Dengue cases occur frequently at Nuevo Leon, Mexico, where Aedes aegypti (L.) and Ae. albopictus (Skuse) are present. Ae. albopictus is considered the second vector of dengue. Because it bites humans outdoors during the day, the mosquito plays an important role in transmission of dengue virus (DENV). However, no previous studies at Nuevo Leon indicated the role of the mosquito outdoors. To assess Ae. albopictus for dengue virus, mosquitoes were collected from April to October 2010 at five localities at Guadalupe and Santiago, Nuevo Leon, (Northeast) Mexico, by using two methods: engine backpack aspirator and ovitraps. In total, 1,836 Ae. albopictus and 833 Ae. aegypti mosquitoes were collected by ovitrap and engine backpack aspirator methods. Groups of mosquitoes were processed by RT-PCR. Examination for DENV infection of mosquitoes showed one positive group of four female Ae. albopictus from an ovitrap. This research provided information that showed transovarial transmission of dengue virus in Ae. albopictus occurred naturally, maintaining endemic levels of disease at a study site.

Sensitivity of monitoring Aedes aegypti (L.) populations was determined to identify the distribution of dengue virus (DENV) during epidemics in Quintana Roo. From September to November 2012, we used a motorized aspirator to collect 2,144 female Ae. aegypti from 569 homes. These were grouped into 220 to use semi-nested RT-PCR for DENV, and positive groups were analyzed individually. Five groups (2.27%) were positive for DENV. Individual analysis yielded eight groups that tested positive, six with DENV-2, one DENV-1, and one DENV-4. The latter was not reported by the surveillance system that year. The mean number of female mosquitoes per household was 3.77 ± 5.71, and the rate of viral infection of Ae. aegypti was 0.4%. Most infected mosquitoes (49%) were concentrated in 10% of the houses. Monitoring Ae. aegypti infected with DENV has the potential to complement the current system of clinical and entomological surveillance.

Serology of West Nile virus vectors and non-human reservoirs was surveyed at Acapulco, Jose Azueta, and Ometepec, three Pacific Coast localities of Guerrero State, Mexico. The objectives of this study were to use enzyme-linked immnosorbent assay (ELISA) to assess West Nile virus antibodies of bird and equine serum samples and use reverse transcription of polymerase chain reaction (RT-PCR) to detect the virus in field-collected resting mosquitoes. Forty birds trapped using mist nets yielded 10% seroprevalence. Similarly, 18.6% of 102 equine blood samples had West Nile virus. In addition, 4,854 mosquitoes were caught using motorized backpack aspirators and grouped into 116 pools. Of the 16 species and seven genera, no mosquito was positive for West Nile virus. Our study demonstrated West Nile virus seroprevalence on resident birds and equines in Guerrero State, Mexico.

In October 2011, the State Health Department announced that several laboratory-confirmed cases of dengue had occurred among residents in two neighborhoods of Benito Juarez, Quintana Roo State, Mexico. To identify the dengue virus serotype(s) temporally and spatially associated with the cases, entomologic-based virus surveillance was initiated in October 2011 in both neighborhoods. Adult mosquitoes were collected from 88 houses by CDCbackpack aspirator, and all female Aedes aegypti L. (n = 419) were individually homogenized and assayed in pools of as many as 10 by reverse transcriptionpolymerase chain reaction (RT-PCR) using dengue virus-specific primers. Five (12%) of 41 pools were positive for dengue virus RNA. The individual mosquitoes that comprised the pools were analyzed separately by RT-PCR using dengue virus serotype-specific primers. Six mosquitoes were positive for dengue virus serotype-2 (DENV-2) RNA, three of which were collected in the same house. The mean number of female Ae. aegypti collected in each house was 4.76 ± 6.19. The overall dengue virus-infection rate in female Ae. aegypti was 1.4%. Interestingly, most (60%) of mosquito females were collected only from 15 (17%) houses. In summary, we provide evidence of recent DENV-2 transmission in Quintana Roo State.

Secondary dengue virus infections are a major risk for developing dengue hemorrhagic fever. Recent exposure to infectious bites of Aedes aegypti (L.) females in previously diagnosed dengue cases fulfills the epidemiological model of dengue hemorrhagic fever. A study was comprised of 357 (89.2%) dengue and 43 (10.8%) dengue hemorrhagic fever cases confirmed by laboratory tests and clinical manifestations. An entomological survey was done in homes and backyards. Concurrently, a questionnaire was used to assess the impact of healthpromotion campaigns through knowledge of the vector and its epidemiological role. Seventy-six (28.4%) of the 268 (67.0%) total wet or dry oviposition sites were positive for the presence of larvae or pupae, while adult Ae. aegypti were found in 32 (8.0%). One hundred thirty-two (33%) householders who formerly had dengue fever or dengue hemorrhagic fever had knowledge of either larval or adult dengue vector stages. According to gender distribution, 145 (36.2%) and 14 (3.5%) of the males confirmed with cases of dengue and dengue hemorrhagic fever lived in houses with 17.9 and 2% of the Ae. aegypti larval and pupal habitats. Houses with females who had dengue and dengue hemorrhagic fever were 212 (53%) and 29 (7.3%), with containers with immature Ae. aegypti in 19.4 and 7%, respectively. Background: Aedes aegypti, the ‘yellow fever mosquito’, is the primary vector to humans of dengue and yellow fever flaviviruses (DENV, YFV), and is a known vector of the chikungunya alphavirus (CV). Because vaccines are not yet available for DENV or CV or are inadequately distributed in developing countries (YFV), management of Ae. aegypti remains the primary option to prevent and control outbreaks of the diseases caused by these arboviruses. Permethrin is one of the most widely used active ingredients in insecticides for suppression of adult Ae. aegypti. In 2007, we documented a replacement mutation in codon 1,016 of the voltage-gated sodium channel gene (para) of Ae. aegypti that encodes an isoleucine rather than a valine and confers resistance to permethrin. Ile1,016 segregates as a recessive allele conferring knockdown resistance to homozygous mosquitoes at 5–10 mg of permethrin in bottle bioassays. Methods and Findings: A total of 81 field collections containing 3,951 Ae. aegypti were made throughout Me´xico from 1996 to 2009. These mosquitoes were analyzed for the frequency of the Ile1,016 mutation using a melting-curve PCR assay. Dramatic increases in frequencies of Ile1,016 were recorded from the late 1990’s to 2006–2009 in several states including Nuevo Leo´n in the north, Veracruz on the central Atlantic coast, and Yucata´n, Quintana Roo and Chiapas in the south. From 1996 to 2000, the overall frequency of Ile1,016 was 0.04% (95% confidence interval (CI95) = 0.12%; n = 1,359 mosquitoes examined). The earliest detection of Ile1,016 was in Nuevo Laredo on the U.S. border in 1997. By 2003–2004 the overall frequency of Ile1,016 had increased ,100-fold to 2.7% (60.80% CI95; n = 808). When checked again in 2006, the frequency had increased slightly to 3.9% (61.15% CI95; n = 473). This was followed in 2007–2009 by a sudden jump in Ile1,016 frequency to 33.2% (61.99% CI95; n = 1,074 mosquitoes). There was spatial heterogeneity in Ile1,016 frequencies among 2007–20

Pyrethroid insecticides prolong the opening of voltage-dependent sodium channels in insect nerves to produce instant paralysis and “knock-down.” Many insects have evolved knock-down resistance through nonsynonymous mutations that reduce pyrethroid binding in the channels. In 2006 we discovered one such mutation in the arbovirus mosquito vector Aedes aegypti, called Ile1,016, that confers very high knockdown resistance to the pyrethroid insecticide permethrin in mosquitoes homozygous for this mutation. We examined collections of Ae. aegypti from México during 1996–2009 and found that the overall Ile1,016 frequency increased from <0.1% in 1996–2000, to 2%–5% in 2003–2006, to 38.3%–88.3% in 2007–2009 depending upon collection location. We also demonstrate a strong linear relationship between the frequency of Ile1,016 homozygotes and knockdown rate in bioassays and speculate that widespread use of permethrin-based insecticides in México may be impacting the frequency of Ile1,016. Such a rapid increase is predicted by a simple model of positive directional selection acting on a recessive allele. Unfortunately this model also predicts rapid fixation of Ile1,016 unless there is a negative fitness associated with Ile1,016 in the absence of permethrin and if insecticidal pressure can be reduced.

OBJECTIVE: Novel, inexpensive solutions are needed for improved management of vector-borne and other diseases in resource-poor environments. Emerging free software providing access to satellite imagery and simple editing tools (e.g. Google EarthTM) complement existing geographic information system (GIS) software and provide new opportunities for: (i) strengthening overall public health capacity through development of information for city infrastructures; and (ii) display of public health data directly on an image of the physical environment. METHODS: We used freely accessible satellite imagery and a set of feature-making tools included in the software (allowing for production of polygons, lines and points) to generate information for city infrastructure and to display disease data in a dengue decision support system (DDSS) framework. FINDINGS: Two cities in Mexico (Chetumal and Merida) were used to demonstrate that a basic representation of city infrastructure useful as a spatial backbone in a DDSS can be rapidly developed at minimal cost. Data layers generated included labelled polygons representing city blocks, lines representing streets, and points showing the locations of schools and health clinics. City blocks were colour-coded to show presence of dengue cases. The data layers were successfully imported in a format known as shapefile into a GIS software. CONCLUSION: The combination of Google EarthTM and free GIS software (e.g. HealthMapper, developed by WHO, and SIGEpi, developed by PAHO) has tremendous potential to strengthen overall public health capacity and facilitate decision support system approaches to prevention and control of vector-borne diseases in resource-poor environments.
