydrogels, 3D macromolecular networks with hydrophilic groups, present articular cartilage-like features such as high water content and load-bearing capacity. In this study, 3D porous polyethylene glycol diacrylate (PEGDA) hydrogels were fabricated combining the gas foaming technique and a UV-based crosslinking strategy. The 3D porous PEGDA hydrogels were characterized in terms of their physical, structural and mechanical properties. Our results showed that the size of the hydrogel pores can be modulated by varying the initiator concentration. In vitro cytotoxicity tests showed that 3D porous PEGDA hydrogels presented high biocompatibility both with human chondrocytes and osteoblast-like cells. Importantly, the 3D porous PEGDA hydrogels supported the viability and chondrogenic differentiation of human bone marrow-derived mesenchymal stem/stromal cell (hBM-MSC)-based spheroids as demonstrated by the positive staining of typical cartilage extracellular matrix (ECM) (glycosaminoglycans (GAGs)) and upregulation of chondrogenesis marker genes. Overall, the produced 3D porous PEGDA hydrogels presented cartilage-like mechanical properties and supported MSC spheroid chondrogenesis, highlighting their potential as suitable scaffolds for cartilage TE or disease modelling strategies.

This work proposes a 3D-printed sensor based on fiber Bragg grating (FBG) technology for respiratory rate (RR) and heart rate (HR) monitoring. Each sensor is composed of a single FBG fully encapsulated into a 3D-printable Flexible , during the printing process. Sensors with different material thicknesses and infill densities were tested. The sensor with the best metrological properties was selected and preliminary assessed in terms of capability of monitoring RR and HR on three users. This work proposes an optical sensing network to monitor pressure and temperature in specific areas of a wheelchair to prevent pressure ulcers and to monitor the position of the wheelchair user by analyzing its pressure distribution. The sensing network is composed of six optical fiber Bragg grating (FBG)-based sensor cells. Each sensor cell is built from a polylactic acid (PLA) base and has two FBGs, one embedded in epoxy resin to monitor pressure variations (FBG P ) and another without resin to monitor temperature (FBG T ). Once produced, all sensor cells were experimentally characterized for pressure and temperature variations, resulting in an average pressure sensitivity of 81 ± 5 pm/kPa (FBG P ) and -5.0 ± 0.4 pm/kPa (FBG T ), and an average temperature sensitivity of 25 ± 1 pm/°C (FBG P ) and 47.7 ± 0.7 pm/°C (FBG T ). The sensor cells were then placed in six specific areas of a wheelchair (four in the seat area and two in the shoulder blade area) to carry out experimental tests, wherein the response of the sensors to a specific sequence of relief positions was tested. During the execution of the test, the optical signal of all sensors was monitored, in real time, with the pressure and temperature values detected in each zone of the wheelchair. In addition, random position changes were performed in order to evaluate the precision of the proposed sensing network in the identification of such positions.

Developing technologies capable of constantly assessing and optimizing day-to-day activities has been a research priority for several years. A key factor in such technologies is the use of highly sensitive sensors to monitor in real-time numerous parameters, such as temperature and load. Due to their unique features, optical fiber sensors became one of the most interesting and viable solutions for applications dependent on those parameters. In this work, we present an optical fiber load sensor, called load cell, based on Fabry-Pérot hollow cavities embedded in a polymeric material. By using the load cells in a parallel configuration with a non-embedded hollow cavity, the optical Vernier effect was generated, allowing maximum sensitivity values of 0.433 nm N -1 and 0.66 nm °C -1 to be attained for vertical load and temperature, respectively. The proposed sensor's performance, allied with the proposed configuration, makes it a viable and suitable device for a wide range of applications, namely those requiring high thermal and load sensitivities.

The bifunctional enzyme soluble epoxide hydrolase (sEH) is found in all regions of the brain. It has two different catalytic activities, each assigned to one of its terminal domains: the C-terminal domain presents hydrolase activity, whereas the N-terminal domain exhibits phosphatase activity. The enzyme's C-terminal domain has been linked to cardiovascular protective and anti-inflammatory effects. Cholesterol-related disorders have been associated with sEH, which plays an important role in the metabolism of cholesterol precursors. The role of sEH's phosphatase activity has been so far poorly investigated in the context of the central nervous system physiology. Given that brain cholesterol disturbances play a role in the onset of Alzheimer's disease (AD) as well as of other neurodegenerative diseases, understanding the functions of this enzyme could provide pivotal information on the pathophysiology of these conditions. Moreover, the sEH phosphatase domain could represent an underexplored target for drug design and therapeutic strategies to improve symptoms related to neurodegenerative diseases. This review discusses the function of sEH in mammals and its protein structure and catalytic activities. Particular attention was given to the distribution and expression of sEH in the human brain, deepening into the enzyme's phosphatase activity and its participation in brain cholesterol synthesis. Introduction: Cervical cancer screening is an important tool in public health. Liquid-based cytology (LBC) has been performed at the studied hospital for 7 years. The present study compares the performance of 2 LBC techniques with conventional cytology., Objective: Our objective is to verify the sensitivity for the detection of neoplastic and preneoplastic epithelial atypia, as well as the positive predictive value of the 3 methodologies., Methods: We analyzed retrospectively 24,529 cases and evaluated the conventional cytology, ThinPrep®, and BD SurePath® performance categorizing the results according to the Bethesda system. We also compared the level of unsatisfactory samples, the presence of elements from the squamocolumnar junction, and the detection of pathogenic microorganisms., Results: ThinPrep® (1.43%) showed superior sensitivity over BD SurePath® (0.91%) and conventional cytology (0.71%) in terms of the detection of high-grade lesions; however, in terms of squamous atypia as a whole (ASC-US+), BD SurePath® (6.44%) proved to be more sensitive than conventional cytology (5.28%) and ThinPrep® (3.73%)., Conclusions: The results show the advantage of implementing LBC in routine screening for cervical lesions. In this work, a fiber Bragg grating (FBG) based sensing system for wheelchair pressure ulcer prevention was developed. Six FBGs were strategically positioned in a wheelchair to monitor the more prominent bone areas, namely scapulas (right (SR) and left (SL)), ischiatic zone (right (IR) and left (IL)), and heels (right (HR) and left (HL)). The sensing architecture was tested by a female user during pressure relief exercises, to verify its effectiveness on pressure monitoring. The proposed system proves to be a compact and reliable solution for wheelchair pressure ulcer prevention, making it a suitable alternative to existing conventional electronic sensors, with the advantage of being immune to electromagnetic interferences and usable in humid environments. In addition to the pressure, the breathing rate was also monitored. By combining the proposed sensing architecture with a wheelchair user detection software, it is possible to create alerts for the user to know when a new position should be adopted, in order to relieve the pressure in a specific area, thus avoiding one of the biggest problems for such patients, pressure ulcers.

Fabry-Perot interferometric (FPI) sensors are an accurate and well-established sensing technology that are used to monitor a wide range of parameters such as strain, temperature, and refractive index, among many others. Nevertheless, due to the limited number and high cost of existing interrogation techniques for FPIs, its use is often restricted to discrete measurements, not being so explored for dynamic applications. The development of an alternative interrogation technique for a high rate of acquisition may propel this type of sensor into less explored fields such as dynamic biomedical applications. In this work, we present the theoretical and experimental analyses of an FPI sensing architecture by using an alternative high rate dynamic acquisition methodology, based on frequency to amplitude conversion, where the FPI spectral shift is detuned by the convolution of the optical light source with the FPI interference pattern. The good agreement between the theoretical and experimental results verified the reliability of the proposed methodology. The purpose of this study was to compare the linear polymerization shrinkage of different restorative resin-based composites (RBCs) using fiber Bragg grating (FBG) sensors. Five RBCs were evaluated: Zirconfill ® (ZFL); Aura Bulk-Fill (ABF); Tetric ® N-Ceram Bulk-Fill (TBF); Filtek TM Bulk-Fill (FBF); and Admira Fusion-Ormocer ® (ADF). Ten samples per resin were produced in standardized custom-made half-gutter silicone molds. Two optical FBG sensors were used to assess temperature and polymerization shrinkage. Light curing was performed for 40 s and polymerization shrinkage was evaluated at 5, 10, 40, 60, 150, and 300 s. Statistical analysis was accomplished for normal distribution (Shapiro-Wilk, p > 0.05). Two-way repeated measures ANOVA with Greenhouse-Geisser correction followed by Bonferroni's post-hoc test was used to analyze the linear shrinkage data ( p < 0.05). ZFL showed the highest linear shrinkage and ADF the lowest. Shrinkage increased for all RBCs until 300 s, where significant differences were found between ADF and all other resins ( p < 0.05). Among bulk-fill RBCs, TBF showed the lowest shrinkage value, but not statistically different from FBF. The ADF presented lower linear shrinkage than all other RBCs, and restorative bulk-fill composites exhibited an intermediate behavior.

Magnetic field sensing is an important issue for many application areas, such as in the military, industry and navigation. The current sensors used to monitor this parameter can be susceptible to electromagnetic interferences, however due to their advantages over the traditional sensors, the optical fiber devices could be an excellent alternative. Furthermore, magnetic fluid (MF) is a new type of functional material which possesses outstanding properties, including Faraday effect, birefringence, tunable refractive index and field dependent transmission. In this paper, the optical fiber magnetic field sensors using MF as sensing element are reviewed. Due to the extensive literature, only the most used sensing configurations are addressed and discussed, which include optical fiber grating, interferometry, surface plasmon resonance (SPR) and other schemes involving tailored (etched, tapered and U-shaped) fibers.

This paper presents the characterization of polymer optical fibers (POFs) submitted to the catastrophic fuse effect towards intensity-variation-based sensing of strain, transverse force, temperature, and moisture. In the experiments, POFs with and without the fuse effect are tested and the results are compared with respect to the sensitivity, linearity, and root mean squared error (RMSE). The fused POFs have higher linearity and lower RMSE than non-fused POFs in strain and transverse force sensing. Also, the sensitivity of the fused POFs is higher in transverse force and temperature sensing, which can be related to the higher sensitivity to the curvature that the transverse force creates on the POF and to the more significant variations of the refractive index with temperature increase. Additionally, the fused POFs present lower moisture absorption than the non-fused POFs. The presented results indicate a great potential of the fused POFs intensity-variation-based sensing applications of various physical parameters.

In an era of unprecedented progress in sensing technology and communication, health services are now able to closely monitor patients and elderly citizens without jeopardizing their daily routines through health applications on their mobile devices in what is known as e-Health. Within this field, we propose an optical fiber sensor (OFS) based system for the simultaneous monitoring of shear and plantar pressure during gait movement. These parameters are considered to be two key factors in gait analysis that can help in the early diagnosis of multiple anomalies, such as diabetic foot ulcerations or in physical rehabilitation scenarios. The proposed solution is a biaxial OFS based on two in-line fiber Bragg gratings (FBGs), which were inscribed in the same optical fiber and placed individually in two adjacent cavities, forming a small sensing cell. Such design presents a more compact and resilient solution with higher accuracy when compared to the existing electronic systems. The implementation of the proposed elements into an insole is also described, showcasing the compactness of the sensing cells, which can easily be integrated into a non-invasive mobile e-Health solution for continuous remote gait monitoring of patients and elder citizens. The reported results show that the proposed system outperforms existing solutions, in the sense that it is able to dynamically discriminate shear and plantar pressure during gait.

This Letter presents, for the first time, to the best of our knowledge, the dynamic mechanical analysis of a polymer optical fiber (POF) that was previously damaged by the catastrophic fuse effect. The variation of the fiber Young's modulus was evaluated with respect to the increase of temperature, humidity, and frequency of strain cycles. The obtained data for the fused POF are compared with the ones for the same POF without the fuse effect. The results show the feasibility of the fused POF for sensor applications, such as strain and acceleration measurement, since it presents temperature sensitivity almost two times lower in temperatures between 26°C and 90°C and Young's modulus 2.3 times lower than those obtained with the bare fiber. The Young's modulus variation with the humidity is 1.5 MPa/%RH in a humidity range of 66-96%. In addition, the fused POF presented a variation of its dynamic modulus with the frequency increase four times lower than non-fused POFs on the range of 0.01-100.00 Hz. These results pave the way for future applications of fused POFs as sensing elements.

Peptide YY (PYY) belongs to the neuropeptide Y (NPY) family, which also includes the pancreatic polypeptide (PP) and NPY. PYY is secreted by the intestinal L cells, being present in the blood stream in two active forms capable of crossing the blood brain barrier, PYY (1-36) and its cleavage product, PYY (3-36). PYY is a selective agonist for the Y2 receptor (Y2R) and these receptors are abundant in the hippocampus. Here we investigated the mechanisms by which PYY (3-36) regulates intracellular Ca 2+ concentrations ([Ca 2+ ] i ) in hippocampal neurons by employing a calcium imaging technique in hippocampal cultures. Alterations in [Ca 2+ ] i were detected by changes in the Fluo-4 AM reagent emission. PYY (3-36) significantly increased [Ca 2+ ] from the concentration of 10 -11 M as compared to the controls (infusion of HEPES-buffered solution (HBS) solution alone). The PYY (3-36)-increase in [Ca 2+ ] i remained unchanged even in Ca 2+ -free extracellular solutions. Sarcoplasmic/endoplasmic reticulum Ca 2+ -ATPase pump (SERCA pump) inhibition partially prevent the PYY (3-36)-increase of [Ca 2+ ] i and inositol 1,4,5-triphosphate receptor (IP3R) inhibition also decreased the PYY (3-36)-increase of [Ca 2+ ] i . Taken together, our data strongly suggest that PYY (3-36) mobilizes calcium from the neuronal endoplasmic reticulum (ER) stores towards the cytoplasm. Next, we showed that PYY (3-36) inhibited high K + -induced increases of [Ca 2+ ] i , suggesting that PYY (3-36) could also act by activating G-protein coupled inwardly rectifying potassium K + channels. Finally, the co-infusion of the Y2 receptor (Y2R) antagonist BIIE0246 with PYY (3-36) abolished the [Ca 2+ ] i increase induced by the peptide, suggesting that PYY (3-36)-induced [Ca 2+ ] i increase in hippocampal neurons occurs via Y2Rs., (Copyright © 2017 Elsevier Ltd. We propose a novel polymer optical fiber (POF) sensing system based on fiber Bragg gratings (FBGs) to measure foot plantar pressure. The plantar pressure signals are detected by five FBGs, in the same piece of cyclic transparent optical polymer (CYTOP) fiber, which are embedded in a cork insole for the dynamic monitoring of gait. The calibration and measurements performed with the suggested system are presented, and the results obtained demonstrate the accuracy and reliability of the sensing platform to monitor the foot plantar pressure distribution during gait motion and the application of pressure. This architecture does not compromise the patient's mobility nor interfere in their daily activities. The results using the CYTOP fiber showed a very good response when compared with solutions using silica optical fibers, resulting in a sensitivity almost twice as high, with excellent repeatability and ease of handling. The advantages of POF (e.g., high flexibility and robustness) proved that this is a viable solution for this type of application, since POF's high fracture toughness enables its application in monitoring patients with higher body mass compared with similar systems based on silica fiber. Fiber Bragg gratings are widely used optical fiber sensors for measuring temperature and/or mechanical strain. Nevertheless, the high cost of the interrogation systems is the most important drawback for their large commercial application. In this work, an in-line Fabry-Perot interferometer based edge filter is explored in the interrogation of fiber Bragg grating dynamic measurements up to 5 kHz. Two devices an accelerometer and an arterial pulse wave probe were interrogated with the developed approach and the results were compared with a commercial interrogation monitor. The data obtained with the edge filter are in agreement with the commercial device, with a maximum RMSE of 0.05 being able to meet the requirements of the measurements. In an era of unprecedented progress in technology and increase in population age, continuous and close monitoring of elder citizens and patients is becoming more of a necessity than a luxury. Contributing toward this field and enhancing the life quality of elder citizens and patients with disabilities, this work presents the design and implementation of a noninvasive platform and insole fiber Bragg grating sensors network to monitor the vertical ground reaction forces distribution induced in the foot plantar surface during gait and body center of mass displacements. The acquired measurements are a reliable indication of the accuracy and consistency of the proposed solution in monitoring and mapping the vertical forces active on the foot plantar sole, with a sensitivity up to 11.06 ?? pm / N . The acquired measurements can be used to infer the foot structure and health condition, in addition to anomalies related to spine function and other pathologies (e.g., related to diabetes); also its application in rehabilitation robotics field can dramatically reduce the computational burden of exoskeletons’ control strategy. The proposed technology has the advantages of optical fiber sensing (robustness, noninvasiveness, accuracy, and electromagnetic insensitivity) to surpass all drawbacks verified in traditionally used sensing systems (fragility, instability, and inconsistent feedback).

It is well recognized that stress or glucocorticoids hormones treatment can modulate memory performance in both directions, either impairing or enhancing it. Despite the high number of studies aiming at explaining the effects of glucocorticoids on memory, this has not yet been completely elucidated. Here, we demonstrate that a low daily dose of methylprednisolone (MP, 5mg/kg, i.p.) administered for 10-days favors aversive memory persistence in adult rats, without any effect on the exploring behavior, locomotor activity, anxiety levels and pain perception. Enhanced performance on the inhibitory avoidance task was correlated with long-term potentiation (LTP), a phenomenon that was strengthen in hippocampal slices of rats injected with MP (5mg/kg) during 10days. Additionally, in vitro incubation with MP (30-300µM) concentration-dependently increased intracellular [Ca 2+ ] i in cultured hippocampal neurons depolarized by KCl (35mM). Animal venoms have been widely recognized as a major source of biologically active molecules. Bothriurus bonariensis, popularly known as black scorpion, is the arthropod responsible for the highest number of accidents involving scorpion sting in Southern Brazil. Here we reported the first attempt to investigate the neurobiology of B. bonariensis venom (BBV) in the insect and mammalian nervous system. BBV (32 μg/g) induced a slow neuromuscular blockade in the in vivo cockroach nerve-muscle preparations (70 ± 4%, n = 6, p < 0.001), provoking repetitive twitches and significantly decreasing the frequency of spontaneous leg action potentials (SNCAPs) from 82 ± 3 min(-1) to 36 ± 1.3 min(-1) (n = 6, p < 0.05), without affecting the amplitude. When tested in primary cultures of rat hippocampal cells, BBV induced a massive increase of Ca(2+) influx (250 ± 1% peak increase, n = 3, p < 0.0001). The disturbance of calcium homeostasis induced by BBV on the mammalian central nervous system was not accompanied by cellular death and was prevented by the co-treatment of the hippocampal cells with tetrodotoxin, a selective sodium channel blocker. The results suggest that the biological activity of BBV is mostly related to a modulation of sodium channels function. The integration of fiber Bragg grating (FBG) sensors in lithium-ion cells for in-situ and in-operando temperature monitoring is presented herein. The measuring of internal and external temperature variations was performed through four FBG sensors during galvanostatic cycling at C-rates ranging from 1C to 8C. The FBG sensors were placed both outside and inside the cell, located in the center of the electrochemically active area and at the tab-electrode connection. The internal sensors recorded temperature variations of 4.0 ± 0.1 °C at 5C and 4.7 ± 0.1 °C at 8C at the center of the active area, and 3.9 ± 0.1 °C at 5C and 4.0 ± 0.1 °C at 8C at the tab-electrode connection, respectively. This study is intended to contribute to detection of a temperature gradient in real time inside a cell, which can determine possible damage in the battery performance when it operates under normal and abnormal operating conditions, as well as to demonstrate the technical feasibility of the integration of in-operando microsensors inside Li-ion cells., Competing Interests: The authors declare no conflict of interest.

Human umbilical cord blood is an attractive source of stem cells; however, it has a heterogeneous cell population with few mesenchymal stem cells. Cell reprogramming induced by different methodologies can confer pluripotency to differentiated adult cells. The objective of this study was to evaluate the reprogramming of fibroblasts and their subsequent neural differentiation after co-culture with umbilical cord blood mononuclear cells. Cells were obtained from four human umbilical cords. The mononuclear cells were cultured for 7 d and subsequently co-cultured with mouse fibroblast NIH-3T3 cells for 6 d. The pluripotency of the cells was evaluated by RT-PCR using primers specific for pluripotency marker genes. The pluripotency was also confirmed by adipogenic and osteogenic differentiation. Neural differentiation of the reprogrammed cells was evaluated by immunofluorescence. All co-cultured cells showed adipogenic and osteogenic differentiation capacity. After co-cultivation, cells expressed the pluripotency gene KLF4. Statistically significant differences in cell area, diameter, optical density, and fractal dimension were observed by confocal microscopy in the neurally differentiated cells. Contact in the form of co-cultivation of fibroblasts with umbilical cord blood mononuclear fraction for 6 d promoted the reprogramming of these cells, allowing the later induction of neural differentiation.

Background: In general, surgeries currently tend to be less invasive and cardiac surgery has started to follow this trend., Objective: To evaluate the evolution of one hundred patients undergoing minimally-invasive coronary artery bypass grafting., Methods: Access to the heart was attained through a small; 6-cm thoracotomy, located in the 4th left intercostal space, starting at the nipple. Through the same intercostal space, 3 cm after the primary incision, a 6.5-mm optical device was inserted at 30º. Where the saphenous vein was used, the pericardium was opened above the aorta and the latter was partially clamped with a systolic pressure of 80 mmHg, with the proximal anastomosis being carried out in the conventional manner. The distal anastomoses were carried out in the conventional manner. The procedure was performed off-pump using single lung ventilation., Results: The mean age was 63.9 ± 10.66 years. Sixty-eight (68%) patients were males. Fifty-three (53%) were in functional class III or IV. Left ventricular function was normal in fifty-three (53%) patients. Forty-two (42%) had undergone previous angioplasty. A total of 153 anastomoses were performed, ranging from 1 to 3. The average ventilation time was 4.06 ± 4.08 hours. Seventeen (17%) patients had atrial fibrillation and eight (8%) had pneumonia. There were two deaths in this series., Conclusion: Revascularization was safe with low mortality and morbidity. With the advent of new devices, this surgery may have a greater applicability.

Purpose: To investigate the clinical, perimetric, contrast sensitivity and visual evoked potentials findings in patients on longterm treatment with amiodarone., Methods: We enrolled 14 patients (age range 47-78 years, mean age 62.7 years) on chronic amiodarone therapy (mean cumulative dose of 519 g), and 14 age-matched control subjects. Participants were submitted to ophthalmological examination, kinetic visual field testing (Goldmann), standard achromatic automated perimetry (central 30-2, Humphrey), computerized isoluminant chromatic contrast sensitivity (CCS) and pattern visual evoked potentials evaluation (VEP). Data from baseline, 6-month and 1-year follow-up examinations were analysed. The main outcome measures were: initial and final corrected visual acuity; corneal, optic disc and fundus abnormalities; mean deviation (MD) and pattern standard deviation (PSD) central 30-2 perimetry parameters; visual field loss on kinetic perimetry; indices of CCS (tritan, protan and deutan axes), and pattern VEP latency (L-VEP) and VEP amplitude (a-VEP)., Results: We found a statistically significant difference between the patients and controls' baseline L-VEP and a-VEP measurements (p < 0.001). We found no significant correlation between prolonged L-VEP and duration of amiodarone use (r = 0.07). After a 1-year follow-up, we evidenced an average increment in L-VEP of 1.96 ms and a-VEP measurements showed a decline of 0.69 micro V. In cumulative dose-specific analyses, mean differences between baseline and 12-month L-VEP measurements were more evident in those taking higher than 200 g cumulative amiodarone doses (p = 0.03). We found abnormal tritan CCS results in 19 eyes (68%). Of these, four eyes belonged to patients with no ocular disease (diabetes, cataracts). No significant differences were observed between the baseline and 12-month clinical eye examinations, central 30-2 perimetry and CCS results., Conclusion: We found a significant prolongation in pattern L-VEP and a significant reduction in pattern a-VEP in patients on longterm treatment with amiodarone compared to control subjects. Nevertheless, current data are insufficient to recommend appropriate methods of visual screening.
