Abstract

We present radial velocity measurements of the very bright ($V\sim5.7$) nearby F star, DMPP-4 (HD 184960). The anomalously low Ca II H&K emission suggests mass loss from planets orbiting a low activity host star. Periodic radial velocity variability with $\sim 10$ ms$^{-1}$ amplitude is found to persist over a $>4$ year timescale. Although the non-simultaneous photometric variability in four TESS sectors supports the view of an inactive star, we identify periodic photometric signals and also find spectroscopic evidence for stellar activity. We used a posterior sampling algorithm that includes the number of Keplerian signals, $N_\textrm{p}$, as a free parameter to test and compare (1) purely Keplerian models (2) a Keplerian model with linear activity correlation and (3) Keplerian models with Gaussian processes. A preferred model, with one Keplerian and quasi-periodic Gaussian process indicates a planet with a period of $P_\textrm{b} = 3.4982^{+0.0015}_{-0.0027}$ d and corresponding minimum mass of $m_\textrm{b}\,\textrm{sin}\,i = 12.2^{+1.8}_{-1.9}$ M$_\oplus$. Without further high time resolution observations over a longer timescale, we cannot definitively rule out the purely Keplerian model with 2 candidates planets with $P_\textrm{b} = 2.4570^{+0.0026}_{-0.0462}$ d, minimum mass $m_\textrm{b}\,\textrm{sin}\,i = 8.0^{+1.1}_{-1.5}$ M$_\oplus$ and $P_\textrm{c} = 5.4196^{+0.6766}_{-0.0030}$ d and corresponding minimum mass of $m_\textrm{b}\,\textrm{sin}\,i = 12.2^{+1.4}_{-1.6}$ M$_\oplus$. The candidate planets lie in the region below the lower-envelope of the Neptune Desert. Abstract

The Dispersed Matter Planet Project targets stars with anomalously low Ca II H&K chromospheric emission. High precision, high cadence radial velocity measurements of the F8V star HD 38677 / DMPP-1 reveal four short period planets. DMPP-1 has log(R'HK) = -5.16 which probably indicates the presence of circumstellar absorbing gas arising from an ablating hot planet. The planets have P$_{\rm orb}$ ~ 2.9 - 19 d, i.e., a compact planetary system with super-Earth (~3 M$_\oplus$) to Neptune-mass (~24 M$_\oplus$) planets. These irradiated planets may be chthonian: remnant cores of giant planets after mass-loss while crossing the Neptune desert. Modelling the possible long-term activity indicators while searching for Keplerian signals modifies the recovered planetary signals. A priori inferences about the presence of short period planets allowed the efficient discovery of the DMPP-1 planets. Abstract

We generalize the KPZ equation to an O(3) $N=2j+1$ component model. In the limit $N \to \infty$ we show that the mode coupling equations become exact. Solving these approximately we find that the dynamic exponent $z$ increases from $3/2$ for $d=1$ to $2$ at the dimension $d\approx3.6$. For $d=1$ it can be shown analytically that $z=3/2$ for all $j$. Methods: The cost-effectiveness of pregabalin versus usual care for treatment of CLBP-NeP was evaluated over a 12-month time horizon using the incremental cost-effectiveness ratio (ICER). Quality-adjusted life years (QALYs), derived from the five-dimension, five-level EuroQol (EQ-5D-5L) questionnaire, was the measure of effectiveness. Medical costs and productivity losses were both calculated. Expected costs and outcomes were estimated via cohort simulation using a state-transition model, which mimics pain state transitions among mild, moderate, and severe pain. Distributions of pain severity were obtained from an 8-week noninterventional study. Health care resource consumption for estimation of direct medical costs for pain severity levels was derived from a physician survey. The ICER per additional QALY gained was calculated and sensitivity analyses were performed to evaluate the robustness of the assumptions across a range of values. Results: Direct medical costs and hospitalization costs were both lower in the pregabalin arm compared with usual care. The estimated ICERs in the base case scenarios were approximately ¥2,025,000 and ¥1,435,000 per QALY gained with pregabalin from the payer and societal perspectives, respectively; the latter included indirect costs related to lost productivity. Sensitivity analyses using alternate values for postsurgical pain scores (0 and 5), initial pain severity levels (either all moderate or all severe), and the actual EQ-5D-5L scores from the noninterventional study showed robustness of results, with ICERs that were similar to the base case. Development of a cost-effectiveness acceptability curve showed high probability (≥75%) of pregabalin being cost-effective. Conclusion: Using data and assumptions from routine clinical practice, pregabalin is cost-effective for the treatment of CLBP-NeP in Japan. Abstract

The effect of hydrostatic pressure in the range of 0–10 GPa on the structural, electric, and electronic properties of hybrid organic–inorganic ferroelectric perovskites TMCM–MnCl3and TMCM–CdCl3(TMCM = trimethyl chloromethyl ammonium) is predicted from first-principles density functional theory calculations. These materials differentiate themselves in the family of organic–inorganic ferroelectrics by their unusually high Curie temperature and piezoelectric constants and therefore are of both scientific and technological significance. Computations predict (i) the (co)existence of multiple competing phases with different polarizations and band gaps tunable by the pressure; (ii) the presence of two regions of structural response to pressure and their atomistic origin; (iii) increase of spontaneous polarization of up to 18% (19%) for TMCM–CdCl3(TMCM–MnCl3) driven by the significant volume decrease under pressure; and (iv) 36% decrease in the band gap of TMCM–MnCl3and 10% increase in the band gap of TMCM–CdCl3under 10 GPa.

Abstract

Aotearoa’s (New Zealand’s) biological heritage is in decline due to threats such as climate change and habitat destruction. Aotearoa’s biological heritage and the wider environment are critical to the Māori world view and culture and Māori have long advocated for greater engagement in efforts to reverse this decline. One negative outcome of localised declines in biological heritage is a concomitant loss of local Māori language (dialectical) terms. Compounding this is the growing use of standardised Māori terms that can displace local dialectical terms. This also runs the risk of losing the associated mātauranga (knowledge) that is inherent in the meaning of these local terms for their unique flora and fauna. Retaining this biocultural knowledge is considered important and could play a role in conservation efforts. This collaborative research addressed the concerns articulated by a Māori biological heritage expert about the loss of their own unique local Māori terms for flora and fauna. The research explored ways to retain and empower local indigenous biocultural terms via the creation of a static visual educational resource for Tūhoe–Tuawhenua youth displaying the forest vegetation of their rohe (area that defines a tribe’s traditional mandate or authority). The plants in the final resource are identified by their local Māori term and their corresponding scientific name. Depicting ecological accuracy in the artwork was a specific requirement of the kaumātua and created some unique outcomes in how the artwork formed. The approaches employed in this research and an analysis of the results and wider implementation are discussed.

38. Novel Temperature-Dependent Development Rate Models for Postdiapause Egg Eclosion of Three Important Arthropod Pests Found in Commercial Christmas Tree Plantations of Southern Québec, Canada

Abstract

The balsam twig aphid Mindarus abietinusKoch (Hemiptera: Aphididae), the spruce spider mite Oligonychus ununguis(Jacobi) (Acari: Tetranychidae), and the pine needle scale Chionaspis pinifoliae(Fitch) (Hemiptera: Diaspididae) are three important pests for the Christmas tree industry of Québec, Canada. Temperature-dependent development rates for postdiapause overwintering eggs of these species have never been studied for populations of southern Québec, where the vast majority of Christmas trees are grown. The accumulation of physiological time in growing degree days (GDD) is a standard tool to predict egg eclosion dates for pests, when first generation immatures are most vulnerable to insecticidal control. Development rates for postdiapause eggs were tested at four or five constant temperatures in the 11–23°C range under controlled conditions, based on time before hatching for eggs collected in a balsam fir plantation in late winter. The standard linear model and three published nonlinear models were fitted to the data and compared for their ability to estimate key biological temperature parameters and to predict development rates of postdiapause eggs. Validation of model predictions of egg eclosion time on balsam fir in the field was reasonably accurate using the classical linear model where field-accumulated GDD were calculated with a particular interpretation of the average method based on daily maximum and minimum temperatures. Le développement des œufs hivernants postdiapausants, en fonction de la température, n’a jamais été étudié pour les populations du sud du Québec, où cette culture est concentrée. L’accumulation du temps physiologique en degrés-jours de croissance (DJ) est un outil essentiel pour prédire la date d’éclosion des œufs de ravageurs, alors que les immatures de première génération sont vulnérables aux traitements insecticides. Le taux de développement des œufs hivernants a été mesuré en conditions contrôlées pour quatre ou cinq températures constantes dans la gamme 11–23°C, calculé selon le temps de développement jusqu’à l’éclosion d’œufs collectés sur le sapin baumier. Le modèle linéaire standard et trois modèles non linéaires publiés ont été évalués pour leur capacité à prédire les paramètres biologiques de réponse à la température et le taux de développement. La prédiction de l’éclosion des œufs sur le terrain a été validée avec le calcul du temps physiologique accumulé en DJ avec une interprétation particulière de la méthode des moyennes basée sur les températures quotidiennes maximales et minimales, en association avec le modèle linéaire. Cela a permis de prédire l’éclosion printanière sur le terrain à environ cinq jours près.

Udgivelsesdato: 1996-May-15, A high background of read-through transcripts from degenerate human L1 retrotransposons is present in almost all human cell types. This prevents the detection of RNA transcripts from potentially functional elements. To overcome this, we have developed an RNase protection assay based on the reconstructed consensus sequence for the 5' end of the major L1 family. In the human Ntera2D1 teratocarcinoma cell line, this assay readily detected L1 transcripts that were located primarily in the cytoplasm and where 20% were in filterable particles. By this assay, potentially functional L1 elements are also transcriptionally active in lymphocytes from some but not all normal individuals. Together with the full length protection product, there were three other discrete L1 RNAs, two of which (305 and 275 bases) were transcribed from the 5' end of the L1 element. These smaller L1 RNAs do not appear to be derived from transcripts from divergent L1 families but are either discrete shorter transcripts or specifically processed products from longer initial transcripts.

46. Modified-cytosine restriction-system-induced recombinant cloning artefacts in Escherichia coli.

Abstract

We have tested whether, and to what extent, recombinant clones from DNA segments with 5-methylation of cytosines recovered in methylation-restrictive (mcr+) hosts contain mutations. We constructed a model system in which the tetracycline-resistance-encoding gene (tet) from pBR322 was cloned into the plasmid pGEM3Zf+. The central region of tet was removed from the construct, methylated in vitro and then religated back into the unmethylated remainder of the construct. The central region of tet was either (1) methylated with a combination of four bacterial methyltransferases (M.AluI, M.HaeIII, M.HpaII plus M.HhaI) or (2) methylated with M.SssI which methylates at all CpG dinucleotides. These two protocols generated theoretical levels of DNA methylation in the central fragment of 10.5% and 33%, respectively. The construct was transformed into a series of isogenic (recA+) bacterial strains that were mcrA+ mcrB+C+, mcrA+ mcrB-C+, mcrA- mcrB+C+, mcrA- mcrB-C+ or mcrA- delta mcrBC, and also into a set of isogenic recA- derivatives of these strains. With the two methylation protocols, there was an average 48- and 141-fold reduction, respectively, in the number of transformants recovered from the recA+ mcr+ hosts compared with a methylation-tolerant host (mcr-). Of the clones recovered in recA+mcr+ hosts, > 20% of clones had an inactivating mutation in tet. The majority of such mutant clones contained deletions that frequently extended into the unmethylated portion of tet and even into the plasmid sequences beyond the end of the polylinker. Abstract

Efficient recovery of clones from the 5' end of the human L1 dispersed repetitive elements necessitates the use of deletion mcr- host strains since this region contains a CpG island which is hypermethylated in vivo. Clones recovered with conventional mcr+ hosts seem to have been derived preferentially from L1 members which have accumulated mutations that have removed sites of methylation. We present a revised consensus from the 5' presumptive control region of these elements. This revised consensus contains a consensus RNA polymerase III promoter which would permit the synthesis of transcripts from the 5' end of full length L1 elements. Such potential transcripts are likely to exhibit a high degree of secondary structure. In addition, we have determined the flanking sequences for 6 full length L1 elements. The majority of full length L1 clones show no convincing evidence for target site duplication in the insertion site as commonly observed with truncated L1 elements. These data would be consistent with two mechanisms of integration of transposing L1 elements with different mechanisms predominating for full length and truncated elements. Abstract

While numerous susceptibility loci for epithelial ovarian cancer (EOC) have been identified, few associations have been reported with overall survival. In the absence of common prognostic genetic markers, we hypothesize that rare coding variants may be associated with overall EOC survival and assessed their contribution in two exome-based genotyping projects of the Ovarian Cancer Association Consortium (OCAC). METHODS: The primary patient set (Set 1) included 14 independent EOC studies (4,293 patients) and 227,892 variants, and a secondary patient set (Set 2) included six additional EOC studies (1,744 patients) and 114,620 variants. Because power to detect rare variants individually is reduced, gene-level tests were conducted. Sets were analyzed separately at individual variants and by gene, and then combined with meta-analyses (73,203 variants and 13,163 genes overlapped). RESULTS: No individual variant reached genome-wide statistical significance. A SNP previously implicated to be associated with EOC risk and, to a lesser extent, survival, rs8170, showed the strongest evidence of association with survival and similar effect size estimates across sets (Pmeta = 1.1E-6, HRSet1 = 1.17, HRSet2 = 1.14). Rare variants in ATG2B, an autophagy gene important for apoptosis, were significantly associated with survival after multiple testing correction (Pmeta = 1.1E-6; Pcorrected = 0.01). CONCLUSIONS: Common variant rs8170 and rare variants in ATG2B may be associated with EOC overall survival, although further study is needed. IMPACT: This study represents the first exome-wide association study of EOC survival to include rare variant analyses, and suggests that complementary single variant and gene-level analyses in large studies are needed to identify rare variants that warrant follow-up study., Other Research Unit
