Abstract: 16 pages; International audience; tUnderstanding differences in phenology for varieties of a given species is important for making informeddecisions on variety choice in the context of climate change and breeding new cultivars. Phenologicalmodels that incorporate temperature as a key environmental factor can be used to describe the differencesin phenological timing between cultivars. This paper outlines a methodology, based on a phenologicalmodel, for classifying the timing of flowering and veraison for a substantial number of varieties of thegrapevine (Vitis vinifera L.). 95 varieties were characterized for flowering and 104 varieties for veraison.Various statistical measures were used to assess the performance and predictions of the model at thevarietal level: model efficiency, root mean squared error and confidence intervals. The methodologymight be used to understand varietal differences for other species in future studies. Model outputs canbe used in combination with predicted climate change scenarios to assess the suitability of varietiesunder climate conditions of the future.

Abstract: International audience; Background and purpose - A clinical risk score for sudden unexpected death in epilepsy (SUDEP) in patients with drug-resistant focal epilepsy could help improve prevention. Methods - A case-control study was conducted including (i) definite or probable SUDEP cases collected by the French National Sentinel Mortality Epilepsy Network and (ii) control patients from the French national research database of epilepsy monitoring units. Patients with drug-resistant focal epilepsy were eligible. Multiple logistic regressions were performed. After sensitivity analysis and internal validation, a simplified risk score was developed from the selected variables. Results - Sixty-two SUDEP cases and 620 controls were included. Abstract: Background and Aims Condensed tannins, responsible for berry and wine astringency, may have been selected during grapevine domestication. This work examines the phylogenetic distribution of condensed tannins throughout the Vitaceae phylogenetic tree. Methods Green berries and mature leaves of representative true-to-type members of the Vitaceae were collected before ‘véraison’, freeze-dried and pulverized, and condensed tannins were measured following depolymerization by nucleophilic addition of 2-mercaptoethanol to the C4 of the flavan-3-ol units in an organic acidic medium. Reaction products were separated and quantified by ultrahigh pressure liquid chromatography/diode array detection/mass spectrometry. Key Results and Conclusions The original ability to incorporate epigallocatechin (EGC) into grapevine condensed tannins was lost independently in both the American and Eurasian/Asian branches of the Vitaceae, with exceptional cases of reversion to the ancestral EGC phenotype. This is particularly true in the genus Vitis, where we now find two radically distinct groups differing with respect to EGC content. While Vitis species from Asia are void of EGC, 50 % of the New World Vitis harbour EGC. Interestingly, the presence of EGC is tightly coupled with the degree of leaf margin serration. Noticeably, the rare Asian EGC-forming species are phylogenetically close to Vitis vinifera, the only remnant representative of Vitis in Eurasia. Both the wild ancestral V. vinifera subsp. sylvestris as well as the domesticated V. vinifera subsp. sativa can accumulate EGC and activate galloylation biosynthesis that compete for photoassimilates and reductive power.

Abstract: The pip, as the most common grapevine archaeological remain, is extensively used to document past viticulture dynamics. This paper uses state of the art morphological analyses to analyse the largest reference collection of modern pips to date, representative of the present-day diversity of the domesticated grapevine from Western Eurasia. We tested for a costructure between the form of the modern pips and the: destination use (table/wine), geographical origins, and populational labels obtained through two molecular approaches. Significant structuring is demonstrated for each of these cofactors and for the first time it is possible to infer properties of varieties without going through the parallel with modern varieties. These results provide a unique tool that can be applied to archaeological pips in order to reconstruct the spatio-temporal dynamics of grape diversity on a large scale and to better understand viticulture history. The models obtained were then used to infer the affiliations with archaeobotanical remains recovered in Mas de Vignoles XIV (Nîmes, France). The results show a twofold shift between the Late Iron Age and the Middle Ages, from table to wine grape varieties and from eastern to western origins which correlates with previous palaeogenomic results.

Abstract: La virose du court-noué, qui s’est étendue à la faveur de la reconstitution post-phylloxérique, constitue aujourd’hui une préoccupation majeure des vignobles mondiaux. La lutte directe s’avérant hors de portée à l‘heure actuelle, concernant une maladie incurable dont les vecteurs se maintiennent et restent infectieux dans le sol durant de très longues périodes, les efforts de la recherche se concentrent sur plusieurs pistes, parmi lesquelles la voie génétique semble la plus prometteuse et la plus avancée. Si la caractérisation de résistances et tolérances au sein du genre Vitis est exploitée en vue de l’obtention de porte-greffes retardant ou limitant la contamination, les différences d’expressions symptomatiques au sein des variétés de Vitis vinifera ont été très peu abordées jusqu’à présent. Dans cet article, basé sur des observations de terrain et des notations en collection, nous proposons une liste de cépages traditionnels de diverses origines peu ou très peu impactés par la maladie, dans le but de désigner du matériel d’étude pour de futurs travaux.

Abstract: International audience; Relevant aspect of wine geographic origin is related to grapevine varieties used for wine production in specific wine country or region. The grapevine germplasm is highly variable and classified into geographic groups. These classifications were recently confirmed using genetic studies, and further classified varieties into geographicgenetic (GEN-GEO) groups. Polyphenolic compounds are important secondary metabolites in grapes and have an important impact on wine quality and sensory properties. The aim of this research was to investigate the polyphenolic profiles of 50 grape varieties from different GEN-GEO groups. The groups were C2 (varieties from Italy and France), C7 (varieties from Croatia), C8 (varieties from Spain and Portugal). Polyphenolic compounds analysed belonged to the classes of anthocyanins, flavan-3-ols, flavonols, phenolic acids, and stilbenes. The most abundant classes of polyphenols were anthocyanins, followed by flavan-3-ols and flavonols. Using discriminant analysis, the genetic-geographic groups were clearly separated based on their polyphenolic profiles, with all classes of compounds contributing to discrimination.

Abstract: Seedlessness is greatly prized by consumers of fresh grapes. While stenospermocarpic seed abortion determined by the SEED DEVELOPMENT INHIBITOR (SDI) locus is the usual source of seedlessness in commercial grapevine (Vitis vinifera) cultivars, the underlying sdi mutation remains unknown. Here, we undertook an integrative approach to identify the causal mutation. Quantitative genetics and fine-mapping in two ‘Crimson Seedless’-derived F1 mapping populations confirmed the major effect of the SDI locus and delimited the sdi mutation to a 323-kb region on chromosome 18. RNA-sequencing comparing seed traces of seedless and seeds of seeded F1 individuals identified processes triggered during sdi-determined seed abortion, including the activation of salicylic acid-dependent autoimmunity. The RNA-sequencing data set was investigated for candidate genes, and while no evidence for causal cis-acting regulatory mutations was detected, deleterious nucleotide changes in coding sequences of the seedless haplotype were predicted in two genes within the sdi fine-mapping interval. Targeted resequencing of the two genes in a collection of 124 grapevine cultivars showed that only the point variation causing the arginine-197-to-leucine substitution in the seed morphogenesis regulator gene AGAMOUS-LIKE11 (VviAGL11) was fully linked with stenospermocarpy. The concurrent postzygotic variation identified for this missense polymorphism and seedlessness phenotype in seeded somatic variants of the original stenospermocarpic cultivar supports a causal effect. We postulate that seed abortion caused by this amino acid substitution in VviAGL11 is the major cause of seedlessness in cultivated grapevine. Abstract: Background and Aims: Berry weight in the grapevine, as in the tomato, is variable, ranging from less than 1 g to10 g. In the tomato, berry weight depends on cell number and volume but also on carpel number. The aim of this work was to decipher the range of variation and to propose a role for subtraits possibly underlying berry size variation in a highly diverse collection of cultivated grapevines.Methods and Results: Cell division before and after anthesis and cell expansion after anthesis appeared to be the major determinants of flesh weight variation between cultivars. Carpel number varied between cultivars, with two and three carpels per ovary. This trait, however, and also the seed weight did not clearly contribute to berry size variation, although a positive correlation was found between seed weight and number and berry weight at the intragenotypic level, in agreement with previous results.Conclusions: This work deciphered the main anatomical factors underlying variation in berry size in Vitis vinifera;they involved both common fleshy fruits factors but also specific vine factors.Significance of the Study: Further studies of the variation in berry size of the the grape will be assisted by the anatomical factors identified in this study.

Abstract: Background - Grapevine (Vitis vinifera subsp. vinifera) is one of the most important and ancient horticultural plants in the world. Domesticated about 8--10,000 years ago in the Eurasian region, grapevine evolved from its wild relative (V. vinifera subsp. sylvestris) into very diverse and heterozygous cultivated forms. In this work we study grapevine genetic structure in a large sample of cultivated varieties, to interpret the wide diversity at morphological and molecular levels and link it to cultivars utilization, putative geographic origin and historical events. Results - We analyzed the genetic structure of cultivated grapevine using a dataset of 2,096 multi-locus genotypes defined by 20 microsatellite markers. We used the Bayesian approach implemented in the STRUCTURE program and a hierarchical clustering procedure based on Ward's method to assign individuals to sub-groups. The analysis revealed three main genetic groups defined by human use and geographic origin: a) wine cultivars from western regions, b) wine cultivars from the Balkans and East Europe, and c) a group mainly composed of table grape cultivars from Eastern Mediterranean, Caucasus, Middle and Far East countries. A second structure level revealed two additional groups, a geographic group from the Iberian Peninsula and Maghreb, and a group comprising table grapes of recent origins from Italy and Central Europe. A large number of admixed genotypes were also identified. Structure clusters regrouped together a large proportion of family-related genotypes. In addition, Ward's method revealed a third level of structure, corresponding either to limited geographic areas, to particular grape use or to family groups created through artificial selection and breeding. Conclusions - This study provides evidence that the cultivated compartment of Vitis vinifera L. is genetically structured. Genetic relatedness of cultivars has been shaped mostly by human uses, in combination with a geographical effect. The finding of a large portion of admixed genotypes may be the trace of both large human-mediated exchanges between grape-growing regions throughout history and recent breeding.

Abstract: Contact: valerie.laucou@supagro.inra.fr; Using 20 SSR markers well scattered across the 19 grape chromosomes, we analyzed 4,370 accessions of the INRA grape repository at Vassal, mostly cultivars of Vitis vinifera subsp. sativa (3,727), but also accessions of V. vinifera subsp. sylvestris (80), interspecific hybrids (364), and rootstocks (199). The analysis revealed 2,836 SSR single profiles: 2,323 sativa cultivars, 72 wild individuals (sylvestris), 306 interspecific hybrids, and 135 rootstocks, corresponding to 2,739 different cultivars in all. A total of 524 alleles were detected, with a mean of 26.20 alleles per locus. For the 2,323 cultivars of V. vinifera, 338 alleles were detected with a mean of 16.9 alleles per locus. The mean genetic diversity (GDI) was 0.797 and the level of heterozygosity was 0.76, with broad variation from 0.20 to 1. Interspecific hybrids and rootstocks were more heterozygous and more diverse (GDI = 0.839 and 0.865, respectively) than V. vinifera cultivars (GDI = 0.769), Vitis vinifera subsp. sylvestris being the least divergent with GDI = 0.708. Principal coordinates analysis distinguished the four groups. Slight clonal polymorphism was detected. The limit between clonal variation and cultivar polymorphism was set at four allelic differences out of 40. SSR markers were useful as a complementary tool to traditional ampelography for cultivar identification. Finally, a set of nine SSR markers was defined that was sufficient to distinguish 99.8% of the analyzed accessions. This set is suitable for routine characterization and will be valuable for germplasm management

Abstract: International audience; The wild grapevine, Vitis vinifera subsp. silvestris, is an endangered species. In 1999, we initiated an inventory and a characterization of wild grapevine in France. Since this subspecies is dioecious and since, in many cases, wild grapevine have been identified near vineyards, we have been concerned about gene flow between cultivated and wild grapevines. In order to identify such a gene flow, we sampled individuals issued from open pollination in natural environment of the same mother. We identify pollinators by parentage analysis using the 20 SSR markers. Most of the pollen originated from male wild individuals, but evidence of gene flow, even if at a low level, was revealed.

Abstract: International audience; Parentage analysis in grape (Vitis vinifera L.) is becoming more and more popular, but until now relatively small databases were realized. Due to the increasing activities on genetic diversity description in grape, the amount of data should soon be more consistent. Analysis of large scale data set is more difficult and until now it has never been accomplished in grape. The 2,919 different varieties of Vitis vinifera from the germplasm collection of ‘Domaine de Vassal’ have been analyzed using 20 SSR markers well scattered in the genome. This database will be very useful in order to decipher kinship relationships if we can handle such database. In the present paper, we tested a methodology for the analysis of large dataset, using FaMoz software. The method enabled the successful identification of parentage for several selected cultivars with known parentage. The fact that our markers were carefully selected on different linkage group added value to the analysis.

Abstract: Dans la collection de vignes du Domaine de Vassal (INRA, France), l'espèce Vitis vinifera L. est représentée par 5311 introductions, soit 2236 cépages répertoriés et 1090 introductions en cours d'étude. Après description et identification, le potentiel viticole des cépages est évalué. Ces données, recueillies depuis plus de 40 ans, sont maintenant rassemblees dans une banque de données informatisée. Il est ainsi possible de calculer la répartition des principaux caractères sur un grand nombre de cépages (2236). Les génotypes analysés étant d'origines trés diverses, on peut considérer cet échantillon comme une des meilleures représentations disponibles de Vitis vinifera L. Les distributions, moyennes, valeurs modales et extrêmes sont présentées pour les caractères suivants: dates de débourrement et de maturité, port et fertilité, poids moyen des baies et des grappes. La connaissance de la répartition de ces caractères est très utile pour estimer la potentialité des semis , déterminer les cépages "extrêmes" susceptibles d'être utilisés dans des programmes d'amélioration et mieux apprécier Vitis vinifera L. Distribution of the main phenological, agronomical and technical characters of Vitis vinifera L. In the grapevine collection of the ''Domaine de Vassal'' (INRA, France) 5311 accessions belong to Vitis vinifera L. That represents 2236 varieties and 1090 accessions which are still in process of being studied. After being described and identified, the viticultural performances of the varieties are evaluated. These data have been collected since 40 years and are now registred and joined together in a computerized data bank. So, it is possible to calculate distributions and repartitions for the most important characters of a great number of varieties (2236). The origins of these varieties are very diversified and therefore this large sample can be considered like one of the best available representations of Vitis vinifera L. The distribution, average, modal, minimum and maximum values of the following characters are presented: date of budburst, date of maturity, shoot habit, fertility, mean berry weight and mean bunch weight. The knowledge of the repartition of these characters is very useful to estimate the potentiality of seedlings and to determine the ''extreme'' varieties which could be used in different programs of grape improvement. Abstract: L’ampélographie reste un problème d’actualité chez Vitis vinifera L. En effet, reconnaître et identifier une variété est souvent une opération délicate. Ceci est dû au grand nombre de cépages existants et à l’importance de la fluctuation présente au sein de chacun d’eux. Une aide à la reconnaissance est proposée en utilisant par des moyens informatiques un fichier ampélographique portant sur 5 226 clones (2 076 variétés). Ce fichier a été réalisé au Domaine de VASSAL (I.N.R.A., 34340 Marseillan-Plage). Les descriptions intéressent 75 caractères, ce qui représente 272 modalités. Après sauvegarde de ces données sur Mini 6, un programme en Pascal a été mis au point permettant de questionner ce fichier et d’obtenir ainsi des listes d’hypothèses pouvant correspondre aux échantillons testés. Une analyse statistique (A.F.C.) de ces données a permis de mettre en évidence les caractères ampélographiques les plus importants et d’obtenir une image de la population de Vitis vinifera L. contenue dans ce fichier. Deux directions dispersent ainsi le mieux les variétés : - la première oppose les cépages de cuve (petites baies) présentant généralement une densité des poils couchés forte, aux variétés de table (grosses baies), souvent glabres ; - la seconde met en opposition les variétés à caractères juvéniles marqués (pigmentation anthocyanique prononcée, densité des poils dressés importante) avec des cépages plus évolués., Ampelography remains a problem of current concern. The great number of varieties of Vitis vinifera L. and the great variability within cultivars cause difficulty in identifying and recognizing varieties. A method for aiding recognition of varieties using computerized ampelographic data has accordingly been developed. Data were collected from the Research Station of VASSAL (I.N.R.A., 34340 Marseillan-Plage). 5 226 clones representing 2 076 cultivars were described using 75 characters (with 272 values). The descriptions were recorded in a Mini 6 computer and a programme was developed to examine the file and obtain hypothetical listings which might agree with an unknown sample. Statistical analysis (C.F.A.) of these data showed which ampelographic characters were most useful and gave a picture of the population of Vitis vinifera L. The population of varieties was dispersed in two main directions. The first opposed wine-production cultivars with small berries and often a high density of prostrate hairs to table varieties (large berries), which are usually hairless. The second opposed varieties with marked juvenile characters (high anthocyanin colour, high density of erect hairs) to more developed varieties.

Abstract: International audience; Despite the importance of grapevine (Vitis vinifera), relatively few parentages were revealed until now because small databases were analysed. We developed a useful and readable routine for the analysis of large data set, integrating the FaMoz software. The routine was applied to a data set consisting of 2704 individuals of the French national INRA germplasm collection analysed using 20 SSR markers well-scattered over the genome. The large-scale parentage analysis enabled the identification of several kin relationships. The kingroup of Chardonnay, one of the most celebrated international grapevine cultivars, is discussed.
